    冠状病毒(coronavirus, CoVs)是有包膜的正链RNA病毒,其包含最大的已知RNA基因组,最大长度为32 kb。冠状病毒亚科包含了感染人类及动物的重要病毒。可分为四类:α,β,γ和δ冠状病毒(α-,β-,γ-和δ-CoV)。 冠状病毒颗粒至少包含四个标准结构蛋白E(包膜蛋白),M(膜蛋白),N(核衣壳蛋白)和S(刺突蛋白)。 S蛋白既包含受体结合结构域 (receptor-binding domain, RBD),又包含融合相关结构域,使其成为CoV进入过程中的关键蛋白。而本文主要阐述该病毒的最主要毒力因子S(spike)蛋白通过与受体结合、自身水解切割和构象变化入侵人体。目前,针对S蛋白的治疗主要包括中和抗体、减毒或灭活疫苗、亚单位疫苗等手段,新的治疗策略仍需进一步临床验证。

    Abstract:

    Coronavirus (CoVs) are enveloped positive-stranded RNA viruses that contain the largest known RNA genome with a maximum length of 32 kb. Coronavirus subfamily contains important viruses that infect humans and animals. Can be divided into four categories: α, β, γ and δ coronavirus (α-, β-, γ- and δ-CoV). Coronavirus particles contain at least four standard structural proteins E (envelope protein), M (membrane protein), N (nucleocapsid protein) and S (spike protein). The S protein contains both a receptor-binding domain (RBD) and a fusion-related domain, making it a key protein in the process of CoV entry. This article mainly elaborates that the most important virulence factor S (spike) protein of the virus invades the human body through binding to receptors, autohydrolytic cleavage and conformational changes. At present, treatments for S protein mainly include neutralizing antibodies, attenuated or inactivated vaccines, and subunit vaccines. New treatment strategies still require further clinical verification.
