    目的:本研究旨在探讨Klf2能否通过调节炎性反应在ACS发挥作用,为今后防治提供靶点。方法:1. 收集在我院心内科确诊为ACS的患者,选取同时间段冠脉造影狭窄程度<50%的患者为正常组。采用RT-PCR技术及免疫印迹技术检测ACS患者治疗前后患者外周血单个核细胞群中KLF2 mRNA及蛋白表达水平。2. 通过流式细胞术检测ACS患者外周T细胞表面分子的表达水平。3.通过采用RT-PCR技术分别检测各组中心肌标志物及KLF2的mRNA表达,比较ACS组中的KLF2与心肌标志物的相关性。结果:ACS患者的外周血中KLF2 mRNA及蛋白表达水平明显低于对照组,ACS患者外周T细胞表面CD62L和CCR7低于对照组,而CXCR3和CD44的表达水平则明显高于对照组;KLF2与各心肌标志物紧密相关。结论:KLF2在ACS诊断方面可能比心肌标志物效能较高,并为ACS的防治提供新思路。

    Abstract:

    Objective This study was designed to investigate whether Klf2 can play an important role in acute coronary syndrome by regulating inflammatory response, providing a potential treatment for the myocardial infarction in the future. Methods The expressions of Klf2 mRNA and protein were determined by quantitative real time PCR and Western blotting. Flow cytometry was used to detect the expressions of molecules on peripheral T cells of ACS patients. The mRNA expressions of myocardial markers and KLF2 were detected and compared by real-time quantitative PCR and the correlation between KLF2 and myocardial markers was compared in ACS group at the same time. Results The expression of KLF2 protein and mRNA levels of peripheral blood in patients with ACS were significantly lower than those in the control group. The expressions of CD62L and CCR7 on peripheral T cells in ACS patients were lower than those in normal controls, while the expression levels of CXCR3 and CD44 were significantly higher than those in normal controls, and KLF2 was closely related to the myocardial markers. Conclusion KLF2 may be more effective than myocardial markers in the diagnosis of acute coronary syndrome, which provides a new idea for the treatment of ACS.
