      目的:研究心房颤动(房颤)复律后心房肌细胞凋亡水平的变化,以探讨房颤复律后心房组织学重构逆转与心房肌细胞凋亡关系?方法:18只山羊随机分为3组(每组6只):窦性心律组(A组);慢性房颤组(B组);房颤后恢复窦性心律组(C组)?三组均行开胸手术,左心耳缝入起搏电极,A组不起搏,3个月后处死;B?C组持续快速起搏(400 ± 10次/min)制作慢性房颤模型,B组起搏3个月后处死;C组起搏3个月后停止起搏,用心律平静脉注射或直流电复律,维持窦律3个月后处死?取左心房组织,用TUNEL法标记并用流式细胞仪检测心房肌细胞凋亡?结果:慢性房颤组左房心肌细胞凋亡比窦性心律组显著增高(P < 0.05),房颤后复律组左房心肌细胞凋亡仍高于窦性心律组,但比慢性房颤组有所下降(P < 0.05)?结论:房颤复律前后心房肌细胞凋亡水平随心房组织学重构的进展而升高,随重构的逆转而下降?
英文摘要:
      Objective:To investigate the changes of myocardial cell apoptosis in order to explore the association between the atrial reverse remodeling and the myocardial cell apoptosis after chronic atrial fibrillation(AF) conversion to sinus rhythm. Methods:Eighteen goats were randomly divided into three groups(each n = 6):(A)sinus rhythm(SR),(B)Chronic AF,(C)sinus rhythm after AF. Three groups of goats were all underwent open chest surgery to implant the pacemaker electrodes into the left appendages. Group B and C were given continuous and rapid pacing(400 ± 10 bpm) in order to build chronic AF model. Group A was killed after three months without pacing. Group B was killed at the same time. Group C was stopped pacing and converted into sinus rhythm by propafenone and/or direct current. After three months’ sinus rhythm,group C was killed. The atrial tissue was obstained from the left atrium,and was stained by EdUTP-in situ end-labeling and observed by flow cytometry. Results:The apoptotic ratio of the left atrial myocytes in the goats with chronic AF was significantly higher than that in the goats with sinus rhythm(P < 0.05). It was still higher in the goats with 3 months’ SR post-AF than that in the goats with sinus rhythm,but lower than those with chronic AF(P < 0.05). Conclusion:The changes of myocardial cell apoptosis before and after AF conversion suggests that myocardial cell apoptosis is associated with the atrial remodeling and the reverse remodeling.
