      目的:探讨小剂量甲地孕酮(MA)和三苯氧胺(TAM)联合应用对乳腺癌细胞MCF-7生长和凋亡的影响?方法:分别或联合应用TAM?MA作用于MCF-7细胞,应用四甲基偶氮唑蓝(MTT)比色法分析细胞生长和抑制作用,应用流式细胞技术测定细胞周期和凋亡率?结果:TAM可抑制MCF-7细胞生长,阻滞细胞周期于G0/G1期,并可诱导细胞凋亡;小剂量的MA对MCF-7细胞生长与凋亡无明显影响,G0/G1期细胞增多;两者联合应用时,细胞的生长抑制作用加强,凋亡率上升?结论:TAM对MCF-7 细胞具有抑制生长和诱导凋亡作用,加用小剂量的MA后,其抑制生长和凋亡诱导作用加强,两者联合应用具有协同作用?
英文摘要:
      Objective:To study the effect of tamoxifen(TAM) and megestrol acetate(MA) on breast cancer cell line MCF-7. Methods:Estrogen receptor(ER)-positive MCF-7 human breast cancer cell line was treated by TAM or MA alone,or in combination in vitro. Cell proliferation was evaluated by MTT assay. Distribution of cell cycle and rate of apoptosis were determined by flow cytometry(FCM). Results:TAM can inhibit MCF-7 cells proliferation and induce its apoptosis. The cells were arrested in G0/G1 phase;small doses of MA had no significant effect on apoptosis,G0/G1 phase cells increased. When TAM combined with MA,the inhibition effect on proliferation and apoptosis were enhanced. Conclusion:TAM can inhibit MCF-7 cells proliferation and induce its apoptosis. The therapy effect can be improved when two drugs are combined.
