    目的:观察人胎肝间充质干细胞在tat-pdx1蛋白的作用下体外分化为胰岛β细胞的能力以及治疗nod/scid鼠糖尿病的效果-方法:体外分离纯化获得人胎肝间充质干细胞,用tat-pdx1蛋白体外诱导分化为胰岛β细胞,制作nod/scid鼠糖尿病模型,并将诱导后的细胞移植到糖尿病鼠肾包膜下,观察各组小鼠血糖变化并行ipgtt试验-结果:用tat-pdx1蛋白体外诱导分化的细胞稳定表达胰岛素,移植到糖尿病鼠肾包膜下,能快速降低血糖水平,取出移植物,血糖水平再次升高-结论:tat-pdx1蛋白诱导的人胎肝间充质干细胞能有效降低糖尿病鼠血糖,有望成为糖尿病细胞治疗的种子细胞-

    Abstract:

    objective:to observe the therapeutic effects of human fetal liver mesenchymal stem cells (hflmscs) induced by tat-pdx1 protein when transplanted into nod/scid diabetic mice. Methods:the hflmscs which were isolated and purified from fetal liver by adherent method,were induced to differentiate into islet β cells by tat-pdx1 protein. Nod/scid mice models of diabetes mellitus were established by injecting stz. The cells induced by tat-pdx1 protein were transplanted into left renal capsule,the blood glucose of diabetic nod/scid mice was detected. Results:the hflmscs induced by tat-pdx1 protein were expressed the mark of insulin in vitro. The level of mice blood glucose was degraded when the cells were transplanted into left renal capsule and was increased again when the left renal was excised. Conclusion:the hflmscs induced by tat-pdx1 protein can effectively reduce the level of nod/scid mice blood glucose. In the future,they will be the seed cells to treat diabetes mellitus.
