      目的:研究甲状旁腺素(PTH)和不同钙磷饮食对活性维生素D合成的影响?方法:使用正常?高钙低磷或低钙低磷饮食喂养2月龄野生型(WT)和PTH基因敲除纯合子(PTH-/-)小鼠4周,测定血清钙?磷?PTH和1,25-二羟基维生素D3[1,25(OH)2D3]水平,并利用RT-PCR方法检测肾1α-羟化酶[1α(OH)ase]基因表达水平?结果:正常饮食PTH-/-小鼠表现为低钙血症?高磷血症,肾1α(OH)ase基因表达上调,而血清1,25(OH)2D3水平则降低?与正常饮食相同基因型小鼠相比,高钙低磷饮食WT和PTH-/-小鼠血清PTH和1,25(OH)2D3水平降低,肾1α(OH)ase基因表达下调?高钙低磷饮食矫正了PTH-/-小鼠的低钙和高磷血症,肾1α(OH)ase基因表达仍高于WT小鼠,而血清1,25(OH)2D3水平与WT小鼠无差异?与正常饮食相同基因型小鼠相比,低钙低磷饮食WT和PTH-/-小鼠血清PTH和1,25(OH)2D3水平明显升高,肾1α(OH)ase基因表达明显上调?低钙低磷饮食PTH-/-小鼠血清钙严重降低,血清磷仍然较高,肾1α(OH)ase基因表达和血清1,25(OH)2D3水平均较WT小鼠为低?结论:钙磷饮食能够通过PTH依赖性和PTH非依赖性的方式调节活性维生素D的合成?

      Objective:To determine effects of parathyroid hormone (PTH) and different dietary calcium and phosphate on synthesis of active vitamin D. Methods:Two-month-old wild-type(WT) and PTH null (PTH-/-) mice had been fed on a normal diet (1% calcium,0.64% phosphate),high calcium low phosphate diet(2% calcium,0.4% phosphate) or low calcium low phosphate diet (0.005-0.01% calcium,0.4% phosphate) for 4 weeks,and serum calcium,phospharus,PTH and 1,25(OH)2D3 levels were measured. The expression of renal 25-hydroxyvitamin D 1α-hydroxylase[1α(OH)ase]gene was assessed using RT-PCR. Results:Serum calcium levels were decreased,serum phospharus levels were raised. The expression of renal 1α(OH)ase gene was up-regulated,whereas serum 1,25(OH)2D3 levels were reduced in PTH-/- mice on the normal diet compared with their WT littlemates on the same diet. Serum PTH and 1,25(OH)2D3 levels were reduced and the expression of renal 1α(OH)ase gene was down-regulated in WT and PTH-/- mice on the high calcium low phosphate diet compared with genotype-matched mice on the normal diet. Serum calcium and phospharus levels were normal,and the expression of renal 1α(OH)ase gene was still up-regulated with same level of 1,25(OH)2D3 in PTH-/- mice on the high calcium low phosphate diet compared with their WT littlemates on the same diet. Serum PTH and 1,25(OH)2D3 levels were raised significantly and the expression of renal 1α(OH)ase gene was up-regulated dramatically in WT and PTH-/- mice on the low calcium low phosphate diet compared with genotype-matched mice on the normal diet. Serum calcium levels were reduced more dramatically,and serum phospharus levels were less higher,with expression of renal 1α(OH)ase gene and lower serum 1,25(OH)2D3 levels in PTH-/- mice on the low calcium low phosphate diet compared with their WT littlemates on the same diet. Conclusion:Synthesis of active vitamin D was regulated by dietary calcium and phosphate in both PTH-dependent and PTH-independent manners.
