    目的:探讨P物质(substance P,SP)对过敏小鼠腹腔灌洗液中巨噬细胞表面蛋白酶激活受体(protease activated receptors,PARs)表达的影响-方法:通过腹腔注射鸡卵清蛋白(ovalbumin,OVA)建立过敏小鼠模型,再根据腹腔注射的SP的浓度将小鼠分为生理盐水组-0.2-2.0-20.0和200.0 μg/ml SP组,每组6只,同时每组均设有未致敏的对照组-最后一次激发3 h后处死小鼠,取腹腔灌洗液细胞经固定,用流式细胞仪检测巨噬细胞表面PARs的表达-结果:过敏组小鼠腹腔灌洗液中巨噬细胞表面PARs的表达水平较正常组明显降低(P < 0.01),而不同浓度的SP组之间无统计学差异(P > 0.05)-结论:SP对小鼠腹腔巨噬细胞PARs表达无影响,但过敏原可影响巨噬细胞表面PARs的表达-

    Abstract:

    Objective: To study the effect of substance P on the expression of protease activated receptors(PARs) in peritoneal macrophages from normal and allergic mice. Methods:Allergic mice were induced by intraperitoneal injection of ovalbumin(OVA). Mice were divided into normal saline, 0.2 μg/ml, 2.0 μg/ml, 20.0 μg/ml and 200.0 μg/ml SP groups respectively, with 6 mice in each group. The expressions of PARs on the surface of peritoneal macrophages were then analyzed by flowcytometry. Results:PAR-1, PAR-2, PAR-3 and PAR-4 expressions on the surface of peritoneal macrophages were all significantly decreased in allergic mice compared with normal mice(P < 0.01). Substance P had no significant influence on expressions of PARs in macrophages(P > 0.05). Conclusion:The intraperitoneal injection of substance P had no effect on the expressions of PARs in macrophages from mouse peritoneum, but OVA treatment did.
