      目的:探讨胃癌细胞中Ⅲ型β-微管蛋白(Class Ⅲ β-tubulin)?微管相关蛋白tau(MAPT)和凋亡抑制蛋白survivin的表达水平对化疗药物紫杉醇敏感性的预测价值?方法:采用ATP生物荧光法检测54例胃癌原代细胞对紫杉醇的体外药物敏感性,RT-PCR法检测胃癌细胞内Class Ⅲ β-tubulin?MAPT和survivin的mRNA表达水平,分析胃癌细胞对紫杉醇的体外敏感性与上述生物学指标表达水平的关系?结果:Class Ⅲ β-tubulin的mRNA表达水平为1.04 ± 0.22,与分化程度有关(P = 0.029);MAPT的mRNA表达水平为0.80 ± 0.17;survivin的mRNA表达水平为1.00 ± 0.19,与分化程度有关(P = 0.009)?药敏指数与Class Ⅲ β-tubulin表达水平呈负相关(r = -0.372,P = 0.006),与MAPT表达水平呈负相关(r = -0.292,P = 0.032),与survivin表达水平呈负相关(r = -0.340,P = 0.012);Class Ⅲ β-tubulin与MAPT mRNA表达水平呈正相关(r = 0.284,P = 0.037),与survivin mRNA表达水平呈正相关(r = 0.559,P = 0.000)?结论:胃癌细胞中Class Ⅲ β-tubulin?MAPT和survivin的表达水平可能对紫杉类药物的化疗敏感性具有较好的预测价值?
英文摘要:
      Objective:To investigate the potential role of Class Ⅲ β-tubulin,Microtubule-associated protein tau(MAPT) and inhibitor-of-apoptosis protein survivin serving as biomarkers for paclitaxel sensitivity in primary gastric cancer. Methods: Viable tumor cells obtained from the samples of fifty-four patients with gastric carcinoma were tested for their sensitivity to paclitaxel using ATP-based tumor chemosensitivity assay(ATP-TCA). Reverse transcription-PCR was used to analysis the mRNA level of related genes expression. Results:Class Ⅲ β-tubulin mRNA level was negatively correlated with the sensitivity to paclitaxel (r = -0.372,P = 0.006), but related to histological differentiation(P = 0.029). MAPT mRNA level is negatively correlated with the sensitivity to paclitaxel(r = -0.292,P = 0.032). Survivin mRNA level was also negatively correlated with the sensitivity to paclitaxel(r = -0.340,P = 0.012),but related to histological differentiation(P = 0.009). Significant relationship was found between Class IIIβ-tubulin and MAPT or survivin expression, respectively(P = 0.037 or P = 0.000). Conclusion: Our results demonstrate that the mRNA expression levels of Class Ⅲ β-tubulin, MAPT and survivin in gastric cancer might be of good value in predicting the sensitivity to paclitaxel.
