    目的:探讨碱性成纤维细胞生长因子(bFGF)对体外培养的听觉感觉上皮细胞增殖和分化的影响-方法:取新生大鼠听觉上皮细胞进行体外培养,用BrdU和calretinin染色,以观察bFGF对细胞增殖和分化的影响-结果:①100-200 ng/ml bFGF组中的BrdU阳性着染的细胞分别占所观察细胞的36%和35%,与对照组相比有显著差异;而1-10 ng/ml bFGF组中的有丝分裂活动很微弱,分别为3%和7%-②100-200 ng/ml bFGF二组中calretinin阳性着染的细胞数量分别占所观察的细胞总数的7%和7.6%,1-10 ng/ml bFGF组中,calretinin阳性着染的细胞数量分别为0.5%和1.6%-结论:耳蜗上皮细胞的增殖活动和向毛细胞方向的分化受生长因子的调控,bFGF有较强的促增殖和促早期分化的作用-

    Abstract:

    Objective: To evaluate the effects of bFGF on proliferation and differentiation of rat cochlear sensory epithelial cells. Methods: Cochlear sensory epithelial cells of neonatal rats were isolated and cultured with different bFGF concentration. Bromodeoxyuridine (BrdU) labeling and immunohistochemical staining with calretinin were used to identify mitotic status and properties of culture cells. Results:① In 100 ng/ml and 200 ng/ml bFGF group,the BrdU positive cells were 36% and 35%,respectively,while in 1 ng/ml and 10 ng/ml bFGF group,the BrdU positive cells were only 3% and 7%. ② Percentage of calretinin positive cells varied from 0.5%,1.6%,7%,and 7.6% in 1 ng/ml,10 ng/ml,100 ng/ml,and 200 ng/ml bFGF,respectively. The percentage of the BrdU-positive and calretinin-positive cells in high bFGF concentration groups (100 ng/ml and 200 ng/ml) were significantly different compared with the control group. Conclusion: Proliferation and differentiation of cochlear auditory epithelial cells can be regulated by bFGF.
