Effects of basic fibroblast growth factor(bFGF) on expression of β1 integrin subunit and adhesion of osteoblast

    Abstract:

    Objective:To investigate the effects of basic fibroblast growth factor(bFGF) on the expression of β1 subunit of integrin and to explore the effects of bFGF and fibronection(FN) on osteoblast adhesion on bio-derived bone. Methods:The third generation of osteoblast was treated with bFGF(100 ng/ml) and then seeded on bio-derived bone which had been modified with FN(10 μg/ml). Expression of β1 subunit of integrin was detected by Western blot. Cell adhesion was measured by MTT assay. Cell density and appearance was observed by scanning electron microscope. Results:The expression of β1 subunit of integrin was significantly enhanced by bFGF. Both FN and bFGF could enhance osteoblast’s adhesion efficiency on bio-derived bone(P < 0.01). But the osteoblast’s adhesion efficiency was significantly strengthened by combined use of FN and bFGF. They had a significant synergistic effect in statistics(P < 0.01). Combined use of FN and bFGF had a better effect on cell density and appearance than solo use when observed with scanning electron microscope. Conclusion:BFGF can significantly enhance the expression of β1 subunit of integrin and significantly strengthen osteoblast’s adhesion efficiency on bio-derived bone which has been modified with FN. BFGF and FN have a significant synergistic effect on osteoblast’s adhesion.
