    Abstract:

    Objective:To investigate the effect of prostaglandin E2 on the proliferation of human cholangicarcinoma cell and the possible mechanism. Methods:Human cholangicarcinoma cell line(HUCCT1) was cultured and treated with PGE2, and the cell growth rate and viability was measured by cell proliferation assay with WST-1 regent. The intracellular free calcium concentration was measured using laser scanning confocal microscopy with fluo-3/AM as fluorescence probe. The level of cAMP was measured by immunoassay KIT. Results:PGE2 promoted the growth of cholangicarcinoma cell, and increased intracellular free calcium and cAMP in cholangicarcinoma cell. Conclusion:The results that PGE2 could increase the concentration of intracellular free calcium and cAMP suggest that PGE2 may play a role in promoting the cell growth.
