Genotyping study of 168 strains of Mycobacterium tuberculosis isolated from Jiangsu province in VNTR

    Abstract:

    Objective:To investigate the distribution of the genotyping and the characteristics of VNTRs in clinical Mycobaterium tuberculosis(MTB) strains isolated from Jiangsu province with multi-loci of variable number tandem repeats. Methods:168 strains of Mycobacterium tuberculosis were selected from three parts of Jiangsu province. Eleven loci of VNTR from the genome of Mycobacterium tuberculosis were selectd and analyzed for the polymorphism in distribution by means of PCR,agarose electrophoresis and the BioNumerics(Version 3.0)software. Results:It was found that distribution of VNTR from 168 strains of Mycobacterium tuberculosis isolated showed definite polymorphism,and 10 different genotypes could be classified in which most of strains belonged to one genotype(75.1%),and the others were scattered,such as Ⅱ(5.4%),Ⅳ(4.2%),Ⅶ(9.5%). We also found that there was visiblely different among three parts of Jiangsu province. Conclusion:It is concluded that the VNTR loci in the genome of Mycobacterium tuberculosis isolated in Jiangsu showed marked polyporphism,most of the 168 strains belonged to the one genotype(Ⅷ). The distribution of main prevalent strains among three parts were the same(Ⅷ), but other genotypes distribution of strains were different.
