Abstract:Objective:To investigate Paeoniflorin’s inhibition effect on NF-κB in human gastric carcinoma cells and its role in the induction of cell apoptosis. Methods:Paeoniflorin was diluted to different concentrations and treated on the gastric carcinoma cells at different time. Western Blot was used to test the expression of NF-κB in nuclear and cytoplasm. The intranuclear expression of NF-κB was confirmed by ELISA assay. The influence on cell apoptosis of gastric carcinoma cell by Paeoniflorin and 5-FU was estimated by flow cytometry. Results:Paeoniflorin dramaticly inhibitied NF-κB activity in nuclear and cytoplasm. The inhibition pattern of NF-κB exhibited in a time and dose dependence, which was confirmed by Western Blot and ELISA. Moreover,Paeoniflorin had a promotion on cell apoptosis induced by 5-FU. Conclusion:Paeoniflorin could inhibit NF-κB activity in nuclear and cytoplasm of SGC-7901 cells, and enhance cellular apoptosis of gastric carcinoma.
