Objective:To observe the effect of peroxisome proliferator-activated receptor γ agonist(pioglitazone) on leptin-induced proliferation of vascular smooth muscle cells(VSMCs). Methods:By using primary cell culture and MTT colorimetry,the effects of leptin and peroxisome proliferator-activated receptor γ agonist were detected at various concentrations on VSMCs proliferation. Results:Leptin can significantly promote VSMCs proliferation in a dose-and time-dependent manner,whose maximum effect appears at 100 ng/ml and 72 hours(P < 0.05). In additiom,pioglitazone suppressed leptin-induced VSMCs proliferation,especially at 100 μmol/L.(P < 0.05). Conclusion:Leptin can induce the proliferation of VSMCs,and pioglitazone can inhibit this effect. Pioglitazone might be used to treat atherosclerosis, and the mechanism should be involved in that pioglitazone could relieve the leptin resistance.
