    Abstract:

    Objective:To study the micromorphology and genetics of the muscular tissue from the female cadaver of Xi Han Dynasty discovered in Lianyungang in 2002. Methods:Light microscope and electron microscope were used to observe micro morph of the muscular tissue from the female cadaver,which had been fixed in 10% formalin after being unearthed. Besides,DNA Extract kits were used to extract genome DNA from muscular tissue;PCR was used to amplify mitochondrial DNA sequence. Results:Observed by both light microscope and electron microscope,the morph of the muscular tissue from the ancient cadaver sample was in a great mess. Genomic DNA(including mitochondrial DNA) of the cadaver was not obtained by extraction and amplification,respectively. Conclusion:The ancient cadaver fixed by formalin might be disadvantageous of the electron microscope and molecular biology research.
