The expression and clinical significance of OPN and MMP-2 in human pancreatic carcinoma

    Abstract:

    Objective:To investigate the expression and clinical significance of OPN and MMP-2 in pancreatic carcinoma and explore the role of OPN and MMP-2 in the invasion and metastasis of pancreatic carcinoma. Methods:Tumor tissue chip with 178 pancreatic tumor and 36 paraneoplastic tissues were constructed. The location of OPN protein in pancreatic tumor cell lines was detected by immunofluorescence. The expressions of OPN and MMP-2 were investigated by immunohistochemistry assay. Results:The OPN was mainly located on the cell membrane and cytoplasmic in two pancreatic tumor cell lines(PANC-1,SW1990). The expression of OPN and MMP-2 was higher in pancreatic tumor,74.2%(132/178)and 71.3%(127/178)respectively,than in paraneoplastic tissues(P < 0.05). OPN expression was significantly correlated with MMP-2(r = 0.34,P < 0.05). The expression of the OPN protein was related to histological differentiation of pancreatic tumor cells. OPN and MMP-2 expression were higher in cases with lymph node metastases or with nerve infiltration(P < 0.05). Conclusion:OPN and MMP-2 might strengthen the invasion and metastasis of pancreatic carcinoma. OPN and MMP-2 might play synergetic roles in the progression of pancreatic carcinoma.
