Osteoblasts derived from human marrow mesenchymal stem cells on co-cultured tissue engineering bone after transfected by EGFP genes

    Abstract:

    Objective:To explore the transfecting EGFP genes effect into cells on adherence ability of cells to biomaterials,which influences material biocompatibility evalution. Methods:EGFP gene was successflly delivered into osteoblasts induced from human marrow mesenchymal stem cells(MSCs) through electroporation transfection,and the osteblastes were seeded into tissue engineering bone with biomaterials. All cultured cells were detected by inverted microscope,scanning electron microscope,and fluorescent microscope. Results:It was observed that the osteoblasts transfected with EGFP gene had been successfully adhered to tissue engineering bone,and compared with osteoblasts without EGFP gene transfection,there was no obvious difference. Conclusion:It is suggested that transfection of GFP marker does not harm the biocompatibility to biomaterials and proliferative activity of osteoblasts induced from MSCs,which is helpful to understanding biocompatibility of materials accurately.
