Objective:To investigate the chemical synthesis of short interfering RNA (siRNA) and construction of interfering RNA (RNAi) vector of survivin,and their influence on the expression of survivin in HEK293 cell line. Methods:Chemically synthesized siRNA and shRNA vectors were used to silence survivin expression in HEK cell line,and the survivin mRNA and protein expression levels were determined by RT-PCR and Western blot respectively. Results:After transfection of different siRNA and shRNA vectors of survivin gene into the HEK293 cells,the survivin mRNA and protein levels were significantly decreased (P < 0.05). Conclusion:The study proposed that the survivin expression in HEK293 cell line could be silenced by chemically synthesized siRNA or shRNA vector of survivin gene.
