    Abstract:

    Objective:To investigate the clinical significance and relationship of tumor-suppressing gene PTEN and VEGF,FLT1(VEGFR1) mRNA in myleod leukemia cells. Methods:The mRNA expression of PTEN and VEGF in marrow mononuclear cells (MMSCs) from 50 acute myeloid leukemia (AML) patients,10 AML patients in complete remission (CR),10 chronic myelogenous leukemia (CML) patients in chronic phase (CML-CP),10 CML patients in blast crises (CML-BC) and 10 persons as normal controls were detected by real-time fluorescent relative-quantification PCR(FQ-PCR). The correlation and clinical significance of PTEN,VEGF and FLT1 were evaluated. Results:The expression level of PTEN mRNA was lower in AML patients than normal control;and it was lower in CML-BC patients than that in CML-CP patients. PTEN mRNA expression level was negatively correlated with VEGF and FLT1 mRNA,and the low-expression of PTEN gene and high-expression of VEGF and FLT1 gene were associated with the poor prognosis and the higher extra-medullary infiltrations. Conclusion:The low-expression of PTEN mRNA and high-expression of VEGF or FLT1 mRNA in myleod leukemia meand the poor prognosis and the higher extra-medullary infiltrations. PTEN,VEGF and FLT1 mRNA may have synergistic effect on the pathogenesis of myleod leukemia.
