    Abstract:

    Objective:To examine the changes of IL-6 level in the condition of cultured THP-1 with or without human adipocytes,and to explore the relationship between inflammation and obesity. Methods:Co-culture system for monocytic cell line THP-1 and human adipocytes was established through transwell combined with treatment of JNK inhibitor-SP600125. Meanwhile,cultured THP-1 and human adipocytes respectively acted as controls. The accumulation of IL-6 in cultured media supernatant was assayed by enzyme-linked immunosorbant assays (ELISA) for every group. Results:The results of ELLSA showed that the IL-6 was markedly increased in the co-culture of THP-1 and human adipocytes,and the level of IL-6 was decreased when treated with JNK inhibitor. Conclusion:There might be a paracrine loop between the monocytic cell line THP-1 and adipocytes. The level of IL-6 is increased under co-culture condition,while JNK inhibitor could decrease the IL-6 release,which might become a new therapy target for obesity.
