Abstract:Objective:To explore the relationship of the gastric cancer susceptibility and polymorphism of upstream of IL-10 promoter at the site of 1082,819 and 592. Methods:Total of 129 patients with gastric cancer and 98 healthy individuals were enrolled in the study,and divided into patient group and control group. The polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) was applied to detect the base transition in the upstream of IL-10 promoter at the sites of 1082,819 and 592. The H.pylori infection of patients and health individuals was detected by immunoblotting. The SPSS10.0 software was applied to analyze frequencies of all genotypes. Results:There were obviously differences in polymorphism of IL-10-1082 between the gastric cancer group(AA:57.36%,GA:40.31% and GG:2.32%)and control group(AA:75.51%,GA:24.49%). The rate of GA genotype of IL-10-1082 in gastric cancer group was higher than that in control group(P=0.012). Allele G of IL-10-1082 in gastric cancer group was also higher than that in control group(P=0.007). We failed to observe any difference in the genotypes of IL-10-819 and -592 between gastric cancer group and control group. The GA genotype of IL-10-1082 in gastric cancer group was higher than that in the individuals with H.pylori infection in control group(OR=2.65,95% CI:1.16-6.08;P=0.031). Conclusion:The GA genotype or G allele in IL-10-1082 is a risk factor of the occurrence of gastric cancer. There was cooperation of H.pylori infection and the GA genotype in IL-10-1082 on the occurrence of GC.
