Abstract:Objective: To investigate the role of mTOR specific inhibitor RAD001 in inhibiting gastric cancer cell SGC7901 proliferation and the possible mechanism of survivin in the inhibition. Methods:SGC7901 cells were cultivated, treated with RAD001 alone or in combination with cisplatin. MTT, flow cytometry, Immunocytochemistry and Western-blot array were used to investigate the cell proliferation, apoptosis and the expression of survivin in cells. Results:After treated with RAD001 alone or in combination with cisplatin, the cell proliferation were inhibited in a dose-dependment manner, and the inhibition rates increased with the treatment time extending. High concentration of RAD001 could induce apoptosis of SGC7901 cells. The expression of survivin in SGC7901 cells decreased after treatment. Conclusion:RAD001 could inhibit the cell proliferation of SGC7901 and induce the apoptosis through down-regulating the expression of survivin.
