CEACAM6 enhances the ability of invasion and chemo-resistance in pancreatic cancer cells by inducing epithelial-mesenchymal transition(EMT)

    Abstract:

    Objective:To investigate the potential involvement of CEACAM6 in the invasion and chemo-resistance of pancreatic cancer cell SW1990 by inducing epithelial-mesenchymal transition(EMT). Methods:After establishing and screening,CEACAM6-overexpressing cell line SW1990-CEACAM6 was observed by inverted microscope,and the changes of EMT markers were determined by Q-PCR and western blot,the invasion ability was detected by transwell,the chemo-sensitivity to gemcitabine was determined by MTT assay. Results:Q-PCR and Western blot confirmed that SW1990-CEACAM6 cell line was established successfully. Overexpression of CEACAM6 made the pancreatic cell line SW1990 prone to mesenchymal phenotype,such as up-regulation of vimentin and down-regulation of E-cadherin. SW1990-CEACAM6 cell has greater activity of invasion and chemo-resistance to gemcitabine (P < 0.05) than SW1990-EGFP cell. Conclusion:CEACAM6 could promote EMT and enhance malignant potential in terms of invasion and chemo-resistance in pancreatic cancer cell.
