Objective:To investigate if glycine has the protection effect on warfarin induced aortic valve interstitial cells(AVICs)calcification. Methods:Porcine AVICs were isolated,calcium quantification,alkaline phosphatase activation and Alizarin red staining were performed to determine the effect of different concentrations of glycine on calcification,and the mRNA expression of osteogenic markers were determined by RT-PCR. Western blot,Flow cytometry and TUNEL assay were performed to investigate the anti-apoptosis effect on warfarin induced AVICs calcification. Results:Glycine attenuated warfarin induced AVICs calcification,reduced the mRNA expression of osteogenic markers,inhibited the Smad1,5/RUNX2 pathway and decreased the apoptosis of warfarin induced calcification. Conclusion:Glycine inhibits Smad1、5/RUNX2 pathway and decreases the early stage of apoptosis to attenuate warfarin induced AVICs calcification.
