    • Modified Animal Model of Rapid Glomerulosclerosis

      Abstract:To prepare an animal model which is stable on the pathological changes. The preparation of the animal model is easy and rapid, and is close to the human's glomerulosclerosis in pathology.Methods After excise a kidney, daunomycin was injected into rat's tail vein twice. The changes of their serum, urine and pathological changes was evaluated. Results The rats had edema, heavy proteinuria, hypoproteinemia, hypercholesterolemia and typical glomerular segmental sclerosis in pathology in the fourth week. in the ninth week, the BUN and creatinine lev el significantly increased (P<0.05) ,and the rate of ginmerulosclerosis raised to 80%Conclusion That daunomycin animal model can be used as an experimental mode[ of glomerulosclerosis, and the rapid preparation is one of its best advantages.

    • Determination of Urinary Heparin in Bladder Transitional Cell Carcinoma and Its Clinical Significance

      Abstract:To establish a capillary electrophoresis (CE) method to determine the value of urinary heparin in bladder transitional cell carcinoma (BTCC). Methods Reverse polarity CE with a UV-detector 190 nm in a 20kV and pH2. 5 buffer system was used to determine urinary heparin in 21 patients with BTCC and 12 normal individuals. The analysis of variance (ANOVA) was used in this experiment and significance level is 0. 0.5. Results The correlation with heparin peak area and its value is linear. The value of urinary haparin is (0.94± 0. 48) μ/L in 12 normal, (1. 30±0.73) μg/L in 11 patients with Ta~T1 BTCC and (0. 54±0. 45) ug/L in 10 patients with T2~T1 BTCC. Urinary heparin was lower in T2~T1 BTCC than in normal and Ta~T1 BTCC (P<0. 05). Conclusion Reverse polarity CE is a high-resolution, rapid and simple method to separate heparin from urine. Falling of urinary heparin in BTCC plays an important role in the infiltration and transition of the carcinoma cells.

      Abstract:Establishing the research model of arterial endothelial cells in vitro. Methods By means of digesting and scraping, fetal bovine arterial endothelial cells were isolated and cultured in M199 (pH7. 0). Results Vascular endothelial cells can be identified by the "cobblestone" on morphology exhibited by confluent monolayers and positive staining for Ⅷ R: Ag. In the absence of vascular endothelial cell growth factor, Ecs were subcultured to the 18th generation at most. Conclusion Now that human arteries are not readily available, the culture of fetal bovine arterial endothelial cells is a good replacement.

    • The Expression of Human Apolipoprotein Al in Myogenic Cells by Retrovirus Vector

      Abstract:To observe the expression of apolipoprotein A1 in transduced cultured muscle cells. Methods Bicistronic retrovirus vector containing human apolipoprotein A1 cDNA were constructed and packaged it into GP+E-86 and AM12 cell lines which was selected by G418. G418 resistant clone with high virus producing tiler was isolated. Mouse primary myoblasts and C2C12 mouse myoblasts were infected with recombinant virus supernatant. The content of apolipoprotein Al in cell culture medium was detected by ELISA. Results All of transduced mouse pti mary myoblasts and C2C12 cells gained and remained the ability for heterologous expression of human apolipoprotein Al even after differentiation into myotubes. Moreover, stable transduced C2C12 cell clones were generated by G418 selection, which effectively expressed human apolipoprotein Al up to 30 days. Conclusion The use of retrovirus vec tots to genetically modify myoblasts and then implantation back to skeletal muscle might be a safe and feasible strategy for gene therapy of atherosclerosis.

      Abstract:Effects of Bnk Ⅶ and Bmk ⅩⅢ from venom of the scoripion Buthus martensi karsch on spontaneous active potential of the central nerve of insects were investigated. Methods Using extracellular recording, we observed the effect of BmK Ⅶ and BmK ⅩⅢ on spontaneous active potential of the exposed central nerve cord of Periplaneta americana.Results We found that BmK Ⅶ (1 ug/mi) first exerted its reversible inhibition and then fol loved the excitation. BmK ⅩⅢ ( 10 ug/ml has only inhibiting effect, which is irreversible. Conclusion These results indicated that BmK Ⅶ is a mixture including inhibiting and exciting components, while BmK ⅩⅢ has only inhibiting component. The effect of these two toxins possibly through the modulation to sodium channels of the neurons.

    • Study of Anovulatory Menstrual Cycle in Healthy Woman by the Method of Urine Progesterone Enzyme Immunoassay

      Abstract:In this experiment, we investigated the frequency of anovulation in a sample of 38 healthy women by collection two to three consecutive menstrual cycles. Methods Daily morning first urine samples were collected for analysis of urine progesterone (pregnanediol 3 glucttronide), FSH and creatinine by enzyme immunoassay. Results Frequency of anovulatory women was 7. 90%, and anovulatory menstrual cycles frequency was 7. 69%. During early follicular phase, FSH excretion was significantly less in the anovulatory cycles than ovulatory cycles, and progesterone always remained at very low level for the whole cycle. Conclusion Urine progesterone of the enzyme immunoassay can be used to evaluate ovary function and to analyze anovulatory menstrual cycle.

      Abstract:vTo investigate the expression of laminin(LN) on normal endometrium, hyperplastic en dometrJum and endometrlal adenocarcinoma and; to study the relationship between LN's expression and endometrial adenocarcinoma. Methods LSAB immunohistoehemical method was used to study LNts expression on 16 normal, 12 hyperplastic and 27 adenocarcinoma endometrium cases. Results Laminin is widely expressd on epithelia basement membrane(EBM), epithelial/cancer cell(EC) and stromal cell (SC). Among normal, hyperplastie, adenocarcinoma endometrium, LN's expression on EBM (X2=13.06,P<0. 001), EC (X2= 17.82,P<0. 001), SC (X2=16.96,P <0. 01) and EC/SC (X2=27.49,P<0. 001) are significantly different. The LN's expression was compared with clinical and pathologic parameter of endometrial adenoeareinoma, and the clinical stages of endometrial adenocarcinoma compared with LNts expression on EBM (P<0. 05) and on SC (P<0. 01) ,and the pathologic grades of endame trial adenocarcinoma with LN's expression on SC (P<0.05), all those being different in statistics. Conclusion The LN's expression on epithelial/cancer cells and disapearane on epithelial basement membrane, stromal cells are related to endometrial carcinoma; It may be an important sign on the early stage of endometrlal adenocarcinoma.

      Abstract:To investigate the neuroprotective effect and mechanism of TMB 8[8- (N, N-diethy lamino)- n -octyl- 3,4,5- trimethoxybenzoate]. Methods The oxygen-glucose deprivation model of cultured rat neu ron was used to observe the effects of TMB-8 on neuron morphology, permeability and contents of MDA, GSH, TSOD and GSH px in neurons. Results The damaged neurons induced by oxygen glucose deprivation for 1 h and after serum-free culture for 24 h could liberate much LDH; the contents of GSH and the activity of T SOD in neurons were significantly decreased; TMB-8 could concentration dependently reduce the efflux of LDH from the damaged neuron and increase the content of GSH in neuron, the activity of T-SOD in these cells was also increased. Conclusion the neuroprotective role of TMB-8 may be related to the increasing of GSH and T SOD in the damaged neuron.

    • The Application of the Short Test of Mental Status in Functional Assessment of the Brain Injuried People

      Abstract:The purpose was to testify whether STMS assessment was adapted to the cognitive function assessment for the patients with brain injury. Methods The STMS assessment was used in the comparison of the brain injued patient in different stages and different injued cerebral hemisphere, and comparison between the patient and the normal person was then made. Results ①The score of the patient was increasing dramatically with the elapse of time(P<0. 05); ②Comparing the patients in the recovery period with the normal person, the whole score was not significantly different, but still had some difference in some of the subjeets(P<0. 05); ③There was significant difference in the score of STMS in the ability of comprehension and orientation, between the left and the right injuried cerebral hemisphere(P<0.05). Conclusion STMS is a useful assessment method. It can be used in the cognitive function in the brain injuried patient.

      Abstract:To evaluate the relationship between the expression of cathepsin D(CD)and biological be haviour of bladder transitional cell carcinoma (BTCC)and the mechanisms of invasiveness. Methods The expression of CD and BFGF was detected by standard ABC immunohistochemical technique in 33 paraffin embedded tissue speci mens of BTCC. Results All normal transitional ceils were weakly stained. The over expression rate of CD in BTCC group was significantly higher than in normal control group (P< 0. 05).     • Expression and Significance of Ki-67 in Transitional Cell Carcinoma of Bladder

      Abstract:Ki-67 is exclusively present in all stages of cell cycle (C1, S, C2 and mitosis)except for G0, and widely used to determine proliferative activity of various tumors. The relativity of expression of Ki-67 and the biological behaviour of transitional cell carcinoma (TCC) of bladder was evaluated in the study. Methods The paraffin embedded tissue sections from 33 patients with TCC of bladder were reexamined. According to the UICC standard, 21 cases had surperficial tumors and 12 cases had invasive tumors. With the pathological diagnostic standard of WHO, there were 16 cases of G1 , 10 cases of G2 and 7 cases of G3. Meanwhile, there were 21 cases with primary lesions and 12 cases with recurrent lesions. 12 specimens obtained from normal bladders were used as controls. Expres sion of gA 67 was detected by streptavidin-peroxidase (SP) immunohistochemical technique. Results Expression of gA-67 was found in 54. 5%(18/33) of TCCs and 8. 33%(1/12) of controls (P

      Abstract:To study the expression of P185 in breast infiltrating ductal carcinoma and its value. Methods 52 cases of breast infiltrating ducT,al carcinoma were examined for P[85 by the method of S-P immunohistocbemistry. Results 30 cases of breast infiltrating ductal carcinom (57. 7%) were positive in P185 expression. P185 over-expression was not related to histological grade,turnout size and lymph node metastasis (P> 0. 05), but in significantly negative relation to ER expression (P<0.01). Conclusion The examination of P185 and ER in couples is tee ommended and it is related to the selection of effective anticarcinogens.

    • The Experimental Study of Osteolysis Induced by Polyethylene Particles

      Abstract:To study the etiology and the mechanisms of osteolysis induced by polyethylene particles. Methods A rabbit model of resorption of bone at the bone-prosthesis interface induced by polyethylene particles was established. The pathological findings were studied. In vitro human rnonocytes were cultured and stimulated by polyethylene particles. The levels of TNF-a of the supernats were measured by radioimmunoassay. Results There was a layer of interface membrane that contained numbers of maerophages and giant cells around the prosthesis, and there was evidence of active bone resorption and osteolysis. The polyethylene particles were found in the interfacemembrane and engulfed by macrophages. The levels of TNF-a were significantly increased when the monocytes were ex posed to polyethylene particles (P<0.01). Conclusion There is a positive relationship between the accumulation of polyethylene particles and subsequent resorption of bone. The polyethylene particles can stimulate monocytes to produce more TNF-a.

    • Clinical Analysis on 26 SLE Patients with Neuro-Psychological Manifestations

      Abstract:To explore clinical features, treatment and prognosis of neuro psychological lupus erythe matosus (NPLE). Methods Clinical data of 25 SLE patients with neuro-psychoingical manifestations who were hospi talized in Department of Rheumatoingy of our hospital from year 1992 to 1998 were retrospectively summarized. Results In eilninal features, among 26 NPLE patients 13 cases occurred within one year after SLE onset, 11 cases pre seined organic eneephalopathy syndrome, 2 cases encephalin stroke, d cases epilepsy', 2 cases lupoid encephalodynia, 7 cases psychological manifestations or psychosis. In treatment, 12 of 19 servere NPLE patients were intravenously given methoprednisoinne plus immuno suppressive agent pulse dose, in which 10 cases remitted, 2 cases died; the other 7 cases were intravenously treated with methoprednisolone pulse dose alone, in which 4 cases remitted, 3 cases died, compared with the above, P>0. 05, whereas, 6 of 7 cases with psychological manifestations were administrated with oral prednisone, by dose of less than J 00 rog/d, plus anti-psychosis drugs, and then remitted within 2 weeks over 2 months. Conclusion Glucose cortico-steroids plus immuno-suppressive agent pulse dose treatment is efficient for severe NPLE patients and conventional dose glucose corticosteroids plus anti-psychosis drugs treatment is effective for NPLE patients with psychological manifestations.

    • The Blood Conservation Research on Aprotinin Dosages and Effects in Cardiac Surgery

      Abstract:Different aprotinin regimens and their hemostatic effects were comparatively analysed and the more appropriate aprotinin regimen was evaluated and discussed here. Methods Eighty-two open heart patients were divided into three groups. There were high-dose half-dose and low-dose aprotinin groups. Results It was shown that the high-dose aprotinin regimen could reduce postoperative drainage by 54%and 55%during the first 12 and 24 hours,meanwhile the hemoglobin loss and blood transfusion requirement were reduced by 56%and 65%respectively compared to the control group. The half-dose aprotinin regimen could reduce postoperative drainage by 49%and 55%in the first 12 and 24 hours ,which was not significantly different as compared with high-dose aprotinin group. The low-dose aprotinin regimen could reduce tube drainage and blood transfusion requirement by 30%and 26%, which had significant difference as compared to half dose aprotinin group. Conclusion The use of aprotinin could sig nificantly reduce postoperative drainage and blood transfusion requirement,the hemostatic effect was related to apro tinin dose. The half-dose and low-dose aprotinin regimen are recommended.

    • Vitrectomy for Removal of Intraocular Foreign Bodies

      Abstract:To discuss the surgical techniques and effectivity of vitrectomy for intraocular foreign bodies(IOFBs)removal. Methods 32 patients were treated with pars plana vitrectomy and removal of the IOFBs,com bined with lensectomy. IOL implantation,encircling, silicone oil and C3F8 injection. Preoperative visual acuity (VA) with postoperalive VA were compared. Results The successful rate of the extraction of 1OFBs was 100700. Followed 6 months,post ope VA improved obviously. The number of cases with VA(0. 05~0.4)increased from 3 to 13,and that with VA(0. 5 and better)from 2 to 5. Conclusion Vitrectomy for removal of IOFBs is accute and effectient. There fore,the injures are the smallest and post-ope VA can improve as high as possible,
