Abstract:目的:探讨神经生长因子(NGF)对损伤脊髓保护作用的分子机制.方法:采用Allens法以10 g×2.5 cm力致伤大鼠T8脊髓,插蛛网膜下隙导管于术后即刻、2 h,4 h,8 h,12h,24 h,各注入NGF溶液,并与生理盐水组和正常对照组作对照.采用原位杂交法检测N-甲基-D-天门冬氨酸受体1(NMDAR1) mRNA在脊髓中的表达.结果:应用原位杂交法检测,正常组大鼠脊髓中几乎无NMDAR1 mRNA表达,生理盐水组NMDR1 mRNA表达明显增多,NGF组与生理盐水组相比较,NMDAR1 mRNA表达明显减少(P<0.01).结论:NGF能通过抑制脊髓中N-甲基-D-天门冬氨酸受体(NMDAR)的生成,抑制钙内流,抑制NO的生成,从而保护损伤的神经组织.

Abstract:目的:克隆爱泼斯坦-巴尔病毒(EBV)潜伏期膜蛋白2A(LMP2A)基因,进一步研究其基因工程疫苗.方法:用逆转录-聚合酶链反应(RT-PCR)扩增出EBV B95.8株LMP2A全部编码蛋白的基因,并克隆至载体pGEM-T中,通过α-插入失活、PCR及EcoR Ⅰ、Sma Ⅰ限制性内切酶双酶切等鉴定重组质粒;采用Sanger双脱氧终止法测定cDNA片段的核苷酸序列.结果:克隆出LMP2A第1个外显子到第8个外显子的全部编码蛋白的cDNA,测序结果与EB病毒B95.8原型株LMP2A cDNA作同源比较,完全一致.结论:本实验克隆了LMP2A基因的全部编码蛋白的cDNA全长片段.

Abstract:目的:研究显示人皮肤微血管构筑的有效方法.方法:采用酶组织化学的Gomori-Takamatsu钙钴法和(NBT/BCIP)硝基四唑氮蓝/5-溴-4-氯-3-吲哚磷酸盐(NBT/BCIP)显色法,以及CD34和Ⅷ因子相关抗原的免疫组化法,显示人皮肤的微血管构筑.结果:与传统方法比较,该 4 种方法均能良好显示人皮肤的真皮内和真皮下的微血管构筑.免疫组化法的特异性好,但酶组织化学法通过厚切片技术,能更好显示微血管的网状结构.CD34的敏感性高于Ⅷ因子相关抗原.结论:这 4 种方法的初步确立,为皮肤和皮瓣的血运研究,甚至为瘢痕形成和创伤愈合等相关领域的研究,提供了可行的方法学依据.

Abstract:目的:观察去势后大鼠海马结构一氧化氮合酶(NOS)阳性神经元的变化.方法:用黄递酶组织化学染色方法观察切除双侧卵巢后雌性SD大鼠海马结构NOS阳性神经元的形态、分布的变化,并进行计算机图像分析.结果:去势后海马结构NOS阳性神经元分布变化有区域差异性:NOS阳性神经元在下托、海马(CA)一区邻近下托的部分、CA三区、CA四区(CA4)和齿状回(DG)数目明显减少,而在CA二区数目明显增多;CA4和DG的NOS阳性神经元平均密度降低,胞体的平均周长和平均截面积都明显减少.结论:雌激素可能通过影响海马结构NOS的表达来影响学习和记忆.

Abstract:目的:观察白细胞介素-10(IL-10)对白细胞介素-1β(IL-1β)诱导的人系膜细胞(HMC)胞质型磷脂酶A2(cPLA2)基因和蛋白表达及其前列腺素E2(PGE2)释放的影响.方法:利用体外培养的HMC,设立对照组、IL-10处理组、IL-1β处理组及IL-10+IL-1β处理组;采用放免法检测培养上清中PGE2,应用逆转录-多聚酶链反应(RT-PCR)和蛋白质印迹(Western Blot)检测cPLA\-2 mRNA和蛋白水平.结果:正常情况下,HMC低水平表达cPLA\-2 mRNA和蛋白并释放少量的PGE2;IL-1β能显著上调HMC PGE2释放及cPLA\-2 mRNA和蛋白的表达(P值均<0.01);IL-10对基础状态下的HMC PGE2释放及cPLA\-2 mRNA和蛋白表达无明显影响(P值均>0.05),但可呈剂量依赖性地下调IL-1β诱导的PGE2释放及cPLA\-2 mRNA和蛋白表达(P值均<0.01).结论:IL-10抑制IL-1β诱导的HMC PGE2释放及cPLA\-2表达,提示IL-10对HMC具有多方面抗炎作用.

Abstract:目的:研究脊髓损伤(SCI)后脊髓组织星形胶质细胞中胶质纤维酸性蛋白(GFAP)表达的变化,探讨反应性胶质化在脊髓损伤中的作用.方法:SD大鼠25只,随机分成5组(n=5):正常对照组、伤后1天组、伤后4天组、伤后7天组和伤后14天组,用免疫组织化学染色及图像分析的方法观察星形胶质细胞中GFAP的表达.用大鼠综合行为评分法(CBS)对各级大鼠神经功能评分.结果:SCI后星形胶质细胞中GFAP的表达与对照组比较明显增高(P<0.01),1~14天内呈进行性增高趋势;损伤各组的CBS 1~14天内呈降低趋势.两指标有显著的相关性(r=-0.775,P<0.001).结论:SCI后星形胶质细胞通过其反应性胶质化对脊髓的再生和自身修复起着重要作用.

Abstract:目的:观察缺血再灌注后大鼠翼腭神经节一氧化氮合酶(NOS)阳性神经元数目的变化.方法:结扎大鼠双侧颈总动脉不同时间(15、30、60、120 min)后,应用还原型尼克酰胺腺嘌呤二核苷酸磷酸-黄递酶(NADPH-d)组织化学染色法观察并计数翼腭神经节内NOS阳性神经元.结果:缺血30 min组,翼腭神经节内NOS阳性神经元数目明显增加,60 min组与30 min组相比未见明显差异,120 min组则比其它组都显著增加.结论:在缺血再灌注过程中,翼腭神经节内NOS阳性神经元数目增加,可能影响到脑血管周围NOS阳性纤维的NO释放量.

Abstract:目的:探讨白芍总甙对离体豚鼠结肠平滑肌的作用方式及结肠平滑肌M受体在调节结肠运动中的作用.方法:制备豚鼠结肠纵肌和横肌肌条,观察不同剂量白芍总甙及工具药对离体结肠肌收缩活性的效应.结果:白芍总甙使豚鼠结肠离体平滑肌收缩积分和时间显著性增加.且收缩效应的改变具有剂量依赖性.白芍总甙的这种作用能被阿托品抑制.结论:白芍总甙可通过延长结肠收缩时间,增强结肠收缩幅度而调节结肠运动,结肠平滑肌M受体是白芍总甙作用途径之一.

Abstract:目的:在人外周血中有效地诱导、培养树突状细胞(dendriticcells,DC),并鉴定其表面分子的表达.方法:采用密度梯度离心的方法分离人外周血中的单个核细胞,在6孔板上贴壁2h,然后吸去悬浮细胞,用粒细胞-巨噬细胞集落群体刺激因子(GM-CSF)、白细胞介素-4(IL-4)、肿瘤坏死因子-α(TNF-α)细胞因子组合,刺激树突状细胞(DC)增殖、分化.用标有荧光素的单抗标记培养细胞,在激光共聚焦显微镜下拍摄标记有荧光素的细胞,在流式细胞仪上分析所培养的细胞.结果:该细胞直径在10~20μm,培养14天后,CD1a\++48%,CD83\++97%,CD40\++33%,人体主要组织相容性复合物(MHC)Ⅱ\++95%,CD80\++98%,CD86\++93%,CD83是DC的成熟标记.结论:该方法是一种可靠的诱导和培养DC的方法,对于乳腺癌患者还可通过动员等措施进一步提高DC的分离率.

Abstract:目的:探讨早发冠心病患者的心血管病危险因素和冠状动脉病变特点.方法:采用Judkins法对176例胸痛患者行冠状动脉造影,将108例确诊为早发冠心病患者与68例对照者比较,将早发冠心病组按年龄分为不同亚组,探讨早发冠心病组的心血管病危险因素和冠状动脉病变特点.结果:早发冠心病组血浆胆固醇、低密度脂蛋白胆固醇和脂蛋白(a)水平较对照组明显升高,而高密度脂蛋白胆固醇水平降低,高血压、吸烟史和心血管病家族史高于对照组,男性更容易患早发冠心病(P<0.05).两组空腹血糖和甘油三酯水平没有差异.早发冠心病组发病时越年轻,病变发生在左前降支的比例越高.40岁以下组单支血管病变比例明显高于40~55岁组(P<0.05).随着年龄增加,回旋支和右冠状动脉发生病变的比例增加,两支以上血管病变比例明显增加(P<0.05).结论:早发冠心病患者的心血管病危险因素较多,发病时越年轻,单支血管病变(左前降支)比例越高,预防冠心病应从尽早积极控制心血管病危险因素开始.

Abstract:目的:建立一种用尿酶免疫方法监测尿生殖激素促卵泡素(FSH-β)、雌酮结合物(E1C)、孕二醇-3-葡糖苷酸(PdG)在正常月经周期和人工受精(IVF)周期中的动态变化.方法:对11例正常生育年龄妇女用尿酶免疫法测定月经周期中尿FSH-β、E1C、PdG水平,10例同时抽血用放射免疫法测定血FSH、雌二醇(E2)、孕激素(P)水平.用尿酶免疫法测定18例IVF周期治疗的患者尿FSH-β、E1C、PdG水平的变化.结果:10例正常生育年龄妇女28天月经周期中血FSH、E2、糖尿FSH-β、E1C、PdG周期变化的一致性非常好,19个正常月经周期中尿生殖激素水平变化曲线也说明该点.18例IVF周期由于持续大量促性腺激素释放激素对垂体的降调节作用,在排卵期无FSH和LH峰.结论:酶免疫法测定尿生殖激素在IVF周期中、在监测和诱导排卵临床治疗上有良好的应用前景.

Abstract:目的:探讨结直肠癌缺陷基因在大肠腺瘤-癌演序中的蛋白表达及其临床意义.方法:以结直肠癌缺陷基因蛋白胞内部分独特氨基酸链的单抗15041A为一抗,免疫组化ABC法检测21例大肠粘膜、40例伴不同程度不典型增生大肠腺瘤、21例大肠癌组织的蛋白表达情况.结果:结直肠癌缺陷基因蛋白表达率:大肠粘膜及轻度不典型增生大肠腺瘤100%,中度不典型增生腺瘤93%,明显高于重度不典型增生腺瘤(45%)及大肠瘤(33%),P<0.05.结论:结直肠癌缺陷基因蛋白表达缺失与大肠腺瘤演变及大肠癌发展相关,免疫组化法检测结直肠癌缺陷基因蛋白对发现癌前病变、研究大肠腺瘤演变及大肠癌发展具有重要意义.

Abstract:目的∶用实验动物模型同时切除肝脏、胰腺或十二指肠,以探讨残肝再生反应.方法:60只SD大鼠被随机分为4组:①68%肝叶切除组;②68%肝叶切除加50%胰腺切除组;③68%肝叶切除加近端十二指肠切除组;④68%肝叶切除加50%胰腺切除加近端十二指肠切除组.每组中3只大鼠分别于术后12、24、48、72和168h处死并采集血液和肝脏样本.计算肝再生率以及免疫组化证实肝细胞增殖细胞核蛋白抗原(PCNA)的表达.结果:肝、胰切除和肝、胰十二指肠切除可显著减少残肝细胞DNA峰值期的合成,其延迟性肝再生反应延迟至术后168h.结论:起源于胰腺和十二指肠外分泌腺的多种因子对触发肝细胞的增殖可能起显著作用.

Abstract:目的:观察不同孕期胎盘滋养层细胞的超微结构和凋亡现象.方法:取早、中、晚各期胎盘组织,制备成透射电镜标本并观察.结果:细胞滋养层细胞(CT)在各妊娠时期形态结构相似,呈低分化细胞的特点.细胞核常染色质丰富,细胞质中有大量散在游离核糖体,内质网较少,高尔基复合体不发达.合体滋养层细胞(ST)为分化良好的细胞.细胞质内含丰富的扩张内质网,高尔基复合体发达.CT核数/ST核数随妊娠时间增加而减少.中、晚期有些ST核染色质呈边集、固缩等凋亡表现,且多发生在合体结,足月胎盘ST的凋亡指数AI大于中期(P<0.01).结论:妊娠过程中滋养层细胞的形态学变化与ST的凋亡有关.

Abstract:目的:探讨改良巩膜隧道式切口对现代白内障囊外摘除术的影响.方法:对75眼改良巩膜隧道式切口及59眼普通梯形切口的术中虹膜损伤情况、玻璃体脱出后人工晶状体植入情况、术后瞳孔上移情况、术后3个月矫正视力、角膜散光等行对照研究.结果:改良巩膜隧道式切口术中虹膜脱出及色素脱落少,玻璃体脱出后人工晶状体仍可植入,术后瞳孔上移少,角膜散光轻,术后3个月矫正视力、角膜透明情况与普通梯形切口无明显差异.结论:改良巩膜隧道式切口可有效地提高手术安全性,减少切口引起的并发症,值得推广使用.

Abstract:目的:结合断层解剖标本,分析肩关节及其周围结构的磁共振成像(MRI)表现.方法:分两组资料,MRI采用快速自旋回波纵向弛豫时间(T1WIFSE)序列扫描.一组为正常尸体标本,共40具,连续切割断层标本大体解剖与相应层面MRI扫描图像对照分析.另一组为正常肩关节人群,共14例.结果:MRI显示肩袖、盂唇、韧带、肱二头肌长头肌腱等,并且与断层标本相一致.这些结构由于其成分为纤维结缔组织或者纤维软骨,在MRI所有序列均表现为均匀低信号.轴位观察肩袖、盂唇、盂肱上、下韧带,喙肱韧带和肱二头肌长头肌腱较好.斜冠状位观察肩袖、盂唇、喙肱韧带、盂肱下韧带较清楚.喙肩韧带在3个标准体位显示均不满意.结论:MRI清楚显示了肩关节及其相关结构,对于了解正常肩关节的解剖和肩痛的病因病理诊断具有较大的价值.

Abstract:目的:研究糖尿病(DM)患者骨密度的变化,了解DM患者是否易合并骨质疏松及其与相关因素之间的关系,并进一步探讨其骨质疏松的发病机理.方法:采用以色列螺旋电子计算机断层扫描(CT-TWIN),测定78例DM患者及对照组第3腰椎(L3)骨矿含量(BMD),分析其年龄、性别、病程、空腹血糖(FBG)、糖化血红蛋白(HBA1C)、体重指数(BMI)等之间的关系.统计学方法采用SAS和SPSSFORWINDOWS95统计软件分析处理.结果:78例DM患者中,骨量下降及骨质疏松者53例,占68%;DM患者L3骨矿含量明显低于对照组.结论:DM患者较易发生骨质疏松,其发生率与病程、年龄及HBA1C呈负相关,与FBG、BMI相关性不明显.老年女性DM患者BMD比男性下降更为明显,考虑与绝经后卵巢功能丧失有关.

Abstract:ObjectiveTo study the effects of dendritic cells (DC) transfected with recombinant vaccinia virus encoding Epstein-Barr virus (EBV) latent membrane protein 2A(LMP2A) gene,and to provide evidence for further investigation on the therapeutic vaccines against EBV-associated malignancies.MethodsMature DC were transfected with EBV-LMP2A recombinant vaccinia virus (rVV-LMP2A).Before and after the transfection,the expression of surface antigens on mature DC including CD1a,CD83,CD40,CD80,HLA-DR was measured by fluorescence activated cell sorter (FACS) and the function of DC to stimulate allogeneic T cells proliferation was measured by mixed leukocyte reactions (MLR).ResultsLMP2A protein was highly expressed (66.1%) in DC after the transfection of rVV-LMP2A.No significant changes in the primary surface antigens expression and in the MLR were detected during the transfection.Transfected DC still had strong potential in stimulating the proliferation of allogeneic T cells.ConclusionRecombinant vaccinia virus was an effective and non-perturbing vector to mediate the transfection of LMP2A into DC.The functions of mature DC were not affected significantly by the transfection of Vac-LMP2A.This study could provide evidence for the further immunotherapy of EBV-associated malignancies,e.g.nasopharyngeal carcinoma (NPC).``

Abstract:ObjectiveTo elucidate the effect of tetrandrine on acute ischemic renal injury and its relation with apoptosis.MethodsA model for bilateral post-ischemic renal injury in rats was developed by clamping renal pedicles for 45 min.Renal tissular DNA fragmentation analysis and renal tissular HE staining were used.Also quantitative analysis of apoptosis in injured renal tubular epithelium was carried out by using TdT-mediated dUTP nick and labeling (TUNEL).ResultsApoptosis of renal tubular epithelium increased in acute ischemic renal injury.Tetrandrine could remarkably decrease the level of apoptosis in injured renal tubule while protecting renal tissue against the ischemic injuries.ConclusionTetrandrine could adjust the level of apoptosis in renal tubular epithelium and alleviate renal tissular injury.``

Abstract:ObjectiveTo investigate Fas,soluble Fas(sFas)and Fas-ligand(Fas-L)gene expression in thyroid tissues and thyrocytes from patients with Graves disease(GD)and to find the interrelationship between apoptosis and pathogenesis of GD.MethodsThyroid tissues were obtained from 7 GD patients and 3 healthy subjects who died accidentally.Thyrocytes were cultured in Eagle′s medium.Total RNA was isolated from thyroid tissues and cultured thyrocytes.The cDNA was prepared by reverse transcription and amplified for Fas,sFas and Fas-L by polymerase chain reaction(PCR).ResultsFas and sFas mRNA were detected in all samples from both GD and normal thyroid tissues and thyrocytes,but Fas-L mRNA was only found in GD thyroid tissues and thyrocytes.Semi-quantitative analysis showed that when compared with those of normal controls,the Fas and sFas mRNA levels were markedly increased in GD thyroid tissues(P<0.01),whereas in GD thyrocytes only the sFas mRNA levels was significantly elevated(P<0.01).ConclusionGene expression of Fas,sFas and Fas-L showed abnormality in both thyroid tissues and thyrocytes from GD.The increased production of sFas might be involved in the hyperplasia of thyroid gland.``

Abstract:Objective To study the modulation of interleukin-6 (IL-6) generation from human thyroid cells.MethodsThyroid tissues were obtained at surgery from patients with Graves disease.IL-6 in the supernatant of cultured thyrocytes was detected with ultra-sensitive ELISA.mRNA was extracted respectively from primary human cultured thyroidal cells under stimulated and unstimulated conditions,and semiquantitative reverse transcriptase-polymerase chain reaction (RT-PCR) technique was used for produce high concentration IL-6.Thyrotropin (TSH,103mU/L),tumor necrosis factor-α(TNF-α,10-1 U/ml),interleukin-1(IL-1,10 U/ml) and adrenaline(10-5mol/L) significantly increased IL-6 levels in the supernatant of cultured thyroid cells,whereas dexamethasone (10 mmol/L) markedly suppressed IL-6 release from thyrocytes.NaI of 10-4 mol/L results,at a concentration of 10-103 mmol/L,dexamethasone could gradually suppress IL-6 gene expression on human thyrocytes,while TNF-α could obviously stimulate the production of IL-6 mRNA in the range of 10-1-102 U/ml.On the level of 102 U/ml,IL-1 increased the expression of IL-6,but the same effect could not be shown when IL-1 was 10-1 or 10 U/ml.Sodium iodine (NaI) could not affect IL-6 gene from the level of 10-8 to 10-4 mol/L.ConclusionIL-6 produced by thyrocytes might be regulated by many factors that modulate thyroid functions in vitro as well as in vivo.``

Abstract:ObjectiveTo explore the relationship between the mutant genes of ACE,ATN,β3-AR and hypertension in patients with type 2 DM.Methods281 recruited Chinese subjects were divided into two groups according to the mellitus (DM):112 cases.The subjects were genotyped for the ACE gene,the ATN gene and the codon 64 of β3-AR gene polymorphisms by applying polymerase chain reaction (PCR),PCR restriction fragment-length polymorphisms screening with the use of endonuclease.ResultsOur study found that the frequency of D/D genotype and D allele of ACE gene,a/a genotype and an allele of ATN gene in HT patients without DM were increased (P all <0.05);that the frequency of codon 64 mutation of β3-AR gene also increased in HT patients with NGT (P <0.05).In the model of multiple factors non-condition-al Logistic regression analyses,HT had relationship with history of hypertension,age and glucose tolerance (OR=10.745 7,1.780 4,2.034 6;P=0.000 4,0.000 0,0.024 6;respectively),with polymorphism of ATN gene,β3AR gene,ACE gene (OR=2.273 6,1.935 3,1.830 9;P=0.054 3,0.028 7,0.043 2;resceptively).ConclusionThese results suggest that variants of ACE gene,β3AR gene,ATN gene were associated with HT in type 2 DM.``

Abstract:ObjectiveThis research aimed to study the effect of early application of continuous passive motion (CPM) on the drainage volume of knee joints post total knee arthroplasty (TKA).MethodsThe drainage volume was measured 1 h before,during and after CPM on the day and next day of the operation.ResultsThere was no significant difference in the drainage volume before,during and after CPM(P >0.05).ConclusionEarly application of CPM is safe for TKA patients.``

Abstract:Objective To study anticancer effect and ultrastructural influence of phosphorus-32 glass microspheres(32 P-GMS) injected in the hepatocellular carcinoma in nude mice.MethodsThe ultrastructural changes of tumor in both the treatment group and control group were examined by transmission electron microscope.ResultsIn the treatment group,a large number of tumor cells were killed and the death rate of tumor cells was much higher(35%-70%).Ultrastructurally,severe nuclear damage was observed in the dead cells.The early characteristics of necrosis such as margination of heterochromatin were also found in some tumor cells.Besides,well-differentiated tumor cells,degenerative tumor cells and some lymphocytes were seen.The skin and muscle close to the tumor were normal.In the control group,the tumor consisted of poor differentiated tumor cells,in which there were only a few appototic cells(5%).ConclusionThe results suggest that the local administration of 32P-GMS produces obviously the anticancer effect.``

Abstract:ObjectiveTo report a rare case of ALCL (Anaplastic Large Cell Lymphoma) with abundant neutrophils and necrosis and to analyse the histomorphologic subtypes and immunohistologic characteristics of ALCL.MethodsA biopsy of a painless tumor on a 26-year old male′s neck was taken for morphologic and immunohistochemical analyse.ResultsJudging from the morphologic feature of ALCL,the prognosis of the patient is good.ConclusionIt is necessary to do immunohistochemical staining in diagnosing an ALCL since p80NPM/ALK or ALK is the best antibody to distinguish ALCL from other lymphomas.``

Abstract:Dopamine (DA) agonists are playing increasingly important role in the treatment of not only advanced Parkinson′s disease (PD) and in PD patient with levodopa (L-DOPA)-induced motor fluctuations,but also in early treatment of the disease.This shift has been largely due to the demonstrated L-DOPA-sparing effect of DA agonists and their putative neuroprotective effect,based largely on experimental in vitro and in vivo studies.In this article we review the evidence of neuroprotection by DA agonists pramipexole,ropinirole,pergolide,bromocriptine and apomorphine in cell cultures and animal models of nigral injury.Most of the studies suggest that DA agonists exert their neuroprotection via directly scavenging free radicals or increasing the activities of radical-scavenging enzymes,and enhancing neurotrophic activity.The finding that pramipexole can normalize mitochondrial membrane potential and inhibit activity of caspase-3 in cytoplasmic hybrid cells made from mitochondrial DNA of nonfamilial Alzheimer′s disease patients,however,suggests even a broader implication for the neuroprotective role of DA agonists.Although the clinical evidence for neuroprotection by DA agonists is still limited,the preliminary results from several on-going clinical trials are promising.Several longitudinal studies are currently in progress designed to demonstrate a delay or slowing of progresion of PD using various surrogate markers of neuronal degeneration such as 18 F-L-DOPA PET and 123 I β-CIT SPECT.The results of these experimental and clinical studies will improve our understanding of the action of DA agonists and provide critical information needed for planning future therapeutic strategies in PD and related neurodegenerative disorders.``
