      Abstract:Objective;To study the pharmacokinetics of Nerve G rowth Factor(NGF)一liposome in rabbit by isotopic tracing.Methods①NGF was labeled with 'TC ,and its -i-ray counts were detected by isotopic tracing and trichloroacetic acid (TCA) precipitation method. From cpm of -1-ray and NGF content of samples, the standard curve was drawn and it was nearly a line.②9"'TC-NGF was packed with liposome and injected into rabbits (n=5) body from vena auris.③Blood samples of rabbit were collecteda td ifferentti mes (0.5,1,3,5,10,15 ,30,60,90,120,150,180,210,240m in)a fterin jection.T het echniques abovew ere used to detect NGF concentration in rabbit blood and data were input into computer to analyze the pharmacokinetic parameter by software(3P87). Results: The metabolism of NGF-Liposome in vivo in rabbits was fitted to the double-compartments model. With the dose of 40mg/kg,the p harmacokinetics parameters were as the follows:t-α=0.2338 h ,t-β=1.3229h ,A U C= 106.57ng/h.ml-1 Conclusion: NGF-liposome has some slowly-release function compared to NGF.

    • Genotoxic and tumorigenic pyrrolizidme alkaloids in Chinese herbal plants

      Abstract:Pyrrolizidine alkaloids are a class of hepatotoxic and tumorigenic compounds detected in Chinese herbal plants,contaminated foods, and dietary supplements. In this review, the sources, toxicity, genotoxicity, tumorigenicity, and the metabolic pathways,particular the activation pathways leading to hepatotoxicity and tumorigenicity, of pyrrolizidine alkaloids are briefly discussed, with a focus on the most recent important findings concerning the genotoxic mechanism by which riddelliine liver tumors. This mechanism involves the formation of 6,7-dihydro-7-hydroxy-1-hydroxymethyl-5H-pyrrolizine (DHP)-derived DNA adducts and may be general to most carcinogenic pyrrolizidine alkaloids.

      Abstract:Objective:To investigate the protective effects of genistein(gen)on human retinal pigment epithelium (RPE)cells injury induced by KC1 and CoCl2,Methods:Injury was induced by KC1 and CoC12 in RPE cells.The cells viability was measured by M1Tr assay and morphological changes of the cells were recorded.Results:KC1 could significantly decrease the survival rate of RPE cells at 1 2 h,and gen markedly increased the survival rate in a concentration-dependent manner.RPE cells presented progressively destruction of cell membrane at 2 h and pycnosis of cell nuclear at 4 h after 200 mmol/L KCl treatment.At 8 h after KCl treatment.pycnosis and batt-liked cell membrane could be seen Only pycnosis could be observed at 1 2 h.These changes could he inhibited by 200 ~mol/L gen.CoC12 could also inhibit the survival rate of RPE cells and gen could concentration.dependently elevate the survival rate.No significant morphological changes of RPE cells induced by CoC12 were f0und.Conclusion:Gen could be effective agent in protection on the injury induced by KC1 and CoC12 in cultured RPE cells.

    • Preparation and evaluation of nano-magnetoliposomes containing As2O3

      Abstract:Objective:To preparate nano—magnetoliposomes containing As2O3,a completely new form of drug for the targeted chemotherapy and thermotherapy to cancer cells.Metheds:The Manganese—zinc ferrite nanoparticles (Mn1.xZn Fe2O4)were prepared by improved wet-chemical reaction.Their characteristics were measured by transmission electron microscope (TEM )and PE thermal analysis system.The radiofrequence—induced hyperthermia was explored in vitro,and their cytotoxicity was measured by the means of MTT.Th e nano-magnetoliposomes were obtained by the combined methods of rotatory film,ultrasonic and churning at high speed.Their characteristics were measured by TEM,computer magic image analysis system and energy dispersive spectrometer(EDS).The ratio of envelopment of As203 was measured by atom fluorescence spectrometer.Results:Th e average diameter of magnetic particles was about 20-40 nm,round,the cytotoxicity of MnI n Fe2O4 was 0 class.Th e Curie temperature(Tc)of magnetic material varied from 97℃ to 140~C according to different percentages of zinc and manganese in Mn1.xZn Fe204.Th e temperature of magnetic fluid could rise to 35qE-47qE and keep stable in the high frequency alternating electromagnetic field. Th e nano—magneticliposomes contained As2O3 and Mn—Zn ferrite.Th e average diameter of the liposomes was (1 82± 1 25)nm.Th e envelopment ratio of As2O3 was 7 1.1 6% Conclusion:The nano—particles of Mn1. Zn Fe2O4 are perfect carriers for preparating magneticliposomes; Nano—magnetic—liposomes containing As203 have been successfully preparated.

    • Hydrolysis of soy isoflavone conjugates using enzyme may underestimate isoflavone concentrations in tissue

      Abstract:Objective: To investigate the differences of using enzymatic hydrolysis and acid hydrolysis for identification and quantification of isoflavone aglycones from biomatrices. Methods: β-glucuronidase/sulfatase isolated from Helix pomatia for routine enzymatic hydrolysis or 6N HCl was used to release glucuronide and sulfate conjugates in the serum, urine and tissue samples. Profiles of soy isoflavones after enzymatic hydrolysis or acid hydrolysis in several tissues of rat fed with diets containing soy protein isolate were also compared using LC/MS and HPLC-ECD. Results: Acid hydrolysis released more aglycone than enzymatic digestion ( P <0.05) in liver tissue. The total genistein, daidzein and other metabolites were 20% to 60% lower in samples from enzymatic hydrolysis than in acid hydrolysis. Conclusion: These results indicated that unknown factors in tissues reduced the enzymatic hydrolytic efficiency for releasing isoflavone aglycones even in optimized condition. This would underestimate isoflavone tissue concentrations up to 60%.

      Abstract:Objective:To investigate the effect of high glucose on the production of reactive oxygen species and type IV collagen in human mesangial cells and the intervention of tea polyphenols(TPs).Methods:The cells were cultured in high glucose with or without TPs up to 36 h.The supernatants were collected and the activities of SOD and the level of MDA were detected by spectrophotometry.The content of type IV collagen in the media was detected by radi0immun0assay.Immunocytochemistry was used to detect the expression of type IV collagen within the cells.Results:SOD was lowered,MDA increased,while type IV collagen increased in the high glucose media without TPs.However,SOD increased,MDA was lowered,type IV collagen was lowered with TPs compared with that only with high glucose.Conclusion:High glucose contributes to the generation of reactive oxygen species,an increase in type IV collagen produced by human cells and TPs can better control high glucose—induced reactive oxygen species,type IV collagen.

    • In vitro effect of biologically active coating on endothelialisation of Nitinol coils designed for the closure of intracardiac shunt

      Abstract:Objective: To evaluate in vitro the effect of biologically active coating on endothelialisation of Nitinol coils designed for the closure of intracardiac shunt. Methods: Covalently binding procedure was performed with chemical vapor deposition (CVD).Heparin(200 IU/ml ), r-hirudin(21 nmol/ml ), or fibronectin( 15 μg/ml), respectively, were bound to the basic coating( poly( amino- p- xylylene-co-p-xylylen) ). In vitro tests were performed on five different coating groups( n = 2 each) : uncoated Nitinol, basic coating, heparin-,r-hirudin-,and fibronectin-coating. Human umbilical vein endothelial cells(HUVECs) were cultured in the presence of tested coils for 48 and 72 h. Adhesion of HUVECs were evaluated with light microscopy,and with confocal laser scanning microscopy after double-staining of vinculin,phalloidin, Ki67 and fibronectin. Results: After 48 and 72 h of incubation,except HUVECs around the basic coating showing abnormal morphology, HUVECs were able to grow next to all kinds of coils. By CLSM adhering cells were typically confined to maximal three secondary coil windings. HUVECs adhered best to the fibronectin coated coils,followed by the uncoated Nitinol. HUVECs barely adhered to the basic coating. The adhesion to heparin coating or r-hirudin coating was observed with various cell densities. Heparin coating seemed to support cell adhesion better than r-hirudin coating did. The HUVECs assembled fibronectin matrix on the fibronectin coating and uncoated Nitinol coil. Vinculin was expressed on both fibronectin coating and uncoated Nitinol as well, mostly diffused,but occasionally also in focal contacts. Ki67 expression was mainly noticed on the uncoated Nitinol coil and fibronectin coated coil, only occasionally on other materials. The densities and morphology of HUVECs, adhered to the correspohding well surfaces, were similar for all wells. The only exception was the well with the basic coating coil after three days of incubation. In this case, the cell density was diminished, and the cells became more elongated. The cells grown on the well surfaces exhibited a more profound matrix assembly and more pronounced focal adhesions than the cells adhered to the coil surfaces did. Conclusion: Except for basic coating,biologically active substance coated or uncoated coils show no acute cytotoxicity on HUVECs. Due to better cell proliferation,cell matrix assembling,and focal adhesion,only fibronectin coating improves the endothelialisation of Nitinol coils.

      Abstract:Objective:To study the effect of the different development periods to the homeodomain transcription factors Nkx2.2 and Nkx6.1 expression in rat pancreas.Methods:Pancreas were isolated from SD rats on gestational days 1 8.5.new h()rn and adult by microscope.Pancreas were probed by the pancreatic molecule marker—insulin and amylase.A semi—quantitative reverse transeriptase polymerase chain reaction (RT-PCR)was performed to assess the expression of transcription factors Nkx2.2 and Nkx6.1 in diferent developing periods in rat pancreas.Results:Nkx2.2 and Nkx6.1 gene expression increased from gestation to new born and then decreased in adult.Conclusion:Different development periods can regulate the expression of Nkx2. 2 and Nkx6.1 gene.New born period is a special time when loss nutrition from mother body,and initial of autoimmune and digestion system. Fhe resire in cell neogenesis may lead to the increase in the gene expression of the homeodomain transcription factors Nkx2 2 and Nkx6. 1.

    • Expression of PTTG gene and PTEN gene in human breast carcinomas

      Abstract:Objective;To study the expression and correlations of PTTG,PTEN and clinicopathological factors.Methods:In s itu hybridization and PV-9000 Polymer Detection System were used to detect the expression of PTG and PTEN in 71 cases of breast carcinomas (BRCA). Results; Tumor tissue expressed a significantly higher level of PTG and lower level of PTEN at mRNA or protein than the corresponding normal tissue (P<0.05), over-expression PTTG protein in BRCA correlated with lower-differentiation and lymph node metastasis. There were lots of correlations in BRCA between lower-expression of PTEN protein and the tumors'pectoralis invasion, higher pathological grade, and pathologic classification. The expression of PTTG was not correlated with that of PTEN at mRNA or protein level in BRCA. Conclusion: Over-expression of P1TG and lower-expression of PTEN may be related to human breast carcinogenesis.

      Abstract:Objective: To establish the initial symptoms and physical signs of osteosarcoma from the records of the first medical visit and to identify early characteristics of the diseases to shorten the delay of diagnosis. Methods: A group of patients with osteosarcoma in extremities was identified from Nanjing Jinling Hospital. Records from the first medical visit due to symptoms and physical signs related to the bone tumor were obtained from 71 patients with osteosarcoma. Results: The results of the data recorded about the first medical visit,reasons for consultation, pain,trauma,initial diagnosis,delay in diagnosis were reported respectively. Conclusion: Pair an initial symptom of osteosarcoma, and was intermittent but not frequently felt at night. A history of trauma was common, but the clinical course often diverged from what was expected from trauma. The most important clinical feature was a palpable mass. This finding emphasizes that a thorough physical examination is absolutely necessary.

    • Effect of MAPK activation in response to high glucose on human glomerular mesangial cells

      Abstract:Objective:To investigate the activity and possible role of MAPK in mesangial cells in response to high glucose and to research the mechanism of diabetes nephopathy. Methods:The human mesangial cells were cultured and stimulated by high glucose.The MAPK activity was measured by liquid scintilation counting. Production of fibronectin was analyzed by ELISA. The expression of c-fos was assayed by immunochemistry. Results: Compared with the control glucose, high glucose increased the activity of MAPK, the expression of c-fos and the production of FN. When PD98059 were used, these three all decreased. Conclusion: High glucose can active MAPK, increase the expression of c-fos and the production of FN and contribute to diabetes nephopathy.

    • Regulative role of tumor necrosis factor-α on TSG-6 gene expression in both undifferentiated and matured 3T3-L1 adipocytes

      Abstract:Objective:To explore the regulative role of tumor necrosis factor-a(TNF-a)on tumor necrosis factor alpha-stimulated gene-6(TSG-6)gene expression in both undifferentiated and matured 3T3-Ll adipocytes.Methods:3T 3-Ll preadipocytes were cultured and differentiated into the matured adipocytes.Recombinant TNF-a at different co ncentrations( 0.1,1.0,10 .0n g/ml)was added to the Culture medium 2days post-confluent undifferentiated adipocytes(D ay0 )or fully differentiated adipocytes( Day1 0)for various times(0, 0.5, 2.0, 6.0, 12.0, 24.0 h). Total RNA from these adipocytes was extracted and the expression of TSG-6 mRNA was evaluated by RT-PCR.Results:}1 Treatment of un differentiated3 T3-Lla dipocytesw ithT NF-aa tdi ferentco ncentrations (0.1,1. 0,10 .0n g/ml) resulted in the significant decrease in the expression of TSG-6 mRNA. When 0.1 ng/ml TNF-a was applied,the levels of TSG-6 gene expression significantly decreased by 67.30% at 2 h, and 76.05% at 24 h. When 1.0 ng/ml TNF-a was used, the levels of TSG-6 gene expression significantly decreased by 52.79% at 0.5 h, and 99.20% at 24 h. When the cells were treated with 10.0 ng/ml TNFa,TS G-6g enee xpressionw asa lmostfu llyin hibited( 96.14% decreased)at 0 .5h .(2 )Treatment of matured3 T3-Ll adipocytes with TNF-a at different concentrations( 0.1,1.0,10 .0n g/ml)a lsor esultedi nt hes ignificantd ecreasei nt hee xpressiono fT SG-6m RNA.When the cells were treated with 0.1 ng/ml TNF-(x, the levels of TSG-6 gene expression decreased by 33.73% at 6 h, and 97.39% at 12h .W hent reatedw ith1 .0n g/mlT NF-a,th ele velso fT SG-6g enee xpressiond ecreasedb y7 8.68%a t6 h ,which sustained u ntil24 h .Whenter atedw ith1 0.0n g/mlTN F-a,th ele velso fT SG-6g enee xpressiond ecerasedb y9 6.27% at2 h a ndT SG-6gene expression was almost fully inhibited. Conclusion: TNF-a can downregulate the mRNA expression of TSG-6 in both undiferentiated and matured adipocytes. The inhibitive efe ct of TNF-a on TSG -6 gene expression is in dose and time-dependent manne r.

    • Neurophysiological characteristics in infants and young children with auditory neuropathy

      Abstract:Objective: To analyze neurophysiological characteristics in infants and young children with auditory neuropathy (AN) and explore their clinical significance. Methods: Audiological measurements(acoustic immittance, EOAEs, ABR, CM, MLR and ERPs) and peripheral neurological tests were conducted and evaluated in 13 infants and young children with AN. Six of them received highresolution temporal bone CT scans and/or cerebral MRI examinations. Results: All of the children showed type "A" tympanograms with abseatation of stapedial reflexes. EOAEs were normal in 12 of 13 subjects. In one child who had a history of anoxia during the birth, the EOAEs were not elicited. Click-evoked ABRs were absent in 12 of 13 subjects when maximum output of the instrument was reached. The CM potentials were presented bilaterally in all individuals, which were independent of the EOAEs and ABR. Of eight cases tested, all had clear MLR and six showed normal ERPs(P300 and MMN). Peripheral neurological tests and radiological findings were within the normal ranges. Conclusion: The diagnosis of AN in infants and young children should focus on analyzing their neurophysiological characteristics,especially on CM,MLR and ERPs. Combined use of EOAEs, ABR and CM was recommended for hearing screening on newborns with high risk factors.

    • Qualitative Analysis and Law Application of Medical Personnel Receiving Brokerage of Drugs and Appliance

      Abstract:The medical personnel receiving brokerage of drugs and appliance is illegal, howerer, accusing them of bribery remains judicatory controversial. The author expatiated on components and law presumption, made a study of disputes on qualitative analysis of this behavior, and brought out juristic inconsistency with crime of bribery punished. Finally according to legal principle of misdeed that law without an express provision does not criminate in our country criminal law, and referring to License Physician Law and Drugs Management Law etc,the author thought for the general medical personnel who receives brokerage of drugs and appliance, it is a best way to educate; for those without any changes, some administrative punishments are necessany and even it is legal to revoke his/her physician license while remit criminal punishments, for those hospital gorernor, they might be convited of bribery.

      Abstract:Objective:To observe the effects of in tracerebroventricular injection of leptin on the expression of obese gene in adipose tissue and the level of serum leptin and serum insulin, C-peptide in diet-induced obese rats and analyze the correlation among the expression of obese gene and the level of serum leptin and the correlation among serum leptin and insulin and C-peptide.Methods:①Total RNA from the adipose tissue was extracted by using TRIzol isolation kit. Obese gene mRNA expression and semiquantitation were studied by reverse transcription-PCR.②At the time of sacrifice, blood samples were collected by cardiopuncture directly and centrifuged at 2 0 00rlm for 20 minutes.Serum was stored at- 20'C and serum insulin and C –peptide and leptin were detected by using radioimmunoassay and the murine leptin ELISA kit. Results:①The levels of obese gene mRNA expression in retroperitoneal adipose tissue were decreased in obese rats after leptin administration, and lower than in the groups with the saline and the obese control group.In addition,there was no si gnificant difference in all normal groups.② Compared with the ratswith leptin injection and the normal groups, the level of serum leptin in obese rats with the saline and without treatment was increased more significantly. The concentration of serum leptin had significant positive correlation with the level of obese gene expression.③ In obese groups with leptin injection, the levels of serum insulin and C-peptide decreased significantly than those in obese rats with the saline and without treatment. Moreover, there were no obvious changes between the obese groups with leptin injection and all normal groups. The serum insulin and C-peptide concentrations were strongly positively correlated with the level of serum leptin in rats. Conclusion: Leptin could decline the expression of obese gene and the level of serum leptin and insulin and C-peptide in diet-induced obese rats after intracerebroventricular injection so as to improve the leptin and insulin resistance.

      Abstract:Objective: To investigate the points of the clinical diagnosis and surgical treatment for pancreatic and/or duodenal injuries. Methods: Clinical data of 30 patients who suffered from pancreatic and/or duodenal injuries were reviewed. Results: There were 29 cases who received surgical management. Of the 30 cases, 22 cases were cured, seven cases died, and postoperative complications occurred in 16 cases. The cure rate was 73.3%. Conclusion: Pancreatic and/or duodenal injuries are severe abdominal injuries and difficult to treat. The mortality and complication rate are high. The keys to successful treatments for pancreatic and/or duodenal injuries are early diagnosis, careful exploration and proper operational management.

The authors thought it was a wrong judegement of law pr9perty between doctors and patients in the public medical and health cause, which severiously violated the legal rights of both doctors and patients. No matter in continental law system or GB/USA law system, such medical torte and injury compensation are fit into the national compensation. Since our National Compensation Law does not exclude `public hospital’,due to torte,will p ay compensation to citizens,juridical persons and other organizations when their legal rights are damaged, even with the current law, torte and injury compensation for public units should have no obstacles when brought into national compensation.

      Abstract:Objective:The aim of the study was to identify the single nucleotide polymorphisms (SNPs) on type 2-diabetes susceptibility genes located on chromosome 9 D9s171. Methods: Putative type 2-diabetes susceptibility genes were selected based on their known potential involvement in insulin signaling pathway. Sequences of proximal promoter, coding regions (including sequence near splicing site) of each of 4 susceptibility genes were amplified by polymerase chain reaction (PCR). The PCR was carved out using DNA samples from 9 normal controls and patients of 15 type 2-diabetic pedigrees, which had high NPL Scores on D9sl71. PCR products were subjected to the direct bi-directional sequencing, and the sequencing data were analyzed with software Polyphred. SNPs in these genes among type 2 diabetics were detected. Results: A total of 14 SNPs were identified for 4 susceptibility genes; there were 11 high frequency SNPs and 3 low frequency SNPs. Conclusion: SNPs were identified in 4 candidate genes from patients of type 2 diabetes in East China, and these results should be confirmed or rejected in large sample genotyping study and association study in the future.

      Abstract:Objective: To investigate the effects of allitridin on the expression of transcription factor T-bet of cytomegalovirusinduced myocarditis in mice and to analyze its role in anti-cytomegalovirus mechanisms. Methods: Sixty mice were randomly divided into allitridin therapy group, infected controls group and bland controls group. Allitridin therapy group were given to mice via the intraperitoneal same volume of 0.89% sodium chloride and bland controls group were only given with the same volume of 0.89% sodium chloride, without infected MCMV. All experimental mice were sacrificed on day 3, 5, 7 and 14 i.p. ( n = 5 per time point). The expression levels of transcription factor T-bet were measured by RT-PCR. The expression levels of Th1 cytokine IFN-γ were measured by ELISA. Results:MCMV infection could markedly down-modulate the expression of IFN-γ and T-bet mRNA ( P < 0.01). Allitridin significantly up-regulated the expression of Th1 cytokines IFN-γ and Th1-specific transcription factor T-bet mRNA ( P < 0.01). Conclusion: Allitridin up-regulates the expression of transcription factor T-bet mRNA which is an important, but not the only factor to mediate the constitutive expression of Th1 cytokines IFN-γ.

      Abstract:Influence of the metrological control on the hospital culture was caused by the application of engineering-technique in modern medical service. The correlation between the metrological control and the hospital culture was studied in the implication of the hospital culture and the content of the culture environment of hospital.The conclusion showed that th e metrological control was contained in the material culture of hospital,the system culture and the spiritual culture. In the culture environment of hospital, the metrological control is an important factor to maintain the hospital image, to affect the thinking mode of medical personnel, to restrict the behavior of medical personnel, and to promote the accomplishment of hospital aim. The authors pointed out that the metrological control occupied an important place in the culture environment of hospital, which is worthy of close attention.

      Abstract:Objective: To investigate the effects of different anesthesiaes on the activity of aldose reductase(AR) in RBC and the levels of plasma nitric oxide(NO) in patients undergoing gastric cancer operation. Methods: Twenty-eight patients undergoing gastric cancer operation were randomly divided into two groups with 14 cases for each. Anesthesia was maintained with 1.5-2.0MAC isoflurane plus epidural block in group Ⅱ. Venous blood samples were collected to measure the AR ctivity in RBC, the levels of plasma NO and glucose.Results: The plasma levels of glucose were increased significantly at 90 min after incision and kept at the high levels till the 1st postoperative day in group Ⅰ ( P < 0.01 or P < 0.05) as compared with those before anesthesia, the changes in group Ⅱ were same as in group Ⅰ ( P < 0.01) ,but not on the 1st postoperative day. There was no significant difference between the two groups. The activity of AR in RBC was increased significantly and the level of plasma NO was decreased significantly on the 1st postoperative day in group Ⅰ ( P <0.05). In group Ⅱ, AR and NO changed slightly,but these changes were not significant. The extent of change in the activity of AR or the level of NO no the 1st postoperative day in group Ⅱ was less than that in group Ⅰ ( P < 0.05). Conclusion: Polyol pathway in RBC activated under surgical hyperglycemia while the production of NO is inhibited in patients undergoing gastric cancer operation, which can be effectively attenuated by combined general-epidural anesthesia.

      Abstract:Objective:To measure parameters of survival curves of a pulmonary adenocarcinoma cell line SPCA-1 in vitro.M ethods:Two groups were studied in this experiment, which included irradiation alone and combination of irradiation with mild hyperthermia.The former was given single high photon irradiation with 6M X-ray 2, 4, 6, 8街respectively. Hyperthermia was applied to the latter,41.5 degrees Centi grade, 1 h, after having received same irradiation. The cel surviving fractions were determined by clonogenic assay and the parameters of the multi-target/single-hit model, Do, D9, as well as those of the linear-quadratic model, ratio of alpha and beta, were elicited. Results: The parameters of the irradiation combining with hyperthermia, Do and Dq, were 1.390 and 1.426 Gy,whereas they were 1.693 and 2.453 Gy for irradiation alone, respectively. The surviving fraction at 2 Gy(SFZ) was 0.793 versus 0.532 of the combination. The ratio of alpha and beta increased from 0.992 to 7.725 Gy and the SFZ of the linear-quadratic model decreased from 0.743 to 0.459. Conclusion: The values of Do and SFZ show that the radiosensitivity is low relatively, though mild hyperthermia might induce radiosensitization of SPC-A-1 cells.

      Abstract:Objective: To investigate the role and mechanism of adrenomedullin (AM) in diabetic nephropathy. Methods:This research observed the changes of the expression and secretion of AM, TGF-β1 in the cultured human mesangial cells under high glucose conditions and the contents of the laminin and type Ⅳ collagen in the supernatants, and the effect of intervention with AM on the changes.Results: High glucose condition resulted in increase in the expression and secretion of AM, TGF-β1, laminin and type Ⅳ collagen, and AM could reverse the influence of high glucose on the cultured human mesangial cells. Conclusion: The results showed that high glucose condition in one of the stimulating factors of AM, and the renal protective action of AM may be associated with suppression of TGF-β1 and reducing excessive accumulation of laminin and type Ⅳ collagen.

      Abstract:Objective:To summarize the clinical data and investigate the treatment of patients with prematuer ovarian failure.Methods: Thirty-one patients with premature ovarian failure were evaluated from January 1999 to December 2003. Results: Among 31 cases, there were 22 infertile patients and 9 patients with fertility, The average period of amenorrhea was 2.8 years(from 5 months to 7 years),in which there were 7 cases with the abrupt cessation of menstruation and 24 patients with oligomenorrhea and gradually to cessation of menstruation . 16 patients had the symptoms of estrogen deficiency and 48.4% of patients had genital and breast atrophy at different rates. The circulating concentration of estradiol was reduced and follicle-stimulating hormone and luteinizing hormone levels increased. All patients were treated with a combination of estrogen and progesterone. The cure rate of premature ovarian failure was 22.6% and no patient was treated to have a pregnancy. Conclusion: Premature ovarian failure is a disease of multifactors. Further study and emphasis ought to be taken to investigate it.

      Abstract:Objective:To investigate the application value Of rotating fetel head by hand to correct persistent occipitotransverse position and occipitoposterior position during labour, so as to reduce the caesarean section rate Methods:Eighty-nine pregnant women with persistent occipitotransverse position and occipitoposterior position when cervix dilated over 6 cm were performed rotating fetel head by hand, and 72 pregnant women with similar obstetric condition were treated as control group without any intervention.Results:75 of the 89 were successfully rotated into occipitoahead position in the former condition (84.2%),who all underwent vaginal delivery. In study group, the caesarean section rate was 13.5%(12/89), compared with the 65.2%(47/72) of that of the control, and it showed significant difference in the two groups(P<0.01).Conclusion; To perform the rotating timely can reduce the caesarean section rate and lower the complication for lying-in women and babies obviously.

      Abstract:Objective: To investigate the effects of anti-interleukin- 10 monoclonal antibody( anti-IL- 10MAb) on systemic candidiasis. Methods: Control group(only candidiasis injection), and disposal group( candidiasis infection accompanying with anti-IL-10MAb infection) of cyclophosphamide-induced immuno-suppressed murine systemic candidiasis model were set in this study. Colony Forming Units (CFUs) of infected kidneys and spleens were determined using plating dilution method. The histological studies for infected livers, spleens,and kidneys were applied. Levels of interferon gamma(IFN-γ) in spleen tissue homogenare were also measured by enzyme-linked immunosorbent assay. Results: In kidneys, the numbers of CFU of the disposal group were much lower than that of the control group; the numbers of CFU in spleens were similar to the control group. The histopathological scores of the disposal group were much better than that of the control group in kindneys with significant differences( P < 0.01 ). In spleens,the histopathological scores of disposal group were also better than that of control group,but no statistic significant differences were observed ( P > 0.05). And the spleen IFN-γ level of the disposal group was significant higher than that of the control group( P < 0.01). Conclusion: Anti-IL- 10MAb effects on systemic candidiasis was concluded.

      Abstract:Objective:To establish fibroblasts stably expressing human bone morphogenetic protein 2 (hBMP2). Methods:Eukaryonic expression vector(pcDNA3.1-B2) was transduced into NIH3T3 cells using SofastTM, a new generation cationic polymer gene transfection reagent. The positive cell clones were selected with G418. The stable transfection and expression of BMP2 in the NIH3T3 cells were determined by RT-PCR and immunohistochemical stain. Results: BMP2 mRNA was transcripted and expressed in the transfected NIH3T3 cells. Conclusion: With positive compound transfection, outside human BMP2 gene can be successfully transducted into NIH3T3 cells, which is the key step to induce periodontal cells to osseous phenotypes.

      Abstract:Objective:To introduce a fiber-optic fluorescent biosensor for choline.Methods:The choline fiber-optic biosensor designd included a LED,a bifurcated fiber,two kinds of filter,and a spectrometer.T he sensing end of the film was coated with Nafion film dispersed with choline oxidase and oxygen sensitive luminescent Ru(Ⅱ)complex.Results:The lowest limit of this choline fiber-optic biosensor for testing was 1×10-9mol/L with a respondence time T≤10 sec.Meanwhile,the sensor had a high anti-jamming ability.Conclusion:The fluorescence intensity of Ru(Ⅱ)is related to the choline concentration.The choline fiber-optic biosensor proves to be a low-cost,high-performance and portable apparatus.

      Abstract:Objective:To study the structure and wettability changes on the surface of soft-lining material after being treated with low temperature plasma of Ar. Methods: 32 specimens were prepared and 16 specimens were treated with Ar plasma for 20 minutes in plasma equipment.Surface structure and wetting angle were evaluated before and after plasma treatment.Results:(1) The number of C -H was reduced, which indicated crosslinked layer was formed on the surfac‘soft-lining material. (2 The percentage of C-0 increased from 23.56% to 26.18%.③The wetting angle was reduced significantly. Conclusion: Treatment of Ar Plasma can change the surface structure and wettability of soft-lining material.
