Abstract:Objective:To discuss the cytotoxic activity of CTL,stimulated by adenovirus containing the universal DR-restricted Th helper epitope(PADRE) combined with HLA-A1 and HLA-A2 restricted epitopes from mucin4 gene(PADRE/MUC4) infected DCs. Methods:Recombinant adenovirus with PADRE/MUC4 gene was prepared,DCs of HLA-A2 positive donors were induced from PBMC in vitro. PBMC primed with adenovirus infected DC for 3 weeks,the specific cytotoxic activity of CTL was detected by standard 4 h Cr51 release assay and Elispot method. Results:Lymphocytes primed with pAd-CMV-PADRE/MUC4 caused potent cytotoxic responses. By contrast,lymphocytes primed with a GFP expressing adenovirus or mock-infected DCs had no cytotoxic effect. Conclusion:Infection of DCs with an adenovirus encoding PADRE combined with epitopes from mucin4 gene may be one possible strategy for immunotherapy of MUC4-expressing tumors.

Abstract:Objective:To explore the in vitro immune response to tumor associated antigen MUC4-derived cytotoxic T lymphocyte(CTL) epitope-pulsed dendritic cell(DC). Methods:DCs was induced from the HLA-A*0201-positive healthy individuals’ peripheral blood mononuclear cells(PBMC). DCs’ phenotype was identified by flow cytometry. CD8+ T cells were isolated and purified through MACS(magnetic activated cell sorting). Mature DCs were pulsed with each of the five synthesized peptides which were selected as possible CTL epitopes by software analysis previously. Autologous CD8+ T cells from the HLA-A*0201 healthy donor were stimulated with the peptide-pulsed DCs as CTL. CTL activity was assessed by lactate dehydrogenase(LDH) release assay and IFN-γ released by enzyme-linked immuno spot assay(ELISPOT). Results:Mature DCs could be induced from PBMCs through several cytokines’ action. CTL induced by peptide P01204 could lyse T2 cells pulsed with peptide P01204[(9.03 ± 0.24)%,(19.62 ± 0.44)%,(27.93 ± 2.22)% at ratios of 5 ∶ 1,10 ∶ 1,20 ∶ 1] and HCT-116 cells(MUC4+,HLA-A2+)[(7.26 ± 0.18)%,(16.83 ± 0.40)%,(25.23 ± 1.35)% at ratios of 5 ∶ 1,10 ∶ 1,20 ∶ 1]. The number of CTL induced by peptide P01204 which could secret IFN-γ(130.33 ± 6.58) was obviously higher than that in the negative group. Conclusion:Tumor associated antigen MUC4-derived HLA-A*0201-restrictive cytotoxic T lymphocyte(CTL) epitope P01204 could induce PBMCs’ CTL reaction. The CTL could secret immunologic active material to induce the specific target cells’ lysis.

Abstract:objective:To construct the recombinant adenovirus containing pancreatic tumor associated MUC4 antigen and provide the basis for further research on pancreatic tumor vaccine. Methods:The HLA-A1 and HLA-A2 restricted epitopes of MUC4 was predicted by BIMAS and ProPred. The combined polyepitope gene with kozac sequence,sign peptide and PARDE was synthesized. The combined polyepitope was subcloned into pAdshuttle-CMV shuttle plasmid(pAdshuttle-CMV-PE),and then transformed into competent E.coli BJ5183 cells with backbone plasmid already. The homologous recombinant adenoviral plasmid pAdeasy-1-PE was validating with PacⅠ digesting and sequencing to verify their fidelity. Ad293 cells were transfected with PacⅠ linearized recombinant and lipofectamine reagent. Identified by green fluorescence protein(GFP) expression and PCR method,the virus was amplified by infection 293A cells repeatly. Viral particle concentration was determined by TCID50 plus flow cytometer. The virus was transduced into COS-7 cells,and expression of polyepitope protein was demonstrated by the GFP expression and SDS-Page analysis. Results:The recombined adenovirus containing combined polyepitope was constructed successfully and the titer was 10 pfu/ml. Conclusion:A recombinant adenovirus containing combined polyepitope was constructed successfully,a specific protein corresponding with the predicted mass was found in virus transduced COS-7 cells.

Abstract:Objective:To isolate and identify differential expression proteins in embryonic rat testis after maternal exposure to di-nbutyl-phthalate(DBP). Methods:Two-dimensional gel electrophoresis was performed on testes samples from hypospadiac and normal healthy rats. After silver staining,the differential expression proteins were analyzed by ImageMaster 2D analysis software. Results:Nine differential protein spots were found,calculated according to 5-fold expression amount. Five expressed highly in the hypospadiac rat testis but lowly in the normal rat testis,while the other four expressed just the opposite way. Conclusion:The protein profiles of differential expression between the normal and hypospadiac rat testes were established,and some differential proteins were found. The data obtained in this study might be helpful for further studies on the isolation and identification of differentially expressed proteins in hypospadiac rat testes.

Abstract:Objective:To investigate the protective effects of grp75 gene on hydrogen-peroxide(H2O2)-induced cell damage in rat glioma C6 cells. Methods:Cultured C6 glioma cells were transfected with grp75. Assays of MTT,LDH,FCM analysis and hoechst staining were used to investigate the protective effect of grp75 on H2O2 stimulated C6 cells. Results:Transfection of C6 glioma cells with grp75 resulted in the increased cell viability and decreased apoptosis ratio as compared to the control. Conclusion:These data indicated that grp75 might inhibit H2O2-mediated cytotoxic effect on C6 cells.

Abstract:Objective:To establish a rat model with Alzheimer’s disease(AD) induced by incubated Aβ1-40 and study the changes of learning and memory ability and pathology. Methods:Incubated Aβ1-40 was microinjected into the bilateral hippocampus of each rat. Jumplag test and Morris water maze test was used to evaluate rat learning and memory function. The pathological changes were observed by haematoxylin-eosin(HE) and silver staining. Results:The capability of learning and memory was impaired significantly after Aβ1-40 injection. Neuron death,glial reaction and neurofibrillary tangles were found in or around the injection sites. Conclusion:These results suggest that the aggregated and deposition of Aβ1240 in the hippocampus was related to the impairment of learning and memory, and the neuronal degeneration in the rats which showed similar behavior and pathological characterizations of AD. It could be used as an animal model for the prevention and cure of AD.

Abstract:Objective:To investigate the expression of Kiss-1 in the hypothalamus of precocious female rats induced by bisphenol A at various stages of development. Methods:Thirty 21-day-old normal female Sprague-Dawley rats were randomly assigned into three groups of 10 rats:control group 1(prepuberty),control group 2(early puberty),experimental group(bisphenol A treatment group). The rats from the experimental group were gavaged with 400 mg/(kg·d) of bisphenol A at 21 days of age. The two control groups were gavaged with normal saline instead. For the determination of Kiss-1 mRNA expression in the hypothalamus,the rats of the bisphenol A treatment group were sacrificed during the first diestrus and meanwhile the rats of the control group 1 were sacrificed when they were at prepuberty;the control group 2 rats were sacrificed at the first distrust(early puberty). The expression of Kiss-1 mRNA in the hypothalamus among three groups was detected using RT-PCR. Results:Kiss-1 mRNA expression in the hypothalamus in bisphnol A group increased by 2.35 fold,compared with control group 1(P < 0.05). There were no statistical significance in the Kiss-1 mRNA expression between control group 2 and bisphenol A group. Conclusion:Environmental endocrine disruptor bisphenol A could induce precocious puberty,while expression of Kiss-1 might play a role in the progress.

Abstract:Objective:To establish a cell model for ultraviolet damage to the skin and to study whether Eucommia has a protective effect against the UV-induced damage. Methods:Through the method of MTT,the most suitable Ultra Violet Radiation(UVR) time was screened out to best establish the model and found out the optimal concentration of Eucommia Ulmoides Oliv. Three groups were set in this experiments,control,UC and UE. UC were treated by a 30-minute-UVR,UE were treated by Extracts from Eucommia Ulmoides Oliv culture and then a 30-minute-UVR. Microscope and electromicroscope were used to morphologically observe the extent of damage by UVR. Further assessment was executed on the membrane potential of mitochondria and the content of ATP to better understand the extent of mitochondria damage. Results:30-minute-UVR was an ideal choice for the model. Extracts from Eucommia Ulmoides Oliv at the concentration of 0.4 mg/ml was demonstrated the most effective. Electromicroscrope showed cell would decline after a 30-minute-UVR. The protective effect on cell in UE group could be observed morphologically. The result of membrane potential of mitochondria was that UE declined by less extent than UC group compared with the control group.The ATP content ranging from high to low was UC,UE and control group. Conclusion:A damaged cellular model was established by 30 minutes of UV radiation,and the results showed the photoprotective function of eucommia at the concentration of 0.4 mg/ml.

Abstract:Objective:To study the effect of cardiomyocyte microenviroment in vitro on bone marrow mesenchymal stem cell(MSCs) in diffierentiating into cardiomyogenic cell. Methods:Bone marrow cells were isolated form SD rat’s leg bone by their adherence to plastic in vitro,the MSCs were purified by culture-expanded and passaged. After treaded with DNA demethylating agent 5-azacytidine(5-aza). In experiment group the MCSs were grown,which were cultured in dishes seeded with cardiomyocytes from neonatal rats. In control group the MSCs were cultured only. The MSCs differentiation was observed by microscope,immunohistochemistry staining for Demin and cardiac troponin I(cTnI),and the positive time were observed. Results:In control group,after treated with 5-aza for 10 days,the clones of adhenrent fibroblast-like cells were formed,and they connected with adjoining cells 15 days and formed myotube-like structures about 20 days later. But the experiment group they formed myotube-like structures after about 17 days. The results of immunohistochemistry staining indiated that the experiment group’s positive time was earlier than the control group’s. Conclusion:The cardiomyocyte microenviroment are thought to be a key factor in inducing bone marrow mesenchymal stem cells to differentiate into cardiomyogenic cells.

Abstract:Objective:To study the alteration and clinical significance of plasma endothelin-1(ET-1) after transcatheter closure of atrial septal defect(ASD). Methods:Radioimmumoassay was used to measure plasma concentration of ET-1 in 40 ASD patients before,30 minutes after and three days after transcatheter closure of ASD. Results:The plasma level of ET-1 in ASD patients was remarkably higher than that of the control,no matter the ASD patients’ pulmonary arterial pressure was increased or not. In ASD patients,the plasma concentration of ET-1 correlated with the pulmonary arterial pressure,pulmonary to systemic flow ratio(Qp/Qs) and the size of ASD(r = 0.868, r = 0.746 and r = 0.670,respectively(P < 0.01). The plasma level of ET-1 in ASD patients after ASD closure decreased significantly compared with the level before ASD closure. And the plasma concentration of ET-1 in ASD patients three days after ASD closure was lower than that at 30 minutes after ASD closure. Conclusion:The endothelium function in patients with ASD can be improved after transcatheter closure of ASD. The plasma level of ET-1 may be used as a marker for evaluating the effect of transcatheter closure of ASD.

Abstract:Objective:To investigate effects on action potential(AP) of Spraque-Dawley rat by different isomers of amlodipine(Aml). Methods:Calcium-tolerant ventricular myocytes were isolated by enzyme digestion. AP changes in S-Aml,R-Aml and R,S-Aml with concentrations of 0.1,0.5,1.0,5.0 and 10.0 μmol/L were recorded respectively by whole cell configuration patch clamp. Results:①AP maximal velocity,AP amplitude and overshoot had no significant changes in S-Aml,R-Aml and R,S-Aml with concentrations of 0.1,0.5,1.0,5.0 and 10.0 μmol/L(P > 0.05); ②Fifty percent of Action potential duration(APD50) were(36.2 ± 8.2)-(33.9 ± 7.7)-(30.2 ± 6.8)-(22.6 ± 5.1) and (15.1 ± 3.4)ms,respectively(P < 0.05) with the increase of S-Aml concentrations from 0.1 μmol/L to 10 μmol/L;APD50 were (39.2 ± 9.2),(36.7 ± 7.9),(33.8 ± 7.2),(25.4 ± 5.9) and (21.7 ± 5.2)ms,respectively(P < 0.05) with the increase of P,S-Aml concentrations from 0.1 μmol/L to 10 μmol/L;APD50 were(45.1 ± 11.3),(46.2 ± 10.8),(44.9 ± 7.3),(44.8 ± 8.2) and (45.7 ± 9.4)ms,respectively(P > 0.05) with the increase of P-Aml concentrations from 0.1 μmol/L to 10 μmol/L. Conclusion:APDs were gradually shortened with the increase of S-Aml and R,S-Aml concentrations. S-Aml and R,S-Aml might influence APDs by blocking L-type calcium channel;R-Aml had no effect on APD,and might have no influence on L-type calcium channel.

Abstract:Objective:To observe the relationship between overexpressed β1-AR and the contractile amplitude in rat heart failure ventricular myocytes. Methods:Heart failure model was made with isoprenaline. Then,ventricular myocytes were isolated,cultured and infected with adenoviruses containing the sequence for human β1-adrenoceptor. The contents of β1-AR and the single ventricular myocytes contraction amplitudes were tested after the cells were infected with adenoviruses for 24 h. Results:The content in heart failure group was lower than that in control group. Overexpression of β1-AR markedly increased the content. The isoprenaline-stimulated contraction amplitudes in failure group were significantly lower than that in control group. Overexpression of β1-AR improved the contraction amplitudes. Selective β1-AR antagonist CGP20712A descended the isoprenaline-stimulated contraction amplitudes in heart failure myocytes infected with or without adenoviruses. Selective β2-adrenoceptor antagonist ICI118,551 partially decreased the contraction amplitude in heart failure myocytes infected with or without adenoviruses. Conclusion:The results suggested that overexpression of β1-AR increase the contraction amplitude in heart failure myocytes. The effect was directly related to β1-AR. β2-AR participated in the isoprenaline-stimulated contraction amplitudes.

Abstract:Objective:To evaluate the regional myocardial systolic function in the patients with hypertrophic cardiomyopathy(HCM) by TDI imaging. Methods:Doppler myocardial imaging of 83 HCM patients(HCM group) and 30 healthy subjects(control group) was collected,and their regional peak velocity,systolic strain,disaplacement,time to peak systolic strain in the apical,mid and basal segment was measured respectively at septum and lateral wall. The data was compared and analyzed between the two groups. Results:The strain value of HCM group was significantly lower in the place where hypertrophy was happened. There was a significant correlation between midseptal strain and thickness of intraventricular septum. Velocity and displacement was decreased in septum and lateral wall,but have no significant correlation with the extend and site of hypertrophy. No significant difference was found about the time to systolic strain of left ventricle. Conclusion:TDI imaging could detect the global and regional myocardial function of HCM patients,and provide a new quantitative parameter for evaluating the regional myocardial functions.

Abstract:Objective:To investigate the effects of ulinastatin(UTI) on systemic and cerebral inflammatory response and on brain protection during cardiopulmonary bypass(CPB). Methods:Twenty-four patients,undergoing elective cardiac valve replacement were randomly divided into 2 groups:ulinastatin group(Group U,n = 12) and control group(Group C,n = 12). In group U,UTI(1×104 U/kg) was infused intravenously after the induction of anesthesia and during CPB, respectively. Each infusion time was about 1 h. In Group C,normal saline was given instead of UTI. Blood samples were taken simultaneously from artery and jugular bulb after induction of anesthesia(T1),0.5 h after CPB beginning(T2),1 h(T3),4 h(T4) and 20 h(T5) after CPB termination for determination of TNFα,IL-6 and S100B. S100B samples were taken only from jugular bulb. The jugular bulb-arterial gradients of these cytokines(△TNFα and △IL-6) were calculated. Results:The concentration of TNF-α,IL-6,△TNF-αand △IL-6 elevated dramatically at T2,T3,T4,T5 in both groups(P < 0.01 or P < 0.05). TNF-α,IL-6,△TNF-α and △IL-6 at T2,T3,T4,T5 in group C were higher than that at the corresponding time point in group U. Compared with T1 point,the level of S100B increased markedly 30 min after CPB(T2)(P < 0.01),reached peak level at T3 point and began to decrease at T4 point in both groups. The levels of S100B at T2,T3,T4,T5 in Group U were lower compared with group C at the corresponding time point. Conclusion:Systemic and cerebral inflammatory response during CPB could be alleviated by ulinastatin, and Jugular bulb blood levels of S100B Protein decreased by ulinastatin. Ulinastatin had protective effect on the brain injury during CPB.

Abstract:Objective:To evaluate relationship between different inferior wall ST-segment deviation on the initial electrocardiogram in patients with anterior wall acute myocardial infarction and lesion spite and length of anterior descending branch according to coronary arteriongraphy. Methods:According to different inferior wall ST-segment deviation, seventy-eight patients with anterior wall AM1 were divided into two groups:Group A,inferior ST-segment elevation;Group B,inferior ST-segment depression. To Compare its clinical feature and infarction related coronary artery(IRCA) between two groups. Results:As for IRCA,90.63% of group A suffered distal lesion of left anterior descending artery(LAD);80.43% of group B suffered proximal lesion of LAD. 81.25% of group A had reentry of LAD.0% of group B had reentry of LAD. Conclusion:Inferior wall ST-segment depression on the initial electrocardiogram in patients with anterior wall acute myocardial infarction indicated proximal lesion of anterior descending branch. PCI operation success rate of the group B was higher. Inferior wall ST-segment elevation in initial electrocardiogram in patients with inferior wall acute myocardial infarction indicated distal lesion of ALD and reentry of ALD.

Abstract:Objective:To make a comparative analysis of early and mid-term outcomes in two different age groups receiving off-pump coronary artery bypass grafting(OPCAB). Methods:From January,2005 to May,2007,145 consecutive isolated patients undergoing OPCAB were divided into two groups according to their age:the relatively elder group(≥70 years),n = 68 and the relatively younger group(<70 years),n = 77. Early and mid-term outcomes were comparatively analyzed with the combined follow-up of letter,telephone and out-patient clinic re-examination. Results:There were no hospital deaths in each group. There was no significant difference in postoperative average ICU stay and hospital stay time,major cardiac events rate in two groups. But compared with the relatively younger group,the average intubation time,blood transfusion and atrial fibrilation incidence were remarkably higher in the relatively elder group(P = 0.001). Conclusion:OPCAB provides satisfactory early and mid-term clinical outcomes for the elderly as well as younger patients.

Abstract:Objective:To study the clinical pathologic significance of tumor budding in colorectal carcinoma. Methods:The tumor budding of 83 cases of colorectal carcinoma were detected by HE staining and EnVision immunohistochemical staining. Results: Tumor budding was found in 46 cases of colorectal carcinoma(55.4%). Tumor budding was more frequently observed in colorectal carcinoma with lymph node metastasis than in those without lymph node metastasis(P < 0.01). The incident of tumor budding was higher in colorectal carcinoma with lymphatic involvement than in those without lymphatic involvement(P < 0.01). Among pTNM staging,the detection rate of tumor budding in colorectal carcinoma with Ⅲ stage was higher than those with Ⅱ stage(P < 0.01). Tumor budding were not associated with sex,age,tumor size,site,differentiation and venous invasion(P > 0.05). Conclusion:Tumor budding was a potential pathological marker suggesting aggressive or invasive growth of colorectal carcinoma. The detection rates of tumor budding was improved by CKpan immunohistochemistry.

Abstract:Objective:To explore the expression of survivin in bone marrow cells from children with leukemia. Methods:The expression of survivin were detected in 54 children with leukemia and 36 control subjects by RT-PCR. The relationship between the early treatment response and survivin expression was analyzed in all leukemia children. Results:In control sample survivin was not detected,and survivin was detected in 41 leukemia patients. Its positive rate was 79.53%,and expression of survivin in leukemia was significant higher than that of control. Early complete remission rate in survivin positive children with leukemia was significantly lower than that in survivin negative ones. Conclusion:The detection of survivin expression in leukemia might provide a tool for diagnosis,prognosis and MRD of leukemia.

Abstract:Objective:To review the experience of using synchronized intermittent mandatory ventilation(SIMV) in the treatment of neonatal acute lung injury. Methods:Sixty-two neonates suffering from acute respiratory distress syndrome and requiring mechanical ventilation were devided into two groups,32 by SIMV and 30 by intermittent positive pressure ventilation(IPPV). FiO2,PIP,oxygenation index,average time of ventilation and complications were compared between the two groups. Results:FiO2 and PIP dropped in 6 hours of ventilation in SIMV group. Oxygenation index was obviously improved 2 h after use of SIMV and 6 h after use of IPPV. Average time of ventilation was shorter in SIMV group than in IPPV group. The rate of complications was less in SIMV group than in IPPV group. Conclusion:SIMV was superior to IPPV in the treatment of neonatal acute lung injury.

Abstract:Objective:To explore the changes and clinical significances of vascular endothelial growth factor and interleukin-6 serum level in multiple myeloma(MM). Methods:The serum levels of VEGF and IL-6 were detected by enzyme-linked immunosorbent assays in 30 patients with MM. The relationship between the increased serum levels of these two cytokines and MM clinical stage as well as therapic effect was studied. Results:The MM patients had significantly higher VEGF and IL-6 serum levels than the control(P < 0.001). Serum level of VEGF and IL-6 were elevated and correlated with clinical stages. VEGF were highest(mean 327.65 pg/ml) in stage Ⅲ,and there was significant difference(P < 0.001) of the VEGF serum levels between stageⅠand stageⅡ. IL-6 levels increased progressively according to clinic stages and it had significant difference among stages respectively(P < 0.01). The IL-6 and VEGF serum levels were higher in patients with the abnormal Hb and beta2-MG(P < 0.05). No correlation was found between the serum VEGF level and percentage of bone marrow plasma cells and monoclonal Ig levels. Conclusion:An elevated serum level of VEGF and IL-6 was associated with a poor prognosis. The two cytokines might be useful in monitoring the disease progression and triggering the targeted MM therapies.

Abstract:Objective:To analysis the features of apoptosis in placental trophoblasts of patients with severe preeclampsia,and study the relationship between the trophoblastic apoptosis and the etiology and pathophysiology of preeclampsia. Methods:To obtain the evidences of apoptosis in the placental trophoblasts of patients with severe preeclampsia,the techniques of the electron microscope scanning,TUNEL and FCM were applied. Results:①In the ultrastructure of severe preeclamptic placental trophoblasts under the electron microscope scanning,there also were typic morphologic changes of apoptosis including anomalo-nucleus,pyknosis,abnormal distribution of heterochromatin,deeply-stained mitochondrial matrix in cytoplasm,clouding or disarrangement of mitochondrial crest,vacuoles in placental trophoblast cells,and so on. ②Under the detection of TUNEL technique,it was found that there were more numbers of apoptotic trophoblasts in the placenta of severe preeclampsia with fewer apoptotic cells appearing in the placenta of NC. ③In normal placental tissues,no markedly apoptotic peak(Apo peak) was found in the diagrams of FCM. However,there all were the markedly apoptotic peaks in the FCM diagrams of the severe preeclamptic placental tissues. Conclusion:Trophoblstic apoptosis in the placenta may contribute to the etiology or pathophysiology of preeclampsia.

Abstract:Objective:To study the correlation between midpregnancy adiponectin level and gestational diabetes mellitus(GDM). Methods:75 g oral glucose tolerance tests were performed in 305 pregnant women at 24 to 28 weeks of gestation. Fasting maternal blood samples were drawn to determine plasma concentrations of adiponectin,glucose,insulin,free fatty acid,and blood lipids. Blood samples at 1 and 2 h after an oral glucose load were obtained to measure plasma glucose and insulin concentrations. Results:Plasma adiponectin concentrations were significantly lower in women who had GDM than in those who had normal glucose tolerace(NGT)(P = 0.014). The multiple logistic regression model showed that the association between BMI before pregnancy and GDM remained significance(P = 0.017) after adjusting for plasma adiponectin and FFA concentrations. The obese women had significantly lower midpregnancy adiponectin concentrations than non-obese women. In the NGT group,maternal adiponectin concentrations were significantly negatively correlated with plasma fasting insulin,1-h glucose,triacylglycerol and maternal BMI(at blood collection and at delivery),and positively correlated with high-density lipoprotein cholesterol and total cholesterol concentrations.In the GDM group,maternal adiponectin level was negatively correlated with neonatal birth weight and low-density lipoprotein cholesterol concentrations. Conclusion:Midpregnancy lower adiponectin level might be associated with a higher risk of GDM.

Abstract:Objective:To evaluate the effect of inhibiting scar formation and safety of reforming viscocanalostomy with absorbable and non-absorbable pericardial patch implantation. Methods:18 New Zealand white rabbits were equally divided into three groups as experimental groups A,B and control group C. One piece of non-absorbable pericardial patch was implanted into each eye of group A while absorbable one into each eye of group B.Nothing was implanted in group C during reforming viscocanalostomy. Internal structures and lasting time of the scleral space are observed. Results:The formed filter blebs lasted longer in group A and B than group C. The pathology process showed that the spaces in sclera of group A and B were remained while scar formation was found in control group at the twelfth week. Conclusion:Implantation of absorbable and non-absorbable pericardial patches in reforming viscocanalostomy is safe and effective that can inhibit scar formation. Reaction caused by absorbable pericardial patches is lighter than that of non-absorbable ones. Absorbable pericardial patches may lead to better result in inhibiting scar formation.

Abstract:Objective:To investigate the rebuilding of the stereoacuity in intermittent exotropia patient and the related factors affecting the stereoacuity before and after operation. Methods:56 cases of intermittent exotropia were examined before operation and 6 months after operation respectively. Near stereoacuity were tested by Yan shaoming stereogram and distance stereoacuity by Synoptophore. Results:The loss of the near stereoacuity of intermittent extropia patients was less than that of distance stereoacuity. There was no significant difference of the near stereoacuity before and after operation,but the distance stereoacuity were obviously improved by operation. Fusional faculty before operation was essential for the recovery of stereopsis. The difference of the recovery of stereopsis between patients who were younger than 9 years old and older than 9 years were significant. Conclusion:The operation can significantly improve the distance stereopsis of the intermittent exotropia patients. Patients with intermittent exotropia should resort to surgical treatment as early as possible. The early correction of eye position will benefit the restoration of stereopsis and the achievement of functional cure.

Abstract:Objective:To investigate the expression and distribution of aquaporin4(AQP4) in gastric carcinoma tissue and analyze its clinical significance. Methods:The gastric carcinoma tissues were obtained and the expression and distribution of AQP4 in carcinoma and normal stomach tissues were examined by immunohistochemistry,RT-PCR and Western-blot analysis respectively. Results:AQP4 was expressed mainly in the normal gastric tissue basal side of parietal cell and principal cell and absent in carcinoma tissues. Conclusion:AQP4 is mainly expressed in normal stomach tissue and not found in carcinoma tissues.

Abstract:Objective:To investigate the expression of MK protein and its correlation with neovascularization in cervical squamous cell carcinoma,so as to provide experimental foundation for prevention and therapy of CSCC. Methods:The expression of MK protein and microvascular density(MVD) labeled by CD34 were examined by immunohistochemistry S-P method in 58 cases of CSCC and 10 cases of normal cervical epithelium. Results:The expression of MK protein and MVD were significantly higher in CSCC than that in normal cervical epithelium(P < 0.01). Overexpression of MK protein in CSCC was related to tumor size and FIGO staging,but had no relation to age,differentiation degree and lymphnode metastasis. The MVD was significantly higher in the MK protein positive tumors than in negative ones(P < 0.05). Conclusion:The expression of MK protein might play an important role in carcinogenesis and development of CSCC and have close relation with tumor neovascularization. MK could be taken as a new target for prevention and therapy of CSCC.

Abstract:Objective:To investigate the value of 18F-fluorodeoxyglucose(FDG) dual-head coincidence imaging in the diagnosis of recurrent and metastatic differentiated thyroid carcinoma. Methods:Twenty-eight patients with differentiated thyroid carcinoma after post-therapy and radiotherapy with 131I were studied with 18F-FDG dual-head coincidence imaging and 131I whole body imaging. The results were analyzed by calculating its sensitivity, specificity,accuracy,positive predicative value(PPV) and negative predicative value(NPV), compared with thyroglobulin detected. Results:There were 8 true positive cases,13 true negative cases,1 false positive case,6 false negative cases in 28 postoperative patients detected by 18F-FDG dual-head coincidence imaging. The sensitivity, specificity, accuracy, positive predicative value and negative predicative value were 57.1%,92.8%,75.0%,88.9% and 68.4%,respectively. while levels of Tg were more than ≥10 ng/ml,the sensitivity of detecting recurrent carcinoma were higher in cervical lymph lode and bone by 18F-FDG dual-head coincidence imaging. Conclusions:18F-FDG dual-head coincidence imaging holds promising clinical value for diagnosis of patients with differentiated thyroid carcinoma after resection and radiotherapy with 131I, especially for the domestic widespread application of SPECT DHC instrument, which can lower the expense of detection practically.

Abstract:Objective:To explore the feasibility and rationality of low-dose spiral CT scan in the examination for the head of rabbit and provide the basis for the low-dose CT scanning in the head of neonate. Methods:The technical parameters were 120 Kv,3 mm slice thickness and every slice was scanned using tube current of 200,110,70 mAs. Five same regions of interest were measured on every image. The homogeneity and noise level of CT were appraised. The technical parameters were 120 kV,3 mm slice thickness. 200,110,70 mAs were employed in each same slice. CT images were evaluated by four doctors using single-blind method,and 3 degrees including excellent image, good image and bad image. Results:The phantom measurement showed that the lower of CT dose and the lower of homogeneity. The lower of CT dose and the higher of noise leve1. The image quality had no significant diference between five conditions(pitch 1.0,200,110,70 mAs;pitch 1.5,200,110 mAs),(P > 0.05). Conclusion:Low-dose spiral scan for examinating the head of rabbit might not only guarantee image quality but also reduce the radiation during CT scan.

Abstract:Objective:To investigate the association between two polymorphisms of BARD1 gene and the susceptibility of breast cancer in a Chinese population. Methods:Genotyping were performed using PCR-restriction fragment length polymorphism(PCR-RFLP) assay in 422 histological-confirmed breast cancer cases and 476 age frequency-matched cancer-free controls. Results:The genotypes distribution of the BARD1 Pro24Ser(rs1048108) and Arg378Ser(rs2229571) polymorphisms were significantly different between cases and controls. Multivariate logistic regression analysis revealed that variant genotypes of Pro24Se polymorphism(CT/TT) and heterozygotes of Arg378Ser polymorphism were both associated with a significantly decreased risk of breast cancer(adjusted OR=0.64,95%CI=0.49-0.84;adjusted OR=0.54,95%CI= 0.33-0.86,respectively) compared with the wild-type genotype(24CC and 378GG). Furthermore,these two polymorphisms may have joint effect on breast cancer risk. Individuals with more than one protective allele showed a significantly decreased risk compared with those with 0-1 protective allele(adjusted OR=0.67,95% CI=0.51-0.87). Stratified analysis showed that the protective effect of Pro24Ser polymorphism was more evident in subjects with age less than 52,premenopausal status,without family history of cancer and Arg378Ser variant genotypes carriers. Conclusion:Pro24Ser(rs1048108)and Arg378Ser(rs2229571) polymorphisms of BARD1gene were associated with a significantly decreased risk of breast cancer. And these two polymorphisms may jointly affect individuals’ susceptibility. More rigorous laboratory studies and larger epidemiological studies with ethnically diverse populations are warranted to confirm our findings.

Abstract:Objective:To investigate the distribution of the genotyping and the characteristics of VNTRs in clinical Mycobaterium tuberculosis(MTB) strains isolated from Jiangsu province with multi-loci of variable number tandem repeats. Methods:168 strains of Mycobacterium tuberculosis were selected from three parts of Jiangsu province. Eleven loci of VNTR from the genome of Mycobacterium tuberculosis were selectd and analyzed for the polymorphism in distribution by means of PCR,agarose electrophoresis and the BioNumerics(Version 3.0)software. Results:It was found that distribution of VNTR from 168 strains of Mycobacterium tuberculosis isolated showed definite polymorphism,and 10 different genotypes could be classified in which most of strains belonged to one genotype(75.1%),and the others were scattered,such as Ⅱ(5.4%),Ⅳ(4.2%),Ⅶ(9.5%). We also found that there was visiblely different among three parts of Jiangsu province. Conclusion:It is concluded that the VNTR loci in the genome of Mycobacterium tuberculosis isolated in Jiangsu showed marked polyporphism,most of the 168 strains belonged to the one genotype(Ⅷ). The distribution of main prevalent strains among three parts were the same(Ⅷ), but other genotypes distribution of strains were different.
