    • Cloning and sequence analysis of β-actin gene from Aedes albopictus (Diptera: Culicidae)

      Abstract:Objective: To obtain the complete β-actin gene from Aedes albopictus. Methods: Total RNA was extracted from C6/36 cells. Degenerate primers were designed based on the β-actin sequences of An. gambiae, Ae. aegypti, Cx. pipiens pallens and D. melanogaster. By RT-PCR, the product was amplified, purified, cloned into the pGT vector and sequenced. The β-actin sequence was aligned and phylogenetically analyzed by the BLAST program and the CLUSTAL W program. Results: A sequence of 1132 bp including an open reading frame of 1131 bp was obtained(GenBank DQ657949). The deduced protein had 376 amino acids. Aligned to SWISS-PROT, it exhibited a high level of identity with β-actins from Anopheles, Drosophila and Culex at the amino acid sequence level. Phylogenetic analysis indicated that Ae. albopictus β-actin was much more homologous with invertebrate β-actin than with vertebrate β-actin. Conclusion: The gene may be used as the internal control in the experiments of Ae. albopictus.

    • Treatment of herniated lumbar disc by percutaneous laser disc decompression combined with synchronous suction technique through syringe

      Abstract:Objective: To compare the outcomes of percutaneous laser disc decompression (PLDD) and PLDD with synchronous suction through syringe in the patients with herniated lumbar disc(HLD). Methods: Forty-two patients with HLD on MRI and those who did not respond to conservative treatment for 6 weeks were randomly divided into group A and group B. In group A, the patients were treated with PLDD and those in group B with PLDD and synchronous suction through syringe. GaAIAS diode laser at 810 nm was used for the ablation in both groups. The treatment effect was evaluated by modified MacNab’s criteria on the 7th, 30th and 90th day. Results: Evaluated by modified MacNab’s criteria, the percentages of the excellent and good cases in total patients treated for group B and A were 80.95% and 57.14% on the 7th follow-up day, 85.71% and 66.67% on the 30th follow-up day, and 95.24% and 71.43% on the 90th follow-up day, respectively. Conclusion: Synchronous suction technique through syringe during PLDD improves the overall outcome in the treatment of herniated lumbar disc.

    • Comparative study of effects of bone marrow cell vs. Ad5-HGF administration via non-infarct-related artery injection in myocardial infarction in swine

      Abstract:Objective: To evaluate the effect of transplanting bone marrow-derived mesenchymal stem cells(BM-MSCs) or adenovirus5-hepatocyte growth factor(Ad5-HGF) via non-infarct-related artery injection in swine myocardial infarction models. Methods:BM-MSCs were obtained from swine bone marrow and expanded in vitro to a purity of >50%. A myocardial infarction(MI) was created by ligating the distal left anterior descending artery in swine. Either BM-MSCs(5 × 106/ml) or Ad5-HGF(4 × 109 pfu) were transfused via the right coronary artery(non-infarcted artery) four weeks after MI. Gate-controled cardiac perfusion imaging was performed at the end of four and seven weeks after LAD ligation, to evaluate heart function and cardiac perfusion. Morphologic and histologic characteristics of the hearts were also studied. Results:(1)The gate-controlled cardiac perfusion imaging showed that the improvement in LVEF was greater in both treatment groups than in control group at the 4th weeks. (2)In both treatment groups, capillary density was significantly higher than that of control group(P < 0.05). Conclusion:BM-MSCs or Ad5-HGF transplantation via non-infarcted artery administration can stimulate angiogenesis and improve heart function, but there was no difference in therapeutic efficacy between BM-MSCs and Ad5-HGF.

    • No association between thrombospondin-4 A387P polymorphism and acute coronary syndrome in Chinese Han population

      Abstract:Objective: The thrombospondin-4(TSP-4) gene G29926C(A387P) polymorphism was recently reported to be associated with an increased risk of MI(myocardial infarction) in American population. However, several subsequent studies produced controversial findings. The aim of this study was to explore the possible association between TSP-4 A387P polymorphism and ACS(acute coronary syndrome) in Chinese Han population. Methods:A case-control study including 412 patients with ACS and 337 controls free from CAD(coronary artery disease) was conducted. TSP-4 A387P polymorphism was determined by PCR(polymerase chain reaction) and RFLP(restriction fragment length polymorphism) analysis. Results:Slightly decreased frequency of GC genotype was observed in patients with ACS, compared with controls(5.3% vs. 7.1%), but the difference did not reach statistical significance(P = 0.31). Similarly, the prevalence of C allele was 2.7% and 3.6% for ACS and control groups, respectively (P = 0.32). None of homozygote was detected for C allele. Further analyses in subjects subgrouped according to sex and age also showed no association of TSP-4 A387P polymorphism with ACS. Furthermore, after adjustment for conventional risk factors by multiple logistic regression analysis, the carrier prevalence of C allele did not differ significantly between ACS and control groups(OR = 0.85; 95% CI:0.45-1.59; P = 0.60). Conclusion:The present study suggested that the TSP-4 A387P variant showed a low prevalence compared with western populations and failed to associate with an altered risk of ACS in Chinese Han population. The findings further supplement experimental data for TSP-4 gene study of coronary disease.

    • Inhibitory effect and mechanism of chuanxiongzine on multiplication of VSMC

      Abstract:Objective: To study the inhibitory effect of chuanxiongzine on vascular smooth muscle cell (VSMC) proliferation and explore its molecular biology basis. Methods: we selected the VSMC cultured 4~8 generation from rat aorta thoracalis as research object. The objects were divided into four groups(Ⅰ)control group,(Ⅱ)chuanxiongzine(50 μg/ml)group,(Ⅲ)chuanxiongzine (100 μg/ml) group and(Ⅳ) chuanxiongzine(200 μg/ml) group. The inhibitory effect of chuanxiongzine on VSMC proliferation was investigated by cell counting, MTT and 3H-TdR incorporation assay. In order to illuminate the molecular biology mechanism of chuanxiongzine inhibiting VSMCs proliferation, the expression of proliferating cell nuclear antigen(PCNA) and C-myc were detected. Results: Chuanxiongzine could inhibit the proliferation of VSMC significantly in a dose- and time-dependent manner, compared with control group (P < 0.05). The expression of PCNA and c-myc were inhibited obviously and correlated with the concentration of chuanxiongzine (P < 0.05). Conclusion: Chuanxiongzine may play a considerable role in VSMC proliferation process. The inhibitory effect of chuanxiongzine in a dose- and time-dependent manner can be realized via down regulating the expression of PCNA and c-myc. In this study, The great theoretical fundament about Chinese medicine, which is used to treat atherosclerosis(AS), has been obtained.

    • Value of D-Dimers in patients with acute aortic dissection

      Abstract:Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimers were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ ml, P < 0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.

    • Protection of rat islet viability following heme oxygenase-1 gene transfection via adenoviral vector in vitro

      Abstract:Objective: To investigate the effect of Heme oxygenase-1(HO-1) gene transfection on the viability of cultured rat islets, and to explore the potential value of HO-1 gene in islet transplantation. Methods:Recombinant adenovirus vector containing human HO-1 gene(Ad-HO-1) or enhanced green fluorescent protein gene(Ad-EGFP) was generated by using AdEasy system respectively. The rat islets were transfected with Ad-HO-1, Ad-EGFP or blank vector and then cultured for 7 days. Transfection was confirmed by expression of EGFP and human HO-1 protein detected by fluorescence photographs and western blot, respectively. The insulin release upon different concentration of glucose stimulation was detected using insulin radioimmunoassay kit, and stimulation index(SI) was calculated. Glucose-stimulated insulin release was used to assess islet viability. Results:Adenovirus vector successfully transferred HO-1 gene to rat islet cells in vitro, and the insulin release upon high level of glucose stimulation and stimulation index(SI) of Ad-HO-1-infected islets were significantly higher than those of Ad-EGFP-infected islets and control islets(P < 0.05). Conclusion: Adenovirus-mediated HO-1 gene transfection is a feasible strategy to confer cytoprotection and therefore protect the viability of cultured rat islets.

    • Effects of apigenin on cell proliferation of human pancreatic carcinoma cell line BxPC-3 in vitro

      Abstract:Objective: To observe the effects of apigenin on cell proliferation of human pancreatic carcinoma cell line BxPC-3 in vitro. Methods:The inhibitive effects of apigenin at different concentrations(0 -滋mol/L, 100 -滋mol/L, 200 -滋mol/L, and 400 -滋mol/L) on human pancreatic carcinoma cell line BxPC-3 were detected by MTT assays, transmission electron microscope,agarose gel electrophoresis and flow cytometry. The immunohistochemistry was used to detect the expression of Bcl-2 and Bax gene. Results:Apigenin at different concentrations could inhibit the proliferation of human pancreatic carcinoma cell lines BxPC-3, and the inhibitive effect was dose-dependent. The cell cycle of pancreatic carcinoma cells was arrested at G2/M phase. The results of immunohistochemistry showed that the density of apigenin increased, and the expression of Bcl-2 gene was reduced gradually. At the same time the expression of Bax gene was enhanced. Conclusion:Apigenin could inhibit the proliferation of human pancreatic carcinoma cell lines BxPC-3 in vitro. The effect of apoptosis was accompanied with the expression of Bcl-2 decrease and Bax increase.

    • Effects of pioglitazone on proliferation and differentiation of human preadipocytes

      Abstract:Objective: To explore the effects of thiazolidinediones(TZDs) pioglitazone on proliferation and differentiation of human preadipocytes. Methods:Omental adipose tissue biopsies were obtained from 15 patients who were undergoing elective open-abdominal surgery. The primary culture and differentiated induction of human preadipocytes were performed, and the human preadipo-cytes were treated with pioglitazone at different concentrations at proper moments. Dynamic morphological changes of the human preadipocytes were observed, and their proliferation and differentiation were assessed with Colorimetric MTT Assay and Oil Red O Staining. Results:After 24 hours and 72 hours with pioglitazone, 0.1 μmol/L(μmol/ml) pioglitazone increased the MTT values of the human preadipocytes by 25.3% and 34.8%,respectively(P < 0.05), while 1 μmol/L pioglitazone by 27.4% and 26.6%(P < 0.05), compared with the control group without pioglitazone. The human preadipocytes with pioglitazone cumulated more adipose in the endochylema than those without pioglitazone obviously. 0.1 μmol/L pioglitazone increased the differentiation degree of the human preadipocytes differentiated for 8-10 days by 44.81% and 1 μmol/L pioglitazone by 53.76%(P < 0.05). Conclusion:Thiazolidinediones pioglitazone may significantly promote the proliferation and differentiation of the human omental preadipocytes.

    • Effects of poly (ADP-ribose) polymerase inhibitor on early peripheral neuropathy in streptozotocin-diabetic rat

      Abstract:Objective: To explore the effects and mechanisms of poly(ADP-ribose) polymerase(PARP) inhibitor 3-aminobenzamide on nerve lesions in streptozotocin-diabetic rats. Methods: Experimental rats were divided into normal control group(NC group), diabetic control group(DC group)and diabetic group treated with 3-aminobenzamide(DT group).Nerve conduction velocity (NCV),serum superoxide dismutase (SOD) activity and serum malondialdehyde (MDA) concentration,phosphocreatine (Pcr),creatine (Cr) concentration in sciatic nerves were evaluated after 4 weeks. Results: SOD,Pcr activity,and NCV were higher (P < 0.05) and MDA concentration were significantly lower in DT group, compared with DC group (P < 0.01). Meanwhile, ATP and Cr in sciatic nerves were similar in DT group, compared with DC group (P > 0.05). Conclusion: 3-aminobenzamide could alleviate the established functional and metabolic abnormalities of early DPN in the streptozotocin-induced diabetic rat models,which provided a novel approach for prevention and treatment of diabetic neuropathy.

    • The Chemopreventive Effect of Tamoxifen Combined with Celecoxib on DMBA chemically-Induced Breast Cancer

      Abstract:Objective: To investigate the chemopreventive effect of tamoxifen combined with a COX-2 selective inhibitor, celecoxib, on breast cancer in rats chemically induced by 7,12-dimethylben(a)anthracene (DMBA). Methods:DMBA was irrigated into the stomaches of SD female rats to build breast cancer model. A total of 120 rats were divided into four groups: control group, tamoxifen group, celecoxib group and combined group. The incidence rate, latent period, number and volume of breast cancer were detected and analyzed. Results:The tumor incidence rate of tamoxifen group (48.15%, 13/27) and celecoxib group (50.00%, 14/28) were lower than that of control group (85.71%, 24/28), but higher than that of combined group (21.43%, 6/28). The tumor’s latent period of tamoxifen group (97.54±1.85 d) and celecoxib group (96.79±2.89 d) were longer than that of control group (89.50±5.99 d), but shorter than that of combined group (103.67±3.39 d). The average tumor number of tamoxifen group (1.77±0.73) and celecoxib group (1.71±0.61) were less than that of control group (3.50±1.62), but more than that of combined group (1.17±0.42). The average tumor volume of tamoxifen group (1.78±0.71 cm3) and celecoxib group (2.05±1.04 cm3) were smaller than that of control group (6.42±3.96 cm3), but bigger than that of combined group (0.71±0.96 cm3) (P < 0.05 respectively). Conclusion:Celecoxib and tamoxifen are effective drugs in preventing the occurrence of rat breast cancer chemically induced by DMBA. Furthermore, combination of them has better chemopreventive effect.

    • The value of PAPP-A in the diagnosis and prognosis of GTD

      Abstract:Objective: To investigate the value of PAPP-A(pregnancy assouated plasma protein-A) in the diagnosis and prognosis of gestational trophoblastic disease (GTD). Methods: The serum β-HCG(β-human chorionic gonadotropin) and PAPP-A levels of 25 normal pregnant women, 28 patients with complete hydatidiform mole and 38 patients with invasive mole were measured by enzyme linked immunosorbent assay (ELISA) during the periods of diagnosis, treatment and follow-up. Results: Compared with control group, patients with complete mole and invasive mole had higher levels of β-HCG (P < 0.01). But there was no significant difference between the complete and invasive mole group(P > 0.05). The PAPP-A level of complete mole group was significantly higher than that of control group(P < 0.01). The PAPP-A level of invasive mole group was significantly higher than that of complete mole group and control group(P < 0.05). In complete mole group, serum β-HCG and PAPP-A levels of the patients with malignant sequelae were significantly higher than those with benign sequelae (P < 0.05). The β-HCG level had no relationship with the clinical stage of invasive mole. However, the PAPP-A level increased with clinical advancement of invasive mole. The levels of β-HCG and PAPP-A gradually decreased after evacuation in patients with complete moles, but maintained positive or even increased in patients with subsequent malignancy. Conclusion: The PAPP-A level can give us some help not only in early diagnosis of hydatidiform mole and invasive mole, but also in the prognosis of malignant sequelae.

    • Effects of canthaxanthin on H2O2-induced damage of osteoblast

      Abstract:Objective:To investigate the effects of canthaxanthin on the anti-oxidative damage function of osteoblast. Methods: M3T3-E1 osteoblasts were randomly divided into five groups, including control group,model group,canthaxanthin groups[low-dose(1×10-8 mol/L),middle-dose(1 × 10-7 mol/L),high-dose(1 × 10-6 mol/L)]. The activity of cells, superoxide dismutase(SOD), the content of reactive oxygen species(ROS), lipid oxygen(LPO) and membrane fluidity were tested and compared. Results:Compared with canthaxanthin groups,the activity of cells, SOD activity and membrane fluidity in the model group were significantly decreased(P﹤0.01). However, the content of ROS and LPO were significantly increased(P﹤0.01). Conclusion:H2O2 can induce oxidative damage of M3T3-E1 osteoblasts, but canthaxanthin can prevent and decrease its influence.

    • Subjective sleep quality in perimenopausal women and its related factors

      Abstract:Objective: To evaluate the sleep quality and its related factors among perimenopausal women. Methods: A self-designed questionnaire was administered to 506 perimenopausal women. The questionnaire included the influencing factors on the sleep quality, the Pittsburgh Sleep Quality Index(PSQI), the Zung Self-Rating Depression Scales(SDS), the Zung Self-Rating Anxiety (SAS) and the Modified Kupperman Index(KI). Data were analyzed by SPSS11.5. Results: The mean PSQI was 5.97±4.30. Twenty-four percent of perimenopausal women reported poor sleep. Age and perimenopausal symptoms were significantly correlated with sleep quality. The sleep quality of the 45~49 age group was the poorest and the 40~44 age group was the best. The women who had higher Kupperman index were more likely to be poor sleepers. There was no significant correlation between occupation and sleep quality. Night sweat, depression, anxiety, hot flash, stressful life event, and regular exercise were significantly and independently related with sleep quality. Among them, regular exercise was a protective factor of sleep quality. Conclusion: High incidence of poor sleep quality exists among perimenopausal women. Some effective interventions should be taken to improve the sleep quality of perimenopausal women.

      Abstract:Objective:To study the mechanical properties of the plasma- and laser-welded pure titaniums. Methods:Tensile strength was measured with instron testing machine and metallographic change was observed with metallographic microscope. Micro-hardness was also detected. The chemical composition was detected with EDAX. Results:Tensile strength and micro-hardness of plasma-welded titanium had no difference from the control group. The two kinds of joining did not change the chemical composition. Conclusion:Plasma welding can improve mechanical properties and might be suitable for pure titanium welding in restorative dentistry.

      Abstract:Objective: To investigate the inhibitory effect and IC50(50% inhibiting concentration) of the recombinant adenoviral p53 gene (rAdp53) in colorectal cancer cells in vitro and to guide clinical practice. Methods: We evaluated the efficiency(IC50)of the rAdp53 and six kinds of anti-cancer drugs(5-fluorouracil, tegafur, mitomycin c, cisplatin, oxaliplatin, paclitaxel) in human colorectal cancer cell line-174 through the cell culture and MTT chemosensitivity assay to make sure the anti-cancer capability of rAdp53. Expression of p53 protein in transfection cells of colorectal cancer line-174 with rAdp53 was evaluated by immunohistochemical staining. Results: The rAdp53 is a dose- and time-dependent anti-cancer drug, its IC50 is 5.73×1011 VP/ml, but its effect was not obvious when compared with other anti-cancer drugs. In control group, the immunohistochemistry stain was negative. However, rAd-p53 of five different concentrations were all positive in infected colorectal cancer cells with rAd-p53 and the earliest positive result would present 24 hours after infection. Conclusion: The rAdp53 has good anti-cancer efficacy is colorectal cancer cell line-174 in vitro. But its anti-cancer efficacy was less than those of the classical chemical medicine mitomycin c, 5-fluorouracil and cisplatin etc., when it was used alone.

      Abstract:Objective:To explore the effects of splenocytes on graft-versus-host disease(GVHD) and chimerism in murine mixed bone marrow transplantation. Methods:During mixed transplantation, syngeneic bone marrow cells from BALB/c mice were mixed with H-2 haploidentical matched bone marrow cells from CB6F1 mice. At the same time, different infusion dose of splenocytes were transplanted into lethally irradiated groups of BALB/c mice. BALB/c mice of group A hadn’t accepted splenocytes of CB6F1 mice. BALB/c mice of group B were infused with the same dose of syngeneic and CB6F1 mice splenocytes. BALB/c mice of group C accepted the double dose of splenocytes of CB6F1 mice compared with syngeneic splenocytes. After transplantation, the appearance of a GVHD, the level of the chimerism of the recipient mice were observed and detected by histopathological examination on liver, skin, small intestine and flow cytometry. Hematopoietic recovery and the mortality were evaluated by blood routine test and clinical observation. Results:Mice in mixed transplantation groups survived over 50 days. No GVHD manifestation and histopathological alterations appeared in group A, and the chimerism on the 150th day decreased significantly. Mice of group B showed little GVHD manifestation and histopathological changes, and the chimerism on the 150th day was reduced. Mice of group C showed a higher level and more stable of chimerism with grade Ⅱ degree of GVHD histopathological alterations. Typical GVHD manifestation and histopathological alterations appeared in H-2 haploidentical bone marrow transplantation group. The survival rate was only 20% 50 days after transplantation. Conclusion:The proportion and dose of syngeneic and haploidentical MHC splenocytes were of importance to balance GVHD and host-versus-graft reaction(HVGR).

    • Research of combined liver-kidney transplantation model in rats

      Abstract:Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer’s was infused from porta1 veins to donated 1ivers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.

      Abstract:Objective:To establish GFPu-1 clonal cells reflecting the function of the ubiquitin-proteasome system. Methods:GFPu plasmid was transfected into human embryonic kidney(HEK) 293 cells. GFPu-1 cells with stable expression of GFPu were selected by G418. After proteasome inhibitor lactacystin was added into GFPu-1 cells, the distribution and expression of green fluorescent protein(GFP) were studied by confocal microscopy and Western blot. Results:GFPu was distributed diffusely in the nuclear and cytoplasmic compartment of GFPu-1 cells with lactacystin. Compared with the control cells with DSMO, the fluorescence of GFPu significantly increased in GFPu-1 cells with lactacystin. Accummuation of GFPu rose about 2.5 folds(P < 0.05) and β-catenin expression rose about 1.2 fold(P < 0.05). Conclusion:GFPu-1 clonal cells may be a sensitive indicator for the function of the ubiquitin-proteasome system.

      Abstract:Objective:To investigate the expression level of nitric oxide synthases(NOS) and nerve growth factor(NGF) in pterygopalatine ganglion of the allergic rhinitis rats. Methods: The expression level of NOS and NGF was detected in pterygopalatine ganglion of the allergic rhinitis rats using histochemical and immunofluorescence staining methods respectively. Results:The number of labelled NOS-positive neurons and NGF-positive neurons was respectively increased significantly(P < 0.01) in pterygopalatine ganglion of the allergic rhinitis rats. Conclusion:NOS and NGF were probably involved in the pathologic mechanisms of the allergic rhinitis by the nerve regulation.

    • Correlation of MCP-1 expression with angiogenesis in hepatocellular carcinoma

      Abstract:Objective:To investigate the expression of monocyte chemoattractant protein-1(MCP-1) in hepatocellular carcinoma (HCC) and the clinical significance. Methods:Tissue microarray(TMAs) including 36 specimens of non-tumor liver disease and 109 specimens of HCC(containing tumor tissue and adjacent non-cancerous tissue) were prepared. MCP-1, VEGF and CD68 were detected with immunohistochemistry method. Blood capillaries stained for CD34 were counted in HCC, and microvessel density (MVD) was determined. Results:The expression rate of MCP-1 cancer cells, adjacent non-cancerous tissues and non-tumor liver diseases was 40.4%, 67.0% and 44.4% respectively. The expression of MCP-1 was correlated with that of VEGF(r = 0.233,P = 0.015) and MVD(r = 0.234,P = 0.014). MCP-1 expression in TMAs was significantly correlated with the pathological stage(P < 0.05), but not with tumor size, tumor number, blood vessel invasion, differentiation, cirrhosis and prognosis. Conclusion:MCP-1 may play an important role in HCC angiogenesis through inducing the production of angiogenic factors, such as VEGF. But the expression of MCP-1 in HCC can’t be regarded as an indicator of prognosis.

The turnover rate of beds, average admission rate as well as pharmaceutical benefit rate are all lower than the standard set by the Provincial Medical Department. Conclusion: Rigid and flexible management has great affect on the promotion of hospital services and the competitive power. Therefore, it is a worth recommending way of hospital management.

      Abstract:Objective:To establish a plasmid expressive vector coding short hairpin RNA(shRNA) of human cerb-B1 gene in vitro, and to investigate specific inhibition of cerb-B1 gene and epidermal growth factor receptor(EGFR) expression in human colon cancer LoVo cells by shRNA. Methods:DNA templa-primer of cerb-B1 gene was synthetized in vitro and the plasmid expression vectors coding shRNA were established with Pgenesil-1 plasmid. The human colon cancer LoVo cells were transfected by Lipofectamine 2000 with plasmid vector, and then selected for 4 weeks by G418. The expression of cerb-B1 mRNA was assessed by real time PCR and the protein of EGFR was assessed by Western blot method. Results:The digest identification of plasmid vectors confirmed that DNA templa-primer was successfully inserted in the plasmid, and the sequence was in conformity with the designed result. Plasmid expression vectors aiming directly at cerb-B1 gene were successfully established. After the plasmid vectors were transfected into the LoVo cells, the expression of cerb-B1 mRNA was decreased by(81.3 ± 2.8)%, and the expression of EGFR protein was decreased by(73.4 ± 2.3)%, respectively. There was a significant difference to the control plasmid vectors. Conclusion:The plasmid expression vector aiming directly at cerb-B1 gene might suppress cerb-B1 gene and EGFR expression in human colon cancer LoVo cells. The new therapeutic modalities could be used in the treatment for human colon cancer.

      Abstract:Objective: To study the relationships between juvenile internet addiction disorder and parental rearing pattern. Methods: Three hundred and eighteen students from junior high school and universities in the city of Jieyang and Shantou were tested by EMBU and LAD-DQ. Results: The students with internet addiction disorder had a quite different parental rearing pattern from that of the students with non internet addiction disonder in intervention, over-punishment and refusal. Conclusion: Juvenile internet addiction disorder were correlated with the poor rearing pattern of parents.

      Abstract:Objective:To determine the effect of erythropoietin(EP) on the proliferation of mesenchymal stem cells(MSCs) in vitro. Methods:Under serum free culture media, MSCs were incubated with different doses of EP for 24 h(0, 0.25, 0.5, 1, 2, 10 U/ml), and then the proliferation of MSCs was measured by MTT assay. Cell circle stage was measured by flow cytometry assays after incubation of 10 U/ml EP for 72 h. Results:MTT assay showed that EP could enhance MSCs proliferation in a dose-dependent manner. There was significant difference among EP treatment groups and control group,except 0.25 U/ml group. The effects on the proliferation of MSCs was highest in 10 U/ml group. Furthermore,flow cytometry(FCM) assays showed that, compared with control group, EP could significantly decrease G0/G1 ratio, and increase S stage and G2/M stage ratio. Conclusion:EP could enhance MSCs proliferation in dose-dependent manner.

      Abstract:Objective:To investigate the effects of Valsartan on the expression of monocyte chemoattractant protein-1(MCP-1) in the arteria of STZ-induced diabetic rats. Methods:SD rats were randomly divided into NC group,DM group and DV group. Blood glucose(BG), body weight(BW), and HbA1c were observed. the expression of MCP-1 in the arteria of diabetic rats was detected by RT-PCR and Western blot at the 8 th week. Results:The BW of diabetic rats in DM and DV groups decreased significantly compared with NC group(P < 0.01), but no notable difference was found in HbA1c and BG between DM and DV group(P > 0.05). The expression of MCP-1 mRNA in DM and DV groups was much higher than that in NC group(P < 0.01), but it was much lower in DV group than that in DM group(P < 0.01). The expression of MCP-1 protein in DM and DV groups was much higher than that in NC group(P < 0.01), and it was much lower in DV group than that in DM group(P < 0.01). Conclusion:The expression of MCP-1 is much higher in the arteria of diabetic rats. Valsartan could obviously reduce the expression of MCP-1 in the arteria of diabetic rats and ameliorate the inflammation in the arteria.

      Abstract:Objective:To study the relationship between metabolic syndrome(MS) and early-stage diabetic nephropathy(DN) in type 2 diabetes mellitus(T2DM). Methods:478 cases of T2DM were screened for early-stage DN, and some correlated risk factors were analyzed. Results:Patients with MS(64.2%) had a higher prevalence of early-stage DN(36.2% vs. 22.8%, P < 0.01). The proportion of early-stage DN was associated with MS features(16.67%, 26.74%, 36.43%, 45.68%, respectively,P < 0.01). Triglycerides(TG), systolic blood pressure (SBP), glycosylated hemoglobin(HbA1C) and lipoprotein LP(a) were all independent risk factors of early-stage DN(P < 0.05 or P < 0.01). Conclusion:MS is significantly associated with early stage DN in patients with T2DM.

      Abstract:Objective:To explore the effects of astragalus polysaccharide on neurological function deficit and neuron apoptosis and expression of NF-κB(nuclear factor-kappa B) in rats after interacerebral hemorrhage(ICH). Methods:All rats were randomly divided into normal group, sham-operation group, model group and treatment group. ICH model was established by injection of collagenaseⅦ. Sham-operation group was established by saline instead.Treatment group was injected with astragalus polysaccharide(25 mg/kg) through celiac once a day. At 6 hour and on the 1st day, 3rd day and 5th day, the neurological behaviors of rats were observed .By the methods of TUNEL and immunohistochemisty, the dynamic changes of apoptosis and the expression of NF-κB in the tissue surrounding hematoma in were detected ICH rats. Ultrastructures of the cells around the hematoma were observed with transmission electron microscope. Results:Contrast to model group, the score of neurological behavior in treatment group rats was decreased significantly on 3rd and 5th day(P < 0.05), as well as TUNEL positive cells(P < 0.05 and P < 0.01). Significantly difference of NF-κB positive cell between model group and treatment group was found on the first, 3rd and 5th day(P < 0.05 and P < 0.01). The pathological changes of the treatment group were milder than those of the model group 3 days after operation. Conclusion:Apotosis is a pattern of death after intracerebral hemorrhage,and the expression of NF-κB is an important step in apoptotic cell death after ICH. Astragalus polysaccharide can effectively alleviate apoptosis after ICH. Preventing NF-κB expression may be involved in the treatment.

      Abstract:Objective:To investigate the role of extracellular signal-regulated kinases1/2(ERK1/2) in inschemia-reperfusion injuty of spinal cord. Methods:The morphology of spinal cord by hematoxylin and eosin(HE) stain and electron microscope in different ischemia/reperfusion time were observed. The activation of ERK1/2(pERK1/2) was detected by western blot and the localization of pERK1/2 was analyzed by immunohistochemistry. Results:ERK1/2 was activated in spinal cord occluded for 30 min and reperfused for 2 to 24 h. No pERK1/2 was found in neurocyte in the control group by immunohistochemistry. PERK1/2 was stained both in the cytoplasm and nucleus after 30 min of occlusion and 2 h of reperfusion, and stained further after 8 to 24 h of reperfasion. Conclusion:ERK1/2 was activated in the reperfusion of ischemic spinal cord and the stagnation of pERK1/2 in the cytoplasm during reperfusion of ischemic spinal cord may lead to neurocyte apoptosis.

    • Comparison radiofrequency with mechanical gouging in the treatment of knee rheumatoid arthritis

      Abstract:Objective:To analyse the differences between radiofrequency and mechanical gouging in treatment of knee rheumatoid arthritis. Methods:Randomized control and prospective case study was designed, with 21 cases in the radiofrequency treatment group and 17 cases in the mechanical gouging group(control group). All the cases were followed up for an average of 19.5 months(ranging 6 to 39 months) according to Lysholm knee function evaluation score. Results:The Lysholm scores in the treatment group were superior to those in the control group in cases at stage Ⅰand Ⅱ according to ARA staging system(P < 0.05), while there were no statistically significant differences between the two groups in cases at stage Ⅲ(P > 0.05). Conclusion:Radiofrequency is the first choice in treatment of knee rheumatoid arthritis at stage Ⅰand Ⅱ.

      Abstract:Objective:To develop a method for the purification and identification of human Pregnancy-associated plasma protein A. Methods:Pregnancy-associated Plasma Protein A fraction was isolated and purified by DEAE-Sepharose CL-6B ion-exchange and heparin-Sepharose Cl-6B affinity chromatography, denatured SDS-PAGE and Western blot method respectively. Results:The expressed protein, with a relative molecular weight of 200 kD, showed specific reaction with McAb to pregnancy-associated plasma protein A. Conclusion:Pregnancy-associated plasma protein A could be successfully isolated from pregnancy serum.

      Abstract:Objective:To investigate the physiologic characteristics of coagulation parameters during normal pregnancy and puerperium.Methods:One hundred and sixty normal pregnant women were involved in a serial random study to measure prothrombin time(PT),activated partial thromboplastin time(APTT), thrombin time(TT), fibrinogen(Fbg). Blood samples were collected in 10 th-14 th, 20 th-24 th and 30 th-34 th gestational weeks and on the third day of puerperium respectively, The same measurements were also performed on 50 non-pregnant women. Results:PT,APTT and TT were shortened in pregnant women compared with non-pregnant controls. Fbg concentration was always increased in pregrant women versus non-pegnant controls in an increaseng trend with the increase of gestational time. The most significant variations for APTT were recorded up to the 20 th week. From the 20 th week onwards, APTT level remained stable until after the delivery. The level of PT in the mid stage was lower than that in other stages, and there was statistical differences between the perinatal periods. No statistical difference in TT between the different gestational weeks were observed, but the puerperium was significant longer than the gestational period. Fbg level was increased with the increase of gestational time. Conclusion:PT, APTT, TT, Fbg show great variation during pregnancy, and they appear to play a major role in finding the abnormal changes of the pregnant women early.

Methods:The study was conducted with 210 cases of rectal cancer and 439 cases of normal controls. The epidemiological data were collected, and the genotypes of the GSTM1 and GSTT1 were detected by multiplex PCR. Polymorphism of GSTP1(A→G) was detected using PCR-based restriction fragment length polymorphisms(PCR-RFL P). Results:No significant difference was found in the frequencies of the deletion genotypes among the patients and controls No significant difference was found in the frequencies of GSTP1 A/A, A/G and G/G genotypes among the patients and controls. Creased risk of rectal cancer was not significanthy associated with GSTP1 G/G genotypes(adjusted for age, sex and status of the smoking and alcohol drinking(OR = 1.11, 95% CI:0.77-1.60) compared with those with GSTP1 A/A genotypes. Conclusion:The polymorphisms of GSTM1,GSTT1 and GSTP1 may not be associated with the susceptibility of rectal cancer.
