Abstract:Subepidermal autoimmune bullous diseases (SABD) are some autoimmune skin diseases that can present in a variety of forms and can be a challenging disease to treat. An overview of the different forms of SABD are discussed including bullous pemphigoid (BP), epidermolysis bullosa acquisita (EBA), cicatricial pemphigoid (CP), bullous systemic lupus erythematosus (BSLE), and Anti-p200 pemphigoid. Emphasis on recent advancement is presented. In recent years, improved knowledge of the mechanisms of intercellular and cell-matrix adhesion has led to better understanding of the blistering process in some SABD. Defects of such structures cause the subepidermal bullous diseases and have also led to the discovery of new diseases (e.g. anti-p200-pemphigoid). Recent studies have outlined the important role of autoantibodies, mast cell lymphocytes and their cytokines in pathogenesis of SABD.

Abstract:Objective: To construct an eukaryotic expression vector containing Smac gene and study the expression efficiency and specificity of prostate specific antigen(PSA) enhancer/promoter in a possible targeted gene therapy scheme for prostate cancer. Methods: PSA enhancer(PSAE) and promoter(PSAP) sequences were amplified using PCR method. CMV and T7 promoters were deleted from pcDNA3.1-Smac and replaced by the two specific fragments to generate pPSAE-PSAP-Smac. After transfection into different cell lines, the status of cells was observed. And then, we determined the relative concentration of Smac mRNA in RT-PCR. Results: The recombinant plasmid of pPSAE-PSAP-Smac was successfully constructed. And only the prostate cancer cell line PC-3 was suppressed after transfection with pPSAE-PSAP-Smac. However, other nonprostate lines were not. Moreover, the concentration of Smac mRNA regulated by PSA promoter and enhancer was higher in comparison to the CMV promoter-driven control vectors. Conclusion: An expression vector containing the Smac gene (based on elements of the PSA gene regulatory sequences) has been developed and shown to function in prostate cancer cell lines which provides a solid platform for launching clinical studies.

Abstract:Objective: To investigate the incidence and relative risk factors of post coronary artery bypass grafting(post-CABG) atrial fibrillation(AF). Methods: 312 patients with CABG were reviewed and divided into an AF group and a non-AF group. Statistical analysis was used to compare the data between the two groups and screen for risk factors of post-CABG AF. Results: 103/312 (33.01%) patients developed post-CABG AF. Univariate analysis showed that patients in AF group compared with those in non-AF group were more likely to have advanced age (≥70 years), early postoperative withdrawal of β-blockers, hypertension, left atrial enlargement (≥40 mm), a history of AF, prolonged p-wave duration (≥120 ms) and increased number of grafts (≥3). Multivariate logistic regression analysis showed that advanced age (≥70 years), early postoperative withdrawal of β-blockers, hypertension, left atrial enlargement (≥40 mm) and a history of AF were highly related to post-CABG AF. Conclusion: The incidence of AF in patients following CABG was 33.01% in this study. Advanced age, early postoperative withdrawal of β-blockers, hypertension, left atrial enlargement and a history of AF were independent risk factors of post-CABG AF.

Abstract:Objective: To assess the acute and mid-term results of cardiac function improvements and left ventricular outflow tract gradient (LVOTG)changes in 30 patients displaying hypertrophic obstructive cardiomyopathy (HOCM) treated with percutaneous transluminal septal myocardial ablation(PTSMA). Methods: PTSMA was intended for 32 patients comprising of 13 women and 19 men(average years being 54.1 ± 15.5) to be treated in accordance with the following inclusion criteria: The New York Heart Association(NYHA) definition for cardiac functional class III or IV ,or class II but for whom medical therapies were not tolerated or with syncope; intraventricular septal(IVS) and left ventricular posterior wall(LVPW) hypertrophy asymmetrically associated with ratio of IVS to LVPW≥1.3 and LVOTG≥50 mm Hg at rest or ≥100 mm Hg at provocation(Valsalva maneuver). The target vessels were determined by coronary arteriography that demonstrated more than one septal branch and probatory balloon occlusion produced greater than 50% decrease of LVOTG . Once the target vessel established, the alcohol was administrated into septal ventricular via over-the-wire balloon. LVOTG was assessed by means of echocardiography measurements immediately after procedure and 3 months. Simultaneously, cardiac function class was also evaluated. Results: Two patients were abandoned prior to intervention due to inappropriate septal target vessels and DDD Pacemakers were chosed. Immediately after the procedure, resting LVOTG was reduced from 73.8 ± 35.5 to 16.6 ± 7.8 mmHg, at provocation LVOTG from 149.3 ± 42.5 to 61.9 ± 43.0 mmHg(P < 0.0001 each) by echocardiography measurements. After 3 months, the mean New York Heart Association class was reduced from 2.8 ± 0.6 to 1.1 ± 1.0(P < 0.0001) and the LVOTG also remained decrease(28.5 ± 6.4 mmHg at rest and 75.3 ± 11.6 mmHg at provocation). Conclusion: PTSMA is a promising nonsurgical technique for relief of symptoms and reduction of LVOTG in hypertrophic obstructive cardiomyopathy.

Abstract:Objective: To investigate the relationship of matrix metalloproteinase-9 polymorphism to acute coronary syndrome and its affect on the severity of coronary artery disease. Methods: By means of polymerase chain reaction(PCR) and restriction fragment length polymorphism, genotypes of 245 patients with acute coronary syndrome(ACS) and 205 healthy subjects were tested. Genotypes displaying C-1562T functional promoter polymorphism(of the MMP-9 gene) were determined. The relationship between the polymorphism of the MMP-9 gene and ACS and the severity of coronary vessels diseased was analyzed. Results: The frequency of C/T plus T/T genotypes and T allele in patients with ACS was significantly higher than that in healthy subjects(22.1% vs 12.7% and 11.4% vs 6.6% respectively). But they were not associated with the number of coronary arteries diseased. Conclusion: The MMP-9 polymorphism may be susceptible to ACS. But there was not significant difference between the AMI and UAP subgroups.

Abstract:Objective: To evaluate the effect of a new-type sutureless magnetic bilioenteric anastomosis stent that was used to reconstruct the bilioenteric continuity(primarily under the circumstances of severe inflammation after acute bile duct injury in dogs). Methods: Establishing an animal model of acute bile duct injury with severe inflammation and bile peritonitis in dogs. The new-type sutureless magnetic bilioenteric anastomosis stent was used to reconstruct the bilioenteric continuity primarily. Results: The experiment group anastomosis healed well with a mild local inflammation reaction, and the collagen lined up in order without the occurrence of observable bile leakage and infection. Conclusion: It was safe and feasible to use the new-type anastomosis stent to reconstruct the bilioenteric continuity primarily under the circumstances of severe inflammation after acute bile duct injury in dogs.

Abstract:Objective: To observe the expression of TGF-β and TNF-α in the spinal cord injured rat model and discuss the significance of the articular cartilage metabolism. Methods: 36 SD female rats were randomly divided into 2 groups: Rats models of spinal cord injury were implemented by Allen method. T10 laminectomy was performed in the control group. Both groups of rats were killed respectively in 1w, 3w and 6w. Hematoxylin-eosin stain was given to each slice in the model group and control group. Immunohistochemical stain was applied by using ABC method in the expression of TGF-β and TNF-α. Those expressed level were performed in image analysis and statistics process. Results: TGF-β and TNF-α were mainly distributed on the surface layer of the articular cartilage, with a weak expression in control group. The expression of TNF-α in the model group was more significant than that in the control group in the 1w, and still remained an evident difference with that in control group until the 6w(P < 0.05). TGF-β expression of the model group had no remarkable difference with the control group in the 1w(P > 0.05) and prominently became stronger at 6w(P < 0.05). Conclusion: The expression of TNF-α occurred early in the development of spinal cord injury, and the expression of TGF-β became stronger with the revival of spinal neural function. Both expressions were strengthened in articular cartilage in the 3rd week.

Abstract:Objective: To explore the role of matrix metalloproteinase-1,2(MMP-1, MMP-2) and tissue inhibitor of matrix metalloproteinases-1 (TIMP-1) in endometriosis. Methods: The eutopic and ectopic endometria from 40 subjects suffering from endometriosis and regular endometria from 40 subjects(excluding endometriosis) were collected and examined by in situ hybridization technology and western blot assay. Results: Both expressions of MMP-1 and -2 were stronger in ectopic endometrium and eutopic endometrium than in normal endometrium. On the contrary, the expression of TIMP-1 in ectopic endometrium and eutopic endometrium was lower. The differences were significant(P < 0.01). Moreover, there was no relationship among the expressions of MMP-1, 2 and TIMP-1 in ectopic endometrium. Conclusion: The expressions of MMP-1, 2 and TIMP-1 lose balance and lack of periodic changes in ectopic endometrium , which explains the biological invasive behavior of endometriosis. It was suggested that regulating the balance between the MMPs and TIMP-1 should be an ideal therapeutic target to endometriosis.

Abstract:Objective: To study the significance of Leptin and the activity of erythrocyte membrane Ca2+-ATPase(EMCA) in the development of hypertensive disorder complicating pregnancy. Methods: Radioimmunoassay was used to test the level of serum Leptin,and the activity of EMCA was determined chemically in 38 pregnant women with hypertensive disorder complicating pregnancy and 36 normotensive pregnant women. Results: The level of serum Leptin in hypertensive disorder complicating pregnancy(gestational hypertension: 13.76 ± 3.46 ng/ml; preeclampsia:15.76 ± 5.47 ng/ml; eclampsia: 18.32 ± 6.38 ng/ml)was significantly higher than that in normotensive pregnant women(11.33 ± 2.93 ng/ml),respectively. The average EMCA activity of patients with hypertensive disorder complicating pregnancy(gestational hypertension:1.65 ± 0.24 μmol·pi/mg·h ; preeclampsia:1.37 ± 0.19 μ mol·pi/mg·h; eclampsia:1.12 ± 0.14 μ mol·pi/mg·h) was significantly lower than that of normotensive pregnant women(1.83 ± 0.38 μ mol·pi/mg·h),respectively. There was a negative correlation between the level of serum Leptin and the activity of RMCA in hypertensive disorder complicating pregnancy(r = -0.63). Conclusion: Inhibition of EMCA activity of erythrocyte in hypertensive disorder complicating pregnancy may increase cytoplasmic free calcium, which contributes to the development of hypertensive disorder complicating pregnancy. The negative correlation between the level of serum Leptin and the activity of EMCA, also suggested that serum Leptin and the activity of EMCA may play a role in the development of hypertensive disorder complicating pregnancy.

Abstract:Objective: To explore the role of interleukin 4(IL-4), expressions of cyclooxygenase-2 (COX-2) and platelet-derived growth factor(PDGF) in the model of chronic obstructive pulmonary disease(COPD), in the lungs of rats. Methods: Male Wistar rats were used to build up the model of COPD. Rats were randomly divided into the control group and model group, the IL-4 group and the dexamethasone group. The expressions of COX-2, PDGF-A and PDGF-B in the lung tissue were detected by western blotting and RT-PCR. Results: The expressions of COX-2, PDGF-A and PDGF-B in the model group were increased significantly. Those expressions in the IL-4 and dexamethasone group were notably decreased. Conclusion: IL-4 and dexamethasone could interfere in the establishment of COPD. The expressions of COX-2 and PDGF in the lung tissue of COPD were increased significantly and IL-4 and dexamethasone could decrease those expressions.

Abstract:Objective: To investigate the effects of connective tissue growth factor(CTGF) and collagen type I(COL-I) on the pathogenesis of scleroderma and explore the relationship between the level of COL-I and CTGF. Methods: 12 mice model of scleroderma was established by the injection of Bleomycin. The level of CTGF and COL-I were detected by immunohistochemical method. The relationship was analyzed between CTGF and COL-I level. As control group, 12 healthy mice were selected. Results: The levels of CTGF and COL-I in sclerotic models were higher than in normal controls(P < 0.05). It was found that there was a correlation between the level of CTGF and COL-I. Conclusion: CTGF and COL-I played an important role in the hardening process of the skin lesions of the mice model, which may be involved in the pathogenesis of scleroderma.

Abstract:Objective: To study the relationship between drug resistant genetic mutation and drug resistance in Mycobacterium tuberculosis L-form, discuss the internal relationship between drug resistances and drug-resistant related genes and explore the value of PCR-SSCP to clinical application. Methods: A total of 52 clinically isolated strains of tuberculosis L-form were collected among 97 pneumoconiosis patients complicated with tuberculosis. The gene mutations of katG, rpoB and rpsL were detected by PCR-SSCP, and the results were compared with those analyzed by traditional antimicrobial susceptibility test(AST). Results: The gene mutation rates of katG, rpoB and rpsL by PCR-SSCP were respectively 57.70%(30/52), 65.38%(32/52) and 40.38%(21/52). The rate of reversion was 78.85%(41/52) and the result of drug-resistant genes was invariable. The results of AST showed that there were 40(76.92%) multi-drug resistant strains in 52 clinically isolated strains. The number for three-drug resistant strain was 21(40.38%) and that of two-drug resistant was 19(36.54%), but only 12(23.08%) strains were one drug resistant. The rate of total drug-resistance was 100%, but there were 15 strains of allied mutation of three genes, 16 of two mutations and 6 of only one by PCR-SSCP. The coincidences were respectively 71.43%, 84.12% and 50.00%. Then there was no significant difference between the allied mutations of multi-drug resistant gene and the mutations of only one drug resistant gene(P > 0.05). Conclusion: PCR-SSCP technique has a higher sensibility and specificity to detect the genes of katG, rpoB and rpsL in tuberculosis L-form among pneumoconiosis complicated with tuberculosis,and the detecting rate of two drug resistant strains and three drug resistant strains was higher. The combined application of PCR-SSCP and AST has advantages at earlier diagnosis and guidance of clinical medications.

Abstract:Objective: To understand the reactivity of purified protein derivative skin test(PPD test) in HIV-infected persons and to determine the influential factors associated with PPD. Methods: 174 HIV/AIDS patients registered in the local center for disease control and prevention(CDC) participated this study from April to June in 2006. Questionnaire,CD4 count and thoracic roentgenogram were performed for all participants. Results: In this study, response rate of questionnaires was 83.65%. The majority of these participants had a different degree of immunodeficiency that accounted for 93.64%. Female patients had a higher CD4 count than that of males. The total positive rate of PPD was 38.15%. Analysis of single factor in our study indicated that CD4 count, previous tuberculosis history, tuberculosis contact history and thoracic roentgenogram manifestation of patients were related to their PPD diameters. Further analysis of multiple factors also supports the previous conclusion that CD4 count and previous tuberculosis history of patients were risk factors in the PPD test. Conclusion: The PPD test of HIV/AIDS patients could be affected by several factors. For persons infected with HIV, the confirmation of latent tuberculosis infection (LTBI) should be considered the combination effect of previous MTB infection and body cellular immune function.

Abstract:Suppose a case like this:once a patient get a serious disease,which is difficult to be cured or even can not be cured,then he or she go to a hospital to seek help,but suffered with the omission of the doctor. Most patients can not get compensation, for the reason that the patient can not prove the causation between the omission and the damage. Lots of researchers know that is unreasonable, then loss of chance doctrine is developed. Under this doctrine, one theory propose to change the traditional “damage”, and recognizes the lost chance of patients as the damage in the tort law, besides, it complies with the “but for” approach and the balance of probability. In the judicial practice, the doctrine was modified. There are two theories, one is substantial probability that lower the standard of the balance of probability, the other is proportional approach. This article analyzed loss of chance doctrine. It should provide the model and the reference when our country judicial processed similar medical service disputes.

Abstract:Objective: To discuss the clinical and imaging diagnostic rules of peripheral lung cancer by data mining technique, and to explore new ideas in the diagnosis of peripheral lung cancer, and to obtain early-stage technology and knowledge support of computer-aided detecting(CAD). Methods: 58 cases of peripheral lung cancer confirmed by clinical pathology were collected. The data were imported into the database after the standardization of the clinical and CT findings attributes were identified. The data was studied comparatively based on Association Rules (AR) of the knowledge discovery process and the Rough Set (RS) reduction algorithm and Genetic Algorithm(GA) of the generic data analysis tool (ROSETTA), respectively. Results: The genetic classification algorithm of ROSETTA generates 5 000 or so diagnosis rules. The RS reduction algorithm of Johnson`s Algorithm generates 51 diagnosis rules and the AR algorithm generates 123 diagnosis rules. Three data mining methods basically consider gender, age, cough, location, lobulation sign, shape, ground-glass density attributes as the main basis for the diagnosis of peripheral lung cancer. Conclusion: These diagnosis rules for peripheral lung cancer with three data mining technology is same as clinical diagnostic rules, and these rules also can be used to build the knowledge base of expert system. This study demonstrated the potential values of data mining technology in clinical imaging diagnosis and differential diagnosis.

Abstract:The traditional product responsibility system could return and repair the goods or compensate loss only when the products do not conform to national or professional standard, or create the actual harm. It is already insufficient to protect the consumer. From some kind of degree, the appearance of the system of the defective product recalls is the expansion of the traditional product responsibility system. So long as the products exist dangers on people or property, they should be recalled. It is more advantageous in protecting consumer’s legitimate rights and interests. This article attempted to explain the basic connotation of the defective product recalls system, taking the law of contract and the right infringement law as the rationale, from the implementation main body and the recalls procedure to make the preliminary construction to the defective product recalls system.

Abstract:Objective: To present a case of electrical storm (ES) in a female patient with rheumatic valve disease. Methods: A female patient with severe rheumatic valve disease suffered an unexpected ES. She received more than 50 electrical shocks for repeated cardiac arrests due to ES over 16 hours. Then she received beta-blocking agent treatment and had an operation of double valves replacement. Results: ES was suppressed by sympathetic blockade with beta-receptor blocker and finally disappeared after the double pathological valves had been replaced. Conclusion: Increased sympathetic activity plays an important role in the genesis of electrical storm and sympathetic blockade may effectively suppress ES. However, the most important thing in the treatment of ES is to identify and eliminate the underlying cause of ES.

Abstract:Objective:To understand the knowing rate of current farmer’s healthy knowledge, the rate of behavior formation and the healthy demand of Chongming County. To explore health promotion strategy which suit the countryside. Methods:Using the pure random sampling method, carries on the residence questionnaire survey to 500 farmers. Results:The knowing rate of Chongming farmers that prevent and control the common disease was high. The grasping of health life style was also good. But the knowing rates of chronic illness and new disease were low. The farmes thought the most important factor which affects health was“income condition”. Most of them gained the related knowledge from books and periodicals, magazine, television, network or other media. Abstract:Objective:To construct a plasmid pET32a-EC2A for the expression of recombinant extracellular gene of LMP2A of Epstein-Barrin virus(EBV) in escherichia coli(E.coli) and utilize EC2A protein in serological detection for patients with nasopharyngeal carcinoma(NPC). Methods:The plasmid pEC2A containing the recombinant extracellular gene of LMP2A of EBV was digested with BamHⅠ and EcoRⅠ and cloned into the expression vector pET32a. The constructed vector which had been identified by PCR, enzyme digestion and nucleotide sequences analysis was transformed into BL21. After induced with IPTG, the recombinant protein was purified with Ni+ affinity chromatography, at the same time, its antigenic activity was identified by Western blot. The purified protein was used in serological detection for patients with NPC which is associated with EBV. Results:The recombinant plasmid was constructed correctly. SDS-PAGE and Western blot analysis showed that the recombinant protein was about 40 ku. The purified protein could be used as an antigen to detect specific antibodies in the sera of patients with NPC. Conclusion:The recombinant extracellular gene of LMP2A of EBV could be expressed with high performance and the purified protein may be used in serological detection of antibodies for patients with NPC.

Abstract:Objective:To investigate the effect of prostaglandin E2 on the proliferation of human cholangicarcinoma cell and the possible mechanism. Methods:Human cholangicarcinoma cell line(HUCCT1) was cultured and treated with PGE2, and the cell growth rate and viability was measured by cell proliferation assay with WST-1 regent. The intracellular free calcium concentration was measured using laser scanning confocal microscopy with fluo-3/AM as fluorescence probe. The level of cAMP was measured by immunoassay KIT. Results:PGE2 promoted the growth of cholangicarcinoma cell, and increased intracellular free calcium and cAMP in cholangicarcinoma cell. Conclusion:The results that PGE2 could increase the concentration of intracellular free calcium and cAMP suggest that PGE2 may play a role in promoting the cell growth.

Abstract:Objective:To observe the effect of human recombinant hepatocyte growth factor(HGF) on the expression of vascular endothelial growth factor(VEGF) in colorectal carcinoma(CRC) cells in vitro. Methods:Human CRC cell lines SW480, LOVO were cultured with different concentrations of HGF. The culture supernatants were collected to detect the production of VEGF by ELISA assay. The effect of HGF on phosphorylation of its receptor c-Met was detected by Western blot assay. The expression of VEGF mRNA stimulated by HGF was investigated by reverse transcription PCR(RT-PCR). Results:HGF increased the expression of VEGF (mRNA 4-5-fold and protein 3-10-fold) in CRC cell lines; HGF induced the phosphorylation of its receptor c-Met in a dose-dependent manner. Conclusion:In vitro, HGF can up-regulate the expression of VEGF in CRC cells, which correlates with the phosphorylation of its c-Met induced by HGF. Inhibitor of phosphorylation-c-Met, HA, can inhibit the expression of VEGF in CRC cells induced by HGF.

Abstract:Objective:To investigate the effects of proteasome inhibitor PS-341(Bortezomib) on the expression of interleukin-6(IL-6),interleukin-1β(IL-1β) and stem cell factor(SCF) and the apoptosis of U266 cells. Methods:U266 Cells were incubated with RPMI1640 and exposed to 0,50,100,150,200 nmol/L of PS-341 for 4 h, respectively. Then the apoptosis of cells was analyzed by fluorescence microscope and flow cytometry(FCM) with Annexin V-FITC/PI double staining, and the expression of IL-6,IL-1β and SCF was detected by RT-PCR. Results:Fluorescence microscope and FCM showed that PS-341 induced U266 cells apoptosis in a dose-dependent manner; The expression of IL-6, IL-1β and SCF was decreased significantly after PS-341 treatment. The inhibitory effect on IL-6 and SCF was positively related with drug concentration. The expression of IL-1β was decreased gradually when the drug concentration was increased from 0 nmol/L to 150 nmol/L, whereas the expression level of IL-1β remained unchangeable when the drug concentration exceeded 150 nmol/L. Conclusion:Proteasome inhibitor PS-341 could significantly downregulate the expressions of IL-6,IL-1β and SCF in U266 cells, which might be one of the mechanisms for PS-341 for treatment of multiple myeloma.

Abstract:Objective:To determine the effect of selenium on apoptosis and the expression of bcl-2,p53 induced by cadmium in LLC-PK1 cells. Methods:MTT assay was used to observe the effects of selenium on growth inhibition induced by cadmium in LLC-PK1 cells. The effect of selenium was observed in cadmium-induced apoptosis with the fluoromi croscope dyed by acridine orange/ethidium bromide(AO/EB), and the levels of p53 and bcl-2 proteins were detected by Western blot. Results:Selenium inhibited cadmium-induced LLC-PK1 cells growth in a dose-dependent manner, and the selenium pretreatment partially inhibited the cadmium-induced apoptosis. On exposure to 40 μmol/L cadmium for 12 h, the expression levels of p53 proteins were upregulated. At the same time, the level of bcl-2 protein was downregulated in LLC-PK1 cells. The selenium pretreatment inhibited the cadmiumd-induced p53 proteins upregulation, however, did not affect the bcl-2 expression levels. Conclusion:The results indicate that the protective effect of selenium may be related to the inhibition the p53 protein upregulation in cadmium-induced apoptosis.

Abstract:Objective:To investigate the effects of 4’-methery-scutellarein(4-MS) on proliferation and intracellular calcium concentration in human choriocarcinoma JAR cell line. Methods:MTT assay, flow cytometry and laser scanning confocal microscopy were used to detect the effects of 4-MS on JAR cells. Results:4-MS inhibited the growth of JAR cells. Annexin Ⅴ-FITC +/PI- cells and intracellular calcium concentration were gradually increased with the increase of the dose and action time of 4-MS. There were significant differences compared with the control group. Conclusion:4-MS can inhibit the proliferation and induce apoptosis of human choriocarcinoma JAR cell line. Its mechanism is related to the increase of intracellular calcium concentration.

Abstract:Objective:To compare the effects of antenatal ambroxol and dexamethason on rat fetal lung morphogenesis. Methods:Nine pregnant rats were divided into three groups randomly:ambroxol group,dexamethasone group and control group with 3 rats in each group. On the 19th day on gestation, cesareans were taken out and the histologic structures of 6 fetal rat lungs of each pregnant rat were observed with light microscope,electronic microscope and image analysis. Results:Under the light microscope, compared with control group,fetal rat lung in the two treatment groups had more alveolar numbers, larger alveolar space, and thinner alveolar septum(P < 0.01). Under the light microscope, compared with ambroxol group,fetal rat lung in the dexamethasone group had more alveolar numbers, larger alveolar space, and thinner alveolar septum(P < 0.01). Under the electronic microscope, lamellar body and cellular organs such as bioblast in endochylema of the two treatment groups were more common compared with the control group. And lamellar body in the two treatment groups was more anachromasis and condensed compared with the control group. Conclusion:Both ambroxol and dexamethasone could promote the development of fetal rat lung, and dexamethasone had prior effects to ambroxol. However, considering of much side effects of dexamethasone, it is suggested that ambroxol could be used as a better drug to promote fetal lung maturity.

Abstract:Objective:To investigate the effect of inhaled ketamine on airway hyperresponsiveness in allergic asthma rats. Methods:Forty male Brown Norway rats were randomized into five groups:control group C,asthma groups A, ketamine pretreatment group K1,K2 and K3. Airway responsiveness were assessed using a whole body plethysm graphy RT-PCR was used to dectect the mRNA expression of NF--资B. NF--资B levels in bronchioles of all groups were measured by immunohistochemistry. Results:The increase of resistence of exhalation(Re) in group K1,K2 and K3 was significantly lower than that in group A with a dose of 50 -滋g/kg,100 -滋g/kg,200 -滋g/kg of acetylcholine(ACH)(P < 0.01). The decrease of Cldyn in group K1,K2 and K3 was significantly compared with group A with a dose of 50 -滋g/kg,100 -滋g/kg,200 -滋g/kg(P < 0.01) of ACH. NF-кB P65 mRNA expression and NF-кB levels in asthma group was significantly higher than that of control group(P < 0.05), and that of K1,K2,K3 group significantly lower than that of asthma group(P < 0.05). Conclusion:Ketamine aerosol could effectively alleviate the airway hyperresponsiveness in allergic asthma rat.

Abstract:Face to the new history mission of constructing the innovative high school and creative nation, Chinese higher education confronts a new rigorous challenge. Abstract:Objective:To investigate the effects of marrow-derived MSCs on the lung damage in new born rats suffering from hyperoxia. Methods:Marrow-derived MSCs were separated, cultured, amplificated and labeled with BrdU. Thirty-two three-day-old SD rats were randomly divided into four groups(A,B,C,and D). Rats in group B and group A were exposed to 95% oxygen for 7 days, then they were injected with MSCs of 5 × 104 and PBS into tail vein respectively. Rats in group C and group D lived in normal air and were injected with MSCs of 5 × 104 and PBS into tail vein respectively at the same time. Immunohistochemistry(IHC) staining was used to determine the expression of 5-bromo-2-deoxyuridine(BrdU) and vascular endothelial cell growth factor(VEGF) 72 h later. Radical alveolar counts(RAC) of the lungs in the rats were counted under light microscope. Results:The labeled MSCs were detected in the lungs of rats treated with MSCs, but no BrdU-positive cells were found in two untreated groups(A and D). There were significant differences in the levels of VEGF and RAC among four groups(P < 0.05, P < 0.01 respectively). Conclusion:BrdU-labeled marrow-derived MSCs can be implanted into lungs of new born rats suffering from hyperoxia. The protective effect of MSCS on the damaged lungs induced by hyperoxiab might be related to the increased expression of VEGF.

Abstract:Objective:To optimize the isolation and purification of rat islets. Methods:Pancreas of SD rats were digested by collagenase V and purified by Histopaque-1077. The islets were stained with trypan blue to identify the viability, and stained with dithezone to count the yield. Glucose stimulated insulin secretion(GSIS) of islets was performed in vitro. Allogeneic islets transplantation via portal vein was performed in STZ-induced diabetic SD rats and the blood glucose level was observed. Results:The average yield was 646 ± 67 perrat and the livability rate was above 90%. GSIS showed that SI(stimulating index) was 2.6, 1.7 and 1.4 at the 1 st, 3 rd, 5 th respectively. There was significant difference between low glucose groups and high glucose groups(P < 0.05). After transplantation, the blood glucose decreased to 11.1 mmol/L at the 2nd day and kept below 15 mmol/L for 8 days. Conclusion:Collagenase V digestion combined with Histopaque-1077 purification is an effective method for rat islets isolation, and the islet yield is high with good function.

Abstract:Objective:To investigate the effects of troglitazone on growth of thyroid papillary carcinoma cell line IHH-4 and the mechanisms possibly involved. Methods:Papillary thyroid carcinoma cell line IHH-4 was incubated with different concentrations of troglitazone and observed at different times, and the cell viabilities were measured through dimethylthiazol-diphenyltetrazolium bromide(MTT) assay. The cell cycles of the stimulated cells were analyzed by flow cytometer, and the expression of p27 mRNA was evaluated by semi-quantitative RT-PCR. Results:Troglitazone inhibited the proliferation of thyroid carcinoma cells in a dose-dependent and time-dependent manner(P < 0.05). After treated by troglitazone,the proportion of cells in GO/G1 phase increased significantly(P < 0.05), and p27 mRNA expression was up-regulated. Conclusion:Troglitazone inhibits the growth of papillary thyroid carcinoma cells by arresting the G1/S phase transition, so troglitazone could be a choice for the treatment of the aggressive thyroid carcinoma.

Abstract:Objective:To explore the relationship between the expression of glucosylceramide synthase(GCS) gene and multidrug resistance MDR in human leukemia. Methods:Using β-actin gene as internal reference, the level of GCS, mdr1, BCL-2 and BAX gene in 36 clinical multidrug resistance/non-resistance blood samples with acute leukemia were detected by RT-PCR technique. Results:The relative quantities of GCS and mdr1 genes in multidrug resistance group were significantly increased compared with non-resistance group(P < 0.001). The expression of BCL-2 gene in multidrug resistance samples was also significantly higher than that in non-resistance group(P < 0.001), but the expression of BAX gene was significantly lower in multidrug resistance group(P < 0.01). There was a significant difference in BCL-2/BAX ratio between multidrug resistance and non-resistance groups. Conclusion:The high expression of GCS gene is possibly correlated with multidrug resistance in acute leukymia, and cell apoptosis might be one of the main molecular pathology mechanisms of multidrug resistance caused by GCS gene.

Abstract:Objective:To compare the effectiveness, security and survivability of three insulin treatment plans for patients with type 2 diabetes mellitus(T2DM) after operation. Methods:The patients with T2DM receiving surgical intervention and having no or irregular food after operation were enrolled in this study. They were divided into three groups:continuous subcutaneous insulin infusion(CSⅡ) group, long-acting insulin(Lantus) group and continuous intravenous insulin infusion(CVⅡ) group. The fast blood glucose(FBG), postprandial blood glucose(PBG), the control level of blood glucose and hypoglycemia occurrence after treatment were detected. Results:The level of FBG and PBG on the 3rd, 5th, 7th and 10th day after treatment were obviously decreased in both CSⅡ group and Lantus group(P < 0.01). The level of FBG on the 3rd, 5th, 7th and 10th day after treatment were obviously decreased in CVⅡ group(P < 0.01), but the level of PBG was only decreased on the 7 th day(P < 0.05); On the 10 th day, the level of FBG in CSⅡ group was controlled acceptably, and the level PBG was controlled well. The level of FBG and PBG in Lantus group were all controlled acceptably, but they were controlled terribly in CVⅡ group; The compliance of patients was common in CSⅡ group, good in Lantus group and bad in CVⅡ group. There was no hypoglycemia case in CSⅡ group and there were 1 and 4 cases of hypoglycemia in Lantus group and CVⅡ group respectively. Conclusion:For the patients with T2DM receiving surgical intervention and having no or irregular food after operation, insulin pump and Lantus both can control the blood glucose level effectively and have low rate of hypoglycemia. The treatment of CVII has bad control of the blood glucose level and bad compliance. Lantus is a effective, safe and digestible drug for the patient with T2DM after operation.

Abstract:Objective:To investigate the therapeutic effect of Mucosta on chronic erosive gastritis for 8 weeks period. Methods:This study enrolled 60 patients with CEG diagnosed by gastroscopy. After 8 weeks of therapy, the changes of symptoms were observed, and the level of PGE2 and MDA were examined. Results:Fifty-six patients with CEG completed 8 weeks of treatment. In the study group and the control group, symptom scores were significantly decreased(P < 0.05), but there were no significant difference between two groups. A better mucosa restoration on gland density and morphology were observed in the study group(P < 0.05)at the end of 8 weeks. In both groups, the concentration of PGE2 increased significantly and MDA decreased more than before(P < 0.05), and there was significant difference between the study group and the control group in 8 weeks. Conclusion:The gastric protective drug Mucosta can better relieve the symptoms of the patients with CEG, healing the erosive lesion in the stomach, increasing the level of PGE2 and decreasing the level of MDA. Mucosta is an effective drug for CEG.

Abstract:Objective:To study the expression of paxillin and CD44v6 in esophageal carcinoma and to investigate the relationship between the expression of paxillin and CD44v6 and invasion and metastasis of esophageal carcinoma. Methods:Paxillin and CD44v6 expression were detected in 24 normal esophageal mucosa, 94 primary tumor tissues and 83 lymph nodes by SP immunohistochemical method. Results:The positive expression rate of paxillin in primary tumor tissues was related with invasive depth(P < 0.01), clinical staging(P < 0.01) and metastasis of lymph node(P < 0.05). The positive expression rate of CD44v6 in primary tumor tissues was related with invasive depth(P < 0.05), clinical staging(P < 0.01) and metastasis of lymph node(P < 0.05). The expression of paxillin, CD44v6 in primary tumor tissue and metastatic lymph node was much higher than that in non-metastatic lymph node respectively(P < 0.05). Paxillin expression was not correlated with CD44v6 expression in lymph node tissues(P > 0.05). Conclusion:Paxillin and CD44v6 are over expressed in esophageal carcinoma tissue. They play an important role in invasion and progression of esophageal carcinoma and can be used as valuable biomakers to evaluate esophageal carcinoma metastasis.

Abstract:Objective:To study ureaplasma urealyticum(UU) and mycoplasma hominis(MH) infection in the urogenital duct and their drug-resistance. Methods:Mycoplasma culture was carried out with the samples of 682 cases and the drug-resistance was detected at the same time. Results:359 samples from 682 suspected cases cultured were identified to present with positive mycoplasma(52.64%). Among them , 298 samples were infected by UU(43.7%), 8 samples by MH(1.2%), and 53 samples by both UU and MH(7.8%). The detectable rate of UU was far higher than that of MH. Mycoplasma infection was chiefly found in middle-aged people ranging from 21 to 40 years old(86.63%). The positive rate of the female was much higher than that of the male(P < 0.05). The results of drug-resistance showed that mycoplasma was much sensitive to josamycin, pristinmycin, doxycycline, tetracycline, mid-sensitive to azithromycin and erythromycin, and resistant to ciprofloxacin,ofloxacin and clarithromycin. Conclusion:The urogenital mycoplasma infection results mainly from UU, especially in young and mid-aged people. Compared with simple UU or MH infection, mixed infection with UU and MH has significantly greater resistance to a wider variety of drugs. Josamycin, pristinmycin, doxycycline, and tetracycline are the primary choices for urogenital mycoplasma infection.
