Abstract:Objective:To select the specific aptamer of carcinoembryonic antigen (CEA), one of the most attractive molecule for cancer target therapy and imaging. Methods: Seven rounds in vitro selection were performed against the purified CEA protein. Ligand-mediated target purification and Co-immunoprecipitation were adopted to verify the specific binding of the aptamer to the purified and native protein separately. Results:The CEA-specific aptamer which can bind both the purified and native protein with the high specificity was obtained. Conclusion:This is the first time the CEA specific apatmer was produced. The results in this study provides the preliminary evidence for further investigation and application of CEA-aptamer in the future.

Abstract:Objective:To explore the effects of Iptakalim on intracellular free calcium concentration and on the proliferation of cultured rabbit pulmonary arterial smooth muscle cells induced by endothelin-1(ET-1) in vitro. Methods:A cell culture model, [3H]-thymidine([3H]-TdR) incorporation test and confocal microscope were used to observe proliferation and intracellular free calcium concentration([Ca2+]i) of rabbit PASMC induced by ET-1 in vitro. Results:The value of [3H]-TdR incorporation in ET-1 group was increased 1.468 times higher than that in control group. Iptakalim at the concentration of 10-7mol/L, 10-6 mol/L ,10-5 mol/L lowered [3H]-TdR incorporation by(19.8±4.6)%, (41.2±9.5)%, (54.7±10.1)%, respectively, compared with the value of the cells treated with ET-1(P<0.01); The intracellular fluorescence intensity of PASMC in ET-1 group was increased from 73.70±10.12 to 143.84±28.23, significantly higher than that in control group(P<0.01); whereas with Iptakalim,the fluorescence intensity(FI) was only increased from 74.30±10.20 to 86.03±9.82, significantly lower than that in ET-1 group(P <0.01). Conclusion:Iptakalim inhibited proliferation of PASMC and decreased intracellular free calcium concentration of cultured rabbit PASMC induced by ET-1.

Abstract:Objective:To express the melanoma associated gene MAGE-A9 recombinant protein, obtain the anti-MAGE-A9 monoclonal antibody and to examine the expression of MAGE-A9 in hapatocellular carcinoma specimens. Methods:MAGE-A9 cDNA was cloned from human hepatocellular carcinoma tissue by using RT-PCR, and then subcloned into the plasmid pMD18-T. After sequencing, the MAGE-A9 was cloned into the prokaryotic expression vector pBAD/gIII to construct the recombinant expression vector pBAD/gⅢ-MAGE-A9, and was transformed into E. coli TOP10. The recombinant MAGE-A9 protein was expressed under induction of L-Arabinose, and was purified through Hitrap column. The anti-MAGE-A9 monoclonal antibody was generated. The expression of MAGE-A9 in hepatocellular carcinoma specimens was examined through ABC assay. Results:The cDNA sequence of the cloned MAGE-A9 gene was consistent with the reported sequence. By affinity column and SDS-PAGE, the purified MAGE-A9 fusion protein displayed a band of Mr 35,000, and subsequently the anti-MAGE-A9 monoclonal antibody was obtained. We found that MAGE-A9 expressed in the cytoplast of positive cells and MAGE-A9 antigen was detected in 8 cases out of 39 (21%) hepatocellular carcinoma specimens. Conclusion:MAGE-A9 antigen was expressed in a fair proportion of hepatocellular carcinoma specimens, these patients might be suitable candidates for immune involving antigen, encoded by the MAGE-A9 gene.

Abstract:Objective:To investigate the role of 5-HT7 receptor in the pathogenesis of irritable bowel syndrome(IBS). Methods:Rat model of D-IBS was established by intracolonic instillation of acetic acid and restraint stress; Rat model of C-IBS was established by stomach irrigated with 0-4oC cool water daily for 14 d. The content and distribution of 5-HT7 receptor at the brain and bowel was examined by immunohistochemistry and the expression of 5-HT7 receptor mRNA was detected by fluorescence quantitative RT-PCR(Real-time PCR). Results:Immunocytochemistry result showed the 5-HT7 rceptor positive staining at hippocampus and hypothalamus of both C-IBS and D-IBS group was stronger than that of control group(P < 0.01). The 5-HT7R expression at ileum, proximate colon, distal colon of C-IBS group was significantly stronger than that of control group(P < 0.05). Realtime-PCR analysis results showed the expression level of 5-HT7 receptor at hippocampus and hypothalamus of both C-IBS and D-IBS group was increased than that of control group(P < 0.05). At proximal and distal colon of C-IBS group, the 5-HT7 receptor mRNA expression was increased compared with control group(P < 0.05). Conclusion:The up-regulated expression of 5-HT7 receptor at brain and colon may play an important role in the pathogenesis of C-IBS.

Abstract:Objective:To investigate the effects of emodin(EMD) on cell proliferation and correlative cytokines secretion of glomerular mesangial in rats. Methods:The effects of EMD on cell proliferation and IL-6 , TGF-βl secretion of glomerular mesangial in rats were observed. Cell proliferation was measured by MTT method. IL-6 and TGF-βl secretion was detected with ELISA. Results:EMD was able to inhibit the cell proliferation and down-regulate the IL-6 and TGF-βl secretion of glomerular mesangial, as compared to the model group in rats (P<0.05). Conclusion:EMD could significantly inhibit the cell proliferation, and reduce the creation of extracellular matrix(ECM), this indicated that it could play an important role in alleviation and prevention of glomerular sclerosis. The mechanism may be that EMD can reduce the IL-6 and TGF-βl secretion of glomerular mesangial cell in rats.

Abstract:Objective:To determine the relationship between the expression of aquaporin-4(AQP4) after intracerebral hemorrhage and dexamethasone treated. Methods:Collagenase Ⅶ was injected in caudate nucleus in a stereotaxis frame to establish the intracerebral hemorrhage(ICH) animal models. The intracerebral hemorrhage(ICH) rats were randomly divided into four groups: the sham group (group A), the ICH group(group B), low dose-treated group(group C), moderate dose group(group D) and high dose group(group E). The groups were respectively received an intraperitoneal dexamethasone injection with 1 mg/kg, 15 mg/kg, 30 mg/kg, twice a day for three days. The brain water content(BWC), the permeability of blood-brain barrier(BBB) and the expression of AQP4 were observed. Results:Both the BBB disruption and AQP4 expression decreased in treated groups, and the AQP4 expression had a dose-dependent manner in the dexamethasone treatment. And it seemed that low dose dexamethasone was in favor of brain swelling elimination, but the higher dosage had not similar effect. Conclusion:Dexamethesone may play a critical role on expression of AQP4 in the physiopathology of hemorrhagic edema.

Abstract:Objective:To explore a simple and pragmatic method to obtain sufficient olfactory ensheathing cells from human fetus by selective attachment of harvested cells combined with intermittent NT3 nutrition. Methods:DMEM/F12 culture solution including 10% fetal bovine serum or NT3 was used to culture olfactory ensheathing cells intermittently every 48 h. The cell state and growth rates of OECs were observed, and P75 staining was used to estimate the purity of the cells. Results:Human fetal OECs were positive with P75 immunocytochemical staining. OECs in dipolar or tripolar shape formed networks by their processes in vitro. The purity of OECs in “good state”was about 95% at 9 d and 83% on 12 d, respectively. Conclusion:The method of using different attachment rates combined with intermittent NT3 addition is a simple and effective way to culture and purify OECs.

Abstract:Objective:To investigate the effect of MCP-1 on mesenchymal stem cells(MSCs) homing to injured myocardium in a rat myocardial infarction(MI) model. Methods:Rat myocardial infarction model was established by permanent left anterior descending branch ligation. Mesenchymal stem cells from donor rats were cultured in IMDM and labeled with BrdU. The Rats were divided into two groups. Monocyte chemotactic protein 1(MCP-1) expression were measured by in situ hybridization and immunohistochemistry in the sham operated or infarcted hearts at 1, 2, 4, 7, 14 and 28 days post operation in MCP-1 detection group. The rats were injected with MCP-1, anti-MCP-1 antibody or saline 4 days after myocardial infarction in intervention group. Then, a total of 5×106 cells in 2.5 ml of PBS were injected through the tail vein. The number of the labeled MSCs in the infarcted hearts was counted 3 days post injection. Cardiac function and blood vessel density were assessed 28 days post injection. Results:Self-generating MCP-1 expression was increased at the first day, peaked at the 7th day and decreased thereafter post MI and remained unchanged in sham operated hearts. The MSCs enrichment in the host hearts were more abundant in the MI groups than that in the non-MI group(P = 0.000), the MSCs enrichment in the host hearts were more abundant in the MCP-1 injected group than that in the anti-MCP-1 antibody and saline injected groups (P = 0.000). Cardiac function was improved more in MCP-1 injected group than anti-MCP-1 antibody and saline injected groups(P = 0.000). Neovascularization in MCP-1 injected group significantly increased compared with that of other groups(P = 0.000). Conclusion:Myocardial MCP-1 expression was increased only in the early phase post MI. MCP-1 may enhance MSCs homing to the injured heart and improve cardiac function by promoting neovascularization.

Abstract:Objective:To investigate the effection of Arnebia Root oil on the FGF expression in wound surface and the ability to promote wound surface healing. Methods:24 wound surfaces of patients were divided into two groups. Experimental group was treated by Arnebia Root oil and the control was treated by petrolatum gauze. Histology, histochemistry, electron microscope methods and healing rate measurement were used to show the FGF expression and wound healing process. Results:Endogenous FGF were expressed in both of the groups, in which of the experimental group was higher than that of the control group. the wound surface healing rate of experimental group was also higher and paralleled with FGF expression. Conclusion:Arnebia Root oil has effects to promote FGF expression and enhances wound surface repair. The wound healing mechanism between the action of Arnebia Root oil and function of FGF need further investigating.

Abstract:Objective:To investigate the gene expression of osteoprotegerin(OPG) and osteoclast differentiation factor(ODF) in the bone tissue of patients with hip fracture due to osteoporosis. Methods:OPGmRNA and ODFmRNA in the bone tissue in 50 cases of osteoporosis sufferers(over 50 years old) with hip fracture(Observer Group) and 30 cases of hip facture sufferers with no osteoporosis(Control group) were analyzed with the Semi-Quantitative RT-PCR method. Results:The mRNA expressed of ODF, OPG were both high in the patients with hip fracture. In the control group, the expression of OPG mRNA was observed, while the expression of ODF mRNA was very slight. Conclusion:Aged patients contained all signals including OPG, ODF that are essential for inducing osteoclastogenesis and promoting bone resorption.

Abstract:Objective:To explore the epidemiology and etiology for an outbreak of acute respiratory tract infection that occurred in one county of Jiangsu Province, China 2004. Methods: Only cases meeting the case definition were included in the study. We reviewed the medical records of the cases who were admitted to the local hospitals, interviewed cases by a standard questionnaire, and then described the epidemiologic features and analyzed risk factors by means of a case-control study. We collected pharyngeal swab specimens and sent them to different laboratories for isolation and culture. The laboratory used different detection methods such as DIF, PCR, electron microscope examination and microneutralization assay, to identify and then type the positive specimens. Results:A total of 871 cases were reported during the period from April 18 to July 4, 2004. The distribution of onset times presented two peaks, one in late May and another in middle June. The epidemic occurred mainly in the elementary and junior high schools in ten townships of one county, and the mean age of the cases was 12 years (range 7 months to 18 years). The course of the disease was acute, and was characterized by fever accompanied with sore throat and tonsillitis. The WBC count of cases was normal or elevated. The mean duration of illness was 5 days (range 2 to 12 days). No fatalities from illness were reported. A case-control study indicated that the possible risk factors were close contact with a case and/or poultry before onset and sharing of towels among members of the family. The typical CPE was observed through inoculating pharyngeal swab specimens into the HEP-2 cell cultures in different laboratories. An infection of adenovirus type 3 was verified by detecting positive specimens in different methods. Conclusion:This investigation demonstrated that the acute respi-ratory infection in cases was caused by adenovirus type 3. Cases occurred in over 70 schools in ten townships in 2004, and the route of transmission was possibly close contact with cases or droplet transmission.

Abstract:Objective:To further investigate the ability of MRI and 1H-MRS techniques for presurgical evaluation of hippocampal sclerosis. Methods:MRI and 1H-MRS were performed on 30 healthy subjects to determine the confidence levels. Eight patients who were pathologically confirmed hippocampal sclerosis were then studied using the same protocols. The difference of hippocampal formation(DHF) was used to determine atrophy of hippocampus. Areas under the peak of N-acetylaspartate(NAA) ,Creatine(Cr) and Choline(Cho) were measured, and the ratios of NAA/Cr, Cho/Cr, and NAA/Cr+Cho were calculated. NAA/Cr+Cho value was applied to localize the seizure focus. Results:Two patients showed hippocampal atrophy according to DHF value. NAA/Cr ratio decreased significantly in ipsilateral hippocampus compared to that in contralateral hippocampus and control subjects(P < 0.01). Cho/Cr value increased in both ipsi- and contralateral hippocampus in comparison with that in control subjects(P < 0.01). NAA/Cr+Cho ratio, however, significantly reduced in both ipsi-and contralateral hippocampus(P < 0.01) with lowest NAA/Cr+Cho ratio in seizure foci. Six patients could be lateralized by reduced and/or asymmetric NAA/Cr+Cho value. Conclusion:1H-MRS should be a promising diagnostic tool to detect neuron abnormality. 1H-MRS and MRI complement each other in presurgical lateralization of epileptogenic lesion in epilepsy patients.

Abstract:Objective:To study the relationship between plasma treatment time and efficacy. Methods:Test specimens were prepared from an acrylic resin denture material in the size of 2 mm×10 mm×10 mm. Plasma treatment was carried out on the surface of Polymethyl methacrylate(PMMA) at different time. XPS studies, IR spectra studies and measurement of wetting angle were performed. Results: XPS showed the peak corresponding to C-O getting higher as the treatment proceeded, however at 120 seconds, the peak did not increase any longer and partly crossed with the peak at the duration of 60 seconds. IR spectra showed the wave corresponding to C-H was reduced as O2-plasma treatment proceeded, and then changed little. Wetting angle initially decreased dramatically, however, as the reaction proceeded, wetting angle increased slightly. Conclusion:Equilibrium was reached for introducing oxygen-containing groups and changing of C-H. As the treatment proceeded, wetting angle increased slightly.

Abstract:Objective: Early complications were analyzed in those with EvansⅢ type of intertrochanteric fracture treated with operation or nonoperative approaches. Methods:59 cases with Evans Ⅲ type of intertrochanteric fracture between June, 1999 to July, 2006 were admitted in our department. 38 patients were complicated with cardiovascular diseases, such as hypertension and arrhythmia. All the cases were operated. Results:58 cases were operated successfully. One died of pulmonary interstitial fibrosis. Grade data of different age brackets indicated that early infective complications had apparent differences between the operation and the control group. Compared with the control group, the operation group had benefits such as fewer complications, especially the infective complications, and incidence of lower extremities venous thrombosis, and the results showed notable differences. Conclusion:In elderly patients with EvansⅢ type of intertrochanteric fracture, the incidence of pulmonary infection was the highest. Venous thrombosis in lower extremities was the most serious complication. So it will be helpful to perform the quadriceps femories function exercise. The post-operative effect is also related with the operation approach.

Abstract:Objective:To report the clinical manifestations of AIDS with nervous system complications. Methods:We collected the clinical material of AIDS and HIV positive patients who were admitted to our hospital from January 1998 to July 2006, and retrospectively analyzed the 39 cases having nervous system complications, among 146 cases in total. Results:Among 39 cases, there were 3 cases of HIV dementia, 1 case of vacuolar myelopathy, 3 cases of Gullain-barre syndrome, 3 cases of myopathy and 26 cases of secondary opportunistic infection of CNS,including 9 cases of tuberculosis, 6 cases of cryptococcus, 5 cases of toxoplasma, 3 cases of herpes zoster virus, 2 cases of herpes simples virus, 1 case of cytomegalovirus, 1 case of progressive multifocal leukoencencephalitis. 2 cases with central nervous system lymphoma.Among them 12 patients gave up treatment, other patients received anti-HIV treatment and anti-microorganism treatment. 5 patients died of respiratory failure, 2 patients died of multiple organs failure. Conclusion:Up to now, AIDS still has not very good management. So prevention is very important.

Abstract:Objective:To observe the effect of lithium chloride on neuronal apoptosis and GSK-3β of hippocampal CA1 area after transient focal cerebral ischemia/reperfusion in rats. Methods:126 male SD rats were randomly divided into normal group(NO group),sham operation group(SH group),ischemia/reperfusion group(IR group), Li1 group, Li2 group,and Li3 group. The change of neuronal apoptosis in the CA 1 region was examined by Hoechst staining. The expression and distribution of GSK-3β and p-GSK-3β(Ser9) in the cell was examined by immunofluorescence analysis. Results:The apoptotic nuclei could be detected significantly in IR groups(compared with SH group, P < 0.01). Compared to IR group, Li groups dose dependently(1~3 mmol/kg) reduced the apoptotic nuclei in happocampal CA1 area. There was no visible difference among the total amounts of GSK-3β in each group(P > 0.05). P-GSK-3β(Ser9)increased significantly after reperfusion(P < 0.01). Lithium chloride treatment can dose dependently increase the expression of p-GSK-3β(Ser9)(compared with IR groups, P < 0.01). Conclusion:Lithium chloride can dose dependently suppress neuronal apoptosis and GSK-3β expression after MCAO/ reperfusion.

Abstract:Objective:To explore the effect of human growth hormone(GH) on bile duct cancer cell line(QBC939 cell) and pancreatic carcinoma cell line(BXPC-3 cell) in vitro and the possible mechanism. Methods:The QBC939 and BXPC-3 cells during exponential growth stage were harvested and divided into experimental group and control group. The experimental group was separated into four sub-groups(50 μg/L 2 h,50 μg/L 24 h,100 μg/L 2 h,100 μg/L 24 h) according to the dose of GH and culture time. After 2 h and 24 h, Insulin-like Growth Factor1 was detected by using ELISA respectively. The carcinoma cells cultured for 24 h with various GH concentration(50 μg/L,100 μg/L) were made into single cell suspentions, and the samples underwent subsequent cell cycle evaluation. IGF1RmRNA was tested with the method of hybridization in vitro. Results:GH stimulated cell growth in vitro and also elevated S phase percent and proliferation index(PI) of cells after 24 h culture(P < 0.05). The quantum of IGF1 was significantly increased after 2 h culture in GH groups, but there was no statistical significance between GH groups and NS groups 24 h ago. IGF1RmRNA was expressed in two carcinoma cell lines in contrast to blank controls, and the expression boosted up according to the dose of GH. Conclusion:GH stimulates QBC939 and BXPC-3 cell proliferation in vitro,and the possible mechanism is acted by the axis of GH-IGF1.

Abstract:Objective:To investigate the growth inhibition of Oxaliplatin(L-OHP) or Cisplatin(CDDP) in vitro and Livin gene expression induced by the two platinum compounds on human gastric cancer cell line(BGC-823). Methods:The proliferation inhibition ratio was evaluated by MTT in the time of 24, 48 and 72 hours. Different inhibiting concentrations(IC10, IC30 and IC50) of the two platinum compounds were added into the medium of BGC-823 cells. The expression of Livin gene was detected by RT-PCR, and the protein of Livin was assessed by Western-blot. Results:The proliferation of BGC-823 was inhibited in a dose-and time-dependent manner after the treatment of CDDP or L-OHP, especially the latter was dose-dependent significantly than the former. The expression of Livin, which was down-regulated by the two platinum compounds in the earlier period, was up-regulated after 72 hours with CDDP other than L-OHP. Conclusion:L-OHP, which can more significantly inhibit the proliferation of BGC-823 in a dose-dependent manner than CDDP, is a potent inhibitor of Livin. The different influences on the expression of Livin will contribute to the investigation of regulation and function on Livin.

Abstract:Objective:To investigate the reversal effect of the effective component of Verbena Officinalis L. (4’-methery-scutellarein,4’-M-S) on multidrug resistant JAR/VP16 cell line. Methods:MTT[3-(4,5-dimethylthiazol-2-yl) 2,5-dipheny tetrasodium bromide] assay was used to examine the action of 4’-M-S on proliferation of JAR/VP16 cells, determine the available reversal concentration of 4’-M-S and measure the reversal index and ratio on JAR/VP16 cells. Secretory volume of chorionic gonadotrophic hormone(hCG) was measured by monoclonal antibody enzyme-linked immunoquantitation. Cell cycle profile and apoptotic cell rate was examined by flow cytometry(FCM), the morphologic and ultramicrostructural changes were observed with light and transmission electron-microscope(TEM). Results:The resistance of JAR/VP16 to VP16, MTX and ACTD, was enhanced up to 5.02,3.67,2.48 folds and ratios of 81.19%,76.89%,64.08% after exposure of the cells to 20 -滋g/ml 4’-M-S, respectively. Secretory volume of hCG in drug-resistance cell was decreased obviously when treated with combination of 4’-M-S and VP16. Cell cycle was blocked at the period of G0-G1 and G2-M , cell apoptotic rate was increased to 19.71%. The earlier apoptotic changes were observed under TEM in the cells treated. Conclusion:4’-M-S has a strong reversal effect on multidrug resistance in JAR/VP16 cells, which may be achieved by inhibiting growth of JAR/VP16 cells and inducing cell apoptosis.

Abstract:Objective:To investigate the effects of genistein on the the actin cytoskeleton and the adhesion of human breast cancer cells(MDA-MB-435 cells). Methods:The MDA-MB-435 cells were treated with different concentrations of genistein(12.5, 25.0, 50.0 and 100.0 -滋mol/L) for 24 h. Cellular F-actin stained with FITC-phalloidin was examined by fluorescence microscopy and flow cytometric analysis. The adhesive ability of MDA-MB-435 cells to artificial basement membrane(Matrigel) was measured by MTT assay. Results:Genistein significantly induced the depolymerization of cellular F-actin and caused the changes of cellular morphology and the redistribution of the F-actin. Meanwhile, genistein also reduced the adhesion of MDA-MB-435 cells to Matrigel. Conclusion:Genistein induced the less adhesion of MDA-MB-435 cells to Matrigel, which may related to the reorganization of actin cytoskeleton.

Abstract:Objective:To examine the protective effects of eryrthropoietin(EP) on neonatal SD rat cardiomyocytes injury induced by hypoxia/reoxygenation and explore its mechanisms involved. Methods:The cultured neonatal SD rat cardiomyocytes were divided into four groups:①hypoxia/reoxygenation group(H/R):deoxygenation for 2 h and reoxygenation for 4 h;②EP group:EP(10 U/ml) was added 1 h before deoxygenation,then deoxygenation for 2 h and reoxygenation for 4 h;③Wortmannin+EP group(W+EP group):Wortmannin(100 nmol/L), a specific phosphatidylinositol-3-kinase(PI3-K) inhibitor,was added at 30 min before EP incubation;④Control group. Cell apoptosis were measured by flow cytometry.The expression of EP receptor and p-Akt/Akt level were detected by Western blot. Results:EP significantly decreased the apoptotic index induced by hypoxia/reoxygenation and strongly increased p-AKt/Akt level. However, pretreatment with Wortmannin attenuated the cytoprotective effect of EP. Conclusion:Myocardial death and apoptosis are involved in hypoxia/reoxygenation injury,and EP can protect neonatal rat cardiomyocytes from hypoxia/reoxygenation-induced apoptosis. The protective effects are partly associated with PI-3K/AKt pathway.

Abstract:Objective:To evaluate the cytotoxicity of nano-silica coating with sol-gel process of alumina-based ceramic. Methods:Coating gel after heat treatment was analyzed by infrared spectrum(IR), with gel without sintered as control. Human gingival fibroblast cells(HGFC) were cultivated with extractions of alumina-based ceramic with or without nano-silica coating. Relative growth rate (RGR) was analyzed with a methyl thiazolyl tetrazolium(MTT) assay. Results:It can be seen from the IR picture that bending vibration absorption kurtosis of Si-OH vanished after heat treatment. No significant difference was observed between alumina-based ceramic with and without nano-silica coating. Cytotoxical grade(CG) was 0 to 1. Conclusion:Heat treatment may reinforce Si-O-Si net structure of coating gel. Nano-silica coating with sol-gel process of alumina-based ceramic had no significant cytotoxicity to HGFC.

Abstract:Objective:To compare the measurement of knee articular cartilage volume of swine between FLASH(Fast Flow Angle Shot) and MEDIC(Multi Echo Data Imagine Combination) MRI data at 3.0T. Methods:The knees of 20 swine were examined by sagittal(sag) MRI performed with a FLASH water-excitation sequence and a MEDIC. After scanning, knee cartilage plates of swine were resected. Results:Obtained with quantitative MRI were compared with that obtained by direct measurement of cartilage volume of the surgically removed tissue. In the knee joint of swine, average reproducible error for cartilage volume was 2.5 to 3.2% with sag FLASH, and 1.6 to 3.0% with sag MEDIC. Correlation coefficients between MEDIC and FLASH and morphologic analysis ranged from 0.90 to 0.98 for cartilage volume. Random pairwise differences between results obtained with FLASH and MEDIC and morphologic analysis were 4.3% to 6.8% and 2.9% to 6.6%, respectively. Systematic pairwise difference between results obtained with FLASH and MEDIC and morphologic analysis were -0.6% to 1.2% and -1.1% to 2.8%, respectively. Conclusion:Cartilage volume reproducibility was slightly higher for the MEDIC data than in the FLASH data. While sag MEDIC permits accurate analysis of cartilage morphology in all knee cartilage plates at reasonable acquisition times in the knee of swine.

Abstract:Objective:To study the effect and mechanism of cantharidin(protein phosphatase 2A inhibitor) on the protection of pancreatic β-cell’s death and dysfunction. Methods:Twenty type 1 diabetic mice induced by low dose streptozotocin(STZ) were randomly assigned into cantharidin-treated group(n=10) and diabetes mellitus group(n=10). Ten healthy mice were as normal control. The level of fasting blood glucose(FBG) was continuously detected. In the end, the level of fasting serum insulin(FINS) was measured and the morphology of pancreas was observed under optical microscope with HE staining. Results:The level of FBG in cantharidin-treated group was obviously lower than diabetes mellitus group(P < 0.05). The level of FINS in cantharidin-treated group was significantly higher than diabetes mellitus group(P < 0.01). Compared with diabetes mellitus group,the volume and cell number of islets in cantharidin-treated group were increased significantly. Conclusion:Cantharidin could obviously protect STZ-induced diabetes on mice through improving structure and function of survival islets and enhancing the insulin secretion of pancreatic β-cells.

Abstract:Objective:To investigate whether monocyte-macrophages separated from the normal term placenta could differentiate into functional dendritic cells(DCs) in vitro. Methods:Mechanically separated monocyte-macrophages from normal term placenta were cultured in the transendothelial trafficking model for 48 h. The DCs induced from placental monocyte-macrophages were identified for their morphology and phenotype by scanning electron microscope, FACS and for their abilities of inducing proliferation of allogenic T cells by 3H-TdR incorporation assay. Results:After cultured in the transendothelial trafficking model for 48 h, the DC-like cells derived from the normal term placental monocyte-macrophages displayed characteristic morphology and expressed higher level surface antigen of CD80, CD86 and HLA-DR, and could stimulate the proliferation of allogenic T cells. Conclusion:In the transendothelial trafficking model monocyte-macrophages separated from the normal term placenta can be induced into functional dendritic cells(DCs).

Abstract:Objective:To study the different expression of heterogeneous nuclear ribonucleoprotein A2/B1(hnRNP A2/B1) among normal endometria, eutopic and ectopic endometria of patients with endometriosis. Methods:The expression differences among three normal proliferate-stage endometria, three proliferatestage eutopic and ectopic endometria of patients with endometriosis was examined by immunohistochemistry and Western blot. Results:hnRNP A2/B1 was located in the nucleus of both epithelial glands and stroma. The expression level of hnRNP A2/B1 in ectopic and eutopic endometria of patients with endometriosis both were weaker than that in normal endometria(P < 0.05), while the difference between ectopic and eutopic endometria in the patients was not significant(P > 0.05). Conclusion:hnRNP A2/B1 may play an important role in the pathogenesis of endometriosis.

Abstract:Objective:To construct eukaryotic expression vector carrying anti-sense PRLR gene and investigate whether the reco-mbined plasmids could regulate the expression of PRLR mRNA and affect the growth of human breast cancer cell line MCF-7. Methods:PRLR cDNA was inserted into the expression plasmid pcDNA3.1(-) to construct the recombined plasmids p-anti-PRLR that encoding PRLR cDNA in an anti-sense orientation, then the recombined plasmids transfected MCF-7 cells by lipofectin. The stable cell lines were obtained after G418 selection. PRLR mRNA expression was detected by real-time PCR. MTT assay, clone formation test, and the flow cytometry(FCM) were used to evaluate cell proliferation. Results:(1)The eukaryotic expression vector p-anti-PRLR was constructed successfully. (2)Antisense PRLR gene decreased the level of PRLR mRNA in MCF-7 cells. (3)After antisense gene transfection, the proliferation ability of MCF-7 cell was decreased, and the G1 phase cells were increased, but the S phase cells were decreased in cell cycle. Conclusion:The results suggested that antisense PRLR could inhibit MCF-7 cell proliferation, and it might be significant for the breast cancer gene therapy.

Abstract:Objective:To establish a practical HPLC method to determine the concentration of methazolamide in aqueous humor of rabbit eyes for the research of methazolamide drug delivery system. Method:Protein was deposited by 10%(V/V) trichloroacetic acid. Methazolamide was extracted from aqueous humor by using C18 column and mobile phase of methanol ∶ water(25 ∶ 75,V/V). The column temperature was maintained at 40℃, methazolamide was detected at 290 nm, and the flow rate was 1.0 ml/min. Results:A good linear relationship was obtained between the concentration of methazolamide from 0.05 -滋g/ml to 10 -滋g/ml, r = 0.9982. The Relative recovery and absolute recovery of methazolamide were 101.7% and 82.7%, respectively; and RSD of intra-day and inter-day were less than 10%. Conclusion:The improved method could significantly enhance the detective sensitivity without damage to the column and increase the accuracy. The procedure was simple, sensitive, and suitable for determination of methazolamide in aqueous humor with good practicability.

Abstract:Objective:To observe the effect of arsenic trioxide to the whole body and local structure following injection into the foramen mentale of rabbits. Methods:0.5 ml arsenic trioxide with different concentrations of 0.1%, 0.05%, 0.025%, and 0.0125%, were injected into foramen mentale.The changes of body appearance, count of white blood cells and pathologic feature were observed. Results:With suitable arsenic trioxide concentration, mentale nerve was permanently damaged,and no side effect on the whole body. Structures around the nerve was not seen damaged. Conclusion:Suitable arsenic trioxide concentration can damage nerve axons permanently, and can not bring side effect to the structure around the nerve and whole body.

Abstract:Objective:To investigate the correlations between electrogastrogram(EGG) and gastric emptying(GE) in patients with functional dyspepsia(FD). Methods:Fifty-one patients with FD were detected by polygraph ID 4-channel EGG analysis system(EGG) and solid radiopaque marker 5h after a solid meal(GE). Results:31.4% patients with FD showed delayed GE, 72.6% showed abnormal EGG. There were no significant differences between normal EGG groups and abnormal EGG groups in average percentage of postprandial 5 h emptying. In patients who had both abnormal EGG and delayed GE, EGG abnormility mainly exhibited dysrhythmia, and the dominant frequency of EGG were normal. Some patients also exhibited non-enhancive dominant power postprandial. There were no significant differences among patients with mild, moderate and severe symptoms in GE percentage. The patients with severe symptoms had higher dysrhythmia percentage than those in mild and moderate symptoms(P < 0.05), but there was no significant difference between the patients with mild and moderate symptoms. There were no significant difference of GE percentage between Hp negative groups and Hp positive groups, but the dysrhymia percentage of Hp positive groups was higher than Hp negative groups(P < 0.05). There were also no significant differences between active gastritis group and inactive gastris group in both dysrhymia percentage and GE percentage. Conclusion:The abnormality of EGG plays an important role in the pathogenesis of FD. There was no significant relationship between EGG and GE. The patients with severe symptoms and with Hp positive have higher abnormal EGG percentage. Whether the gastritis is active or not, that was no significant correlation with EGG or GE percentage.

Abstract:Objectives:To investigate the effects of tamoxifen(TAM) on the growth of estrogen receptor(ER) negative thyroid carcinomas and the mechanisms involved. Methods:In vitro experiments, papillary thyroid carcinoma cells(IHH-4) and follicular thyroid carcinoma cells(FTC-133) were treated with tamoxifen in different doses and different lengths of time, using the dimethylthiazol-dipheltetrazolium bromide(MTT) method to observe the cell proliferation. Cell cycle and apoptosis were analysed with flow cytometric DNA studies. In vivo experiments the papillary thyroid carcinoma cells were injected into the nude mice subcutaneously, and the mice had been feed with TAM[5 mg/(kg·d)] daily since the second week. The tumor volumes were calculated weekly. Finally the mice were sacrificed and the tumors were weighed separately. Results:In vitro,TAM inhibited the proliferation of the IHH-4 and FTC-133 cell lines in dose-dependent and time-dependent pattern. TAM inhibited the growth of the IHH-4 and FTC-133 cell lines by blocking the cell cycle in G1 phase. Immunohistochemistry and RT-PCR studies showed the ER negative in both cell lines. In vitro,TAM also inhibited the growth of IHH-4 xenografts in nude mice. Conclusions:TAM inhibits the growth of thyroid carcinoma in vitro and in vivo. TAM acts independent of estrogen receptor and could be a potential choice for the treatment of the aggressive thyroid carcinomas clinically.

Abstract:Objective:To study the expressions of TGF-β1, CD31 in papillary thyroid cancer tissues and the correlation with the lymph node metastasis. Methods:The expression of TGF-β1mRNA was detected by in situ hybridization, and the expressions of TGF- β1, CD31 were detected by EnVisionTM immunohistochemistry method in 55 tomour tissues and 10 normal tissues; the expression of CD31 was used as the mark of the vascular endothelial cell for detecting the tumor microvascular density(MVD). Results:The positive expression rate of TGF-β1mRNA in papillary thyroid cancer was 80.00%. The positive expression rate of TGF-β1 in papillary thyroid cancer was 76.37%. The expression levels of TGF-β1 mRNA and TGF- β1 in papillary thyroid cancer tissue were higher than those in normal throid tissue(P < 0.01). There was positive correlation between MVD and TGF-β1. Conclusion:TGF-β1 could could promote the microvascular formation in papillary thyroid cancer tissues and play an important role in the lymph nodes metastasis.

Abstract:Objective:To translate tinnitus handicap inventory(THI) into Chinese and evaluate its quality through clinical use and analysis. Methods:199 patients reporting tinnitus as their primary compliant or secondary to hearing loss were sampled as study participants. They were interviewed with THI Chinese version by investigators and accepted ear and hearing assessment. Data were analysed. Results:The split-half reliability was 0.88,test-retest reliability was 0.98, and Cronbach-蒺s α was 0.90. These results proved the THI Chinese version had good reliability. The THI scores had significant difference in the subjects between outpatients and inpatients. There was significant improvement after treatment in inpatients group. The THI scores were highly correlated with the hearing loss level of the worse ear. Conclusion:THI Chinese version has good reliability and responsiveness. Total score of THI was strong recommended in the research and clinical practice.

Abstract:Objective:To investigate the effects of Tamoxifen on renal interstitial fibrosis in UUO rats. Methods:Feamale SD rats were divided into 3 groups at random:①UUO group(6 rats), ②the treated group(6 rats):UUO + Tamoxifen 10 kg/(mg·d) day by gastrogavage, ③sham operated group(6 rats). Kidney were harvested at day 9 after UUO. The renal tissues were examined by HE,PAS and Masson. Tissue α-SMA protein expression was determined by Immunohistochemistry. Results:UUO for 9 days significantly activated α-SMA protein expression, induced interstitial fibrosis and tubulointerstitial lesion in obstructed kidney of UUO group, compared with sham operated group rats. Conversely daily administration of Tamoxifen significantly inhibited α-SMA expression and decreased interstitial fibrosis and tubulointerstitial lesion in obstructed kidney of treated group. Conclusion:Tamoxifen had beneficial effects on reduction of renal interstitial fibrosis in SD rats with UUO, and this effect may be associated with Tamoxifen inhibits α-SMA expression.

Abstract:Objective:To quantitatively assess macular retinal thickness and display the features of macular morphology using stratus optical coherence tomography(OCT) in patients with type 2 diabetes mellitus. Methods:In a prospective study,OCT was performed in 120 eyes of 75 diabetic patients and 30 healthy eyes of normal control subjects. The diabetic patients were divided into three groups:non-diabetic retinopathy(NDR), non-proliferative diabetic retinopathy(NPDR) and proliferative diabetic retinopathy(PDR). We observed the different tomographic features of diabetic macula and measured macular thickness with the fast macular thickness mapping protocol of the stratus OCT at the same time a day. For each eye,nine different sectors were analyzed(a foveal sector,four parafoveal sectors,and four extrafoveal sectors). Results:There were statistically significant differences in foveal thickness and each of the nine areas among the control eyes,NPDR and PDR groups(P < 0.01). But it had no significant differences between control eyes and NDR group. Visual acuity correlated with retinal thickness at the central fovea in diabetic retinopathy(r =-0.69;P < 0.01). Furthermore,OCT can identify sponge-like retinal swelling, cystoid macular edema(CME), serous retinal detachment,hard exudates,epiretinal membranes(ERM) and other features in diabetic patients. Most eyes had more than one pathologic change in these diabetes OCT demonstrated diabetic macular edema(DME) in 60.8% of eyes,hard exudates in 22.3% of eyes,ERM in 17.5% of eyes and normal foveal structure in 35.8% of eyes. Conclusion:OCT is a useful tool for observating macular thickening and structural changes in eyes with diabetic. OCT can facilitate the diagnosis, treatment and prognosis of diabetic patients,which is especially important in the early stages of diabetic maculopathy.

Abstract:Objective:To explore the genetic association between the regulated on activation, normal T cell expressed and secreted(RANTES) gene promoter -403G/A polymorphism and respiratory syncytial virus(RSV) bronchiolitis in Chinese Han nationality population. Methods:Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) was used to identify the polymorphism at position -403 of the RANTES promoter in 238 patients with RSV bronchiolitis and 288 health controls. The total IgE concentrations in serum samples were measured by enzyme-linked immunosorbent assay(ELISA). Results:Compared to normal groups, no significant difference was demonstrated for genotype and allele frequency of the RANTES -403G/A polymorphism in RSV bronchiolitis. However,both the percentage of the personal atopy history and the familial atopy history showed significant difference among the three genetypes of -403G/A in RSV bronchiolitis group(P < 0.05). Interestingly,compared with those carring the GG genotype,subjects with -403A allele demonstrated a 2.57-fold increased risk of personal atopy history. The IgE level indicated significant difference among the three genetypes in RSV bronchiolitis group. Conclusion:The RANTES promoter -403G/A polymorphism may not be associated with the susceptibility of RSV bronchiolitis in Chinese Han nationality population. However,the -403A allele may be a predisposing factor of the personal atopy history.

Abstract:Objective:To investigate the effects of luteolin on airway remodeling and the expression of IL-13Rα2 in the airway of chronicity asthmatic mice. Methods:Thirty-two BALB/c mices were randomly divided into four groups. The mouse asthma models(asthma group) were established by sensitization and challenge with ovalbumin(OVA), eight mices interfered with luteolin 30 min before challenge(luteolin group), eight mices administrated dexamethasone by intraperitoneal injection 30 min before challenge(dexamethasone group). The healthy control mice were treated with saline instead of OVA. Lung tissu were collected 24 h after the last challenge. The left lung tissue was enbedded in paraffin and sectioned. Henatoxylin and esin(HE) staining were performed,Histologocial changes were detected. The expressions of genes and their prouducts of IL-13Rα2 in airway were detected using RT-PCR and immunohistochemistry(ISH). The expressions of genes of TGF-β in lung tissue were detected using RT-PCR. Results:①HE-stained lung tissue showed there were airway remodeling in each group,the thickness of airway in luteolin group was significantly decreased compared with asthma group and increased compared with normal group.②Wat/Pi and Wam/Pi in luteolin group and dexamethasone group were significantly lower than those in asthma group.③RT-PCR showed that expression of IL-13R2mRNA and TGF-β1mRNA in luteolin group and dexamethasone group were significantly lower than those in asthma group. ④ISH Showed that expression of IL-13Rα2 in luteolin group and dexamethasone group were significantly lower than those in asthma group. Conclusion:Luteolin might inhibit the thicking of airway wall and airway smooth muscle proliferation, and inhibit airway remodeling. Its possible mechanisms might be of inhibiting the expression of IL-13Rα2 in the lung tissue.

Abstract:Objective:To investigate the association between obstructive sleep apnea hypopnea syndrome(OSAHS) and metabolic syndrome(MS) in male Chinese patients. Methods:There were 86 male patients with OSAHS recruited as OSAHS group, which was further divided into mild, moderate and severe OSAHS subgroups. 241 normal male adults(AHI<5) with their age and body weight index (BMI) matched were recruited as control group. The incidence of MS were compared between normal and OSAHS subjects. All subjects investigated were tested for their metabolic parameters of peripheral circulation. In OSAHS subgroup continuous positive airway pressure(CPAP) treatment was performed in 25 subjects(treatment group). Comparison was made for correlation and difference of all parameters between OSAHS and control groups as well as before and after CPAP treatment in treatment group. Results:There was a significant difference between two groups in the following parameters:diastolic and mean arterial blood pressure, circumference of waist(WC) and hip(HC), waist-to hip ratio(WHR) and high density lipoprotein cholesterone(HDL-C). Compared with control group, the incidences of MS were not only significantly higher in OSAHS group as well as in its subgroups, but also became more remarkably high as OSAHS became more severe. Multiple factor stepwise regression analysis showed that OSAHS was the independent risk factor of MS. After CPAP treatment, there was a significant decrease in WC, systolic and diastolic blood pressure, FBG, IR and incidence of MS. Conclusion:OSAHS is closely associated with MS in male patients. OSAHS is one of the independent risk factors. CPAP treatment can improve IR and reduce the incidence of MS.

Abstract:Objective:To study the level of interleukin 19(IL-19)in peripheral blood of children with asthma and investigate its importance. Methods:Three groups were in the experiment:the acute onset group of asthma(21 patients), the catabasis group of asthma(19 patients), and health control group(22 cases ).The level of IL-19 and IgE in peripheral blood was respectively determined with ELISA. The accurate number of EOS in peripheral blood was counted with cytoanalyze. Results:Compared with the children of control group, the levels of IL-19, IgE and the quantity of EOS in peripheral blood of the patients with Asthma were obvious higher(P < 0.01).The level of IL-19 and the quantity EOS of the patients of acute attack group were higher than those of the patients of catabasis group(P < 0.05), but there is no significant difference in the level of IgE between the two groups of asthma. Furthermore, the results of correlation analysis showed that the levels of IL-19 positively correlated to the level of IgE and the quantity of EOS respectively in blood(r = 0.837, P < 0.01;r = 0.742, P < 0.01). Conclusion:The IL-19 level of the children with asthma was always higher than that of health children, and the ascending degree of the IL-19 level are different during various periods of asthma. It may be inferred that IL-19 can play an important role in the pathogenesis of asthma, and observing the change of the level of IL-19 can make for the clinical diagnosis and monitoring the pathogenetic condition of asthma.

Abstract:Objective:To clone envelope glycoprotein K8.1,K8.1A and gL gene of Kaposi’s sarcoma-associated herpesvirus(KSHV)and express them in 293T cells. Methods:K8.1 and gL genes were amplified by PCR with the total DNA of BCBL-1 cells as template. K8.1A genes were amplified by RT-PCR with the total RNA of BCBL-1 cells as template. The PCR fragments were cloned into pcDNA3.1(+) vector to construct recombinant eukaryotic expression plasmids. 293T cells were transfected with the recombinant plasmids. Western blot was performed to evaluate the expressions of K8.1 and K8.1A in 293T cells. RT-PCR was performed to evaluate the transcription of gL in 293T cells. Results:The sequence of K8.1,K8.1A and gL was 100% homology with K8.1,K8.1A and gL gene of KSHV previously registered in GenBank. An interesting band about 35 ku was visible in the result of Western blot,which was consistent with expected size of recombinate protein of K8.1 and K8.1A expressed in 293T. An interesting band about 500 bp was visible in the result of RT-PCR. Conclusion:Glycoprotein K8.1,K8.1A and gL of KSHV were correctly expressed in 293T cells.

Abstract:Objective:To investigate the linical value of acute hypervolemic hemodilution(AHH) combined with controlled hypotension in meningioma surgery. Methods:Thirty meningioma patients of ASA Ⅰ-Ⅱ were randomly divided into AHH group(group A,n = 15) and control group(group B,n = 15). In group A,6%HES was infused(25 ml/min,15 ml/kg) before incision(or after intubatin).Two groups patients were controlled hypotension with isofluran inhaled after incision. A jugular bulb catheter was inserted for blood gas sampling and intravascular pressure recording. Jurgular bulb gases, center pressure(CVP), intracranial pressure(ICP), cerebral perfusion pressure(CPP), mean arterial blood pressure(MAP) and heart rate(HR) were measured before and after the hemodilution. Results:The values of MAP,HR,ICP, Lac and PH had no significant change(P > 0.05) during operation. Jugular bulb oxygen saturation(SjVO2) and oxygen pressure(PO2) of group A had siginificently increased with those before AHH(P < 0.05) and group B. Conclusion:In meningioma surgery, AHH hemodilution combined with controlled hypotension, improved balance of cerebral perfusion and hemodynamics, which is a safe and effective technique.

Abstract:objective:To evaluate the experience of video-assisted thoracoscopic surgery(VATS)for bilateral bullous lung disease in the one-stage operation. Methods:One-stage operation VATS procedures were performed on 53 patients with bilateral ballous lung disease. 11 patients were operated with video-assisted minithoracotomy(VAMT). Results:All patients were cured. The mean duration of chest drainage after surgery was 4.6 days. Postoperative complications occurred in 13 cases,including pulmonary edema,pneumothorax again respiratory failure and so on. The mean postoperative hospital stay was 14.6 days. The patients were followed up 1 month to 5 years. Conclusion:VATS is an effective and preferred method for bilateral bullous lung disease. Advantages of VATS over conventional open techniques for bilateral bullous lung disease include shorter hospital stays, less postoperative pain, and earlier return to work.
