    • Reduced expression of Toll-Like receptor 7 of peripheral monocytes in patients with chronic hepatits B

      Abstract:Objective:To elucidate the role of TLR7 of monocyte-derived DCs in chronic hepatitis B virus infection. Methods:Monocyte-derived DCs were obtained from 10 chronically HBV-infected patients(CHB) and 15 healthy volunteers(control). TLR7 expression on immature and mature DCs were analyzed by FACS Calibur. DCs were pulsed with HBsAg, DCs maturation and ability to stimulate autogeneic T cells were examined. Results:It showed that TLR7 expression was different on monocyte-derived DCs as they go through different maturation stages. TLR7 expression on immature DCs and TLR7 expression on mature DCs were lower in CHB group. The ability of DCs loaded HBsAg to induce T cells producing IFN-γ was not different between the groups. Conclusion:In chronically HBV infection,TLR7 expression on monocyte-derived DCs was reduced and might be involved in a mechanism of persistent HBV infection. DCs pulsed with HBsAg protein stimulated autogeneic T cells producing IFN-γ and induced Th1 immune response.

    • Systematic review of the relationship between genetic polymorphism of cytochrome P450 2E1 at RasⅠ/PstⅠ loci and the susceptibility to lung cancer

      Abstract:Objective:To evaluate relationship between polymorphisms of cytochrome P4502E1 at RasⅠ/PstⅠ loci and susceptibity to lung cancer by means of Meta-analysis. Methods:Studies from 1991-2007 were retrieved through Cochrane Library,Medline,Elsevier,Ovid,Highwire Press,EMCC,CNKI,CBMdisk and VIP database without language limitation to collect relevant papers. Odds ratio of CYP2E1 genotype distributions in the group of patients with lung cancer and the group of healthy control was calculated. The Meta-analysis was applied with RevMan 4.2 software for heterogereity test and pooled OR calculation. Results:Nine case-control studies with 1 123 cases and 1 434 controls were analysed by fixed-effect meta-analysis method. The pooled odds ratio of CYP2E1 genotype c1/c1 vs c1/c2 +c2/c2 was 1.55(95%CI:1.30~1.86),Z statistics were 4.84. An association between CYP2E1 c1/c1 genotype and smoking was not found among cases. The pooled OR of interaction was 0.73(95%CI:0.45~1.15). Z statistics were 1.37. Conclusion:There was enough evidence currently to prove the relationship between CYP2E1 genetic polymorphism and susceptibility to lung cancer. It was demonstrated that the genetic polymorphism of CYP2E1 at RasⅠ/PstⅠ loci was the important risk factor for the susceptibility to human lung cancer, but there was not a definite interaction between the effects of smoking and CYP2E1 c1/c1 genotype on lung cancer risk.

      Abstract:Objective:To investigate the effects of GCP-02,a PPARα/γ dual agonist,on glucose utilization,pre-adipocyte differe-ntiation and cell viability in cultured cells. Methods:GCP-02 was tested for its effects on glucose transportation and consumption in HepG2 cells,on adipocyte differentiation in 3T3-L1 cells,respectively. Viability of HepG2 and WB-F344 cells was also investigated. Results:Viability of WB-F344 cells originated from rat was inhibited while that of HepG2 was not influenced by GCP-02 at the concentration of 10-5 mol/L. GCP-02 increased glucose consumption but not glucose transportation in HepG2 cells. It promoted oil production at the concentrations with no influence on cell viability in 3T3-L1 cells. Conclusion:GCP-02 increased glucose consumption in HepG2 cells,promoted 3T3-L1 pre-adipocyte differentiation,suppressed viability in WB-F344 cells.

    • Pharmacokinetics and relative bioavailability of metformin hydrochloride sustained release tablet in healthy volunteers

      Abstract:Objective:To compare pharmacokinetics and relative bioavailability of metformin hydrochloride sustained release tablet(the test preparation) with metformin common tablet(the reference preparation). Methods:Twenty male healthy volunteers were enrolled in a randomized two-way crossover design with a single-oral dose study(1 000 mg once per day for each preparation) and a multi-oral dose study(1 000 mg once per day for the test preparation and 500 mg twice per day for the reference preparation). The plasma concentrations of metformin were determined by high performance liquid chromatography(HPLC). A two-compartment model was adopted in metformin plasma concentration-time data analysis. Pharmacokinetics parameters were also calculated and bioequivalent was analyzed. Results:The main pharmacokinetic parameters of the test preparation and the reference preparation were as following:In single-dose group,Cmax was(1.61 ± 0.37) and (2.47 ± 0.74)μg/ml;Tmax(3.1 ± 0.6) and (1.5 ± 0.7)h;T1/2(5.1 ± 1.9) and(2.7 ± 0.3)h;AUC0→t(11.08 ± 2.88) and(12.03 ± 2.63)h·μg/ml and the relative bioavailability of the test preparation was(92.6 ± 14.7)%. In Multi-dose group:Cmax was(1.60 ± 0.25) and (1.57 ± 0.40)μg/ml;Tmax(3.2 ± 0.4) and (1.1 ± 0.5)h;T1/2(5.5 ± 3.1) and (3.0 ± 0.6)h;AUCss(13.24 ± 3.08) and (6.99 ± 1.50)h·μg/ml,and the relative bioavailability of the test preparation was(95.3 ± 17.5)%. Conclusion:The test preparation was bioequivalent to the reference common tablet,and showed the characteristics of sustained release.

    • Learning curve analysis of establishing the othotoptic liver transplantation model in rat by a single operator

      Abstract:Objective:To analyze the learning curve of establishing the model of othotoptic liver transplantation in rat by a single operator. Methods:The data of the first 100 consecutive cases of rat orthotopic liver transplantation(ROLT) model were collected for this retrospective study. The models were devided into five phages, of which each group were 20 cases. The donor and recipient operation time, anhepatic time,operation success rate,long-term survival rate and complications rate of each stage were observed and analyzed. Results:The anhepatic times of stage Ⅲ,Ⅳ and Ⅴ decreased significantly compared with the first two stages(P = 0.000). The operation success rate elevated from 35% to 100% steadily and the long-term survival rate raised to 95% with a plat-form survival rate of 55%-60% included(stageⅢ). The complications rate decreased significantly, and the ROLT model was established stably. Conclusion:Learners could grasp these skills by the learning curve with patient and training repeatedly, after which any model suitable for experimental study of liver transplantation could be established successfully.

      Abstract:Objective:To study the mechanism on SGC7901 resistance to the low dose cisplatin. Methods:SGC7901/CDDP was established in vitro. The morphology change between SGC7901/CDDP and SGC7901 was observed by microscope. The mRNA and protein expression of Survivin in SGC7901/CDDP and SGC7901 were detected by RT-PCR and Western-blot,respectively. Results:There were morphological differences between SGC7901 and SGC7901/CDDP. The mRNA and protein expressions of Survivin in SGC7901/CDDP were both higher than in SGC7901. Conclusion:SGC7901 resistance to cisplatin may have association with the increased expression level of Survivin in SGC7901/CDDP.

    • The clinical significance of expression of osteopontin in hepatocellular carcinoma

      Abstract:Objective:To study the clinical significance of osteopontin(OPN) expressions in hepatocellular carcinoma. Methods:Western Blotting was used to detect the expression of OPN in cancerous tissues and paraneoplastic liver tissues from 21 HCC patients. OPN expression was evaluated as well in 12 normal liver tissues. Using immunohistochemistry techniques, the expression of OPN,VEGF and CD34 were measured in 85 surgically removed primary HCCs,10 liver cirrhosis tissues and 10 normal liver tissues. Microvessel density(MVD) was also counted by Weidner method. Furthermore,the correlation between OPN expression and clinicopathologic features was analyzed. Results:The data of Western blotting reveals that OPN was significantly overexpression in HCC and associated closely with early recurrence and metastasis(P = 0.003)-The results from immunohistochemistry demonstrated that OPN in HCC was significantly higher than that in liver cirrhosis and normal tissues(P < 0.05). The OPN was significantly overexpressed in HCC with capsular infiltration(P = 0.03),HBV(P = 0.03) and associated with early recurrence and metastasis(P = 0.0018). A positive correlation between OPN and VEGF expressions was found in HCC(r = 0.71,P < 0.01). And there was also a positive correlation between OPN and MVD expressions in hepatocellular carcinioma(r = 0.44,P < 0.01). Conclusion:Overexpression of osteopontin was common in HCC and was significantly correlated with capsular infiltration,HBV and early recurrence and metastasis. The OPN exoression was closely related to VEGF exoression and MVD in HCC,and it indicated that OPN might participate in carcinioma angiogenesis.

    • The effect of GH on the cell cycle kinetics of cholangiocarcinoma and features of IGFBP3 in vivo

      Abstract:Objective:To explore the effect of recombinant growth hormone on cholangiocarcinoma in vivo,and study the possible mechanism of IGFBP3. Methods:Athymic nude mice were injected with QBC939 cells. When tumor became palpable obviously,animals were randomized to receive GH(GH groups:4 mg 2w,4 mg/kg.s.c. once daily for 2 weeks;2 mg 2w,2 mg/kg.s.c. for 2 weeks;2 mg 3w,2 mg/kg.s.c. for 3 weeks) versus saline control(NS groups). Animals and tumour were weighed,tumor and carcass weight rate were determined. Cells of tumor tissues were evaluated by flow cytometry(FCM). Western blot was used to detect IGFBP3 in mice liver. Results:There was no statistical significance of the difference on the host features(body weight,tumor weight,tumor and carcass weight rate,P > 0.05) or the cell cycle kinetics(G1,G2M,S,PI,P > 0.05) between GH groups and NS groups in vivo. Compare with NS groups,the expression of IGFBP3 in GH groups in mice liver was significantly increased according to the dose of GH[(2 mg/(kg·d):(0.63 ± 0.05) vs (0.28 ± 0.05),and 4 mg/(kg·d):(0.71 ± 0.07) vs(0.28 ± 0.05),respectively,P > 0.01]. Conclusion:GH couldn’t accelerate tumor growth in vivo,and the possible mechanism was acted by the effect of IGFBP3.

    • The combined detection of micrometastases in bone marrow and clinical significance in breast cancer patients with axillary lymph nodes negative

      Abstract:Objective:To study the clinical significance of immunocytochemistry(ICC) and reverse transcription-polymerase chain reaction(RT-PCR) in detection of breast cancer micrometastases in bone marrow of breast cancer patients with lymph nodes negative. Methods:After general anesthesia,all patients underwent bone marrow puncture and performed modified radical mastectomy. Bone marrow micrometastases were detected by ICC and RT-PCR in 62 patients, whose axillary lymph nodes showed negative HE-stain. Results:Among the 62 patients,15 cases(24.20%) presented RT-PCR positive,in which 9 cases(14.52%) were also ICC-positive. RT-PCR and ICC showed good uniformity(kappa = 0.6945),and there was significant difference in detective rate between the two methods(χ2 = 4.1667,P = 0.0412). Conclusion:Even there were no axillary lymph nodes involved,micrometastases still could be detected in bone marrow by ICC and RT-PCR. In addition,RT-PCR is more sensitive than ICC.

    • Study on effect of NGF in the intervention of erectile dysfunction induced by diabetes mellitus

      Abstract:Objective:To investigate the mechanism of action of nerve growth factor(NGF) in penis of rats with diabetic ED by detecting the variation of intracavernous presure(ICP) and Nitric oxide synthase(NOS) containing nerve in the corpus cavernosum of the rat model who use NGF to treat the diabetic ED. Methods:All of 60 male Sprague-Dawley rats were divided into 5 groups:A(n = 6) control group,B(n = 15) diabetic ED group,C(n = 14) NGF intervention group,D(n = 12) insulin intervention group,E(n = 13)NGF plus insulin intervention group(use the insulin by subcutaneous injection and NGF by intraperitoneal injection). Use the STZ i.p injected randomly to establish the dibetic model. Then the rats with diabetic ED were picked up after 8 weeks:B(n = 8),C(n = 8). D(n = 6),E(n = 6). Through 8 weeks of intervention,ICP was measured and the content of neuronal nitric oxide synthase(nNOS) in the erectile tissue was also evaluated by immunohistochemistry Staining. Results:Compared with those in group B,ICP was up-modulate significantly in intervened groups which received NGF or/and insulin intervention and the changes in nNOS staining of those groups were the same(P < 0.05). Conclusion:The ability of combination of NGF with its receptor may also be damaged. It is helpful to use exogenous NGF to ameliorate the neuropathy and improve the erection function of diabetic rats. The abnormal of NGF may play an important role in the generation of diabetic ED and its treatment.

    • Protective effects of Valsartan on over-oxidation injury of neonatal SD rat cardiomyocytes

      Abstract:Objective:To investigate the protective effects of Valsartan(Val) on cardiomyocytes injured by hydrogen dioxide(H2O2)in neonatal SD rat. Methods:The protective effects of various concentration of Val on the model of cultured cardiomyocytes injured by H2O2 in vitro were evaluated by detecting the cell viability. The activity of lactate dehydrogenase(LDH),malondialdehyde(MDA),superoxide dismutase(SOD) were also assayed. Results:Val could protect the myocardial cells from H2O2 injury in a dose-dependent manner(2~10 μmol/L), and Val(10 μmol/L) preconditioning showed the greatest protective effect. Val preconditioning could significantly attenuate the increased LDH 1eve1 and antagonize the elevated MDA level and the reduced SOD activity(P < 0.05). Conclusion:Val might be has the protective effects on cardiomyocytes injured by H2O2 through improving cell antioxidant ablility.

    • Simvastatin could improve the proliferation,migration and anti-apoptosis of endothelial progenitor cells

      Abstract:Objective:To investigate whether simvastatin of different concentrations have influences on the functions of EPCs in vitro. Methods:Total mononuclear cells(MNCs) were isolated from human peripheral blood by Ficoll density gradient centrifugation,and then the cells were plated on fibronectin-coated culture dishes. After 7 days cultured,attached cells were isolated and assessed with a laser scanning confocal microscope. Differentiating EPCs were characterized as adherent cells double positive for DiLDL-uptake and lectin binding. EPCs were treated with simvastatin of different concentrations(0.0001,0.0010,0.0100,0.1000,1.0000 μmol/L),or LY294002(PI3K inhibitor) plus simvastatins(0.01 μmol/L ). The influences of simvastatin on EPCs’ proliferation,migration and apoptosis were assayed with MTT assay,modified Boyden chamber assay and FACS assay, respectively. Results:Simvastatin strikingly improved the ability in proliferating,migrating and anti-apoptosis of EPCs isolated from human peripheral blood(P < 0.05). Simvastatin at 0.01 μmol/L concentration reached the maximum effects on EPCs,while the concentration of simvastatin was further increased more than 0.01 μmol/L,the improvement effects showed a tendency to decline,but the effects at 0.1 μmol/L concentration were still greater than those in control group. And the influences of simvastatin on these functions of EPCs could be blocked by LY294002. Conclusion:The present study established that simvastatin could improve EPCs-蒺 proliferation,migration and anti-apoptosis capability, and the mechanism might be involved in the PI3K/Akt pathway.

    • The expression of HMGB1 between necrosis and apoptosis of myocardiocyte induced by hydrogen peroxide

      Abstract:Objective:To observe the action of hydrogen peroxide(H2O2) inducing cardiomyocytes over-oxidation injury and to investigate the expression of HMGB1 in apoptosis and necrosis of myocardiocyte. Methods:A model for primary culture of cardiomyocytes injuried by H2O2 was established, the morphology of cardiomyocytes was observed, and cardiomyocytes fraction surviving were measured. The activity of lactate dehydrogenase(LDH), superoxide dismutase(SOD),HMGB1 were assayed. Results: Cardiomyocytes survival rate decreased by H2O2 treatment(P < 0.05), and the activity of LDH increased(P < 0.05),whereas the activity of SOD decreased(P < 0.05). Expression of HMGB1 in H2O2 group A was the same as that in control group, whereas the expression of HMGB1 in H2O2 group B was higher than that in control group and H2O2 group A. Conclusion:The HMGB1 expression increased in the necrosis of cardiomyocytes, while it had no change in the apoptosis of cardiomyocytes.

    • The effect of nifedipine on the expression of type I collagen in gingival fibroblasts

      Abstract:Objective:To investigate the effect of nifedipine,a calcium channel blocker,on the expression of collagen in gingival fibroblasts in vitro. Methods:Primarily gingival fibroblasts were cultured and incubated with various concentrations of nifedipine (108-360-1 200 μg/L). Enzyme-linked immunosorbent assay was used to evaluate the amount of type I collagen. Cell proliferation was measured by cell counting. Results:The expressions of collagen and cell proliferation were significantly different among the high concentration groups and the others on 5 th day,especially higher in 360 μg/L and 1 200 μg/L group. Conclusion: Nifedipine can increase the expression of type Ⅰ collagen in gingival fibroblasts and cell proliferation.

    • Changes of substance P in gingival crevicular fluid in response to periodontal treatment

      Abstract:Objective:To investigate the effects of periodontal treatment on the levels of substance P(SP) in gingival crevicular fluid(GCF), resulting in a return to periodontal health. Method:The course of non-surgical treatment was performed in 15 patients. Clinical indexes were measured at periodontitis site before treatment and at 8 weeks after the completion of treatment in each subject. A 30-s sample of GCF was collected from each test site using perio paper strips. The volume of GCF was calculated, and the SP level was analysed by radioimmunoassay. Results:The clinical condition of all test sites improved as a result of the periodontal treatment. The levels(pg/30 s sample) of SP fell from 45.48 before treatment to 10.11 after treatment(P < 0.05). The concentration(pg/ml) of SP in GCF fell from 36.90 to 25.82(P < 0.05). Conclusion:The reduction in inflammation resulting from effective periodontal treatment was associated with a reduction of neurokinin SP in gingival crevicular fluid.

    • Primary culture and identification of uterine leiomyoma cells

      Abstract:Objective:To explore the method of primary culture of uterine leiomyoma cells,which provided a cell line for studying the pathogenesis of uterine leiomyoma and drug screening. Methods:Human uterine leiomyoma cells were cultivated by digestive method and explanted tissue culture in vitro. The surviving cells were observed by phase contrast microscope and identified by immunohistochemistry staining. Results:Many uterine leiomyoma cells were obtained by the above two methods. The cells appeared in spindle shape and showed typical hill-valley pattern under phase contrast microscope. Immunohistochemistry staining with α-Actin demonstrated these cells were positive. Conclusion:Primary culture of uterine leiomyoma cells could be obtained by these two means,with highly purity,and can serve the further studies of the pathogenesis and drug screening of human uterine leiomyoma.

    • High glucose induce production of iNOS by human retinal pigment epithelium cells through activation of the p38 signal pathway

      Abstract:Objective:To study the changes of inducible nitric oxide synthase expression induced by high glucose treatment and the relationship between p38 mitogen activated protein kinase and the level of iNOS expression in human retinal pigment epithelium cells. Methods:The cells were divided into control group, high glucose group, high glucose plus SB203580 group and mannitol group. Cell viability was assessed by the MTT assay. The changes of iNOS and p-p38 MAPK expressed by RPE cells were studied with immunofluorescence staining method in a qualitative way. Results:(1) The treatment of RPE cells with 33 mmol/L glucose caused an obviously decrease of cellular viability. After pretreated with SB203580, cell viability was elevated compare with high glucose group. (2)iNOS expression and the phosphorylation level of p38 MAPK were enhanced obviously by high concentration glucose. After pretreated by SB203580, the high glucose-evoked iNOS expression was significantly inhibited. Conclusion:High glucose could inhibits RPE cells proliferation. RPE cells cultured with high glucose could enhance expression of iNOS of the cells in vitro, at least in part, by activation of p38 MAPK pathway. It suggested that RPE cells under the effecting of high glucose might participate in the pathological reaction of diabetic retinopathy.

    • Evaluation of macular changes before and after successful retinal detachment surgery using optical coherence tomography

      Abstract:Objective:To investigate foveal architecture after routine retinal detachment surgery for macular-off retinal detachment(RD) and analyze the correlation of it and visual acuity. Methods:41 eyes of 41 patients with macular-off RD underwent best-corrected visual acuity(BCVA) measurement and optical coherence tomography(OCT) scan through the fovea before surgery and 1 week,1,3,6,12 months after surgery. Statistical analysis of the data was performed. Results:1 week after surgery, BCVA was 0.05~0.50(P = 0.019), and 41.5 percent of patients showed subfoveal fluid reduction on OCT(P = 0.008). At 1 month, there was no obvious change on BCVA(P = 0.084) and subfoveal fluid(P = 0.053). At 3 month, the average BCVA was 0.1~0.6, which improved above 2 lines was 75.6%. At 12 months, the average BCVA was 0.15~1.00, which improved above 2 lines was 92.7%, and subfoveal fluid was absorbed completely in 95.1% patients. There was statistic different between group of 12 months after surgery and 6 months after surgery(P = 0.0019). Conclusion:Infra-clinical foveal detachment could be detected in the patients with successful retinal reattachment surgery by OCT examination. It could provide the forecast for visual acuity prognosis of the patients with macular-off retinal detachment.

      Abstract:Objective:To observe the nitric oxide synthase(NOS) expression in retina, and to investigate the function of the NOS inhibitor aminoguanidine(AG) in rats with acute intraocular hypertension. Methods:Twenty-seven SD rats were randomly divided into three groups:blank control(unoperational group),experimental group Ⅰ(ocular hypertension group),experimental group Ⅱ(drug-given group). Then each experimental group was divided into subgroups at 12 hours,24 hours,3 days and 7 days after perfusion,and NOS expression in the retina were detected by NADPH-diaphorase histochemistry. Results:The staining of NOS were localized in retinas of all groups. The count of NOS staining cells in the experimental group Ⅰ was more significant than that in the blank control. After AG was injected, the levels of NOS decreased. Conclusions:NO should play some cytotoxicity roles to retina. By inhibiting the activity of iNOS,AG might protect the retina against acute ocular hypertension injury.

    • Prediction of malignant middle cerebral artery infarction by plasma MMP-9 level

      Abstract:Objective:To evaluate the predicting value of the marker of endothelial injury:plasma matrix metalloproteinase-9(MMP-9) level on the progression of acute anterior circulating territory infarction progressing to malignant middle cerebral artery infarction(m-MCA). Methods:Eighty-five patients with acute anterior circulating territory infarction,in which 45 patients progressed to m-MCA,were collected and sampled consecutively. The plasma MMP-9 and C-reactive protein(CRP) of all patients were determined by means of ELISA and immunoturbidimetry,respectively,at admission. And the clinical characters and cranial CT features of the patients were analysed. Results:At admission,the plasma MMP-9 level in the patients with m-MCA(230.0 ± 60.5 ng/ml) was significantly higher than that in the patients with non m-MCA(174.0 ± 56.0 ng/ml)(P < 0.001),while the plasma CRP levels between m-MCA and non m-MCA patients had no significant difference(8.8 ± 3.8 ng/ml vs 8.5 ± 4.0 ng/ml,P > 0.05). The sensitivity and specificity of serum MMP-9≥220 ng/ml to predict m-MCA were 89% and 95%, respectively. Conclusion:The increased level of plasma MMP-9 may be a predictor to the m-MCA proceeding.

    • Clinical observation of two kinds of conjugated estrogen in the treatment of menopausal symptoms

      Abstract:Objective:To compare the efficacy and the safety of conjugated estrogen(CE) in the treatment of menopausal with conjugated equine estrogen(Premarin). Methods:According to the standards ,a total of 60 patients were randomly divided into 2 groups:CE group and Premarin group,a period of 28 days,and from 15 to 28 days plus medroxyprogesterone(MPA) 4 mg Daily.Treatment lasted for 3 periods. This randomly publicly parallel was adopted to compare the hot flashes symptom scores,serum level of estradiol,Kupperman measurement Index and serum lipid. Results:The scores of menopausal symptom,hot flash scores and the serum level of estradiol ascent. TG-HDL-C ascented and LDL-C were reduced after treatment with CE or Premarin(P < 0.001),No significant differences were discovered between the two groups through equivalence margin(P > 0.05). There are no significant differences between the two groups on side effect(P > 0.05). Conclusion:CE is safe and effective in treatment of menopausal symptoms.

      Abstract:Objective:To compare the efficacy and the safety of Remifemin in the treatment of climacteric complaints with Tibolon. Methods:According to the standards,a total of 244 patients were randomly allocated to receive either two tablets of Remifemin or one tablet of Tibolon daily for 12 weeks. This double-blind parallel clinical trial was adopted to compare the Kupperman’s score,Kupperman’s reaction ratio and side effect between the two groups. Results:There was no significant difference between two groups in Kupperman’s scores and Kupperman’s reaction ratio(P > 0.05),but there was significant difference on compound frequency Kupperman’s score and side effect by non-inferiority trial(P < 0.0001). Remifemin showed less side effect compared with Tibolon.(P < 0.001). Conclusion:Remifemin should be effective and safe in relief of climacteric symptoms.

      Abstract:Objective:To investigate the clinical significance of calcaneal quantitative ultrasound(QUS) for the female with QUS,and to investigate the relationship of QUS with DEXA. Methods:QUS and DEXA were performed in 511 female cases,aged 20-83 years. Results:Broadband ultrasound attenuation(BUA),speed of sound(SOS) and stiffness index(SI) of the female calcaneus were moderately related to bone mineral density(BMD) of Lumbar,Femur Neck,Femur Ward’s,Femur Troch,Total(r = 0.447-0.625);BUA,SOS and SI decreased obviously when the females were over 50 years old;BUA,SOS and SI in the calcaneus of the postmenopausal women were significantly less than those of the premenopausal women and decreased with the increase of the age. Those parameters of the osteoporosis group were obviously lower than those of the control group. Conclusion:Calcaneal QUS can indicate the change of female bone quality with the increase of ages. This is an effective method to diagnose osteoporosis and worth of introducing widely.

      Abstract:Objective:To study the cognitive function of middle-age and old patients with type 2 diabetes mellitus(T2DM) and related factors. Methods:Questionnaires were used to investigate 155 patients with T2DM to study the cognitive function and the possible infected factors of memory and non-memory. Results:Cognitive function of the patients with T2DM was not only associated with age(r = -0.366),sex(r = -0.374),educational level(r = 0.574) and incomes(r = 0.268),but also with course of diabetes disease(r = -0.247),the levels of blood glucose(r = -0.269),HBA1C(r = -0.235) and systolic pressure(r = -0.280). The patients with T2DM combined hypertension displayed significant difference to the controls in calculating(P = 0.006) and repeating sentence(P = 0.006) in MMSE,the right numbers of point pictures(P = 0.001) and the expended time of point pictures(P = 0.016). Conclusion:The impairment of cognitive function of middle-age and old patients with T2DM was concerned with age,sex,educational level,incomes,and the levels of blood glucose,combined hypertension and the level of hypertension. Control the level of blood glucose and hypertension effectively should prevent and mitigate the deterioration of cognition of T2DM.

    • Relationship between inflammation factors and peripheral vascular disease in Type 2 diabetes patients

      Abstract:Objective:To investigate the relationship between inflammation factors and peripheral vascular disease(PVD) in Type 2 diabetes(T2DM) patients. Method:The level of inflammation factors including hsCRP,IL-6,TNF--琢 were compared between PVD patients(ABI < 0.9) and N-PVD patients(ABI≥0.9), and the differences in body mass index (BMI),blood pressure (BP),serum glucose,HbA1c,and history were also observed. Result:The hsCRP level in PVD was significantly higher than that of N-PVD(0.61 ± 0.72 vs 0.12 ± 0.93,P < 0.001). The risk of PVD obviously increased when hsCRP was higher than 0.83 mg/L. The patients with cardiocerebrovascular disease had higher level in IL-6 than those who didn’t have such disease in PVD group. The PVD patients were older,while with higher level of fibrinogen ,more prevalent of stroke and lower diastolic blood pressure than that of the N-PVD ones. Conclusion:Inflammation factors had the similar clinical importance for the onset and progress of PVD pateints with cardiocerebrovascular disease. The existence of PVD should be paid affention to when the level of hsCRP was higher than 0.83 mg/L.Perhapse IL-6 was an alarm for initial atherosclerosis. Aging was one of the major risk factors of PVD in T2DM patients. There were unclear relationships between PVD and things such as fibrinogen,hypertension,and cerebrovascular disease.

      Abstract:Objective:To explore anti-reispiratory syncytial virus(RSV) effect of Chinese medicinal Reduning injection in vitro. Method:The technique of cell culture and MTT with ribavirin as positive control to observe anti-reispiratory syncytial virus effect. Results:This drug was found to be a potential inhibitor of RSV in a concentration-dependent manner with the median toxic concentration(TC50) of 3.972 g/L in Hep-2 Cell culture. The median effective concentration(EC50) was 0.428 g/L-1 when drug was added 24 h before infection. The EC50 was 1.160 g/L when drug was added 2 h after virus infection,and it was 1.189 g/L as the drug was added after it mixed with virus 24 h. The therapeutic index(TI) was 9.28,3.42 and 3.34 respectively. Conclusion:Reduning injection has the direct deactivation and blocking effect on RSV. The preventive effect is the most evident in three drug deliveries drug with the same concentration in virto.

    • Expressions and significance of PTTG and PCNA protein in glioma

      Abstract:Objective:To investigate the expressions and their relationship of pituitary tumor transforming gene(PTTG) and proliferating cell nuclear antigen(PCNA) in glioma. Methods:The protein expression of PTTG and PCNA were detected by immunostaining assay using Streptavidin-Peroxidase(SP) mothed in 80 cases of glioma. Results:The positive rate of PTTG in gradeⅠ~Ⅳ were 56.3%,68.2%,80.8%,100.0% respectively. The protein expression of PTTG increased with the increasing of the pathological grade(χ2 = 9.602,P < 0.05). The positive rate of PCNA protein were 37.5%,54.5%,69.2%,93.8% respectively,and the protein expression of PCNA increased with the increasing of the pathological grade(χ2 = 12.147,P < 0.01). The expressions of PTTG have positive correlation with the expression of PCNA protein(γs = 0.557,P < 0.01). Conclusion:The expressions of PTTG and PCNA protein were related to maligant degree of glioma, and might cooperate with eath other in the tumor genesis and progression. They should be considered as the indicators of the biological behavior in glioma.

    • Characteristics of the pelvis osteolysis and its treatment in cementless total hip arthroplasty

      Abstract:Objective:To analyze the specific features of the pelvis osteolysis in cementless total hip arthroplasty and observe the differences between the pre-operative radiological evaluation and the true situation during operation. Methods:Six cementless hips of acetabulum loosening were revised in 2005. The osteolysis and defects were classified according to the method of Paprosky. Results:During the operations 1 case was type Ⅰ defects according to Paprosky which was evaluated non-osteolysis rediographically pre-operation. 3 cases were type Ⅱ defects which were diagnosed type Ⅰ pre-operation,and 2 cases of Ⅲ were diagnosed type Ⅱ pre-operation. 3 hips of type Ⅱ were revised with impacted bone graft and implantation of non-cement cups. 2 hips of type Ⅲ were revised with impacted bone graft and fixed with roof reinforced rings and cemented cups. 1 hip of type Ⅰ was revised with the fixation of a cemented cup within the original metal shell. All the patients were followed for 1 year radiographically and functionally. Conclusion:After cementless total hip arthroplasty,the osteolysis and bone defects in the acetabulum cannot be evaluated accurate radiography. More accurate methods should be used before operation,such as CT.
