Abstract:Objective:To construct rat Pik3cb(phosphatidylinositol 3-kinase,catalytic,beta polypeptide) shRNA eukaryon plasmid express vector, and test its downregulating effect on Pik3cb mRNA expression for the application of RNA interference in restraining vein graft restenosis. Methods:Two shRNA of rat Pik3cb were design and synthesize according to the sequence of Pik3cb in the Genbank, annealed to form double strands and then cloned into pGenesil-1,and then the sequences were examed. After shRNAs were transfected into rat thoracic VSMC through METAFECTENETM,the transfection rate was identified by EGFP. pAkt-473 protein expression was detected by Western blot, and cell apoptosis were obtained through flow-cytometry. Results:The two sequences of synthesized shRNA named pU6-Pik3cb-shRNA-1 and pU6-Pik3cb-shRNA-2 were exactly the same with the design. The 48 h and 72 h transfection rate were 15.7% and 10.1%,respectively. The pAkt-473 protein expression significantly reduced, while the apoptosis cells were more significantly increased in the shRNA groups than the control groups(P < 0.05). The scramble shRNA did not interfere with the VSMC proliferation. Conclusion:Rat Pik3cb shRNA eukaryon express plasmid vectors were constructed successfully and effectively inhibited the proliferation of rat smooth mouse cells.

Abstract:Objective:To detect the expression of PD-1 on allo-recipients’ PBMCs and T cells administrated with immunosuppressant and investigate the significance of positive cooperativity of PD-1 mAb combined with CsA on T cells. Methods:Double and single labeled FCM was used to detect the expression of PD-1 on PBMCs and T cells of patients. The mixed lymphocytes assay(MLA) was used to establish a model of the transplant rejection in vitro. MTT was used to detect the cooperativity of PD-1 mAb and CsA. Results:Compared with normal person,PD-1 was highly expressed on T cells in allo-recipients who administrated with immunosuppressant. In mixed lymphocytes assay, the expression of PD-1 seemed to be induced by CSA,and PD-1 mAb plus CsA displayed a positive cooperativity, which would owe to the low intensity of costimulatory signal. Conclusion:PD-1 might be used as a new target of immune interference of allograft survival.

Abstract:Objective:To investigate the association between Klotho gene polymorphisms and the risk of gastric cancer in Jiangsu population. Methods:In a case-control study of 151 patients and 145 controls,we genotyped G-395A in the promoter region and C1818T in exon 4 of Klotho gene by PCR-restriction fragment length polymorphism(PCR-RFLP) methods. The association between the genetic polymorphisms and the risk of the cancer was further examined by multivariate analysis. Results:The frequency of the A allele of G-395A polymorphism in patients(39.7%) was significantly higher than that in controls(28.2%,P = 0.038), with the adjusted OR=1.66 and 95℅CI 1.02~2.72,which was more pronounced among subgroups of youger males, smokers and drinkers. However, the frequency of C1818T did not show any significant difference between the patients and control subjects(P = 0.80). The two polymorphisms were in linkage disequilibrium(D = 0.449,r = 0.187) and in compliance with Hardy-Weniberg equilibrium in control. None of the haplotypes were significantly associated with the presence of the gastric cancer. Conclusion:The G-395A polymorphism in Klotho gene may positively contribute to the etiology of gastric cancer in Jiangsu population.

Abstract:Objective:To explore the synergistic anti-tumor effect of alcohol extract of verbena officinalis(EVO) and paclitaxel (PTX). Methods:S180 cells were cultured in vitro, and the growth inhibition effects of the two drugs used respectively and in combination were detected by MTT methods. S180 transplant model was established in male ICR mice, and the tumor growth inhibition effects of the two drugs were calculated by measuring tumor volume in every two days. Results:In vitro experiment showed that cell growth was significantly inhibited by PTX when EVO was added concomitantly at a dose of 20 mg/ml. Moreover,in vivo experiment demonstrated that EVO(1 g/kg) efficiently enhanced the anti-tumor effect of PTX at a dose of 10 mg/kg. Conclusion:A subclinical dose of EVO could greatly improve the anti-cancer effect of paclitaxel, which provide a new strategy to amplify the efficiency of chemotherapeutics without increasing its dose.

Abstract:Objective:To investigate the effects of a novel extracorporeal blood purification therapy-coupled plasma filtration adsorption(CPFA) on the function of monocytes in the multiple organ dysfunction syndromes(MODS) patients with severe infection. Methods:This study was a pospective, pilot, crossover clinical trial. A total of seven patients diagnosed MODS with severe infection were selected in this study. Patients were randomly allocated to both 10 hrs of coupled plasma filtration adsorption plus hemodialysis(CPFA,treatment A) and 10 hrs of high volume hemo- filtration(HVHF,treatment B) with 12 h interval in random order, that was to say each patient receiving A+B or B+A randomly. Plasma monocyte HLA-DR expression and leukocyte TNF--琢 production(both spontaneous and lipopolysaccharide stimulating) were tested before and after the treatment. TNF--琢 production from normal human monocytes(THP-1 cell) incubated with patients’ plasma obtained at 0,5,10 h of the treatment was also measured. Results:①The blood monocyte HLA-DR expression(%) increased after CPFA(84.32 ± 4.63 vs. 73.65 ± 11.52,P = 0.037) but no change after HVHV(P > 0.05).②Spontaneous and lipopolysaccharide inducing TNF--琢 production from patients’ whole blood increased over time with CPFA(P < 0.05). This increase was negative in HVHF group(P > 0.05). ③Patients’ serum suppressed production of TNF-a by cultured monocytes(THP-1). This suppression effect decreased over time with CPFA(P < 0.05). However,no significant difference was seen in HVHF group(P > 0.05). Conclusion:CPFA was superior to HVHF in improving the antigen presentation ability of monocyte and restoring leukocyte responsiveness to lipopolysaccharide. These findings suggest a potential role for CPFA in the treatment of MODS.

Abstract:Objective:To assess the value of tumor markers(TMs) protein chip diagnostic system C12 in the diagnosis of gastrointestinal cancer(GIC). Methods:The sera of 329 pathologically-confirmed GIC patients were detected by the C12 protein biochip system. The most relevant TMs and the contribution of various combinations of TMs to the improvement of diagnosis were determined. Results:The diagnostic rates of C12 biochip system in 329 GIC patients were 13.73%,33.33%,38.30%,58.03% for stagesⅠ,Ⅱ,Ⅲ and Ⅳ patients, respectively, and the overall diagnostic rate was 39.21%. There were statistically significant difference(P < 0.01) among all clinical stages(contingency table chi-square test), stage Ⅰversus stage Ⅲ,stage Ⅰversus stage Ⅳ,and stage Ⅱ versus stage Ⅳ. The other comparisons didn’t have statistical significance(P > 0.05). Among all the 12 TMs,the top three positive rates of 27.36%,19.76% and 19.45% were obtained from CEA,CA242 and CA19-9,respectively,which were correlated with the stage of GIC. Conclusion:The C12 biochip diagnostic system has some value in the diagnosis of advanced GIC,but its sensitivity for early stage is poor. The miniature and effective diagnostic system is needed in clinic urgently.

Abstract:Objective:To analyze the CT indicating features of hepatic high perfusion disorders at the hepatic arterial phase during the MSCT 3-phases liver scanning and the relationship with fundamental diseases, and discuss the mechanism. Methods:One thousand one hundred and forty-eight cases were conducted with CT plain scanning at the hepatobiliary region. After the contrast injection at the elbow vein, the 3-phases scanning,i.e. arterial, portal venous and balance phase, was conducted at the time of 25 s,60 s and 180 s, respectively. Results:One hundred and fifty cases of hepatic high perfusion disorders were found at the arterial phase and the abnormal shape of wedge, triangle, block or irregular of uniform high density. Most of high perfusion disorders appeared at the edge of the liver. Part of them was still shown slightly high density at the portal venous phase, and they appeared equal-density at the balance phase. Conclusion:MSCT 3-phases enhancement scanning is the best method for studying the hepatic high perfusion disorders at present. Abstract:Objective:To know about the knowledge of migrant workers in Jiangsu area about AIDS,including the basic knowledge, means of transmission, and its clinical symptoms, and analyze the influencing factors on their knowledge. Methods:1668 migrant workers in the factories and construction sites were investigated in Nanjing and Kunshan of Jiangsu Province, and the univariate analysis and multivariate logistic regression analysis were used to explore the influencing factors on their situation of knowledge. Results:In general, the migrant workers in Jiangsu area lack knowledge about AIDS. Most of them know little about the means of non-aids transmission and the clinical symptoms of AIDS in particular, and the percentages of workers knowing about the two aspects are 26.62% and 27.27%, respectively. The percentage of women having knowledge about AIDS is lower than that of men,and workers with higher educational level know more about AIDS. Conclusion:AIDS health education should be carried out urgently among migrant workers to improve their knowledge about it.

Abstract:Objective:To collect the data of drug resistance to evaluate the prevalence trend of 10 anti-tuberculosis drugs commonly. Methods:The study objectives were from 2001 to 2006. Susceptibility tests(absolute concentration methods for INH,SM,RFP,EMB,TH1321,PAS,AN,KM,OFLX and CPM) were done in all specimens. Results:1800 tuberculosis cases involved in the study. The culture positive rate was 70%. The rate of the tuberculosis anti-multidrugs was 54.9%. Conclusion:The rate of the tuberculosis mutidrug resistant is very different. Some of drug’s resistance trend were rising yearly. The anti-tuberculosis drug resistant surveillance were established. The reliable data on the status of anti-tuberculosis drug resistance results provided the evidences for improvement and assessment on TB control and clinical drug use.

Abstract:Objective:To investigate the distribution of clinical fungus infection and the analysis of antibiotic resistance. Methods:Fungi were identified with bioMérieux Inc’s ATB Expression microbe identification system,ID32C strip and CHROMagar chromogenic substrate. Drug susceptibility test was executed with ATB Fungus drug susceptibility plate. Results:Candida albicans, candida tropicalis and candida glabrata are the main microorganism at clinical fungal infections; and they are distributed mainly among respiratory ward, cardiovascular ward, intensive care unit and infective ward; fungi show drug susceptibility to amphotericin B and 5-flucytosin and drug resistance against fluconazole to some extent. Conclusion:Clinic suspect samples should be tested in time, and antifungal drugs should be applied on the basis of drug susceptibility test to minimize the incidents of deep fungal infections.
