    • Biocompatibility of Nitinol slices designed for the closure of CHD covalently coated by chitosan/recombinant-hirudin in vitro

      Abstract:Objective:To assess the effect of bioactive coatings on biocompatibility of flat Nitinol slices designed for the closure of congenital heart defects in vitro. Methods:①The experiment was divided into six groups: control group, uncoating group, chitosan group(Chi group), chitosan/r-hirudin(Chi/r-Hir) low-dose group, Chi/r-Hir middle-dose group and Chi/r-Hir high-dose group. ②The hemocompatibility of biomaterials was evaluated in vitro in a dynamic model with whole human blood by the hemolysis test and anticoagulant activity such as partial thrombin time(APTT), prothrombin time(PT) and thrombin time(TT) were measured as well. Surface characterizations such as protein adsorption, platelet adhesion and aggregation were observed by scanning electron microscopy. ③Humanumbilical vein endothelial cells(HUVECs) were cultured in the presence of tested slices for 72 h. Adhesion and proliferation of HUVECs were evaluated with light microscopy and fluorescence microscope by double-staining of fibronectin(Fn) and Ki67. Results:①The hemolysis rates of biomaterials were less than 1.7%. ②White blood count(WBC), red blood count(RBC) and platelet count (PC) were normal and no difference was observered between each groups. ③Chi/r-Hir significantly prolong the APTT and TT than those of other groups(P < 0.001). the protein adsorption, platelet adhesion and aggregation were reduced by Chi/r-Hir coated. furthermore, there was dose-effect relationship of differential concentration Chi/r-Hir coated(Dunnetts, P < 0.05). ④After 72 h of incubation, without abnormal morphology, HUVECs were able to grow on to all kinds of slices. The adhesion of the cells on the slices by fluorescence microscopy through the application of Fn immunofluorescent antibody labeled HUMECs decreased as following:Chi>Uncoating>Chi/r-Hir coating. The proliferation of the cells on the slices by fluorescence microscopy through the application of Ki67 immunofluorescent antibody labeled HUMECs decreased as following:Chi=uncoating>Chi/r-hir coating. The cells grown on the surfaces of Chi/r-Hir were not as good as on the Nitinols slices. Conclusion:In vitro, Chi/r-Hir coated with nitinol slices present good anti-coagulation, but adhesion and proliferation of HUVEC is not beneficial.

      Abstract:Objective:To construct the recombinant pEGFP-N1/hIL-1ra and test its transient expression in human gingival fibroblast cells, this may be useful of the treatment of periodontitis. Methods:Total RNA was extracted from human mammary cancer tissue. RT-PCR was performed to amplify the hIL-1ra encoding gene. Then PCR product was purified and cloned into pMD18-T. After DNA sequencing,both of the recombinant vector pMD18-T/hIL-1ra and plasmid pEGFP-N1 were digested with HindⅢ and BamHⅠ respectively. IL-1ra gene fragment was ligated into plasmid pEGFP-N1. The recombinant plasmid DNA was transformed into E.coli competent cells TOP 10 and positive clones were selected and tested. After being transfected by pEGFP-N1/hIL-1ra,the transient expression of IL-1ra gene in human gingival fibroblast(HGF) cells were detected by fluorescence microscope and RT-PCR. Results:hIL-1ra encoding gene fragment was 531 bp and was inserted into the eukaryotic expression vector pEGFP-N1/hIL-1ra correctly. And HGF cells,which were identified by both fluorescence microscope and RT-PCR,had a transient expression of IL-1ra after transfection. Conclusion:The new recombinant expression vector pEGFP-N1/hIL-1ra was constructed successfully and the HGF cells which could express IL-1ra transiently were also successfully made.

      Abstract:Objective:To construct E.coli expression vector of human spasmolytic polypeptide(hSP),express recombinant hSP and underlie the base of function analyses. Methods:The hSP gene encoding mature peptide was amplified by reverse transcriptase polymerase chain reaction,and then inserted into the expression vector pET32a. Recombinant plasmid pET32a-hSP was transformed into the E.coli Origami B(DE3). Recombinant proteins were assayed with SDS-PAGE and Western blot. Results:It was proved that the fragment amplified was inserted into the expression vector pET32a correctly by PCR and gene sequencing. Tricine SDS-PAGE analyse proved that the molecular weight of hITF was about 32 kD and Western blot demonstrated that the expression proteins have much good antigenicity and specificity. Conclusion:hSP E.coli expression vector was successfully constructed and recombinant hSP was expressed. This research lay foundations for the further function studying of hSP.

      Abstract:Objective:The purpose of this study was to detect the signaling pathways involved in angiotensinⅡ(AngⅡ)-induced mesangial cell(MC) proliferation. Methods:The incorporation of 3H-thymidine(3H-TdR) and cell count were used as the measure of mesangial cell proliferation. Reactive oxygen species(ROS) production was determined by DCFDA fluorescence. Nicotinamide adenine dinucleotide phosphate(NADPH) oxidase activity was examined by lucigenin chemiluminescence. c-Jun animoterminal kinase(JNK) activation was assayed by Western blot. Results:AngⅡ time-dependently and dose-dependently increased intracellular ROS production in cultured human MCs as early as 3 min and peak at 60 min. Incubation with different dose of AngⅡ(1 nmol/L,10 nmol/L, and 100 nmol/L AngⅡ) for 60 min, ROS production increased for 1.82-, 2.92-, and 4.08-fold, respectively. AngⅡ-induced ROS generation was inhibited by the AT1R antagonist losartan but not the AT2R antagonist PD123319, as well as NADPH oxidase inhibitors diphenyleneiodonium sulfate(DPI) and apocynin. In contrast, inhibitors of other oxidant-producing enzymes, including rotenone, allopurinol, indomethacin, nordihydroguiaretic acid, ketoconazole and G-nitro-L-arginine methyl ester(L-NAME) have no effect on AngⅡ-induced ROS production and MC proliferation. AngⅡ treatment induced translocation of cytosolic of p47phox and p67phox to the membrane and p47phox and p67phox gene expression. Conclusion:NADPH oxidase-derived ROS involved in AngⅡ-induced JNK/AP-1 activation and mesangial cell proliferation.

      Abstract:Objective:To investigate the expression of melatonin receptors(MT1,MT2) in BMSCs from adolescent idiopathic scoliosis patients. Methods:Twenty-eight volunteers were divided into two groups:AIS group consisted of 18 patients and control group consisted of 10 subjects. The human bone marrow anticoagulated by heparin was obtained from anterior superior iliac spine,and the BMSCs were isolated by density gradient centrifuge from the mononuclear cells,and then were cultivated and serial subcultivated in vitro. P3 cultures were analyzed by the flow cytometry to determine the surface antigens,and expressive intensity of melatonin receptors(MT1,MT2)was detected by RT-PCR. Results:Mononuclear cells were cultivated and subcultivated to P3 culture in vitro,which were analyzed by the flow cytometry,and demonstrated that the expanded mononuclear cells expressed mesenchymal cell markers. There was no statistical difference of melatonin receptors expression in BMSCs between AIS group and control(P > 0.05). Conclusion:The expression of melatonin receptors in BMSCs were comparable between AIS patients and controls,which indicated that melatonin receptors would not involved in the pathogenesis of AIS in the stem cell level.

    • Effect of rosiglitazone on the expression of HO-1 and cell survival of HUVECs cultured in high glucose condition

      Abstract:Objective:To investigate the effect of rosiglitazone on the expression of heme oxygenase-1 and cell survival in human umbilical vein endothelial cells cultured in high glucose condition. Methods:HUVECs in vitro were treated with rosiglitazone at different concentrations(ranging from 0 to 30 μmol/L) for 48 h or exposed to10 μmol/L rosiglitazone for 0-24,48 and 72 h,respectively. Gene expression was analyzed at mRNA level by RT-PCR or at protein level by Western blot. The trypan-blue exclusion test was used to study the influence of rosiglitazone on cell survial. Results:Compared to the control groups,the expression of HO-1 was induced by rosiglitazone in a dose and time-dependent manner(P < 0.05),and it could make the cell survival increase(P < 0.05). Conclusion:Rosiglitazone could induce the expression of HO-1 gene in HUVECs cultured in high glucose condition and make cell death atc of endothelial cells decrease. This indicated that rosiglitazone could elevate the capability of the cells counteracting the oxidative stress in high glucose condition,which could protect the vascular function.

    • Inhibitory effects of As2O3 and Aspirin on cell proliferation in human gastric carcinoma cell line SGC-7901

      Abstract:Objective:To investigate the inhibitory effects of As2O3 and Aspirin on cell proliferation in human gastric carcinoma cells(SGC-7901 cell lines). Methods:The inhibition of proliferation,apoptosis and cell cycle of gastric cancer cells SGC-7901 were detected by MTT assay and the flow cytometry and light microscope. Results:①After 72 hours,MTT assay showed the growth of the SGC-7901 cells was inhibited by As2O3(4 μmol/L), Aspirin(2 mmol/L),and As2O3+Aspirin(2 μmol/L + 1 mmol/L). And the inhibitory rates were 31.04%,16.11% and 24.19% respectively. It also showed the inhibitory effects among negative control group,two single drug groups(As2O3 group,Aspirin group) and drug combination group(P < 0.05). ②Flow cytometry showed the apoptosis rates were 1.55% in negative group,1.81% in As2O3 group,1.64% in Aspirin group and 19.01% in As2O3+Aspirin group,which showed statistical significance(P < 0.05). ③The proportion of cells at G0/G1 phase was significantly decreased while the proportion of cells at G2/M phase was increased in As2O3 group. The proportion of cells at G0/G1 phase was significantly increased while the proportion of cells at S phase and G2/M phase were decreased in Aspirin group. And the increasing of proportion of cells at G0/G1 phase and G2/M phase were found in drug combination group,while proportion of cells at S phase decreased. Conclusion:As2O3 and Aspirin could inhibit the growth of gastric cancer cell SGC-7901 by inducing apoptosis and cell cycle arrest. It showed that inhibitory effect on proliferation of SGC-7901 cell line in drug combination group(As2O3+Aspirin) were markedly enhanced and their dose was decreased.

    • Role of Bad in anti-apoptotic mechanism of insulin during post-ischemia reperfusion myocardium injury in rats

      Abstract:Objective:To observe anti-apoptotic effect of insulin,and explore the role of Bad in anti-apoptotic mechanism of insulin during post-ischemia reperfusion injury in myocardium of rats. Methods:Forty-six male Sprague-Dawley rats were randomly divided into 4 groups:①I/R group was subjected to ischemia for 30 min and reperfusion for 180 min;②INS group was subjected to intravenous infusion of Insulin(60 U/L,4 ml/kg·h) at 5 min before 3 h of reperfusion;③INS+LY group was subjected to intravenous infusion of LY294002(0.3 mg/kg) at 10 min before 3 h of reperfusion and intravenous infusion of insulin and LY294002(30 -滋g·kg-1·h-1) at 5 min before 3 h of reperfusion;④SHAM group was subjected to placing a silk around the left anterior descending coronary artery without a slipknot. At the end of 4 h of reperfusion,apoptosis of cardiomyocytes were assessed through TUNEL and DNA Ladder;Immunochemistry and Western-blotting were used to analyse the expression of phosphorylated Bad in all groups. Results:①Compared with I/R group,apoptosis index(AI) were significantly decreased,and expression of phosphorylated Bad was prominently higher in INS group. ②There were a remarkably increase in AI and lower expression of phosphorylated Bad in INS+LY group than that in INS group. Conclusion:①Ischemia and reperfusion resulted in lower expression of phosphorylated Bad in myocardium of rats;②Insulin exerted an anti-apoptotic effect in myocardium of rats during ischemia and reperfusion;③its mechanism may be involved in higher expression of phosphorylated Bad via PI3-K/Akt pathway.

    • Effects of Trichostatin A on hTERT-mRNA expression and proliferation in human anaplastic thyroid carcinoma cell

      Abstract:Objective:To investigate the effect of Trichostatin A on the growth of human anaplastic thyroid carcinoma cell line in vitro. Methods:Human anaplastic thyroid carcinoma DRO cell line was treated with Trichostatin A of different concentrations and time. The antiproliferative effect was measured by methyl thiazolyl tetrazolium(MTT). Flow cytometry was used to examine apoptosis and cell cycle when TSA was 1.0 -滋mol/L in concentration and the exposure to DRO was 24 h,48 h and 72 h,respectively. The leves of hTERT-mRNA,p21-mRNA in cells were detected by RT-PCR,with TSA for 48 h. Results:After being treated with 0.1 -滋mol/L,0.2 -滋mol/L,0.5 -滋mol/L,1.0 -滋mol/L,and 1.5 -滋mol/L TSA,the prolifergation of DRO cells was significantly inhibited. IPCM showed that there were obvious morphologic changes after TSA treatment. Changes in the cells cycle occurred 48 h after treatment. hTERT expression was down-regulated after treatment with TSA. Conclusion:TSA significantly inhibited growth of the DRO cells and mechanism may be related to a down-regulation in hTERT expression.

    • Effect of rosiglitazone on C reactive protein induced growth,apoptosis and the expression of nuclear factor kappa B in human umbilical vein endothelial cells

      Abstract:Objective:To investigate the effect of rosiglitazone(RSG)on C reactive protein(CRP) induced growth,apoptosis and the expression of nuclear factor kappa B(NF-κB) in human umbilical vein endothelial cells(HUVECs),and explore the roles of rosiglitazone in the mechanism of anti-atherosclerosis. Methods:HUVECs cultured at the 5th generation were divided into eight groups:Normal control group(NG),CRP 5 μg/L group(CRP 5),CRP 20 μg/L group(CRP 20),CRP 100 μg/L group(CRP 100),RSG control group(RSG),RSG and CRP 5 group,RSG and CRP 20 group,RSG and CRP 100 group. Cells were intervened by RSG(5 μmol/L) for 12,24 and 48 hours. RT-PCR was used to detect the expression of NF-κB. The growth and apoptosis of HUVECs were also observed. Results:The expression of NF-κB and the apoptosis were obviously higher in the each mass concentration CRP group than that in the NG(P < 0.05), more obviously when the concentration was 100 μg/L(P < 0.05),and the growth change was opposite. The mRNA expression level of NF-κB and the apoptosis in cells treated by RSG were lower significantly than that of control group(P < 0.05),the growth was higher than that of control group,and it was significantly higher with the increase of the time. Conclusion:CRP promoted the apoptosis and inhibited the growth of Certain density CRP can induce the expression of NF-κB,which indicated that CRP can activate the inflammation response, and play a vital role in the pathology mechanism at coronal AS. Rosiglitazone can effectively reduce the inflammation effect of CRP thus to delay the occurrence of the AS.

    • The neuroprotective effects of MK-801 on Aβ42 transgenic drosophila model of Alzheimer’s disease

      Abstract:Objective:To explore the neuroprotective effects of the N-methyl-D-aspartate(NMDA) receptor antagonist MK-801 on Aβ42 transgenic drosophila model of Alzheimer’s disease. Methods:Drosophila was designed into four groups:normal control,normal drosophila exposed to MK-801,Aβ42 transgenic drosophila and Aβ42 transgenic drosophila exposed to MK-801. And the olfactory associative learning assay,climbing assay and survival assay were performed in four groups of drosophila. Results:Compared with Aβ42 transgenic drosophila group,the associative learning ability,climbing ability and life span of Aβ42 transgenic drosophila applied with MK-801 group were significantly improved. Conclusion:In Aβ42 transgenic drosophila model of Alzheimer’s disease,MK-801 can reduce the neurotoxic effect induced by Aβ42.

    • An experimental study on treatment of early osteoarthritis with BMSCs transplantation

      Abstract:Objective:To analyze the effects of BMSCs transplantation on early osteoarthritis in rabbits. Methods:Seventy-two New Zealand white rabbits were randomly and equally divided into normal control group, early osteoarthritis control group and BMSCs transplantation group. Injection of BMSCs was given to the rabbits in the transplantation group on the fourth week after arthritis induction. The general information of animals,serum TNF-α and IL-1β levels,MRI,gross morphological and pathohistological changes were observed as well as Mankin scores. Results:Compared with the early osteoarthritis group,the rabbits of BMSCs group had a better gait;the serum TNF-αand IL-1β levels were decreased;MRI showed the diminution of low signal area,the thickness of articular cartilage got close to normal and without arthroedema; The surface of articular cartilage became evenness by gross morphology but the luster below the mark;histopathology indicated hyalo-chondrogenesis and to line up in order with equal thickness. Conclusion:BMSCs can induced into chondrocytes in articular cavity and be effective for early osteoarthritis in rabbits.

    • The effects of topiramate and sodium valproate retard tablet on cognitive function in adult with epilepsy

      Abstract:Objective:To evaluate the effects of topiramate(TPM) and sodium valproate retard tablet(VAP) on cognitive function in adult patients with epilepsy. Methods:Sixty-two adult patients,newly diagnosed as epilepsy,were randomly divided into 2 groups:TPM Group(n = 30) and VAP Group(n = 32). The cognitive function of all the patients in these two groups were evaluated by the Revised Adult Wechsler memory scale(WMS-RC) and event related potentials(ERPs) before treatment and after two months administration of TPM/VAP. Results:①Before administration of TPM:Memory Quotient(MQ) < 100,accounting for 26.67%;In the ERPs,the P3 wave latency >359.9ms,accounting for 33.33%. Before administration of VAP:MQ < 100,accounting for 28.0%;In the ERPs,the P3 wave latency > 359.9ms,accounting for 31.25%. There was not significant difference between these two groups with respect to the cognitive function(P > 0.05). ②After administration of TPM,in TPM Group the score of sequence counts of 1 to 100 declined(P < 0.01),the score of sequence counts of 100 to1 declined(P < 0.05), the score of digit cumulate cognitive function declined(P < 0.05),the mistakes of cards recognition increased(P < 0.01), the function of cards remembrance declined(P <0.01), the mistakes of cards reclamation increased(P < 0.01),the mistakes of word association increased(P < 0.01), the object touch was more difficult(P < 0.01), the function of story apprehension declined(P < 0.01)and the natural order and anti- natural order repeat of digit were more difficult(P < 0.01). In the ERPs, the N1 wave latency prolonged(P < 0.05) and the P2,N2 and P3 waves latencies all prolonged significantly(P < 0.01),while the amplitude of P3 wave declined(P < 0.01). After administration of VAP,in VAP Group:the score of WMS-RC TEST in every item did not change significantly(P > 0.05). In the ERPs,the N1 wave latency shortened(P < 0.05) and the P2,N2 and P3 waves latencies all shortened significantly(P < 0.01),while the amplitude of P3 wave did not increase significantly(P > 0.05). ③The transient memory of WMS-RC was reversely correlated with the P3 wave latency(P < 0.05). Conclusion:After administrateration of TPM,patients with epilepsy who were in the drug titration phase were more likely to suffer from many cognitive disturbers;Patients’ cognitions were improved by administration of VAP;After examination of ERPs and WMS-RC ,the results of evaluation of cognitive function will be more objective and comprehensive.

    • Evaluation of surface electrocardiogram clues for differential diagnosis of idiopathic left ventricular tachycardia and wide QRS complex supraventricular tachycardia

      Abstract:Objective:To evaluate the value of surface electrocardiogram clues for the differential diagnosis of idiopathic left ventricular tachycardia and wide QRS complex supraventricular tachycardia. Methods:Surface ECGs and clinical data of 75 patients with wide QRS complex tachycardia of right bundle branch block type and without organic heart disease were analyzed retrospectively, and the final diagnoses were made by intracardiac electrophysiological study. Results:The valuable surface ECG clues for the diagnosis of idiopathic left ventricular tachycardia were as follows:(1) evidence of atrioventricular dissociation, (2)evidence of capture or fusion beats, (3)axis of “no man’s land”(-90°~180°) or left axis deviation, (4)negative direction S wave is predominating in inferior wall leads(Ⅱ-Ⅲ and avF),(5)QRS morphology in lead Ⅰ was characterized biphasic Rs wave; QRS morphology in lead avL was Rs or qR or qRs;QRS morphology in lead V6 was characterized biphasic rS wave;(6)it occurs transient reversible T change during sinus rhythm after tachycardia termination. And the valuable surface ECG clues for the diagnosis of superventricular tachycardia with wide QRS complex were as follows: (1)normal axis or right axis deviation, (2)positive direction R wave is predominating in inferior wall leads(Ⅱ-Ⅲ and avF); QRS morphology in lead avL was biphasic rS or QS;(3)QRS morphology in lead V6 was characterized biphasic Rs wave. Conclusion:Surface ECG clues are valuable for differential diagnosis of wide QRS complex tachycardia of right bundle branch block type and without organic heart disease. Several features are extracted in this text for differential diagnosis idiopathic left ventricular tachycardia and supraventricular tachycardia of wide QRS complex.

    • Investigation of prognosis relationship between anti-cardiolipin antibodies?D-Dimer and the patients with acute cerebral infarction

      Abstract:Objective:To explore relationship between anti-cardiolipin antibodies(ACA)-D-Dimer and acute cerebral infarction. Methods:The content of ACA-D-Dimer in blood serum of 187 acute cerebral infarction patients was detected in the morning after fasting over night using ELISA methods, and compared with 135 average healthy cases. Results:The ACA positive rate in acute cerebral infarction group was much higher than control and the difference was statistically significant(P < 0.01); The content of D-Dimer in ACA-positive group was significantly higher than that in ACA-negative group; As the cerebral infarction worsenly, the IgG-ACA positive rate became higher as well as the content of D-Dimer; ACA in the 1st,2nd,3rd and 4th week were compared, suggesting that ACA positive rate reduced as time changed(P < 0.01); ACA positive status was closely related with cerebral infarction patients who were 50 years old or younger. Conclusion:The dynamic changes of ACA-D-Dimer in blood serum might be helpful in predicting prognosis of acute cerebral infarction.

    • Prognostic value of cytoplasmic μ heavy chain in childhood immature B-cell acute lymphob- lastic leukemia

      Abstract:Objective:To investigate the prognostic value of cytoplasmic μ heavy chain(C-滋) in childhood immature B-cell acute lymphoblastic leukemia, in order to provide evidence for rational treatment protocol. Methods:From Jan 1, 2001 to March 31, 2005, 161 patients with immature B-cell acute lymphoblastic leukemia received ALL-XH-99 protocol treatment in our hospital. Baseline data were obtained for age, sex, peripheral white blood cell (WBC) count, immunophenotype, P170 and fusion gene. Absolute peripheral blood juvenile cell count of the eighth day of prednisone treatment, bone marrow smear of the eighteenth day of induction treatment, bone marrow minimal residual disease(MRD) after induction treatment, and risk classification were also assessed. Results:Kaplan-Meier analysis showed that the five-year event-free survival(EFS) rate of Cμ positive patients was 56.7 ± 0.88%, significantly lower than that of the Cμ negative patients(74.1 ± 0.05%,P < 0.01). χ2 analysis indicated that the prognostic difference was unrelated to biological characters such as sex, age, peripheral WBC count when first diagnosed, P170 level, myeloid marker (My), fusion gene(BCR/ABL or MLL/AF4), prednisone window trial and bone marrow smear of the nineteenth day of treatment. Cox regression risk analysis demonstrated that Cμ positive, WBC≥50×109/L before treatment, MRD positive and fusion gene positive were independent risk factors(P < 0.05). Conclusion:Cμ positive is an important risk factor of poor prognosis for childhood immture B-cell lymphoblastic leukemia, and it was independent of CD10 negative MLL gene rearrangements.

    • A clinical study of paclitaxel-based or oxaliplatin-based regimens in the treatment of advanced gastric cancer

      Abstract:Objective:To compare the efficacy,toxicity and safety of paclitaxel-based or oxaliplatin-based regimens in the treatment of patients with advanced gastric cancer. Methods:Eighty-four patients with advanced gastric cancinoma were divided into 2 groups. Forty-four patients enrolled in group A were given paclitaxel-cisplatin-fluorouracil triplet regimen and 40 patients in group B received oxaliplatin-fluorouracil couplet regimen. Each patient received chemotherapy for at least 2 cycles and was evaluated every 2 cycles. Results:Objective results in the Group A showed:ORR 50%,DCR 84.1%, mTTP 7.0 months,mOS 11.2 months,one-year survival 34.2% and in Group B were:ORR 50%,DCR 75%,mTTP 6.5 months,mOS 10.8 months,one-year survival 33.6%. There was no significant difference in ORR,DCR,mTTP,mOS and 1-year survival(P > 0.05) between two groups. The main toxicity in group A was bone marrow depression and alopecia. The main toxicity in group B was neurotoxicity. Conclusion:Both paclitaxel-based regimen and oxaliplatin-based regimen showed comparable and good efficacy in the treatment of advanced gastric cancinomas. The toxicities caused by the two regimens were mild and tolerable,respectively.

    • Adenosine triphosphate based tumour chemosensitivity assay in breast carcinoma of clinical research

      Abstract:Objective:To investigate the heterogeneity of chemosensitivity in breast carcinoma using adenosine triphosphate based tumour chemosensitivity assay(ATP-TCA) and the feasibility of individual chemotherapy. Methods:ATP-TCA was used to determine the effect of 6 single drugs in surgical specimens from 75 patients with breast carcinoma. And 60 patients with breast carcinoma were used as matched control group. Results:All above these specimens were detected successfully. The sensitivity of six drugs was 25.33% for THP,21.33% for E-ADM,20.00% for DTX,13.33% for MTX,12.00% for gemcitabine,6.67% for 5-Fu in these specimens respectively. There were the heterogeneity of the in vitro chemosensitivity in breast carcinoma and had statistical significance(P < 0.05). After three years followed up,the relapse rate of therapy group was lower than that of the matched control group with statistical significance(P < 0.05). Conclusion:The use of the ATP-TCA may provide a method of selecting appropriate anti-cancer drugs of breast carcinoma in chemotherapy of individualization.

      Abstract:Objective:To assess the infuence of preload on Tei index(including PW-Tei and TDI-Tei) to evaluate left ventricular function. Method:Twenty-seven patiens with PDA and 32 patients with VSD without other diseases were studied. PW-Tei,TDI-Tei,LVEF,Vp,LVEDD and LVESD were calculated before and after the surgical operation. Results:Either PW-Tei or TDI-Tei increased while the LVEF,LVEDD and Vp decreased signicantly after operation. Conclusion:Both the PW-Tei and the TDI-Tei were influenced by preload when they were used to evaluate the left ventricular function.

    • Study on relationship between CT staging and CT enhancement of gastric cancer and tumor angiogenesis

      Abstract:Objective:To study the relationship between CT Staging and CT enhancement of gastric cancer and tumor angiogenesis and provide useful information for clinic therapy. Methods:Enhanced two-stage spiral CT scanning was given before operation to 30 cases who suffered from gastric cancer and were confirmed pathologically after operation. The 30 cases include 24 men and 6 women (average age was 63). CT enhancement values of pathologic region were determined. Tumor specimens of 30 cases were collected after operation. Two-step CD 34 mono-clone antibody and VEGF mono-clone antibody immunological and histo-chemical staining were performed on corresponding research area. TNM and CT staging were also performed based on clinical,CT and pathological findings. Statistical research was carried out on CT enhancement values,CT staging,TNM staging,pathological grading and results of CD 34 and VEGF staining. Results:Conformity rate of TNM staging and CT staging of 30 cases of gastric cancer was 90%. CT enhancement value was positively correlated with TNM and CT staging. CT enhancement value was positively linearly correlated with MVD. Its equation was Y = 49.4125+0.5067X. Difference of CT enhancement values between VEGF high and low expression groups was significant(P < 0.05). There was no relation between pathological grading and CT enhancement value(P > 0.05). Conclusion:Staging can be made by CT presentation before operation. CT enhancement value can reflect tumor angiogenesis of gastric cancer. Pathological grading has no direct assistance in the study of CT enhancement value.

    • Application in flap design with ultrasonography and anatomic evaluation of supratrochlear artery

      Abstract:Objective:To study the character of supratrochlear artery(STA) with ultrasonography and anatomy and its application in designing forehead flap. Methods:A total of 30 STAs of 15 normal adults were detected by ultrasound and 12 STAs of 7 adult cadavers were dissected to study their calibers,courses and the relationship to the surrounding soft tissue. Results:Thirty STAs of normal adults and 12 STAs of cadaver controls were displayed well. The STA where it started in normal adults was(1.20 ± 0.17)mm in outer diameter and(1.40 ± 0.16)cm away from the midline of the face,and in adult cadaver controls was(1.28 ± 0.22)mm and(1.33 ± 0.14)cm. Twenty-six STAs in normal adults and 10 STAs in cadavers ascended inward. There was no statistial difference between the two groups(P > 0.05). Conclusion:The position of STA was permanent but some variation,which can be clearly displayed by ultrasonography. Ultrasonography is a good method to help design the forehead flap.

      Abstract:Objective:Breast-conserving therapy has been one of the important operation therapies for breast carcinoma in early stage. This article discussed the therapeutic manner and clinical prognosis of canonical breast-conserving therapy. Methods:41 patients with breast carcinoma,Ⅰstage with 17 cases,Ⅱ stage with 23 cases, Ⅲstage with 1 cases,who underwent canonical breast-conserving therapy, postoperative radiotherapy, chemotherapy , endocrinotherapy and targeted therapy were followed up ranging from 6 to 27 months and observed for the clinical curative effect. Results:up to now, the local recurrent rate and distant metastasis rate is zero,the patients are satisfied with their self-image. Conclusion:the canonical breast-conserving therapy is relative to the control of local recurrent rate-perfect hairdressing effect and the long-survival rate.

      Abstract:Objective:To study relationship between anxiety,depression and quality of life in patients with functional dyspepsia(FD). Methods:Total of 78 patients with FD(according to Rome Ⅲ criteria)were evaluated by Zung’s self-rating anxiety scales(SAS)and Zung’s self-rating depression scales(SDS) and Chinese version of the MOS 36-item short form healthy survey(SF-36). Results:The SDS and SAS scores were significantly higher in FD patients than those in Chinese normal. The SF-36 scores in all dimensions including physical functioning,role of physical,pain,vitality,role of emotional,mental health,social functioning and general health in FD patients were significantly lower than that in Chinese normal. The SF-36 scores in all dimensions had a negative association with SDS and SAS scores(P < 0.01). Conclusion:There was close relationship between anxiety,depression state and quality of life in FD patients.

      Abstract:Objective:To analyze the rate of HIV infection among drug users from Jiangsu in 2005~2008, understand the status of HIV infection and risk factors among drug users,and evaluate the effect of intervention on drug users. Methods:The data of surveillance among drug users in Jiangsu during 2005~2008 were analyzed by SPSS11.5. Results:Total of 7739 drug users were detected for HIV and HCV and the infection rate of HIV and HCV was 0.7% and 34.04%, respectively. The trend of injection rate among drug users was down from 2005 to 2008 and had significant difference that in(P < 0.01). The rate of HIV infection in sharing of syringes group was higher than the no-sharing group(P < 0.01). The trend of rate actives who never used condom in commercial sexual was down from 2005 to 2008(P < 0.01). In addition,HCV infection among drug users by injection was higher than that without injection(P < 0.01). The rate of HCV infection Compared with group, the rate of HIV infection in group was down from 2005 to 2008(P < 0.01). Conclusion:The risk really existed that bloodborne diseases,such as HIV and HCV,were transmitted and spread among drug users. It is necessary to enhance surveillance,education on the prevention of the diseases and behavioral intervention for drug users so as to reduce prevalence of bloodborne infectious diseases such as HIV and HCV.

    • Effect of the promoter polymorphism of tumor necrosis factor alpha 308 G/A gene on the susceptibility to ulcerative colitis:A meta-analysis

      Abstract:Objective:To investigate the association between the polymorphism of tumor necrosis factor alpha 308 gene(TNF-α-308) promoter and the risk of ulcerative colitis(UC) using the method of meta-analysis. Method:The database of PubMed, EMBase, and CNKI(Chinese), Wanfang(Chinese) were searched for published case-control studies of the association between the polymorphism of TNF-α-308 gene promoter and UC. Data were extracted using a standardized form and the meta-analysis was performed. Results:Fourteen eligible studies, 8 of Europeans, 2 of Americans, and 4 of Asians including 3 East Asians, were included in the meta-analysis. However, stratification by ethnicity indicated that there was significant association between TNF2 allele and UC in East Asians(OR=3.12, 95% CI 1.44~6.77,P = 0.004). Conversely, there was no association between TNF2 allele and UC patients from the European(OR 0.73,95% CI 0.41~1.31,P = 0.30). The OR for the TNF2/2+TNF2/1 genotype verse TNF1/1 genotype in samples overall and in each ethnic group showed a similar trend to those for the TNF2 allele. Conclusion:In East Asians, the TNF2 allele confers a significant risk for developing UC. There is no association between the polymorphism of TNF-α-308 gene promoter and UC in Europeans.

    • Analysis on implement effect of PPM-DOTS tuberculosis control project in Lianyungang city

      Abstract:Objective:To study implement effect of PPM-DOTS tuberculosis control project in city. Methods:The indexes of case-detection and cure rate were compared with before the project and after the project. Results:After the project finished, the new smear positive notification rate increased from 1.87/100 000 before the project to 37.38/100,000. After the project,and the smear detecting rate increased from 42.08% to 73.91%. The cure rate of new smear positive tuberculosis and the retreatment smear positive tuberculosis increased from 50.00% to 92.32% and from 55.55% to 86.49%. Conclusion:Through the PPM-DOTS implemented, the new smear positive detecting rate was increased obviously and the treatment of new smear positive pulmonary tuberculosis was improved in city areas.
