Abstract:Objective:To explore the regulative role of leptin on NYGGF4 gene expression in matured human adipocytes. Methods:Human preadipocytes were cultured in vitro and differentiated into matured adipocytes. The matured adipocytes were treated with 100 ng/ml leptin for 6 hours and 24 hours,respectively. Total RNA from these adipocytes was extracted and the levels of NYGGF4 mRNA expression were evaluated by real-time quantitative RT-PCR. Results:Treatment with 100 ng/ml leptin for 6 hours or 24 hours significantly decreased NYGGF4 gene mRNA expression in human matured adipocytes (P < 0.05). Conclusion:Leptin could downregulate the expression of NYGGF4 gene mRNA in human matured adipocytes.

Abstract:Objective:STAT(signal transducer and activators of transcription) is one kind of cytoplasm transcription factor which plays an important role in promoting malignant tumor to genesis,proliferation and apoptosis. This research is to approach the association the single nucleotide polymorphisms(SNP)in STAT3 with biological behavior of gastric cancer. Methods:Using SYBR GREEN PCR with Tm-shift primers to detect genotyping of the SNP in STAT3 in 93 gastric cancer patients. Results:SNP in STAT3 of 18 gastric cancer patients,which confirmed by gene sequencing are same as the results by SYBR GREEN PCR with Tm-shift primers. Stratified analysis indicated that an elevated risk of vessel invasion and nerve invasion was observed in subjects who carried at least one G allete (GG or CG) compared with CC genotype (P < 0.001). Conclusion:SYBR GREEN PCR with Tm-shift primers can be one of the methods to detect STAT3 SNP. SNP in STAT3 was associated with clinicobiological behaviors of gastric cancer,such as vessel invasion and nerve invasion. The expression of STAT3 G allele indicates high risk of vessel invasion and nerver invasion.

Abstract:Objective:To study the expression of TSPYL5 in gastric carcinoma and adjacent gastric tissues,and the effect of the expression of TSPYL5 on the biobehavion of gastric carcinoma. Methods:RT-PCR was used to detect the expression of TSPYL5 mRNA in 46 samples of human gastric cancer and normal gastric tissue,and the relationship between the expression of TSPYL5 and oncogenesis,lymph node metastasis,depth of tumor invasion and grade in gastric carcinoma was also analyzed. Meanwhile the effeet of TSPYL5 on AGS cells was analyzed by MTT in vivo. Results:Of 46 gastric cancer tumor tissues,26 tissues were detected the expression of TSPYL5 mRNA. The rate of TSPYL5 mRNA in the primary gastric cancer tissues was significantly higher than that in the adjacent non-cancerous tissues(56.5%). The TSPYL5 positive isolates had more lymph node metastases than that of TSPYL5 negative ones(P > 0.05). Furthermore,the expression of TSPYL5 mRNA in TNM and none metastasis signifcantly decreased(P < 0.05). TSPYL5 suppressed the growth of AGS cells. Conclusion:TSPYL5 may be had a significance effect on development and progress in gastric cancer.

Abstract:Objective:To investigate the role of PARP activation and AIF translocation in intestinal injury after ischemia-reperfusion (I/R). Methods:The intestinal ischemia/reperfusion(I/R) model in rats was established by clamping both the celiac trunk and the superior mesenteric artery for 30 min,followed by reperfusion for 2h. The animals were assigned to different groups:I/R group,Drug group(3-AB,PARP inhibitor,30 mg/kg i.v.) and Sham group. The morphological changes of intestine,PARP-1 activation,apoptosis and the expression of PARP-1 cleavage p85 and AIF were detected by means of HE,immunohistochemical staining for poly(ADP-ribose),TUNEL and Western blot respectively. Results:In I/R group,the extent of intestinal injure and PARP-1 activation,the expression of PARP-1 cleavage p85,AIF and TUNEL positive-staining were elevated significantly. In Drug group,these variables were statistically lower than those in I/R group,but higher than those of Sham group. Conclusion:These findings indicate that PARP activation and AIF translocation contribute to intestinal injury after ischemia-reperfusion;PARP-1 inhibitor has the potential protection for intestinal I/R by inhibiting PARP-1 activation and preventing AIF translocation.

Abstract:Objective:To study the therapeutic effects and security of combination of simvastatin with berberine on the patients with hyperlipemia. Methods:In clinical trial,a total of 99 patients with hyperlipidemia in our hospital were divided into 3 groups. Group A:SIMVA,Group B:BBR,Group C:combination of BBR with SIMVA. After 8 weeks of treatment,the Blood lipid,liver and kidney functions in the three groups were detected and compared. Rats fed the HFHC diet were randomly divided into 5 groups. They were the HFHC diet control group treated with saline (n=6),SIMVA at 6 mg/(kg·d)(n=9),BBR at 90 mg/(kg·d)(n=9),BBR (90 mg/[kg·d]) plus SIMVA(6 mg/[kg·d])(n=10),and SIMVA at 12 mg/(kg·d)(n=9),respectively for 30 days. Serum LDL-C,total cholesterol (TC),triglyceride (TG) and LDLR mRNA levels were assayed. Results:After 8 weeks of treatment,in the group of the patients with hyperlipemia,combination of BBR with SIMVA was more effective than the SIMVA or BBR monotherapy,markedly effective cases in the former group were 1-fold higher than that of SIMVA alone. The total effective rate of group C was 100%. Serum TC,TG were significantly different before and after simvastatin treatment(P < 0.01). Combination therapy reduced serum TC,TG and LDL-C,the levels of them were all significantly lower than these after simvastatin single-used in the hyperlipemia patients(P < 0.01). Comparing the three groups,in decreasing TC,TG,LDL-C,group C was more effective than group A (P < 0.01). Animal experiments piont out that the therapeutic effects of combination of half-simvastatin with berberine is correspond to entire amounts of simvastatin. We had not found any adverse effects in group C. Coadministration of berberine with simvastatin up-regulated LDLR mRNA to a level significantly higher than that of the monotherapies (P < 0.001). Conclusion:As compared with monotherapies,the combination shows an improved lipid-lowering effect with reduction of serum LDL cholesterol. Adding BBR to simvastatin could reduce the dose of simvastatin by at least 50%.The combination therapy display a new and effective regimen for hyperlipemia.

Abstract:Objective:To investigate and compare the differences on morphology and growth characteristics of mesenchymal stem cells(MSCs)derived between bone marrow of aplastic anemia(AA)patients,myelodysplastic syndrome(MDS)patients and normal individuals. Methods:MSCs were isolated from bone marrow sample of AA patients,MDS patients and normal individuals and cultured in vitro. The morphology and growth curves feature were observed. Results:MSCs derived from AA patients and normal individuals were similar in cell morphology. Both were fibroblast-like spindle. But MSCs from AA patients grew slower than those of the normal control group. The time of colony-forming of the MSCs from AA patients was longer. And the time required for the MSCs from AA patients reaching 90% confluency was also longer than that of the normal control group. The proliferation capacity of MSCs from AA patients at passage 3 was weaker than that of normal individuals. While,MDS patients and normal individuals derived MSCs were similar in cell morphology as well as the time of cell colony-forming and cell reaching 90% confluency. There was no difference in the proliferative ability of MSCs at passage 3 between MDS patients and normal control group. Conclusion:The morphology of MSCs showed a similar characteristics among the three groups. But compared with MSCs from normal individuals,MSCs from AA showed significant slower growth rate and weaker proliferative ability. Whereas,there were no significant differences between MSCs from MDS and those of normal control group in morphology,growth rate and proliferation capacity.

Abstract:Objective:To investigate the effect of exogenous SIRPα1 gene on the proliferation and apoptosis of MCF-7 and explore the molecular biology mechanism. Methods:SIRPα1 gene was transfected into SIRPα1-negative MCF-7 with lipofectamine. Cell were stimulated by EGF and the expression of JNK and pho-JNK were examined by Western blot. The proliferation activity was assessed by MTT and the apoptosis of MCF-7 was detected by TUNEL. Results:After SIRPα1 was transfected into MCF-7,there was no change of JNK protein expression but a remarkable decrease of p-JNK. In addition,the expression of SIRPα1 also reduced MCF-7 cell vitality and induced cell apoptosis. The intracellular part of SIRPα1 can be phosphorylated in response to a variety of growth factors. But there was no significant change of cell apoptosis after EGF stimulated MCF-7 which were transfected with exogenous SIRPα1 gene. Conclusion:SIRPα1 can induce cell apoptosis,inhibit cell proliferation in MCF-7 cell line and serve as a potential anti-oncogene.

Abstract:Objective:To explore the effects of the human neural stem cells(hNSCs) and human umbilical cord blood cells(hUCBCs)on the treatment of cerebral ischemic rats and the proliferation and differentiation status of these cells in the ischemic brain tissue of rats. Methods:HNSCs were separated from 10~13 weeks brains of spontaneous abortion human embryo and cultured;hUCBCs were gotten from the placentas of full-term babies;both hNSCs and hUCBCs were marked with BrdU(5 -滋mol/L) for two days. The middle cerebral artery occlusion(MCAO) rat models were made and the stem cells were injected into the tail vein one day later. The Neurological Severity Scores(NSS) tests were undertaken in all rats of the three groups at 0~35 days after injection. Immunohistochemical method was used to check the differentiation and migration of stem cells in vivo. Results:Neural stem cells from human embryonic brains had been successfully cultured,and found to form typical neurospheres in suspension. The hUCBCs had the capacity of proliferation in vitro. Twenty-one days later,the neurologic function of rats with injection recovered much better than those without injection,as evidenced by the NSS(P < 0.05). There was no statistical difference of NSS between the two injected groups at every time point before and after injection(P > 0.05). Within the brain tissue,BrdU positive cells were distributed throughout the damaged brain of recipient rats,the majority of these cells localized in the ischemic hemisphere,few of these cells were observed in the contralateral hemisphere(P < 0.05). Significantly more BrdU positive cells were found in the brain tissue at the 21,28 and 35 days points respectively than that at the 14th days after injection(P < 0.05). More BrdU positive cells were found in the rats received hNSCs the rats injected with hUCBCs. Nestin positive cells were found within the damaged brains at every time point. Among all the BrdU positive cells of hNSCs group,73.8% of these cells were positive for the astrocyte marker glial fibrillary acidic protein(GFAP),16.7% were positive for the CNPase and 9.5% expressed the neuronal markers neuron specific enolase(NSE). Among all the BrdU positive cells of hUCBCs group,74.5% of these cells were positive for GFAP,15.4% were positive for CNPase and 10.1% expressed NSE. There was no statistical difference between the two groups(P > 0.05). Conclusions:HNSCs and HUCBCs had multipotential differentiation properties in vivo and differentiated into three main types of neural cells by the influcence of the ischemic microenvironmental signals. Intravenous injection of hNSCs and hUCBCs could effectively improve the neurologic function of ischemic rats. Besides of hNSCs,hUCBCs injection may be an efficient method to treat ischemic cerebrovascular disease.

Abstract:Objective:To study the effect of JAK/STAT signaling pathway on extracellular release of high mobility group box 1 (HMGB1) in endotoxin-induced hepatocytes. Methods:HMGB1 concentration was determined in the culture medium of 100 μg/L lipopolysaccharide-induced BRL-3A hepatocytes by enzyme-linked immunosorbent assay (ELISA),and the change of HMGB1 mRNA expression was detected by RT-PCR method. The effects of JAK/STAT pathway inhibitors (tyrphostin AG 490 and fludarabine) with various concentrations on HMGB1 mRNA expression and extracellular release were investigated. Results:The level of HMGB1 mRNA expression and HMGB1 concentration in the culture medium significantly increased after BRL-3A hepatocytes induced by lipopolysaccharide for 24 hours(P < 0.01). 25 μmol/L tyrphostin AG 490 and 50 μmol/L fludarabine partly inhibited HMGB1 mRNA expression and extracellular release in lipopolysaccharide-induced BRL-3A hepatocytes(P < 0.05). Conclusion:Lipopolysaccharide induces hepatocytes to release HMGB1,which is related to intracellular JAK/STAT signaling pathway.

Abstract:Objective:To study the effect of X-ray on expression levels of X-ray repair cross-complementing gene2(XRCC2)and XRCC3 in lung cancer cells,and explore the effect of DNA homologous recombination repair mechanism in radiotherapy of lung cancer. Methods:Cell inhibition ratio was measured using MTT assay. The expression levels of XRCC2 mRNA and XRCC3 mRNA in lung cancer cell line A549 were measured by RFQ-PCR assay. Results:The rate of proliferation inhibiting of lung cancer cells increased in line with the prolong radiation time of X-ray(P < 0.05)and the increase of radiation dose(P < 0.05),but there was no difference between the groups of 16 Gy and 32 Gy(P=0.211). The expression levels of XRCC2 and XRCC3 mRNA in these lung cancer cells increased significantly after treated with X-ray,and then decreased. The expression levels of XRCC2 and XRCC3 mRNA peaked at 48 h after X-ray treatment(P < 0.05). The expression levels of XRCC2 and XRCC3 mRNA increased markedly with the increase of radiation dose(P < 0.05). Conclusion:X-ray has a fairly obvious function to decrease proliferation of lung cancer cell,and could increase the expression levels of XRCC2 and XRCC3 mRNA in lung cancer cells. The results indicate that DNA homologous recombination repair mechanism may play an important role in resistance to radiotherapy in lung cancer cell.

Abstract:Objective:To study the differentiation potential of human umbilical cord mesenchymal stem cells into osteogenic,adipogenic,myocardiogenic and endothelial cells. Methods:The human umbilical cord mesenchymal stem cells were cultured and differentiated in the osteogenic,adipogenic,cardiomyogenic and endothelial medium. At the end of the culture,the cells were stained with Von Kossa or Oil red-O solution to reveal osteogenic and adipogenic differentiation. Immunocytochemistry and immunofluorescence were performed to investigate differentiation of the cells to cardiomyocytes and endothelial cells. Results:The human umbilical cord mesenchymal stem cells were positive for Von Kossa staining and have the ability to accumulate different amounts of lipid vacuoles after cultured in osteogenic and adipogenic medium. Immunocytochemistry showed that differentiated cells were strongly stained with cardiac sarcomeric α-actin and cardiac myosin after 5-azacytidine treatment. Immunofluorescence studies confirmed their endothelial phenotype with expression of known endothelial cell markers including CD31 and CD34 after VEGF treatment. Conclusion:Our results demonstrated that human umbilical cord mesenchymal stem cells have the ability to differentiate into osteogenic,adipogenic,myocardiogenic and endothelial cells.

Abstract:Objective:To investigate the expression of hypoxia inducible factor-1α(HIF-1α) and the Bcl-2/adenovirus E1B 19 kDa-interacting protein 3 (BNIP3) in human non-small cell lung carcinoma(NSCLC) and their correlation. Methods:The expression of BNIP3 and HIF-1α in 45 tumor samples,45 para-cancer tissues and 14 control normal lung tissues were examined by reverse transcription-polymerase chain reaction analyses (RT-PCR) and SP immunohistochemical method. Multivariable analysis was used to analyze the correlation of the HIF-1α and BNIP3 expression on histological types,differentiation,TNM clinical staging,lymphatic metastasis,age and cigarette smoking. Results:Compared with the control group,the mRNA levels of HIF-1α and BNIP3 in lung carcinoma tissues were much higher. There was a significant difference between tumors and the control(P < 0.01). BNIP3 mRNA levels were positively correlated with HIF-1α levels(r=0.68,P < 0.01). In lung carcinoma,the positive expression rates of HIF-1α and BNIP3 proteins were 53.3% and 66.7% respectively,which were higher than those in para-cancer tissues and normal lung tissues(P < 0.01). The positive correlation between HIF-1α and BNIP3 expression was highly significant(rs=0.38,P < 0.01). The expression of HIF-1α was correlated with histological types and TNM clinical staging(P < 0.05). No significant correlation was found between BNIP3 expression and patients’ clinicopathologic parameters. Conclusion:The high expression levels of HIF-1α and BNIP3 were closely related in non-small cell lung carcinomas,both of them played important roles in the carcinogenesis and development of lung carcinoma.

Abstract:Objective:To investigate the inhibition of N-acetylcysteine(NAC) on the inflammation of diabetic nephropathy rats. Methods:A rat model of DN was established. The rats were divided into control group,DN group and NAC group randomly. Urinary albumin excretion rate(UAER),hs-CRP,IL-6 and TNF-α in serum were determined. The expression of NF-κB in renal tissue was detected by immunohistochemistry 8 weeks after treatment. Pathomorphologic changes of nephridial tissues were observed. Results:①Compared to control group,UAER,hs-CRP,IL-6 and TNF-α in serum were significantly higher(P < 0.01 or P < 0.05),activity of NF-κB in nephridial tissue increased(P < 0.01 or P < 0.05) and significant changes of pathomorphology in nephridial tissues were observed in DN group and NAC group. ②Compared to DN group,UAER,hs-CRP,IL-6 and TNF-α in serum were significantly lower(P < 0.05),activity of NF-κB in nephridial tissue was lower(P < 0.05) and improvement of pathomorphology in nephridial tissues were observed in NAC group. Conclusion:The protective effect of NAC on the kidney of the DN rats may be partly related with the inhibition on inflammation.

Abstract:Objective:To investigate the feasibility of microbubble mediating Ang-1 gene therapy of lower limb ischemia by using ultrasound. Methods:Fifteen rabbits with the left femoral artery ligation were randomly and averagely devidied into 3 groups:group A was locally intramuscular injected with the mixture of plasmid pEGFP/Ang-1 and microbubble and accepted ultrasound local radiation,group B was locally intramuscular injected with the plasmid pEGFP/Ang-1,and group C was injected with NS. Four weeks post-treatment,angiography was applied to evaluate the collateral circulation,and the expression of Ang-1 was evaluate by immunochemical staining. Results:In the matter of the collateral circulation,the degree of brown in cytoplasm,new vascular of the treatment region,group A was superior to the group B,the group B was superior to the group C. Conclusion:Microbubble mediating Ang-1 gene therapy by using ultrasound could promote the collateral circulation and new vessels,it may be a new methods for the treatment of limb ischemia diseases.

Abstract:Objective:To investigate the pathologic changes of kidney vessels after intra-arterial infusion of pingyangmycin(PYM)or pingyangmycin-lipiodol emulsion(PLE)in dogs. Methods:Thirty-one canis familiaris were divided into control group,PYM group,and PLE group. Right renal arteries were selectively infused with PYM solution (PYM group) or PLE (PLE group) by Seldinger’s technique and selective catherization. The pathologic changes of kidney blood vessels were observed at the following 1,3,7,14,28,56 days of PLE group and 1,7,14 days of PYM group. Results:The lesions of renal vessels in PYM group were slight,characteristics of which was focal mild intima swelling of glomerular capillaries at 1st and 7th day and nearly recovery at 14th day. After PLE treatment within 7 days,fibrinoid necrosis occurred in glomerular capillary loops and tunica media of arterioles,mainly interlobular arteries and afferent glomerular arteriole,companied by thrombosis and inflammational cells infiltration around blood vessel. Since 14th day,proliferation of vascular smooth muscle cells and fibrosis of vessel wall have increasingly become apparent,ultimately resulting in stenosis and occlusion of damaged arteries. Conclusion:The lesion of renal vessels were earlier and more lasting by PLE renal arterial injection compared with PYM. PLE may be used as one new embolism agent in embolization.

Abstract:Objective:To explore the effect of liguistrazine on the expression of hypoxia-induced factor-1α(HIF-1α)in neonatal rats with hypoxic-ischemic brain damage(HIBD). Methods:Models of 7-day-old neonatal rats with HIBD were established by occluding the left common carotid artery and subjected to inhalation of 8% oxygen. The mice of treated group were injected with liguistrazine intraperitoneally(50 mg/kg) before and after the operation. The expression of HIF-1α in cortex was measured by RT-PCR and immunohistochemical staining. Results:Although the levels of HIF-1α mRNA and protein expression at 2 h after HIBD were higher than those of sham group,there was no significance(P > 0.05) .While the expression of HIF-1α mRNA and protein were remarkably higher in liguistrazine treated group than those of HIBD group(P < 0.05). Conclusion:Liguistrazine can increase the expression of HIF-ɑ mRNA and protein in neonatal rats with HIBD,which may be the molecular mechanism of its neuroprotective effect.

Abstract:Objective:To evaluate the utility of HCVcAg detection system in clinical diagnosis of hepatitis C infection. Methods:HCVcAg and anti-HCV in sera of 214 hepatitis C patients,60 health control and 40 patients with other hepatic disease were detected by ELISA,and sera HCV RNA was measured by real-time quantitative PCR. Meanwhile,the sera from 78 patients with HIV infection were screened for HCV infection. Results:162 patients (75.7%) were HCVcAg positive in 214 hepatitis C patients. The positive ratio of HCVcAg increased with the HCV RNA copies. The HCV RNA copies were highly related with the HCVcAg with HCV RNA copies over 106/ml. Of 78 samples with HIV,16 were HCV RNA positive,15 were HCVcAg positive,9 were anti-HCV positive. Conclusions:HCVcAg is a serological marker for early identification of HCV. The measurement of HCVcAg significantly shortens the window period of HCV detection,especially for the individuals with negative anti-HCV detection results who use immunosuppressant,and is substituted for HCV RNA detection in where PCR cannot be carried out.

Abstract:Objective:To explore the correlation between the endoscopic diagnosis and pathologic diagnosis of CG (chronic gastritis) in older patients. Methods:A control experiment was carried out between gastroscopic diagnosis and pathologic diagnosis of 352 senior patients with CG. Results:225 cases of NAG(non-atrophic gastritis) and 127 cases of CAG (chronic atrophic gastritis) were diagnosed by endoscopic examination. Comparing to pathologic diagnosis, the accuracy of NAG was 98.67%, while the accuracy of the CAG was 66.93%. A significant improvement of correlation(85.50%) was obtained when combining several endoscopic characteristics. Conclusion:Correlation between gastroscopic diagnosis and pathologic diagnosis was relatively high for NAG, while relatively poor for CAG, but macroscopic diagnosis was indicative to pathological changes for CAG, especially when several endoscopic characteristics coexisted. Currently, the endoscopic diagnosis can't substitute for mucosal pathologic examination yet.

Abstract:Objective:To detect changes of neural response on happy facial expressions in first-onset major depressive disorder(MDD)by antidepressant treatment investigated with functional magnetic resonance imaging (fMRI). Methods:Thirteen patients with first onset MDD and 14 normal age-matched volunteers underwent fMRI and patients underwent scanning twice during an 8-week period. Blood oxygenation level dependent(BOLD) was processed using SPM2 to compare the function of whole brain between the two groups exposure to neutral and positive pictures. Results:Decreased activation of left laterial superior frontal cortex exposure to happy pictures was found in patients compared with control group(P < 0.001,uncorrected). Symptomatic improvement after antidepressant was associated with reversion of left laterial superior frontal cortex(P < 0.001,uncorrected). Conclusion:Changes in BOLD of fMRI associated with symptomatic improvement indicate that fMRI may be a useful marker of antidepressant treatment response.

Abstract:Objective:To identify the characteristic 1H Nuclear Magnetic Resonance spectroscopy(NMR) regions of sera from the patients with esophageal squamous cell carcinoma (ESCC). Methods:1H NMR spectroscopic analysis was performed on serum specimens obtained from 60 preoperative patients with ESCC and 30 healthy controls. Results:The characteristic fingerprint was profiled with 1H NMR spectroscopic data in healthy controls. In the meantime,1H NMR spectra of sera from 60 preoperative patients with ESCC were measured and compared with healthy controls. We found that the 1H NMR spectra of sera from the patients with ESCC most differed from healthy controls at the extent of 1.0~1.1 ppm and 3.0~3.8 ppm. Conclusion:Resort to the characteristic absorption peak,our data suggest that 1H NMR spectroscopy contribute to the early diagnosis on suspicious ESCC by analysis of sera from patients.

Abstract:Objective:To evaluate the application value of ultrasound-guided thrombin injection (UGTI) against simple compression repair in the treatment of small interventional femoral pseudoaneurysms(PSA). Methods:Thirty patients with small interventional femoral arterial pseudoaneurysms firstly received simple compression repair. The pressure should not beyond the endurability of the patients and the throbbing of the feet arterial should be palpable. With 48 hours of compression,12 patients have been cured with satisfactory outcomes. The rest eighteen patients,who can not endure the therapy or who haven’t been successfully cured,received percutaneous puncture with 20 G needle under ultrasound guidance,and were treated by intracavitary thrombin injection. The concentration of thrombin injection was 200 U/ml,the total volume dose was no more than 500 U. Results:Twelve patients were successfully cured at the first time by simple compression repair. No recurrence and complication were found in a three-month followup. The other eighteen patients,who can not endure the therapy or who haven’t been successfully cured by this therapy,were successfully treated afterwards by ultrasound-guided thrombin injection (UGTI). No complication occurred during the operations or after the operations. and no recurrence and complication were found in a three-month followup. Conclusion:During the treatment of interventional femoral pseudoaneurysms (PSA),simple compression repair is first recommended if the body of PSA is small,and the neck is narrow. Otherwise it should be treated by thrombin injection under sonographic guidance for better outcomes.

Abstract:Objective:To compare the effect of right ventricular septum(RVS) pacing and that of right ventricular apex(RVA) pacing on the ventricular wall motion and cardiac function. Methods:RVS pacing was conducted in 20 patients,and RVA pacing was conducted in other 20 patients(sick sinus syndrome or Ⅲ° atrioventricular block). Three months after installment of the dualchamber pacemakers,tissue Doppler imaging was used to evaluated the time to peak in systolic phase of interventricular septum and left ventricular. CI,SV,LVDd and LVEF were also measured by conventional 2-Dimensional echocardiography. Results:Compared with RVA pacing,RVS pacing improved CI-SV- LVEF more. RVS pacing significantly shortened the time to peak in systolic phase and improved the consistency of wall motion as compared with the RVA pacing. Conclusion:Compared to RVA pacing,RVS pacing fulfiled a more physiological depolarization and significantly improveed ventricular performance,including cardiac function and cardiac synchronism.

Abstract:Objective:To assess the diagnostic accuracy of 16-slice computed tomographic angiography (CTA) in detecting intracranial aneurysms compared with digital subtraction angiography(DSA) and surgical findings. Methods:356 cases with suspected intracranial aneurysms received CT scan,and the results were compared to DSA and intraoperative findings retrospectively. Results:Combining CTA,DSA and intraoperative findings,215 aneurysms were detected in 195 of the 356 patients. Five aneurysms were missed when using CTA. Nine aneurysms detected by CTA were not clearly displayed at DSA,five were proved at surgery and four were not performed by surgical treatment. There was no significant difference in sensitivity between 16-slice CTA and DSA. Conclusion:16-slice CTA has a very high sensitivity,specificity,and accuracy of detecting aneurysms,and it is sensitive enough to replace conventional DSA in the triage,diagnosis,and treatment planning in patients with intracranial aneurysms.

Abstract:Objective:To evaluate the value of 64-slice CT coronary angiography(64SCTCA)in the diagnosis of coronary heart disease. Methods:With 64SCTCA, the images of 118 cases were analyzed retrospectively. Results:There were 116 of 118 cases showed the main branches of coronary arteries successfully by 64SCTCA examination. 77 cases of coronary artery lesions of various branches were found, including 61 cases totally 82 branches of coronary artery stenosis. There are 2 cases of coronary artery stent placement got the other coronary artery branches postoperative stenosis, 2 branches restenosed in 10 cases of coronary stent placement. 2 branches occluded in 3 cases of coronary artery bypass surgery. 5 cases of myocardial bridge were detected. 80 plaques were detected from 77 cases. Conclusion:64SCTCA showed coronary artery stenosis clearly and could be used to screen for coronary heart disease. It has unique advantage on evaluation of coronary plaque and postoperative situation, such as coronary stent placement and coronary artery bypass surgery. Furthermore, diagnosis of myocardial bridge by 64SCTCA was superior to coronary angiography.

Abstract:Objective:To study the relationship of the changes of amount of neutrophils to concentration of IL-6,TNF-α during cardiopulmonary bypass. Methods:The serum levels of TNF-α,IL-6 were measured by Enzyme Linked Immunosorbent Assay(ELISA) method in patients undergoing heart valve replacement,and the amount of neutrophils was also measured. Serial blood samples were collected at the following intervals respectively:before CPB,40 min after CPB and 0 h,2 h,6 h,24 h after the end of CPB. Results:The serum levels of TNF-α,IL-6 and the amount of neutrophils after CPB were significantly higher than that before CPB(P < 0.01).The serum levels of TNF-α and IL-6 gradually increased after the beginning of CPB. The serum level of TNF-α reached the peak at 2 h after the end of CPB,and that still maintained a high level at 24 h after the end of CPB. The amount of neutrophils had the similar change. The serum level of IL-6 reached the peak at 6h after the end of CPB,then began to decrease but still higher than the concentration before CPB significantly. The concentration of TNF-α and IL-6 was correlated positively to the time of CPB and ACC(P < 0.01). There were correlations between the amount of neutrophils and the concentrations of TNF-α and IL-6. Conclusion:Cardiac surgery with cardiopulmonary bypass (CPB) leads to a systemic inflammatory response syndrome(SIRS) with secretion of cytokines and neutrophils. The time of CPB and ACC influences release of TNF-α and IL-6,so it may change the degree of SIRS in patients after operation. There are correlations between the amount of neutrophils and the concentrations of TNF-α and IL-6 during CPB.

Abstract:Objective:To study the influence of chemotherapy combined with thymopetidum on cellular immunity of patients with gastrointestinal malignant tumor in the later stage. Methods:80 cases were divided into two groups:testing group (40 cases,with thymopetidum and chemotherapy) and control group (40 cases,with chemotherapy only). 25 cases without malignant tumors were selected as normal group. The activity of T-lymphocyte subgroup and its cytokines as IL-2(Th1) and IL-10 (Th2) in peripheral blood of each group were investigated before treatment and at the tenth day after treatment of two weeks. Results:Before treatment,T-lymphocyte subset(CD3+,CD4+ and the ratio of CD4+/CD8+) and IL-2 of patients with gatrointestinal malignant tumor were lower than those of normal group(P < 0.05),while the values of CD8+ and IL-10 were higher obviously compared with normal group(P < 0.05). After treated for two weeks,CD3+,CD4+ and the ratio of CD4+/CD8+ were higher than those of before treatment in two groups(P < 0.05),and the changes in testing group were more obviously than those in control group(P < 0.05). In testing group,IL-2 was lower and IL-10 was higher than before treatment(P < 0.05),while there was no significant change after treatment in control group. The efficacy of testing group was higher than that of control group(P < 0.05). Conclusions:The immunity of T-lymphocyte subgroup was suppressed in the patients with gastrointestinal malignant tumor and the expression of Th2 type cytokines was enhanced. The cellular immunity of patients was improved by killing the tumor cells and removing the immunosuppressive agent after the chemotherapy together with Thymopetidum.

Abstract:Objective:To evaluate the efficacy and toxicity of docetaxel combined with oxaliplatin and xeloda on treatment of advanced gastric carcinoma. Methods:Thirty-two patients were treated with docetaxel 35 mg/m2 on day 1,8,combined with oxaliplatin 130 mg/m2 on day 1,and xeloda 1 250 mg/(m2·d) twice daily,on day 1~14,with 28 days as a course of treatment. All the patients were treated with more than two courses of chemotherapy. Results:All patients were assessable for toxicity,and 30 of 32 patients were assessable for response. In evaluable 30 patients,CR 1 case,PR 12 cases,SD 9 cases,and PD 8 cases. The response rate was 43.3%(13/30). Median TTP was 5.8 months. The major toxicities were bone suppression and alopecia. Conclusion:Docetaxel combined with oxaliplatin and xeloda are feasible,effective and safe for patients with advanced gastric carcinoma.

Abstract:Objective:To observe the efficacy and security of Hemabate in prevention and cure of postpartum hemorrhage after cesarean section(CS) on parturient with hepatitis B. Methods: Three hundred cases of parturients with hepatitis B accepted cesarean section,whose liver function were abnormal but blood clotting function normal,were divided into 3 groups randomly,and named Hemabate group,Oxytocin group and Combined group respectively. They were all medicated at the moment after childbirth. The postpartum hemorrhage volumes in 2 hours and 24 hours,the ratios of postpartum hemorrhage and the total effective rates were compared among the 3 groups. The advise reactions and the changes of liver function including alanine aminotransferase(ALT),aspartate aminotransferase(AST),totle bilirubin(TB) after medication were observed. Results:The ratios of postpartum hemorrhage in uterine inertia of Hemabate group and Combined group were 22% and 20%,much lower than that of Oxytocin group (54%)(P < 0.01). The postpartum hemorrhage in Hemabate group volumes in 2 hours and 24 hours were (271.54±73.96)ml and (360.97±90.12)ml,which were both much lower than those in Oxytocin group(P < 0.01). The total effective rates of Hemabate group and combined group were 96%,which was higher than that of Oxytocin group (87%). The levels of ALT,AST,TB in Oxytocin group were higher than Hemabate group and combined group in different extent and there were significant differences in ALT and AST(P < 0.05),but there was no significant differentce in ALT,AST and TB between Hemabate group and Combined group(P > 0.05). Some patients suffered from nausea,vomiting,diarrhea,high blood pressure,etc., may almost recover without any disposal. Conclusion:The Hemabate has notable efficacy and slide adverse reaction in prevention and cure of postpartum hemorrhage after CS because of weak uterine contraction on parturient with hepatitis B. Further more,it had nearly no influence on liver function and had good security.

Abstract:Objective:To investigate the effect of diet modification on the weight of the pregnant women with gestational abnormal glucose metabolism. Methods:24 pregnant women with gestational abnormal glucose metabolism were recruited by the glucose challenge or tolerance test,and they have been pregnant for 26 to 28 weeks. The food intake frequency for 3 months before the test was investigated. Nutritional therapy was designed for each women based on the nutrtional assessment,individual status,etc. The change on the level of serum glucose and the weight gain were observed. Results:After the modification,the intake of whole-grain cereals,soybean products,marines,vegetables increased obviously,while the intake of oil,grains,meats decreased significantly,and the intake of energy,fat,carbohydrates was also reduced. The level of 2-hour-postprandial glucose and the weight gain per week decreased. Conclusion:The weight gain can be reduced by diet modification for the pregnant women with gestational abnormal glucose metabolism.

Abstract:Objective:To evaluate the effect of intravitreous injection with triamcinolone acetonide(TA) on macular edema caused by retinal vein occlusion. Methods:Examined by opthalmoscopy,optic coherent tomography(OCT),and fundus fluorescein angiography(FFA),42 patients(42 eyes) with macular edema caused by retinal vein occlusion were intravitreously injected with 0.1 ml TA(40 g/L). During the 3~6 months follow-up period,the visual acuity,intraocular pressure(IOP),inflammatory extent,manifestation of lens and fundus were observed,the retinal thickness was examined by OCT,and vascular leakage was detected by FFA. The visual acuity and retinal thickness were analyzed by ANOVA for Randomized Block Design. Results:After 3 months treatment,the average visual acuity was 0.40±0.27,which improved 3 lines in 33 eyes and not changed in 4 eyes,while the visual acuity was 0.17±0.17 before treatment,the difference was statistically significant(P < 0.05). The average macular thickness was(332.3±196.5)μm 3 months after treatment,while the macular thickness was(537.4±201.6)μm before treatment,the difference was statistically significant(P < 0.05). Three patients who recurred macular edema after 3 months were injected again,one occurred cataract . Three patients had the temporary high IOP(>21 mm Hg),and returned to normal IOP in 1 week without any drugs. The result of FFA showed less leakage of fluorescein after treatment. There were no severe complications such as vitrous hemorrhage,retinal detachment and endophthalmitis. Conclusion:Intravitreous injection with 4mg TA can be used to treat macular edema from retinal vein occlusion effectively,and improve the visual acuity. In those ischemic retinal vein occlusion patients who have recurrent macular edema,TA can be reinjected,but the long-term curative effects and complications should be further observed.

Abstract:Objective:To observe the curative effects of compound glycyrrhizin combined with ligustrazine on patients with alcoholic liver disease. Methods:160 cases of alcoholic liver disease were randomly divided into four groups:control group,compound glycyrrhizin group,ligustrazine group and combined treatment group. The parameters of hepatic function and fibrosis were observed four weeks after treatment. Results:The parameters of hepatic function were improved after treatment and fibrosis,TGFβ1,IL-6,Leptin,TNF-α after treatment in four groups were lower than those before treatment(P < 0.05). The parameters of hepatic function,fibrosis,TGFβ1,IL-6,Leptin and TNF-α after treatment among control group,compound glycyrrhizin group and ligustrazine group had no significance(P > 0.05). Compared with control group,compound glycyrrhizin group and ligustrazine group,the parameters of hepatic function,fibrosis,TGFβ1,IL-6,Leptin,TNF-α of combined treatment group after treatment were obviously improved(P < 0.05). Conclusion:Compound glycyrrhizin combined with ligustrazine are effective for the treatment of alcoholic liver disease.
