Abstract:Objective:To discuss two different ways of staining methods in evaluating myocardial ischemia-reperfusion injury. Methods:Anesthetized SD rats were subjected to myocardial ischemia reperfusion injury(ischemia 30 mins/reperfusion 6 hrs). Hemodynamic parameters were measured upon the completion of reperfusion in rats. The hearts were stained with TTC or Evans blue-TTC double-staining,separately,to determine the extent of myocardial damage. All the rats used in the experiment had the same LV morphology and the similar function. Results:The Evans blue-TTC double-staining method can measure the ratio of infarct areas-to-total left ventricular areas(I/R ratio),While the TTC staining can only measure the ratio of infarct areas-to-risk areas(I/T ratio). Compared with the I/T ratio,the I/R ratio has higher relevance with the cardiac function. Conclusion:The Evans blue-TTC double-staining method can determine the degree of myocardial damage induced by the ischemia-reperfusion injury more objectively and accurately than the TTC staining method.

Abstract:Objective:To study the effect of N-acetylcysteine(NAC)on cardiocytes injury induced by chronic intermittent hypoxia (CIH) by establishing an animal model of obstructive sleep apnea syndrome(OSAS). Methods:Thirty-three ICR mouse were divided into 3 groups randomly:control,CIH and NAC groups. Malondialdehyde(MDA) and superoxide dismutase(SOD) in the cardiocyte homogenates were measured before and after the treatment of NAC using thiobarbituric acid(TBA) and xanthine oxidase methods respectively. In addition,the concentration of cardiac troponin I(cTnI) in the serum was detected by ELISA. Results:① The SOD activity and MDA concentration of CIH group were significantly higher than those of other groups(P < 0.05). The MDA concentration of NAC group was lower than that of control group(P < 0.01). ② The cTnI concentration of CIH and NAC groups was significantly higher than that of control group(P < 0.01). ③The degeneration of muscular tissues,transverse striation blurred,and fabric effusion were observed in tissue pathological section in CIH and NAC groups. However,normal tissue was found in control group. Conclusion:The cardiocytes were injuried by producing activation oxygen in OSAS animals,and this effect can be partially inhibited by NAC.

Abstract:Objective:To investigate the cytoprotection and mechanisms of glycine(Gly) on the ATP-depleted SH-SY5Y cells of neuroblastoma. Methods:SH-SY5Y cells were exposed to ATP depletion in the presence or absence of glycine. The cell injury was assessed by measurements of ATP concentration,lactate dehydrogenase (LDH)release and ceu activity. Glycine receptor expression was analyzed by Western blot. Results:Gly was found to ameliorate a reduce in ATP level within 60 minutes of treatment with rotenone and antimycin A. After the treatment for 6 hours,Gly could reduce the LDH release and enhance cell activity. Meanwhile,glycine receptor was overexpressed under the stress,which was decreased by addition of glycine to cells. Conclusion:These results demonstrated that Gly protected SH-SY5Y cells from ATP depletion injury and alleviated over-expression of glycine receptor under stress.

Abstract:Objective:To study the effect of tamoxifen(TAM) and megestrol acetate(MA) on breast cancer cell line MCF-7. Methods:Estrogen receptor(ER)-positive MCF-7 human breast cancer cell line was treated by TAM or MA alone,or in combination in vitro. Cell proliferation was evaluated by MTT assay. Distribution of cell cycle and rate of apoptosis were determined by flow cytometry(FCM). Results:TAM can inhibit MCF-7 cells proliferation and induce its apoptosis. The cells were arrested in G0/G1 phase;small doses of MA had no significant effect on apoptosis,G0/G1 phase cells increased. When TAM combined with MA,the inhibition effect on proliferation and apoptosis were enhanced. Conclusion:TAM can inhibit MCF-7 cells proliferation and induce its apoptosis. The therapy effect can be improved when two drugs are combined.

Abstract:Objective:To observe the changes of resistance to daunorubicin(DNR) and the expression of multidrug resistance associated protein-1(MRP1) in acute monocytic leukemia cells THP-1 treated by cyclosporin A(CsA), and explore CsA’s function on chemotherapy sensitization to THP-1 cells as well as the related mechanism. Methods:IC50 of DNR to THP-1 cells, content and distribution of DNR in THP-1 cells treated by CsA were examined by MTT method and confocal laser scanning microscopy method, respectively. Expression amount of MRP1 were detected by immunocytochemistry and Western blot. Results:After treated with CsA, IC50 decreased obviously(1.536 4 ± 0.175 1 vs 0.558 8 ± 0.054 7,P < 0.01), content of DNR increasing obviously(21.40 ± 1.71 vs 131.96 ± 16.45,P < 0.01), DNR distributing uniformly,and expression of MRP1 increased(0.266 5 ± 0.004 2 vs 0.316 9 ± 0.006 2,P < 0.01). Conclusion:CsA can increase the sensibility of THP-1 cells to DNR, not through decreasing the expression of MRP1 in THP-1 cells. This might show that the resistance of THP-1 cells to DNR can’t be reduced by inhibiting the activity of MRP.

Abstract:Objective:To investigate the expression of vascular endothelial growth factor (VEGF)-C,VEGF-D and their receptor-3 (VEGFR-3) in patients with non-small cell lung cancer(NSCLC),and the relationship with lymphangiogenesis and lymph node metastasis and their clinicopathological significance. Methods:Forty specimens of the NSCLC and eleven benign pulmonary diseases were studied. The expressions of VEGF-C,VEGF-D,VEGFR-3,LVYVE-1 and Prox-1 protein in specimens of NSCLC and benign pulmonary diseases tissues were studied by immunohistochemical technique. Microlymphatic vessel density (MLVD) was counted. Results:①Among 40 cases of NSCLC,the positive rates of VEGF-C,VEGF-D and VEGFR-3 were 77.50%(31/40),67.50%(27/40)and 62.50%(25/40) respectively,which were significantly higher than those of the paraneoplastic and pulmonary benign diseases tissues(P < 0.01).②Significant correlations between VEGF-C and VEGFR-3(r=0.409,P=0.005),VEGF-D and VEGFR-3(r=0.492,P=0.000) expression were observed in NSCLC;while no correlation between VEGF-C and VEGF-D(r=0.256,P=0.093) was observed. ③The expressions of VEGF-C,VEGF-D and VEGFR-3 protein in NSCLC were not correlated with age,gender,site and dimension of lesion,types of histological and degree of differentiation,but correlated significantly with lymph node metastasis (P < 0.05) and PTNM stage(P < 0.05). ④In the center of NSCLC cancerous tissues among 40 cases,the microlymphatic vessel densities marked by LYVE-1 and Prox-1 were 4.22 ± 1.25 and 1.99 ± 1.49 respectively,which were significantly lower than those of the pulmonary benign diseases tissues(P=0.000). However,the MLVDs in cancerous invasive edge were significantly higher than those in the center of cancerous tissues and those in the pulmonary benign diseases tissues (P=0.000). The MLVDs in the positive VEGF-C,VEGF-D and VEGFR-3 groups were significantly higher than those in the negative groups (P < 0.05). MLVDs were correlated significantly with lymph node metastasis (P=0.000) and PTNM stage (P=0.000). Conclusion:Non-small cell lung cancer cells may secrete lymphangiogenetic growth factors VEGF-C,VEGF-D and their receptor VEGFR-3,which induce the growth of lymphatic vessel endothelium and lymphangiogenesis by VEGF-C,VEGF-D/VEGFR-3 signaling pathway,and further accelerate lymphatic metastasis of NSCLC. VEGF-C,VEGF-D and VEGFR-3 might be acted as molecular phenotypes of lymphangioghesis in NSCLC and important markers for evaluating lymphatic metastasis and prognosis in patients with NSCLC. As specific markers of lymphatic vessel endotheliocyte,LYVE-1 and Prox-1 could identify blood vessels and lymphatic vessels strictly and evaluate vascular system of tumor fairly exactly.

Abstract:Objective:To investigate the expression and correlation of ERK1/2 and Survivin in esophageal cancer. Methods:The expression of ERK1/2 and Survivin on 40 esophageal cancer tissues and 20 normal esophageal tissues were detected by immunohistochemistry. The protein expression of ERK1/2 and Survivin and the consequent changes by PD98059 were detected by Western blot analysis. The correlation of ERK1/2 and Survivin in Eca109 cells was detected by co-immunoprecipitation analysis. Results: Among 40 esophageal cancer tissues,the positive immunolabelling rate of ERK1/2 and Survivin was 87.5% and 90.0% respectively,while among normal esophageal tissues the positive rate was 40.0% and 5.0%. The protein expression levels of ERK1/2 and Survivin in esophageal cancer tissues were higher than those in paraneoplastic tissues. There was coexpression of ERK1/2 and Survivin proteins. PD98059 down-regulated the expression of Survivin in Eca109 cells. Conclusion:The expression of ERK1/2 and Survivin is remarkably up-regulated in esophageal cancer tissues than in paraneoplastic tissues. There is positive correlation between the expression of ERK1/2 and Survivin in Eca109 cells. Inhibition of ERK1/2 by PD98059 induces down-regulation expression of Survivin. The expression of Survivin on esophageal cancers may depend on ERK1/2 expression.

Abstract:Objective:To investigate the effects of the abnormal expression of myo-inositol oxygenase (MIOX) on the expression of TGF-β1 in rat renal tubular epithelial cells under high glucose. Methods:① In vitro, rat renal tubular epithelial cell line NRK-52E was maintained in dulbecco’s modified eagle’s medium (DMEM) containing 5.6 mmol/L glucose and 10% fetal bovine serum (FBS). Then the cells were treated with D-glucose at different concentrations (ranging from 15 to 30 mmol/L) for 48 h, D-mannitol served as control. Gene expressions were analyzed at mRNA level by semi-quantitative RT-PCR and at protein level by Western blot. ② The expressions of MIOX mRNA and protein were observed in NRK-52E cells treated with TGF-β1 at different concentrations (ranging from 1 to 25 mmol/L) for 48 h. ③ Transient transfection of the normal NRK-52E cells with anti-sense oligodeoxynucleotide (ASODN) for 8 h was performed to knockdown MIOX gene using LipofectamineTM 2000. The cells were then treated with D-glucose at concentration of 30 mmol/L for 48 h. Gene expressions were analyzed at mRNA level by RT-PCR and at protein level by Western blot. Results:① In vitro, the expression of MIOX mRNA and protein in NRK-52E cells was induced by high D-glucose in a dose-dependent manner. Paralleled increases were observed with respect to TGF-β1 (P < 0.05). ② There was no significant difference of the MIOX mRNA and protein levels in NRK-52E cells between each group after the stimulation of TGF-β1 at different concentrations (ranging from 1 to 25 mmol/L,P > 0.05). ③ Transient transfection of the normal NRK-52E cells with ASODN significantly attenuated the increase of TGF-β1 expression in normal NRK-52E cells induced by high D-glucose (P < 0.05). Conclusion:High glucose can induce the expression of MIOX and TGF-β1 gene in NRK-52E cells in a dose-dependent manner. MIOX might play a role in the progression of diabetic nephropathy in rat renal tubular epithelial cells through the activation of cytokine TGF-β1.

Abstract:Objective:To investigate the changes of T helper 17 cells and the level of IL-17 in bronchial asthmatic mice. Methods:Twenty-four BABL/c mice were randomly divided into two groups,asthma group and control group. The asthmatic mouse models were established after ovalbumin(OVA) sensitization and challenge. The airway inflammation of lung tissue was observed by hematoxylin and eosin(HE) staining. The ratio of Th17 cells to mononuclear cells in the spleen was detected by flow cytometry. The level of IL-17 in bronchoalveolar lavage fluid(BALF) was measured by enzyme-linked immunosorbent assay(ELISA). The cell numbers in BALF were counted as total cells,eosinophils and neutrophils. Results:Elevated inflammation were observed in the lung and airway tissues of asthmatic mice when compared with controls. Not only the frequency of Th17 cells in spleen but also the level of IL-17 in BALF of asthmatic mice were significantly higher than those of normal controls(all P < 0.01). The numbers of total cells and eosinophils in BALF of asthmatic mice were significantly higher than those of normal controls(all P < 0.01). Conclusion:The study showed that the increasing number of Th17 cells in spleen and level of cytokine IL-17 in BALF are characteristic features in asthmatic mouse model. IL-17 may be associated with the recruitment of eosinophils into airways in asthma.These results demonstrate that Th17 cells may play an important role in the pathogenesis of asthma.

Abstract:Objective:To observe the prevention and therapeutic effects of alendronate combined with simvastatin on the osteoporosis in rats. Methods:Thirty SD female rats were randomly divided into six groups:the ovariectomized group(A), the OVX with alendronate treatment group(B), the OVX with alendronate and the simvastatin treatment group(C), the sham operation group(D), the sham operation with alendronate treatment group(E), the sham operation with alendronate and simvastatin treatment group(F). Two weeks after OVX operation,medical intervention was given into each group except of group A. After three months, all rats were sacrificed. We observed the change of rats’ femoral bone density(BMD), bone gamma-carboxyglutamic-acid-containing proteins(BGP), osteoprotegerin(OPG) and osteoprotegerin ligand(OPGL). Results:The level of BMD in group A was lower than that in the group D(P < 0.05). The level of BMD raised in group B compared to the group A, and in group C BMD raised compared to the group B. The level of BGP in group A was higher than that in the group D(P < 0.05). The level of BGP decreased in group B compared to the group A, and in group C BMD decreased compared to the group B. The level of OPG in group A was lower than that in the group D(P < 0.05). The level of OPG increased in group B and C, compared to the group A. The level of OPGL in group A was higher than that in the group D(P < 0.05). The level of BGP degraded in group B and C, compared to the group A. Conclusion:Combined application of alendronate and simvastatin has better effect than simply application of alendronate on the osteoporosis in rats,and maybe take influence by OPG and OPGL.

Abstract:Objective:To investigate the expression of cyclinD1,P16 and cyclinA1 in the fibroblasts of keloids and hypertrophic scars,and explore the relationship between the genesis of two different kinds of scars. Methods:Normal skin tissues were used as control group,and the expression of cyclinD1,p16 and cyclinA1 were screened in hypertrophic scars and keloids by immunohistochemical SP method. Results:In keloids,positive expression rate of cyclinD1 is 91.7%, that obviously higher than 45.5% in the hypertrophic scar group as well as the control group(P < 0.05);81.8% as the positive rate of p16 in hypertrophic scar group is significant higher than the rate of keloid group,which is 33.3% as well as the control group(P < 0.05);the rate of cyclinA1 in keloids (75.0%) is higher than hypertrophic scar group (27.3%) as well as the control group(P < 0.05);cyclinD1 and cyclinA1 in hypertrophic scars, as well as the expression rate of P16 in the keloid group,are higher than that in the normal skin group,but the difference are not significant. Conclusion:In the progress of scar formation,the imbalance expressions of cyclinD1 and p16,as one pair of the positive and negative cell cycle regulatory factors and positive regulatory factor of cyclinA1,play important roles of the prognosis of scars.

Abstract:Objective:To observe the effects of recombinant human erythropoietin(rhEPO)on skin flap in the SD rats,and discuss the possible mechanism. Methods:A dorsal,caudally based random flap measuring 2 cm × 9 cm was created in all 24 SD rats which were divided into two groups:control group(Group A,n=12)with saline injected through the intraperitoneal route after operation,and treated group(Group B,n=12)with single-dose rhEPO 5 000 IU/kg injected through the intraperitoneal route after operation. At 24 th hour postoperatively,the myeloperoxidase(MPO)activity and the amount of malondialdehyde(MDA) were evaluated on the flap tissue samples of 6 rats chosen randomly. The flap viability and immunohistochemical(CD34)methods which can evaluate the microvascular network were applyed on the rest rats at day 7 after operation. Results:There was significant decrease in MPO and MDA in treated group compared with control group,and a significant increase in the flap viability in treated group was observed(P < 0.05). Conclusion:rhEPO can decrease the MPO activity and amount of MDA in the tissues of random flap at the first 24 h after operation and increase microvessel density and viability at day 7 after operation.

Abstract:Objective:To study the effects of different primers and data analysis methods on quantitative real-time PCR. Methods: Four pairs of different primers were designed for target genes CCND1 and C-JUN using Primer Express 2.0 software. These primers were then used for quantitative PCR amplification and the differences in expression levels between paclitaxel sensitive and resistant lung cancer cell lines were calculated using 2(-DeltaDeltaC(T))method,Pfaffl method and the relative standard curve method,respectively. Results:The expression levels obtained from different primers were significantly different (P < 0.05). In the three kinds of calculation methods,Pfaffl method has no significant difference with the relative standard curve method (P > 0.05),however,2(-DeltaDeltaC(T))method has significant differences with other two kinds of methods (P < 0.05). Conclusion:The amplification efficiency of primers have significant impact on quantitative PCR analysis. The Pfaffl analysis method in the evaluation of relative real-time PCR may be more accurate.

Abstract:Objective:To assess left ventricular(LV)systolic sequence and synchrony by speckle tracking imaging during right ventricular septum pacing and right ventricular apex pacing. Methods:Myocardial imaging of LV apical views and parasternal LV short axis(SAX)views was performed in 9 patients with right ventricular septum pacing(DDDspt group),15 patients with right ventricular apex pacing(DDDapx group)and 13 healthy subjects(control group).The time to peak systolic longitudinal strain(TsL)in the LV apical views,the time to peak systolic radial strain(TsR)and systolic radial strain rate(TsrR)in the LV SAX views were measured. The standard deviation of TsL,TsR and TsrR(TsL-SD,TsR-SD and TsrR-SD)and the maximal temporal difference of TsL,TsR and TsrR(TsL-d,TsR-d and TsrR-d)of 18 segments were used as an indicator of LV systolic dysynchrony,to evaluate the contraction sequence of the LV wall by analyzing TsL. Results:Compared with control group and the DDDspt group,TsL-SD,TsR-SD,TsrR-SD and TsL-d,TsR-d,TsrR-d were significantly higher in DDDapx group (P < 0.05). No significant difference was found between DDDspt group and control group. Left ventricular contraction sequence was similar in DDDspt group and control group,but it changed during right ventricular apical pacing. The contraction of the whole LV delayed in DDDapx group compared with DDDspt group and control group,most segments showed significant differences(P < 0.05). Conclusion:DDDapx group showed significant LV longitudinal,radial systolic dyssynchrony compared with DDDspt group,by inducing different electromechanical activation sequence. Speckle tracking imaging is a useful modality to evaluate systolic sequence and synchrony of the LV during pacing at variant right ventricular sites.

Abstract:Objective:To investigate the safty and feasibility of active-fixation lead in patients with right ventricular outflow tract septum(RVOTS)pacing. Methods:Eighty patients were divided into two groups randomly. One group underwent the right ventricular outflow tract septum(RVOTS)with the active-fixation lead. The other group underwent the right ventricular apex(RVA)pacing with the passive-fixation lead. The parameters in and after operation of two groups were recorded and compared accordingly. Results:Operations went smoothly without complications in two groups. The active-fixation lead group had one case of wire dislocation,while the passive-fixation lead group also had one. Terms of X ray exposure time in active group were significantly longer than those in passive group(19.8 ± 6.4 vs 10.3 ± 4.8,P < 0. 01). After implantation of active electrode,the pacing thresholds were higher than those underwent passive electrode(0.61 ± 0.23 vs 0.48 ± 0.17,P < 0. 05). However,there was no difference between the two groups in pacing thresholds at one and three month point during follow-up. Pacing impedance and R-wave sensing between two groups had no significance. Meanwhile,there was no difference in the QRS duration between two groups. Conclusion:It is safe and feasible to pace in RVOTS with active electrode.

Abstract:Objective:To investigate the changes of right ventricular velocities,strain and strain rate in patients with atrial septal defect(ASD)after transcatheter closure. Methods:Using speckle tracking imaging(STI),the 6 segments of longitudinal peak systolic velocities,peak systolic strain rate(SR),and end systolic strain of the RV free wall and the septum were calculated repeatedly in 25 patients with ASD before surgery and the 3rd day after surgery,and 25 healthy subjects as controls. LVESV,LVEDV,LVSV,RVESV,RVEDV and RVSV were calculated. LV and RV ejection fraction(EF)was measured by Biplane Simpson method. Results:Compared with before closure of ASD,a significant reduction in RV long-axis dimension was noted in 72 hours after ASD closure. In patients with ASD,significantly higher peak systolic velocities were recorded from the RV free wall,compared with that in healthy control subjects. After transcatheter closure of the defect,we observed a significant reduction of peak systolic velocities,which returned to normal physiologic value. Before ASD closure,end systolic strain values were significantly higher,while peak systolic SR values showed a trend to increase,however,no significance compared with control. After transcatheter ASD closure,peak systolic SR and end systolic strain showed a diminishing tendency. However,the difference was not significant. In the patients with ASD,significant higher end systolic strain values and peak systolic velocities were found at the interventricular septum,followed by a significant reduction after ASD closure. Peak systolic SR did not differ significantly from normal values and remained unchanged after closure of the defect. Conclusion:RV longitudinal systolic deformation were not significantly influenced by chronic ventricular volume overload and sudden abolishment of this volume overload. These findings suggest that SR/S are relatively load-stable measures of contractile function in clinic. In contrast,myocardial velocities are load-dependent obviously.

Abstract:Objective: To evaluate the efficacy of heart-type fatty-acid binding protein (H-FABP) for early diagnosis of AMI. Methods:One hundred and four consecutive patients admitted to the hospital within 6 h after the onset of chest pain were recruited in the study. A rapid test kit for qualitative determination of H-FABP was applied to detect the whole serum H-FABP level in all patients at the time of admission. Serum troponinT (cTnT), creatine kinase-myocardial band(CK-MB), and myoglobin(MYO) were also measured simultaneously. Results:Among 104 recruited patients, 51 had a final diagnosis of AMI,and 53 were excluded from AMI, among whom 17 were diagnosed as UAP, 16 as SAP, and 20 as non-cardiogenic chest pain. By comparing the results of the biochemical tests with the standard diagnosis,the sensitivity of H-FABP,MYO,CK-MB and cTnT in diagnosis of AMI were 86.27%, 68.63%, 47.06% and 47.06% respectively. The sensitivity of H-FABP was significantly higher than that of MYO, CK-MB and cTnT (P < 0.05 for all). In terms of specificity, the numbers are 84.90%, 84.90%, 96.23% and 98.11% respectively. The specificity of H-FABP was lower than that of cTnT(P < 0.05), but there was no significant difference between H-FABP and MYO or CK-MB; Within 3 hours after the onset of symptome appearance, the respective sensitivity of H-FABP,MYO,CK-MB and cTnT in diagnosis of AMI was 79.17% ,58.33%, 37.50% and 33.33%;Within 3~6 hours, the respective sensitivity was 92.59%,77.78%,55.56% and 59.26%. In both of the two time stage , the sensitiviy of H-FABP were significantly higher than that of CK-MB and cTnT(both P < 0.05); Within 6 hours after the onset of chest pain, the sensitivity of H-FABP + cTnT was significantly higher than that of MYO+CK-MB + cTnT (92.16% vs 76.47%, P < 0.05), but the differences between the specificity of the two were not statistically significant. Conclusion: The rapid qualitative H-FABP test kit has high efficacy in the diagnosis of AMI as early as within 6 hours of symptom onset. Combining H-FABP and cTnT provides higher sensitivity than the MYO,CK-MB and cTnT combination in the diagnosis of early AMI.

Abstract:Objective:To study the changes of serum IL-4, IL-10 and IFN-γ in patients with different pathological types of primary glomerular disease and its clinical significance. Methods:Twelve cases with FSGS, 12 cases with focal proliferative glomerulonephritis, 12 cases with MsPGN, 12 cases with MN, 12 cases with lupus nephritis and 12 normal controls were involved in this experiment,and their serum levels of IL-4,IL-10,IFN-γ were measured with sandwich ELISA. Results:The levels of IL-4 and IL-10 in MsPGN, MN, FPGN increased, while the level of IFN-γ in them decreased. The level of serum IL-4 and IL-10 in FSGS decreased, while the IFN-γ level increased. The IFN-γ level in LN highly increased,and the IL-4 and IL-10 level increased a little. And results showed that there was negative-correlation of IL-4 level with serum C-reactive protein, and positive-correlation with serum C3 level. Conclusion:The Th2 cells play a dominant role in patients with MsPGN, MN and FPGN, while the Th1 cells play a dominant role in patients with FSGS; there is a high-level interaction between Th1 and Th2 in lupus nephritis patients, and the Th2 cells play a dominant role. The pathological kinds of primary glomerular disease could be initially detected by measuring the level of IL-4, IL-10 and IFN-γ, combining with some other clinical characteristics.

Abstract:Objective:To investigate antibiotic resistance and carbapenemase genotype in Acinetobacter baumannii. Methods:Twenty-two strains of Acinetobacter baumannii resistant to carbapenem and 3 strains susceptible to carbapenem were collected. Their MIC were done by Kribry-Bauer methods and microbroth dilution. The genes were amplified by polymerase chain reaction(PCR), and the PCR products were sequenced. Results:The 22 strains were multi-drug resistance. OXA-23 gene was identified in 18 of the 22 strains, but the gene was not identified in 3 susceptible strains. OXA-24 was not identified in either resistant or susceptible strains. The sequenced results were absolutely homology with blaOXA-23 gene. Conclusion:Production of OXA-23 in Acinetobacter baumannii is one of the molecular mechanisms responsible for carbapenem resistance, and the code gene was blaOXA-23.

Abstract:Objective:To investigate the association between the potentially functional polymorphisms in IL-12A and IL-12B and risk of cervical cancer. Methods:The case-control study was conducted in Jiangsu female population, including 404 cervical cancer cases and 404 cancer-free controls frequency-matched by age and residential areas. The genotype of the IL-12A rs568408, rs2243115 and IL-12B rs3212227 polymorphisms were determined by polymerase chain reaction restriction and fragment length polymorphism (PCR-RFLP) assays. Results:The IL-12A rs568408 GA/AA and IL-12B rs3212227 AC/CC variant genotypes were associated with a significantly increased risk of cervical cancer [adjusted odds ratio (OR)=1.43,95% confidence interval(CI)=1.06~1.93 and adjusted OR=1.30,95% CI=0.97~1.75, respectively], compared with their corresponding wild-type homozygotes. Moreover, a significant gene-gene interaction of these two loci were evident in the risk of cervical cancer (P for multiplicative interaction=0.048); subjects carrying variant genotypes of both loci had a 1.82-fold (95% CI=1.28~2.57) increased risk of cervical cancer. In the stratified analyses,the combined genetic effect was more pronounced in patients with early-stage tumors and those with more parities. Subjects carrying rs568408 AG/AA and rs3212227 AC/CC genotypes and having more than 2 parities showed a 6.00-fold (95% CI=2.86~12.56) elevated cervical cancer risk (P for multiplicative interaction=0.046). Conclusion:These findings indicate that IL-12A rs568408 and IL-12B rs3212227 may individually and jointly contribute to the risk of cervical cancer, and may modify cervical cancer risk associated with parity.

Abstract:Objective:To explore the relationship between IL-1α-889C/T polymorphism and the susceptibility to pediatric epilepsy. Methods:IL-1α-889C/T polymorphism in 117 patients with pediatric epilepsy and 95 healthy individuals controls were analyzed with polymerase chain reaction restriction and fragment length polymorphism(PCR-RFLP). Results:Multivariate Logistic regression analysis revealed that individuals carrying at least one -889 T variant allele (CT + TT genotypes) had a significant increased risk for pediatric epilepsy (adjusted OR = 2.91,95% CI:1.27~6.69) , compared with the wild-type genotype (-889CC) . Furthermore, individuals with epilepsy or febrile seizures family history had a significantly higher risk (adjusted OR = 6.83, 95% CI: 2.30~20.29), compared with those with both CC genotypes. Conclusion:These findings support the hypothesis that IL-1α-889C/T polymorphism may contribute to the risk of developing pediatric epilepsy.

Abstract:Objective:To investigate the color difference of natural teeth and two types of metal ceramic restorations when the light source changed. Methods:According to the CIE L*a*b* color scale ,on the condition of illuminants of D65,A and CWF,the color parameter and spectral reflectance curves of natural teeth and two types of metal ceramic restorations of A2 shade were measured with a spectrophotometer(PR-650). Metameric index was calculated to determine the metameric color differences between natural teeth and two types of metal ceramic restorations. Results:CIE L,a,b values were influenced by the different illuminants in both natural teeth and two types of metal ceramic restorations. The spectral reflectance curves for natural teeth and metal ceramic restorations of A2 shade differed greatly,while there were more than 5 crossing points in each curve. The metameric index between natural teeth and tow types of metal ceramic restorations under illuminant A were 2.03 and 2.46,respectively,2.84 and 2.89 under illuminant CWF. The metameric index between metal ceramic restorations of different kinds were 1.09 under illuminant A,and 0.40 under CWF. Conclusion:Color differences were found between natural teeth and metal ceramic restorations of A2 shade relative to the different illuminants. The metameric effect between natural teeth and metal ceramic restorations of A2 shade is significant.

Abstract:Objective:To investigate the significance of contrast-enhanced ultrasound in quantitative diagnosis of liver fibrosis and cirrhosis. Methods:Twenty cases of liver fibrosis and 20 cases of liver cirrhosis proven by biopsy were enrolled in this study while 10 cases without liver disease history served as controls. A real time gray-scale contrast enhanced ultrasound was performed with SonoVue bolus injection to determine hepatic artery to vein transit time (HA-VTT). Results:There is a significant difference of the HA-VTT among the three groups(P < 0.05). HA-VTT in hepatic cirrhosis group was shorter than controls(P < 0.01). Conclusion:Determination of HA-VTT were considered to be a new diagnostic method in quantitative diagonosis of hepatic fibrosis and cirrhosis.

Abstract:Objective:To investigate the effect of treatment of the elder patients suffering from cervical spinal stenosis. Methods:Thirty-five elder patients (65~79 years old) with cervical spinal stenosis were accepted with different surgeries including the anterior resection the posterior longitudinal ligament combining plate fixation with bone graft or the posterior laminectomy combining lateral mass screw fixation with bone graft from October 2004 to October 2008. The spinal function recovery and improvement rates were analyzed by Japanese Orthopedic Association(JOA) score system. Results:All patients were followed up 7 to 30 months (average 18.4 months). JOA score was 5 to 11 points (average 7.8 points) before operation. JOA score was 9 to 16 points (average 14.2 points) after operation. The clinical effect of operation was excellent. Bone graft integration was observed in all cases. Conclusion:For elder patients suffering from the cervical spinal stenosis,it is possible to get reliable result using appropriate method,complete decompression,firm fixation and effective fusion.

Abstract:Objective:To study the reasons of misdiagnosis and postoperative recurrence of dermatofibrosarcoma protuberan(DFSP)and find out effective ways to improve its cure rate. Methods:The clinical and pathological data of 36 patients with DFSP was submitted to a retrospective study. All 36 patients underwent wide excisions with intraoperative frozen sections. The surgical margins were between 2.5 and 3.0 cm. Three patients with relapse history were treated with 125I radioactive seeds implantation. Results:The clinical misdiagnosis rate at first diagnosis was high to 63.8%(23/36). The operative wounds of all 36 extensive resections were covered with skin grafts or flap transplantations,and the primary healing rate was 95%. The median follow-up period was 3.4(1~6)years and 4(11.1%)cases recurred. Three patients with 125I radioactive seeds implantation were followed up for 1 year to 2 years and none of them had recurrence. There were neither distant metastasis nor death in all cases. Conclusion:The high misdiagnosis rate of DFSP is mainly attributed to untypical early symptoms. The diagnosis of DFSP depends on pathological examination. Wide excision is the crucial measure to improve cure rate and interstitial brachytherapy is an effective adjunctive therapy.

Abstract:Objective:To evaluate the effects of implementation of 3-years’s HIV/AIDS behavior intervention program, and provide evidence for developing intervention strategies. Methods:Two counties in every south, middle and north part of Jiangsu province were selected as representative, used same approach of two-stage cluster sampling to recruit entertainment based female sex workers (FSWs). Questionnaires survey,blood and cervical secretion collection were conducted in 2006 and 2008 respectively. Results:FSWs selected came from similar group during the two cross investigation. After intervention, the awareness rate of AIDS related knowledge increased from 69.2% to 89.4%, and the condom use rate in the previous sexual intercourse increased from 72.5% to 90.2%. The self-protection consciousness of FSWs got to be reinforced, and stigma and discrimination behaviors towards AIDS reduced. The prevalence rate of Syphilis was 4.4% in 2008, a little bit lower than in 2006. While the prevalence rate of Chlamydia trachomatis declined dramatically from 23.1% in 2006 to 10.7% in 2008. The occurrence of incorrect behaviors, for example, immediately vaginal douching after sexual intercourse or took antibiotics to prevent AIDS/STD reduced continuously. In multivariate Logistic analysis, the factors associated with condom use were: registration in Jiangsu province, high number of clients, and good skills at refusal and negotiation to clients. AIDS knowledge looked no statistical association with the condom use. Conclusion:The implementation of scaling up HIV/AIDS behavior intervention program was proved to be positively effective in Jiangsu.
