    • The application of label-free quantitative method in mouse spermatogenesis related proteome

      Abstract:Objective: To identify the proteins differently expressed between 3-week-old and adult mouse testes by label-free quantitative approach. Methods:Proteins were extracted respectively from 3-week-old and adult mouse testes,and digested by trypsin and subsequently analyzed by Nano LC-(liquid chromatography)-LTQ Orbitrap Mass Spectrometric Method. Using this method,the proteins were identified,while at the same time,the relative quantitation of protein expression was calculated according to peak intensity. Results:A grand total of 319 non-redundant proteins were identified,and 30 proteins among them were observed to exhibit significantly differential expression between 3-week-old and adult mouse testes(P < 0.05). These proteins primarily participate in morphogenesis,contraction,endocytosis and fertilization. These processes were tightly related with the spermiogenesis and sperm function. Conclusion:The label-free quatitative proteomic technique provided simple,high- throughput and low-cost measurement of protein expression levels. As such,it has a broad prospect in identifying related proteins involved in testis development,spermatogenesis and sperm function.

    • Abnormal expression of anti-apoptotic gene XIAP in chronic lymphocytic leukemia and its clinical significance

      Abstract:Objective: Several mechanisms were found in the chronic lymphocytic leukemia (CLL) cell apoptosis-resistant process. The objective of this study was to investigate expression of anti-apoptotic gene XIAP in CLL patients and its clinical association with established prognostic factors(IgVH mutation status and CD38 expression). Methods: A total number of 37 CLL patients and 28 age-matched normal control were enrolled in this study. XIAP mRNA expression was detected by SyberGreen-based real-time PCR, clinical relationship was analyzed by GraphPad Prism software. Results:XIAP median expression was 0.04040(95% CI∶0.01610-0.06471) in CLL group compared to 0.07502(95% CI:0.04383-0.1062) in normal control group (P=0.0025); further sub-groups analysis implicated that XIAP expression was higher in drug-resistant group than that in drug-sensitive without statistical difference(P=0.3978), IgVH non-mutated status and positive CD38 expression were associated with higher XIAP expression. Conclusion:Abnormal XIAP mRNA expression as found in CLL patients with significant correlation with established prognostic factors and potential role in drug resistance, and XIAP may become a new supplement of CLL prognostic markers.

    • Release and correlation of matrix metalloproteinase-9 and inflammatory markers in patients undergoing coronary artery bypass grafting

      Abstract:Objective: To assess the release and timing of matrix metalloproteinase-9(MMP-9)and inflammatory markers in patients taken coronary artery bypass grafting(CABG) with cardiopulmonary bypass(CPB) and off-pump coronary artery bypass surgery(OPCAB). Methods:Eighteen patients each subjected to coronary artery bypass grafting and off-pump coronary artery bypass surgery respectively were included in this study. In the CPB group,artery blood samples were collected for biochemical measurements at the following time points:immediately prior to the induction of anesthesia,15 min after initiation of CPB,termination of CPB,2 and 6 h after termination of CPB. In the OPCAB group,time points were as follows:T0,before induction of anesthesia;T1,after completion of anastomose of left anterior descending coronary atery;T2 after completion of all anastomoses;T3,at 2 h,T4,at 6 h after completion of all anastomoses. Results:The levels of MMP-9,TNF-α and IL-8 in CABG group were higher significant than in OPCAB group(P < 0.01). Concentration of MMP-9,TNF-α and IL-8 peaked at termination of CPB in CABG group(P < 0.01). The correlaton between MMP-9 and TNF-α,MMP-9 and IL-8 were significant(r,0.72 and 0.56,respectively). Conclusion:Cardiopulmonary bypass induced the release of MMP-9. TNF-α and IL-8 might amplify the release of MMP-9. The release of MMP-9 could contribute to the CPB-induced inflammatory reaction.

    • Study on activation of NF-κB p65 in rat hippocampal formation after experimental seizure and neuroprotective effects of W-7

      Abstract:Objective: To explore the meaning of activation of NF-κBp65 in rats hippocampal formation and the protective effects and mechanisms of calmodulin inhibitor W-7 on PTZ-induced rats. Methods:Fifty-eight healthy adult Sprague-Dawley rats were randomly divided into 5 groups: control group(i.p.NS), PTZ group(i.p.PTZ 65 mg/kg) and W-7 preteated groups (i.p.W-7 25 μmol/kg, 50 μmol/kg, 100 μmol/kg, followed by i.p.PTZ65mg/kg after 10min of W-7 treatment). Behaviors of rats were observed according to the criteria of Racine. The expression of NF-κBp65 in the hippoacampus of rats as examined by means of immunohistochemistry and Western blotting analysis. Results:Immunohistochemical staining and Western blotting analysis showed that NF-κBp65 was expressed in the hippocampus of all groups. Compared with control group, the expression of NF-κB p65 was increased in PTZ group(P < 0.01), while it was decreased in W-7 groups(P < 0.05,P<0.01). Conclusion:The expression of NF-κB p65 is increased in the hippocampus of PTZ-induced rats. W-7 has protective effects on PTZ-induced rats by depressing expression of NF-κB p65.

    • Role of cyclin kinase inhibitor p21 in the regulation of bufalin on mesangial cell proliferation

      Abstract:Objective: To investigate the role of p21 in the regulation of bufalin on the mesangial cell (MC) proliferation. Methods: The effects of different concentrations of bufalin (0.01,0.04,0.08,0.16 μmol/L) on MC proliferation induced by PDGF-BB(20 ng/ml) was evaluated by MTT test; Flow cytometry and RT-PCR were utilized for cell cycle analysis and p21 DNA expression,,respectively. Western blot was used to analyze protein expression of p21. Results: After twelve hours of PDGF-BB stimulation, MC was cultured with different concentrations of bufalin from 0.04 μmol/L to 0.16 μmol/L for 24 hours, respectively. Proliferation of GMC induced by 20 ng/ml PDGF-BB was inhibited significantly by bufalin in a dose-dependent manner(P < 0.05). Similar results were observed in 0.08 μmol/L bufalin, which blocked the PDGF-BB-induced progression from G0/G1 to S phase of the cell cycle in synchronized cells(P < 0.05). In addition, we found that bufalin reversed PDGF-BB-induced down-regulation of cyclin-dependent kinase inhibitor p21 on both gene and protein level(P < 0.05). However, 0.08μmol/L bufalin had no effects on normal mesangial cell proliferation (P > 0.05). Conclusion: Bufalin may inhibit the PDGF-BB-induced MC proliferation by increase of p21 expression.

      Abstract:Objective: To investigate the susceptibility of the cyclin D1 gene(CCNDl)G870A polymorphism to childhood acute lymphoblastic leukemia(ALL) in a southern Chinese population. Methods:Three hundred and forty-six children with ALL and 520 matched cancer-free controls were genotyped for the CCNDl G870A polymorphism by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)and sequencing. Results:The distribution of three CCND1 G870A genotypes among cases was not significantly different from that among controls. Among ALL patients,the phenotype distribution was not signifcantly different when stratified by treatment branch and immunophenotype of ALL. Conclusion:The results showed that the G870A polymorphism of CCND1 was not associated with the risk of ALL in a southern Chinese population.

      Abstract:Objective: To transfect human angiotensin Ⅱtype 1 receptor(AT1R)gene to human embryonic kidney(HEK)293 cells coexpressing human amyloid-beta protein precursor(APP)and presenilin-1(PS1)gene(APP-PS1-HEK293 cells),select high AT1R expression clone,and verify. Methods:Using pcDNA3.1-AT1R to transfect APP-PS1-HEK293 cells,pcDNA3.1-EGFP as control,selecting positive clones by G418(600 μg/mL),amplifying,then verifying them by RT-PCR,Immunofluorescence and Western blot. Results:RT-PCR,Immunofluorescence and Western blotting indicated that APP-PS1-HEK293 transfected by AT1R gene highly and stably expressed AT1R. Conclusion:The establishment of the stably transfected cell line expressing the target gene provides a solid experimental foundation for further studies on the role of the AT1R gene in the pathogenesis of Alzheimer’s disease.

      Abstract:Objective: To investigate apoptosis and related molecular mechanism caused by nano FeOx powders in hepatoma cell lines HepG2 and Bel-7402 exposed to extremely low frequency altering magnetic field (EMF). Methods: Proliferation and apoptosis of cell lines were detected by CCK-8 kit and flow cytometer using a apoptosis kit,respectirely. Western blot was utilized to detect expression of protein associated with apoptosis and heat shock protein. Results: The proliferation of cells absorbed by nano FeOx powders was inhibited due to the EMF exposure, the inhibited ratio of HepG2 and Bel-7402 cells were (35.54±3.2)% and(76.31±2.3)%(P < 0.05), respectively. The exposure of EMF induced early apoptosis of cells, the apoptosis ratios of HepG2 and Bel-7402 absorbed by nano powders were(18.64±6.68)% and (23.34±2.16)%,respectirely(P > 0.05). Western blot was utilized to analyze the mechanism of cells apoptosis, and the results indicated that the expression of Bcl/Bax in Bel-7402 cells was had no difference, but the apoptosis ratio was similar in two cell lines the expression of Hsp-27 in Bel-7402 was higher than HepG2 cells. Conclusion: The inhibition of proliferation and kigh early apoptosis of Bel-7402 and HepG2 loading magnetic nano FeOx powders under EMF exposure indicated that this kind of treatment could be applied as a potential method for treatment of liver tumor in future.

    • Inhibitory effects of soy isoflavone on proliferation of rat vascular smooth muscle cell induced by angiotensin Ⅱ

      Abstract:Objective: To investigate the effects of soy isoflavone on the proliferation of cultured rat vascular smooth muscle cell (VSMC) induced by angiotensin Ⅱ. Methods:VSMCs were primary cultured by tissue sticking method. Cell proliferation model was established by stimulation with AngⅡ. Cell proliferation was measured by MTT assay and CellTiter-Glo■ assay to observe the effects of various concentrations of soy isoflavone and NG-nitro-L-arginine methyl ester(L-NAME, 100 μmol/L) on VSMC proliferation induced by AngⅡ. Nitric oxide (NO) level was measured by Griess reagent. Nitric oxide synthase (NOS) and inducible nitric oxide synthase (iNOS) levels were detected by chemical colorimetric method. mRNA expression of iNOS was measured by reverse transcription polymerase chain reaction (RT-PCR). Results: Soy isoflavone ranging from 0.1~1.0 μmol/L inhibited cell proliferation in a dose-dependent manner. The inhibitory effects were partly blocked by 100 μmol/L of L-NAME. Soy isoflavone markedly increased NO, NOS and iNOS levels, and increased iNOS mRNA expression in VSMC. Conclusion: Soy isoflavone could inhibit VSMCs proliferation induced by AngⅡ. The increase of NO secretion may participate in this progress.

    • Interaction between parathyroid hormone related protein with Bmi-1 in vitro

      Abstract:Objective: To identify the interaction between protein PTHrP and Bmi-1, and to further explore the underlying signaling pathway of PTHrP. Methods: The PTHrP gene was amplified by RT-PCR and inserted into corresponding sites of expression vector pGEX-2TK, and the fusion protein expressed in E.coli BL21 induced by IPTG. After identifying, the interactions between protein PTHrP and Bmi-1 were characterized by GST pull-down assay and Western Blot. Results:The new recombinant expression vector pGEX-2TK/mPTHrP was constructed and expressed successfully our. experiment confirmed the interactions between the protein PTHrP and Bmi-1. Conclusion:PTHrP specifically interacted with Bmi-1, which may modulate PTHrP downstream signaling pathway.

    • Abnormal expression of LTβR on peripheral blood monocytes in patient with SLE and its possible inducing factors

      Abstract:Objective: To observe expression of lymphotoxin beta receptor (LTβR) molecule on monocytes from peripheral blood of systemic lupus erythematosus(SLE) patients, and to explore regulating factors. Methods:Peripheral blood mononeuclear cells(PBMCs) from the patients and controls were separated, and then LTβR molecule expression was detected by FACS. Monocyte were separated from PBMC by the immunomagnetic beads, and then the mRNA level of LTβR was detected by RT-PCR. Besides, the changes of LTβR expression on the monocyte cell lines(U937 and THP1) and nomal peripheral blood monocyte were investigated after stimulation with LPS,PMA or DEX in vitro. Results:The PBMCs from SLE patients expressed abnormaly high levels of LTβR molecule, which did not express on the PBMCs from normal controls. In cell culture in vitro, DEX up-regulated the espression of LTβR on U937 cell, but did not on THP1 cell and normal peripheral blood monocyte. In U937 and THP1 cell cultures, LTβR expression on cell membrane displayed a temporary decrease,but the LTβR mRNA did not show any changes after treatment with PMA;while no changes were found in LTβR expression on normal peripheral blood monocyte after PMA treatment. Neither the U937 and THP1 cell lines, nor the normal peripheral blood monocyte, did not display any changes in LTβR expression after LPS treatment. Conclusion:The LTβR molecule highly expresses on peripheral blood monocyte from SLE patient, and this abnormal expression may be not related to the active of monocyte or the treatment of glucocorticoid medicine. On the other hand, DEX may be a factor that can influence this molecule expression of monocyte in an earlier stage of development.

    • Development of a new method based on NASBA for enterovirus fast detection

      Abstract:Objective: To develop a new nucleic acid sequence-based amplification (NASBA) method which is based on fluorescence probe for the detection of enterovirus and clinic diagnosis. Methods: The special primers and probes were designed based on the nucleic acid sequences of relative enterovirus in Gene bank. The commercial immunomagectic beads were conjugated with the primers and probes using nanotechnology. The conjugated products were amplified by NASBA and sequentially detected by NucliSens reader. The results were validated by real time-PCR. The sensitivity, repetition and specificity of the method were evaluated respectively. Results: The method could detect enterovirus with high specificity and stability as well as no interactions with other respiratory virus such as RSV. Conclusion: The new NASBA method is convenience, fast, sensitive and specific for enterovirus detection, which could be applied for regular surveillance and urgent investigation of outbreaks of enterovirus in the near future.

    • The efficacy and safety of Tirofiban in percutaneous coronary intervention for non-ST elevation acute coronary syndrome patients

      Abstract:Objective: To evaluate the safety and short-term outcomes of domestic tirofiban in patients with non-ST elevation acute coronary syndrome (NSTEACS) undergoing percutaneous coronary intervention (PCI). Methods:Two hundred and forty-seven patients with NSTEACS were enrolled in the study and divided into two groups: the tirofiban group(n=125) and the control group (n=122). In addition to low molecular weight heparin, patients in the tirofiban group received a dose of tirofiban 0.1 μg/(kg·min) infusion for up to 24 hours after PCI. TIMI grade, platelet aggregation rate and bleeding rate in both groups were investigated and compared before and after PCI. Major adverse cardiac events were also recorded in hospital and during 3 months’ follow-up. Results: Ninty-six percent of patients in the tirofiban group compared with 86.9% in the control group obtained TIMI grade 3 flow(P < 0.05). Platelet aggregation rate in both groups were decreased, and it was lower in the tirofiban group(P < 0.01). There were no serious bleeding complications and MACE in-hospital and follow-up period. Conclusion:Domestic tirofiban is effective on improvement of TIMI grade and prognosis of patients with NSTEACS after PCI.

    • Effect of dietary patterns on the maternal weight changes and neonatal birth weight in pregnant women with gestational abnormal glucose metabolism

      Abstract:Objective: To learn the effect of dietary patterns(low glycemic index dietary patterns)on the maternal weight changes and neonatal birth weight in pregnant women with gestational abnormal glucose metabolism. Methods:Ninety-two pregnant women with gestational abnormal glucose metabolism were recruited with the glucose challenge or tolerance test,they have been pregnant for 24 to 28 weeks. Forty-six pregnant women with gestational abnormal glucose metabolism received diet management(low glycemic index dietary patterns). The food intake frequency for 3 months before the test was investigated of diet management group(dietary patterns regulation group) that dietary patterns regulation was designed for each women based on the nutrtional assessment,individual status,etc. Low glycemic index dietary patterns were adopted by diet management group. Other 46 pregnant women with gestational abnormal glucose metabolism without diet management who only received routine obstetrical checkup. The changes of the weight gain in pregnant women and neonatal birth weight were observed. Results:The maternal weight changes of pregnant women who received diet management increased(13.04±4.12) kg from pre-gestation to delivery and(3.04±3.77) kg from 29 to delivery,which was significantly lower than those of non-diet management pregnant women(16.52±4.47) kg from pre-gestation to deliver and(7.06±3.13) kg from 29 to deliver(P < 0.01). Incidence of fetal macrosomia was significantly lower than 6.5% vs 30.4%(p < 0.01)between diet management group and non-diet management group,respectively. Incidence of maternal and fetal outcomes decreased obviously,with diet management group. Conclusions:dietary patterns(low glycemic index dietary patterns)would be beneficial for better pregnancy outcome.

    • Perioperative psychological supporting therapy combined with patient controlled epidural analgesia for labor analgesia

      Abstract:Objective: To observe the effects of perioperative psychological supporting therapy(PPST)for labor analgesia combined with patient controlled epidural analgesia(PCEA). Methods:120 parturients were randomly assigned to the PPST group,PCEA group,combined group or control group.The variables of analgesia,neonatal outcome,anxiety state,mode of delivery,delivery procedure,postpartum hemorrhage and oxytocin use were assessed. Results:The degree of anxiety in PPST group and combined group were relieved after PPST(P < 0.05). PPST group had a more effective analgesia than control group,but less efficacious than PCEA group and combined group(P < 0.05).With the similar analgesia between PCEA group and combined group,Combined group had a less dosage of PCEA(P < 0.05). Conclusion:Coadministration of PPST with PCEA could be beneficial for relieving labor analgesia and reducing the medication.

    • Effects of HFOV plus SIMV on premature newborns with hyaline membrane disease

      Abstract:Objective: To evaluate the effects of high frequency oscillatory ventilation (HFOV) plus synchronized intermittent mandatory ventilation(SIMV) on treating critical hyaline membrane disease(HMD:grade Ⅲ~Ⅳ). Methods: A total of 53 infants with critical HMD in the NICU of Huaian Maternal and Child Health Hospital from January 2005 to January 2010 were enrolled and divided into two groups: research group (HFOV plus SIMV) and control group(SIMV, or A/C )according the mechanical ventilation mode. X-ray, blood gas analysis, outcome and complications (pneumothorax, intracranial hemorrhage, bronchopulmonary dysplasia) were compared between the two groups. Results: The alteration of X-ray (after 24h treatment), PO2(after 12 h and 24 h treatment) and outcome in research group were better than control group(all P < 0.05). Compared with control group, the incidence of pneumothorax in research group was significantly lower(P < 0.05). But no statistic significance was found in incidence of intracranial hemorrhage and bronchopulmonary dysplasia between two groups(P < 0.05). Conclusion: HFOV plus SIMV can improve lung oxygenation and the outcome, reduce the incidence rate of pneumothorax without increasing the risk of intracranial hemorrhage. In the treatment of critical HMD, HFOV plus SIMV was superior to conventional mandatory ventilation.

    • A comparison of curosurf and calsurf on preterm infants respiratory distress syndrome

      Abstract:Objective: To evaluate the efficacy and safety of two natural surfactants (curosurf and calsurf) on the treatment of preterm infants respiratory distress syndrome(RDS). Methods:From January 2007 to July 2009, we conducted a trial at the neonatal intensive care unit of Nanjing Children’s Hospital. Eighty-one premature babies suffered from RDS and requiring mechanical ventilation(MV)were recruited and divided into two groups:54 in curosurf group and 27 in calsurf group. The surfactant was administered by endotracheal intubation. Blood gas index,ventilator parameters,oxygenation index(OI),the duration of ventilator dependence and hours of oxygen requirement,time in hospital and incidence of complications before and after surfactant administration were analyzed and compared. Results:The oxygenization and ventilator parameters of the two groups showed significant improvements at 24h after administration in comparison with those before surfactant therapy(both P<0.05). There were no statistical differences in the duration of MV or oxygenation,hospitalization time. No statistically significant differences were found in incidence of mortality or complications,which included retinopathy(ROP)and necrotizing enterocolitis(NEC),pneumonorrhagia,pneumatothorax,patent ductus arteriosus (PDA). Both groups had sustained decreasing mean airway pressure(MAP)and OI after surfactant therapy,but time-effect curves were different. MAP and OI of curosurf group decreased most significantly within the initial 4 hours after administration (P < 0.01)compared to the calsurf group,which mainly decreased between the 4th hour and the 8th hour after administration(P < 0.05). Conclusion:In the treatment of preterm infants with RDS,the efficacy and safety of curosurf and calsurf were similar,while curosurf works earlier than calsurf. We should adjust the MV parameters properly in accordance with the different time-effect curves of the two surfactants,so as to reduce complications.

    • The value of conventional MRI and 1H-MRS in early diagnosis of brain damage in neonatal hyperbilirubinemia

      Abstract:Objective: To investigate the characteristics of magnetic resonance imaging(MRI) in brain,and changes in brain metabolites proton MR Spectroscopy(1H-MRS) of neonate with hyperbilirubinemia,and the relationship between those changes and common diagnostic parameters of brain injury. Methods: Thirty-six children with neonatal hyperbilirubinemia(serum bilirubin >290 -滋mol/L)were inspected by MRI and MRS,serum SB100B were also detected,and neonatal behaviral neurological assessment(NBNA)were scored at day 7 after birth. The incidence of brain injury based on single parameter was calculated. Results:Conventional MRI showed symmetrical high signal of TWI in globus pallidus of 12 patients,of which Multi-voxel 1H-MRS revealed that the relative concentrations of NAA/Cr and Cho/Cr were much lower than the others,but there was no significant difference in the relative concentrations of Cho/NAA. The incidences of brain damage dignozed by a single parameter were as follows:clinic diagnosis 22.2%,MRI 33.3%,MRS 41.7%,NBNA 38.9%,S100B 36.11%,respectively. Conclusion:Combination of MRI and MRS,will be more helpful for early diagnosis of brain damage due to hyperbilirubinemia and provides reliable clinical evidence for early intervention.

      Abstract:Objective: To detect the presence of common viral pathogens in clinical specimens of respiratory tractand to understand the current situation of viral infection, children’s acute respiratory tract infection(ARTI) in Nanjing Children's Hospital. Methods: Deep nasopharyngeal secretions(NPS) specimens were collected in Hospitalized children diagnosed with ARTI. A variety of viral pathogens including respiratory syncytial virus(RSV), influenza virus(IFV), parainfluenza virus(PIV), human metapneumovirus (HMPV), adenovirus (ADV), rhinovirus (HRV) and enterovirus(EV) were detected using reverse transcription polymerase chain reaction(RT-PCR). Meanwhile, the clinical and epidemiological manifestations of ARTI caused by these viral pathogens were analyzed. Results:A total of 212 cases affected with viral pathogens were detected in 340 specimens (62.35%). Among them, 132 cases were RSV infection (62.3%), 45 cases IFV infection (21.3%), 13 cases PIV infection (6.1%), 13 cases HMPV infection (6.1%), 4 cases ADV infection (1.9%), 2 cases HRV Infection (0.9%), and 7 cases EV infection (3.3%). Conclusion:RSV infection is most common in ARTI children. Few children have multiple viral infections.

      Abstract:Objective: To evaluate the long-term outcomes and safety of laser in situ keratomilesis (LASIK) for myopia. Methods: 39 cases (77 eyes), who underwent LASIK, were divided into two groups: myopia ≤-6.0 D and >-6.0 D . All patients were followed-up for 10 years after surgery. The main parameters for evaulation were predictability, stability, safety and efficacy. Uncorrected visual acuity (UCVA), best-corrected visual acuity (BCVA), refractions, topography, corneal thickness and postoperative complications were recorded at 10 years. Results: At the 10th year, 63% of eyes with myopia ≤-6.0 D and 42% of eyes with myopia>-6.0 D had 1.0 or better UCVA, 27% of eyes with myopia ≤-6.0 D and 58% of eyes with myopia>-6.0 D had less than 1.0 UCVA . 98% of eyes with myopia ≤-6.0 D and 72% of eyes with myopia>-6.0 D had 1.0 or better BCVA(P < 0.05). There were significant differences at values of Diff value on anterior and posterior surface,Irreg at 3 mm and 5 mm zone obtained from Orbscan Ⅱand corneal thickness between pre- and post- operation(P < 0.01). Mean corneal thickness at the 10th year postoperatively was higher than attempted residual corneal thickness after ablation, it suggested that the stroma of cornea proliferated. Conclusion:LASIK surgery is safe and has predictable and stable results in refractive and visual outcomes in correcting myopia on long-term follow-up.

      Abstract:Objective: The purpose of this study was to evaluate the potential benefit of an ultrasonic device in apical surgery on the outcome of treatment when comparing with conventional technique using a bur. Methods:A case-control study was used in a standardized treatment protocol. 98 patients were treatment with an ultrasonic device or with a bur for preparation. One year after treatment the results were sorted and numbered by another clinician. The results were evaluated by a third dentist who was blinded for the therapy. Results:The overall success rate in the ultrasonic group was 90.0% and in the group treated with a bur 82.8%(P > 0.05). In premolars,the difference in success rate was 84.6% in ultrasonic group and 70.8% in conventional group(P > 0.05). Conclusion:The use of an ultrasonic device in apical surgery improved the outcome of treatment. Ultrasonic preparation could get better cannel obturation than conventional technique.

    • Comparison of planter turbine drill and traditional chisel technique on removal impacted mandibular third molars

      Abstract:Objective: To compare effects of planter turbine drill and traditional chisel technique on removal of impacted mandibular third molars. Methords:Three hundred and thirey cases with impacted third molars of middle or low site were randomly assigned to two groups:test group including 160 cases received treatment of planter turbine drill; control group including 170 cases received treatment of traditional chisel technique. The duration of extraction and discomfort of the patients, and post-operational complications were determined and analyzed. Results:The test group had shorter duration of extraction, less discomfort, and less post-operational complications such as ache,swelling, limited opening of mouth, than those of the control group. Occurrences of dry socket and fracture of lingual alveolar bone were lower than the control(P < 0.01). Conclusion:The planter turbine drill has a superiority for application on extraction of impacted third molars compared with traditional chisel technique.

      Abstract:Objective: To study the agreement and reproducibility of probing depth(PD) measurements between the electronic computerized periodontal probe for constant pressure and the conventional manual periodontal probe before mechanical treatment in patients with chronic peridodontitis. Methods:PD values of total 144 periodontal sites from 12 patients with chronic periodontitis were duplicately measured using electronic periodontal probe and conventional manual probe before instrumental therapy by two examiners at two visits for the 1st day and the 7th day, respectively, Analysis of the agreement by intraclass correlation coefficient (ICC) and reproducibility between the two different periodontal probes on PD measurement was performed by paired t-test. Different examiners,different visits time, different periodontal pocket depth, different probes(methods) were taken into consideration as possible factors leading to the difference. Results:The agreement and reproduciblity of PD values from two different probes were good (P > 0.05)both between two examiners and two visits. In addition, by linear regression model the difference of PD values between two tools is contributed to the periodontal pocket depth(t=72.42,P<0.001) other than examiners(t=0.26,P=0.793),visits(t=-0.17,P=0.866)or periodontal probes(t=-0.44,P=0.662). Conclusion: Electronic periodontal probe and manual periodontal probe, which are used by examiners trained normally and strictly, are both reliable tools for pretreatment measuring PD values in chronic peridodontitis.

      Abstract:Objective: To propose a simple image segmentation method,which is based on the teeth image acquired by oral cavity endoscope. Methods:Firstly,removing the sequins of the image by the open operation of morphology;Secondly,removing the noise of image by the value of filtering;Lastly,segmenting the image through the parameters gained by the gray value of the image,detecting the edge of the teeth image and corners. Results:This method can separate the teeth out from the image effectively,and the corners can offer some available parameter for the 3D reconstruction of the teeth. Conclusion:The method can segment the teeth from the image effectively.

      Abstract:Objective: To investigate the clinical change of cytokines expression in severely burned patients. Methods:Forty-nine burned patients with burn surface exceeding 35% TBSA and healthy controls of same number were enrolled in this study. The peripheral blood of patients were collected at 6 h,12 h and 1,2,7,14 post-burn days. The T lymphocyte proliferation ability was analyzed by CCK-8 and the levels of IL-2-IL-4-IL-6-IL-8 in patients and healthy donors were detected by ELISA. Results:Compared with that in healthy controls,the proliferation ability of T lymphocyte and the levels of IL-2,IL-4 were significantly decreased in patients on 6 h,12 h and 1,2,7,14 post-burn days(all P < 0.05),but the levels of IL-6,IL-8 were significantly higher than those of healthy controls(both P < 0.05). Conclusion:The immunological function of T lymphocyte in severely burned patients is depressed persistently,and the pro-inflammatory cytokines could participate in the pathological process of inflammatory response following burn.

    • Anti-asthma mechanism of Xiansheng Kechuanling oral liquid in asthma rat model

      Abstract:Objective: To investigate possible anti-asthma mechanisms of Xiansheng kechuanling Oral Liquid in asthma model rats.Methods:Sixty rats were randomly divided into normal control group, asthmatic model group and treatment group with 20 rats in each group. Rat asthma model were established by egg albumin induction. The rats of treatment group were treated with Xiansheng kechuanling Oral Liquid (10 ml/kg) once daily for 7 days continuously. The concentrations of IL-4, IL-5 and IL-13 in rat plasma were determined using ELISA. Results:The symptom of asthma in treatment group was more improved than that in model group. Furthermore, the concentrations of IL-4, IL-5 and IL-13 in plasma were significantly decreased in treatment group than those in model group. Conclusion:These findings suggest that Xiansheng kechuanling Oral Liquid inhibits IL-4, IL-5 and IL-13, which is likely to be one of its anti-asthma mechanisms.

      Abstract:Objective: To investigate the efficacy of decision tree incorporated with indocyanine green clearance test on hepatic reserve function in patients with hepatocellular carcinoma. Methods:Eighty-two patients with hepatocellular carcinoma confirmed by pathology were enrolled. The presence of ascites, serum bilirubin level and indocyanine green retention rate at 15 minutes were elevated. Results:During strict application of this decision tree to 82 hepatectomies,no postoperative mortality(overall morbidity of 0%) and no major complications were observed. Conclusion:The decision tree can provide more precise information of liver function so as to avoid liver failure of patients with hepatocellular carcinoma.

    • Correlations between the utilization of first-generation cephalosporins antibiotics and resistance rates of staphylococcus aureus

      Abstract:Objective: To evaluate the correlation between the utilization of first-generation cephalosporins antibiotics and resistance rates of staphylococcus aureus(MSSA) from 2001 to 2009, to provide theoretical evidence for clinical antimicrobial medication choice. Methods:DDDs(DDDs/1000) of first-generation cephalosporins antibiotics and resistance rates of MSSA were counted, and correlation of two variables was analyzed by Sperman’s non-parametric correlation using SPSS17.0 software. Results:During 9 years there was overall upward trend in resistance rates of MSSA. The resistance rates of penicillin, cefuroxime were the highest, while resistance rate of glycopeptide was lowest. Resistance of co-trimoxazole, minocycline had decreased, including DDDs of first-generation cephalosporins was decreased year by year. There were respectively perfectly positively correlation between DDDs of first-generation, cefradine and cefadroxil and resistance rate of minocycline(r=1.000,P < 0.01).DDDs of cephradine and first-generation cephalosporins and resistance rates of penicillin, cefepime, clindamycin and amikacin were respectively negatively correlated(r= -0.900,P < 0.01;r=-0.967,P < 0.01;r = -0.967, P < 0.01;r=-0.917,P < 0.01;r=-0.817,P < 0.01;r=-0.733,P < 0.05;r=-0.950,P < 0.01;r=-0.783,P < 0.05);The DDDs of cephradine and resistance rates of piperacillin /tazobactam, cefazolin, erythromycin, levofloxacin were negatively correlated(r=-0.683,P < 0.05;r=-0.917,P < 0.01; r=-0.750,P < 0.05;r=-0.881,P < 0.01 respectively).The DDDs of cefadroxil and resistance rates of cefuroxime were perfectly positively correlated(r=1.000,p < 0.01). Conclusion: The DDDs of first-generation cephalosporin and resistance rate of MSSA were significantly correlat, and clinical doctors should pay more attention on the usage of cephalosporin antibiotics.
