Abstract:Objective: To explore the applied characteristics of back-propagation artificial neural network (BPANN) on studying the genetic variants in PPAR-γ and RXR-α gene and type 2 diabetes risk in a Chinese Han population. Methods: With BPANN as fitting model based upon data gathered from type 2 diabetes patients(n=591) and normal controls (n=724), the mean impact value (MIV) for each input variables and sequencing the factors according to their absolute MIVs were calculated. The results from BPANN were compared with multiple logistic regression analysis,and multifactor dimensionality reduction (MDR) method was used to consider the joint effects of PPAR-γ and RXR-α gene. Results:By BPANN analysis, the risk factors of diabetes mellitus were serum adiponectin level, hypertension, waist, rs4240711, rs3132291, rs3856806, diabetes mellitus family history,no alcohol drinking hyperlipoproteinmia, smoking, age, body mass index, rs1045570, gender, rs2920502, rs6537944, rs4842194, rs17827276 and rs1801282. However, only 8 factors were statistically significant in multiple logistic regression analysis, arrayed according to the important valne: hypertension, diabetes mellitus family history, waist, no alcohol drinkiy, smoking, rs4240711, rs4842194 and serum adiponectin level. Model X1 X2 X3(rs3856806, rs3132291, rs4240711) was the best model (CV Consistency=10/10, P=0.0107) with MDR method. Conclusion: These results suggested that the interactions of PPAR-γ and RXR-α gene might have important role in the susceptibility of T2DM. Neural network could be used to analyze the risk factors of diseases and more complicated relationships (main effects and interactions) between inputs and outputs, better than using the traditional methods.

Abstract:Objective: To determine whether bone marrow mesenchymal stem cells (BM-MSCs) are presented in the different fractions of bone marrow cells and whether the efficiency of colony forming unit-fibroblastic(CFU-f) formed by BM-MSCs is regulated by 1,25(OH)2D3 and prostaglandin E2(PGE2). Methods:Total bone marrow cells or non-adherent bone marrow cells derived from total bone marrow cell cultures for 1 day were separated into the mononuclear cell fraction and the granulocyte/erythrocyte fraction or the mononuclear cell fraction, the granulocyte fraction and the erythrocyte fraction by density-gradient centrifugation and were cultured in the absence or presence of 10-8mol/L 1,25(OH)2D3 or 10-7 mol/L PGE2 for 10 days. The resulting cells were stained with methylene blue and the number of CFU-f was counted. Results:①The CFU-f formed by mononuclear cell fraction accounted for about 10% of the total CFU-f, and CFU-f formed by granulocyte/erythrocyte fraction, granulocyte fraction and erythrocyte fraction accounted for about 90%,47% and 35% of the total CFU-f, respectively; ②The fractions derived from the non-adherent bone marrow cells accounted for about 71% of the total CFU-f, which was more than those formed by the fractions derived from total bone marrow cells; ③ Treatment of 1,25 (OH)2D3 and PGE2 enhanced the CFU-f formation of the mononuclear fraction and the granulocyte fraction derived from total bone marrow cells and the mononuclear cell fraction, the granulocyte fraction and the erythrocyte fraction derived from non-adherent bone marrow cells. Conclusion: Our results indicate that BM-MSCs are presented in the different fractions of bone marrow cells,and 1,25(OH)2D3 and PGE2 play a role in stimulating proliferation of BM-MSCs.

Abstract:Objective:To package the recombinant adenovirus containing HIV-1 Vif gene by using AdEasy system and detect the effect of Vif protein expression on the latent infection and lytic replication of KSHV. Methods: The fragment of Vif gene from expression vector pCI-neo-Vif was cloned into the shuttle plasmid pAdTrack-CMV. With the digested recombinant plasmid pAdTrack-Vif and the backbone plasmid pAdEasy-1, the homologous recombination took place in the E.coli BJ5183,and the recombinant adenovirus plasmid was generated. The recombination adenoviruses were packaged and amplified in the AD293 cells. Then the virus titer was checked by observing the expression of GFP. After BCBL-1 cells were infected by the recombinant adenovirus with 1 MOI, the mRNA transcripts and protein expression of Vif in BCBL-1 cells were detected by RT-PCR and Western blot. Then, the expressions of KSHV ORF26 mRNA and vIL-6 protein were detected by Real-time PCR and Western blot, respectively. Results: The recombinant adenovirus carrying HIV-1 Vif was packaged successfully with the virus titer of 2.5×108 TU/ml. After infected with adenovirus, more than 80% BCBL-1 cells could express GFP, and the exact band of Vif was detectable by RT-PCR and Western blot. Moreover, the expression of KSHV ORF26 mRNA and vIL-6 protein were down-regulated by Vif protein. Conclusion: The recombinant adenovirus carrying HIV-1 Vif was packaged successfully,and Vif protein expressed in BCBL-1 cells could inhibit KSHV lytic replication and enhance KSHV latent infection.

Around the world,as the important component of legal system,Independent Review Board (IRB) plays a significant role in protecting human testing subjects. However there exist many problems in the practice of IRB and the supervision over it. The article suggests that we would draw inspiration from American relative laws about IRB.

Abstract:Objective:To investigate the expression protein of HHV-8 K13 in human endothelial and prostate cancer cells respectively, and evaluate the effect on the proliferation of the two cell lines. Methods:The recombinant plasmid PEF-K13-Flag-IRES/Puro was transiently trancfected into EA.HY926 cells and PC-3 cells. The expression of K13 protein in both cells was analyzed by Western blot. The function of K13 protein in cell cycle and cell proliferation were measured by flow cytometry and MTT assay, respectively. Results:The specific band of K13 protein was detected at the expected place by Western blot. In addition, the results of the flow cytometry and MTT assay showed that the HHV-8 K13 protein could significantly inhibit the proliferation in human endothelial and prostate cancer cells. Conclusion:HHV-8 K13 protein might inhibit the growth of human endothelial and prostate cancer cells.

Abstract:Objective: To observe the abnormal DNA methylation inhibits the miRNA expression in pancreatic cancer cell line, and DNA methyltransferase inhibitor on the miR-615-5p of the methylation status and expression in the pancreatic cancer cell line PANC-1. Methods: Methylation microarray was used to screen in pancreatic cancer cell line PANC-1 in the abnormal methylation of miRNA. PANC-1 cells were treated with demethylating agent 5-aza-2-adeoxycytidine (5-aza-dC) in vitro, Methylation specific polymerase chain reaction (methyla-tion- specific PCR, MSP) combination of bisulfite modified DNA sequencing method (bisulfite-sequencing PCR, BSP) were used to detect miR-615-5p methylation changes before and after treatment. The changes of miR-615-5p expression was detected by quantitative RT-PCR before and after treatment. Results: Methylation was significantly higher in pancreatic cancer cell line PANC-1 than in normal tissue by MSP and BSP methods. miR-615-5p reduction in methylation was associated with the expression reactivation in the pancreatic cancer cells PANC-1 treated with demethylation agent 5-aza-dC. Conclusion: This study demonstrates that aberrant DNA methylation of the miR-615-5p CpG island is closely linked to their inappropriate silencing in pancreatic cancer cell line PANC-1.

Abstract:Euthanasia is related with physic,law,philosophy and so on and there are two attitudes towards it. In combination with jurisprudential base,the article analyzed the concept and related issues of euthanasia,pointed out the legality of euthanasia in terms of the value of law,the right of subsistence,the illegality hindered of the subject and the principle of crimes and penalties prescribed by law,then indicated the reasonableness of the practice of euthanasia. Also,some constructions and assumptions were presented to do some efforts to the legislation of euthanasia in China.

Abstract:Though reinterpreting the characteristics and significance of holism of traditional Chinese medicine from the view of existentialism, phenomenology and structuralism, the author believes that traditional Chinese medicine is a recognizing framework based on Chinese language, and it is Chinese structuralism with a different method compared with the western culture, all the theories of traditional Chinese medicine are based on the self-evidence of living world. Scientific spirit and humane features of traditional Chinese medicine lie in healthy living perspective and considering its subject under the field of medicine as a whole alive human being.

Abstract:Objective: To investigate the role of mTOR specific inhibitor RAD001 in inhibiting gastric cancer cell SGC7901 proliferation and the possible mechanism of survivin in the inhibition. Methods:SGC7901 cells were cultivated, treated with RAD001 alone or in combination with cisplatin. MTT, flow cytometry, Immunocytochemistry and Western-blot array were used to investigate the cell proliferation, apoptosis and the expression of survivin in cells. Results:After treated with RAD001 alone or in combination with cisplatin, the cell proliferation were inhibited in a dose-dependment manner, and the inhibition rates increased with the treatment time extending. High concentration of RAD001 could induce apoptosis of SGC7901 cells. The expression of survivin in SGC7901 cells decreased after treatment. Conclusion:RAD001 could inhibit the cell proliferation of SGC7901 and induce the apoptosis through down-regulating the expression of survivin.

Abstract:Objective: To evaluate the energy metabolism of skeletal muscle in sprinters using 3.0 tesla phosphorus magnetic resonance spectroscopy(31P-MRS). Methods: 31P-MRS was acquired from quadricep muscles in 14 trained sprinters and 16 non-trained healthy volunteers. Concentration of inorganic phosphate(Pi),phosphocreatine(PCr)and adenosine triphosphate(ATP) was measured by matlab software. Concentration of adenosine diphosphate(ADP) and intracellular pH value was calculated from the biochemistry reaction equilibrium. Results: Concentration of PCr and ATP in the muscle of trained sprinters were significantly higher than those in the volunteers. Sprinters had lower pH value in skeletal muscle than volunteers did. No difference was observed in the concentration of ADP between two groups. Conclusion: Trained athlete have higher content of ATP and PCr indicating a more efficient mitochondrial phosphorlation system,and 31P-MRS has important role in evaluating training effects in sports.

Abstract:Objective: To determine whether histamine H3 receptor in the hypothalamic paraventricular nuclei (PVN) regulates the function of hypothalamic-pituitary-adrenal (HPA) axis in rats with asthma. Method: Rat model for asthma was established by Elwood’s methods. A stainless steel jacket was implanted into the PVN using a technique of central three-dimensional position, and R-(α)-methylhistamine [R-(α)-MeHA, a H3 receptor agonist] 2 μg or thioperamide (a H3 receptor antagonist) 5 μg was injected into the PVN. Levels of histamine in PVN were detected by high-performance liquid chromatography, and the concentrations of CRH in median eminence, ACTH and CORT in plasma were detected by the radioimmunoassay. Relative levels of IL-10 in both hypothalamus tissue and plasma were measured by double-ligand-linked immunosorbent assay. Result: Levels of histamine in PVN decreased significantly, whereas the concentrations of CRH in median eminence, ACTH and CORT in plasma and the levels of IL-10 in both hypothalamus tissue and plasma were all increased significantly in acute episode asthma rats with treatment of R-(α)-MeHA compared with control rats, and these responses were blocked by treatment of thioperamide, a H3 receptors specific antagonist. Conclusion: Specific activation histamine H3 receptors in PVN can improve the function of HPA in rats with asthma.

Abstract:As one of our government point work in 2010,the reformation of medicine and health system has received extensively attention from our community and how to establish medicine and health system which is suitable to our national conditions has become one of hot society issues. Based on the target of reformation of medicine and health system,the status of public hospital and private hospital in quantity,resource allocation and so on,the article indicated that the problem of difficulty and high cost of getting medical service was not resolved although the investment in health care was enhanced by government in recent years. Under the background mentioned above,the authors advised that “one hospital,two systems” should be implemented as the reform model to solve the contractions between non-profit insufficiency of public hospital and inadequate funding of government.

Abstract:Objective:To investigate the gene expression profiles of squamous esophageal cancer carcinoma(ESCC). Methods:The cDNA retro-transcribed from both 3 cases(groupⅠ) and 6 cases(groupⅡ),which consisted of equal quantity of both squamous esophageal cancer tissues and adjacent normal tissues,were labelled with Cy5 and Cy3 fluorescence as probes. The mixed probes were hybridized with a piece of Biostar H-140s single dot human whole gene chip. It was scanned by scanarray 4 000 instrument. The acquired image was analyzed by GenePix Pro 3.0 software. Results:There were 1 855 genes in groupⅠ and 613 genes in the groupⅡ,respectively,whose Cy5/Cy3 ratio was greater than 2.0 or less than 0.5. Furthermore, there were 60 genes, whose Cy5/Cy3 ratio wasgreater than 2.5 or less than 0.25 simultaneously in bothⅠgroup and Ⅱ group, including 30 ESTs in GenBankTM. There are 26 genes that has not been reported relating to squamous esophageal cancer,except 4 genes of epithelial membrane protein 1(EMP1),secreted phosphoprotein 1(SPP1),ubiquitin-conjugating enzyme E2C(UBE2C) and retinoblastoma-like 2 (p130) (RBL2). Conclusion:Microarray may play important roles in exploring the gene change in ESCC with and without family history, to illustrate the mechanism on the developement of ESCC.

Abstract:Objective:To investigate the expression and significance of C/EBP homologous protein/growth-arrest and DNA damage inducible gene 153(CHOP/GADD153) in preeclampsia. Methods: Semiquantitative RT-PCR,immunohistochemistry and Western-blot were used to investigate the expression of CHOP/GADD153 mRNA and protein levels in placenta of 30 women with preeclampsia and 35 normal pregnant women. Results: ①The expression of CHOP/GADD153 mRNA was significantly higher in preeclampsia group(13.416 ± 1.516) than in the normal pregnant group(2.615 ± 0.659,P < 0.01). ②The mean density of CHOP/GADD153 in preeclampsia placenta(0.156 ± 0.008) was significantly higher than that in the th normal pregnant women group(0.143 ± 0.009,P < 0.01). ③The expression of CHOP/GADD153 protein levels was significantly higher in preeclampsia group(16.210 ± 1.217)than in the normal pregnant group(2.710 ± 0.799,P < 0.01). Conclusion:Up regulation CHOP/GADD153 mRNA and excess expression of CHOP/GADD153 protein levels in placenta may be involved in the pathophysiological processes of preeclampsia.

Abstract:Staff’s basic medical insurance(SBMI)has made remarkable achievements as so far,but it also has some problems. Based on data from Shuyang county Jiangsu province,the major objective of the article is to research the general evaluation of the institution from population sociology indicators and satisfactory degree on the local policy and designated medical institution. The multiple regression analysis was used to find the relevant influencing factors. The result showed that the general satisfactory degree on SBMI of the participant was 3.7,and the following factors could deeply influence the satisfactory degree. They were the pay scale of the medicine and clinics project,the pay policies of participant,the range of medicines in medicine catalogue,the medical treatment charge of individual responsibility in the hospital,convenience of the procedure in hospital and so on.

Abstract:Objective: To investigate the expression of protease-activated receptor-2(PAR-2) and microvessel density(MVD) in normal cervix, carcinoma in situ of uterine cervix and invasive carcinoma of cervix. Methods:Immunohistochemistry method was used to examine the expressions of PAR-2 and MVD in 70 specimens of cervical squamous cell carcinoma, 20 carcinoma in situ of uterine cervix , and 15 normal cervical specimens. Result: The expressions of PAR-2 and MVD were higher in cervical invasive carcinoma than those in normal cervical tissues(P < 0.01). The expressions of PAR-2 and MVD in positive and negative lymph nodes metastasis of pelvic cavity were statistical significance(P < 0.01). The expression of MVD was significantly higher in PAR-2 positive group than that in negative group. Conclusion:PAR-2 and MVD are closely associated with the invasion and metastasis of cervical carcinoma.

Abstract:Objective:To investigate the mechanism of M-譈ller cell adhesion to vitreous gel. Methods:Porcine M-譈ller cells were extracted and cultured,identified with immuno-recognition with glial fibrillary acidic protein(GFAP),vimentin and glutamine synthetase(GS) antibody. Cells were collected and incubated with/without integrinα2β1 and CD44 antibody before application to fresh porcine vitreous for 2 hours,and then washed with PBS for 3 times. Remaining cells on the vitreous were counted. Results:① Cells collected were 97.7% positive for GFAP,94.6% positive for vimentin,86.0% positive for GS,respectively. Cells adhered to vitreous after 2 hours of culture,formed net-like structure and enter vitreous gel after 72 hours;② Pre-incubation with integrinα2β1 antibody significantly decreased the remaining cell,while no such significance was observed in the cells incubated with CD44 antibody. Conclusion:M-譈ller cell can adhere to vitreous and proliferate,in which the interaction with type Ⅱ collagen is an important mechanism.

Abstract:Objective: To study the association of N-terminal pro-brainnatriuretic peptide(NT-proBNP), tumor necrosis factor-α(TNF-α) levels and the extent of coronary artery disease(CAD). Methods:One hundred and twenty CAD patients and thirty-five non-CAD patients were included in this study. All of the subjects were verified with coronary angiography. In the light of the types of CAD, CAD patients were assigned into three groups (SAP group, UAP group, AMI group), the comparisons of NT-proBNP and TNF-α levels were performed among the SAP, UAP, AMI and non-CAD groups. According to the number of suffered coronary artery, CAD patients were divided into groups of single-vessel, double-vessel and three-vessel. The associations of the levels of NT-proBNP and TNF-α with CAD severity were also evaluated. Results:We found a significant association between the types of CAD and changes of NT-proBNP or TNF-α level. We also observed that change of TNF-α, not NT-proBNP, was significantly associated with the number of vessels of suffered coronary artery. Conclusion:NT-proBNP and TNF-α may be useful markers of the extent of CAD.

Abstract:Objective:To establish the evaluation of the median values for serum free β-human chorionic gonadotropin(β-HCG),pregnancy-associated plasma protein A(PAPP-A)and fetal nuchal translucency thickness(NT)in first-trimester prenatal screening in Nanjing region and investigate the standard program of the cutoff value for clinical soft-marker and its significance in prenatal diagnosis. Methods: The concentrations of β-HCG and PAPP-A in 1 684 gravidas with pregnancy of from 8 to 13+6 weeks were measured by time-resolved fluoroimmunoassay(TRFIA),and fetal NT value in their gestation from 11 to 13+6 weeks was determined by ultrasound scanning. Then the pregnancy outcomes of gravidas were followed up. Results: ① The median values ofβ-HCG in different gestigational week were 32% higher than their embedded parameters of software,those of PAPP-A were close to their embedded parameters in different gestigational week except in the 8th week of pregnancy,and those of NT values were 15% less than their embedded parameters;② when the positive rate of prenatal screening was 5%,the cutoff value of high risk in 21-trisome(Down’s Syndrome)was about 1∶550,and 4 cases with chromosome abnormalities were detected;③ The multiples of the median(MOM)values of serum PAPP-A with adverse pregnancy outcome in first-trimester prenatal screening were significantly lower than those with normal pregnancy outcome, and NT values were markedly higher than those with the normal pregnancy outcome. Conclusion: The standard cutoff values of the median soft-markers in serum and ultrasound scanning may be different in diverse regions,thus it is necessary to further investigate its applicability of the embedded parameters with foreign software;What′s more the MOM value of single soft-marker in first-trimester prenatal screening is important on the pregnancy outcome assessment,and it is benefit to detect other chromosomal abnormalities.

Abstract:Objective-structured clinical examination(OSCE) was applied to students of grade three during the implementing of basic surgery,through which students’ clinical skill could be evaluated. Some teachers’ and students’ opinions were drawn from face-to-face investigation,and a thorough recognition of the organization and outcome could be used to direct the improvement of OSCE and the clinical skill of students.

Abstract:Surgical practice teaching-and-learning is a eritical important part for training of medical talents. But the traditional way has many limits:laws and rules in medicine,coordination of patients,resources of teaching-and-learning and so on. For the reason mentioned above,the authors develop and construct the virtual surgery teaching-and-learning system,which has more interactions between computer and students,is operated easily and not restrained by the time and the place. By comparison with traditional way more attention can be triggered among students,and effects of teaching-and-learning on surgical practice can be strengthened.

Abstract:Objective: To study the feasibility of enucleation of leiomyoma of esophagus with diameter less than 1 centimeter using video-assisted thoracoscope. Methods:Total 8 patients were recruited, who had leiomyoma of esophagus with diameter less than 1 centimeter excised successfully using video-assisted thoracoscope between August 2006 and April 2010. There were 5 male and 3 female. Age ranged from 35 to 64 years with mean age 48 years old. The diameters of tumors ranged from 0.5 to 1.0 centimeter with mean value 0.72 centimeter. The tumor of 3 cases located in midpiece of esophagus, which had been excised successfully via right thorax, whereas another 3 cases with tumor located in inferior one third of esophagus and the other 2 cases with tumor located in the gastro-esophageal juncture, which had been excised successfully via left thorax. Results:There was no death in 8 cases, all recovering successfully. The operation cost 60 to 180 minutes with average of 120 minutes. There was no injury of mucous membrane of esophagus, and also no one was compelled to open-chest operation during surgery. No one had complications such as fistula, diverticulum and so on. Conclusion:It is safe and feasible for enucleation of leiomyoma of esophagus with diameter less than 1 centimeter using video-assisted thoracoscope.

Abstract:Objective: To evaluate the value of high-frequency ultrasonography(US)-guided 14- gauge(14G)core-needle biopsy(CNB)in diagnosing and typing lymphoma. Methods: Retrospective analysis was made on 30 patients diagnosed as lymphoma by pathology,and all of them had performed high-frequency US-guided 14G CNB. The locations of lesion involved neck,supra-clavicle,armpit,inguen,mammary glands,erect muscles,gluteal region,pars dorsalis and root of thigh. Pathology examination included hematoxylin and eosin staining and immunhistochemical assays. Results: Twenty-eight patients were diagnosed as non-hodgkin lymphoma(NHL)by high-frequency US-guided 14G CNB with accuracy of 93.3%(28/30),and two false-negative patients were found. We got the histologic subclassification in 26 cases of the 28 patients diagnosed as lymphomas on the basis of high-frequency US-guided 14G CNB. There were no complications of severity found in our study. Conclusion: The technique of high-frequency US-guided 14G CNB in diagnosing lymphoma has high diagnostic accuracy,high success rate,and low complication rate,and has important value in clinical application.

Abstract:Objective:To explore the value of intima-media thickness quantification technology and Sono CT in evaluating the vascular pathology of carotid artery in patients with type 2 diabetes mellitus. Methods: The carotid artery of 124 patients with type 2 diabetes mellitus and 82 healthy controls were detected respectively. The IMT of the the posterior wall of the carotid artery, and the presence of atherosclerotic plaque were observed. The patients were divided into DM1 group and DM2 group according to with or without hypertention, and the course of disease. Results: In DM1 with hypertention and DM2 without hypertention, the prevalence rates of carotid IMT are 81.5%, rates of carotid plaque are 78.2%, while in control group the data were 40.2% and 31.7%. There was significant difference between the two groups (P < 0. 05). The prevalence rates of carotid plaque of DM2 are higher than DM1, mean carotid IMT of DM2 which detected by QLAB are higher than that of DM1, with highly significant difference.The prevalence rates of carotid IMT and carotid plaque of patients with more than 5 years course of disease are higher than those patients with less than 5 years. Conclusion: The main change of carotid lesions in type 2 diabetes mellitus is the carotid IMT. The second change is carotid plaque. Sono CT technology can improve the resolution capability of the fine structure, increase the visibility of the rim of vascular lesions. The mean IMT measured by QLAB is superior to the maximum IMT in evaluating the vascular pathology of carotid artery in diabetes patients. It is especially valuable for the early diagnosis and guidance treatment of type 2 diabetes mellitus.

Abstract:Objective: To analyze the incidence rate and and management of adverse reaction in dual-source CT coronary arteriography(DSCT-CCTA). Methods: Total 1405 patients who received DSCT coronary angiography examination were enrolled in this study from Jan 1, 2010 to May 18, 2010. Considering both the allergic history and the pre-examination management, the incidence rate and the medical managements of adverse reaction were recorded and analyzed. Results: There were 79 patients who had a history of drug allergy or other allergic diseases. Adverse reaction occurred in 18 patients of those patients. The other 1 326 patients had no history of allergic disease, and adverse reaction occurred in 35 patients (P < 0.01). Among those patients of allergic history, 42 cases received pre-examination treatment of Dexamethasone. There was no significant difference of the incidence rate of adverse reaction between those patients with or without pre-examination treatment (P > 0.05). However, allergic shock occurred in two patients without pre-examination treatment of Dexamethasone. Those 53 patients with adverse reaction recovered after proper medical treatment. Conclusion:When adverse reaction occurs during the DSCT-CCTA, patients should be treated individually according to the degree of adverse reaction. Adverse reaction was highly correlated to allergic history. Pre-examination treatment of Dexamethasone could not reduce the incidence rate of adverse reaction significantly, but may relieve the degree of adverse reaction.

Abstract:Objective:To explore the effect factors of serve Hand-foot-mouth disease(HFMD)among children. Methods: A hospital-based case-control study was conducted on two groups of children matched by sex,age and hospital of time. Interview was carried out with uniform designed questionnaires between the case-group with serious complication and control-group with mild symptoms. Cox’s proportional hazards regression analysis was then performed. The pathogen of HFMD was confirmed by RT-PCR. Results: Time intervals between onset date and admission date(OR=1.668,95%CI:1.307-2.128),heating(OR=5.181,95%CI:1.939-13.844)-fever duration(OR=1.562,95%CI:1.142-2.135),stiff neck(OR=3.188,95%CI:1.055-9.634),the higher number of white blood cell(OR=1.038,95%CI:1.013-1.064)and neutrophilic granulocyte(OR=1.235,95%CI:1.108-1.378),and Enterovirus71 infection(OR=5.069,95%CI:2.472-10.393)were risk factors of HFMD among children with serious symptoms,However,lymph-cell counts increase(OR=0.968,95%CI:0.944-0.992),Children infected with CoxA16(OR=0.191,95%CI:0.094-0.388)were the protection factors. The risk factors were found with multivariate analysis,and were associated with time intervals between onset date and admission date(OR=1.668,95%CI:1.122-2.477),the higher number of white blood cell(OR=1.304,95%CI:1.080-1.575)and neutrophilic granulocyte(OR=1.088,95%CI:1.001-1.184),The protections factors was associated with CoxA16 infection(OR=0.169,95%CI:0.038-0.753).Conclusion: To strengthen clinical surveillance and etiology surveillance of HFMD disease,early diagnosis and early treatment can effectively prevent children developing severe hand-foot-and-mouth disease possibility.
