    • The role of intracellular protein O-glycosylation in cell adhesion and disease

      Abstract:Post-translational protein modification, including phosphorylation, is generally quick and reversible, facilitat-ing rapid biologic adjustments to altered cellular physiologic demands. In addition to protein phosphorylation, other post-translational modifications have been identified. Intracellular protein O-glycosylation, the addition of the simple sugar O-linked N-acetylglucosamine (O-GlcNAc) to serine/threonine residues, is a relatively recently identified post-translational modification that has added to the complexity by which protein function is regulated. Two intracellular enzymes, O-GlcNAc transferase and O-GlcNAcase, catalyze the addition and removal, respec-tively, of O-GlcNAc to serine and threonine side-chain hydroxyl groups. Numerous proteins, including enzymes, transcription factors, receptors and structural proteins have been shown to be modified by intracellular O-glyco-sylation. In this review, the mechanism and relevance of O-GlcNAc protein modification are discussed in the con-text of cell adhesion and several representative diseases.

    • Activation of Rac1-PI3K/Akt is required for epidermal growth factor-induced PAK1 activation and cell migration in MDA-MB-231 breast cancer cells

      Abstract:Epidermal growth factor (EGF) may increase cell motility, an event implicated in cancer cell invasion and me-tastasis. However, the underlying mechanisms for EGF-induced cell motility remain elusive. In this study, we found that EGF treatment could activate Ras-related C3 botulinum toxin substrate 1 (Rac1), PI3K/Akt and p21-actived kinase (PAK1) along with cell migration. Ectopic expression of PAK1 K299R, a dominant negative PAK1 mutant, could largely abolish EGF-induced cell migration. Blocking PI3K/Akt signalling with LY294002 or Akt siRNA remarkably inhibited both EGF-induced PAK1 activation and cell migration. Furthermore, expression of dominant-negative Rac1 (T17N) could largely block EGF-induced PI3K/Akt-PAK1 activation and cell migration. Interestingly, EGF could induce a significant production of ROS, and N-acetyl-L-cysteine, a scavenger of ROS which abolished the EGF-induced ROS generation, cell migration, as well as activation of PI3K/Akt and PAK, but not Rac1. Our study demonstrated that EGF-induced cell migration involves a cascade of signalling events, in-cluding activation of Rac1, generation of ROS and subsequent activation of PI3K/Akt and PAK1.

    • Aspirin suppresses growth of human gastric carcinoma cell by inhibiting survivin expression

      Abstract:Regular use of aspirin (ASA) could reduce the risk of gastric cancer although the precise mechanism remains unclear. Down-regulation of survivin may be one of the cyclooxygenase-independent mechanisms whereby ASA induces apoptosis of gastric cancer cell. In this study, we investigated the effect of ASA on the growth, apoptosis and survivin expression of gastric cancer cell line SGC7901. The survival of cells treated with 3.0 and 10.0 mmol/L ASA for 24 h was decreased by 44.6% and 88.5%, respectively. ASA at 3.0 mmol/L inhibited the viability of SGC7901 cells in a time-dependent manner. Apoptosis analysis showed similar results with MTT assay. ASA at 3.0 and 10.0 mmol/L decreased the mRNA transcript levels of survivin and reduced survivin protein levels in SGC7901 cells also in a time-dependent manner. Our findings indicated that ASA inhibited the proliferation of SGC7901 by sup-pressing survivin at both the transcriptional and translational level.

    • Effect of water storage on fluoride release and mechanical properties of a polyacid-modified composite resin (compomer)

      Abstract:We evaluated the effect of water storage on fluoride release and mechanical properties of compomer restorative material. Fluoride release was recorded using a specific fluoride electrode. Flexural properties and fracture tough-ness were measured using a universal testing machine. Vickers hardness was measured using a micro-hardness tester. There was initial burst of fluoride release up to 1 w, which was diminished to a low level in 1 mon and remained relatively constant over 6 mon. Flexural strength and hardness were increased up to 1 mon followed by a gradual decrease up to 6 mon. Flexural modulus was decreased gradually up to 6 mon. Fracture toughness was increased during the first week and gradually decreased over the storage period. We concluded that flexural prop-erties, fracture toughness, Vickers hardness and fluoride release of compomer were sensitive to water as well as storage time. There was a significant effect of fluoride release on the studied mechanical properties.

      Abstract:Cambridge University and Oxford University have been implementing their distinctive educational pattern of collegiate system. Two types of parallel school or college, one for teaching, research, social services, science and technology development and other acedemic activities, named “school”, or “faculty” or “department” and the other for undergradautes’ educational activities and talent training, named “college”, are established in university. Colleges are responsible for enrolling students who major in different specialties and offer the students who live and study in the same college academic guidance and life coaching. Simultaneously employed in a certain college, the majority of course’s staff in a school/department have dual attributes, tutoring small-classed undergraduates and guiding both their life and academic activities. This educational pattern offers undergraduates of different specialties and backgrounds a growing environment of mutual communiciation and learning, builds a strong bond between course’s staff and students by getting involved in their students’ study and offering them comprehensive guidances in the course of their development, and establishes a talent training platform based on human-orientedness, persoanlized instruction and individuality development. The connotation of the educational pattern of collegiate system is just the core value needed to be remodelled and solidified in the higher education reform and construction in our country.

    • Localization of epileptic foci in Children with childhood absence epilepsy by magnetoencephalography combined with synthetic aperture magnetometry

      Abstract:This present study was aimed to investigate the localizable diagnostic value of magnetoencephalography (MEG) combined with synthetic aperture magnetometry (SAM) in childhood absence epilepsy (CAE). Thirteen CAE pa-tients underwent MEG detection at resting state and after hyperventilation, and then the epileptic foci were located by SAM. In the thirteen CAE patients, epileptic foci were found in five cases (38.5%) , and they were all located in the bilateral frontal lobe, suggesting that the frontal lobe in some CAE patients may serve as the epileptic foci. Our findings indicate that MEG combined with SAM could be of diagnostic value in localizing the epileptic foci in certain CAE patients.

      Abstract:This research investigated the treatment experience of patients in 38 township hospitals of Shuyang during July 2010 to September 2010 by spot-questionnaires, to analyze the satisfaction on township hospitals and understand the quality of health care serves of Shuyang township hospitals and non-governmental hospitals’ image among general population. The highest score item is the comfort level, followed by convenience and safety, economy got the lowest score. Shuyang township hospitals should find the exact position, make a development plan; build brands with fidelity, strengthen doctor’s professional ethics; improve diagnosis and treatment levels; control cost of patients.

    • ApoB/apoA1 is an effective predictor of coronary heart disease risk in overweight and obesity

      Abstract:We investigated the relationship of apoB/apoA1 ratio and coronary heart disease (CHD) in persons who were overweight or obese. The subjects were divided by the body mass indexes (BMI) into the normal weight group (n=397, BMI<24 kg/m2) and the overweight group (n=400, BMI>24 kg/m2). Our results showed that the over-weight group had higher blood pressure [(130.15±19.01) mmHg vs (123.66±18.70) mmHg] and higher levels of blood sugar [(7.09±2.89) mmol/L vs (6.21±2.59) mmol/L], triglyceride [(1.93±1.19) mmol/L vs (1.44±0.85) mmol/L], total cholesterol [(4.26±1.06) mmol/L vs (4.09±0.99) mmol/L], low-density lipoprotein cholesterol (LDL-C) [(2.56±0.75) mmol/L vs (2.39±0.72) mmol/L], and apoB [(0.83±0.27) mg/L vs (0.78±0.23) mg/L], and a higher apoB/apoA1 ratio (0.83±0.27 vs 0.75±0.25) and lower levels high-density lipoprotein cholesterol [(1.10±0.26) mmol/L vs (1.21±0.31) mmol/L] and apoA1 [(1.04±0.20) mg/L vs (1.08±0.22) mg/L] than those of the normal weight group (all P < 0.05). The prevalence of CHD in the over-weight group in the lowest LDL quar-tile was almost twice greater than that of the highest apoB/apoA1 quartile, compared with the subjects in the low-est apoB/apoA1 quartile. The higher apoB/apoA1 quartile was in agreement with the higher prevalence of CHD. In the overweight and obesity group, the area under ROC curve (AUC) was the highest for apoB/apoA1 (0.655). The cut-off point of apoB/apoA1 for optimal sensitivity and specificity was at 0.80, with a sensitivity of 57.19% and a specificity of 71.72%. In conclusion, apoB and apoA1 were simple clinical indicators, and the apoB/apoA1 ratio was closely related with CHD in overweight and obese patients. The apoB/apoA1 ratio may provide some useful information in the differential diagnosis.

      Abstract:Integration of statistical data, related literatures and policy documents were conducted to analyze the present situation of urban community doctor in Nantong, to find out problems and provide evidences of further improving measurements. The group of urban community doctors in Nantong is insufficient and of low quality. In addition, there are also shortages in the training and incentive mode which is applied in the training of community doctors. All above are obstacles of improving the service ability of community doctors. Therefore, the following measures are suggested: to take directional training plan and priority policies to establish incentive policies, to improve the doctors’ treatment and the performance appraisal policy, to ascend social status of urban community doctors and to increase opportunities of career development, to conseguently establish a stable and expanded medical group for urban community, which could pave the way to cultivate and bring in more talents to the community and enhance the corresponding service quality.

    • Bisoprolol reverses down-regulation of potassium channel proteins in ventricular tissues of rabbits with heart failure

      Abstract:Remodeling of ion channels is an important mechanism of arrhythmia induced by heart failure (HF). We in-vestigated the expression of potassium channel encoding genes in the ventricles of rabbit established by volume-overload operation followed with pressure-overload. The reversible effect of these changes with bisoprolol was also evaluated. The HF group exhibited left ventricular enlargement, systolic dysfunction, prolongation of cor-rected QT interval (QTc), and increased plasma brain natriuretic peptide levels in the HF rabbits. Several potas-sium channel subunit encoding genes were consistently down-regulated in the HF rabbits. After bisoprolol treat-ment, heart function was improved significantly and QTc was shortened. Additionally, the mRNA expression of potassium channel subunit genes could be partially reversed. The down-regulated expression of potassium channel subunits Kv4.3, Kv1.4, KvLQT1, minK and Kir 2.1 may contribute to the prolongation of action potential duration in the heart of rabbits induced by volume combined with pressure overload HF. Bisoprolol could partially reverse these down-regulations and improve heart function.

    • Cardioprotective activity of alcoholic extract of Tinospora cordifolia (Willd.) Miers in calcium chloride-induced cardiac arrhythmia in rats

      Abstract:The present study investigated the antiarrhythmic activity of alcoholic extract of Tinospora cordifolia (T. cor-difolia) in CaCl2 induced arrhythmia. CaCl2 (25 mg/kg) was administered by intravenous infusion (iv) to produce arrhythmia in rats. The animals were then treated with T. cordifolia extract (150, 250, and 450 mg/kg) and vera-pamil (5 mg/kg,iv). Lead II electrocardiogram was monitored. Plasma calcium, sodium and potassium levels were measured. In CaCl2 induced arrhythmia, heart rate was decreased by 41.10%, T. cordifolia at 150, 300, and 450 mg/kg decreased the heart rate by 26.30%, 29.16%, and 38.29%, respectively, and verapamil reduced the heart rate by 9.70% compared to the normal group. The PQRST waves were normalized and atrial and ventricular fi-brillation was controlled in rats treated with verapamil and T. cordifolia. CaCl2 increased calcium and sodium lev-els and decreased potassium levels in blood. T. cordifolia dose-dependently decreased calcium and sodium levels and increased potassium levels. Hence, T. cordifolia can be used in antiarrhythmic clinical settings and beneficial in atrial and ventricular fibrillation and flutter and may be indicated in ventricular tachyarrhythmia.

    • Relationship between nerve injury-induced protein gene 2 polymorphism and stroke in Chinese Han population

      Abstract:The aim of present study was to investigate the relationship between nerve injury-induced protein 2 (NINJ2) gene polymorphism and stroke in Chinese Han population. Fifty-two patients with large-artery atherosclerosis (LAA) infarction, 85 patients with small-artery occlusion lacunar (SAO) infarction, 50 patients with intracerebral hemorrhage (ICH) and 66 controls were included. Genotypes and alleles frequencies of the two single nucleotide polymorphisms (SNPs) of NINJ2 among different groups were analyzed and compared. In regard to rs12425791, the frequencies of the AG and AA+AG genotypes of the LAA and SAO groups were significantly higher than those in the control group; the frequency of the A allele of the SAO group was significantly higher than that of the con-trol group. In regard to rs11833579, there were not any significant differences between the case and the control groups. The SNP rs12425791 is significantly associated with ischemic stroke, and the A allele increases the sus-ceptibility to stroke. The SNP rs11833579 is not significantly associated with stroke.

    • Effects of minocycline on the expression of NGF and HSP70 and its neuroprotection role following intracerebral hemorrhage in rats

      Abstract:The present study was aimed to investigate the effects of minocycline (MC) on the expression of nerve growth factor (NGF) and heat shock protein 70 (HSP70) following intracerebral hemorrhage (ICH) in rats, and explore the neuroprotective function of MC. Seventy-eight male SD rats were randomly assigned to three groups: the ICH control group (n = 36), ICH intervention group (n = 36) and sham operation group (n = 6). The ICH control group and ICH intervention group were subdivided into 6 subgroups at 1, 2, 4, 5, 7 and 14 d after ICH with 6 rats in each subgroup. Type IV collagenase was injected into the basal nuclei to establish the ICH model. All rats showed symptoms of the nervous system after the model was established, and the sympotsm in the ICH control group were more serious than the ICH intervention group. The number of NGF-positive cells and HSP70-positive cells in the ICH intervention group was higher than that of the ICH control group. MC administration by intraperito-neal injection can increase the expression of NGF and HSP70. MC may inhibit the activation of microglia, the in-flammatory reaction and factors, matrix metalloproteinases and apoptosis, thus protecting neurons. The change of the expression of NGF and HSP70 may be involved in the pathway of neuroprotection by MC.

      Abstract:Eysenck Personality Questionnaire Revised short Scale for Chinese(EPQ-RSC)and subjective well-being index scale were applied to investigate the personality of college students and the relation of their personality and subjective well-being from 236 college students in Nanjing. Extraversion was affected significantly by environment. The college students from urban were easier to be extravert than that from countryside. The relation of learning stress and neuroticism was very significant. There is significant difference of neuroticism among different types of college students. There is significant difference of extraversion and neuroticism among different healthy states college students. The economic of college students did not affect the traits of their personality significantly. The relation of extraversion and subjective well-being was very significant. The relation of neuroticism and subjective well-being was very significant. The degree of the relations is in the middle level, but the forward is positive and the latter is negative. The relation of the traits of the personality of college students and their subjective well-being is close.

      Abstract:objective:to develop a sandwich elisa system for quantitative measurement of the human connective tissue growth factor(ctgf), and to detect the sera ctgf level of human. methods:after labeled the purified monoclonal antibodies(mabs) with horseradish peroxidase (hrp) by sodium oxidation method, the antibody-pair matching(for capturing and detection of antigen, respectively) was performed using anti-ctgf mab as coating antibody and hrp labeled anti-ctgf mab as labeled antibody,the optimal concentrations of anti-ctgf mabs were defined by titration. standard curve was performed using purified ctgf protein,the effectiveness of this sandwich elisa system was evaluated by sensitivity, reproducibility and recovery rate. the ctgf levels in sera of health people, hbsag(+) without hepatocellular carcinoma (hcc) patients and hbsag(+) with hcc patients were measured by this system. results:the optimal paired antibodies were anti-ctgf mab 3h5 and hrp labeled anti-ctgf mab 4c5 with optimal concentrations of 40 μg/ml and 1∶4 000 respectively. the sensitivity of this elisa system was 1.46 ng/ml. the average coefficients of variations were 4.59% within assay and 5.70% between assays. the recovery rate was (98.73±4.84)%. the ctgf levels in sera of 40 health people, 76 hbsag(+) without hcc patients and 35 hbsag(+) with hcc patients were [2.65(2.16±3.18) ng/ml], [5.53(2.74±8.23)ng/ml],[8.77(4.81±13.65) ng/ml],respectively. the difference among these groups showed great significance(p < 0.001). conclusion:a sandwich elisa system for quantitative measurement of human ctgf was successfully established. the serum level of ctgf in hbsag(+) with hcc patients raises significantly compare with hbsag(+) without hcc patients and normal group. this suggests that ctgf may be a potential biomarker for the forewarning of hbsag (+) with hcc.

      Abstract:objective:to evaluate the expression level and clinical significance of adiponectin gene on abdomen adipocyte in the patients with gastrointestinal cancer. methods:the study included the samples from surgical patients who were mainly divided into two groups, the cancer group(n=39) and the control group(n=20). then the gastrointestinal cancer group contained the metastasic group (n=21) and the no-metastasic group(n=18). total rna were collected from adipocyte, and the expression level of adiponectin have been detected with real-time pcr. results:the expression level of adiponectin mrna in cancer group was significantly lower than control group(p < 0.05), and the expression level in the metastasic group(3.85±1.41) was lower than the no-metastasic group(4.90±1.06), but there was no significance(p > 0.05). conclusion:the expression level of abdomen adipocyte adiponectin gene may be involved in the gastrointestinal tumor generation, which can provide theoretical foundation for a new treatment.

      Abstract:objective:to develop a 5-fluorouracil(5-fu)-resistant human pancreatic cancer cell line patu8988/5-fu and investigate its biological characteristics and the preliminary mechanisms of chemoresistance in pancreatic cancer. methods:patu8988 was exposured to increased 5-fu progressively for 10 months to obtain stable 5-fu-resistant patu8988/5-fu in vitro. the morphological features and growth characters of patu8988/5-fu were observed. mtt assay was used to observe its sensitivity of drug resistance and cross resistance. the growth curve was observed and the doubling time was calculated. flow cytometry assay was used to evaluate the cell cycle distribution,apoptosis rate and accumulation of rh123 in two cell lines. the expressions of hif-1α and mdr1 mrna were detected by rt-pcr. results:the acquired multidrug resistance(mdr) cell line of patu8988/5-fu was established successfully. compared with the parental cell,patu8988/5-fu showed great changes in biological characteristics. the drug resistance indexes of cell line patu8988/5-fu to 5-fu, adriamycin (adm) and gemcitabine were 36.98,2.76 and 2.42,respectively. compared with the parental cell,patu8988/5-fu cell exhibited a lower growth rate,prolonged population doubling time,and cell distribution decreased in s phase and increased in g2/m phase. the expressions of hif-1α and mdr1 mrna were significantly higher,while the accumulation of rh123 in cytoplasm and the apoptotic ratio after treated with different concentration of 5-fu were significantly lower in patu8988/5-fu cell than in the parental cell. conclusion:the acquired mdr cell line of patu8988/5-fu was established successfully after exposure to increased 5-fu. hif1α maybe contribute to acquirement of drug resistance by inhibiting apoptosis and upregulating the expression of the main drug resistant related genes mdr1.

      Abstract:objective:to study the effect and mechanism of sphingosine kinase inhibitor ski-Ⅱ on the apoptosis of human gastric cancer cell sgc7901. methods:sgc7901 cells were cultured and treated with ski-Ⅱ alone or in combination with cisplatin(ddp).the apoptosis of the tumor cells was analyzed by flow cytometry;the morphological changes were observed by electron microscopy;the expression of sphk1,nf-κb,bcl-2 and bax were detected by immunocytochemistry and western blot. pearson analysis was used to detect the correlations between sphk1 and nf-κb,nf-κb and bcl-2. results:after treated for 48 h,the apoptosis rates in sgc7901 treated with ski-Ⅱ 5,10 μmol/l,ddp 2.5 mg/l,ddp 2.5 mg/l+ski-Ⅱ 5 μmol/l and ddp 2.5 mg/l+ski-Ⅱ 10 μmol/l were (40.39±1.06)%,(45.58±0.75)%,(47.27±1.13)%,(53.64±1.11)% and (66.98±2.32)%,respectively,compared with the control group[(18.46±1.64)%],the differences were significant(p < 0.05). apoptotic bodies were found in sgc7901 cells by electron microscopy. the expression of bax was increased,but the expression of sphk1,nf-κb and bcl-2 was decreased. and the correlations between sphk1 and nf-κb,nf-κb and bcl-2 were positive. conclusion:ski-Ⅱ could induce the apoptosis of sgc7901 cells through down-regulating the expression of sphk1 and nf-κb,and increasing the ratio of bax/bcl-2.

      Abstract:objective:to observe the radiosensitization effects of human recombinant endostatin(endostar) combined with radiation by establishing subcutaneous transplantation tumor model of nasopharyngeal carcinoma in balb/c nude mice. methods:the nasopharyngeal carcinoma model in balb/c nude mice was established by subcutaneous transplantation of cne1 cell. the diameters of the tumor in the control group(group a), endostar treated group(group b), radiation treated group(group c) and endostar combined with radiation group(group d) were measured every 4 days, the growth curves of tumor were observed, tumor volumes and expected value/observed value(e/o) were calculated. the inhibition rates of tumor by endostar and(or) radiation were calculated according to tumor weight. endostar was injected around tumor 20 mg/kg daily for 10 days, the irradiation dose of local tumor was 20 gy. the expression of hif-1α in tumor was detected by immunohistochemistry staining, apoptosis of tumor cells was observed by transmission electron microscopy. results:in combination with radiotherapy, endostar exhibited significant inhibition of nasopharyngeal carcinoma, the average tumor size of group d[(765.697±78.725)mm3] was significantly lower than group b [(2340.186±370.606)mm3], group c [(1454.028±180.371)mm3] and group a [(3112.452±720.764)mm3] after treated for 4 weeks, tumor weight of group d [(0.523±0.180)g] was lower than group b [(1.294±0.346)g], group c [(0.926 ± 0.233)g] and group a [(1.849±0.596)g,p < 0.05], and the inhibition rate of group d(86.11%) was significantly higher than group c(60.45%) and group b(27.12%). immunohistochemistry staining showed the expression of hif-1α decreased in endostar treated group, transmission electron microscope observed the decreased number of tumor cells and significantly increased apoptosis in group d compared with other groups. conclusion:endostar inhibits the growth of nasopharyngeal carcinoma in nude mice and has radiosensitization effect.

      Abstract:objective:to observe the proliferation and differentiation of xenogeneic bone mesenchymal stem cells (bmscs) in the thymus and to explore the feasibility of intrathymic inoculation with xenograft-bmscs in inducing immunotolerance. methods:bmscs of guinea pigs were cultured in medium to p3,and labeled with fetal bovine serum-fe2o3 and 4,6-diamidino-2- phenylindole(dapi).the marked bmscs were inoculated into thymuses gland of rats. the thymuses were sliced up and dyed with perls solution seven days later and observed directly under the fluorescenc microscope. results:bmscs survived long term in the micro-environment of the xenogeneic thymuses,and had the ablitity to differentiate into various cell types. the xenogeneic bmscs had the capablilty of multiple rounds of division. conclusion:it is feasible to induce immune tolerance by intrathymic inoculation with xenograft-bmscs.

      Abstract:objective: to explore the effects of saturated fats diets,polyunsaturated fats diets and monounsaturated fats diets on the expression of x-box binding protein 1(xbp-1)in liver tissues of rats. methods:wistar rats(n=48)were randomly divided into normal control group(n),saturated fatty acid diets group(s),monounsaturated fatty acid diets group(m),polyunsaturated fatty acid diets group (p). the rats in group n were fed with a regular lower fatty acids diet containing 10.3% fat,24.2% protein,and 65.5% carbohydrate as percentage of total calories. the rats in group m,p and s were fed with regular diets mixed with 15% tea oil,soybean oil,lard oil respectively,containing 35.4% fat,17.4% protein and 47.2% carbohydrate as percentages of total calories. the blood sample was collected by cardiac puncture after rats were anesthetized with diethyl ether for the biochemical analysis,insulin resistance was evaluated by glucose infusion rate (gir) of hyperinsulinemic euglycemic clamp technique. after 8 weeks,the rats were killed with phenobarbital sodium after hyperinsulinemic euglycemic clamp test,hepatic xbp-1 were detected by western blot and immunohistochemistry. results:① compared with group n,the amounts of food intake were significantly lower in group s,m and p(p < 0.05). adjusted for the amount of food intake,serum tc,tg in group s were significantly higher than group n(p < 0.05),but there was no detectable difference among n,p and m groups(p > 0.05). compared with group n,fins and fbg were significantly higher in group s and p groups(p < 0.05),gir was significantly lower in s,p and m groups(p < 0.05). ②immunohistochemistry analysis showed positive xbp-1 localized in both cytoblast and cytoplasm with intense brown staining. further more,western blot for xbp-1 in liver tissues showed that xbp-1 protein levels increased in group p and s [(1.09±0.16),(1.15±0.21)]compared with group n and m [(0.85±0.15),(0.79±0.14),p < 0.05]. conclusion:it may be one of the mechanisms of insulin resistance that the higher expression of xbp-1 and in liver tissues induced by polyunsaturated fatty acid and saturated fatty acid diets,simultaneously,it suggested that endoplasmic reticulum stress(ers)can be a core mechanism involved in triggering insulin resistance.

      Abstract:objective:to study the relationship of protection of telomeres 1(pot1) mrna expression,telomerase activity in lymphocytes with the development of type 2 diabetic mellitus(t2dm). methods:one hundred and sixty-two cases with t2dm were test group,162 cases of healthy were control group,real-time quantitative pcr was used to analysis pot1 mrna expression,semi-quantitative trap-elisa was used to analysis telomerase activity. results:the expression of pot1 mrna and telomerase relatively activity in test group were obviously higher than those in control group(p < 0.01). meanwhile,the pot1 mrna expression and telomerase activity were significantly higher in t2dm patients with passable or poor controlled blood glucose compared with controls(p < 0.01),and they were also significantly higher in t2dm patients with different stage of diabetic retinopathy compared with controls(p < 0.01),significant differences also existed among the patients whose time course of t2dm between 5 to 10 years or above 10 years and time course of t2dm less than 5 years or controls(p < 0.01). conclusion:pot1 mrna expression and the telomerase activity were significantly increased with the progression of t2dm;they might be the important evidences in disease prognosis.

      Abstract:objective:to investigate the effect of metformin on the proliferation of rat mesangial cells induced by high glucose and the possible mechanism. methods:the rat messangial cells (hbzy-1) were cultured in vitro. ①cultured hbzy-1 cells were divided into four groups:normal glucose(5.6 mmol/l),normal glucose plus metformin(200 mmol/l),high glucose(30 mmol/l),high glucose plus metformin at 50,100,200,400 mmol/l,respectively,and cultured for 6,12,24 h. the proliferation of hbzy-1 cells was detected by micro-tetrazolium salt colorimetric enzyme reaction test(mtt). ②cultured hbzy-1 cells were divided into four groups:normal glucose(5.6 mmol/l),normal glucose plus metformin(200 mmol/l),high glucose(30 mmol/l),high glucose plus metformin(200 mmol/l),and cultured for 12 h. the protein levels of tgf-β1,p-ampk and ampk were measured by western blot. results:as compared to normal cells,exposure of hbzy-1 cells to d-glucose at concentration of 30 nmol/l caused a time-dependent increase in the proliferation of hbzy-1 cells and reached the highest level at approximately 12 h. the addition of metformin significantly attenuated the high glucose-enhanced proliferation of hbzy-1 cells (p < 0.01) in a dose-dependent manner. but no notable difference was found between normal glucose plus metformin group and normal glucose group(p > 0.05). as compared to normal cells,the levels of tgf-β1 protein significantly increased,and the levels of p-ampk significantly decreased in high glucose group(p < 0.01). metformin significantly attenuated the increase of tgf-β1 expression and increased the expression of p-ampk in hbzy-1 cells exposured to high d-glucose(p < 0.01). conclusion:high glucose contributes to an increased proliferation of hbzy-1 cells. metformin can significantly reverse high glucose up-regulated proliferation of hbzy-1 cells through inhibiting the expression of tgf-β1 protein and increasing the expression of p-ampk protein in hbzy-1 cells.

      Abstract:objective: to investigate the effect of iptakalim(ipt),a novel atp sensitive potassium(katp) opener,on the expression of proliferating cell nuclear antigen (pcna) mrna in primary cultured human pulmonary artery smooth muscle cells. methods:under germfree conditions,the smooth muscular layers were separated from human pulmonary arterioles(3rd~4th divisions). human pulmonary artery smooth muscle cells were cultured by tissue block method and were divided into following groups:control group,hypoxia group,hypoxia+iptakalim or pinacidil group and hypoxia+iptakalim or pinacidil+glibenclimide group. fluorescence quantitative reverse transcription-polymerase chain reaction(fq-rt-pcr)was used to measure the expression level of pcna mrna. results:the expression of pcna mrna in hypoxia group increased(3.90±0.24) times higher than that in control group. ipt at the concentrations of 10-7,10-6 and 10-5 mol/l lowered the expression of pcna mrna by (26.00±2.01)%,(55.33±4.04)%,(79.00±1.00)%,respectively,compared with that in hypoxia group(p < 0.01). glibenclimide (gli) at the concentration of 10-5 mol/l antagonized the effect of ipt with the same concentration on the expression of pcna mrna,compared with that in hypoxia + ipt 10-5 mol/l group(p < 0.01). conclusion:the expression of pcna mrna in human pulmonary artery smooth muscle cells under hypoxic conditions increased,while ipt has an inhibitory effect on the expression of pcna mrna in a concentration-dependent manner through activating katp channels.

      Abstract:objective:to observe the effects of lithium chloride(licl)pretreatment on remifentanil-induced hyperalgesia in rats model of incisional pain. methods:fourty-two male sprague dawley(sd) rats were randomly divided into seven groups(n=6):control group(c),incisional pain group(i),remifentanil+ incisional pain group(r),incisional pain+licl group(il),remifentanil+incisional pain+low-dose licl group(rl1),remifentanil+incisional pain+medium-dose licl group(rl2),remifentanil+incisional pain+high-dose licl group(rl3). licl 0.4 ml were administered to il group and rl1~3 groups intraperitoneally ten minutes before anesthesia with sevoflurane,the dose of licl in il group and rl1~3 groups were 36,9,18,36 mg/kg respectively,accordingly saline were administered intraperitoneally to group c,i and r. r group and rl1~3 groups were pumped subcutaneously with remifentanil(40 μg/kg)at the speed of 0.8 ml/h at fifteen minutes after anesthesia with sevoflurane,while group c,i and il were pumped subcutaneously with saline accordingly. all groups except c were made the model of incisional pain. at t0(24 hours before operation)and t1(6 hours after operation),t2(24 hours after operation),t3(48 h after operation)respectively,paw withdrawal thermal latency(pwtl) and paw mechanical withdrawal threshold(pmw) were detected on the rats’ right hindpaw in all groups. results:at t1,t2,t3,the pmw and pwtl were significantly lower in group r and significantly higher in group il then those in group i(p < 0.05). comparing with group r,the pmw and pwtl in rl groups were significantly higher(p < 0.05). conclusion:in rat model of incisional pain,remifentanil could induce hyperalgesia on the area of adjacent to the incision;lithium chloride could increase the pain threshold of incision and prevent the remifentanil-induced hyperalgesia.

      Abstract:objective:to observe the effect of electro-acupuncture(ea) on serum testosterone(t) and spermatogenesis in normal rests at different developmental stages. methods:sprague-dawley rats were assigned into four groups:juvenile group (jg),early-puberty group (epg),later-puberty group (lpg)and adult group(ag). each group was subdivided into four teams:ea at acupoints(ewa),ea at non-acupoints(ena),immobilization control(ic),normal control(nc). ea(3 hz)at ewa or ena was performed for 20 min per day in different developmental group of rats for 10 days. sperm number was counted and serum level of t was measured after ea treatment. results:the results showed that the serum level of t in the epg,lpg and ag rats were significantly depressed(p < 0.05). sperm counts in the epg,lpg and ag groups were also significantly reduced. conclusion:the results suggested that repeated stimulation with ea is a good option to reduce the serum t and sperm quantity of male rats after the puberty period.

      Abstract:objective:to investigate the antibacterial activity of grapefruit seed extract mouthwash against streptococcus mutans and explore the potential mechanism. methods:the minimal inhibitory concentration (mic) and the minimal bactericidal concentration(mbc) of the grapefruit seed extract mouthwash against s.mutans were detected using double dilution. and the cell wall alteration of s.mutans treated with grapefruit seed extract mouthwash was observed by using a scanning electron microscope(sem). results:the mic and mbc determined for grapefruit seed extract mouthwash were 46.875 mg/l and 93.75 mg/l. both rupture of the cell wall and release of the intracellular components were observed in the grapefruit seed extract mouthwash-treated group by sem. conclusion:the grapefruit seed extract mouthwash can inhibit the growth of s.mutans and kill them in vitro,this suggests that grapefruit seed extract mouthwash has an antibacterial action on the s.mutans by affecting the normal structure and function of the cell wall of s.mutans.

      Abstract:objective:to analyze hla-a,b and drb1 alleles high-resolution polymorphism and haplotypes in the han population in jiangsu province. methods:pcr-ssop and pcr-sbt methods were applied for hla high-resolution genotyping of 644 unrelated healthy donors in jiangsu branch of chinese national marrow donor program (cmdp) registry. the allele frequencies at hla-a,b,and drb1 loci were estimated by direct counting method and the haplotype frequencies were estimated using arlequin software 3.01. results:a total of 29 alleles for hla-a,60 alleles for hla-b,and 36 alleles for hla-drb1 were found. the most common alleles were a*1101(17.86%),b*4601(9.55%)and drb1*0901(15.76%). the most common a-b-drb1 haplotype was a*3001-b*1302-drb1*0701(5.98%),the most common a-b haplotype was a*3001-b*1302(6.68%),the most common b-drb1 haplotype was b*1302-drb1*0701(7.22%)and the most common a-drb1 haplotype was a*3001-drb1*0701(6.06%). conclusion:the high-resolution results show the characteristics of hla distribution in jiangsu han population,and it is helpful in hla matching in transplantation,as well as to population genetics and disease association studies.

      Abstract:objective:to investigate the associations between potentially functional polymorphisms of interleukin 23 receptor(il23r) and esophageal cancer risk. methods:a case-control study with 1 045 esophageal cancer cases and 1 047 cancer-free controls frequency matched by age and gender was conducted,and the genotypes were determined by the taqman assay. logistic regression analysis was used to evaluate the odd ratios(ors)and 95% confidence intervals(cis)of rs1884444 t>g and rs6682925 t>c with susceptibility of esophageal cancer. results:the il23r rs1884444 tg/gg variant genotypes were associated with a significantly increased risk of esophageal cancer(adjusted or=1.20,95%ci=1.01~1.43),compared with tt genotype,il23r rs6682925 tc/cc genotypes were associated with a marginal increased risk of esophageal cancer(adjusted or=1.17,95%ci=0.98~1.40) compared with tt genotype,and the risk effects were more predominant in young subjects(age≤60 years) and no-drinkers for both two snps. conclusion:our findings indicated that il23r rs1884444 and rs6682925 may contribute to the risk of esophageal cancer in chinese.

      Abstract:objective:to investigate the association between the mir-146a gene polymorphism(rs2910164) and the susceptibility of ovarian cancer in jiangsu province. methods:in a hospital-based case-control study of 131 patients with newly diagnosed ovarian cancer and 221 cancer-free controls frequency-matched by age,the polymorphism of mir-146a gene was analyzed by polymerase chain reaction-restriction fragment length polymorphism(pcr-rflp). the risk of ovarian cancer was compared between carriers of different genotypes. age,age at menarche,parity,number of abortions and menopausal status of cases were further stratified to investigate their effects on the risk of ovarian cancer. results:no significant difference was found between the mir-146a polymorphism of patients and of controls. conclusion:our results showed that the mir-146a g>c polymorphism may not contribute to the risk of ovarian cancer in jiangsu province.

      Abstract:objective:to investigate the susceptibity of c-reaction protein(crp) gene c+1444t polymorphisms to acute coronary syndrome (acs). methods:from nov 2003 to may 2008, 443 inpatients with acs (acs group) were recruited, the control group was consisted of 285 subjects who were free of coronary artery disease confirmed by coronary angiography. the crp gene c+1444t polymorphisms were determined by polymerase chain reaction and restriction fragment length polymorphisms analysis. results:there were no significant differences in the frequencies of cc, ct and tt genotype between acs group and control group (83.7% vs 86.7%, 14.4% vs 12.3% and 1.8% vs 1.1%,p were 0.292, 0.439 and 0.540, respectively). the prevalence of c allele were 91.0% and 92.8% for acs group and control group, respectively(p=0.243). furthermore, after adjusted for multiple factors such as age, sex, smoking, systolic pressure, diastolic pressure, blood glucose, and blood lipids, the multivariate logistic regression analysis indicated that there was no significant correlation between the crp gene c+1444t polymorphisms and acs(p > 0.05). conclusion:the present findings suggest that the crp gene c+1444t polymorphisms may be no related to acs susceptibility in the chinese han population of suwan region.

      Abstract:objective:to detect the serum epstain-barr virus(ebv)latent membrane protein 1(lmp1)antibody and to appraise its clinical significance in patients with nasal polyps. methods:there were 20 patients with nasal polyps who were managed by functional endoscopic sinus surgery(fess) in experimental group,20 cases of healthy volunteers in control group. the anti-lmp1 antibody in sera of two groups before operation and 15,30,90,180 days after operation were detected by elisa and western blot using glutathione-ebv-lmp1 extracellular domain fusion protein(gst-lmp1)as the antigen. results:the average levels of serum anti-ebv-lmp1 antibody displayed by absorbance of elisa in nasal polyps group and control group were significantly different(0.976±0.131 vs. 0.400±0.102,p < 0.01). the levels of anti-ebv-lmp1 antibody in the patients with nasal polyps before operation and 15,30,90 and 180 days after surgery were 0.976±0.131,0.921±0.138,0.899±0.168,0.576±0.144 and 0.521±0.117,respectively. the anti-ebv-lmp1 levels in sera of patients with nasal polyp decreased significantly in postoperative day 90 and 180 compared with those before operation(p < 0.01),but there was no significant difference among the levels before operation and in postoperative day 15 and 30. western blot also showed the same changes. conclusion:nasal polyp may be closely related to ebv infection,the serum anti-lmp1 antibody level in the patients with nasal polyp declined following effective treatment,and it may be used as a valid indicator of clinical therapeutic efficacy.

      Abstract:objective:to analyze the relationship of seropositive rate of hbsag with hepatitis b vaccination in a randomly selected general population in three counties of jiangsu province,and improve the comprehensive control against hepatitis b. methods:with cluster random sampling in different regions,questionnaire survey was conducted and blood samples were collected to detect hbsag by elisa. results:the standardized seropositive rates of hbsag were 4.61%,6.88% and 10.06% in zhangjiagang,danyang and taixing respectively,and the seropositive rate of hbsag among people over 18 years old who were born before programmed immunization started was significantly higher than that among people under 18 years old. the hepatatis b vaccine inoculation rates were 39.56%,19.66% and 41.04% in zhangjiagang,danyang and taixing respectively. in the population over 18 years,the seropositive rate of hbsag among people who were inoculated with hepatitis b vaccine was lower than that among people who hadn’t been inoculated. conclusion:the programmed hepatitis b immunization of newborns has got great achievement and hepatitis b vaccine inoculation among adults also obtained notable results. the immunization for the adults with hepatitis b vaccine should be improved to better control the infection of hbv.

      Abstract:objective:to analyze the epidemiological characteristics and the influence factors of the antiretroviral treatment patients(arts) in jiangsu province. methods:according to the base data of arts during 2005~2009,excel database was established and statistical analysis was performed using spss 17.0 software. results:there were 732 arts during 2005~2009 totally and the number of arts increased by year. ninety-five point five percent of the arts were from local province;the ratio of male to female was 2.1∶1;the average age was 41.9 years old;67.3% of the arts were married;patients were mainly detected and diagnosed in hospital;67.76% of the arts were infected with hiv through sexual transmission,while the proportion of men who have sex with men(msm)had the trend to increase;90% of the arts had the symptoms of fever,diarrhea and so on before treatment;the average number of cd4+ t cell was only 111.1 counts/μl;the median of the interval time between hiv confirmation and art started was 116 days. conclusion:it’s not early for the aids to start art in jiangsu province because aids patients were diagnosed late. earlier detection and diagnosis of hiv positive cases should be strengthened.

      Abstract:objective:to find out the problems in diagnosis and case report and to provide useful information for standardization of viral hepatitis diagnosis and report by analyzing the reported viral hepatitis cases in taixing. methods:the clinical and laboratory classification of the viral hepatitis patients reported in taixing during 2005~2009 were collected and analyzed with excel 2007 and spss 13.0 retrospectively. results:the reported incidence rate of viral hepatitis was 31.01/100 000~46.63/100 000 during 2005~2009,which showed an increased trend. the incidence of chronic hepatitis b in adult was obviously unstable;the incidence of hepatitis b without classification was 6.25/100 000~13.40/100 000,accounting for 31.0%~70.2% of total. compared to 2005,the incidence of hepatitis c increased(linear trend χ2=82.04,p < 0.01) while the hepatitis a decreased gradually(linear trend χ2=6.10,p < 0.05). the non-abc hepatitis accounted for 16.6%~23.0% of hepatitis. according to medical records and laboratory testing,the percentage of correct diagnosis of hepatitis a and e was 66.7%(12/18). the classification coincidence rate of hepatitis b was 51.35%(19/37). most hepatitis c patients[68.42%(13/19)] were anti-hcv positive without symptoms. more than half of non-abc hepatitis(53.97%) could be reclassified. conclusion:the reported incidence of chronic hepatitis b in taixing during 2005~2009 was unjustified,this point out that the reported incidence of hepatitis was inaccurate and the accuracy of categorization,classification of hepatitis should be improved.

      Abstract:objective:to explore the data management and quality control of the large-scale epidemiological survey study through an example. methods:the quality control was applied in the whole process including quality control system,staff training,field organization,laboratory,data entry and so on,and the data quality was assessed by data checking. results:after the original forms had been verified and the abnormal data had been revised,the error rates of people and the error rates of variable of 3 randomly selected projects were all about 10% and less than 0.5%,respectively. conclusion:there are many factors that can affect data quality in the medical research. it is essential for data quality to develop the reasonable programs of data management and quality control according to the actual situation,although considerable manpower and material resources are required.

      Abstract:objective: to assess the correlation between neoplastic microvessel density (mvd) and a series of pathological characteristics and study the prognostic value in postoperative patients of thymic epithelial tumors. methods:mvd in 66 cases of thymic epithelial tumor was evaluated by the expression of monoclonal cd34 antibody. survival analysis of 62 patients with 16~178 months follow-up was calculated using the kaplan-meier method and differences in survival were analyzed using the log rank test. results:there were no significant correlation between mvd and some pathological characteristics including the patients’ age (p > 0.05) and gender (p > 0.05). although bigger tumor (≥11.0 cm) developed higher mvd value,no significant difference was found compared with smaller tumor(<11.0 cm). mvd values were significantly different among subtypes of who histological classifications(p < 0.05)and clinical stages of masaoka’s staging system(p < 0.01). no positive correlation was found between mvd value and the status whether the patients had paraneoplastic myasthenial gravis or not(p > 0.05). the survival curves showed that the increased mvd,who histological classifications and masaoka’s staging system were valuable prognostic predictors of thymic epithelial tumors. conclusion:tumor angiogenic activity might play an important role in capsular and widely neighbouring invasion of thymic epithelial tumors. mvd together with who histological classifications and masaoka’s staging system seem to be more helpful for clinical prognostic estimation.

      Abstract:objective:to explore the diagnostic value of the fractional exhaled nitric oxide(feno) for asthma in children. methods:the patients with unkown-caused respiratory symptoms including wheezing, cough and breathlessness were enrolled from jun to sep in 2010. feno was measured by nitric oxide analyzer. bronchial challenge test and bronchodilator test were defined as golden standard for asthma diagnosis in children. the diagnostic value of feno was assessed and the optimal operating point of feno testing was determined by the means of the receiver operating characteristic(roc) curve. results:a total of 87 patients were enrolled, in which 52 cases were diagnosed as asthma by positive result in bronchial challenge test or bronchodilator test, and the other 35 cases were non-asthma. the level of feno in asthma group was higher than non-asthma group[(59.77±42.48)ppb vs(25.26±22.60)ppb, p < 0.05]. the level of feno was correlated with the counts of eosinophile granulocyte(r=0.546,p < 0.01) but not correlated with fev1%(r=-0.151,p > 0.05). area under roc curve was 0.818. the optimal diagnostic cut-off point was 34.5 ppb which was capable of differentiating asthma and non-asthma with sensitivity of 0.712, specificity of 0.686, positive predictive value of 0.755, negative predictive value of 0.605 and accuracy of 0.689. conclusion:determination fo feno may be helpful in the diagnosis of asthma in children.

      Abstract:objective:to investigate the risk factors for candidemia in pediatric intensive care unit(picu)patients. methods:a population-based case-control study of picu patients was conducted from 2001 to 2010. patients with candidemia infection were identified using laboratory records,and control patients without candidemia infection were selected from picu rosters. following multivariate analysis,logistic regression analysis was performed to determine risk factros and predicted probabilities of candidemia. results:one hundred and seven patients with candidemia infection were identified. factors independently associated with candidemia included presence of a central venous catheter [odds ratio (or)=25.68, 95% confidence interval (ci):8.72~97.47],malignancy(or=5.34,95%ci:1.80~15.22),use of vancomycin for ≥4 days(or=5.57,95%ci:1.74~17.49),and receipt of agents with activity against anaerobic organisms for ≥4 days(or=3.66,95%ci:1.31~7.53). predicted probability of having various combinations of the aforementioned factors ranged from 0.004 to 0.36. conclusion:future efforts should focused on validation of these risk factors identified in different picu population and development of interventions for prevention of candidemia infection.

      Abstract:objective: to investgate the influence of mothers’ hepatitisa b virus (hbv) dna quantity on immunologic response of infants inoculated with hepatitis b vaccine combined with hepatitis b immunoglobulin(hbig). methods:from feb 2008 to feb 2010,128 full-term infants born in nanjing second hospital affiliate to southeast university with negative hbv dna and hepatitis b surface antigen(hbsag),together with positive hbsag of their mothers were selected and divided into three groups(a,b and c) according to their mothers’ hbv dna quantity,the control group (d) was composed of 21 infants with their mothers had negative hbsag. all groups were given the same preventive inoculation,and the concentrations of hepatitis b surface antibody(hbsab)were examined at the same time. results:the average concentrations of hbsab in groups a,b,c and d were(240.19±79.34),(248.58±57.07),(240.87±61.97) and(266.03±70.93)mu/ml in one-month infants,(155.87±97.78),(155.72±141.11),(196.90±141.51) and(175.26±105.34)mu/ml in two-month infants,and(766.86±314.27),(754.30±363.56),(687.18±326.45) and (888.68±247.83)mu/ml in seven-month infants,respectively. there was no significant difference among these 4 groups at the same timepoint(f=0.666,p=0.574;f=0.884,p=0.451;f=0.857,p=0.466,respectively). conclusion:the prenatal hbv dna quantity had no effect on the levels of hbsab in 1,2 and 7 months infants. the antibody produced in the first month after combined immunization and decreased in the second month. the immunologic response of infants was not affected by mothers’ hbv dna quantity.

      Abstract:objective:sperm dna fragmentation has been assessed as a positive predictor of fertility potential in recent years. the aim of this study is using the acridine orange test(aot) to analyze the relationship between sperm dna fragmentation indexes(dfi) and semen conventional parameters of infertility men. methods:semen samples were collected from 376 infertility male patients in our andrology outpatient department.the samples were analyzed by improved neubauer haemocytometer chamber and computer-aided sperm analysis(casa) system to obtain concentration and motility parameters. all the samples were stained by shorr staining method and the smears were analyzed according to tygerberg strict criteria recommended by world health organization(who) to obtain morphology parameters. the dfi as marker of sperm dna damage determined using the aot. results:significant negative correlations were noted between dfi and sperm concentration(r=-0.150,p=0.003),motility(r=-0.114,p=0.028)and morphology(r=-0.155,p=0.003). no significant correlations were found between dfi and sperm progressive motility (r=-0.089,p=0.085). conclusion:sperm assessment for dna status using ao is reliable and shows good correlation with sperm count,motility and morphology. assessment of sperm dna fragmentation with aot measures a new parameter of male fertility.
