    • The relationship between B7-H4 expression and prognosis of stage T1 renal cell carcinoma

      Abstract:Objective: To investigate B7-H4 expression in stage T1 renal cell carcinoma and its role in evaluation of prognosis. Methods: B7-H4 expression was detected in kidney specimen from 102 cases,whose clinical features and renal cell carcinoma recurrence rate were also recorded. Results: There was no significant difference in the clinical and pathological features between the B7-H4 positive and negative groups;the disease free survival rate and metastasis rate were significantly higher in the B7-H4 positive group than those of the negative group. Tumor size and clinical stage have a significant correlation with the B7-H4 expression in renal cell carcinoma recurrence after surgery(P < 0.05). Conclusion: The B7-H4 expression is closely related to the development and immune evasion of renal clear cell carcinoma,and is valuable in predicting the prognosis of patients with renal cell carcinoma.

    • Evaluation of the cytotoxicity of Adper Prompt self-etch system of dental bonding agent on human gingival fibroblasts by flow cytometry

      Abstract:Objective:To evaluate the cytotoxicity of Adper Prompt self-etch system of bongding agents on human gingival fibroblasts by flow cytometry. Methods:Human gingival fibroblasts (HGFs) of passage 5 were cultured when cells covered 85% of the flask bottom. Different concentration (1.00 -滋l/L,0.50 -滋l/L,0.25 -滋l/L) of Adper Prompt were added- into medium respectively,and no Adper Prompt in medium as the negative control group. Cells were collected after 4 d and 7 d with 0.25% trypsin,fixed with 75% ethanol. The percentages of DNA content and apoptosis in different cycle phase were measured using flow cytometry,and the cell proliferation indexes(CPI) of different groups were calculated. Results:The peak of apoptosis rate was obvious in 1.00 -滋l/L group,and the peak of aneuploidy cell proliferation appeared in the cells treated with 1.00 -滋l/L Adper Prompt for 4 d. As compared with the control group,the DNA percentage of G2/M phase in 0.50 -滋l/L and 0.25 -滋l/L group increased significantly(P < 0.05) for both 4 d and 7 d,and while for 4 d,the DNA percentage of S phase in 1.00 -滋l/L group increased significantly (P < 0.05). The CPI of 7d is lower than that of 4d in the same concentration group. Conclusion:The cytotoxicity of Adper Prompt on human gingival fibroblasts in vitro is obvious,and it must be used very carefully in the clinic and kept away from gingives.

    • A study on relationship between life-meaning feeling and suicidal intension of college students

      Abstract:In this article 1 017 college students were investigated with PIL and SIOSS. Results showed that only 22.9% had definite life purpose and 6.0% had suicidal intension. Additionally, college students’ life-meaning feeling had significantly negative correlation with suicidal intension. Multiple regression analysis showed that college students’ life value, life attitude and life independence could predict their suicidal intension, of which life value gave the best prediction. Therefore, college students’ suicidal intension could be predicted by their life-meaning purpose, and suicide could be prevented in advance.

      Abstract:Objective:To study work family conflict situation of clinical doctors. Methods:Work family conflict scale and perceived social support scale were used. Results:Work interference with family (WIF) of clinical doctors was higher than family interference with work (FIW). Both male’s WIF and FIW were higher than female. FIW of unmarried was higher than married. WIF of clinical doctors with children was higher than those without children,which had significant negative correlation with the children’s age. WIF was significant positive correlation with the average working time of one week. FIW was significant negative correlation with the perceived social support. Conclusion:Work family conflict of clinical doctors has professional character. Doctor’s gender,family situation,working time and social support has different effect on the WIF and FIW.

      Abstract:Animal has no moral,but this does not negate that the virtue is form the basis of human life. Baby who cannot use language,cannot give the conceptually reasoning cannot grow up into mature practitioner without person’s biological basis. Mature practitioner cannot keep the identity without the body of animal personality. Based on recognition of the biological basis of virtue,the relationship between virtue and reason should be clarified probably,and the dilemma of rationalism moral philosophy should be solved.

      Abstract:Objective:To understand the trend of residents’ health literacy in Jiangsu province through the continuity surveillance over the years. Methods:The health literacy levels in different factor,such as age,gender were analyzed. And all the data was standard analyzed using statistical software. Results:Overall,the health literacy level was improved steady,and the similar trend was happened in different gender,education and age groups,but the changing trends were not exactly same in different group. The improving range of students’ correct recognition was higher than the residents in Jiangsu province. Conclusion:In recent years,the trend of residents’ health literacy was advanced. However,the intervention strategy should be enacted according to the different characteristic of the residents,and the institutionalization of regular intervention and evaluation work of the basic knowledge and skills of residents’ health literacy should be carried out.

      Abstract:Objective:To compare the differences between medical students and non-medical students in the aspects of rates of sexual act incidence and rates of sexual act protection during the last 10 years of China,and discuss the influence of medical education for current sexual acts in order to provide foundations for developing sexual education. Methods:Domestic published documents about current sexual act situation in both medical and non-medical universities were collected from January,2002 to January,2012. The documents were evaluated according to the specific standard,and the required documents were analyzed by Meta. General statistic description was applied if data heterogeneity was too obvious to apply Meta. Results:The number of qualified documents was 15 in total. Due to the apparent heterogeneity,general statistic description was applied. In medical students the rate of sexual incidence was lower and the rate of sexual protection was higher than non-medical students apparently,and the both rate were rising year by year recently. Conclusion:The problem of sexual acts in the universities is not optimistic,and the response is necessary.

      Abstract:Objective:To observe the intervention effect of systematic health education in patients with gestational diabetes mellitus,and to find the scientific,convenient,economic and effective diet treatment and health promotion method. Methods:120 patients with gestational diabetes mellitus were randomly arranged into two groups,60 women in case group who were received systematic health education and others in control group who were received traditional health education. Related knowledge,attitudes and eating habits of the two groups were observed. Results:The knowledge survey score and the attitude score increased after the intervention in systematic health education group were significantly higher than those in traditional group. The eating habits of the systematic education group were also superior to the traditional education group. Conclusion:The systematic health education in the pregnant women with gestational diabetes mellitus is helpful in control of blood sugar and obstetrical disease.

    • A study on chronic diseases management mode based on electronic health records

      Abstract:Objective:To study the application of the electronic health information record in chronic disease management mode. Methods:Management conditions of three kinds of chronic disease that was hypertension,diabetes mellitus and malignant tumor in Gulou district of Nanjing city from October 2008 to October 2011 were studied,which were divided into three periods as Period A from October 2008 to September 2009,Period B from October 2009 to September 2010 and Period C from October 2010 to September 2011. The area number,filing management number,filing management rate,standardized management number and specification management rate at the three periods were investigated. Results:By the end of September 2011,598 800 regulative electronic health records had been completed with filing rate 72.43%,among which 333 400 were standard managed with standardized management rate 55.68%. Period C and Period B were significantly higher than Period A(P < 0.05),Period C in filing management number and filing menagement rate were statistically higher than Period B(P < 0.05),and the difference between Period B and Period C in standardized management number and specification management rate were statistically meaningless(P > 0.05). Conclusion:Chronic disease information management mode can effectively integrate medical resources and realize the scientific management of chronic diseases’ prevention and control. It can realize the dynamic observation of individuals and groups of related health problems,regularly evaluate,timely find problems and respond to intervene and control,and effectively improve the chronic disease management.

    • A study on influencing factors and coping measures of rising maternal cesarean section rate in Nanjing Maternal and Child Health Care Hospital

      Abstract:Objective:To analyse the cause of rising maternal cesarean section rate and its influencing factors in Nanjing city. Methods:Cluster sampling was used to collect 4 793 parturient women choosing cesarean section in Nanjing Maternity and Child Care Hospital from January to December,2011 and questionnaire was used to the maternal and their family members. Results:In the indications of cesarean section,social psychology factors accounted for 69.27%,economic factors (family income and health insurance,etc) accounted for 17.24%,medical indications factors accounted for 13.49%. Maternal choosing cesarean section with highly educated or high income or high aged was significantly more than the normal cesarean delivery mothers (P < 0.01). Many factors Logistic regression showed that the related risk factors of choosing cesarean section are:age,education level,family income,medical insurance,marital status,maternal and family psychological intervention. Conclusion:The following measures should be helpful to reduce cesarean section rate:paying attention to delivery knowledge education and perinatal care to parturient women,strengthening the screening of high-risk groups and standardized management,widely conducting painless delivery and hospital services,exactly grasping the cesarean section indications and opportunity.

      Abstract:Objective:To investigate the clinical pathway for cesarean section in the application effect of single disease. Methods:The object of study was parturient women choosing cesarean section that satisfied the conditions for clinical pathway of cesarean section from January to December,2010, which divided into clinical path group and control group. The preoperative length of stay,the average hospitalization days, cost of hospitalization, complications, and patient satisfaction and disputes in the two groups were analyzed. Results:In clinical pathway group, preoperative length of stay, total days of hospital stay were reduced, total cost of hospitalization was decreased slightly, and patients’ satisfaction was improved. Conclusion:The implementation of clinical pathway is helpful to guarantee the medical quality and control the medical expense. It is an effective,feasible method of diagnosis and treatment.

Methods:According to the characteristics of the students of different phase,the teaching department of stomatological college cooperated with several enterprises to develop kinds of teaching methods,such as universal education of professional knowledge,digitalized experimental teaching,operating skill race,knowledge lecture of leading edge,and crosswise scientific research items by corporate sponsorship and technical support. These methods offered kinds of studying pathway for the students. The assessments of the efficiency of cooperation of school and enterprise using in the stomatological teaching had also been done after these teaching activities. Results:The cooperation of school and enterprise using in teaching realized the modernization,diversification and interests of teaching methods. So that it was suitable for stomatological education because of its professional and could be used as assistant teaching form of school education. This cooperation improved the teaching quality of stomatological education and achieved better teaching efficiency. Conclusion:The cooperation of school and enterprise has wide application prospect to use in the stomatological teaching. And it is the effective way to improve the teaching quality.

      Abstract:Objective:Problem-based Learning(PBL)has been suggested as a key educational method of knowledge acquisition to improve medical education. We used PBL and conventional teaching method in obstetrics and gynecology clinical teaching to clarify which curricula fit medical education better. Methods:Sixty-one interns in obstetrics and gynecology department of the First Affiliated Hospital of Nanjing Medical University were randomly divided into two groups:experimental group using PBL teaching method(n = 31)and control group using conventional teaching method(n = 30). The experimental group used PBL rating scale and the evaluation form of teaching quality,and the control group used the evaluation form of teaching quality. Results:Tow parts of “build clinical thinking” and “promote a sense of teamwork” got the highest score in PBL rating scale and “build professional knowledge” and “help to learn other courses” had the lowest score. Experimental group had statistically higher score in three aspects than the control group(P < 0.05),which were teaching content,teaching methods and teaching effectiveness. Conclusion:Comparing to conventional teaching methods,PBL is conducive to develop students’ teamwork and problem-solving skills,improve medical students’ clinical thinking ability and core competitiveness. Researches deserve closer attention in the future PBL teaching method.

    • Appreciation of cultural background diversity is a key component of successful medical parasitology education of international students

      Abstract:Increasing cultural and scientific exchange between Chinese educational institutions and foreign students brings new challenges to Chinese educators. Most Chinese educators grow up in a monolithic culture with no dietary restriction imposed by religions. Consequently,many medical parasitology instructors do not have enough appreciation of the impact of religions on the transmission of parasitic diseases. In addition,different geographic distributions make some parasites and their vectors less relevant teaching materials for some international students. The medical parasitology course for international students should not be a mere replica of the course for Chinese students. To inspire the motivation of the students and improve the quality and long-term benefit of medical parasitology course,teachers should take into consideration the diversity in cultural background and epidemiological difference,make necessary adjustments in the contents of the course,and respect the cultural customs of the students.

      Abstract:This article chose funded papers as high-level papers,analyzed the citations of these papers,and investigated the differences in citation between funded papers and non-funded papers. The results suggested that there was no difference in citation frequency of single paper between funded papers and non-funded papers,while there was significant difference in citation rate between funded papers and non-funded papers. It could be got that citation rate of papers may be used to reflect the levels of journals.

    • A single administration of fluoxetine induces lipid accumulation in mouse liver

      Abstract:Objective:To observe the effects of fluoxetine on the lipid metabolism in mouse liver and investigate the mechanisms. Methods:All mice received an intraperitoneal injection of saline (control),fluoxetine (10 mg/kg or 25 mg/kg) or clozapine (25 mg/kg). The mice were sacrificed at 6h or 24h after injection,and the livers were collected. Liver neutral lipid was determined by oil red O staining. Liver triglyceride and total cholesterol were measured by test kits. Liver sterol regulatory element-binding protein 1c (SREBP1c),acetyl-CoA carboxylase 1 (ACC1) and fatty acid synthase (FAS) as well as carboxylesterase 1 (CES1) and carboxylesterase 3 (CES3) protein levels were determined by Western Blot. Results:Acute fluoxetine treatment induced hepatic lipid accumulation in mouse liver. Furthermore,the expression of ACC1 and FAS was evidently elevated while the expression of triacylglycerol hydrolases,CES1 and CES3 was suppressed by fluoxetine. Meanwhile,the expression of SREBP1c,a lipid-modulating transcription factor,was also stimulated by fluoxetine. Conclusion:These data suggest that fluoxetine may upregulate lipogenesis via SREBP1c,as well as downregulate lipolysis in mouse liver,resulting in hepatic lipid accumulation.

    • Generation and identification of human monoclonal antibodies to the rabies virus

      Abstract:Objective:To generate human monoclonal antibodies (mAbs) against the rabies virus (RV) by using the mice carrying human immunoglobulin transloci for immunization and further to identify their characters. Methods:The human IgM transgenic mice were immunized with the inactivated RV(CTN strain) as immunogen. The human anti-RV mAbs were screened and produced by using the classical hybridoma technique in a combinatory high throughput screening (HTS) strategy. The fully human mAbs were identified via the double antibody sandwich enzyme-linked immunosorbent assay(ELISA),and the specificity of mAbs was determined by the indirect ELISA,Dot Blot and Western Blot. In addition,the affinity of mAbs was detected by BiaCore X-100 system. Results:Two anti-RV hybridoma cell lines 9E3 and 9F2,which can produce human IgM mAbs and recognize the CTN strain of RV specifically in the indirect ELISA and the Dot Blot assays,have been established. The results of Western Blot demonstrated that the two prepared mAbs specifically recognize glycoprotein(G protein) of the rabies virus. The equilibrium dissociation constants (KD) of 9E3 and 9F2 were 2.62 × 10-10 mol/L and 4.06 × 10-11 mol/L,respectively. Conclusion:Two anti-RV fully human IgM mAbs with specificity and high affinity were generated. The first-phase preparations of mAbs could be the bases for the screening immunoprophylaxis neutralized monoclonal antibody and immunotherapy on the rabies.

    • CD4+CD25+Foxp3+:a marker of Treg related to SLE pathogenesis

      Abstract:Objective:To investigate markers of regulatory T cells in the pathogenesis of systemic lupus erythematosus (SLE).Methods:Peripheral blood mononuclear cells (PBMCs) were isolated from SLE patients and healthy donors;Flow cytometry (FCS) was used to detect the expressions of CD25 and Foxp3 in CD4 and CD8 T subsets and analyze the changes in the ratios of Foxp3+/CD4+,CD25+Foxp3+/CD4+,Foxp3+/CD4+CD25+,Foxp3+/CD8+,CD25+Foxp3+/CD8+,Foxp3+/CD8+CD25+,and their relationships with SLEDAI,anti-dsDNA antibodies,complement C3/C4,serum IgG,kidney damage and leucopenia. Results:① Compared with healthy donors,the percentage of CD25+ cells in CD4+T cells did not change significantly in SLE patients,but the ratios of Foxp3+/CD4+,CD25+Foxp3+/CD4+ and Foxp3+/CD4+CD25+ significantly reduced in patients with mid- and high-active SLE,and the ratios were also negatively correlated to SLEDAI. Particularly,the ratio of CD25+Foxp3+/CD4+ in SLE patients was related with anti-dsDNA antibody,decrease of complement C3/C4 and increase of IgG;and the reduce of CD25+Foxp3+/CD4+ ratio was remarkable in SLE patients with renal damage or Leucopenia and patients in initial disease stage. ② Foxp3+ cells ratio of CD8+ cell population was similar between SLE patients and healthy controls,except that the ratio of Foxp3+/CD8+CD25+ decresed in mid- and high-active SLE patients,but had no relationship with disease activity. Conclusion:CD4+CD25+Foxp3+ is an important marker for evaluation of regulatory T cells,which reflects the deficient quantity of Treg,and might reflects the deficient function of Treg by the relationship between the cell percentage and SLE disease activity,immune disorder and tissue lesion. The study confirmed that CD4+CD25+Foxp3+ as the marker of Treg is significant for evaluation of immune system functional status of SLE patients in clinical.

Methods:Forty healthy male Sprague Dawley rats were randomly and equally divided into 4 groups,ie,saline-injected group,AD group,clenbuterol group and ICI118,551 group. The AD rat model was established by amyloid-β1-40(Aβ1-40) injection into hippocampus. Then the clenbuterol group and ICI118,551 group were peritoneal injected by clenbuterol of 0.5 mg/kg and ICI118,551 of 1 mg/kg respectively for 20 days after they were injected Aβ1-40 injection. The learning and memory ability were determined by Y-maze,and the activity of AChE and ChAT in hippocampus was tested by chemistry method. Nissle staining was used to verify neuron apoptosis. Results:Compared with the AD rats,the learning and memory ability of rats injected with clenbuterol were significantly decreased(P < 0.01),correspondingly,the activity of AChE and ChAT decreased. While in the rats of the ICI118,551 group,both their learning and memory ability and the activity of AChE and ChAT were increased compared with those in the AD rats(P < 0.05). Conclusion:The β2-AR agonist clenbuterol might aggravate the injury induced by Aβ1-40,while the β2-AR selective antagonist ICI 118,551 can improve AD pathological process. These results suggested that the level of acetylcholine in brain tissue may be involved in the possible mechanisms of AD.

      Abstract:Objective:To investigate the effects of early use of recombinant human tumor necrosis factor receptor-Fc fusion protein(rhTNFR-Fc) on pulmonary arterial hypertension (PAH) induced by monocrotaline (MCT) in rats,and to explore its mechanism. Methods:Total 24 adult male rats were randomly divided into control group (C group),rhTNFR-Fc group (R group),MCT group(M group) and rhTNFR-Fc combined monocrotaline group(M+R group). PH was induced by intraperitoneal administration of MCT at a concentration of 60 mg/kg for the rats of M group and M+R group on day 1. rhTNFR-Fc(0.4 mg/kg) or saline(1 ml) was injected subcutaneously every other day for day 2 to day 21. After 3 weeks,the mean pulmonary arterial pressure(mPAP),tha ratio of right ventricle to left ventricle and septum[RV/(LV+S)] were measured,microscope was used to detect the morphologic changes of small pulmonary arteries as calculating the percentage of vascular wall thickness to vascular external diameter(WT%). The expression of Tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6) in lung homogenates were measured by ELISA method,and the protein level of NF-κB in lung tissue was measured by Western blot. Results:① The levels of mPAP, RV/(LV+S) and WT% were higher in M group than those in C group,and early-use of rhTNFR-Fc can inhibit the increase of mPAP and the rise of WT% in MCT-induced rats(P < 0.05). ② rhTNFR-Fc can inhibit the increase of IL-6 and TNF-α in lung of the rats induced by MCT(P < 0.05). ③ rhTNFR-Fc can inhibit the increased expression of NF-κB in pulmonary tissue of MCT-induced rats(P < 0.05). Conclusion: These results suggest that rhTNFR-Fc can ameliorate the progression of PAH induced by MCT in rats,which was probably through inhibiting NF-κB signal pathway and decreasing inflammatory cytokines expression of IL-6 and TNF-α.

      Abstract:Objective:To investigate the effects of small interference RNA (siRNA) against ribosomal protein L22(RPL22) on ET-1 induced proliferation of human pulmonary arterial smooth muscle cells (HPASMC). Methods:The experimental models of primary cultured HPASMC induced by ET-1 were established in vitro. HPASMC were transfected with siRNA-RPL22,incubated in smooth muscle cell culture medium (SMCM). The mRNA and protein expresson levels of RPL22 in HPASMC were measured by real-time PCR and Western blot,respectively. The HPASMC proliferation was evaluated by PCNA expression and flow cytometry analysis. Results:Compared with control groups,RPL22 expression in HPASMC was significantly inhibited after siRNA-RPL22 transfection(P < 0.05),and HPASMC proliferation was significantly inhibited after siRNA-RPL22 transfection. Conclusion:The data suggest that interruption of RPL22 expression can inhibit HPASMC proliferation. It’s a novel finding needs to be further explored.

      Abstract:Objective:To investigate the inhibitory effects of budesonide on the expression of OX40(CD134),airway inflammation,and airway hyperresponsiveness in a murine model of asthma. Methods:Total 18 BALB/c mice were randomly divided into 3 groups. They were control group,ovalbumin (OVA) group and budesonide group. Mice were sensitized and challenged by OVA. Airway responsiveness to acetylcholine chloride was measured. Hematoxylin & eosin staining was used to assess the inflammatory cell infiltrates. Levels of IL-4 and IL-13 in bronchoalveolar lavage fluid(BALF),and total IgE and OVA-specific IgE(OVA-sIgE) in serum were detected by ELISA. The protein expression of OX40 was determined by Western blot analysis. Results:The airway resistance in the OVA group was obviously increased in a dose-dependent manner by administration of ACh,whereas only a slight increase could be detected in the control group. There were no significant differences in baseline airway resistance among three groups(P > 0.05). Treatment with budesonide led to a sharp decrease in airway resistance compared with the OVA group (P < 0.05). The number of eosinophils and total inflammatory cells in BALF in the OVA group increased significantly compared with the control group (P < 0.05),which was significantly decreased by treatment with budesonide(P < 0.05). The levels of the IL-4 and IL-13 in BALF,and the levels of total serum IgE and OVA-sIgE were significantly increased in OVA-sensitized/challenged mice compared with the control group (P < 0.05). Administration of budesonide reduced the levels of those Th2 cytokines in BALF,and the levels of total serum IgE and OVA-sIgE,when compared with the OVA group(P < 0.05). Treatment with budesonide increased the ratio of OX40,compared with that in the OVA group (P < 0.05). Conclusion:Budesonide could inhibit the airway inflammation and hyperresponsivenes by down-regulating the expression of OX40.

      Abstract:Objective:To observe the curative effect of GuQing granules on the adjuvant arthritis rats,and detect whether GuQing granules could regulate the expression of acid-sensitive ion channel 3(ASIC3) in articular cartilage. Methods:The rats were randomly divided into five groups:blank control group,adjuvant arthritis model group,positive control group(aspirin 50 mg/kg),GuQing granules group of thearapeutic dose(8.4 g crude drug/kg),GuQing granules group of high dose(16.8 g crude drug/kg). Except the rats in blank control group,all the rats were injected intradermally with complete Freund’s adjuvant(CFA) at left toes to induce adjuvant arthritis model. Ten days after injection,all rats of each group were administration with corresponding drugs for 10 days. The weight,volume of right foot and arthritis scores of rats were recorded.,the pathological changes of rat ankle joint were observed by optical microscope,and the mRNA and protein expression of ASIC3 in rat knee cartilage were analyzed by RT-PCR and Western blot. Results:GuQing granules significantly inhibited the paw swelling of rats ankle after treatment,the mRNA and protein expression of ASIC3 in knee cartilage decreased after GuQing granules treatment. There was no significant difference between the positive control aspirin group and GuQing granules treatment group. Conclusion:GuQing granules can reduce the inflammation of adjuvant arthritis rats,protect articular cartilage and down-regulate the expression of ASIC3.

      Abstract:Objective:To evaluate the effect of the combined use of glutamine and growth hormone on the intestinal permeability and the bacterial translocation of rats with obstructive jaundice(OJ). Methods:The study was designed as a prospective,randomized and controlled animal experiment. The common bile duct was ligated to obtain obstructive jaundice model. Sixty Sprague-Dawley rats were randomly assigned into 6 groups with 10 rats in each group:Group Ⅰ,Control Group with normal saline(1.5 ml/d) intragastric administration;Group Ⅱ,Sham-operated Group with normal saline(1.5 ml/d) intragastric administration;Group Ⅲ,OJ Group with normal saline(1.5 ml/d) intragastric administration;Group Ⅳ,OJ Group with Glutamine(Gln) of 0.2 g/(kg·d) intragastric administration;Group Ⅴ,OJ Group with Growth hormone(GH) of 2 U/(kg·d) subcutaneous injection;Group Ⅵ,OJ Group with Gln of 0.2 g/(kg·d) intragastric administration and GH of 2 U/(kg·d) subcutaneous injection. On postoperative 9th day,each rat received dual saccharides by intragastric administration. On day 10 postoperatively,24 h urine sample of each rat was collected for L/M test,while liver,spleen,mesenteric lymph node(MLN) and the inferior caval blood samples of each rat were obtained for bacterial culture and serum biochemical test,respectively. Results:Except that there was no significant change in the L/M value between groupⅠand Ⅱ(P > 0.05),and between group Ⅳ and group Ⅴ (P > 0.05),there were significant differences between any other two groups in six groups(P < 0.05). The bacterial translocation ratio in group Ⅵ was significantly lower than that in group Ⅳ or Ⅴ(P < 0.003 3 respectively) whereas there was no significant difference in bacterial translocation ratio between group IV and V (P > 0.003 3). The bacterial translocation rate in group Ⅳ was significantly lower than in group Ⅲ(P < 0.003 3);The bacterial translocation ratio in group Ⅵ was significantly higher than that in group Ⅰ or Ⅱ(P < 0.003 3 respectively),while it was similar between groupⅠand Ⅱ(P > 0.0033). Conclusion:The combined administration of Gln and GH is more effective than using each agent alone on reducing intestinal hyperpermeability and bacterial translocation ratio in obstructive jaundice.

      Abstract:Objective:To investigate the expression and gender disparity of protein tyrosine phosphatase receptor-type O (PTPRO) in human hepatocellular carcinoma(HCC),the correlation of PTPRO with carcinoma recurrence after hepatectomy,and whether PTPRO can be served as a desirable prognostic index. Methods:PTPRO expression of HCC tissue and adjacent tissue in 59 male and 22 female patients were determined by real-time PCR and immunohistochemistry(IHC),as well as estrogen receptors(ERs). Gender disparity of PTPRO and ERs and the correlation between PTPRO and ERs were analyzed by t test,correlation analysis and F test;The correlation between PTPRO level and post-hepatectomy recurrence rate of HCC patients were analyzed by Kaplan-Meier curve and log-rank test. Results:Expression of PTPRO and ER-琢 in HCC tissues were significantly lower than adjacent cirrhotic tissues(P < 0.000 1);Expressions of PTPRO and ER-琢 of adjacent tissue was lower in male than in female (P < 0.05);Expression of PTPRO was negatively correlated to recurrence of HCC patients(P < 0.000 1). Conclusion:PTPRO is a potential tumor suppressor with gender disparity,and can serve as a candidate for predicting recurrence after surgical resection of HCC.

      Abstract:Objective:To elucidate the role of antisense oligodeoxynucleotide of p55PIK inhibiting the proliferation of thyroid carcinoma cells. Methods:The lentivirus package system carrying the antisense RNA of p55PIK was constructed. The FTC236 cell was infected by the virus. Western blot and Realtime PCR were used to confirm the protein content and mRNA level of p55PIK. Then MTT assay and BrdU/PI corporation assay were used to analyze the proliferation of stable cells. Results:Two stable cell lines,one expressing p55PIK antisense RNA and the other expressing scramble antisense RNA,were constructed,and the p55PIK antisense RNA could knockdown the target protein’s expression(P < 0.01). In MTT assay,the absorbance ratio of the cells expressing p55PIK antisense RNA is (59 ± 19)% of that in the cells expressing scramble antisense. The BrdU/PI corporation assay showed that the DNA synthetize ratio of cells expressing p55PIK antisense RNA decreased by 11.6% compared with that of cells expressing scramble antisense. Conclusion:The p55PIK antisense can inhibit proliferation of the thyroid carcinoma cells by knockdown the p55PIK protein expression.

      Abstract:Objective:To explore the expression of microRNA-335(miR-335) in non-small cell lung cancer(NSCLC) and its effect on migration,invasion and proliferation of NSCLC cell. Methods:Real-time quantitative PCR was used to detecte and compare the expression of microRNA-335 in 12 pairs of non-small cell lung cancer,adjacent normal tissue,NSCLC cell line SPCA-1 and normal epithelium cell line 16HBE. The expression of miR-335 in SPCA-1 cell was down-regulated by anti-miR-335 transient transfection with Lipofectamine 2000,and the effect was identified by real-time quantitative PCR. Cell migration and invasion was measured in vitro by wound healing assay and transwell assay. The influence of miR-335 on SPCA-1 cell proliferation was detected by MTT rassay and colony formation assay. Results:Compared with adjacent normal tissues,miR-335 in non-small cell lung cancer was significantly up-regulated(P < 0.05). Smilar to that,the expression of miR-335 was significantly increased in SPCA-1 cell compared to 16HBE cell(P < 0.05). The miR-335 expression of SPCA-1 cell was significantly down-regulated 24 hours after anti-miR-335 transfection(P < 0.01). In SPCA-1 cell,decreased miR-335 significantly inhibited cell migration and invasion,with the inhibiting rates of (42.8 ± 2.7)%(P < 0.01) and (73.25 ± 4.4)% (P < 0.01),respectively. But,no effect on SPCA-1 cell proliferation was observed when the expession of miR-335 was inhibited. Conclusion:The expression of miR-335 is up-regulated in NSCLC. Down-regulating expression of miR-335 significantly inhibites migration and invasion of SPCA-1 cell while has no effct on cell proliferation.

      Abstract:Objective:To evaluate the impact of the single nucleotide polymorphism (SNP) in Janus kinase 2(JAK2) rs1887427 on gastric cancer risk. Methods:In this hospital-based,case-control study,the genotypes were identified by polymerase chain reaction-restriction fragment length polymorphism protocols. Results:The frequencies of the wild and variant genotypes in cases were significantly different from those of controls (P = 0.027). Compared with individuals with the wild genotype AA,subjects with the variant genotypes (AG + GG) had a significantly higher risk of gastric cancer (P = 0.023,adjusted OR = 1.71,95% CI = 1.08-2.71). Moreover,in stratified analysis,the elevated gastric cancer risk was especially evident in younger individuals,males and nonsmokers (P < 0.05 for all). Conclusion:The JAK2 polymorphism is associated with an increased risk of gastric cancer in Chinese population.

      Abstract:Objective:To analyze the structure characteristics of the breast cancer susceptibility protein 1(BRCA1) gene sequence by bioinformatics methods and predict the relationship between mutations of functional coding regions in BRCA1 and the pathogenic genetic effects. Methods:The molecular phylogeny,conservation,selection pressure,domains,three-dimensional structure and missense of BRCA1 were analyzed by online Bioinformatics analysis tools such as Maximum Likelihood,ClustalW,SMART,SELECTON and so on. Results:Total 195 fixed-residue sites(10.5%)and 393 conservative sites(21.1%),of which distribution is non-randomness,were obtained. A big difference in the BRCT domain was founded between human and other species,which indicated the role of BRCT domain in BRCA1 gene of various species is difference. The high pathogenic mutation rate in conservative sites was confirmed by correlation analysis. Conclusion:Taking full advantage of relevant resources,the BRCA1 sequence structure analysis based on bioinformatics methods can deepen our understanding of the correlation of BRCA1 gene mutation and tumor.

      Abstract:Objective:To investigate the effects of mitochondrial damage and drug resistance of 5-FU-resistant human pancreatic cancer cell line Patu8988/5-FU cultured by cobalt chloride(CoCl2). Methods:Patu8988/5-FU were cultured in medium containing different concentrations of CoCl2(0,6.25 μmol/L,12.50 μmol/L,25.00 μmol/L,50.00 μmol/L,100.00 μmol/L,200.00 μmol/L,400.00 μmol/L). MTT assay was used to detect the cell proliferation activity and drug resistance of Patu8988/5-FU. JC-1 fluorescence staining with flow cytometry was used to analyze the mitochondria membrane potential. The mRNA expressions of HIF-1α and MDR1 were detected by RT-PCR technique. P-gp pump function of Patu8988/5-FU cells’ membrane was tested by Rh123. Results:With CoCl2 concentration rising or 100 μmol/L CoCl2 treatment time extension,the proliferation activity of Patu8988/5-FU cells was degressive,the depolarization of mitochondrial membrane potential was increased. Drug resistance index of Patu8988/5-FU to 5-FU in normoxia and hypoxia reaches to 28.11±3.19 and 52.08 ± 3.53,respectively(P < 0.01). The mRNA expressions of HIF-1α and MDR1 in Patu8988/5-FU cells is higher in hypoxia than in normoxia (P < 0.01). The accumulation amount of Rh123 in cytoplasm of Patu8988/5-FU cells was significantly lower in hypoxia than in normoxia(P < 0.01). Conclusion:CoCl2 induced chemistry hypoxia of Patu8988/5-FU cells in a time-and-dose dependent manner. CoCl2 mimicing hypoxia dramatically increased resistance of Patu8988/5-FU cells to 5-FU,and maybe through upregulating the expression of HIF-1α and MDR1.

      Abstract:Objective:To investigate the expression profile of Mcl-1 in cervical cancer and its relationship with the clinical pathological characteristic,and to assess its clinical significance. Methods:Mcl-1mRNA expression both in cervical cancer and in corresponding normal tissue was detected by reverse transcription fluorescent quantitation polymerase chain reaction method. Mcl-1 protein expression was detected by immunohistochemistry method. The correlation of Mcl-1 expression and clinicopathological features of cervical cancer was analyzed. Results:Mcl-1mRNA was higher in cervical cancer tissue when compared with corresponding normal tissue. Mcl-1 was overexpressed in cervical cancer tissue(positive rate of 53.3%) when compared with corresponding normal tissue (positive rate of 11.1%). High expression of Mcl-1 was significantly correlated with histological grade,tumor size and lymph node involvement(P < 0.05). Conclusion:Our results suggest that Mcl-1 may play an important role in cervical cancer and have potential as biomarker and therapeutic target.

    • The relationship between Modic changes and the degree of disc degeneration in degenerative spondylolisthesis

      Abstract:Objective:To study the relationship between Modic changes and the degree of disc degeneration in patients with degenerative spondylolisthesis before surgery. Methods:From January 2009 to December 2011,44 cases of patients who had the degenerative spondylolisthesis were treated by operation. Preoperative MRI was performed to classify the Modic changes type. Pfirrmann grading assessment and modified Pfirrmann grading assessment were also perfomed. Then,the relationship between Modic changes and the degree of disc degeneration were analyzed in these patients with degenerative spondylolisthesis. Results:Among 44 cases,12 cases had no Modic changes,Pfirrmann grades were 3.92 ± 0.29,modified Pfirrmann grades were of 5.92 ± 0.90;16 cases were ModicⅠ type,Pfirrmann grades were 4.31 ± 0.60,modified Pfirrmann grades were 6.88 ± 1.54;15 cases were Modic type Ⅱ,Pfirrmann grades were 4.27 ± 0.46,modified Pfirrmann grades were 6.87 ± 0.83;Only 1 case of Modic type Ⅲ was found. Statistic analysis showed that modified Pfirrmann grades were significantly different between ModicⅠtype group and no change group(P < 0.01);Differences were also exsit between Modic Ⅱ type group and no change group(P < 0.01);But,there was no significant difference between ModicⅠtype group and Modic Ⅱ type group(P > 0.05). Conclusion:In degenerative spondylolisthesis,there is a clear connection between the Modic changes and the degree of disc degeneration according to the Pfirrmann and modified Pfirrmann grading system assesment.

      Abstract:Objective:To improve the capability of ultrasound-guided fine-needle aspiration biopsy (US-FNAB) in diagnosing thyroid diseases. Methods:Total 1 016 US-FNAB cytopathological cases were analyzed retrospectively from May 2011 to Jan 2012 in Department of Endocrinology,the First Affiliated Hospital of NJMU. Meantime,44 histological pathological reports from these patients were analyzed. Results:Among 1 016 US-FNAB cytopathological reports,72 cases (7.09%) were labelled as malignant or suspicious for malignancy while confirmative cytopathological diagnosis for capsula glandulae thyroideae,chronic lymphocytic thyroiditis (CLT) and subacute thyroiditis were seen in 132 (12.99%),165 (16.24%),and 56 (5.51%) cases. By combining cytological result of US-FNAB and histological pathological results of sugery,the sensitivity,specificity,positive predictive value,negative predictive value,false positive rate and false negative rate were calculated as following 65.22%,76.19%,75.00%,66.67%,25.00% and 33.33%. Total positive cell rate was 85.24%. Conclusion:US-FNAB showed great importance in differentiation between benign and malignant thyroid lesions and in diagnosing capsula glandulae thyroideae,chronic lymphocytic thyroiditis and subacute thyroiditis.

      Abstract:Objective:To test the hypothesis that ventilator-associated pneumonia(VAP) after cardiac surgery with cardiopulmonary bypass(CPB) requires TLR4-dependent signaling. Methods:Twenty patients were categorized into two groups based on development of VAP following CPB,group with VAP (group A) and group VAP without (group B). The blood samples of left atrium were collected in both groups at the time points of preoperation(T1),20 min(T2),40 min(T3) and end(T4) of CPB. Expression of TLR4 on granular leukocyte was detected with flow cytometry. The cinical data of all cases in two groups were compared. Results:VAP occurred in 8 patients. CPB time in group A was statistically longer than in group B. The expression of TLR4 on granular leukocyte in group A at the time points of T3 and T4 were significantly higher than those in group B. Conclusion:Development of VAP after cardiac surgery with CPB was promoted by long CPB time via TLR4-dependent signaling activated.

      Abstract:Objective:To observe the correlation of single-nucleotide polymorphisms(SNPs) of chitinase 3-like protein 1(CHI3L1)-329 G/A and coronary artery disease(CAD) in Jiangsu province. Methods:Total 391 patients were enrolled,including 189 patients with CAD and 202 controls. The SNP at CHI3L1 -329 G/A was genotyped by ligase detection reaction. Results:Analysis of genotype frequencies did not reveal any significant difference between CAD patients and controls. There were significant differences in the frequencies of A allele and A allele carriers among SAP,UAP,and MI groups(P < 0.05 respectively). No associations existed between the SNP and the severity of coronary artery stenosis(All P > 0.05). Conclusion:These results did not support an involvement of SNP rs10399931 of CHI3L1 -329 G/A in predisposition to CAD in Jiangsu area.

    • Clinical effects of oxiracetam for treatment of vascular cognitive impairment in varying degree

      Abstract:Objective:To investigate the effects of oxiracetam treatment for vascular cognitive impairment(VCI) in varying degrees. Methods:Sixty patients with VCI were selected. Among them,30 patients were with vascular cognitive impairment no dementia(VCIND)and 30 patients with vascular dementia(VD). All patients received conventional base treatment and the oxiracetam treatment. The event-related potentials P300(ERP-P300) and Montreal cognitive assessment(MoCA) of all patients were observed and compared before and after the treatment. Results:After the treatment,the scores of MoCA of all patients showed improvement(VCIND:t = 11.886,P < 0.01;VD:t = 2.242,P < 0.05). After the treatment,ERP-P300 of the VCIND group was improved significantly,the latency period of ERP-P300(P300-PL) was significantly shortened(t = 5.799,P < 0.01) while the amplitude(P300-Amp) was significantly increased(t = 17.603,P < 0.01);but the ERP-P300 of the VD group had no obvious improvement(PL:t = 1.414,P > 0.05;Amp:t = 1.069,P > 0.05). There were significant differences in the scores of MoCA,the latency period and amplitude of ERP-P300 between VCIND group and VaD group(t = 2.135,4.402,7.682,P < 0.05). Conclusion:Oxiracetam are beneficial in the treatment of VCI,especially effective to the patients with VCIND.

    • The effects of group training on psychological health and self-esteem of new prisoners

      Abstract:Objective:To study the effects of group training on psychological health and self-esteem of new prisoners. Methods:An experimental study was conducted among new prisoners. The experimental group consisting of 10 new prisoners accepted psychological intervention by group training,while the control group of 10 new prisoners was not exposed to any intervention. Effects were assessed by comparing the scores of Symptom Checklist 90(SCL-90)and Rosenberg Self-Esteem Scale(RSES) before and after the experiment. Results:There was no significant difference in the scores of SCL-90 and RSES between experimental group and control group(P > 0.05). After group training,the scores showed great improvement in Somatization,Obsessive-Compulsive,Interpersonal sensitivity,Depression,Anxiety,Photic anxiety,Paranoid ideation,Psychoticism and Self-esteem in experimental group(P < 0.05 or < 0.01). Compared with the control group,the scores in Somatization,Interpersonal sensitivity,Depression and Anxiety were significantly decreased while the score of Self-esteem significantly increased(P < 0.01). Conclusion:Group training is an effective method of improving psychological health and self-esteem of new prisoners.

      Abstract:Objective:To research the mRNA expressions of Stat3 and Bcl-2 in process of malignant change of oral premalignant lesions. Methods:The mRNA expressions of Stat3 and Bcl-2 were detected in normal oral mucosa,simple hyperplasia epithelia,dysplasia epithelia and oral squamous cell carcinoma (OSCC) by RT-PCR. Results:The positive rate of Stat3 and Bcl-2 and the semiquantitative value(Stat3/GAPDH,Bcl-2/GAPDH) increased stepwise from normal mucosa,hyperplasia epithelia,dysplasia epithelia to OSCC. The expression of Stat3 was linear positively correlated to that of Bcl-2. Conclusion:There is some relation among Stat3,Bcl-2 and the process of malignant change of oral premalignant lesions.

    • Analysis on epidemiological distribution of serum uric acid and the relationships between serum uric acid and blood lipid and glucose in children and adolescents

      Abstract:Objective:To describe the epidemiological distribution of serum uric acid in children and adolescents,and to provide theoretical basis for early prevention of hyperuricemia-related diseases. Methods:Stratified cluster sampling method was used to select children and adolescents in the age group of 5-15 years in two rural and two urban township schools in Shuyang County,Jiangsu province. With written informed consent,a total number of 1 159 children’s parents were interviewed,and the questionnaire included demographic information,behavior and lifestyles factors. Body height and weight of children were measured by physical examination at the same time of interview. Blood sampling and lab analysis on serum uric acid (UA),fasting glucose (GLU),total cholesterol (TC) and total triglyceride (TG) were conducted among 971 subjects. Results:Significant negative linear correlation was found between UA and GLU (r = -0.11,P = 0.02) among males,whereas no relationship was observed between UA and age,TC,TG and BMI(P > 0.05). For females,UA was observed to be significantly correlated with age and GLU (r = -0.09,P = 0.039;r = 0.13,P = 0.004),but not with TC,TG and BMI(P > 0.05). Logistic regression analysis indicated that UA elevation was associated with BMI among males,and was associated with age and GLU among females. The corresponding odds ratios (ORs) and 95% confidence intervals (95%CIs) were 2.32(1.24~4.35),0.90 (0.84~0.96) and 1.52(1.13~2.04). Conclusion:Age,GLU and BMI might be important determinants of serum UA level of children and adolescents,and have significant differences between males and females.

    • Evaluation of the diet quality among students in the technical college with diet balance index

      Abstract:Objective:To evaluate the diet quality of students in the technical college with Diet Balance Index(DBI),so as to provide the basis for nutritional intervention based on the Dietary Guidelines. Methods:Total 747 students were randomly selected by cluster sampling,their food intakes were investigated with the 24-hour record. The diet quality was evaluated by DBI-07 scoring. Results:Poor food intake was the main problem among these students,while excessive food intake problem also existed. The diet mode C was the main mode for most of the students. Conclusion:There are nutritional problems among the students in the technical college. Targeted nutritional education should be carried out,and the intake should be increased for fruits,milk,eggs and fish to improve the nutritional quality of the students.
