    • The paternalism about “Forced mental illness”

      Abstract:There are three paternalisms in the “Mental health law”,state paternalism,medical paternalism and family paternalism. They are the cornerstone of the normal development mental health. But they also may be violations of the legitimate rights and interests of patients,even the initiator of evil about the normal citizen’s “Forced mental illness”. Practice shows that,if one of the three forces is too strong and be lack of constraints,it is possible to cause “Forced mental illness” phenomenon. Therefore,“Mental health law” should focus on the three forces and take them into an effective processs,so that they can restrict each other,and arouse positive interactions.

      Abstract:Township health centers(THCs) of counties implementing health Ⅺ project were surveyed to promote performance management. Policies on performance management were summarized. The questionnaire investigation of all THCs’ principals on the conditions of performance management was performed. Directors of health bureau and some THCs were interviewed. The contents,subject,frequency,methods and motivational mechanism of performance evaluation were comprehended through the above methods. In the end,several policy recommendations were put forward.

      Abstract:The study of idiopathic outflow tract ventricular arrhythmias supported by the National Science and Technology Pillar Program in the Twelfth Five-year Plan Period is a large-scale and multi-center study,with characteristics such as long time participation,a large sample size et al. By reviewing the project preparation and implementation,some of the experience was summarized. Before the implementation,we tried our best to unify people’s thinking through repeated discussions to ensure the feasibility;to meet regularly to strengthen the communication between the participating centers;to do the preparatory work by reasonable division of responsibility to the people;to communicate patiently with the patient and supervise strictly to ensure the quality and progress of the implementation. This has laid a good foundation and accumulated management experience for the further study of this program.

      Abstract:Objective:The purpose of this study was to determine the characteristics of social support and its impact on the mental health of female needy students in a medical university. Methods:Stratified random sampling method was used to select 442 female needy students in a medical university of Fujian province. Symptom checklist-90(SCL-90) and Social Support Revalued Scale(SSRS) were applied to conduct the survey. Results:Statistical significance in the total score of social support and the score of external support among different level of poverty (P < 0.01) were found. While subjective social support was highly associated with mental health. Curve regressive relation exits between subjective social support,the use to social support and depression,psychopathy. Conclusion:The current financial assistant system and the psychological guidance,social support for female needy students should be improved,which is based on the perspective of sexism.

      Abstract:Surgical practice teaching and regional anatomy are practical medical courses. However,surgical practice teaching and regional anatomy belong to different subjects,including clinical medicine and basic medicine. There are not enough effective convergence and communication between them. For better training medical talents,we broke traditional curriculum framework,combined surgery with regional anatomy teaching and created a practice teaching that contains four stages:“surgical skill training on model and small animals-the autopsy-watching operation,video teaching and virtual surgery-surgical experiments on big animals”. The teaching reform effectively promotes the integration of surgery practice and regional anatomy,the teachers’ teaching level and the students’ knowledge.

Oncology is not a duplication of other subjects. It contains multidisciplinary principles. The students will acquire the ability of complex analysis from this course and be qualified as a specialist.

      Abstract:Medical elements in literature are interaction result of ancient Chinese medicine and classic literature. Four Great Classics in Ming and Qing Dynasty describe the medical activities at great length with four distinct characteristics:cognition,art,exaggeration and fiction. Based on the analysis of above features,Eco-translatology sheds new light on translation strategies of medical elements in literature which are adapted and selected from three viewpoints,namely linguistic,cultural and communicative dimensions. The paper hence,taking Four Great Classics as examples,tries to find out translation principles and methods for the medical elements in ancient Chinese classics.

    • The role of PGE2 on β1-integrin expression and its related mechanism in hepatocellular carcinoma

      Abstract:Objective:To investigate the effect of prostaglandin E2 (PGE2) on the expression of β1-integrin and its related signaling pathway in Huh-7 cells. Methods:Huh-7 cells were treated with PGE2,EP1 receptor agonist(17-phenyltrinor Prostaglandin E2,17-PT-PGE2),EP1 receptor antagonist SC19220,nuclear factor-κappaB(NF-κB)inhibitor PDTC. Western blot and immunofluorescence test were employed to detect the expression of β1-integrin and activation of NF-κB in Huh-7 cells. Results:When Huh-7 cells were treated with 5 μmol/L PGE2 for 24 h,the level of β1-integrin was increased by 129.48% (P < 0.01). EP1 receptor agonist (5 μmol/L 17-PT-PGE2) mimicked the effect of PGE2, and increased the expression of β1-integrin by 216.34%(P < 0.01). EP1 receptor antagonist SC19220(10 μmol/L) suppressed PGE2-mediated expression of β1-integrin by 34.51% (P < 0.05). In immunofluorescence assays,NF-κB translocated into the nucleus induced by 17-PT-PGE2 in Huh-7 cells. Further,Western blot assays showed that the level of Phospho-NF-κB-P65 was increased by 209.27%(P < 0.01)after treatment of 5 μmol/L 17-PT-PGE2 for 120 min. NF-κB inhibitor PDTC decreased EP1-mediated expression of β1-integrin by 63.49% (P < 0.01). Conclusion:PGE2 might up-regulate the expression level of β1-integrin through EP1 receptor in Huh-7 cells,which was partly related to the NF-κB signaling pathway.

    • Correlational study between prolactin promoting breast cancer cell proliferation and microRNA expression profiling change

      Abstract:Objective:To explore changes of microRNA expression profiling in human breast cancer cell based on prolactin(PRL) treatment,and to understand their potential roles in breast cancer initiation and progression. Methods:Human breast cancer T-47D cells,in which PRL receptor(PRLR) is highly expressed,were treated with or without human recombinant PRL,two group cells were subjected to detect their proliferation,cell cycle and apoptosis alteration by the MTT assay and flow cytometry method (FCM). miRNA expression profile of PRL treated and untreated cells were analyzed by Solexa sequencing technology,miRNAs were validated by real-time PCR and undertaken bioinformatic analysis. Results:In MTT results,it showed that T-47D cell proliferation ability was enhanced by PRL treatment. FCM revealed that G1 phase cells were reduced,while S phase cells were increased. Meanwhile,cell apoptotic rate was reduced in PRL group. Two miRNAs expression profiles were successfully analyzed from PRL-treated and untreated T-47D cells by Solexa sequencing technology. Within the Solexa sequencing results,821 and 798 miRNAs were detected in the two libraries,respectively. 428 miRNAs were co-expressed,and 42 miRNAs were significantly differentially expressed. Four miRNAs were validated by real-time PCR,their intracellular expression was consistent with Solexa sequencing data. Besides,86 and 115 novel miRNAs were also detected in the two libraries,and 46 novel miRNAs were co-expressed. Conclusion:PRL can enhance breast cancer cell proliferation ability,and miRNAs expression profiles in T-47D cells treated with or without PRL are successfully analyzed by Solexa sequencing technology. Series of PRLR signaling pathway related miRNAs were harvested. This study provides a reference for elucidating the complex miRNA-mediated regulatory networks of PRL/PRLR signaling pathway that affect breast cancer tumorigenesis and progression.

    • The mechanism of up-regulated TLRs in ovarian cancer incidence and development

      Abstract:Objective:To investigate the role of transduction molecules and modulatory factors of signal pathways of Toll-like receptors(TLRs) in ovarian cancer. Methods:We collected peripheral blood mononuclear cell (PBMC) from 24 ovarian cancer patients,22 benign diseases,and 22 healthy females. Total RNA of the PBMCs was extracted,and reverse transcripted into cDNA. Expression levels of TLR1~TLR9 mRNA were then determined by real-time quantitative PCR. PBMCs were then either unstimulated (-) or stimulated with agonists for TLRs and assessed for IL-1β,IL-6,IL-12P40 by real-time PCR. Results:TLR1~TLR9 were all expressed in PBMC of the three groups and the expression levels of TLR2,TLR6 mRNA in patients with ovarian cancer were higher than the healthy controls (TLR2:F = 3.27,P < 0.05;TLR6:F = 2.21,P < 0.05). Ovarian cancer patients aslo showed increased TLR2 levels compared to benign diseases group(F = 3.24,P < 0.05).The expression of TLRs has no difference between benign diseases group and the healthy controls. After stimulated with HKLM for 24 h,ovarian cancer group demonstrated a significant up-regulation in expression of IL-1β,IL-6,IL12-P40. The expressions of IL-1β,IL-6,IL12-P40 at the mRNA level were significantly higher in ovarian cancer group compared to healthy controls (F = 2.24,P < 0.05;F = 2.94,P < 0.05;F = 2.35,P < 0.05) and benign diseases group (F = 2.02,P < 0.05;F = 2.50,P < 0.05;F = 2.85,P < 0.05). Furthermore,the level of IL-1β,IL-6,IL12-P40 mRNA had no difference between benign diseases group and healthy controls. Conclusion:TLR2 and TLR6 were highly expressed in ovarian cancer patients,inflammatory reaction mediated by IL-1β,IL-6,IL-12P40 may be related to the growth and development of ovarian cancer.

    • The shotgun analysis of brain proteins composition changes in filial mice after the pregnant mice exposure to PFOS

      Abstract:Objective:To clarify the changes in the filial mice brain proteins after the pregnant mice exposure to Potassium perfluorooctanesulfonate(PFOS). Methods:The 3 days pregnant mice were randomly divided into 2 groups. The mice in the control group were orally administered with the normal saline. The mice in the other group were orally administered with 0.5mg/kg PFOS for 7 days. The filial mice(within 24 hours) born from the control group(marked as A group) and 0.5 mg/kg PFOS-exposed group (marked as B group) were killed and the brain proteins were collected and homogenized respectively. Changes in brain proteins of the filial mice of A group and B group were examined by shotgun analysis. Results:Compared with A group,the categories of brain proteins in B group were significantly changed and some uncharacterized proteins were detected. Conclusion:Orally administration of PFOS to the pregnant mice results in the abnormal changes in the brain proteins of the filial mice.

    • The effects of peptide hydrogel composite scaffold on the proliferation and differentiation of RAT adipose-derived stem cells

      Abstract:Objective:To study the effects of peptide hydrogel composite scaffold on the proliferation and early osteogenic differentiation of rat adipose-derived stem cells(ADSCs). Methods:Nano-Hydroxyapatite/Collagen(nHAC) scaffold was coated by 1% peptide hydrogel. nHAC without peptide coating was set as control. They were characterized by field emission scanning electron microscope (FESEM) and confocal laser scanning microscope (CLSM). ADSCs were chosen to seed onto the nHAC coated and uncoated with peptide hydrogel. CCK-8 kit was used to observe cell proliferation. The alkaline phosphate(ALP) activity was detected the early osteogentic differentiation. Results:Peptide coating was formed on the surface of scaffold. ADSCs adhered to and fully extended on the peptide hydrogel composite scaffold under CLSM. The data of proliferation and ALP continually increased in both groups. There was no difference in cell proliferation at 1 d,3 d(P > 0.05). At 5 d,7 d,ADSCs on the coated nHAC proliferated more significantly(P < 0.05). There was no difference among 3 d,5 d and 7 d in the ALP activity (P > 0.05). Conclusions:The peptide hydrogel composite scaffold could improve adhesion and proliferation of ADSCs,and also can improve early osteogentic differentiation of the cell, although not so obvious as the former two.

    • Effect of PGC-1α on the survival rate and reactive oxygen species of injured neurons induced by oxygen-glucose deprivation

      Abstract:Objective:To explore the protective effects and mechanism of Peroxisome proliferative activated receptor-γ activation of auxiliary factor 1 Alpha(PGC-1α) on injured neurons induced by oxygen-glucose deprivation(OGD) in embryonic mice.Methods:Cortical neurons cultured for 7 days were randomly divided into four groups:normal control group,oxygen-glucose deprivation group(OGD);OGD + PGC-1α overexpression plasmid group(overexpression group) and OGD+no-load plasmid group(no-load group). Overexpression group and the no-load group pretreatment with PGC-1α overexpression plasmid and no-load plasmid respectively before OGD treatment. 24 hours after OGD,cell survival rate was detected by MTT(Methylthiazolyldiphenyl-tetrazolium bromide),observe the expression of PGC-1α was using Western blot technology,and the flow cytometry was used to monitor the content of Reactive Oxygen Species (ROS). Results:Compared with the normal control group,the cell survival rate in OGD group decreased(P < 0.01),the survival rate was increased in overexpression group due to pretreatment of PGC-1α overexpression plasmid,which was compared with OGD group and no-load group with a significant difference(P < 0.01). The ROS of OGD groups were significantly higher than that of the normal group (P < 0.01);the content of ROS was decreased in overexpression group,and was significantly lower than that of the OGD group and no-load group (P < 0.01). Conclusion:The study indicated that PGC-1α can significantly reduce the ROS content of oxygen-glucose deprivation neurons and improve its survival rate,to protect the neurons.

    • The effect of transbronchial gene transfer of human interleukin-10 gene to donor lung ischemia-reperfusion injury in the rat’s lung transplantation

      Abstract:Objective:To investigate the effect of transtracheal transfering the adenovirus-mediated human interleukin 10 (hIL-10)gene on lung ischemia-reperfusion injury(IRI),and posttransplant lung function in SD rat lungs. Methods:Thirty-six male SD rats were divided into 3 groups randomly:blank control group,empty vector-control group and gene transfer group. The donors of three groups received 0.5 ml of normal saline,5×109 PFU of Ad-5 or 5×109 PFU of Ad-5-hIL-10,respectively. After 24 h of in vivo transfection,lungs were harvested and stored in low-potassium dextran solution(LPDG) at 4℃ for 18 h. Then the left lung orthotopic transplantation was performed. After 4 h of reperfusion,arterial blood gas analysis and wet-to-dry(W/D) weight ratios were determined,superoxide dismutase (SOD) activity,malondialdehyde (MDA) contents and myeloperoxidase (MPO) activity were detected. Tumor necrosis factor-α(TNF-α) and interferon-γ(IFN-γ) were measured in graft tissue by ELISA. Besides,hIL-10 was detected by immunohistochemistry. Graft pathohistological changes were examined under a light microscope. Results:Empty vector and blank control groups showed a significant lung ischemia-reperfusion injury(IRI),while gene transfer group’s lung IRI reduced significantly. The lung tissue edema,interstitial inflammation and exudation in gene transfer group were obviously reduced. Expression of hIL-10 gene was obviously incrdased when detected by immunohistochemistry (IHC). Partial pressure of oxygen (PaO2) levels in the gene transfer group were higher than in empty vector and blank vector groups (P < 0.01),respectively. W/D ratios were reduced in hIL-10 lungs (P < 0.05) compared with the other groups. MDA contents and MPO activity were reduced in tissue of the gene transfer group(P < 0.01). But the activity of SOD was increased significantly (P < 0.01). TNF-α and IFN-γ were reduced in tissue of the gene transfer group(P < 0.01) measured by ELISA. Conclusion:Transtracheal adenovirus mediated gene transfection of hIL-10 can obviously lighten IRI after lung transplantation and improve early lung function effectively after lung transplantation.

      Abstract:Objective:To investigate the beneficial effects of ginsenoside Rg1 (Rg1) in the treatment of cardiac hypertrophy induced by transverse aortic contraction(TAC) and the potential molecular mechanisms in rats. Methods:Adult male SD rats were conducted in the study;twenty-four rats which are managed to present left ventricular hypertrophy(LVH) after TAC were randomly divided into two groups:the TAC group and the Rg1 group. Another 12 rats which was absence of the aortic artery ligated after analogous operation was included in the Sham group. The administration of Rg1 was conducted at 10 mg/(kg·d) for 4 weeks. After that,the left ventricular mass index (LVMI) were examined as hypertrophic parameters;left ventricle sections were stained with hematoxylin and eosin for ventricular wall thickness and myocardial microstructure. The mRNA transcription of ANP,BNP and HIF-1α was detected by Real-time quantitative PCR. The protein expression of HIF-1α was evaluated through Western blot test. Results:Compared with TAC group,the treatment with Rg1 significantly reduced the LVMI,the size of cardiomyocyte,and the mRNA expression of transcripts of ANF,BNP (P < 0.05,respectively). In the Rg1 group,the protein and mRNA levels of HIF-1α were significantly increased comparing to the control group(P < 0.05,respectively). Conclusion:Ginsenoside Rg1 attenuates the left ventricular hypertrophy and improves the cardiac function in the experimental rat model after TAC undertaken. This beneficial effects may be partially attributed to the increase of HIF-1α expression.

      Abstract:Objective:To study the protective effect and mechanism of pyrroloquinoline quinine(PQQ) against UVA induced human skin fibroblasts. Methods:Human skin fibroblasts were cultured in 6-well plate in vitro with an equal number of cells(104~106 per well). HSF exposed to UV irradiation at a dose of 9 J/cm2. Pyrroloquinoline quinine(50,100,200 ng/ml) and 8-methoxypsoralan (80 ng/ml) were added into the cell culture medium before and after the irradiation. 8-MOP was used to enhance UVA absorption. After 72 hours of irradiation, SA-β-Gal staining was performed to evaluate the senescence state,change of mitochondria membrane potential was examined by JC-1 staining using fluorescence microscope,JC-1 staining was also used to test mitochondria membrane depolarization by flow cytometry(FCM),and real-time PCR was used to determine mRNA expression of senescence-associated gene of mmp1,mmp3. Results:Cells cultured with lower dosage PQQ (50 ng/ml) exhibited negative SA-β-Gal staining. JC-1 staining showed the change of mitochondria membrane potential was reversed by lower dosage PQQ (50 ng/ml). Mitochondria membrane depolarization was relieved lower dosage PQQ(50 ng/ml). Meanwhile,the gene expression of senescence-associated signals of mmp1,mmp3 decreased in PQQ group compared with control group. Conclusion:Pyrroloquinoline quinine can protect human skin fibroblasts from aging induced by UVA irradiation via mitochondria pathway.

      Abstract:Objective:To investigate expression and significance of PRL-3 in thyroid papillary carcinoma and its relationship with clinicopathologic features. Methods:The expression of PRL-3 in primary thyroid papillary carcinoma,the adjacent non-tumor tissues and lymph node metastasis of the thyroid papillary cancer(LMNs) were detected by Real-time PCR and SP immunohistochemistry. Results:The transcription expression of PRL-3 mRNA was higher in primary thyroid papillary carcinoma or LMNs than in the adjacent non-tumor tissues(P < 0.05) .The positive expression rates of PRL-3 were also higher in primary thyroid papillary carcinoma and LMNs than in the adjacent non-tumor tissues(P < 0.05). The positive rates of PRL-3 protein were higher in primary thyroid papillary carcinoma with lymph node metastasis than in primary thyroid papillary carcinoma without lymph node metastasis(P < 0.05). The positive rates of PRL-3 were higher in tumor of size > 2 cm than in tumor of size≤2 cm (P < 0.05).Conclusion:The expression of PRL-3 is closely related to the tumor size and lymph node metastasis. The high expression of PRL-3 in PTC may forecast the lymph node metastasis,and it may be a molecular marker to predict lymph node metastasis in papillary thyroid carcinoma.

      Abstract:Objective:To investigate the expression of Sox2 in human bladder cancer and evaluate the effect of Sox2 on the proliferation of bladder cancer cell line T24. Methods:Immunohistochemical and immunofluorescence staining was used to detect the expression of Sox2 in bladder cancer. The constructed pCDNA3.1-SOX2-EGFP plasmid was transfected into bladder cancer cell line T24. The expression of Sox2 in T24 was analyzed by RT-PCR and Western blot. The function of Sox2 protein in cell proliferation was measured by MTT assay. Results:The high expression of Sox2 was detected in 68.6% of bladder cancer specimens. The pCDNA3.1-SOX2-EGFP plasmid was successfully constructed. In addition, MTT assay showed that Sox2 could significantly promote the proliferation in human bladder cancer cell line T24. Conclusion:The expression of Sox2 was detected in bladder cancer. Sox2 might promote the growth of human bladder cancer cell line T24.

      Abstract:Objective:To explore the expression of vascular endothelial growth factor(VEGF) in esophagus of patients with portal hypertension and its relationship with the dynamics of portal vein and spleen. Methods:Thirty-six patiens who were undergone selective devascularization were received Doppler ultrasound examination, and the diameter and mean flow rate of portal vein and splenic vein were mesured before operation, and the blood flow were calculated. The esophagus tissue was reserved and FPP(free portal hypertension) was measured by puncturing the right gastroepiploic vein during the operation. Immunohistochemistry was used to detect the expression of VEGF in esophagus tissues. Results:The ranking of VEGF expression in esophagus tissues:0(-)3 cases;1-4(+)10 cases;5-8(++)18 cases;9-12(+++)5cases. The positive rate was 91.7% and the high expression rate of VEGF in esophagus is 63.9%. The diameter and flow of portal vein and splenic vein of the VEGF high expression group is higher than those of the VEGF low expression group(P < 0.01). The velocity of portal vein of the VEGF high expression group is lower than that of the VEGF low expression group (P < 0.01),but there was no obviously significant in the velocity of splenic vein between two groups(P < 0.01). The free portal pression of the VEGF high expression group is obviously higher than that of the VEGF low expression group(P < 0.01). Conclusion:The expression of VEGF in esophagus tissues of portal hypertension patients was high, and its high expression was related to parameters of portal and splenic hemodynamics and free portal pression. VEGF-mediated angiogenesis was one of the factors that caused hyperperfusion of splanchnic circulation in portal hypertension patients.

      Abstract:Objective:To test whether the level of serum inflammatory factors could predict the occurrence of ventilator-associated pneumonia(VAP) undergoing cardiopulmonary bypass(CPB) cardiac surgery. Methods:Twenty valve replacement patients were categorized into two groups based on the occurence of VAP following CPB,VAP group(A) and NVAP group (B). The blood samples of left atrium were collected in both groups at the time points of preoperation(T1),20 min after CPB(T2),40 min after CPB(T3) and end(T4) of CPB. Expression of serum IL-10,IL-1β and TNF-α were detected by ELISA. The common and CPB data of all cases in two groups were compared. Results:There was no significant difference in TNF-α and IL-1β in each time point between VAP group and NVAP group. The expression of IL-10 and IL-10/TNF-α ratio in NVAP group at the time points of T3 and T4 were significantly higher than VAP group (P < 0.05). The occurance of VAP was correlated with the CPB time. Conclusion:The increase of the expression of IL-10 could significantly decrease the occurence of VAP.

      Abstract:Objective:To observe the clinical effect of melanocytes transplantation on vitiligo with coating method in 32 patients.Methods:The medium was prepared with recombinant human granulocyte colony-stimulating factor(rhG-CSF) as the major growth factor,mixed with carbomer gel to constitute the system of coating on the treatment of vitiligo with melanocytes transplantation.Therapeutic evaluation was assessed with certain standard. Results:All patients had repigmentation for more than 20%,in which the repigmentation of six cases was more than 90%,11 cases for 70% to 80% and 15 cases for 30% to 50%. The total effective rate was 100%. The 84.4% of the patients showed evenly repigmentation. No obvious mottling was observed. Conclusion:Melanocytes transplantation on vitiligo with coating method is safe and effective.

      Abstract:Objective:To explore the application value of the single step dilatational tracheostomy(SSDT) in critically ill patients in the intensive care unit(ICU). Methods:Consecutive sixty-four patients were randomly divided into SSDT group(32 cases) or traditional surgical tracheostomy(ST) group(32 cases). The operation time,incision size,blood loss,changes of vital signs,and postoperative complications such as infection rate,incidence rate of subcutaneous emphysema/pneumothorax,infection rate,and procedure-related mortality were observed. Results:The operation time,amount of blood loss,size of operative incision and changes of vital signs in the SSDT group were superior to those in the ST group (P < 0.05). While the rate of overall complications including wound bleeding,subcutaneous emphysema/pneumothorax,overflow and incision infection was lower in the SSDT group(9.4%) compared with the ST group (53.1%) (P < 0.05). Conclusion:Compared with the ST,the SSDT is more effect,safe,and simple with less perioperative complications for critically ill patients.

      Abstract:Objective:To investigate the association of mitochondrial NDUFB6 gene with type 2 diabetes by single nucleotide polymorphism (SNP) detection and genotyping. Methods:Sequences of coding regions (including sequence near splicing site) of NDUFB6 gene was detected by PCR-sequencing in 15 type 2 diabetics and 9 normal controls. To high frequency SNPs,PCR-sequencing was used in 192 type 2 diabetics and 192 normal controls for geno-typing. Results:A total of 6 SNPs were identified for NDUFB6 gene;there were 3 high frequency SNPs and 3 low frequency SNPs. One SNP(-1211C > T) located in the promoter region of NDUFB6 had significant difference of genotype distribution between type 2 diabetics and controls (P < 0.05). Conclusion:The NDUFB6 gene -1211C > T polymorphism may associate with type 2 diabetics in Chinse.

      Abstract:Objective:To analyze the changes of carotid-femoral and carotid-radial pulse wave velocity in patients with obstructive sleep apnea hypopnea syndrome (OSAHS)associated with hypertension,and effect factors on these changes. Methods:One hundred and sixty research objects were classified into four groups according to the hypertension history and polysomnography(PSG):control group (40 patients),hypertension group (40 patients),OSAHS group (40 patients),and OSAHS with hypertension group (40 patients). Automatic pulse wave velocity measuring system was applied to examine carotid-femoral pulse wave velocity(c-fPWV) and carotid-radial pulse wave velocity(c-rPWV) as the parameters reflecting central elastic large arterial and peripheral muscular medium-sized arterial elasticity respectively. Results:Compared to control group,hypertension,OSAHS,and OSAHS with hypertension groups had significantly higher c-fPWV(F = 23.056,P < 0.05),and OSAHS with hypertension group had significantly higher c-fPWV than hypertension group[(10.72 ± 1.06)m/s]vs[(10.09 ± 1.05)m/s,P < 0.01]. Compared to control group,hypertension,OSAHS with hypertension group had significantly higher c-rPWV(P < 0.01). C-fPWV correlated significantly with c-rPWV (all P < 0.01) by both Pearson and partial correlation analysis adjusted by age,body mass index,waist-hip ratio,systolic blood pressure,diastolic blood pressure and heart rate. Multiple stepwise regression analysis showed that the age,systolic blood pressure and heart rate were positively while female gender was negatively correlated with c-fPWV (R2 = 0.537,P = 0.000). C-rPWV was positively correlated with diastolic blood pressure(R2 = 0.052,P= 0.041).Conclusion:Our study showed that arterial stiffness was increased in OSAHS patients,and with OSAHS exacerbations,c-fPWV value increased. Arterial stiffness was more severe in the patients with OSAHS and hypertension. The change of c-fPWV of patients with OSAHS was related to age,female gender,systolic blood pressure and heart rate. C-rPWV was associated with diastolic pressure. The PWV would be an applicable functional index in early diagnosis of vascular pathological changes and in medical evaluation . Examining the change of large arterial elasticity has important clinical value in OSAHS patients.

      Abstract:Objective:To explore the epidemic characteristics of infective diarrhea disease in Jiangsu province,and to provide scientific basis for effective strategy for general control and monitoring of diarrhea disease. Methods:The epidemiological characteristics and etiologic characteristic of infective diarrhea disease in Jiangsu from 2004 to 2011 were analyzed. A total of 246 hospital of county and above level were investigated to understand the capability of breadboard detection. Results:The incidence of infective diarrhea disease in Jiangsu has been decreasing by years from 2004 to 2010,the incidence bounced in 2011. The average incidence rate was 43.79/100 000. There was a visible supernumerary tendency of constituent ratio in other diarrhea disease. The typical and aestival peak was observed in summer,but there was a regularity and specificity which “V” type distribution was observed from August to October. With occupational distribution,peasants,students,workers and scattered children were the major infected population. Piecemeal decrease of incidence rate was following with aetatis increase. The incidence rate of male was higher than female.The incidence rate and capability of breadboard detection in southern part were higher than central region,and central region were higher than north part. Conclusion:The incidence of infective diarrhea disease has been well controlled. There was a visible supernumerary tendency in epidemiological characteristics. Further step should be taken for the capability of laboratory,diagnose and positive report.

    • Separation and analysis of protein complexes in plasma membrane

      Abstract:Objective:Discuss the methods of plasma membrane and its protein complex separation and qualitative. Methods:We evaluated the application of “density gradient ultracentrifugation”and“aqueous two-phase partition”to separate high-quality plasma membrane. Western blot and transmission electron microscope were used in the following research to validate the purity of the separated cell membrane. Results:We applied the blue-native method to process the one-dimensional separation of the protein complex in the cell membrane and 23 kinds of protein complex were separated. We also processed the second-dimensional separation of the 23 kinds of complex and identified 135 proteins, of which 78 were proteins correlated to plasma membrane and 57 were from endomembrane system. Conclusion:Our results show that the combination of different kinds of purification methods can effectively increase the protein separation rate and separation purity. High purity membrane on plasma membrane proteome research has important significance.

      Abstract:Objective:Synthesis of L-2,6-dimethylphenylalanine and L-2,4,6-trimethylphenylalanine through enzymatic resolution. Methods:Racemic 2,6-dimethylphenylalanine and 2,4,6-trimethylphenylalanine were synthesized through reaction of diethyl acetamidomalonate with 2,6-dimethylbenzyl bromide or 2,4,6-trimethylbenzyl bromide,respectively. The DL-amino acids were then enzymatically resoluted by L-amino acylase. Results:Racemic 2,6-dimethylphenylalanine and 2,4,6-trimethylphenylalanine were synthesized through reaction of diethyl acetamidomalonate with 2,6-dimethylbenzyl bromide and 2,4,6-trimethylbenzyl bromide in yield of 87.9% and 86.0% respectively;DL-2,6-dimethylphenylalanine was successfully resoluted by L-amino acylase,but it was failed in the resolution of 2,4,6-trimethylphenylalanine. Conclusion:L-amino acylase can be used in the resolution of DL-2,6-dimethylphenylalanine,but it is ineffective in the resolution of 2,4,6-trimethylphenylalanine.
