      Abstract:Objective:To study the protective effects of nicotine on cartilage of monoiodoacetic acid(MIA)-induced rat osteoarthritis model. Methods:A total of 40 rats were randomly divided into four groups(control group,MIA group,MIA+Nic 0.25 mg/kg group and MIA+Nic 0.5 mg/kg group). Each group rats were sacrificed on 30th day after they received the first intra-articular injection of MIA(1mg) surgery. The right knee samples were carefully dissected. Generally light microscope and histological pathological examination were used to observe the destruction of articular cartilage. Reverse-transcription PCR was used to detect the gene expression of type II collagen and aggrecan. The protein expression of phosphor-Akt was measured by Western blot test. Results:Compared with the MIA group,nicotine treatment significantly decreased the light microscopy score and the Mankin’s score(P < 0.01) of the knee samples,and significantly increased the mRNA levels of typeⅡ collagen and aggrecan (P < 0.01) and the protein levels of phosphor-Akt (P < 0.01). Conclusion:Nicotine attenuates the severity of cartilage degeneration in the experimental rat model. These protective effects may partially benefit from the increase of phosphor-Akt expression.

      Abstract:Objective:This study evaluated myocardium damage under the irradiation of ultrasound combined with homemade nitric oxide (NO) microbubbles. Furthermore,we intended to investigate the stromal cell-derived factor-1(SDF-1) expression in rat myocardium after the intervention of ultrasound combined with NO microbubbles. Methods:Thirty-two rats were divided into four groups randomly. Ultrasound (1MHz,1W/cm2) was applied to the anterior chest for ten minutes in group one (US+NOMB),group two (US+MB) and group three (US+PBS),group four was set for contrast. Four rats were sacrificed in each group four hours later and myocardium was harvested for HE staining. Serum was also collected for myocardial enzyme testing. One week later,all rats were sacrificed,and RT-PCR and western blotting were performed for testing SDF-1 expression. Results:HE staining showed that there were no significant differences in pairwise comparison. Creatine kinase and lactic dehydrogenase were higher in the group one and the group two than those in the group three and group four (P < 0.05),but there was no significant difference between the group one and the group two (P > 0.05). Troponin I was lower in the group one than that in group two,but higher than those in the group three and group four (P < 0.05). RT-PCR and Western blotting demonstrated that the expression of SDF-1 was higher in the group one than that in the group two,group three and group four,and all the P values were less than 0.05. Conclusion:NO microbubble,as a novel microbubble,is safe for rat myocardium. Moreover,it can induce a higher SDF-1 expression than common lipidic microbubbles,which shows great potential for stem cells homing.

      Abstract:Objective:To explore effects of H2S on expression of COX-2 on macrophages(RAW264.7) and formation of macrophage-derived foam cells. Methods:RAW264.7 foam cell model induced by low-density lipoprotein (Ox-LDL) is established,then added with different concentrations of H2S. Detection of foam cell formation is conventionally practiced by Oil Red O staining. The expressions of COX-2 mRNA and protein are detected by real-time qPCR and Western blot respectively. Results:A significant increase of the production of COX-2 and the deposition of intracellular lipid was observed. Different concentrations of H2S can considerably reduce the expression of COX-2 and the deposition of intracellular lipid induced by the Ox-LDL,especially the concentration of 250 μmol/L and the difference is statistically significant(P < 0.05). Conclution:H2S can significantly reduce the expression of COX-2 in macrophages(RAW264.7 )and inhibit the formation of foam cells .

      Abstract:Objective:To investigate the effects of gabapentin(GBP) on free calcium concentration[Ca2+]i of injury DRG in an experimental rat model with neuropathic pain following spinal nerve ligation. Methods:Thirty five male SD rats aged 4~6 weeks were used in this study. Left L5 spinal nerve was ligated between DRG and sciatic nerve and cut distal to the ligature to reproduce neuropathic pain model. On the 15th postoperative day,these animals were decapitated. L5 DRGs were isolated and the neurons in the ganglion were enzymatically dissociated in SNL group and Sham group. Flow cytometer was used to monitored the resting[Ca2+]i and high K+(50 mmol/L)-evoked[Ca2+]i with or without GBP in L5 DRG. Results:GBP attenuated high K+(50 mM)-evoked[Ca2+]i in a dose-dependent manner(P < 0.05 or P < 0.01),but no effect on resting[Ca2+]i. Conclusion:GBP significantly inhibited high K+-evoked[Ca2+]i in injured DRG neurons in neuropathic pain rats,which may be a peripheral way of GBP which involved in the anti-nociception mechanism.

      Abstract:Objective:To explore circadian pattern of β-klotho and regulation function of core circadian gene BMAL1. Methods:The circadian expression patterns of β-klotho in cell level and liver tissue of mouse were detected by Real-time PCR .Plasmids expressed BMAL1 and CLOCK were prepared,then transfected into HepG2 cells to analyze β-klotho expression level by RT-PCR method. Furthermore,the expression level of β-klotho in the BMAL1 knockout mice was also detected. Results:β-klotho displays a robust circadian expression pattern. At the molecular level,core clock elements BMAL1 and CLOCK coordinately activate β-klotho transcription. Conclusion:β-klotho is a new clock-controlled gene.

      Abstract:Objective:To investigate the level of hyaluronan synthase 2(HAS2) expressed in normal zone fibroblasts(NFs) and cancer-associated fibroblasts(CAFs),as well as its impact on squamous cancer cells of the tongue. Methods:CAFs and NFs were received from carcinoma and para-carcinoma tissue of the same tongue cancer patient,respectively. The cell phenotype was verified by immunocytochemistry and Western blot. The invasiveness of Cal27 in NFs,CAFs and control group was compared by transwell chamber assay. The expression of HAS in fibroblasts was measured by Western blot and real-time RT-PCR. HAS2 was knocked down by siRNA,and the invasiveness of Cal27 in HAS2-interferenced group and control group was surveyed by transwell chamber assay. Results:α-SMA was expressed in CAFs but not NFs. Cal27 had highest invasiveness in CAFs group in comparison with NFs group or control group (P < 0.01). The level of HAS2 expression was 6 times higher in CAFs than that in NFs determined by real-time RT-PCR(P < 0.01) and 2 times higher determined by Western blot. The invasiveness of Cal27 was largely inhibited by down-regulation of HAS2 in CAFs in contrast to control group(P < 0.01). Conclusion:CAFs promoted the invasiveness of oral tumor cells mostly by higher level of HAS2 in vitro,and inhibition of HAS2 in CAFs may be a new therapy for anti-tumor invasion and metastasis.

      Abstract:Objective:MicroRNA-199a-3p (miR-199a-3p) is down-regulated in many kinds of tumors,including osteosarcoma. This study was aimed to investigate the effect of miR-199a-3p on apoptosis of human osteosarcoma cells (U2-OS). Methods:Human osteosarcoma cells(U2-OS) were transfected with miR-199a-3p mimics or negative control mimics (NC mimics) as negative control. MiR-199a-3p expression level was detected by quantitative real-time reverse transcriptase polymerase chain reaction (qRT-PCR). Expression level of myeloid cell leukemia-1 (MCL-1) protein and cleavage of poly ADP-ribose polymerase(PARP) were detected by Western blot. Cell apoptosis was detected by flow cytometry. Results:Compared with control group,miR-199a-3p expression level of experiment group was significantly up-regulated,MCL-1 protein expression was reduced,cleavage level of PARP was increased and cell apoptosis rate of experiment group was significantly increased. The differences between experimental and control groups have statistical significance(P < 0.05). Conclusion:MiR-199a-3p can dramatically promote apoptosis of human osteosarcoma cells.

      Abstract:Objective:The aim of this study was to investigate the interventional effects of breviscapine on myocardial tissue of and neuropeptide Y level of diabetic rats. Methods:Experimental diabetes model of wistar rats were established by intraperitoneal injection of streptozotocin. They were randomly divided into untreated group,insulin treatment group,low dose breviscapine intervention group,high dose breviscapine intervention group and normal control group. The body weight and blood glucose at state of empty stomach were measured before execution. Furthermore,cardiac mass index and heart coefficient of rats were measured after excution. NPY levels in plasm and myocardial tissue were measured by radioimmunoassay(RIA). The pathologic changes of myocardium were observed using HE staining. Results:Cardiac mass index,heart coefficient and NPY levels in plasma and myocardial tissue of diabetic rats were significantly increased. Various pathological changes were observed in diabetic rats. TRI and breviscapine intervention improved myocardial tissue pathological changes of diabetic rats,the effect of high dose intervention of breviscapine is similar to that of insulin therapy. NPY levels were decreased in plasma during breviscapine intervention,while increased in myocardial tissue. The results showed that NPY levels were significantly higher in diabetic rats as compared with control group. Conclusion:Breviscapine didn’t reduce the level of blood glucose,however,it improved physiological functions,such as myocanrdium hypertrophy and cardiac function in rats with diabetic cardiomyopathy.

    • The relationship between serum uric acid levels and metabolic diseases in elderly

      Abstract:Objective:To study the relationship between hyperuricemia and disease related to metabolic in the elderly. Methods:All 7304 adults above 60 years were enrolled in 2009~2010 at the medical examination center of the First Affiliated Hospital of NJMU. The medical history,the biochemical index and the general physical data were evaluated. The sex-specific cutoff for serum uric acid quartiles and multiple stepwise by multinomial Logistic regression model was used to analyze the relationship between uricemia and ages,sex,cholesterol,and other factors related to metabolism. Results:The incidence rates of hyperuricemia in men were higher than that in women at the same age group. The uric acid level increased with the ages. BMI,systolic blood pressure and diastolic blood pressure,fasting plasma glucose,especially TG and TG/HDL-C were positively correlated with blood uric acid(P < 0.05). The blood uric acid level was also positively correlated with blood pressure grades. Blood uric acid level of those with IFG was significantly higher than that of diabetic patients. The incidence of diabetes mellitus increased in hyperuricemia group than the non- hyperuricemia group. The blood uric acid level was positively associated with blood glucose in men,no such correlation observed in women. When the sex-specific cutoff for serum uric acid quartiles and multinomial Logistic regression model were used,the analysis revealed that ages,BMI,blood pressure,cholesterol,high density lipoprotein,cholesterol,TG/HDL-C,fasting plasma glucose and blood uric acid levels were significantly positive correlated. Conclusion:The blood uric acid level was closely related to ages,BMI,hyperlipidemia,hyperglycemia,hypertension and other constitution of metabolism. It is important to investigate the possible benefits of anti-hyperuricemia treatment for preventing complications in cardio-celebral-vascular system.

      Abstract:Objective:To evaluate the activity of FORT on assessing ovarian response and predicting pregnancy outcome by FORT in IVF-ET. Methods:Three thousand and two hundred and nighty-two patients of controlled ovarian stimulation,divided into low,medium and high groups based on FORT,were investigated and each activated by rFSH 225 U. The controlled ovarian stimulation data of these patients and their laboratory index were analyzed. Results:Antral follicle count (AFC) is the lowest in the high FORT group and the highest in the low FORT group,preovulatory follicle count (PFC),as well as E2 with injection of HCG in the same day,are the highest in the high FORT group and the lowhest in the low FORT group. The number of retrieved oocytes,the total number of embryos and the number of high quality embryos escalate increased consistently with the FORT in these group,and the differences were significant amony group amparison comparison. Meanwhile,the embryo implantation rate and the clinical pregnancy rate also differ statistically in these groups and reach to the highest level in the high FORT group. Condusion:FORT is an important tool to study the reactivity of ovary. The high FORT group has better pregnancy outcome.

    • Expression and clinical significance of FcγRⅡ and VEGF-A in colorectal cancer

      Abstract:Objective:To investigate the expression of FcγRⅡFc fragment of IgG,low affinity IIa,receptor(FcγRⅡ) and vascular endothelial growth factor A(VEGF-A) in colorectal cancer and analyze their association,and consequantly explore the mechanism of FcγRⅡin colorectal cancer. Methods:Expression of FcγRⅡand VEGF-A and their association with clinicopathological features were determined using real time polymerase chain reaction(RT-PCR) in 56 cases of colorectal cancer patients. The distribution of the clinicalpathologic and the expression of VEGF-A and FcγRⅡwere analyzed using Fisher’s exact test and the Chi-squared test. Spearman’s rank correlation was used to analyze the correlation between FcγRⅡand VEGF-A expression. Results:FcγRⅡmRNA expression in colorectal cancer tissues was significantly lower than that in matched adjacent normal tissues. FcγRII mRNA expression was significantly related to the patients’ infiltration depth and clinical stage(P < 0.05). VEGF-A mRNA expression in colorectal cancer tissues was significantly higher than that in matched adjacent normal tissues. VEGF-A mRNA expression was significantly related to the patients’ age,tumor size,infiltration depth and clinical stage(P < 0.05). In colorectal carcinoma, FcγRⅡand VEGF-A mRNA expression was significantly negatively correlated (r = -0.104, P = 0.002). Conclusion:The expression of VEGF-A and FcγRⅡwas associated with the clinical features of colorectal cancer. Expression of FcγRII was negatively correlated with VEGF-A. This study indicates that FcγRⅡand VEGF-A expression is related to the tumor invasion and metastasis. FcγRⅡ may become a new cancer immunotherapy target.

    • The expression of MMP-14 in gastric carcinoma and its clinical significance in peritoneal metastasis

      Abstract:Objective:To investigate the expression and clinical significance of MMP-14 in gastric carcinoma and its role in peritoneal metastasis. Methods:The mRNA expression of MMP-14 in peritoneal washing samples were deteced by RT-PCR in gastric carcinoma(98 cases) and normal control(30 cases). Immunohistochemical staining S-P method was used to detect the expression of MMP-14 in gastric carcinoma(98 cases) and normal gastric mucosa(20 cases). Results:The expression rates of MMP-14 were 76.5%(58/98) in gastric cancer tissues and 5%(1/20)in normal gastric mucosa tissues,which were significantly higher than those in the normal tissues respectively (P < 0.01). The MMP-14 mRNA positive rate of peritoneal washing samples in gastric cancer was higher than that in normal control. The positive expression rate of MMP14 was related to the depth of invasion,the size of tumor,lymph node metastasis,macroscopic type and clinial stage (P < 0.05,0.01),but not related to age,gender and differentiation of tumor(P > O.05). Conclusion:The high expression of MMP-14 in gastric carcinoma and peritoneal washing samples suggests that MMP-14 may be involved in the development of gastric cancer and peritoneal metastasis process.

    • Expression and clinical significance of SALL4 gene in cervical cancer and cervical intraepithelial neoplasia

      Abstract:Objective:To investigate the expression pattern and significance of SALL4 gene in CIN and cervical cancer. Methods:The expression of SALL4 mRNA in tissues and plasma of 56 cases of cervical cancer,120 cases of CIN,35 cases of normal cervical or chronic cervicitis(control group) was detected by Real-time PCR. The correlation analysis was conducted to assess the relationship between SALL4 expression in tissues and plasma of cervical cancer group. The levels of SALL4 in plasma were detected by enzyme linked immunosorbent assay. Results:The tissue levels of SALL4 mRNA expression in the the control group,CINⅠ group,CINⅡ~Ⅲ group and cervical cancer group were(4.58 ± 0.64) ×10-4,(6.86 ± 0.72)×10-4,(27.03 ± 2.85)×10-4 and(40.8 ± 5.50)×10-4. The plasma levels of SALL4 mRNA expression were (0.14 ± 0.05)×10-4,(0.16 ± 0.07)×10-4,(0.49 ± 0.08)×10-4 and (2.18 ± 0.22)×10-4. The concentration of SALL4 in plasma were (237 ± 45)-滋g/L,(256 ± 59)-滋g/L,(713 ± 62)-滋g/L and(1 135 ± 107)-滋g/L,respectively. With the progress of cervical disease,the expression of SALL4 was increasing gradually,the expression of SALL4 in cervical cancer group was higher than that in other groups(P < 0.01). A positive correlation between SALL4 expression in tissues and plasma was observed in cervical cancer group(r = 0.684,P < 0.01).Conclusion: SALL4 expression was up-regulated in the CIN and cervical cancer tissues and plasma,and it may be probably associated with the initiation,progression of cervical cancer..

Methods:Tweenty patients undergoing CABG were randomly divided into dopamine group and control group. The hemodynamics changes (HR,MAP,CVP,CI,et al) and the related hepatic vein flow indicators(hepatic vein diameter,hepatic vein flow,et al) were measured by Transesophageal echocardiography. These data were recorded after general induction,5 minutes after cardiopulmonary bypass beginning,30 minutes after cardiopulmonary bypass beginning,and 30 minutes after off-pump,and the liver and kidney changes were compared between preoperation and postoperation. Results:Blood pressure decreased in the two groups after cardiopulmonary bypass,heart rate of dopamine group was higher than that of the control group after offpump(P < 0.05). The liver blood flow of dopamine group during cardiopulmonary bypass increased compared with preoperation(P < 0.05),but these was no significant difference compared with control group. Creatinine and blood urea nitrogen of dopamine group was lower after surgery compared with control group(P < 0.05). Conclusion:Continous infusion of dopamine with 2 μg/kg-min could increase hepatic blood flow,but it needs further reaearches of appropriate dose of dopamine to getting a better clinical effect.

    • Effects of early intervention on physical and neurobehavioral development of premature infants

      Abstract:Objective:To explore effects of early intervention on physical and neurobehavioral development of premature infants during infant period. Methods:One hundred and tweenty-six premature infants were randomly divided into intervention group (n = 63) and control one (n = 63). They were treated with early intervention in intervention group,while a routine physical examination was given to control group. The physique,mood,sleep and intellectual development indexes were observed and recorded regularly,and then compared between two groups. Results:The measured values of weight,height,and head circumference were significantly greater in the intervention group(P < 0.01). At corrected 3 months old,the sleep conditions were significantly better in the intervention group(P < 0.05). The number of crying in intervention group was significantly less during the physical examination process of the corrected 6 months (P < 0.05). The developmental quotients in intervention group were remarkably increased,when they were corrected 6 and 12 months old (P < 0.01). Conclusion:Early intervention is beneficial for the physical development of premature infants,and is helpfur for reduce of sleep problems,negative emotion of crying,and improvement of intelligence development of premature infants.

    • Correlation between renal volume normalized by BSA and renal function in patients with renal tumors

      Abstract:Objective:To assess the correlation between renal volume measured on contrast-enhanced CT images normalized by BSA (body surface area) and renal function in patients with renal tumors. Methods:Thirty-four patients with renal tumors underwent abdominal contrast-enhanced CT and radionuclide renography within one week before surgery. Residual bilateral renal cortical volume (RCV) and renal medullary volume (RMV) on arterial phase images were measured by a semiautomatic software. Renal parenchymal volume (RPV) was the sum of RCV and RMV. Glomerular filtration rate (GFR) measured by radionuclide renography was considered as the reference standard. RPV,RCV,RPV normalized by BSA(RPVBSA=RPV/BSA),and RCV normalized by BSA(RCVBSA=RCV/BSA) were correlated with GFR using the Pearson correlation coefficient,respectively. Results:The correlation between RPV / RCV and GFR presented r = 0.474,P = 0.005 and r = 0.448,P = 0.008,respectively. RPVBSA and RCVBSA had a higher degree of correlation (r = 0.659,P < 0.001 and r = 0.627,P < 0.001) with GFR. Conclusion:RPV and RCV measured on arterial phase images could estimate renal function,while RPVBSA and RCVBSA in patients with renal tumors had a better correlation with renal function before surgery.

    • ROC curve analysis of dual-normal parameters model for imaging diagnostic tests

      Abstract:Objective:To amaylze imaging diagnostic test data using receiver operating characteristic curve (ROC) combined with dual-normal model parameter method. Methods:Continuous data of a single common imaging diagnostic test and ordered segment information of two completely diagnostic tests were selected for analysis. The former group consisted of 49 cases of gastric neoplastic lesions was examined by conventional MRI and diffusion-weighted imaging(DWI) to analyze the difference of apparent diffusion coefficient (ADC) between gastric benign and malignant placeholder and determine the best cut-off point;the latter group consisted of 55 cases of Solitary pulmonary nodules(SPN) with HRCT and PET/CT data. Two types of data were analyzed by ROC analysis software (ROCKIT). The "gold standard" of diagnostic data was pathological results of significance(α = 0.05). Results:Area under the ROC curve(Az) of the former diagnostic tests was 0.963 5;the parameters a was 2.347 6,and the relevant parameter b was 0.844 7;Youden index corresponding to the maximum cut-off point was 1.915. The area under the ROC curve obtained by the latter diagnostic experiments HRCT and PET/CT were 0.812 7 and 0.959 0,respectively. The correlation coefficient was 0.827 9. Z test of the area under the curves of both group got the one-tailed P value of 0.000 1. Conclusion:The dual-normal parameters model combined with ROC curve analysis software ROCKIT could systematically analyze the imaging diagnostic data. ROC evaluation can be achieved in a single diagnostic test for continuous data to determine the optimum operating point and ordered categorical data on the diagnostic performance comparison.

      Abstract:Objective:To evaluate the mechanical properties of longitudinal section of carotid plaque in patients with T2DM,and compare the indexes of velocity,strain and strain rate with normal controls. Methods:One hundred subjects were divided equally into diabetic group and control group according to blood glucose level. Acuson Sequoia C2000(Siemens) was used to collect the image data of carotid plaque. Velocity vector imaging technique was performed to detect the motion information of 5 regions of interest on the surface of carotid plaque,and obtain the positive peak,negative peak and peak difference of velocity,strain and strain rate. Results:All indexes of the diabetic group were smaller than the control group. In the two groups,the indexes of velocity were firstly decreased and then increased. And in the diabetic group,the proximal end of shoulder was greater than the remaining points for strain and strain rate. Conclusion:Velocity vector imaging is an effective technique for evaluating the mechanical properties of carotid plaque in patients with T2DM.

    • Rapid and sensitive detection of EGFR drug response mutation by Junction probe technology

      Abstract:Objective:Epidermal growth factor receptor(EGFR)mutation,particularly E746_A750del and L858R,is thought to be associated with clinical outcomes of patients with non-small cell lung cancer(NSCLC) receiving TKIs treatment,suggesting that EGFR mutation test may play an important role in clinical decision. So,establishment of a rapid,simple and cost-effectiveness EGFR mutation test method can effectively relieve the financial burden of patients. Methods:We established a rapid and sensitive assay for detecting EGFR mutation by asymmetrical PCR and Junction Probe (APCR-JP). In this study,37 NSCLC tissue samples were detected by APCR-JP assay. Results:Seventeen samples (45.9%) carried 19del mutation and 8 samples (21%) had L858R mutation,which are consistent with the results of direct sequencing. Conclusion:These results suggest that APCR-JP could be used in EGFR mutation detection and provide imformation for clinical decision.

      Abstract:Objective:To investigate the long-term effect of bone marrow-derived cells (BMC) therapy on left ventricular systolic function in patients after acute myocardial infarction (AMI) by performing meta-analysis of randomized controlled trials with follow-up ≥2 years. Methods:Specific terms were used to conduct a systematic literature search of MEDLINE,EMBASE,the Cochrane Library and the Cochrane Central Register of Controlled Trials,and the China Biological Medicine Disk database from their inception to July 2012. A standardized form was used to extract information,and RevMan version 5.0 was used to analyze outcome data quantitatively. Results:Six trials comprising 551 patients were included. Compared with controls,BMC therapy significantly improved left ventricular ejection fraction (LVEF) (5.09%,95% CI:2.67% to 7.52%,P < 0.000 1). Subgroup analysis revealed that significant improvement of LVEF induced by BMC therapy could be observed in patients with baseline LVEF≤42%,but disappeared in those with baseline LVEF> 42%. Conclusion:Intracoronary BMC infusion in patients with AMI may further increase LVEF and improve left ventricular systolic function on top of standard therapy,and beneficial effects could last for more than 2 years. The findings need to be validated in the future.

    • Hydrocortisone for treating hypotension in preterm infants:A meta-analysis

      Abstract:Objective:To discuss the efficacy of hydrocortisone for treating hypotension and reduction of vasopressor requirements in preterm infants. Methods:A search in Cochrane Library,PubMed,Ovid,Springer was performed to identify relevant articles according to the inclusion criteria. Standard meta-analytic techniques,including random and fixed effects models,were used to calculate combined effect size correlations and significance levels. Results:Seven studies were included,with total 144 newborns,144 cases in Hydrocortisone group and 93 cases in Hydrocortisone and inotrope group. Random effects meta-analysis showed that hydrocortisone increases blood pressure (r = 0.74,95%CI:0.16 to 0.91) and reduces vasopressor requirement (r = 0.72,95%CI:0.0124 to 0.96). Conclusion:Hydrocortisone is effective in treatment of refractory hypotension and decreasing vasopressor requirements in preterm without an increase in short term adverse consequences. However,the long term safety data on the use of hydrocortisone is unknown.
