    • The association between CD8+ T cell subsets and immunopathology during Schistosoma japonicum infection

      Abstract:Objective:To study the role of CD8+T cell subsets in the development of liver immunopathology after Schistosoma japonicum (S japonicum)infection. Methods:Splenocytes were prepared from spleens of mice with S. japonicum infection at 0 and 8 weeks,and the proportions of Tc1,Tc2 and CD8+T regulatory cells in T cells were determined by FACS. The relationship between CD8+T cell subsets and liver immunopathology were analyzed. Results:Compared to the control group,the spleen mononuclear cells (MNC)in S. japonicum infected mice were increased significantly (P < 0.001). In addition,the proportions and the absolute numbers of T cells and CD8+T cells in spleen of S. japonicum infected mice were significantly increased(all P < 0.05). Furthermore,the proportions of Tc1,Tc2 and CD8+T regulatory cells were significantly increased at 8 weeks post-infection(all P values < 0.05)and the proportions of Tc2 cells increased more significantly than Tc1 cells. Multiple linear regression analysis showed that Tc2 and regulatory CD8+T cells were the main factors affecting liver immunopathology during S. japonicum infection. Conclusion:Tc1,Tc2 and CD8+ T regulatory cells in splenocytes increased significantly after S. japonicum infection. Tc2 and regulatory CD8+ T cells play critical roles in promoting and inhibiting the liver immunopathology,respectively.

      Abstract:Objective: To explore the neuroprotective mechanism of ischemic preconditioning in spinal core ischemia. Methods:Fifty adults rats were randomly assigned to 4 groups: the sham group receiving anesthesia and surgical preparation(group A, n=5),the ischemia/reperfusion (I/R)group adminstrated to 0.5 h ischemia followed by 3,6,12,or 24 h reperfusion (group B, n=20),the IPC group receiving three cycles of 5 min ischemia followed by 5 min of reperfusion (group C, n=5),and the IPC+Ⅰ/R group (group D, n=20). HE and immunohistochemical staining were used to evaluate the spinal neuron. Autophagic activity was investigated by electron microscopy and immunohistochemistry. The autophagosome marker LC3-Ⅱ and the autophagy-associated protein Beclin-1 were detected by Western blot. Results: In the Ⅰ/R group,LC3-Ⅱ was significantly elevated after 3 h of reperfusion,but declined significantly by 24 h. At 24 h,rats in the I/R group exhibited extensive spinal damage and decreased neuronal survival. In the IPC+Ⅰ/R group, neuronal death was reduced and expression of LC3-Ⅱ sustained throughout the 24 h reperfusion period. Conclusion:Ischemic preconditioning can sustain the beneficial effects of autophagic lysosomal degradation during Ⅰ/R by inhibiting autophagic cell death.

    • Effects of PEP-1-SOD1 combined with neural stem cells transplantation on the neurologic function of rats with traumatic brain injury

      Abstract:Objective:To investigate the effects of neural stem cell (NSC)transplantation combined with PEP-1-SOD1 on recovery of neurologic function of rats with traumatic brain injury. Methods:A total of 80 healthy SD rats were randomly divided into four groups including the control group (with injection of physiological saline into injury region),the NSCs transplantation group (suspension of NSCs),the SOD1 group (PEP-1-SOD1 protein),and the NSCs transplantation + SOD1 group (suspension of NSCs + PEP-1-SOD1 protein). AQP4 mRNA and AQP4 expressions were determined by quantitative polymerase chain reaction (QPCR)and Western blot assay at the 4th day after traumatic brain injury. Bederson neurobehavioral assessment was performed 24 hours,3 days,1,2,3 and 4 weeks after traumatic brain injury. Morris water maze test was performed to determine neurologic function of rats from day 23 to 28 after traumatic brain injury. Immunohistochemistry and pathology changes were determined at 4 weeks after injury. Results:The expressions of AQP4 mRNA and AQP4 in the control group were higher than those in the NSCs transplantation group and SOD1 group,and the expressions in the NSCs transplantation group and SOD1 group were higher than those in the NSCs transplantation+SOD1 group(P < 0.05). Bederson scores at 4 weeks after traumatic brain injury were lower than those at 24 hours in all groups. At 4 weeks after traumatic brain injury,the scores in the NSCs transplantation+SOD1 group were lower than those in the control group(P < 0.05). Morris water maze test showed that neurologic function in the NSCs+SOD1 group was significantly more improved than that in the control group (P < 0.05). Immunohistochemical staining showed that the number of BrdU positive cells in the NSCs transplantation+SOD1 group was higher than that in the control group,NSCs transplantation group,and SOD1 group(P < 0.05). Conclusion:PEP-1-SOD1 combined with transplantation of neural stem cells can significantly improve the neurological function in rats with traumatic brain injury.

      Abstract:Objective:To study whether parathyroid hormone regulates the differentiation of bone mesenchymal stem cells into osteoblasts through Notch signaling pathway. Methods:Six PTH+/+ and six PTH-/- mice of 8-week-old were used in this study. Bone mesenchymal stem cells obtained from mouse femurs were isolated and cultured,then osteogenic differentiation was induced. Western blot was performed to detect the expression of Notch signaling pathway at day 3,7,10,14. The alkaline phosphate (ALP)activity was detected to evaluate the early osteogentic differentiation at day 3 and 7. Western blot was performed to detect the expression of Runx2 and OCN proteins. Results:The receptor and ligand of Notch signal pathway and the osteogenic index was increased in the wild type group and PTH gene knockout group after osteogenic differentiation. The expression of Jagged1 and Notch1 protein in PTH gene knockout group was significantly lower than that of the wild type group (P < 0.01). Compared with the wild type group,Runx2 and OCN protein levels were lower in PTH gene knockout group,with the same trends as ALP results. Conclusion:The deficiency of endogenous PTH inhibited the differentiation of bone marrow mesenchymal stem cells into osteoblasts,by reducing the expression of Notch1 and Jagged1 proteins.

    • Application of bioluminescence imaging on early intrapulmonary micrometastases tumors in mouse model

      Abstract:Objective:To investigate the ability of bioluminescence imaging (BLI)used for early detection of lung tumors and quantitative measurement of tumor size. Methods:The luciferase activity of SPC-A1-luc cell was detected in vitro by BLI. SPC-A1-luc cells were subcutaneously injected into nude mice to establish mice subcutaneous tumor model,and the subcutaneous tumor was imaged every week by BLI and measured by caliper. SPC-A1-luc cells were intravenously injected into nude mice to establish intrapulmonary micrometastases model,BLI and micro-CT were used to monitor tumor growth and quantitatively analyze tumor size. Results:SPC-A1-luc cells stably and highly expressed luciferase,and the luciferase activity was significantly correlated with cell numbers(R2=0.9966)and tumor volume(R2=0.9849). Bioluminescence signals were diffused distribution in bilateral lung at the first week. On the third week,quantitative analysis showed that bioluminescence signals were the lowest and approximately 600 tumor cells were growing in the lungs. However,intrapulmonary micrometastases could not be detected by micro-CT until the 6th week, it was three weeks later than BLI. Conclusion:Intrapulmonary micrometastases could be early detected and quantitatively analyzed by BLI,which has higher sensitivity than micro-CT.

      Abstract:Objective: To explore the effects of over-expression of GPC5 on the biological function of human lung adenocarcinoma A549 cancer-initiating cells(CICs). Methods: Lentivims-induced GPC5 over-expression was used for biological functional analyses in human lung adenocarcinoma A549 cell line, including cancer-initiating cell microspherescounting by light microscopy, CD133 positive cells measured by flow cytometry, migration and invasive activity detected by Matrigel and Transwell assay, as well as epithehal-mesenchymal transition-related protein expression detected by Western blot. Results: Compared with the control group, the numbers of cancer-initiating cell microspheres remarkly decreased(P < 0.05) and the percent of CDl33 positive cells was reduced (P < 0.05) in the GPC5 over-expressed group. The over-expression of GPC5 also inhibited the invasion and migration ability of CICs, and led to the down-regulation of Vimentin and N-cadherin, as well as the up-regulation of E-cadherin. Conclusion: The over-expression of GPC5 can inhibit the genesis, invasion and migration of human lung adenocarcinoma A549-CICs, and regulate the expression of EMT related proteins.

      Abstract:Objective:To study the role and significance of phosphatase and tensin homolog deleted on chromosome ten(PTEN)protein for the occurrence and development of endometriosis (EMs)in vitro experiments. Methods:Lentiviral vector was used to overexpress PTEN and silence PTEN expression in human primary endometrial cells; flow cytometry was used to detect the changes in cell cycle and apoptosis in the different PTEN expression groups after lentivirus infection. Results:Cell cycle analysis showed that after transfection of overexpressed lentivirus of PTEN gene,the proportion of G0/G1 phase in human primary endometrial cells was increased,compared with the control group(P <0.05). The proportion of G2/M phase was gradually decreased,and cell cycle was arrested in G0/G1 phase; In the blank group,the overexpressed PTEN group,and the siPTEN group,cell apoptosis rates were(11.70 ± 0.03)%,(15.80 ± 0.14)%,and(5.33 ± 0.08)%,respectively. Conclusion:Overexpression of PTEN could significantly increase apoptosis rate and inhibit cell cycle of human primary endometrial cells. PTEN has a certain inhibiting effect on the occurrence and development of endometriosis.

    • Expression of YKL-40 in small cell lung cancer and its clinical significance

      Abstract:Objective:To study the relationship between the expression of YKL-40 and clinicopathologic characteristics and prognosis in small cell lung cancer (SCLC). Methods:The expression of YKL-40 in 105 samples of SCLC tissues and 40 samples of para-cancerous tissues were examined by immunohistochemistry. Results:The expressions of YKL-40 in cancer tissues and para-cancerous tissues were 72.4% and 22.5%,respectively(both P < 0.01). The expression of YKL-40 was not associated with gender,age and smoking,but associated with clinical stage and lymph node metastasis (both P < 0.05). Median overall survival (OS)of SCLC patients was 16.3 months. Median OS in patients with YKL-40 expression was 12.4 months,which was shorter than that in patients without YKL-40 expression(25.6 months),and there was statistically differences (P < 0.05). Univariate analysis showed that clinical stage and the expression of YKL-40 were related to the prognosis of SCLC patients,while multivariate analysis showed that YKL-40 was an independent prognosis factor of SCLC patients. Conclusion:YKL-40 may be a molecular marker to predict the relapse,metastasis and prognosis of SCLC patients.

    • Expression profile of circular RNA in colorectal cancer

      Abstract:Objective:To analyze the expression profile variation of circular RNA(circRNA) between normal colorectal tissues and colorectal cancer(CRC) tissues. Methods: Human circRNA microarray was performed to detect the difference of circRNA expression between 6 cases of CRC tissues and 6 cases of normal colorectal tissues. After pretreatment and homogenization of raw data,the circRNAs with differental expression were screened out and then analyzed by hierarchical clustering and scatter volcano diagram. Results: Compared with normal colorectal tissues, there were 892 circRNAs differentially expressed which have more than 2 times variation and significant difference(P < 0.05). Among them, 412 increased and change′s fold more than 2 times,while 480 reduced and change′s fold more than 2 times. Conclusion: The circRNA expression profile of CRC changed significantly in comparison with the normal colorectal tissues, the different circRNAs may be associated with the development of CRC.

    • Resveratrol promotes the browning of white adipocytes via suppressing miR-27b expression

      Abstract:Objective:To study the role of resveratrol in regulating the browning of mouse primary white adipocytes and find out the related molecular mechanisms. Methods:Primary subcutaneous preadipocytes were isolated from the groin of C57BL/5J mice and induced for differentiation. The cells were treated with different concentrations(0-50 -滋mol/L) of resveratrol to analyze the resveratrol′s effect on the browning of white adipocytes. Real-time PCR was performed to detect the mRNA expression of BAT-specific genes and miR-27. mimic-miR-27b was transfected into primary cells to increase miR-27b expression,then,resveratrol (50 μmol/L)was performed to stimulate these cells. We used real-time PCR and Western blot to measure mRNA and protein levels of BAT-specific genes in these cells. Results:Resveratrol promoted the expression of BAT-specific genes and suppressed miR-27b expression in a concentration-dependent manner in mouse white adipocytes. Overexpression of miR-27b significantly inhibited the resveratrol action on the mRNA expression of BAT-specific genes. At the protein level,overexpression of miR-27b also suppressed the resveratrol action on the PRDM16 and UCP1 protein levels in mouse white adipocytes. Conclusion:Our data showed that resveratrol promoted the browning of mouse white adipocytes via suppressing miR-27b expression. MiR-27b might provide a potential therapeutic target for the treatment of obesity and metabolic syndrome.

    • Role of curcumin in suppressing the inflammation after the myocardial infarction in rats and its underlying mechanisms

      Abstract:Objective: To investigate the inhibitory effects of curcumin on the inflammation after myocardial infarction (MI) in rats and its underlying mechanisms. Methods: The Wistar rat MI models were established through the ligation of the left anterior descending coronary artery. Infiltration of inflammatory cells and the expression of related cytokines at different time points after MI were detected,and then we tried to uncover the underlying molecular mechanisms suppressing the inflammation by curcumin with Western blot. Results: Compared with the control group, the use of curcumin significantly improved cardiac function in 28 d after MI, and reduced the inflammatory cells infiltration of infarction area after MI 1 d, 7 d, 14 d and 28 d; further studies suggested that curcumin decreased inflammatory cytokines such as TNF-α, IL-1β and IL-6 in rats after MI at different times, and suppressed the activation of NF-κB and JAK-1/SATA3 signaling pathways. Conclusion: Curcumin can inhibit inflammation after MI by signaling pathways to reduce infiltration of inflammatory cells in infarct area and reduce the secretion of inflammatory cytokines, thus improve hemodynamics, reduce ventricular remodeling and maintain cardiac function.

    • Association between interleukin-6 promotor-634C/G polymorphism and coronary artery disease

      Abstract:Objective: To explore the association of-634C/G polymorphism of interleukin-6 (IL-6) gene with coronary artery disease(CAD). Methods: A total of 86 CAD patients and 91 healthy controls were selected for this study. The PCR-based restriction fragment length polymorphism (PCR-RFLP) technique was performed to assess the genotypes frequencies. The concentration of serum IL-6 was measured with ELISA. The relations of the levels of serum IL-6 between groups were analyzed. Results: The circulating IL-6 levels in the CAD group[(26.57 ± 6.68) ng/L] were significantly higher than those in the control group[(6.89 ± 2.32) ng/L,P < 0.05]. The distribution of the IL-6 -634C/G genotypes (CC,CG, and GG) was 65.9%,3 1.9% and 2.2% in the controls,and 57%, 29% and 14% in the CAD group, respectively(P < 0.05). Compared with the wild type CC,the G allele carriers(CG+GG genotypes) had higher circulating IL-6 levels[(34.575 ± 6.618)ng/L vs.(16.758 ± 5.182)ng/L, P < 0.05] in the CAD group. Conclusion:These findings suggest that IL-6 -634C/G polymorphism is associated with CAD and the G allele carriers have increased circulating IL-6 levels.

    • Gene sequence determination and analysis of Hantavirus isolated strains in Jiangsu province from 2009 to 2013

      Abstract:Objective:The gene sequence of Hantavirus isolated strains during 2009 and 2013 was analyzed at molecular level to provide evidence for the prevention and control of this disease. Methods:The serum of patients with acute hemorrhagic fever with renal syndrome(HFRS)and rat lung specimens were collected from 2009 to 2013. Hantavirus IgM and IgG antibody in serum samples and antigen in mouse lung samples were detected using colloidal gold and IFA assay,respectively. With VERO E6 cell line,Hantavirus was isolated,then analyzed using RT-PCR assays,and genome was sequenced using high-throughput sequencing technology. Finally,sequence alignment and phylogenetic analysis were accomplished with MEGA6.0 software. Results:With the closest genetic distance,isolated 11 virus isolates and Shanxi LR1,Sichuan isolates s85-46 and Hantavirus strain 76-118 prototype were all classified as Hantaan (HTN)virus. In addition,strains isolated in Jiangsu 2002 (A9,HTN type)and strains in Zhejiang and Anhui on different evolutionary branches,indicated a larger genetic distance. Conclusion:Both Hantavirus predominant strains switchover and host transfer have happened in Jiangsu. Surveillance should be strengthened to guarantee the efficacy of the vaccine.

      Abstract:Objective: Atrial fibrillation (AF) is the most common cardiac arrhythmia in clinical practice. Unfortunately, the sensitive serum biomarkers of atrial remodeling in AF remain unclear. The aim of this study was to determine sensitive serum markers for estimating atrial structural remodeling of AF. Methods: Right and left atrial specimens were obtained from 90 patients undergoing valve replacement surgery. The patients were divided into the sinus rhythm(n = 30),the paroxysmal atrial fibrillation(n = 30),and the persistent atrial fibrillation (n = 30) groups. CIB1, calcineurinA, calcineurinB,nuclear factor of activated T3(NF-AT3),nuclear factor of activated T4(NF-AT4), collagen Ⅰ and Ⅲ protein expression in atria were measured. We also tested the levels of the carboxyl-terminal peptide of pro-collagen Ⅰ(PICP),the N-terminal typeⅠprocollagen propeptides, the N-terminal type Ⅲ procollagen propeptides, and TGF-β1 in serum using an enzyme immunosorbent assay. Results: Key protein expression in calcineurin system was increased in the AF groups,especially in the left atrium. In patients with AF, the expression levels of nuclear NF-AT3 and NF-AT4 were correlated with those of collagens Ⅰ and Ⅲ in the atria and with PICP and TGF-β1 in blood. Conclusion:These data support the hypothesis that PICP and TGF-β1 levels may be sensitive serum biomarkers to estimate atrial structural remodeling with at AF.

    • A nested case-control study on risk factors of hypertension in Changzhou rural areas

      Abstract:Objective:We sought to use large sample size cohort to investigate the related risk factors of hypertension in rural communities of Changzhou,the results would help take intervention measures for onset hypertension. Methods: In this nested case-control study,3 400 new cases with hypertension were included,and 3 400 non-hypertension controls were randomly matched by sex and age. The related risk factors were detected by single and multi-variate Logistic regression. Results: Single-variate Logistic regression showed that drink, body mass index (BMI), total cholesterol (TC) and triglyceride(TG) were associated with hypertension; further multi-variate logistic regression showed that drink,overweight,obesity, TC and TG increased the risk of hypertension and low weight reduced the risk. Conclusion: Hypertension was related to drink,overweight, obesity and lipoprotein level in rural communities of Changzhou. Taking integrated intervention measures of high risk groups is an important way to reduce the risk of hypertension.

    • Efficacy and safety of modafinil on residual sleepiness in patients with obstructive sleep apnea syndrome treated by continuous positive airway pressure

      Abstract:Objective: To evaluate the efficacy and safty of modafinil for residual sleepiness(RS) in patients with obstructive sleep apnea syndrome (OSAS) treated by continuous positive airway pressure (CPAP). Methods: Thirty-two male patients with mild-to-moderate OSAS under CPAP treatment were selected. All of them had Epworth seepiness scale(ESS)>10. The study included three stages (T1,T2 and T3). Following baseline evaluation (T1),patients were randomly allocated to the modafinil or placebo group. The second stage(T2): all subjects were instructed to take a piece of modafinil or placebo (100 mg) for 7 days. The third stage (T3): patients were instructed to take two pieces of modafinil or placebo(200 mg) for 28 days. Patients underwent a final evaluation on the last day of the third stage. The evaluations at T1 and T3 consisted of routine blood, blood biochemical tests, routine urine, electrocardiogram, nocturnal polysomnography (PSG) and multiple sleep latency test (MSLT). In addition, ESS was constantly detected during T1, T2 and T3 . Results:Before and after treatment, there were no significant changes both in Apnea Hyponea Index (AHI) and the per-night use time of CPAP between the modafinil and placebo groups. After treatment, comparing with the placebo group,the mean sleep leatency was lengthened and ESS score was decreased in the modafinil group,and both differences had statistical significance (P < 0.05). In addition,in the modafinil group,changes of ESS was significantly greater in T3 with 200 mg than that in T2 with 100 mg (F=0.719,P=0.015). Adverse drug reactions occurred in 62.5% patients in the modafinil group, such as dry mouth, nausea,headache and so on. there was no significant difference between the reactions (P=0.41),and all of these adverse drug reactions were mild to moderate in severity. Conclusion: Modafinil was effective and well tolerated for improving RS in OSAS patients under CPAP treatment.

Methods: A total of 30 patients of newly diagnosed type 2 diabetes were selected to examine the oral glucose tolerance test (OGTT), insulin, C peptide, haemoglobin A1C (HbA1c), weight, waist circumference, blood pressure and miR-375 expression level changes in serum before and after 12 weeks of liraglutide treatment. Data analysis was performed using paired t test and Wilcoxon signed rank test. Results: MiR-375 expression and HbA1c were decreased after liraglutide treatment (P < 0.05). Areas under the curve of blood glucose 0-30 min and 0-120 min were significant increased (P < 0.05). Conclusion: After liraglutide treatment, insulin resistance and functional changes in pancreatic islet β cells of diabetics were improved, it may be related to the decrease of miR-375 expression level in serum.

      Abstract:Objective:To explore the expression level of miR-203 in the peripheral blood mononuclear cells (PBMCs) of patients with systemic lupus erythematosus (SLE), and to analyze the relationship between the expression level and clinical manifestations. Methods: The PBMCs of 31 SLE patients, 16 healthy people were collected,and then SLE patients were divided into two groups, including the lupus nephritis group (21 cases) and the no kidney involvement group (10 cases). The expression level of miR-203 in PBMCs was detected by real-time fluorescence quota polymerase chain reaction (qRT-PCR). The expression levels of miR-203 in each group were compared to analyze the relationship between the expression level and clinical manifestations. Results: The expression level of miR-203 was significantly lower in the SLE group compared to the control group (P < 0.05). The expression level of miR-203 was significantly lower in the lupus nephritis group than that in the no kidney involvement group and the control group (both P < 0.01). The expression level of miR-203 was negative correlated to 24 hours urine protein, acute kidney disease index (P < 0.01). Conclusion: The expression of miR-203 was decreased in SLE patients, indicating that miR-203 may play a role in the pathogenesis of SLE. And miR-203 may be associated with the occurrence of lupus nephritis.

    • Clinical distribution and analysis of antimicrobial resistance among Acinetobacter baumannii isolated from 2012 to 2014 in Xuzhou

      Abstract:Objective:To investigate the specimen source,clinical distribution and antimicrobial resistance of Acinetobacter baumannii isolated from Affiliated Hospital of Xuzhou Medical College from 2012 to 2014. Methods:Acinetobacter baumannii isolates were identified by BD Phoenix 100 automatic microbial identification system (USA). The susceptibility testing was performed by Kirby-Bauer diffusion method,and the antimicrobial susceptibility data were analyzed by WHONET 5.6 software. Results:The isolated Acinetobacter baumannii of 2012-2014 strains were 1213,1492 and 1712,respectively,accounting for 53.5%,44.4% and 39.9% of the non-fermentative gram negative bacilli,respectively. The Acinetobacter baumannii strains were mainly isolated from sputum specimens (89.2%),and the Acinetobacter baumannii infection mainly distributed in emergency intensive care unit (ICU)(37.5%). Except for levofloxacin,amikacin,polymyxin and sulfamethoxazole compound,the resistance of Acinetobacter baumannii to cephalosporins,carbapenem,quinolones and aminoglycoside reached more than 80.0% in 3 years. The resistance rate of imipenem increased from 83.1% in 2012 to 87.1% in 2013,subsequently,the rate decreased to 82.8% by 2014. Likewise,the resistance rates of meropenem were 84.9%,87.7% and 85.1% from 2012 to 2014,respectively. Nevertheless,the resistance rate of polymyxin was the lowest,ranging from 1.5% to 2.9%. Conclusion:Antimicrobial resistance rate of Acinetobacter baumannii isolated from 2013 exhibited an apparent increasing compared to that from 2012,and then decreased in 2014,but still very serious,which should be paid more attention to. Therefore,monitoring of the resistant of Acinetobacter baumannii and the management of clinical rational use of antimicrobial drugs should be strengthened.

    • Effects of rapid plasma regain titer of syphilis during pregnancy on perinatal outcome and serologic outcome of the newborn

      Abstract:Objective:To analyze the relationship between rapid plasma regain (RPR) titer of syphilis and adverse pregnancy outcomes in pregnant women and RPR positive rate of newborns. Methods:From Jan 1,2009 to Fab 1,2014,192 pregnant women with syphilis were screened by RPR test and treponema pallidum particle agglutination (TPPA) test before delivery and some of the newborn were screened for peripheral blood at birth and 6 months,so that to find the relationship between RPR titer of syphilis in pregnant women and RPR positive rate and negative conversion rate of the newborns,as well as the adverse pregnant outcomes in pregnant women with syphilis. Results:RPR titer of newborns was closely relative to their mothers. When RPR titer of pregnant women was 1∶1 and 1∶2,PRP positive rate of the newborns was 68.4% and 87.0%,respectively. When RPR titer of pregnant women was higher than 1∶8,PRP positive rate of the newborns reached 100% and higher than those with mother′s RPR titer was ≤1∶4 (P < 0.05). When RPR titer of pregnant women was ≥1∶8,the RPR negative conversion rate of the 6-month newborns was decreased,compared that with mother's RPR titer was ≤1∶4(P < 0.05). With the rise of RPR titer of pregnant woman,risk of malformation,stillbirth,preterm birth,congenital syphilis,neonatal death and adverse newborn outcome increased. Preterm birth,congenital syphilis and neonatal malformation rates were increased when RPR titer of pregnant women was ≥1∶8,compared that when RPR titer of pregnant women was ≤1∶4(P < 0.05). Conclusion:With the increase of RPR titer syphilis in pregnant women,PRP positive rate of the newborns was also increased. In addition,when RPR titer of pregnant women was ≥1∶8,PRP positive rate of the newborn reached 100%,the RPR negative conversion rate of the 6-month newborns was decreased, and occurrence rates of malformation,preterm birth and congenital syphilis were significantly increased.

      Abstract:Objective:To study stress distribution of All-on-Four distal implants with different tilt angles and lengths on edentulous maxilla using the three-dimensional finite element method. Methods:A female patient with edentulous maxilla was selected. After data collecting and three-dimensional reconstruction,a three-dimensional finite element model of edentulous maxilla with All-on-Four implant restoration was established. According to the tilt angles and lengths of the distal implants,there were four groups:Group A:30°,11.5 mm; Group B:30°,13 mm; Group C:45°,11.5 mm; Group:D 45°,13 mm. 100 N vertical static force was loaded on the first premolar,the second premolar and the first molar to simulate chewing, respectively Equivalent stress values of implants on the edentulous maxilla model were calculated using ABAQUS software. Results:Under the vertical static load,stress concentration was found around the implant neck in the cortical bone area. With the increase of the distal implant length,the stress decreased slowly in all regions. With the tilt angle of distal implant increasing,the distribution of stress became more dispersed and the stress in implant neck and apical areas significantly reduced. Conclusion:The stress distribution and magnitude of distal implant did not change significantly with the increase of the implant length,while the implant tilt angle of 45° was more favorable for stress dispersion than the implant tilt angle of 30°.

      Abstract:Objective: To investigate the effect of computer aided design/compnter aided manufacturing (CAD/CAM) enamic blocs with different thickness on the fracture load. Methods: Fifty test specimens were fabricated with isolated tooth in the study. These specimens were divided randomly into two groups:the experimental group (Vita enamic)and the control group (Vitablocs MarkⅡ). In each group,these specimens were divided randomly into five differential thickness sub groups (1.0 mm,1.5 mm,2.0 mm,2.5 mm and 3.0 mm). Every specimen was cemented on the differential thickness bloc with panavia f resin adhesive system. They were stored in distilled water at 37℃ for 24 hours prior to fracture testing. Then, the fracture load was tested in a universal testing machine. Results: Accompany with the increase of thickness of bloc,the increase of fracture load was detected in both groups. When the bloc thickness lied from 1 mm to 2 mm,the fracture load in both groups was similar. But from 2.5 to 3 mm,the fracture load of Vita Enamic was higher than that of Vitablocs MarkⅡ . Conclusion: The fracture load between the Vita enamic and Vitablocs MarkⅡ was similar. Only when the thickness is over 2.5 mm,the mechanical property of Vita enamic was super to that of Vitablocs MarkⅡ.

    • Diagnostic accuracy of ABVS versus HHUS for benign and malignant lesions in breasts:a meta-analysis

      Abstract:Objective:To assess the diagnostic value of automated breast volume scanning (ABVS) versus hand-held ultrasound(HHUS) in benign and malignant breast tumor by meta-analysis. Methods: The Cochrane Library,Web of Science,the PubMed,EMBASE,VIP,CBM,CNKI and Wanfang databases (From date of creating database to 2015/03/07) were searched for studies on the diagnosis of breast cancer with ABVS and HHUS according to inclusion criteria,and then STATA12.0 software was performed to analyze the selected documents. Results: A total of 17 studies were included after screening. The pooled diagnostic odds ratio, specificity and sensitivity of ABVS and HHUS were 95.21 (53.85-168.36)and 28.88 (16.15-51.66), 0.90 (0.86-0.93) and 0.84(0.78-0.89), 0.91(0.88-0.93)and 0.84(0.79-0.88), respectively. Conclusion:The results of meta-analysis suggest that ABVS has higher value than HHUS in the diagnosis of benign and malignant breast tumor.

    • Measurement of 25-hydroxyvitamin D in preterm infants and its related factors

      Abstract:Objective:To measure the content of 25-hydroxyvitamin D in preterm infants, and to investigate factors contributing to it. Methods: A total of 100 infants that were born in Maternal and Child Health Hospital and the 3rd People's Hospital of Wuxi from 05/2012 to 09/2013 were selected as subjects, 50 of which were preterm infants and the other 50 were full-term. Basic situations of birth were recorded and subjects were divided into groups according to different purposes. 25-hydroxyvitamin D in preterm and full-term infants and their corresponding mothers were measured by enzyme-linked immunosorbent assay;calcium, phosphorus, alkaline phosphatase (ALP) were measured using conventional biochemical method;bone mineral density (BMD) values were measured by quantitative ultrasound (QUS). Results: ① 25-hydroxyvitamin D in preterm infants was significantly lower than that detected in full-term infants (P < 0.01). ② 25-hydroxyvitamin D of the preterm group A (28 w≤gestational age≤32 w) was significantly lower than those of the group B (32 w≤gestational age≤37 w), group C (37 w≤gestational age≤39 w) and group (39 w≤gestational age≤42 w) (both P < 0.01). Infants were also divided into 3 groups according to neonatal body weight. All pairwise comparision showed significant difference (both P < 0.01). ③Preterm infants were divided into 3 groups: the regular implement, sporadic implement, and no implement of 25-hydroxyvitamin D groups. All pairwise comparision of the three groups showed significant difference (P < 0.05);④25-hydroxyvitamin D level of preterm infants was positively correlated with gestational age and neonatal body weight (P < 0.05). However, in full-term infants, none of these factors showed significant correlation (P > 0.05). ⑤There was no significant difference in serum calcium, phosphorus, and ALP between preterm and full-term infants (P > 0.05). In the preterm group, 25-hydroxyvitamin D was negatively correlated with ALP (P < 0.01). ⑥BMD difference between preterm infants and full-term ones was significant (P < 0.01). BMD of preterm infants was positively linked to gestational age, neonatal body weight and 25-hydroxyvitamin D level (P < 0.05), and was negatively linked to alkaline phosphatase(P < 0.05). In full-term infants, BMD was significantly correlated with gestational age and 25-hydroxyvitamin D level(P < 0.05). Conclusion: ① 25-hydroxyvitamin D is significantly lower in preterm infants than that in full-term ones; ②Gestational age, birth weight, and vitamin D or calcium supplementation of the mother have great influence on 25-hydroxyvitamin D level of neonatal preterm infants.

    • Application value of 99Tcm-DTPA renal dynamic imaging for diagnosis and treatment duration of renal mass

      Abstract:Objective:To investigate the application value of 99Tcm-DTPA renal dynamic imaging in the diagnosis and therapy of renal single mass. Methods:A total of 61 patients with renal mass were retrospectively collected from January 2010 to January 2015. All patients accepted 99Tcm-DTPA renal dynamic imaging and blood biochemical test while CT enhanced imaging and ultrasound examination were performed in most cases. Renal perfusion index (PI)and glomerular filtration rate (GFR)was evaluated by 99Tcm-DTPA renal dynamic imaging. 99Tcm-DTPA renal dynamic imaging was performed in 32 cases with renal malignant mass and 2 cases with benign tumor about 6 months later after operation. Results:The diagnostic sensitivity and accuracy of 99Tcm-DTPA renal dynamic imaging for renal malignant mass was 37.5% and 63.9%,respectively,lower than that of CT enhanced imaging,while the diagnostic sensitivity,specificity and accuracy was 66.7%,85.7% and 75.0%,respectively,when the long diameter of the renal mass was more than 4 cm. If diagnostic criteria were designed as PI ratio of the renal mass to normal contralateral kidney(T1/K1)was no more than 1,and GFR ratio of the renal mass of functional phase to contralateral(or adjacent)symmetrical kidney tissues(T2/K2)was almost 0,the diagnostic sensitivity increased to 83.3%. Nephron-sparing surgery was preferred when moderate-to-serious injury of the contralateral kidney was seen on 99Tcm-DTPA renal dynamic imaging. Six months after operation,the renal PI was decreased and GFR was increased,otherwise,the renal function inadequacy was found,some patients needed dialysis treatment. Conclusion:99Tcm-DTPA renal dynamic imaging,evaluating PI,GFR of the kidney and the renal mass,is useful for the diagnosis,operation guiding and prognosis prediction.

    • Imaging of bone metastasis from lung cancer with 99Tcm-3P4-RGD2

      Abstract:Objective:To investigate the value of imaging with 99Tcm-HYNIC-PEG4-E[PEG4-c(RGDfk)]2(99Tcm-3P4-RGD2) as radiotracer in the dectection of lung cancer bone metastases. Methods: A total of 45 patients (25 males and 20 females) with lung cancer were retrospectively studied in our hospital from July 2012 to September 2014. All patients received 99Tcm-3P4-RGD2 imaging (RGD imaging,including plane and tomography) and 99Tcm-MDP imaging(MDP imaging,including plane and tomography),meanwhile undergoing SPECT-CT fusion imaging. The final diagnosis was made based on history, pathology,and 6 months follow-up. RGD imaging in the diagnosis of lung cancer with bone metastasis lesions was investigated. Results:Within 122 bone metastasic lesions, 102 was osteolytic, 20 was osteoblastic metastases (5 were demonstrated no bone density changes)and there were 51 benign bone lesions. Regional accumulation of 99Tcm-3P4-RGD2 (T/NT:6.90 ± 2.31) was higher than that of 99Tcm-MDP (1.18 ± 1.24,P=0.010). The diagnotic sensitivity of RGD imaging to osteolytic metastases was 84.8%, higher than that of MDP imaging(25.0%) while the diagnostic sensitivity of RGD imaging to osteoblastic metastases was 55.9%, lower than that of MDP imaging(91.2%)(both P < 0.05). When combined with 6 slice-CT imaging, the diagnostic specificity difference of bone metastases between RGD fusion imaging and MDP fusion imaging was insignificant. The diagnostic sensitivity and accuracy of MDP fusion imaging(92.6% and 93.4%) were higher than those of RGD fusion imaging(78.7%,P=0.008;80.3%,P=0.010). Furthermore, RGD imaging also detected primary lung cancer and lymph node metastases. Conclusion: Specifically targeting integrin αvβ3, which was highly expressed on osteoclast and lung cancer cells,99Tcm-3P4-RGD2 imaging has great potential for detection of lung cancer osteolytic metastasis.
