    • The progress of UHRF1 in epigenetics regulation as well as in tumorogenesis and theranostics

      Abstract:Epigenetics is a discipline focusing on maintenance of phenotype through successive mitosis and the mechanisms mainly include DNA methylation and histone modification.Ubiquitin-like with PHD and ring finger domains 1(UHRF1)is a unique molecule,which is a main “hub protein” involved in epigenetic information integration as well as links DNA methylation and histone modification to ensure faithful epigenetic memory inheritance. Recent researches suggest that UHRF1 overexpression is abundant in many kinds of cancers,and it is involved in cancer occurrence and progression. This paper reviews the mechanism of epigenetics as well as the progress of UHRF1 in tumor diagnosis and treatment.

    • Clinical value of serum miR-148a as a potential biomarker for hepatocellular carcinoma

      Abstract:Objective:To explore the clinical value of serum miR-148a as a potential biomarker for hepatocellular carcinoma(HCC). Methods:Quantitative real-time polymerase chain reaction (qRT-PCR)was used to inrestigate the levels of serum miR-148a in HCC patients,the patients with benign liver diseases and healthy volunteers were used as control groups. Results:Serum levels of miR-148a were significantly decreased in HCC cases than those in benign and healthy controls,P < 0.001. Receiver operating characteristic curve(ROC)showed that the area under the curve(AUC)in HCC group vs. benign group was 0.666,95% Confidence interval(CI),0.581-0.744;the diagnostic sensitivity was 67.7% and specificity was 59.2%,P < 0.001. The AUC in HCC group vs. healthy group was 0.746,95%CI,0.663~0.818;the diagnostic sensitivity was 43.6% and specificity was 96.1%,P < 0.001. In addition,serum miR-148a was significantly associated with tumor size (P=0.011) and TNM stage (P < 0.001),whereas,no significant correlation of serum miR-148a with other clinical parameters,P > 0.05. The level of serum miR-148a in postoperative HCC patients was significantly higher than that of preoperation,P=0.023. However,the level of serum miR-148a in postoperative patients with recurrence or metastasis was significantly decreased,and even lower than that of preoperative group,P < 0.05. Kaplan-Meier analysis demonstrated that the decreased serum miR-148a was closely correlated with shorten overall survival of HCC patients,P < 0.001. Besides,Cox regression model further indicated that serum miR-148a was an independent prognosis marker for HCC patients. Conclusion:The down-regulated serum miR-148 was related to disease condition,malignant progression and prognosis of HCC patients. Moreover,serum miR-148 might act as a potential marker for diagnosis and differential diagnosis,evaluation of disease,observation of curative effect and judgment of prognosis for HCC.

    • Constration of IL-15 knockout MGC-803 cell line by CRISPR/Cas9 technology

      Abstract:Objective:To construct IL-15 knockout MGC-803 cells by using CRISPR/Cas9 genome engineering technology. Methods:Guid RNA was designed by trageting the second exon of IL-15 gene,which encoded its homeodomains. PX458-gRNA recombinant plasmid was constructed and transformed into competent Stbl3. To confirm the efficitiveness of designed gRNA,the recombinan was screened and sequenced. The expression of IL-15 gene of MGC-803 cells sorted through FACS was detected by ELISA, and the cell cycle was detected by FACS. Results:Compared with negative control group,the expression of IL-15 transfected with PX458-IL-15-gRNA plasmid was significantly lower (P < 0.001). Conclusion:The IL-15 konctout gastric cancer cell line(MGC-803 cells)had been successfully constructed by using CRISPR/Cas9 system,which lays the foundation for further study of the mechanism and founction of IL-15 in tumors.

    • miR-145 regulates cisplatin resistance of human gastric cancer cell line via targeting IGF1R and IRS1

      Abstract:Objective:To investigate the possible role of miR-145 in the formation of cisplatin resistance in human gastric cancer cell line. Methods:Expression of miR-145 was assayed by quantitative real-time PCR. MTT (3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide) and clonogenic assays were used to detect cell viability and the drug-resistance phenotype changes of-cancer-cells associated with up-regulation or down-regulation of miR-145. Dual-luciferase activity assay was used to testify the target genes of miR-145. Protein expressions were measured by western blot,immunohistochemistry or Immunofluorescence staining. Flow cytometry was used to detect CDDP induced apoptosis. Results:We found that miR-145 was significantly down-regulated in both gastric cancer tissues and various gastric cancer cell lines. In addition,it was down-regulated in cisplatin-resistant gastric cancer cell line SGC7901/cisplatin (DDP) and the down-regulation of miR-145 was concurrent with the up-regulation of IGF1R and IRS1,compared with the parental SGC7901 cell line,respectively. In vitro drug sensitivity assay demonstrated that over-expression of miR-145 sensitized SGC7901/DDP cells to cisplatin. The luciferase activity of the above proteins 3'-untranslated region-based reporters constructed respectively in SGC7901/DDP cells suggested that IGF1R and IRS1 were the direct target genes of miR-145. Enforced miR-145 expression reduced its target proteins level,inhibited SGC7901/DDP cells proliferation and enhanced SGC7901/DDP cells to DDP-induced apoptosis. Conclusion:Our findings suggested that hsa-miR-145 could modulate cisplatin resistance of human gastric cancer cell line at least in part by targeting IGF1R and IRS1.

      Abstract:Objective:To investigate the expression of c-Myc in non-small cell lung cancer (NSCLC)tissues,and to explore the prospects of the c-Myc gene silencing in NSCLC gene therapy. Methods:The expression levels of c-Myc mRNA in 84 NSCLC tissues and the corresponding para-cancerous tissues were detected by RFQ-PCR(real-time fluorescence quantitative-PCR). C-Myc siRNA was transfected into NSCLC H1299 cells by LipofectAMINE 2000. Cisplatin toxicity experiments further validated effect of inhibiting tumor cells with c-Myc gene silencing. The expression level of c-Myc mRNA in H1299 cells was detected by RT-PCR,and the effect of inhibiting H1299 cells was detected by CCK-8 (Cell Counting Kit-8). Results:The expression level of c-Myc mRNA in NSCLC tissues was higher than that in the corresponding para-cancerous tissues (P < 0.05). C-Myc siRNA was successfully transfected into NSCLC H1299 cells. After transfection,the expression level of c-Myc mRNA was down-regulated in H1299 cells. Effect of inhibiting tumor cells increased significantly when the c-Myc gene silencing and cisplatin were in combination (P < 0.05). Conclusion:The expression level of c-Myc mRNA in NSCLC tissues is higher than that in the corresponding para-cancerous tissues. The lower expression of c-Myc can significantly inhibit NSCLC H1299 cells. It is suggested that c-Myc may act as an oncogene in NSCLC gene therapy.

      Abstract:Objective:To observe the ability of self-renewal and differentiation of mouse spermatogonial stem cells(SSCs)in vitro. Methods:Mouse spermatogonial stem cell line was established stably. The capacity of SSCs self-renewal in vitro was detected via EdU Proliferation Kit; Cell proliferation was observed by Live cell Imaging System. Cell apoptosis was tested by TUNEL method. The capacity of SSCs differentiation was observed by retinoic acid (RA) induction. Results:After incubation with EdU for 2 hours,the proliferation rate was analyzed as an average of 40.75% by EdU Flow Cytometry Assay Kits. The dividing stem cells were clearly catched via short-term live cell imaging. The signal of apoptosis of SSCs was nearly undetected by TUNEL methods. The expression of genes(C-kit,Scp3 and Stra8)marking differentiation was increased after RA induction(P < 0.05,n=3). Conclusion:Similar as SSCs in vivo,SSC line in vitro also has the capacity of self-renewal and differentiation. It provides us a good platform to carry out the function of the related genes and proteins during mouse spermatogenesis.

      Abstract:Objective:To construct heart-specific lncRNA-uc.167 transgenic mice and analyze their phenotype. Methods:After double enzyme digestion by Sfi1 and Asc1,Vector plasmid pRP. ExSi Isl1 and purpose fragment uc167-pMD18■ Tplasmid were recycled and connected. And then, the uc.167 gene was cloned into the downstream of the Isl promoter specifically expressed in myocardial tissue to construct the transgene expression vector. Finally,we made the resulting vector linearization,established uc.167 transgenic mice by micro injection and detected the genotype by PCR. The phenotype of transgenic mice was preliminarily analyzed by cardiac histology and echocardiography. Results:We successfully constructed uc.167 transgenic mice. Echocardiography displayed that transgenic mice had no obvious abnormality in cardiac structure and function compared with the wild type mice. Heart weight index was not significantly changed both in positive and negative transgenic mice. HE staining suggested ventricular size and ventricular wall thickness had no significant difference and there were also no abnormal phenomenon including the change of cell size,inflammatory infiltration and cell edema in myocardial cell. Masson staining showed that both positive and negative transgenic mice had no significant change in myocardial fibrosis. Conclusion:Uc.167 transgenic mice did not appear obvious cardiac malformations,and cardiac structure and function were not significantly abnormal.

    • Effects of levosimendan on calcium transient in norepinephrine-cultured neonatal rat ventricular myocytes

      Abstract:Objective:To explore the alterations of the calcium transient in the development of heart failure,and the impact of levosimendan(LeV)on intracellular Ca2+ dynamics. Methods:The neonatal rat ventricular myocytes in culture were divided into 4 groups,normal,NE alone (10 μmol/L),NE+LeV1(0.1 μmmol/L), and NE+LeV2(1 μmmol/L). Calcium transients of myocytes loaded by Fluo-3/AM were observed under Laser scanning confocal microscope. Results:(1)Compared with the control,calcium wave dispersed,conducted slowly,and exibited dyssynchrony of Ca2+ release in NE group,NE accelerated the cultured cell beating rate(26.7 ± 4.3 vs. 11.6 ± 3.6,P < 0.01),decreased peak systolic Ca2+(128.37 ± 65.44 vs. 155.33 ± 61.77,P < 0.05),shortened time to peak(Ttp;0.413 ± 0.324 vs. 0.212 ± 0.050,P < 0.01)and decay time(Tau;1.162 ± 0.524 vs. 0.722 ± 0.169,P < 0.01)of calcium transient,except Ca2+ transient amplitude and resting Ca2+.(2)NE in the presence of LeV increased synchrony of calcium transient,reduced Ttp(0.212 ± 0.044 vs. 0.413 ± 0.324,NE+LeV1 vs. NE,P < 0.01 and 0.205 ± 0.062 vs. 0.413 ± 0.324,NE +LeV2 vs. NE,P < 0.01 respectively)and Tau (0.735 ± 0.269 vs. 1.162 ± 0.524,NE +LeV1 vs. NE,P < 0.01; and 0.753 ± 0.152 vs. 1.162 ± 0.524,NE + LeV2 vs. NE,P < 0.01),had no effect on cell beating rate,peak systolic or resting Ca2+,and transient amplitude.(3)There were no concentration difference in the presence of Levosimendan. Conclusion:Our results show that calcium transient gets weaker and slower,exibiting dyssynchrony of Ca2+ release in failing rat ventricular myocyte. Levosimendan exerts positive inotropic effect through accelerating the velocity of calcium signaling propagation and synchronzing the calcium release.

    • The effect of zoledronate on gene expression of calmodulin and calcineurin during osteoclast differentiation

      Abstract:Objective:The study is aimed to examine the effects of zoledronate(ZOL) on gene expression of calmodulin and calcineurin during differentiation of osteoclasts. Methods:RAW264.7 cells were divided into 2 groups,group A and group B. The cells in both groups were induced with 100 ng/ml RANKL(ligand of receptor activator of nuclear factor κB)for 4 d for osteoclast differentiation. At the second day of RANKL induction,cells in group B also treated with 1×10-6 mol/L ZOL for 48 h. Four days later,the cells were harvested and examined for osteoclastogenesis,as well as gene expression of both calmodulin and calcineurin. Results: TRAP positive multinuclear cells,number and size of resorption lacunaes in group B were 8.8 ± 2.3,6.7 ± 1.2 and 997.1 ± 14.8 μm2 respevtively,which were significantly lower than those (19.7 ± 2.9,13.3 ± 1.5 and 1676.9 ± 24.9 μm2)in group A (P < 0.01). mRNA and protein level of calmodulin in group B decreased about 51% and 78.7%,respectively when compared with group A (P < 0.05),and those of calcineurin also decreased about 37% and 69.5%,respectively(P < 0.05). Immunofluorescent cytochemistry showed obvious decrease of fluorescent intensity of calmodulin and calcineurin protein in group B when compared with group A. Conclusion:ZOL significantly inhibits osteoclastogenesis and down-regulates gene expression of calmodulin and calcineurin,which may play a role in ZOL-induced inhibition of osteoclastogenesis.

      Abstract:Objective:To investigate the new method of curing the type Ⅰ Diabetes by cultivating and trans-planting the composite artificial scaffold of MIN-6 β cells. Methods:Silk-collagen type Ⅳ scaffold was prepared by the method of freeze-drying. We implant MIN-6 β cells on the silk-collagen type Ⅳ scaffold and then observe the distribution situation of the growing cells and the vitality differences between those cells that growing on the ordinary plane and the composite scaffold. Reaction difference between those cells was compared. After transplantation,the change of the blood glucose level was monitored and the function of cell secretion was tested. Results:More β structure appeared after the silk- collagen type Ⅳ scaffold cross-linked by the methanol. The vitality of those cells cultivated on the scaffold was obviously better than those on the ordinary plane (P < 0.05). Both groups had a good stimulation reaction on both low & high concentration glucose,and the scaffold groups had a better cell secretory function than that of the ordinary plane groups(P < 0.05). MIN-6 β cells were dyed to brown and the cells still had a nice insulin secretory function. Conclusion:MIN-6 β cells which are three-dimensionally cultivated by the silk-collagen type IV scaffold have better vitality and relatively long-lasting insulin secretory function.

    • Involvement of P2X7 receptor and ATP in Müller cell gliosis induced RGC-5 cells apoptosis

      Abstract:Objective:To explore the possible involvement of Müller cell gliosis,ATP/ P2X7 receptors and RGC-5 apoptosis in vitro. Methods:We cultured and purificated rat retinal Müller cells,RGC-5. The concentration of extracellular ATP was quantified by bioluminescence assay. After adding the medium of DHPG-activated Müller cells Medium(conditioned medium /CM)to RGC-5 cells,TUNEL assay and Western blot of anti-and pro-apoptotic proteins were used to observe RGCs apoptosis. Immunofluorescence and Western blot were also used to detect the P2X7 expression of RGC-5 with CM. Results:DHPG induced an increasing ATP release from Müller cells. After adding the CM to RGC-5 cells,TUNEL assay showed a significantly increase RGCs apoptosis while P2X7 receptor blocker BBG reduced apoptosis of RGCs. Moreover,CM treatment of RGC-5 cells significantly increased Bax protein level and decreased Bcl-2 protein level,which was also mimicked by BzATP and blocked by BBG,respectively. Conclusion:The current study suggests that DHPG-activated Müller cells could aggravate RGCs apoptosis,while the release of ATP from Müller cells and the activation of P2X7 receptor in the RGC-5 might be involved in this process.

    • Effects of estrogen on lipid metabolism and glucose of female rats

      Abstract:Objective:To investigate the effect of estrogen on lipid metabolism and glucose. Methods:Fotry 7-week-old female Sprague-Dawley rats were randomly divided into 4 groups,the control group(Con),sham-operated group(sham),ovariectomy(OVX)group and OVX rats given high dose E2 treatment (OVX+ E2). After being weighted,each group of rats were given corresponding processing. Nine weeks later,the change of visceral fat weight,total cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL),high-density lipoprotein(HDL) and glucose were measured. Results:Compared with the control group,the body weight and abdominal fat of OVX group increased significantly. There was a significant difference in the level of TG,TC and LDL between the control and OVX groups. But in the high dose E2-treated OVX group it showed no significant differences. Compared with sham operation group rats,fasting blood glucose was of no difference in OVX group,but existed impairing glucose tolerance. Conclusion:The estrogen level was negatively correlated with the body weight,lower estrogen induced abnormal lipid metabolism and insulin resistance. But this phenomenon can be reversed after treated with estrogen.

    • The preliminary experience of child and older marginal donation in adult liver transplantation

      Abstract:Objective:To explore the application of Donation after citizen’s death(DCD)and age-related marginal donor for liver transplantation in clinical liver transplantation. Methods:From December 2013 to October 2015,3 cases of DCD age-related marginal donors for liver transplantation were included in the Liver Transplantation Center,the First Affiliated Hospital of Nanjing Medical University. The clinical situation of liver transplantation was analyzed,retrospectively,including clinical data of both donors,and recipients, and short-term recovery of recipients. Results:Among the 3 cases,2 cases were performed by Chinese Ⅲ DCD ethical standard,and 1 cases was performed by Chinese Ⅱ DCD ethical standard. Among the 3 cases,one case was 7 years old,the other two cases were 79 years old and 83 years old. Three recipients recovered smoothly and liver function recovered well. The recipients by children liver appeared hallucinations,auditory hallucination and delusion of neuropsychiatric symptoms after 1 month,and healed after treatment. Conclusion:The study suggests that to some extent,as long as the scientific evaluation and reasonable application of the appropriate receptors,the liver transplantation of DCD marginal donors can obtain good results. Age should not be an independent taboo for liver transplant. On the basis of adequate assessment,the age-related marginal donor for liver transplantation is feasible.

    • The application value of renal dynamic imaging and serum cystatin C for kidney GFR evaluation in patients with hydroperinephrosis

      Abstract:Objective:To investigate the application value of renal dynamic imaging and serum cystatin C (Cys C) for kidney function evaluation in patients with hydronephrosis. Methods:A total of 72 patients with hydroperinephrosis and 15 healthy people was retrospectively studied in our hospital from May 2011 to July 2015. All patients accepted 99Tcm-DTPA renal dynamic imaging (Renal imaging) and serum cystatin C examinatioin for GFR calculation with rGFR as gold standard. Results:The diagnostic sensitivity and specificity of 99Tcm-DTPA renal dynamic imaging for hydroperinephrosis was 100%. There was no significant difference between I-GFR and rGFR in mild hydroperinephrosis goup while C-GFR was higher than rGFR. The I-GFR value was higer than rGFR in patients with moderate hydroperinephrosis while it was lower than rGFR in patients with severe hydroperinephrosis. There was no significant difference between C-GFR and rGFR in patients with moderate or severe hydroperinephrosis. 99Tcm-DTPA renal dynamic imaging,quantifying GFR of the kidney without hydroperinephros is and it also can effectively evaluate GFR of the kidney with hydroperinephros is when combined with serum Cys C. Conclusion:Renal dynamic imaging has great advantage for early dectction and GFR evaluation of the healthy kidney in patients with hydroperinephros. And it has great potential for precise GFR evaluation of the separate kidney in patients with moderate to severe hydroperinephros when combined with serum Cys C.

    • Determination of the application of CBP therapy performed on SAP by monitoring inflammatory cytokines

      Abstract:Objective:The purpose of this study is to investigate the relationship between the intervention time and the effect of continuous blood purification(CBP)therapy performed on severe acute pancreatitis(SAP)patients by monitoring the changes of inflammatory cytokines. Methods:A total of 33 SAP patients were hospitalized and treated with CBP therapy. The patients were randomly assigned into three groups(A,B,C)according to the time of CBP therapy performed after the onset of SAP,which is,0~24 h,24~48 h and 48~72 h,respectively. The CBP treatment is continuity veno-venos hemofiltration (CVVHF)for 12 h daily for 3 consecutive days. Blood sample of the patients were collected pre and post CBP therapy. Presence of TNF-α,IL-1,IL-6,IL-8,blood amylase(AMY),C-reactive protein(CRP)and APACHE Ⅱ score were measured and calculated. Results:It showed that the indicators for condition evaluation(AMY,CRP,APACHE Ⅱ score)and the level of inflammatory cytokines (TNF-α,IL-1,IL-6,IL-8),were lower post CBP therapy than that pre CBP therapy in all three groups of SAP patients. The decrease in the indicators for condition evaluation such as AMY,CRP and APACHE Ⅱ score in group C (48~72 h)was statistically lower than those in group A and B (0~24,24~48 h). However,the decrease in the level of inflammatory cytokines such as TNF-α,IL-1,IL-6,and IL-8 was higher in group C(48~72 h)than those in group A and B. Conclusion:CBP therapy can effectively reduce the level of inflammatory cytokines in SAP patients,and also improve their overall condition. SAP patients who received CBP therapy within 48 h show a better improvement in their condition by effectively reducing the level of inflammatory cytokines.

      Abstract:Objective:To investigate the crown-root an-gle of central incisor of early youth,and to analyze its correlation with cephalometric measurement of hard tissue. Methods:Forty-two malocclusion pations were enrolled to measure the crown-root angle of the 168 central incisors with MIMICS 10.0 software,and to measure three-dimensional and lateral cephalogram two-dimensional SPSS21.0 statistical software was used for the analysis of the correlationship between the crown-root angle and the results of cephalometric measurement. Results:In early youth,there is no significant correlation between the crm angle of the upper central incisor with the 8 cephalometric indexes. The cr angle of the upper central incisor is positively associated with the protrusion of upper central insicor,and is negatively correlated with SNA. The crm angle and the cr angle of the lower central incisor are positively associated with ANB and the protrusion of the lower central insicor. Conclusion:Some cephalometric indexes of hard tissue are related to the crown-root angle of central incisor of early youth,which has the clinical reference value.

    • The association between vitamin D,parathyroid hormone levels and the product of heart rate and systolic blood pressure in community-dwelling adults

      Abstract:Objective:To investigate the relationship between the serum vitamin D,parathyroid hormone and the heart rate-systolic blood pressure product (RPP)among adults. Methods:Based on the previous cross-sectional study,data analyses were carried out on 621 permanent residents aged≥40 years old. Analysis of variance and generalized liner model analysis were used to analyze the relationship between the 25 (OH)D,PTH and heart rate,systolic blood pressure,RPP. Results:Compared to participants with 25(OH)D≥20 μg/L,those with 25(OH)D<10 μg/L had higher heart rate and RPP,by increasing (5.0 ± 1.2) beats/min (P < 0.01)and(754.3 ± 187.8) beats/min-mmHg (P < 0.01),respectively. Among the high PTH group,heart rate increased 10.7 beats/min(P=0.001)and RPP increased 1 279.4 beats/min-mmHg(P=0.011)in participants with 25(OH)D<10 μg/L compared to those with 25(OH)D≥20 μg/L. After adjustment for age,gender and other variables,heart rate and RPP also increased 8.2 beats/min(P=0.017)and 1 041.8 beats/min-mmHg (P=0.06)in those with 25 (OH)D<10 μg/L. While in the low PTH group,heart rate and RPP increased 3.7 beats/min (P=0.018)and 639.5 beats/min-mmHg (P=0.007)respectively,and the adjusted RPP increased 595.7 beats/min-mmHg(P=0.012)in participants with 25(OH)D<10 μg/L compared to those with 25(OH)D≥20 μg/L. Conclusion:Vitamin D deficiency is associated with the increase of heart rate and RPP, which suggestes low vitamin D status may increase cardiac work,especially combination of vitamin D deficiency and elevated PTH maybe play a co-effect on the cardiac work.

    • Prognostic value of miR-210 in breast cancer:A meta-analysis

      Abstract:Objective:To summarize the predicting role of increased miR-210 expression for survival in patients with breast cancer. Methods:All of relevant studies were identified by searching PubMed,EMBASE,Web of Science and CNKI. Pooled hazard ratios(HRs)with 95% confidence intervals(CIs) for overall survival(OS),recurrence-free survival(RFS),and disease-free survival(DFS)were calculated by Stata 12.0 software. Results:A total of 585 cases from 6 eligible studies were involved for this global meta-analysis. For OS,the pooled HR of higher miR-210 expression in breast cancer was 2.29(95%CI:0.89~5.91,P= 0.086). And for DFS/RFS,the pooled HR of higher miR-210 expression in breast cancer was 2.81(95%CI:1.35~5.87,P=0.006). Conclusion:This meta-analysis suggests that high expression of miR-210 may predict poor survival in patients with breast cancer.

    • A meta-analysis on the effect of early minimal feeding on low birth weight infants of China

      Abstract:Objective:To evaluate the effect of early minimal feeding on low birth weight infants of China. Methods:We searched CBM,CAJ,Wanfang and CQVIP databases using keywords since databses established. Also we searched MEDLINE for Chinese papers. Results:Tweenty-four researches were included.(1)Time of parenteral nutrition in early minimal feeding group is shorter than control group WMD= -5.73;95%CI=[-6.68,-4.78];P < 0.00001;(2)Weight gain per day in early minimal feeding group is more than control group WMD= 3.64;95%CI=[1.85,5.44];P < 0.0001;(3)Time of returning to birth weight in early minimal feeding group is shorter control group WMD=-2.59;95%CI=[-3.35,-1.83];P < 0.00001;(4)Incidence of NEC in early minimal feeding group doesn’t show statistical significance compared with control group OR= 0.70;95%CI=[0.35,1.41];P=0.32;(5)Incidence of feeding intolerance is lower than control group OR= 0.38;95%CI=[0.24,0.60];P < 0.0001;(6)Incidence of hypoglycemia is lower than control group OR= 0.19;95%CI=[0.10,0.35];P < 0.00001. Conclusion:The early minimal feeding group has an advantage over control group in time of parenteral nutrition,weight gain,time of returning to birth weight,incidence of NEC,feeding intolerance and hypoglycemia.

    • Associations between four polymorphisms(FokⅠ,ApaⅠ,BsmⅠ,TaqⅠ)of vitamin D receptor gene and type 1 diabetes:A meta-analysis

      Abstract:Objective:The vitamin D receptor(VDR)gene polymorphisms have been suggested to be involved in the development of type 1 diabetes(T1D). However,the results to date have been inconclusive. We performed a meta-analysis to examine the associations. Methods:Literature was retrieved from PubMed,Web of Science and Chinese databases WanFang. Pooled odds ratios(ORs)with 95% confidence intervals (CIs) were calculated using a random effect model. Results:The results indicated that BsmⅠ and ApaⅠ were associated with an increased risk of T1D (B vs b:OR=1.10,95%CI:1.04~1.18,P=0.003,AA vs aa:OR=1.24,95%CI:1.05~1.46,P=0.012,),while the FokⅠ and TaqⅠ polymorphisms were not. In the subgroup analysis by ethnicity and area,the increase risk of T1D remain in the East Asian and Latino subgroup for the BsmⅠ polymorphism(B vs b:OR=1.52,95%CI:1.33~1.74,P < 0.001 for East Asian;OR 1.51,95%CI:1.25~1.82,P < 0.001 for Latino),East Asian subgroup for the ApaⅠ polymorphism(AA vs aa:OR 1.99,95%CI:1.20~3.29,P=0.007). Whereas no significant association was found in other population. Conclusion:Our meta-analysis suggests that the BsmⅠ and ApaⅠ polymorphism may be a risk factor for susceptibility to T1D in Asians. Moreover,BsmⅠ polymorphism may be a risk factor in Latinos. However,the FokⅠ and TaqⅠ polymorphisms are unlikely to be associated with increased T1D risk in the populations investigated.
