Abstract:Objective:To investigate the association of five single nucleotide polymorphisms (SNPs)(rs1800975,rs11615,rs2228000,rs2228001,rs238406) in DNA repair genes with the risk of breast cancer and it’s pathologic characteristics and related hormone receptor levels in Nanjing region. Methods: Genotypes of 5 SNPs in 350 breast cancer female patients and 350 healthy controls were determined by the Sequenom MassArray. Immunohistochemical experiment was used to measure the expression level of estrogen receptor (ER),progesterone receptor (PR) and human epidermal growth factor receptor-2 (HER-2). Logistic regression was applied to analysis the association of different genotypes with the breast cancer risk and it’s pathologic characteristics and related hormone receptor (ER,PR,HER-2) levels. Results: Both rs11615 TC/TT and rs2228000 TT genotype present an increased risk for breast cancer. In contrast,subgroup analysis indicated that rs1800975 AA and GA genotype exert a decreased breast cancer risk in the subgroup of postmenopausal and ER negative,respectively. Besides,rs2228000 T allele showed significant association with PR,HER-2 positive breast cancer patients. Additionally,rs11615 had an increased breast cancer risk in the subgroup of ER and PR positive,however,rs2228001 showed a decreased breast cancer risk in patients with T3 stage,PR,or HER-2 negative state. Conclusion:Rs2228000 genotype is associated with the increased breast cancer risk. Morever,for specific polymorphism,the tumor characters,levels of ER,PR,HER-2 were related to the occurrence or prevention of breast cancer.

Abstract:Objective:To discuss the expression of B7-H4 protein in breast cancer tissues as well as the relationship among clinical pathological parameters. Methods:Immunohistochemical technique was employed to investigate the expression of B7-H4 in 68 cases of breast cancer and 30 cases of benign fibroadenoma. And statistical analysis was carried out to study the relationship between B7-H4 and pathological parameters. Results:The B7-H4 positive rate of breast cancer was 75%,significantly higher than fibroadenoma tissues (17%)(P < 0.05). And highly expression of B7-H4 was negatively correlated to estrogen receptor (ER) but positively correlated to human epidermal growth factor 2 correlation (HER-2), independent from other factors such as age,size of tumor and clinical stage. Conclusion:Aberrant B7-H4 expression in breast cancer but not benign fibroadenoma implied that B7-H4 may play an important role in the development of breast cancer.

Abstract:Objective:To investigate the expression of miR-486 in serum of patients with gastric cancer and its value to auxiliary diagnosis. Methods: The serum of 100 patients with pathological confirmed GC, 30 gastric polyps and 50 healthy controls were recruited. Serum miR-486 expression was determined by fluorescent quantity PCR. Serum CEA, CA199 and CA724 were determined by chemiluminescence method. Comparison of differences of serum miR-486 between groups was performed by Mann-Whitney U test. Analysis the relationship between serum expression of miR-486 in gastric cancer patients and its clinical pathological characteristics was performed by χ2 test. The relationships between miR-486 and CEA, CA199, CA724 were examined by Spearman correlation analysis. ROC curve and AUC (95% CI) were used to evaluate their diagnosis efficiency. Results: The relative expression of miR-486 in GC, gastric polyps, healthy controls were 0.454 (0.225, 0.653), 0.121 (0.083, 0.166), 0.063 (0.013, 0.119),respectively. The relative expression of miR-486 in GC was significantly higher than that in gastric polyps and healthy controls (P < 0.001). The difference between gastric polyps and healthy controls is not significant (P > 0.05). The differences of expression of miR-486 in serum of GC in tumor differentiation (P=0.046), TNM(P=0.013) and the occurrence of lymph node metastases (P=0.006)were statistically significant except for age, gender, tumor area (P > 0.05). The relative expression of miR-486 in GC had a correlation with CEA (r=0.213, P=0.034),CA724 (r=0.297, P=0.003) but no obvious correlation with CA199 (r=0.037, P=0.714). AUC of serum miR-486 was 0.839, 0.774~0.904 (95%CI) in discriminating GC from healthy controls. Conclusion: The relative expression of miR-486 in GC was significantly higher than that in gastric polyps and healthy controls, serum miR-486 may be a biomarker for the auxiliary diagnosis of GC.

Abstract:Objective:To investigate the level of nucleolin in serum of colorectal cancer patients and its clinical significance. Methods:The levels of serum nucleolin in 172 patients with colon cancer,24 patients with rectal cancer and 80 healthy controls were determined by enzyme linked immunosorbent assay (ELISA). The relationship between serum nucleolin level in colon cancer and clinical features and pathological features were analyzed. Results:The serum nucleolin level in colon cancer patients,rectal cancer patients,and healthy controls were 228.9 ± 212.4 pg/mL,115.1 ± 34.5 pg/mL and 101.1 ± 32.4 pg/mL. The serum nucleolin level in colon cancer patients was significantly higher than that in healthy controls (P < 0.001),and there was no significantly difference between rectal cancer patients and healthy controls (P > 0.05). The sensitivity and specificity of serum nucleolin detecting in colon cancer were 91.9% and 83.8%,and serum carcino-embryonic antigen (CEA)were 68.6%,85.0%,respectively. The level of nucleolin in serum of infiltrating type colorectal cancer was significantly higher than that in protruded type and ulcerative type (P < 0.05). There was no correlation with the serum nucleolin level in colon cancer and age,gender,tumor size,differentiation degree,degree of invasion,lymph node metastasis,distant metastasis,TNM stage,and Ducks stage. Conclusion:The detection of serum nucleolin level in colon cancer patients has a high sensitivity and specificity,and the sensitivity is better than CEA and the specificity is as similer as CEA. Serum nucleolin may be potential valuable for colon cancer diagnosis.

Abstract:Objective:This study investigated the expression and biological role of toll-like repeptor-4 (TLR4) in human breast cancer proliferation and metastasis. Methods:Lipopolysaccharide (LPS) was used to stimulate MCF-7,expression of TLR4 mRNA and protein was analyzed by RT-PCR,Real-time PCR,FCS and Western blotting;MMP-2,MMP-9 and VEGF were measured by RT-PCR and Real-time PCR on the mRNA levels and ELISA on the protein levels;MyD88 was analyzed by Western blotting;The cytokines in the supernatants were measured by CBA;The cell proliferation was analyzed by MTT;The invasion was analyzed by wound healing assay. Results:Using LPS to stimulate MCF-7,TLR4 mRNA and protein increased obviously. However,the proliferation had no significant changes. In addition,the activation of TLR4 promoted the expression of MMP-2,MMP-9 and VEGF notablely. LPS promoted the invasion of MCF-7. Moreover,the stimulation of LPS significantly increased the expression of MyD88,and caused the secrection of IL-6. Conclusion:This study indicated that TLR4 may participate in the progression and metastasis of human breast cancer.

Abstract:Objective:To investigate the effects of major vault protein (MVP)on the proliferation of vascular smooth muscle cells(VSMCs). Methods:The wild-type mouse VSMCs were isolated,and cultured,and the human aortic smooth muscle cells(HAoSMCs)were used. Western blot analysis was used to determine the MVP,α-SMA and SM22α. Expression of MVP stimulated by platelet-derived growth factor (PDGF-BB)or 15% fetal bovine serum (FBS) was measured. MVP siRNA was used to downregulate the MVP expression in HAoSMCs. Cell proliferation CCK-8 assay was conducted to detect the proliferation ability of VSMCs in the case of MVP deficiency or downregulation. Results:After the treatment of PDGF-BB or 15% FBS,the MVP expression of VSMCs was downregulated in a time-dependent and dose-dependent manner. Compared with wild-type mouse VSMCs,the proliferation of MVP in knockout mouse VSMCs was suppressed. MVP siRNAs inhibited FBS-induced HAoVSMCs proliferation. Conclusion:MVP may played crucial effects in the proliferation of the VSMCs.

Abstract:Objective:To study whether hypoxia and reoxygenation (H/R)injury induces the necrosis of cardiomyocyte via regulation of programmed necrosis (necroptosis). Methods:Neonatal rats ventricular myocytes were isolated from neonatal Sprague–Dawley rats. The incubated cells were subjected to reoxygenation (4 h)after hypoxia (2 h). The effect of H/R injury on the necrosis of cardiaomyocytes was determined by PI (propidium iodide)staining. Wsetern blot and Co-IP were performed to test the protein expression of receptor interacting protein (RIP)1 and 3,the formation of RIP1/RIP3 complex Ⅱ,and the ubiquitination level of RIP1 and RIP3. Results:When subjected to reoxygenation (4 h) after hypoxia (2 h),the protein expression levels of RIP1 and RIP3 were significantly increased compared with those of control. Besides,H/R injury also promoted the formation of RIP1/RIP3 complex Ⅱ and upregulated the ubiquitination of RIP1 and RIP3. Conclusion:Hypoxia and reoxygenation injury could induce the necrosis of cardiomyocytes,which may be involved in regulation of RIP1/RIP3 dependent programmed necrosis signaling pathway.

Abstract:Objective:To observe the effects and study the mechanisms of slow-releasing hydrogen sulfide donor GYY4137 on the vasodilation of thoracic aortic in spontaneously hypertensive rats (SHRs). Methods:Male SHRs at 12 weeks of age were randomly divided into 4 groups treated with different dosages of GYY4137:0(SHR group),10(GYY10 group),25(GYY25 group)or 50(GYY50 group)mg/kg/day. GYY4137 was given by intraperitoneal injection once daily for 4 weeks. Age-matched normotensive Wistar-Kyoto(WKY)rats served as controls (WKY group). SHR and WKY control groups received the same volume of physiological saline. During treatment,systolic blood pressure (SBP)was measured by the tail-cuff method. After 4 weeks,vasodilation of thoracic aortic was measured. The degree of oxidative stress was evaluated by DHE staining,malondialdehyde (MDA)and total antioxidant capacity(T-AOC). The expression of p85α,Akt,p-Akt(Ser473),eNOS and p-eNOS(Ser1177)was detected by western blot. Results:After treatment,GYY4137 improved endothelium-dependent vasodilation of thoracic aortic in SHRs,lessened the superoxide generation,decrease MDA level,enhanced T-AOC,and increased the protein level of p85α and the phosphorylation of Akt and eNOS. Conclusion:GYY4137 improved endothelium-dependent vasodilation of SHRs,which may be associated with the suppression of oxidative stress and activation of PI3K/AKT/eNOS signal pathway.

Abstract:Objective:To explore the immunogenicity of severe fever with thrombocyte-penia syndrome virus(SFTSV)glycoprotein Gn. Methods:SFTSV glycoprotein Gn gene and optimal Gn gene were amplified by polymerase chain reaction(PCR)respectively. The gene product were cloned to pJW4303 to construct recombinant plasmids and all constructs were identified by sequencing,and then transiently transformed into HEK293T cells to measure the Gn expression by Western blot. We immuned the BALB/c mice with recombinant plasmids and blank vector,and verified its immunogenicity by enzyme-linked immunosorbent method. Results:The recombinant plasmids pJW4303-WSP-Gn and pJW4303-tPA-Gn-opt were constructed success-fully. The pJW4303-WSP-Gn transiently expressed Gn antigen in cell lysates of HEK293T cells in vitro,and pJW4303-tPA-Gn-opt in supernatants as same as cell lysates. The specific IgG antibodies induced by pJW4303-tPA-Gn-opt was earlier and higher than that of pJW4303-WSP -Gn. Conclusion:SFTSV glycoprotein Gn showed good immunogenicity. The recombinant plasmid pJW4303-tPA-Gn-opt promoted the synthesis and secretion of Gn,and showed better humoral immunogenicity than pJW4303-WSP-Gn.

Abstract:Objective:To explore the role of HRD1 in apoptosis and lipid deposition caused by oxLDL in human umbilical vein endothelial cells (HUVECs). Methods:HUVECs were incubated with 0 μg/ml oxLDL as control group,or with 50 μg/ml oxLDL as oxLDL group. Western blot was used to analyze expression of HRD1 protein. HRD1 expression in vascular tissue was detected by immunofluorescence analysis. HRD1 effects on cell apoptosis and lipids deposition of HUVEC was detected by western blot and oil red o staining analysis. Results:Expression of HRD1 protein decreased significantly in oxLDL group than that in control group. Meanwhile,HRD1 also expressed in the athrosclerostic plaques,and predominantly in the endothelial cells. oxLDL induced the significant increases in apoptosis and lipids deposition of HUVEC. While the HRD1 overexpression by the infection of Ad-HRD1 markedly inhibited the Cleaved PARP and Cleaved Caspase 3 expression and lipids deposition caused by oxLDL. Conclusion:HRD1 inhibited the apoptosis and lipids deposition of HUVEC induced by oxLDL.

Abstract:Objective:To investigate the effect of elevated retinoic acid inducible gene-I (RIG-I)on the growth of NIT-1 pancreatic β-cell and possible mechanisms during this process. Methods:Different concentrations of retinoic acid (RA)were used to activate RIG-I with different periods. Cell viability was assessed by MTT colorimetric assay. RIG-I was detected using real-time RT-PCR and Western blot analysis. Cell cycle distribution was measured by FACS analysis,and cell cycle protein p27 was further detected by Western blot analysis. Results:Retinoic acid upregulated the mRNA and protein level of RIG-I in NIT-1 cells (P < 0.05). Elevated RIG-I significantly inhibited the viability of NIT-1 cells in a dose-dependent manner (P < 0.05). Furthermore,elevated RIG-I induced an increase in the proportion of cells at the G1 phase and a corresponding decrease in the number of cells at the S phase (P < 0.05). Additionally,elevated RIG-I caused a significant increase of p27 protein (P < 0.05). Conclusion:The results suggest that activated RIG-I induced upregulation of p27 in pancreatic β-cell,which inhibits proliferation through cell cycle arrest at the G1 phase,and ultimately results in an overall β-cell mass deficiency.

Abstract:Objective:To explore effects of PI3K/mTOR dual inhibitor NVP-BEZ235 on proliferation and apoptosis in nasopharyngeal carcinoma (NPC). Methods:We treated NPC cells CNE1 and SUNE1 with NVP-BEZ235 in different concentrations,MTT assay was used to detect inhibition of cell proliferation by NVP-BEZ235. Inhibitors-induced apoptosis in NPC cells was evaluated by propidium iodide (PI)staining assay,4',6-diamidino-2-phenylindole (DAPI)staning and Transferase-mediated FITC-12-dUTP nick-end labeling (TUNEL)assay. Western blot analysis was used to detect the proteins of PI3K/mTOR pathway. Tumor xenografts were established by injection of CNE1 cells into nude mice to observe the growth inhibitory effect by NVP-BEZ235 and effects of the inhibitor on PI3K/mTOR pathway and apoptosis in vivo. Results:① NVP-BEZ235 selectively inhibited proliferation of NPC cells rather than normal nasopharyngeal cells. ② NVP-BEZ235 selectively inhibited the phosphorylation of PI3K/mTOR downstream signal molecules. ③ Exposure of NPC cells to NVP-BEZ235 resulted in G1 growth arrest and significant apoptosis. ④ In CNE1 nude mice xenograft models,orally NVP-BEZ235 efficiently attenuated tumor growth and shrinked tumor volumes,and induced apoptosis through blocking phosphorylation of PI3K/mTOR downstream molecules in vivo. Conclusion:NVP-BEZ235 selectively inhibited proliferation of NPC cells and induced apoptosis through G1 growth arrest and inhibiting the phosphorylation of PI3K/mTOR downstream signal molecules both in vivo and in vitro.

Abstract:Objective:The aim of this study was to determine the stress-strain properties of Beagle Dog’s periodontal ligament(PDL)and compare response characteristics of PDL in different root anatomic level and different loading rate status in uniaxial tensile stage for further exploration of the elasticity modulus of PDL. Methods:4 Beagle Dogs were killed before the bone-PDL-tooth complex was cut and the transverse samples were made. The experimental samples were divided into 3 groups according to the anatomic level of the root(cervical margin,mid-root,and apical),and each group contained 8 samples. Samples were randomly selected by rate of v1=0.05 mm/s and the other four samples by v2=0.01 mm/s. The uniaxial tensile test was performed and stress-strain curves of PDL was obtained. The elastic modulus was obtained by calculating the slope of the straight line in each segment,and paired t test analysis of influence of tensile rate on the elastic modulus of the PDL was carried out. The randomized block design and variance analysis was set to evaluate the difference caused by different anatomic level of the tooth root. Results:All samples of the stress-strain diagram showed a bilinear property. In different rates,the elastic modulus of linear region differed significantly in mid-root(P=0.016 < 0.05)and apex(P=0.016 < 0.05)but cervical margin represented no significant difference(P > 0.05). and the elastic modulus of toe region differ significantly in mid-root(P=0.001 < 0.05)and cervical margin(P=0.002 < 0.05)but no significant difference in apex(P > 0.05). The difference of elastic modulus was not significant between the different anatomical level of the root (P > 0.05)but the difference between the size of toe region was significant(P < 0.05). Conclusion:The tensile stress strain curves of Beagle Dog’s PDL can be fitted to the bilinear property. The elasticity modulus varies from 0.021 ± 0.003 MPa to 1.150 ± 0.235 MPa. The tensile rate can significantly affect the elastic modulus of the PDL. Difference caused by anatomic levels of PDL was not significant.to elasticity modulus but significant to the size of toe region.

Abstract:Objective:To find a new technique for determine sagittal screw angle(SSA)cervicothoracic spine (C6-T6) pedicle screw placement by measuring the angles between different anatomic landmarks and between planes of upper vertebral end-plate. Methods:CT images of 60 cases (thirty males and thirty females) of normal adult human cervicothoracic segments (C6-T6) were analyzed in thiss study,The angles between the plane of the upper vertebral end-plate and the line between the vertex of the spinous process and the upper one (method one); the lower one (method two); the superior margin line of spinous process (method three); the left vertebral laminae plane of the same spine (method four); the right vertebral laminae plane of the measuring same spine (method five)were measured ,respectively. One or several methods was be chosen as a new placement technique for cervicothoracic spine(C6-T6)pedicle screw surgery on SSA by the reference values of each method in C6-T6 and comparing the pros and cons of different methods. Results:The angles between the plane of the upper vertebral end-plate and the line between vertex of spinous proscess and adjacent ones were 100° to 110°with great deviation in C6 to C7,and from T1 to T6,the angles approached 90°with smaller deviation;the angles between the plane of the upper vertebral end-plate and the left vertebral laminae plane decreased gradually from C6 to T6. The vertebral laminae plane was 90° in the vertical position. Conclusion:If the angles of pedicle screw agianst the vertebral laminae plane are about 90° at C6 to T6,the sagittal srew surgery are 0°,the vertebral laminae plane can be the identification marker for cervicothoracic spine(C6-T6) pedicle screw surgery on SSA.

Abstract:Objective:To study the expression of vascular endothelial growth factor(VEGF) and it’s receptors FLT-1 and FLK-1 in rats with portal hypertensive gastropathy. Methods: Wistar rats were devided into two groups:PHG group whose portal hypertension was established by ligating the portal vein,and the sham-operation(SO)group served as control. After two weeks portal vein pressure was measured by physiological pressure capsule. Immunhistochemistry was used to detect the expression of VEGF and it’s receptors,and RT-PCR was used to detect the mRNA expression of VEGF and the receptors in both groups. Results: The pressure of PHG group was higher significantly than that in SO group(P < 0.05). And the PHG group has a significantly higher expressions of VEGF and the receptors compared with SO group(P < 0.05),the same of mRNA expressions. Conclusion: VEGF and the receptors may participate in the pathogenesis of portal hypertensive gastropathy.

Abstract:Objective:To evaluate the safety and effect of transcatheter closure of atrial septal defect(ASD)in patients aged over 60 years old. Methods:Fifty-one over 60-year-old patients underwent transcatheter ASD closure,and their clinic data and procedural outcome were analyzed in this study. Meanwhile,all the data and outcomes were compared with other age groups. Results:Patients above 60 years old with ASD (60ASD)accounted about 7.9%(51/647)of adult ASD patients. The incidence of atrial fibrillation in 60ASD (27.45%)was significantly higher than that of ASD patients aged 18~60 years (59ASD)(3.52%). Compared with 59ASD,the cardiac remodeling showed more obvious effects in 60ASD. Those effects included atrial and ventricular enlargement,increasing atrioventricular chamber pressure and pulmonary vascular resistance. However, the correlation analysis suggested there was weak correlation between age and pulmonary arterial pressure,left atrial diameter,right atrial diameter,left ventricular end diastolic diameter,right ventricular diastolic diameter,and pulmonary resistance (r=0.263,0.5,0.215,0.174,0.153,0.335,respectively). Successful rates of procedure and major complications didn’t show significant difference between 60ASD and 59ASD. Right cardiac remodeling after procedure was significant,as the right atrial/ventricular overload was alleviated and diameter was reduced no matter which age group was the patient in. However,the improvement of 60ASD cardiac remodeling was less than 59ASD. Conclusion:Transcatheter ASD closure is effective and safe among ASD patients over 60 years old.

Abstract:Objective:To explore the change and relationship between serum homocysteine level and other common risk in acute ischemic stroke. Methods: According to the level of homocysteine,we divided 392 cases with acute ischemic stroke into two groups:ischemic stroke with hyperhomocysteinemia group (HHcy group,n=88 cases)and ischemic stroke with normal homocysteine level group (nHcy group,n=304 cases). The clinical characteristics,the level of blood glucose,blood lipid,uric acid,D-dimer and glycosylated hemoglobin (HbA1c)were retrospectively analyzed. Results:In the HHcy group,the male patients were more than the nHcy group. The level of serum homocysteine and HbA1c in the HHcy group was higher than the nHcy group,and the infarct size was larger than the nHcy group,all P < 0.05. The result of correlation analysis of serum homocystiene and other common risk factors showed that the level of Hcy was positively related to the level of uric acid and D-dimer.(r:0.144 and 0.129 respectively,all P < 0.05). Conclusions:Ischemic stroke caused by hyperhomocysteinemia is common in men and the infarct size more lager,and correlation to the level of uric acid and D-dimer

Abstract:Objective:To explore the relationship between serum uric acid and islet beta cell function in newly diagnosed type 2 diabetes. Methods:We analyzed 605 newly diagnosed type 2 diabetes inpatients. We detected serum uric acid,cholesterol,triglyceride etc. and conducted OGTT and insulin release tests on each patient and total insulin level (InsAUC120),early phase insulin secretion index(INSR30),total insulin secretion index(INSR120)as well as beta cell function index(HOMA-β)were calculated. The population was divided into high and low uric acid groups respectively in the male and female groups. The relations between serum uric acid and InsAUC120,INSR30,INSR120 as well as HOMA-β were evaluated. Results:①The patient with hyperuricemia had higher InsAUC120,INSR30,INSR120 and HOMA-β than patient with hypouricemia;②The serum uric acid revealed significantly positive relationships with InsAUC120,INSR30,INSR120 and HOMA-β;③The correlations still existed after adjusting for possible influencing factors by multivariate regression analysis. Conclusion:Serum uric is an independent factor for indicating the islet beta cell secretion capacity and can play a potential serum marker of the insulin secretion ability.

Abstract:Objective:To retrospectively study the angiographic findings of intractable bleeding in head and neck,and evaluate the safety and effectiveness of transcatheter arterial embolization (TAE) for these patients. Methods: Forty-eight patients underwent TAE for intractable bleeding in head and neck from January 2009 to March 2015. Through analyzing medical history and angiographic features of these patients,effectiveness and complications were evaluated according to etiologies and angiographic findings,respectively. Results: The 48 patients included 15 cases of malignant head and heck tumor and 33 cases of benign diseases. There were 36 cases presenting with positive angiographic findings and 12 cases with negative findings. There were no significant difference on positive angiography rate between malignant diseases and benign diseases (P=0.106). Of the cases with negative findings there were 8 patients with idiopathic epistaxis and 4 patients with other diseases. The negative angiography rate of idiopathic epistaxis was higher than that of other diseases (P=0.001). Forty-six patients achieved hemostasis. Recurrence occurred in 5 patients. One patient developed visual defect after embolization. No significant differences were found in recurrence rates between malignant and benign diseases or between positive and negative angiographic findings (P=0.339,P=0.785,respectively). Conclusion: TAE is safe and useful for intractable bleeding in head and neck. Patients with idiopathic epistaxis are prone to present with negative angiographic finding. It seems effective for patients with epistaxis presenting with negative angiograms to receive prophylactic bilateral sphenopalatine artery embolization.

Abstract:Objective:To investigate the association between IR and the vascularization of thyroid nodules in women. Methods:A cross-sectional,population-based study was conducted in Nanjing,China,from June to November 2011. 1,786 female thyroid nodule cases were enrolled in the study. Power Doppler was performed to measure the flow patterns,resistive index(RI),and vascular index(VI)of thyroid nodular vascularization. Fasting insulin,fasting glucose,2-hour oral glucose tolerance test,glycosylated hemoglobin(HbA1c),homeostasis model assessment of IR(HOMA-IR),and body mass index were also tested. Results:HOMA-IR(R = 0.36,P < 0.001)and HbA1c(R = 0.20,P < 0.001)were positively correlated with thyroid nodular flow patterns,and the positive correlations were more significant in participants with large nodules (volume ≥ 0.2 mL). Additionally,in participants with large nodules,HOMA-IR and HbA1c were positively correlated with RI(HOMA-IR:β = 0.41,SE = 0.03,R = 0.43,P < 0.001;HbA1c:β = 0.23,SE =0.03,R = 0.23,P < 0.001)and VI(HOMA-IR:β = 0.47,SE = 0.04,R = 0.53,P < 0.001;HbA1c:β = 0.34,SE = 0.11,R = 0.37,P < 0.001). Conclusion:IR and hyperglycemia were positively correlated with the flow patterns,RI,and VI of thyroid nodules,especially in large nodules. The findings suggest a pivotal role of IR in the distribution,construction,and density of thyroid nodular vascularization,which might contribute to the growth and the progression of thyroid nodules.

Abstract:Objective:To study the clinical values of detection for SPOCK2 protein in serums of preterm infants and to determine a novel risk factor and therapeutic target against serious lung injuries of premature neonates. Methods:Upon diagnostic criteria of Respiratory Distress Syndrome(RDS),premature infants were divided into Group RDS(17 cases)and control(21 cases without RDS). ELISA and Western blot were performed to investigate the serum levels of SPOCK2 protein in every groups and expressions of SPOCK2 in human lung fibroblasts,respectively. The noncoding region in 3 terminal of SPOCK2 gene was analyzed to scan candidate binding sites of microRNA with software TargetScan online and followed verification was performed by fluorescence real-time quantification PCR. Person method was used to analyze the correlation between SPOCK2 and microRNA. The lentiviral vector of mir-124-3p was constructed and transfected into human lung fibroblasts. The regulatory effect of mir-124-3p on SPOCK2 expression was studied by Western blotting. Results:The serum levels of SPOCK2 protein in Group RDS were 16.43 ± 0.54 μg/L,which was higher than that of control(7.24 ± 0.43 μg/L,t=12.81,P < 0.01). There were several microRNAs located in the 3 terminal of SPOCK2 gene,including mir-124-3p,mir-25-3p,and mir-122-5p. However,mir-25-3p and mir-122-5p failed to be observed in serums of preterm infants. The expression of mir-124-3p was negatively correlative with that of SPOCK2 protein in preterm infants with lung injury(r=-0.645 9,P=0.012 6). Mir-124-3p overexpression markedly suppressed the expression of SPOCK2 protein in human fibroblasts. Conclusion:The detection of SPOCK2 protein in serum benefitted evaluating severity of preterm infants with RDS. Mir-124-3p down-regulated the expression of SPOCK2 protein,which might be helpful to build a novel therapeutic strategy against lung injury of premature neonates.

Abstract:Objective:To investigate the adherence status and interrelated factors of highly active antiretroviral therapy(HAART)on men who have sex with men(MSM)in Jiangsu province. Methods:Totally 184 new HIV AIDS patients who received antiretroviral therapy in 2013—2014 were recruited with purposive sampling method in this study. All participants were asked to complete one questionnaire. Results:Among the 184 HIV/AIDS patients,76.1% of participants had a good adherence to highly active antiretroviral therapy (HAART). Univariable analysis showed that employment,smoking and social support (family,friends and other support)were the relative factors which influence the adherence among HIV/AIDS patients. Multivariable analyses showed that employment,smoking,family and other support were the major risk factors (P < 0.05). Conclusion:The medication adherence to HAART of HIV/AIDS patients in MSM is low. The related professional departments should strengthen the compliance education before HAART.

Abstract:Objective:To investigate the incidence and correlates of HIV-1 drug resistance among men who have sex with men(MSM)on highly active antiretroviral therapy (HAART). Methods:We conducted a prospective cohort study in Jiangsu province to estimate the incidence of HIV-1 drug resistance (HDR). Cox regression model was used to investigate the correlations of HDR. Results:During the follow-up period,23 MSM acquired drug resistance,and the incidence was 2.72 per 100 person-years. The independent risk factors of HDR were the CD4+T cell count (less than 200 copies/μl)before ART (HR:3.210;95%CI:1.333~7.733),and the use of nevirapine(NVP)on initial regimen (HR:2.852;95%CI:1.194~6.817). Conclusion:The incidence of HDR among MSM on ART was low. Early treatment of MSM infected with HIV-1 was recommended according to our study. It’s better to use EFV rather than NVP when initiate ART.
