      Abstract:Objective:To investigate if taurine has the protective effect on angiotensinⅡ-induced hypertension. Methods:AngiotensinⅡosmotic mini-pumps were implanted to mice subcutaneously to induce hypertension. Caudal arterial pressure was tested,and Hematoxylin-eosin staining and Masson staining were performed to determine the effect of taurine on hypertension and hypertension induced vascular remodelling;the mRNA levels of fibrotic markers were determined by RT-PCR;Western blot was performed to investigate the effect of taurine on molecular mechanisms of angiotensinⅡ-induced hypertension and hypertension induced vascular remodelling. Results:Taurine inhibited angiotensinⅡ-induced hypertension and consequent vascular remodelling,reduced the mRNA levels of fibrotic markers,and inhibited the MAPK pathway during the process. Conclusion:Taurine cowd inhibit MAPK pathway to attenuate angiotensinⅡ-induced hypertension and consequent vascular remodelling.

      Abstract:Objective:To investigate if glycine has the protection effect on warfarin induced aortic valve interstitial cells(AVICs)calcification. Methods:Porcine AVICs were isolated,calcium quantification,alkaline phosphatase activation and Alizarin red staining were performed to determine the effect of different concentrations of glycine on calcification,and the mRNA expression of osteogenic markers were determined by RT-PCR. Western blot,Flow cytometry and TUNEL assay were performed to investigate the anti-apoptosis effect on warfarin induced AVICs calcification. Results:Glycine attenuated warfarin induced AVICs calcification,reduced the mRNA expression of osteogenic markers,inhibited the Smad1,5/RUNX2 pathway and decreased the apoptosis of warfarin induced calcification. Conclusion:Glycine inhibits Smad1、5/RUNX2 pathway and decreases the early stage of apoptosis to attenuate warfarin induced AVICs calcification.

      Abstract:Objective:To explore the role and mechanism of miR-155 overexpression in the development of zebrafish embryos. Methods:The expression of miR-155 during each stage of embryogenesis was detected by quantitative RT-PCR. miR-155 mimic was microinjected to the 4 blastomeres of 4-cell stage embyos to overexpress the gene. The expressions of developmental related marker genes were detected by quantitative RT-PCR,and ETS1 mRNA was used to rescue embryos after overexpression of miR-155. Results:The expression of miR-155 was found in the whole embryonic development period and obviously increased in the periods of organogenesis. Overexpression of miR-155 leads to retardation of embryo development,delayed membrane depletion,abdominal and pericardial effusion,tail shortening and thickening,and down regulated the expression of genes related to liver,intestine,heart and muscle development. ETS1 can reduce the developmental malformation caused by overexpression of miR-155. Conclusion:Overexpression of miR-155 may affect embryonic development of zebrafish through negative regulation of ETS1.

    • Establishing a method for expansion of human regulatory T cells with high efficiency in vitro

      Abstract:Objective:To establish a method to isolate and culture regulatory T cells in vitro with high efficiency. Methods:CD4+CD25+CD127low T cells were isolated by fluorescence-activated cells sorting and cultured in vitro with anti-CD3-anti-CD28-coated microbeads and IL-2 in 2 weeks. Count cells numbers to evaluate the expansion ability,phenotype of expanded cells were identified with flow cytometry and the suppressive function was examined by mixed lymphocyte reaction. Results:A mean expansion of (755.5 ± 213.5) fold was obtained after cultured for 2 weeks. In addition,the expression of CD4,CD19,CD8,and CD25 in the expanded cells are(98.3 ± 1.04)%,(0.039 ± 0.021)%,(0.443 ± 0.239)% and(97.6 ± 1.35)% respectively. FOXP3+ cells accounts for(87.7 ± 5.5)% and Helios+ cells accounts for (73.3 ± 2.9)% of expanded cells. Cultured cells shows highest suppressive capacity with(84.39 ± 1.98)% suppression of activated PBMC at responder∶Treg ratio of 1∶1. Conclusion:The research provides a method to obtain large quantity of Tregs with high efficiency in vitro. The expanded cells remain their original phenotype and have ability of immune suppression which has great significance in cell therapy for treating autoimmune diseases and transplant rejections.

      Abstract:Objective:To study the expression of Netrin-1 in pancreatic tissue in diabetic mice and explore the significance in the development of type 2 diabetes mellitus. Methods:C57BL/6J mice were divided into the normal group and the diabetic model group. RT-PCR and Western blot were used to detect the mRNA and protein expression of Netrin-1 in pancreatic tissue in mice;Ins-1 cells was cultured in medium with normal and abnormal concentration of glucose. MTT was used to detect the cell viability. RT-PCR and Western blot were used to detect the mRNA and protein of Netrin-1 respectively. Results:The mRNA and protein expression of Netrin-1 increased in pancreatic tissue of diabetic mice. While high concentration of glucose up-regulated the mRNA and protein expression of Netrin-1 in ins-1 cells. Conclusion:The level of Netrin-1 increases in pancreatic tissue in diabetic mice which correlates with the abnormal concentration of glucose. Netrin-1 may participate in the pathophysiology of diabetes.

      Abstract:Objective:To investigate the expression of IQGAP3 in cholangiocarcinoma(CCA) and its clinical significance. Methods:RT-PCR and Western blot were performed to determine the levels of IQGAP3 in CCA tissues and CCA cell lines. Colone formation assay,CCK8 and EdU proliferation experiment were used to reveal the role of IQGAP3 in CCA cell proliferation in vitro. Western blot and phosphorylated proteinchip array were used to verify the possible mechanisms of IQGAP3 in regulating the proliferation of CCA cells. Results:IQGAP3 was overexpressed in the CCA tissues and cell lines. IQGAP3 promoted the proliferation of CCA cell lines in vitro. The expression of IQGAP3 was significantly correlated with tumor size,pathological grading and AJCC TNM staging in CCA patients combined with clinical sample data. Kaplan-meier survival analysis showed that the survival time of the CCA patients with high IQGAP3 expression was significantly lower than that those with low IQGAP3 expression. The expression level of IQGAP3 was positively correlated with the expression of WNK1. Conclusion:IQGAP3 was overexpressed in CCA and might promote the proliferation of CCA cells by up-regulating WNK1.

      Abstract:Objective:To explore the protective effect of fibroblast growth factor 21(FGF21)on liver ischemia-reperfusion(IR)injury in mice and the possible mechanisms. Methods:Thirty C57BL/6 mice were randomly divided into 3 groups:sham-operated group,IR+NS group(mice were injected with 2 mL/kg saline by tail vein just before ischemia induction)and IR+FGF21 group(mice were injected with 2 mL/kg FGF21 before operation). The mouse partial liver model of IR injury was established. Twelve hours after reperfusion,the mice were sacrificed and the serum and liver samples undergoing IR injury were collected. The ALT and AST levels in serum were determined. Liver histological damage was evaluated with hematoxylin-eosin(HE)staining and TUNEL detection. Levels of malondialdehyde(MDA),superoxide dismutase(SOD),glutathione(GSH)and reactive oxygen species(ROS)in liver tissue were also determined. Results:Liver tissues in IR+FGF21 group were more slightly damaged than those in IR+NS group(P < 0.01). The ALT and AST levels in IR+FGF21 group were significantly lower than those of IR+NS group(P < 0.01). The levels of plasma MDA and ROS contents in IR+FGF21 group were significantly lower than those of IR+NS group(P < 0.01). The levels of plasma SOD and GSH in IR+FGF21 group were higher than those of IR+NS group(P < 0.01). Conclusion:FGF21 can alleviate liver IR injury probably by inhibiting oxidant stress in mice.

    • Influence of miR⁃939 on NSCLC cell invasion and EMT process

      Abstract:Objective:The purpose of this study was to examine the expression and biological role of miR-939 in human non-small cell lung cancer(NSCLC)and epithelial-mesenchymal transition(EMT)progress. Methods:Quantitative real-time polymerase chain reaction(qRT-PCR)was used to detect the expression of miR-939 in NSCLC. Transwell analysis was used to determine the effect of miR-939 on cell invasion. EMT-related markers were detected by Western blot. Results:The expression of miR-939 in NSCLC tissues was significantly higher than that in adjacent non-cancerous tissues,and closely related to TNM staging and lymph node metastasis. Meanwhile,compared with normal lung epithelial cells,the expression of miR-939 in NSCLC cell lines was also significantly increased. The invasive ability of miR-939 cells was obviously decreased after down-regulation of miR-939 expression(P <0.05). In addition,miR-939 down-regulation caused E-cadherin increasing and Vimentin decreasing. Conclusion:miR-939 may serve as an adverse factor to promote cell invasion,thus affecting the development of NSCLC.

    • HER3 affects irradiation⁃induced migration and invasion in Luminal A breast cancer cells

      Abstract:Objective:To investigate the effect of knockdown of human epidermal growth factor receptor-3(HER3)expression on migration and invasion with irradiation(IR)in breast cancer cells and its molecular mechanism. Methods:Stable transfected breast cancer cells MCF-7 and ZR75-1 were established by short hairpin RNA(shRNA)to knockdown the expression of HER3 gene. The transfection efficiency was detected by Western blot. The control group transfected with empty virus and HER3 knockdown group were irradiated by 6-Gy X-ray. Cell healing assay and Transwell assay were used to analyze the migration and invasion ability of breast cancer cells. Western blot was used to detect the expression of Claudin-1. Results:After knockdown of HER3,HER3 protein expression in MCF-7 and ZR75-1 cells was significantly decreased. After IR,the migration and invasion increased,and the expression of Claudin-1 protein increased. However,knockdown of HER3 and combined IR reduced the migration and invasion of breast cancer cells,and decreased the expression of Claudin-1. Conclusion:Knockdown of HER3 reduces the migration and invasion of Luminal A breast cancer cells after IR. Its mechanism may be through the down-regulation of Claudin-1 expression.

      Abstract:Objective:To observe the effect of blocking electronic respiratory chain on the development of early female germ cells. Methods:We labeled normal,control and treatment group with germ cells associated markers:DDX4(DEAD Asp-Glu-Ala-Asp box polypeptide 4),γ-H2AX(the phosphorylated form of Histone H2AX),OCT4(POU domain,class 5,transcription factor 1)and STRA8(stimulated by retinoic acid gene 8) to assay the effects of the early female germ cells after blocking the electron respiratory chain by antimycin A(AMA). Results:12.0 dpc(days postcoitum)female mice gonads were cultured in vitro,and we found aberrant gonadogenesis after treatment with AMA,showing obviously decreased ATP(adenosine triphosphate)and increased ROS(reactive oxygen species),thus,leading to the abnormal meiosis initiation. Conclusion:The electron respiratory chain was highly linked with the gonads development in female mice.

    • The role of long noncoding RNA NANCI⁃NKX2.1 signaling pathway in lung tissue of neonatal mice with bronchopulmonary dysplasia

      Abstract:Objective:To investigate the role of NANCI-NKX2.1 signaling pathway in the process of bronchopulmonary dysplasia(BPD) formation in neonatal mice. Methods:A total of 32 neonatal mice(C57BL/6J) were randomly divided into 4 groups:interference group,negative control group,normal saline control group and blank control group,with 8 pups in each group. One microlitre of normal saline with lncRNA NANCI adenovirus and control adenovirus were administered intranasally to lungs of newborn mice in interference group and negative control group at postnatal days 2 respectively. One microlitre of normal saline was administered intranasally to lungs of newborn mice in normal saline control group .No interference treatment was processed in blank control group.The mice were sacrificed 7 days after birth and lung tissue samples were collected. Hematoxylin-eosin(HE)staining was used to observe pathological changes in lung tissues. RT-qPCR was used to measure the expression of NANCI. RT-qPCR and Western blot were used to measure the mRNA and protein expression of NKX2.1,SPC and AQP5. Results:HE staining in interference group showed thinner alveolar septum,increased alveolar cavity and decreased number of alveoli and septum. Lung tissue biopsy also showed decreased RAC values(P < 0.05).The relative expression of NANCI decreased(P < 0.05);the expression of NKX2.1,SPC and AQP5 mRNA and protein were decreased(P < 0.05). Conclusion:NANCI-NKX2.1 signaling pathway may play an important role in BPD neonatal mice.

    • The protective effect and mechanism of sevoflurane postconditioning on renal ischemia/reperfusion injury in mice

      Abstract:Objective:To explore the effect of sevoflurane postconditioning on renal ischemia/reperfusion injury,and to determine whether its protective mechanism is related to the anti-inflammation and anti-oxidation. Methods:Male C57BL/6 mice were randomly and equally divided into four groups:sham operation group(Sham group),sham operation+ sevoflurane postconditioning group(Sham+Sevo group),renal ischemia/reperfusion injury group(IR group)and renal ischemia/reperfusion injury + sevoflurane postconditioning group(Sevo-Post C group). The renal ischemia/reperfusion injury model was induced by left renal pedicle clamping for 30 min followed by 24 h reperfusion and right nephrectomy. The mice were inhaled 2% sevoflurane for 1 h at the start of reperfusion in Sevo-Post C group. After 24 h reperfusion,renal functional parameters,serum mediator concentrations and markers of oxidative stress in kidney tissues were determined,and renal histopathological analysis was performed. The protein including p-Akt,p-GSK3β,Akt and GSK3β were determined by Western blot. Results:Serum creatinine(Scr),blood urea nitrogen(BUN),TNF-α,IL-6,and MDA concentrations were significantly increased after renal I/R as compared with the sham group(P<0.05). Sevoflurane postconditioning reduced Scr,BUN,TNF-α,IL-6,and MDA levels(P<0.05),and decreased histological scores(P<0.05). The phosphorylation levels of renal Akt and GSK3β were elevated after renal ischemia/reperfusion injury,and sevoflurane postconditioning further increased the phosphorylation levels of them as compared with the IR group(P<0.05). Conclusion:Sevoflurane postconditioning protected against renal IR injury possibly through the anti-inflammatory and antioxidative effects induced by the activation of Akt/ GSK3β signialing pathway.

    • Establishment of the standardized lower limb ischemia model in rabbits

      Abstract:Objective:To explore the standard method of lower limbs ischemia model establishment in rabbits to provide a consistent condition for the subsequent study on the basis of ischemia. Methods:Twelve New Zealand rabbits were divided into 2 groups(Group I and Group F)according to a random number table with 6 rabbits in each. The rabbits were ligated at the lower segment of the left common iliac artery(cross of internal and external iliac artery)in the group I,or at the start level of left femoral artery(inguinal ligament)in the group F,respectively,followed by DSA to observe the lower limbs blood supply,and measurement of vastus medial shell temperature at the 3rd and 10th day of ligation. Results:No blood supply of branch artery was observed in group I whereas there were still branches from the external iliac artery to the lower limb(P < 0.05). Conclusion:Ligation at the lower segment of the common iliac artery can promise to block the blood supply for the lower limb,which can be used to establish a consistent lower limb ischemia model.

    • Analysis of fatty acid composition and content in different brain regions of rats by gas chromatography

      Abstract:Objective:To compare the differences of fatty acid composition and content in different brain regions of rats,and to provide experimental evidence for clarifying the distribution of fatty acids in the brain. Methods:Lipids were extracted from the brain regions with methanol/dichloromethane,and fatty acids were methylesterified using methanol/acetyl chloride. Fatty acid methyl esters were then assayed by gas chromatography. Results:Five brain regions including cerebellar vermis,striatum,hippocampus,cerebral cortex and white matter,contained at least 31 fatty acids. There were significant differences in the contents of the ten kinds of fatty acids(including C20∶0,C22∶0,C23∶0,C24∶0,C20∶1,C22∶1,C24∶1,C20∶5n-3,C22∶5n-6 and C22∶6n-3)among the five brain regions. Conclusion:The fatty acid compositions in the five brain regions of rats are basically the same and some fatty acid contents are significantly different.

    • Effects of type 2 diabetes on postoperative acute renal injury and other complications of off⁃pump coronery artery bypass grafting

      Abstract:Objective:To investigate the effects of type 2 diabetes on postoperative acute renal injury (AKI) and other complications of off-pump coronery artery bypass grafting(OP-CABG). Methods:Retrospectively analysed 894 cases of patients with OP-CABG from January 2012 to September 2017 in our hospital,including 318 patients with diabetes mellitus(DM)and 576 patients without DM. The statistical differences was analyzed using Graphpad Prism. Results:In total 894 patients of this study,there were significant differences in serum creatinine(SCr),glomerular filtration rate(eGFR),and creatinine clearance(CCr)after surgery between two groups. At 48h,SCr has the highest T-value(t=3.36)and P < 0.01. The intra-group mean of SCr was the highest at 12 h,and the deviation showed a significant increase,while the majority of patients fell back to preoperative level at 48h. Even in some patients,CCr,SCr and eGRF were smaller than preoperative values after coronary artery bypass surgery. Conclusion:Compared to patients without DM,patients with DM were more prone to AKI after OP-CABG,which appeared most at 12 h after surgery and had significant inter-group and intra-group difference in this time. Individual kidney showed a trend of deterioration or heal. In the two groups,the process of the AKI was not parallel to the process of systemic inflammatory response. The former healed at 48 h,while the latter continued to be aggravated.

    • Detection of colonic mucosal phosphorylated α⁃synuclein in patients with Parkinson’s disease and its clinical significance

      Abstract:Objective:To investigate the expression of phosphorylated α-synuclein(pSNCA)in colonic mucosa of Parkinson's disease(PD)patients with constipation and its relationship with non-motor symptoms. Methods:Ten constipated patients with PD(PDC)and 10 gender- and age-matched patients with functional constipation(FC)were enrolled in this study,and the clinical data and non-motor symptoms were evaluated. Two specimens of ascending and descending colonic mucosa were obtained from each subject by endoscopic biopsy. pSNCA and neurofilament protein(NF)were stained by immunohistochemistry and colocalized. Finally,the correlation between pSNCA and non-motor symptoms was conducted to explore the mechanism of non-motor symptoms in PD. Results:All PDC patients displayed pSNCA(10/10)in the colonic mucosa and 90%(9/10)in the control group,with no significant difference between the two groups. However,there was a statistically significant difference between the two groups in the expression of pSNCA(P<0.05),and the PDC group was more significant. The level of pSNCA in PDC patients was related to H-Y stage(r=-0.59,P<0.05),but not with disease duration,UPDRS-Ⅲ score and LEDD. The severity of constipation in PD was associated with UPDRS-Ⅲscore(r=0.67,P<0.05). In addition,the severity of non-motor symptoms(including anxiety,depression,sleep disturbances,fatigue,constipation)in patients was not related to the deposition of pSNCA in the intestine. Conclusion:The deposition of pSNCA may be involved in the development of constipation and Parkinson’s disease,but not corelate with the non-motor symptoms of Parkinson’s disease.

    • Association between estrogen receptor alpha gene PvuⅡ polymorphism and fragility fracture in postmenopausal women with heart failure

      Abstract:Objective:To evaluate the effect of the PvuⅡ polymorphism(rs2234693,C>T)of the estrogen receptor alpha(ESR1)gene on fragility fractures in elderly postmenopausal women with heart failure(HF). Methods:This was a hospital-based case-control study of postmenopausal women > 60 years of age with HF,including 80 fracture patients and 80 controls between January 2009 and January 2014. The PvuⅡ genotype was determined using a polymerase chain reaction-restriction fragment length polymorphism assay. Results:Fragility fracture was more common in smokers and those with a history of fragility fracture and less common in subjects who exercised daily. The PvuⅡ T allele was present in 83.75% of patients with fractures vs. 62.50% of controls. Carriers of the variant rs2234693T allele had increased risk of fracture(P < 0.05). Compared with the common genotype,the CT+TT rs2234693 genotype was associated with significantly increased risk of fracture(P=0.036,adjusted odds ratio=1.323,95% confidence interval=1.263~2.787). In stratified analyses,the association between the risk of fracture and the rs2234693 variant was more prominent in younger individuals(≤ 67 years)and non-smokers. Conclusion:The PvuⅡ polymorphism(rs2234693,C>T)of the ESR1 gene may contribute to the susceptibility of fragility fractures in elderly postmenopausal women with HF.

      Abstract:Objective:The aim of the study was to investigate the diagnostic value of glucose transporter 1(GLUT1) gene expression in differentiating malignant thyroid nodules from benign ones. Methods:Reverse transcription Quantitative Real-time PCR(RT-qPCR)was used to detect the expression of GLUT1 mRNA in 112 thyroid nodules(42 benign nodules and 70 malignant ones)and 52 peripheral normal thyroid cells of fine needle aspiration(FNA)specimens. Furthermore,immunohistochemistry(IHC)was used to detect the expression of GLUT1 protein in postoperative thyroid nodule tissues. Receiver operating characteristic(ROC)curve was plotted to evaluate the diagnostic performance of GLUT1 gene in diagnosing thyroid carcinoma. Results:The expression of GLUT1 mRNA can be detected in FNA specimens,and it was in line with the expression of GLUT1 protein in IHC. PCR results showed that there was no significant difference in the expression of GLUT1 mRNA between benign nodules and normal thyroid cells(P > 0.05). Compared to benign nodules,the expression of GLUT1 mRNA in thyroid cancer increased significantly(P < 0.001). The sensitivity,specificity and area under the curve(AUC)of GLUT1 gene in the diagnosis of thyroid cancer were 62.9%,92.9% and 0.797,respectively. The expression of GLUT1 gene in thyroid cancer was not correlated with sex,age,tumor size,number of nodules,BRAFV600E mutation or lymph node metastasis(P > 0.05),but significantly correlated with AJCC staging(P < 0.001). Conclusion:GLUT1 gene detection had high specificity in diagnosing thyroid cancer,making it an important reference for the preoperatively differentiation of benign and malignant lesions.

      Abstract:Objective:To observe the effect of negative balance fluid therapy during early postoperative phase on patients undergoing liver transplantation. Methods:One hundred and sixteen patients who received orthotropic liver transplantation in the hospital were randomized into an experimental group(60 cases)receiving negative balance fluid therapy and a control group(58 cases)receiving routine venous transfusion . We observed the recovery of liver function after surgery,the time of the postoperative recovery,the length of hospital stay, the volumes of patients’ ascites,pulmonary complications and other postoperative adverse events. Results:In the experimental group,the time of the postoperative recovery and the length of hospital stay were shorter than those in the control group,the volumes of patients’ ascites and pulmonary complications were significantly less than those in the control group. Liver function after surgery is better than control group. All the differences reached statistical significance(P < 0.05). Conclusion:To the patients,negative balance fluid therapy is safe and effective,and this measure can accelerate the postoperative recovery of these patients. This measure can play an active role in ERAS (enhanced recovery after surgery) treatment.

    • A retrospective analysis of clinical characteristics and prognostic factors for 51 cases of follicular lymphoma grade 3A and grade 3B

      Abstract:Objective:The aim of this study was to analyze the clinical characteristics,treatment response and outcomes of follicular lymphoma(FL)grade 3A and grade 3B. Methods:Data of 51 patients,diagnosed as FL grade 3,were collected. Statistical analysis was performed to analyse the differences in clinical characteristics and outcomes. Results:Out of 51 FL3 cases,including FL3A in 34 cases(67%)and FL3B in 17 cases(33%),20 cases had diffuse large B cell lymphoma(DLBCL)component. The percentage of FL3B patients with DLBCL component was significantly higher than that of FL3A patients. Correlation analyses showed that FL3A was significantly associated with BCL2 positive expression(P=0.046),whereas FL3B was significantly associated with MUM1 positive expression(P=0.002)and early Ann-Abor staging(P=0.046). All patients received first-line immunochemotherapy includes rituximab and chemotherapy. There was no significant difference between the two groups in response to treatment. Survival analyses showed that FL3B was an independent risk factor for progression free survival(PFS)in FL3 patients(P=0.048,HR=3.271,95% CI:1.012~10.573). However,there was no significant difference between the two groups in terms of overall survival(OS). FL3 co-existence with DLBCL component predicted shorter PFS(P=0.003)and OS(P=0.04). The co-existence of DLBCL component was an independent prognostic factor for PFS and OS in FL3 patients(PFS:P=0.012,HR=4.291,95% CI:1.373~13.405;OS:P=0.013,HR=2.392,95% CI:1.199~4.771). Conclusion:FL3A and FL3B had significant differences in clinical characteristics and outcomes. FL3B patients had shorter PFS. Moreover,FL3 coexistence with DLBCL component suggested poor prognosis.

      Abstract:Objective:To investigate the correlation between serum 25-hydroxyvitamin D,serum parathyroid hormone levels and the biomarkers of heart failure (BNP and sST2) in patients with HFrEF. Methods:A total of 152 patients with HFrEF who were admitted to the Department of Cardiology of Nanjing Drum Tower Hospital from September 2017 to February 2018 were selected and 31 people with normal heart function was selected as the control group. levels of 25-hydroxyvitamin D were measured using electrochemiluminescence,parathyroid hormone levels were measured by chemiluminescence. The condation of chronic heart failure and lack of vitamin D,parathyroid hormone levels was analyzed. Results:Compared with healthy controls,patients with heart failure had significantly lower 25-hydroxyvitamin D levels and significantly increased PTH levels(P < 0.001). Patients in HF with different 25-hydroxyvitamin D levels had a significant difference in heart function classification(P=0.003). Patients with 25-hydroxyvitamin D deficiency had increased BNP and sST2 levels significantly,with statistical significance(P=0.016,P < 0.001);Correlation analysis revealed that NYHA grades,BNP levels,sST2 levels were negative associated with 25-hydroxyvitamin D,VD/PTH respectively and positively correlated with PTH. Logistic multivariate regression analysis of independent risk factors for heart failure revealed that both 25-hydroxyvitamin D and PTH levels were independent risk factors for heart failure. Conclusion:The 25-hydroxyvitamin D levels in patients with HFrEF are lower than those in healthy people,and serum parathyroid hormone levels are higher than those in healthy people. The level of 25-hydroxyvitamin D in patients with heart failure was negatively correlated with the severity of cardiac function. PTH levels were positively correlated with the severity of heart function. 25-hydroxyvitamin D/PTH levels were negatively correlated with the severity of heart function;and 25-hydroxyl vitamin D and PTH levels are independent risk factors for heart failure.

    • The influence of different treatments on the root surface of teeth with severe periodontitis

      Abstract:Objective:This study aimed to evaluate the treatment of four different methods on root surfaces of teeth affected by severe periodontal disease,adhesion and proliferation of Porphyromonas gingivalis(P. gingivalis)and human periodontal ligament cells(hPDLCs)on root surfaces. Methods:Severe periodontal affected teeth(n=100)were extracted and treated with scaling and root planning(SRP),and root slices were prepared. These slices were randomly divided into four groups:SRP group,Periocline group,EDTA group and Nd:YAG group. Scanning electron microscopy(SEM)and atom force microscope(AFM)were used to determine the morphology and the roughness of the root slices. The proliferation of P. gingivalis and hPDLCs was evaluated by cell count kit-8(CCK-8). Results:The obvious smear layer was observed in SRP group,whereas it was almost removed in other three groups. The roughness of the root surface of Periocline group was the lowest while the Nd:YAG group was the highest. After 4 hours and 1 day,the amount of P. gingivalis adhered to the root surfaces of Periocline group and Nd:YAG group was significantly less than that to other groups. The attachment of hPDLCs to the root surfaces showed a time dependent manner,and the number of attached cells was higher in Periocline group and Nd:YAG group. Conclusion:SRP combined with Periocline or Nd:YAG laser is more conducive to the attachment and proliferation of hPDLCs,and decrease the adhesion of P. gingivalis at the early stage.

The expression of FoxO1 is closely related to tumorigenesis,development and prognosis. FoxO1 is widely expressed in human tissues such as lung,heart,liver,pancreas,skeletal muscle,testis and ovary.       Abstract:Recombinant human endostatin(rh-Endostatin) is an anti-angiogenic targeting drug with multi-targets. The mechanisms of rh-Endostatin include not only regulating the surface proteins of endothelial cells and intracellular signaling pathways at the microscopic molecular level,but also remodeling the microenvironment of tumors at macro level,and then improving the overall therapeutic effect of tumors. Numerous studies have shown that anti-angiogenic therapy(such as rh-Endostatin) can work together with chemo-radiotherapy,immunotherapy,and other targeted therapies to combat tumors. Our work will give a detailed review of the mechanisms and application of rh-Endostatin and its research progress in different tumors.
