    • The immunoregulation effect of n⁃3 polyunsaturated fatty acids on T cells in NOD mice

      Abstract:Objective:This study aims to observe the effects of n - 3 poly unsaturated fatty acids(n - 3 PUFA)on immunological functions of T cells in NOD mice. Methods:Spleen cells were separated from Balb/c WT mice and NOD mice. The CD4+ T cells were obtained from magnetic bead separated spleen cells and divided into four groups:wild type group,NOD mice untreated group(control), NOD mice treated with(docosahexaenoic acid,DHA)and(eicosapentaenoic acid,EPA)groups. The NOD mice T cells were treated with EPA and DHA respectively for 24 h. After the stimulation and activation of PMA and lonomycin,the viability of T cells was tested by CCK-8 kit,the differentiation of Th cells was analyzed by flow cytometer,and the secretion of inflammatory cytokines was measured by ELISA Kits. Results:EPA and DHA could inhibit the viability of T cells from NOD mice,suppress the naive T cells’differentiation into Th1 cells,balance the ratio of Th1 and Th2,and down-regulate the secretion of IL-6 and IL-17. Conclusion:n-3 PUFAs could suppress some immune functions of T cells by inhibiting their viability,regulating the Th1/Th2 polarization and inhibiting the secretion of IL-6 and IL-17.

      Abstract:Objective:This study aims to investigate the effect of integrated stress response inhibitor(ISRIB)in apoptosis and endoplasmic reticulum stress of SH-SY5Y cells induced by amyloid β-amyloid protein1-42(Aβ1-42)and to clarify its mechanism. Methods:SH-SY5Y cells were divided into normal control group,Aβ group,ISRIB group and ISRIB+Aβ group. The survival rate of cells in each group was detected by MTT colorimetric assay. Western blotting was used to examine the expression of endoplasmic reticulum stress marker glucose-regulated protein 78(GRP78)and activation of protein kinase R-like ER kinase(PERK)-eukaryotic initiation factor 2α(eIF2α)-activation transcription factor 4(ATF4)signaling pathway,together with C/EBP homologous protein(CHOP). Meanwhile,apoptosis-related proteins were also detected,including Bcl-2、Bax and cleaved-caspase3. Results:Compared with Aβ group,ISRIB significantly increased the survival rate of SH-SY5Y cells(P < 0.01),significantly decreased the activation of p-PERK(P < 0.01),p-eIF2α(P<0.05),ATF4(P < 0.05)and CHOP(P < 0.05),increased Bcl-2/Bax level(P < 0.05)and decreased cleaved-casepase3 expression(P < 0.01). Conclusion:ISRIB could inhibit the apoptosis of SH-SY5Y cells induced by Aβ1-42,and its mechanism is related to the inhibiting the activation of eIF2α pathway and apoptotic signaling pathway induced by ERS.

    • Effects of 11β⁃HSD1 inhibitor—BVT.2733 on the expressions of specific genes related to metabolism and inflammation of multiple organs in obese mice

      Abstract:Objective:This study aims to to assess the effect of BVT.2733,a selective 11β-hydroxysteroid dehydrogenase type 1 inhibitor,on energy metabolism and inflammation-related gene expression in hepatic,skeletal muscular,cardiac muscular,renal and splenic tissues in C57BL/6J mice. Methods:C57BL/6J mice were fed at a normal fat diet(NC)or high fat diet(HFD). HFD treated mice were then administrated with BVT.2733(HFD+BVT)or vehicle(HFD). We examined the effects of BVT.2733 on energy metabolism-related genes and typical inflammatory genes expression in hepatic,skeletal muscular,cardiac muscular,and renal tissues by RT-PCR. Results:The expression of glucose consumption-related genes such as UCP2 and GLUT4 increased significantly,while mRNA level of PDK2 and PDK4,as the negative regulator of glucose oxidation,increased significantly in the skeletal and cardiac muscle of HFD+BVT mice. In liver,11β-HSD1 inhibitor significantly reduced the key genes expression of lipid synthesis,including SREBP and FAS. Interestingly,11β-HSD1 inhibitor also significantly inhibited the critical glucose reabsorption genes expression of SGLT1 and SGLT2 in kidney. On the other hand,there was significant inhibition of the typical inflammatory gene,such as TNF-α,MCP1,IL-6 and IL-5,both in skeletal and cardiac muscles. Conclusion:BVT.2733 had a remarkable effect on the expressions of specific genes related to metabolism and inflammation in skeletal muscular,cardiac,hepatic,renal and splenic tissues in obese mice.

    • Role of PARP⁃1 in attenuation of myocardial ischemia⁃reperfusion injury treated by dexmedetomidine in rats

      Abstract:Objective:This study aims to evaluate the role of poly(ADP-ribose)polymerase-1(PARP-1)in attenuation of myocardial ischemia-reperfusion injury treated by dexmedetomidine in rats. Methods:Forty eight healthy male Sprague Dawley rats,8~12 weeks old,weighing 200~240 g were selected. The rats were then randomly divided into 3 groups(n=16):Sham operation group(S group),lung ischemia-reperfusion group(I/R group)and I/R+dexmedetomidine group(I/R+Dex group). In S group,the anterior descending branch was only exposed but not ligated;Myocardial I/R was induced by occlusion of anterior descending branch of left coronary artery for 30 min followed by 120 min of reperfusion in I/R group and I/R+Dex group. Dexmedetomidine 5 μg/kg were injected intraperitoneally 15 min before reperfusion in I/R+Dex group while physiological saline was injected in S and I/R groups. The mean arterial pressure(MAP),heart rate(HR)and heart rate systolic blood pressure product(RPP)were observe and recorded before ischemia(T0),30 min of ischemia(T1) and reperfusion of 60 min(T2) and 120 min reperfusion(T3). At the end of 120 min reperfusion,hearts were removed for determination of the myocardial infarct size in the left ventricular myocardial tissues. The expression of PARP-1 activity markers(PAR)and apoptosis related protein Caspase-3 in myocardial tissue were investigated by Western blot. TNF-a and IL-6 levels were examined by ELISA. Results:Compared with the time point of T0,the MAP,HR and RPP were significantly decreased at time point of T1-T3 in I/R group and I/R+Dex group(P<0.05). Compared with S group,MAP,HR and RPP were significantly decreased at time point of T1-T3,myocardial infarct size,TNF-a and IL-6 levels,PAR and Caspase-3 expressions were increased in I/R group and I/R+Dex group(P<0.05). Compared with I/R group,MAP,HR and RPP were significantly decreased at time point of T1-T2 while increased at T3,myocardial infarct size,TNF-a and IL-6 levels,PAR and Caspase-3 expressions were decreased in I/R+Dex group(P<0.05). Conclusion:Dexmedetomidine can attenuate myocardial ischemia-reperfusion injury.The mechanism may be associated with reducing the activation of PARP-1,and then inhibit inflammation and apoptosis in myocardial tissue.

      Abstract:Objective:This study aims to elucidate the role and molecular mechanism of JWA in regulating cell energy metabolism and inhibiting the migration of pancreatic cancer cells. Methods:Human pancreatic cancer Panc1 and Bxpc3 cells were treated with the JWA small molecule agonist JAC4. The assays of wound healing and transwell were used to determine cell migration;the mitochondrial energy metabolisms of cells were measured by Seahorse XF energy metabolism analyzer. The pancreatic cancer cells Panc1 were used to establish series vivo metastatic mice models. Western blotting was performed to determine the levels of protein expression in related molecules. Results:The mRNA expressions of JWA was significantly lower in pancreatic cancer tumor tissues and pancreatic cancer cells compared with that in adjacent normal tissues and normal pancreatic cells;JAC4 inhibited migrations of pancreatic cancer Panc1 and Bxpc3 cells,and inhibited pancreatic cancer metastasis in mice and reduces serum LDH levels. The mechanistic evidences indicated that JAC4 positively regulates FOXO3a through AMPK signaling pathway led to the activation of mitochondrial complex Ⅲ(UQCRC2),and enhanced the oxidative phosphorylation;at the same time,JAC4 suppressed glycolysis of pancreatic cancer cells. Conclusion:The inhibiting role of JAC4 on pancreatic cancer cells was realized by mitochondria metabolic reprogramming through the JWA/AMPK/FOXO3a/UQCRC2/FAK signaling.

    • The relationship of CCR1 expression in hepatocellular carcinoma tissue and the prognosis of patients

      Abstract:Objective:This study aims to investigate the expression of C -C motif chemokine receptor 1(CCR1)protein in human hepatocellular carcinoma(HCC)and its correlation with clinicopathological factors and prognosis. Methods:Immunohistochemistry (three - step method)was used to detect the expression of CCR1 protein in tissue microarray constructed by 288 cases of HCC. Univariate and multivariate Cox regression models were performed on follow-up data. Results:The expression of CCR1 in HCC tissues was significantly higher than that in adjacent tissues. The tumor-free survival and overall survival of CCR1 positive expression group were significantly lower than those of CCR1 negative expression group. Multivariate Cox regression models showed that positive expression of CCR1 was an independent risk factor for tumor-free survival and overall survival after surgery. Conclusion:Detection of CCR1 expression in HCC is helpful for the prediction of postoperative recurrence

    • AIM2 expression predicts response of rectal cancers to pre⁃operative chemoradiotherapy and its relationship to prognosis

      Abstract:Objective:This study was aimed to determine whether Melanoma 2(AIM2)expression predicts response to neoadjuvant chemoradiotherapy (nCRT) and outcomes for patients with locally advanced rectal cancer (LARC). Methods:The study examined the AIM2 expression in 122 pre-treatment biopsies of LARC by immohistochemistry(IHC). The associations between AIM2 and clinical characters were analyzed. Results:High expression of AIM2 in tumor cells was observed in 26 biopsy specimens. There was no significant correlation between high AIM2 expression in tumor cells and gender(P=0.316),age(P=0.283),differentiation(P=0.356),CEA serum level(P=0.468),tumor location(P=0.309),clinical tumor status(P=0.284),and clinical node status(P=0.264). There were 65(67.7%) specimens with high expression of AIM2 in 96 postoperative specimens(P < 0.001). Univariate and multivariate analysis showed that AIM2 high expression in tumor cells could predict LARC resistance to nCRT:25/26(96.2%)cases with AIM2 high expression tumor showed poor response,while 78/96(81.3%)cases with AIM2 low expression tumor showed good response(P < 0.001). After a median follow-up time of 51 months,multivariate analysis showed that high-expression of AIM2 in tumor cells was not correlated with patients’ recurrence free survival(RFS)(P=0.634)or overall survival(OS)(P=0.932). Conclusion:AIM2 expression levels might be a useful indicator of nRCT sensitivity for LARC. The administration of AIM2 signaling pathway antagonist may have the potential to increase rectal cancer radiosensitivity.

    • High expression of ZNF692 is a predictor of poor prognosis in patients with gastric cancer

      Abstract:Objective:This study aims to investigate whether Zinc finger protein 692(ZNF692)can be used as an independent predictor of poor prognosis in patients with gastric cancer. Methods:Immunohistochemical(IHC)staining was used to detect the expression of ZNF692 in gastric cancer tissues and adjacent tissues of 191 cases. Results:The expression of ZNF692 in gastric cancer tissues was significantly higher than that in adjacent tissues. There was no significant difference in the clinicopathological features between patients with high ZNF692 expression and those with low ZNF692 expression(P > 0.05). However,overall survival(OS)of patients with ZNF692 high expression was significantly shorter than that of patients with ZNF692 low expression(P=0.000 1). The mean survival time of patients with ZNF692 high expression(30.1 months)was significantly shorter than that of patients with ZNF692 low expression(42.6 months)(P < 0.01). Cox multivariate risk regression model analysis showed that ZNF692 could be used as an independent predictor of poor prognosis in patients with gastric cancer. Conclusion:High expression of ZNF692 is one of poor prognosis predictors in patients with gastric cancer.

    • Analysis of overall survival and prognostic factors in elderly biliary tract cancer patients treated with radical resection

      Abstract:Objective:This studt aims to explore the prognostic factors in elderly operable biliary tract cancer patients treated with radical resection. Methods:The clinicopathologic and follow-up data of 143 elderly patients(>65 years old)with biliary tract cancer treated with radical resection from January 2011 to December 2015 were analyzed retrospectively. Results:Univariate analysis showed that depth of tumor invasion,lymph node involvement,histological differentiation,lymphovascular invasion,resection margin,lactate dehydrogenase(LDH)level were correlated to recurrence and metastasis of biliary tract cancer after radical resection. Multivariate analysis showed that depth of tumor invasion,histological differentiation and LDH level were independent factors affecting the prognosis of elderly biliary tract cancer patients. Conclusion:The overall survival of elderly operable biliary tract cancer patients was correlated with tumor invasion, histological differentiation and LDH level before radical resection.

      Abstract:Objective:This study aims to compare differences in the incidence and severity of non-motor symptoms in Parkison’s disease(PD) patients with different motor phenotypes,and to further study the influencing factors. Methods:From October 2016 to April 2018,91 patients were diagnosed as primary PD in Jiangsu Subei People’s Hospital were included in this research. PD patients were administrated with non-motor scales and motor scales,including non- Motor Symptoms Questionnaire(NMSQ),Autonomic Symptom Scale(SCOPA-AUT),Hamilton Depression Rating Scale(HDRS),Pittsburgh Sleep Quality Index(PSQI),MoCA,and Mini-Mental State Scale(MMSE),the unified Parkinson’s Disease Rating Scale(UPDRS)Ⅱ and Ⅲ,according to UPDRS Ⅱ and Ⅲ score,all patient were divided in to 12 cases of tremor type(TD),53 cases of postural instability and gait difficulty(PIGD)and 16 cases uncertain type,the differences of scores in the non-motor scales between the TD group and PIGD group were compared,and the influencing factors of the differences were further analyzed. Results:There were no significant differences in age,gender,duration of disease,MMSE,HDRS,FSS,and PSQI scores between the two groups(P > 0.05),education level(P=0.031),age of onset(P=0.038),and H-Y grade(P=0.011),UPDRS-Ⅲ total score(P=0.011),SCOPA-AUT score(P=0.019),NMSQ score(P=0.009)were statistically different. And the influencing factors of the MoCA score,NMSQ score and SCOPA-AUT score was further analyzed,the education level and UPDRS-Ⅲ total score were the influencing factors of MoCA score,P value was <0.001,0.003,respectively;HDRS score and H-Y scale are the influencing factors of SCOPA-AUT score,P value was < 0.001 and 0.044;Age,HDRS score,FSS score and UPDRS-Ⅲ total score are the influencing factors of NMSQ score,P value was 0.031,<0.001,0.008,0.005,respectively. Conclusion:The age,severity of depression,fatigue and motor symptoms are influencing factors of non-motor symptoms,the occurrence of depression and H-Y scale are the influencing factors of the occurrence of autonomic nervous symptoms,and the level of education and the severity of motor symptoms are the influencing factors of the cognitive impairment.

      Abstract:Objective:To analyze the differential expression of exosomal miRNAs deriving from plasma between recurrent spontaneous abortion(RSA)and healthy-control subjects. Methods:This study enrolled 3 patients with RSA who were admitted to Nanjing Maternity and Child Health Care Hospital,and a cohort of 3 age and gestation-matched healthy-control subjects. Peripheral blood samples were collected on the first day after curettage surgery. Trancmission electron microscopy was used to identify exosomes derived from plasma samples. Sequencing was performed to identify the differentially expressed exosomal miRNAs between the RSA group and the control group. Target gene prediction,gene ontology analysis,and pathway analysis were performed for miRNAs with significant differential expression. All subjects provided written informed consent for their participation in the study. Results:By comparing with the control group, 24 significantly differential expressed exosomal miRNAs were screened in RSA group,of which 11 were down-regulated and 13 up-regulated. Conclusion:The exosomes contain abundant miRNAs which expressions are significantly different. These exosomal differential miRNAs may be a promising biomarker in the activation of RSA,needing further investigation.

    • The medium-term outcomes of 3D printed customized porous titanium acetabular prosthesis replacement for acetabular bone defection

      Abstract:Objective:This study aims to explore the feasibility,safety,efficacy and medium-term outcomes of 3D printed customized porous titanium acetabular prosthesis in treating acetabular bone defection. Methods:Twelve patients with acetabular bone defection[8 male and 12 female,with a mean age of(62.25±12.21)years,range 42-82 years]were treated with 3D printed customized porous titanium acetabular prosthesis from January 2016 to September 2017. The 3D printing techniques was used for reconstructing and designing the optimal acetabular prosthesis for the personalized acetabular bone defection of each patient. The acetabular prosthesis was subjected to porous processing. The 3D printing of porous titanium acetabular prosthesis and post-processing was subsequently performed. The acetabular prosthesis was implanted through the lateral approach of hip joint. The operative duration,intraoperative blood loss and complications were recorded. All patients were followed up postoperatively. Pain was assessed using the visual analog scale(VAS),and the hip joint function was evaluated using the Harris score and gait analysis. The patients underwent anterior-posterior radiography and computer tomography of the hip joint aiming to observe the restoration of lower limb alignment and the position of the acetabular prosthesis. Results:The mean operative duration of 12 patients was(2.23±0.7)h(1.1-3.5 h),and the mean blood loss was(406.67±219.29)mL(150-820 mL). All patients were followed up for 12 to 26 months,with an average of 15.33 months. All patients were satisfied with functional restoration. The VAS scores ranged from 5.17±1.53 preoperatively to 1.67±0.78 postoperatively(t=9.225,P < 0.001),and Harris scores ranged from 47.67±7.15 preoperatively to 86.75±4.52 postoperatively(t=-27.380,P < 0.001)with statistically significant difference,respectively. The image suggested good restoration of lower limb alignment and position of the acetabular prosthesis,and a close contact between the titanium acetabular prosthesis and acetabular bone defection without loosening. Conclusion:The 3D printed customized porous titanium acetabular prosthesis could reconstruct the defect of acetabular and increase the initial stability of hip joint. The method could reduce the difficulty of the hip reconstruction,and the patients could obtain good hip joint function.

    • Value of leukocyte and procalcitonin on the prognosis of cardiac arrest due to acute non-infectious diseases in the early stage

      Abstract:Objective:This study aims to study the value of leukocyte and procalcitonin on the prognosis of cardiac arrest caused by acute non-infectious diseases in the early stage. Methods:Forty-six patients with cardiac arrest admitted to our hospital from November 2016 to May 2019,who returned to spontaneous circulation by cardiopulmonary resuscitation(CPR)without acute infectious diseases,were selected. They were divided into two groups according to the survival at 30 d after cardiac arrest or not. The correlation between the prognosis and the changes of leukocyte and procalcitonin after cardiac arrest was analyzed statistically. Results:①There was significant difference in the levels of leukocyte and procalcitonin between the two groups in the initial 48 hours after cardiac arrest;② For the death group,the level of leukocyte increased immediately after cardiac arrest,reached the peak around 24 h and decreased in 48 h;③The level of procalcitonin of the death group also increased to the peak about 24 h after cardiac arrest. Conclusion:The level of leukocyte and procalcitonin increased significantly after cardiac arrest. The large increase of leukocyte in 24 h after cardiac arrest may indicate a poor prognosis. However,the procalcitonin level in the early stage and its increase cannot predict the death risk of patients within 30 days after cardiac arrest.

    • Durability evaluation of zirconia ceramic containing phosphate ester monomer bonded to resin

      Abstract:Objective:This study aimed to evaluate the durability of resin bonding to an yttria-stabilized tetragonal zirconia polycrystals(Y-TZP)using two different primers containing phosphate ester monomer [10-methacryloyloxydecyl dihydrogen phosphate(MDP)]. Methods:A total of 80 zirconia plates were prepared and assigned to 4 groups to build bonded specimens according to surface treatments(sandblasted with alumina particles;sandblasted with alumina particles followed by application of Z-Prime Plus or Clearfil Ceramic Primer;tribochemical silica coating followed by application of silane). Bonded specimens were built using resin luting cement and tested for shear bond strength(SBS). X-ray photoelectron(XPS)was used to investigate the formation of chemical bond on the surface of Y-TZP after conditioned with two types of MDP containing primers. Release of the element phosphorus from MDP-mediated Y-TZP/resin-bonded interfaces were evaluated via inductively coupled plasma-mass spectrometry(ICP-MS). Results:The two types of MDP containing primers increased the SBS in short time,compared to negative control group,the tribochemical silica coating group obtained the highest SBS. After thermocycling,no statistical difference was observed in tribochemical silica coating group,but SBS decreased significantly in groups sandblasted and conditioned with two types of MDP containing primers(P < 0.05). XPS analysis detected -P-O-Zr bond on the surface of Y-TZP conditioned with MDP-containing primers. Phosphorus was detected by ICP-MS from the solutions used for immersed the Y-TZP/resin-bonded specimens. Conclusion:Water negatively affects bonding between zirconia and resin mediated by MDP-containing primers.
