    • Effects of rapamycin on ovarian function in obese mice

      Abstract:Objective:This study aims to observe the intervention effect of rapamycin on ovarian dysfunction caused by maternal obesity and evaluate the safety of drugs. Methods:A female obesity mice model with high fat diet was established and then randomly divided into the treatment group injected with rapamycin and the control group injected with PBS. After 24 weeks,the number of ovulations was counted and the oocytes were collected for spindle operation. Immunofluorescence staining was used to evaluate the effect of rapamycin on ovulation function and oocyte quality in obese mice,and the expression of ovarian-related genes in protein and mRNA was detected by Western blot and RT-qPCR. Results:The weight of ovarian in rapamycin treated mice decreased. Expression of phosphorylated ribosoma lprotein(P-rps6) and the phosphorylated 4E binding protein1P(P-4EBP1) was significantly decreased in rapamycin-treated obese mice,and the number of superoved oocytes increased compared with the control. But results of the oocyte spindle staining showed no significant difference in oocyte quality between two groups. The mRNA expression level of tumor necrosis factor-α(TNF-α) in the ovary was increased after rapamycin treatment. There were no significant changes in serum estrogen and follicle stimulating hormone between two groups. Conclusion:Rapamycin has a tendency to improve ovulation function by inhibiting the mTOR signaling pathway. It was not found that rapamycin had a significant effect on oocyte quality. This study provided experimental basis for research of rapamycin on fertility in obese women in the future.

    • Effects of hyperandrogen on human embryonic stem cells induced to differentiate into hypothalamic neural progenitors

      Abstract:Objective:In this study,human embryonic stem cells(hESCs) were induced to differentiate into hypothalamic neural progenitor cells in vitro,and the effects of different concentrations of androgen on the differentiation of hypothalamic neural progenitor cells were compared. Methods:The quality of human embryonic stem cell line CCRM22 was identified,CCRM22 was induced to differentiate into hypothalamic neural progenitor cells in vitro,then 1×10-8 mol/L and 1×10-7 mol/L testosterone were added to the differentiation medium(anhydrous ethanol as cosolvent control). The differentiated cells were collected at different stages of differentiation,and the differentiation efficiency was compared. The differentiated cells were identified by RT-qPCR,flow cytometry and immnofluorescence,and the expression of genes related to reproductive function regulation was detected. Results:The differentiation efficiency of CCRM22 into hypothalamic neural progenitor cells was more than 85%,but testosterone treatment decreased its differentiation efficiency;differentiated cells expressed nerve cell marker NESTIN and hypothalamic neural progenitor cell specific marker NKX2.1,expressed both KISS1 and androgen receptor(AR),and the expression of AR was positively correlated with testosterone concentration. Conclusion:Human embryonic stem cells were successfully induced into hypothalamic neural progenitor cells. High concentration of testosterone inhibited the differentiation of hESCs into hypothalamic neural progenitor cells. This cell model can be used to study the effects of hyperandrogenic environment in early pregnancy on hypothalamic nerve cell differentiation and development of offspring in women with polycystic ovary syndrome in vitro.

      Abstract:Objective:This study aims to observe the role of small RNA 2’-O-methyltransferase Henmt1 during mouse spermatogenesis by generation of Henmt1 mutant mouse model. Methods:Henmt1 mutant mouse were constructed by CRISPR/Cas9 system(clustered regularly interspaced short palindromic repeats and CRISPR-associated protein 9)and analyzed the phenotype of mutant mice by computer-aided sperm analysis,HE staining and immunofluorescent staining. Results:Henmt1 mutant mouse model were successfully generated. Henmt1 mutant males are sterility with spermatogenic arrest and up-regulation of retrotransposon LINE1. Conclusion:This study found that HENMT1 plays an important regulatory role during spermatogenesis.

      Abstract:Objective:This study aims to investigate the expression pattern of heat shock protein DNAJB8[DnaJ heat shock protein family(HSP40)member B8] in spermatogenesis. Methods:Quantitative real-time PCR and Western blot were used to determine the transcript levels and protein abundance of DNAJB8 in various mouse tissues,testes at different developmental stages and different germ cell populations. The subcellular localization of DNAJB8 was further assessed by immunofluorescence analysis. Results:DNAJB8 was specifically expressed in the mouse testis. The transcripts of DNAJB8 mRNA increased significantly at postnatal day 21(P21),when round spermatids appeared. However,the protein of DNAJB8 was detected until in P35 mouse testis,where elongating spermatids were highly enriched at this stage. Immunostaining on frozen adult mouse testis and spermatozoa showed that DNAJB8 was observed in late stages of spermatids,and its signal was also present in middle-piece flagellum of spermatozoa. Conclusion:DNAJB8 may function in the elongation of round spermatids during late stages of spermiogenesis.

      Abstract:Objective:In order to detect the putative off-target effects of CRISPR/Cas9 system,the knockout mouse model of RNA methyltransferase Nsun5 gene were constructed using wild-type Cas9 and nickase Cas9-D10A,respectively,and performed off-target analysis of F0 mice to compare the specificity of the two strategies. Methods:Paired sgRNAs targeting the third exon of Nsun5 were designed and co-injected with Cas9 mRNA and Cas9-D10A mRNA respectively into mouse one-cell fertilized eggs to generate Nsun5 knockout founders. Off-target effects were analyzed in founder mice injected with CRISPR/Cas9 and Cas9-D10A,respectively. Results:CRISPR/Cas9 and Cas9-D10A were used to establish the Nsun5 gene knockout mouse model successfully. The founder mice obtained from the two variants were subject to off-target analyzation,and no off-target sites were detected. Conclusion:Nsun5 gene knockout models were successfully generated by the both strategies without off-target sites,paving the way for further studying the biological functions of Nsun5.

      Abstract:Objective:This study aims to explore the influence and mechanism of mechanical properties of extracellular matrix on biological behaviors of oral squamous carcinoma cells. Methods:Two kinds of oral squamous carcinoma cells,CAL27 and HSC3,were cultured on hydrogel with different mechanical properties. Cell morphological changes were observed by microscope. The location and expression of laminin β2(LAMB2),collagen VI α1(COL6A1) and E-cadherin(CDH1) were detected by immunofluorescence. The mRNA expression level of SOX2,NANOG,LAMB2,COL6A1,integrin α6β4(ITGA6 and ITGB4) and CDH1 were detected by qPCR. Results:The mechanical properties of hydrogel changed the adhesion of oral squamous carcinoma cells,and the high mechanical properties of hydrogel increased cell adhesion. The biological behavior of cells on hydrogel with high mechanical properties is similar to those on traditional cell culture plates. CAL27 formed tight multicellular tumor spheroid on hydrogel with low mechanical properties,while HSC3 formed loose cell clusters. On hydrogels with low mechanical properties,the expression of LAMB2,COL6A1,integrin α6β4 in the two kinds of cells were increase. Cell stemness markers(SOX2,NANOG)and E-cadherin(CDH1)in CAL27 decreased with mechanical properties increased in hydrogels,whereas HSC3 were opposite. Conclusion:The mechanical properties of extracellular matrix can regulate cell adhesion,extracellular matrix protein synthesis,tumor cell stemness and intercellular adhesion. The mechanical properties of extracellular matrix may change the biological behavior of cells by changing the protein expression of extracellular matrix. However,the specific cell biological behavior is related to the specificity of cell line.

      Abstract:Objective:This study aims to construct osteoclast-special Trio gene knockout mouse model and explore the effects of Trio on osteoclasts. Methods:Osteoclast-special Trio gene knockout mice were cultivated by cre-loxp recombination system. Micro-CT was used to scan femurs. The histologic sections of femurs were stained with HE staining,and tartrate resistant acid phosphatase staining to observe number of osteoblasts and osteoclasts. Moue bone marrow cells were treated with macrophage colony stimulating factor(M-CSF) and receptor activator of nuclear factor-κ B ligand(RANKL) for differentiation to discover the effects of Trio on osteoclasts. Results:The mice of Trio knockout group showed small body size. The result of Micro-CT revealed bone mass and related indexes increased. Histologic sections staining displayed that the bone mass of femur increased and osteoclasts decreased in gene knockout mice. In vitro,the number of osteoclasts differentiated from bone marrow cells decreased in mice of Trio knockout group. Conclusion:osteoclast-specific Trio gene knockout mouse model was successfully established. The bone mass of Trio gene knockout mice increased,and osteoclast differentiation was inhibited.

      Abstract:Objective:This study aims to observe the effect of microRNA-1 (miR-1) on the development of neural crest cells and craniofacial region in the knockout zebrafish model. Methods: miR-1 of zebrafish was knocked out by CRISPR / Cas9 gene knock-out system. In situ hybridization was used to detect the expression of differentiation related genes in neural crest cells, and qRT-PCR was used to detect the expression of apoptosis related genes. Results: In the knockout group, the mandible was atrophic and the pigment cells appeared delayed. In situ hybridization showed that the expression of tfap2a,dlx3b, ngn1 and snailb decreased 24 hours after fertilization. qRT-PCR results showed that miR-1 affected the expression of apoptosis related genes. Conclusion: miR-1 is involved in the regulation of neural crest cells and affects craniofacial development, which may play a role by regulating apoptosis related pathways.

    • Effects of vitamin A on behavior of autistic model rats

      Abstract:Objective:This study aims to explore the effect of vitamin A(VA)on the autistic model rats which were induced by sodium valproate(VPA). Methods:The 12.5 d pregnant Wistar rats were given intraperitoneal injection of VPA(600 mg/kg) or normal saline. Then the male offsprings were randomly divided into three groups:control group(VPA,n=11),vitamin A treatment group(VPA+VA,n=12)and saline group(SAL,n=11). On the 7th day after birth,rats in the VPA+VA group were given VA(50 000 U/2.5 kg)by gavage for 7 days,other two groups were given the same amount of soybean oil by gavage for 7 days. Three groups of rats were tested the vitamin A levels in blood in the the 21st day and the 49th day after the birth. The experiments as open-field,three chamber experiment,marble bury and water maze tests in the 35th day after birth were carried out among 3 groups of rats. Results:Compared with the SAL group,repeated behavior and the spontaneous activity level were increasing significantly in the VPA group,the social intercourse ability,spatial learning and memory ability were decreasing in the VPA group(P < 0.05). After vitamin A treatment,compared with the VPA group,the social ability,spontaneous activity,the spatial learning and memory ability were obviously improved in the VPA+VA group(P < 0.05). However,there were no significant improvement on the repetitive behavior of VPA+VA group(P > 0.05). Conclusion:Vitamin A partially improves the behavior of VPA-induced autistic model rats.

      Abstract:Objective:This study aims to investigated and summarized the risk factors and clinicopathological characteristics of synchronous multiple early gastric cancer(SMEGC) to help clinicians improve the detection rate of SMEGC. Methods:Medical records of patients with early gastric cancer (EGC) or high-grade intraepithelial neoplasia (HGIN) wete retrospectively reviewed form June 2014 to December 2018. In total,517 patients were included. Results:SMEGC was diagnosed in 35 patients(6.8%). Male (P=0.028),chronic atrophy gastritis(P=0.024),and moderate to severe intestinal metaplasia(P=0.048) were independent risk factors for the SMEGC. Also,there were no significant differences in survival rates between patients with SMEGC and patients with single neoplasm(P=0.976). Further,main or minor lesions were defined according to Moertel standard. Main and minor lesions showed significant positive correlation in size(r=0.779,P=0.001). Macroscopic types were identical in 51%(P=0.02)between main lesions and minor lesions. Conclusion:Therefore,the endoscopic physicians need a more meticulous endoscopic examination and find the possibility of SMEGC considering characteristics of main and minor lesions.

    • Selection of treatment for early gastric cancer

      Abstract:Objective:This study aims to compare the difference of clinicopathological characteristics of endoscopic submucosal dissection(ESD) and surgery of early gastric cancer(EGC) patients,and to explore the difference of the absolute and expanded criteria for ESD,so as to provide a basis for choosing the best treatment for EGC. Methods:The clinicopathological data of 495 EGC lesions in the First Affiliated Hospital of Nanjing Medical University were prospectively observed,of which 163 patients treated with ESD,and 332 patients treated with surgery. Univariate and multivariate analyses were performed on 153 patients(observation group)in the ESD group and 183 patients(control group)in the surgical group who met the endoscopic resection criteria. Finally,the lymph node metastasis of surgical group was analyzed separately. Results:①Multivariate analysis showed that endoscopic forceps biopsy with carcinoma,flat type,ulcer,and submucosal invasion were influence factors for the selection of surgery(OR=25.970,3.970,7.244,5.038,P < 0.05). ② In the ESD group,the overall accuracy of indication selection was 93.87%(153/163). The pathology of 10 lesions met the surgical criteria were undifferentiated. ③Multivariate analysis showed that tumor location,tumor size,depth of invasion,and lymphovascular invasion were independent risk factors for lymph node metastasis(OR=4.433,2.097,3.510,43.701,P < 0.05).④In the surgical group,the lymph node metastasis rate of of patients suitable for absolute criteria and expanded criteria of ESD were1. 96% and 3.79%,respectively. There were no statistically significant differences in lymph node metastasis rate between patients met absolute criteria and expanded criteria(F=0.428,P=0.51). Conclusion:Some of patients who met the extended indications of ESD chose surgery,and some of them could use ESD. The treatment accuracy and curative resection rate of undifferentiated patients were low. The tumor located in the middle-third of the stomach,the tumor size>2 cm,submucosal invasion and lymphovascular invasion were independent risk factors for lymph node metastasis.

    • Mutation analysis of coexisting genes with DNMT3A p.R882 in acute myeloid leukemia

      Abstract:Objective:This study aims to investigate the coexisting mutation of DNA methyltransferase 3A(DNMT3A)p.R882 in patients with acute myeloid leukemia(AML)and its correlation with some clinical parameters. Methods:There were 49 target genes detected by high-throughput DNA sequencing,and the mutations of DNMT3A p.R882,exon 9 of CALR gene,exon 12 of NPM1 gene,FLT3-ITD,TAD and BZIP of CEBPA were detected by genomic DNA-PCR combined with Sanger sequencing. Results:①Forty-one patients with DNMT3A p.R882 mutations were detected in 301 patients,most common in M5 subtype;nearly all DNMT3A p.R882 mutations(97.6%,40/41)carried other gene mutations,with an average of 3.17 mutations per patient,including 9 double gene mutations,12 co-existing 3 gene mutations and 19 coexisting ≥ 4 gene mutations. ②The most common genetic mutation associated with DNMT3Ap.R882 is NPM1(n=27,65.9%),followed by FLT3-ITD(n=18,43.9%),IDH1(n=9,22.0%),TET2(n=8,19.5%)and NRAS(n=6,14.6%),and so on. ③The leukocyte level of patients with ≥ 4 gene mutations was higher than that of patients with double or three gene mutations,but there was no significant difference in leukocyte,hemoglobin or platelet level(P>0.05). The peripheral leukocyte level of patients with FLT3-ITD gene mutations was higher than that of wild patients(P=0.034),but there was no significant difference in age,hemoglobin and platelet level. And there was no significant difference in median age and peripheral leukocyte level between NPM1,IDH1 or TET2 mutants and wild type. ④Compared with the wild type,patients with NPM1 mutations had a higher complete remission(CR)rate,while the patients with IDH1 mutations had a lower CR rate(P=0.010,0.025). But there was no significant difference in CR rate between the patients with FLT3-ITD on TET2 mutations and the wild type. Conclusion:More than 95% of DNMT3A p.R882-mutated AML coexisted with additional gene mutations,the number and type of coexisting mutations had a certain impact on the clinical characteristics of patients.

    • Effects and mechanisms of simvastatin pretreatment on liver function in hepatocellular carcinoma(HCC) patients undergoing Pringle’s hepatectomy

      Abstract:Objective:This study aims to investigate effects and mechanisms of simvastatin pretreatment on liver function in HCC patients undergoing Pringle’s hepatectomy. Methods:Total 73 HCC patients were randomly divided into control group(n=35)and simvastatin pretreatment group(patients were given oral simvastatin 20 mg/d,n=38). Pre-hepatectomy hepatic biopsies were harvested after laparotomy(prior to hepatic portal occlusion),and post-hepatectomy hepatic biopsies were obtained after reperfusion(prior to abdominal closure). Serum was collected before operation(Pre-Op)and on the first day after operation(POD-1). Serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL)and albumin(ALB)were measured to evaluate liver injury,and the levels of inflammatory cytokines interleukin(IL)-1,IL-6,inducible nitric oxide synthase(INOS)and tumor necrosis factor(TNF)-α were analyzed by reverse transcriptase polymerase chain reaction(RT-PCR). The levels of serum malondialdehyde(MDA) and superoxide dismutase(SOD) were detected by kits according to the manufacturer’s directions. Endoplasmic reticulum stress-related protein C/EBP-homologous protein(CHOP) was analyzed by Western blot. Results:The serum levels of ALT,AST and TBIL in POD-1 were significantly lower in simvastatin group than those in control group(P < 0.05),and RT-PCR results showed the levels of IL-1,IL-6,INOS and TNF-α, were significantly lower in simvastatin group than those in control group(P < 0.05). Furthermore,serum MDA was lower in simvastatin group than that in control group(P < 0.05);SOD in simvastatin group was significantly higher than that in control group(P < 0.05). Western blot results displayed that the expression of CHOP was lower in simvastatin group than in control group. Conclusion:Simvastatin pretreatment for HCC patient effectively alleviates liver injury after Pringle’s hepatectomy,and its mechanism could be related to the inhibition of oxidative stress and endoplasmic reticulum stress.

    • Effects of myocardiac fibrosis indicators on patients with sick sinus syndrome

      Abstract:Objective:This study aims to screen some effective indicator to foretell the extent of atrial fibrosis in patients with sick sinus syndrome(SSS). Methods:Total 95 patients were 40-80 years old,who were consecutively enrolled in cardiovascular department of the Affiliated Jiangning Hospital of Nanjing Medical University from January 2017 to January 2018,and were divided into two groups. One group is control,another group is patients who were diagnosed as SSS. All the patients received echo-cardiogram and dynamic electrocardiogram examination. And transforming growth factor-beta 1(TGF-β1),N-terminal pro-brain natriuretic peptide(NT-proBNP),propeptide of type Ⅰ procollagen(PINP),procollagen Ⅲ N-terminal propeptide(PⅢNP) in the blood of these patients were also measured. Besides,the amplitude of P wave in lead Ⅱ were measured in electrocardiogram of every patient. Then,differences in all parameters between the two groups were compared. Results:There were no statistical differences in age,sex,uric acid,blood glucose and diameter of left or right atrial between two groups,but the amplitude of P wave in electrocardiogram of SSS group was significantly lower than that of control group[(0.09±0.01)cm vs.(0.15±0.02)cm,P=0.001]. The levels of TGF-β(P=0.002),PⅢNP(P=0.017),NT-proBNP(P=0.001) in SSS group obviously higher than those in control group. Besides,after one year’s follow-up,the variation of TGF-β1,PⅢNP,NT-proBNP and the amplitude of P wave in lead Ⅱ between SSS and healthy control group has statistical significance. Conclusion:The levels of TGF-β1,PⅢNP,NT-proBNP in serum and the amplitude of P wave in electrocardiography can predict the morbidity and development of SSS

      Abstract:Proton pump inhibitor(PPI)was widely used in clinic as a first-line drug for the treatment of gastroesophageal reflux disease and peptic ulcer. As the most commonly used prescription drug in digestive department,its usage is increasing year by year. With the long-term use of drugs,a variety of adverse reactions had been reported in recent years. Several studies had shown that use of PPI would cause the disorder of gastrointestinal flora,and was positively correlated with increased risk of intestinal infection. This paper summarized the correlation between PPI and gastrointestinal flora.

    • Progress in diagnosis and treatment of sagittal synostosis

      Abstract:Sagittal synostosis(SS)is the most common non-syndromic craniosynostosis in children,manifesting itself in language-motor development delay,intracranial pressure increase,cognitive dysfunction,abnormal mental activities and seizures. At present,the main diagnostic methods are physical examinations,clinical manifestations and imaging examinations,and the main treatment of SS is operative treatment currently. However,there are many surgical methods selected in terms of different ages of children,degree of cranial suture closure,and degree of cerebellar tonsillar hernia caused by increased cranial pressure. Meanwhile,controversy persists in choosing surgical treatments. The purpose of this review is to investigate the pathogenesis,diagnostic standards,therapeutics of SS,and evaluate preoperative and postoperative neurodevelopment of SS,which can provide convincing evidence for future diagnosis and treatment of SS.

      Abstract:In recent years,with the continuous promotion and improvement of enhanced recovery after surgery(ERAS),rapid recovery has become an important part of perioperative management of gastrointestinal tumors. Under the influence of ERAS,the average hospital stay of patients has been significantly shortened,but some complications(such as anastomotic leakage) are difficult to be detected early,thus increasing the risk of hospital readmission. A large number of literatures proposed that some inflammatory factors were significantly correlated with postoperative anastomotic leakage and had important predictive value in its occurrence,but its predictive value in risk management and control in ERAS has not been reported. In this paper,research progress of inflammatory factors in predicting anastomotic leakage in postoperative ERAS management of digestive tract tumors was reviewed,in order to search for more sensitive predictors to guide patients discharged safely.
