Abstract:Spermatogenesis,including mitosis of spermatogonia,meiosis of spermatocytes and spermiogenesis of spermatids,is one of the most complicated cell differentiation processes in human body. The process of meiosis begins after puberty. For a century,scientists have been working on developing and optimizing culture systems that can support this process in vitro. In recent years,significant progress has been made in the study of in vitro spermatogenesis. The in vitro spermatogenesis can be achieved by in vitro cell culture and gonadal tissue culture. The establishment of in vitro spermatogenesis system is important for the development of theories of human reproductive biology,in vitro evaluation of infertility-related environmental factors,and technologies of fertility preservation.

Abstract:Objective:This study aims to explore the effects of Cyclin D1 expression regulated by GCN5 and the acetylation modification of SOX9 by GCN5 in the glomerular mesangial cells(GMC)stimulated by sublytic C5b-9. Methods:Firstly,GCN5,SOX9 and Cyclin D1 expression in the renal tissue of Thy-1 nephritis(Thy-1N)rats(in vivo)and in sublytic C5b-9-treated GMC(in vitro)was examined using Western blot. Then the plasmids of pIRES2-GCN5 and shGCN5 or these plasmids with Cyclin D1 promoter plasmids were respectively transfected rat GMC,and Cyclin D1 gene initiation,transcription and expression were measured by luciferase reporter gene assay,real-time PCR and Western blot. Meanwhile,SOX9 acetylation was detected by co-immunoprecipitation(Co-IP) and Western blot in the GMC exposed to sublytic C5b-9 or transfected GCN5 overexpression or enzyme activity mutation(ΔGCN5)plasmids. Results:The expression level of GCN5,SOX9 and Cyclin D1 was significantly up-regulated both in vivo and in vitro,and GCN5 gene overexpression increased Cyclin D1 promoter activity,mRNA transcription and protein expression;GCN5 gene knockdown made for the opposite results. Besides,SOX9 acetylation was also elevated upon sublytic C5b-9 stimulation and GCN5 gene overexpression;SOX9 acetylation was reduced upon GCN5 knockdown. Conclusion:GCN5 can promote Cyclin D1 expression and acetylate SOX9 in GMC.

Abstract:Objective:This study aims to evaluate the effect of Nerve injury-induced protein 1(Ninj1)in myeloid-derived macrophages on liver ischemia/reperfusion(IR)-induced immune inflammatory response. Methods:Peripheral blood mononuclear cells(PBMC) were collected from 6 patients who undergone hepatic portal occlusion(hepatic portal occlusion group)during hepatectomy and another 6 patients who had no hepatic portal occlusion(control group). RT-PCR and Western blot were performed to compare the expression of Ninj1 in PBMC of pre-operation and post-operation between the two groups. Murine bone marrow derived macrophages(BMDM)treated with lipopolysaccharide(LPS)were used to evaluate the role of Ninj1 in regulating macrophage inflammatory response in vitro. A murine liver partial warm ischemia reperfusion model was established to determine the role of Ninj1 in liver IR injury and hepatic inflammation. Results:In the hepatic portal occlusion group,the mRNA and protein levels of Ninj1 in PBMCs of post-operation were significantly higher than that in PBMC of pre-operation(P < 0.05),while no obvious change was observed in the control group. Compared with the PBS group,Ninj1 was significantly upregulated in murine BMDM in response to LPS stimulation. Knockdown of Ninj1 in BMDM by siRNA in vitro markedly reduced LPS-induced inflammatory cytokine secretion(P < 0.05). Specific blockade of Ninj1 in macrophages obviously attenuated liver IR injury and hepatic inflammation in vivo(P < 0.05). Conclusion:Liver IR significantly activated Ninj1 in patients’ PBMCs. Inhibition of Ninj1 can mitigate macrophage inflammatory response leading to attenuation of liver IR injury in mice.

Abstract:Objective:This study aims to investigate the effect of proton pump inhibitors on adhesion and inflammation of human umbilical vein endothelial cells(HUVEC)and its potential mechanism. Methods:Western blot and qRT-PCR were used to detect the mRNA and protein expression of interleukin-6(IL-6),intercellular adhesion molecule-1(ICAM-1)and vascular cell adhesion molecule-1(VCAM-1)in HUVECs after treating with 10 μmol/L lansoprazole for 0,24,48,72 and 96 h,protein levels of calcineurin(CaN)and nuclear factor of activated T cells(NFAT)were evaluated at the same time. In addition,protein levels of NFAT,IL-6,ICAM-1 and VCAM-1 were evaluated after treating with lansoprazole,CaN inhibitor FK506,and lansoprazole combined FK506 for the further research in mechanism. Results:Lansoprazole could time-dependently increase the expression of CaN and nuclear translocation of NFAT,and further increase the expression of IL-6,ICAM-1 amd VCAM-1. Calcineurin inhibitor could relieve the NFAT nuclear translocation and vascular inflammation induced by lansoprazole. Conclusion:Lansoprazole may induce endothelial cell inflammation and increase cell adhesion by activating CaN.

Abstract:Objective:This study aims to explore the role of ‘Brain-gut’ regulation in low-frequency auricular electro-acupuncture on maternal separation combined with enema induced visceral hypersensitivity rats. Methods:Visceral hypersensitivity was established by maternal separation combined with acetic acid enema in SD rats,which were randomly divided into the normal control(NC) group,model control(MC) group,auricular acupuncture group 1(AA1)and auricular acupuncture group 2(AA2). Rats in the AA1 group and AA2 group were received electroacupuncture stimulation(2 Hz,30 min/d)for consecutive 10 days. The visceral sensitivity of rats was evaluated by electromyographic(EMG)to graded colorectal distension. The pathological change in distal colon were observed by HE staining. The mRNA expressions of hypothalamic corticotropin releasing factor(CRF),corticotropin releasing factor receptor1(CRF1R),corticotropin releasing factor receptor 2(CRF2R),nesfatin-1,and colonic serotonin receptor 4(5-HT4R)were detected by quantitative real-time PCR. Results:Rats in MC group show higher EMG activity,and mRNA expression of hypothalamic CRF,nesfatin-1,CRF1R,and CRF2R compared with NC group(all P < 0.05). The colonic 5-HT4R mRNA expression in MC group shows a decreased tendency(P=0.067). In contrast,AA treatment significantly reduced the area under the curve(AUC)(P < 0.05). Further,compared to MC group,the mRNA expressions of hypothalamic CRF,nesfatin-1,CRF1R,and CRF2R of AA group significantly decreased(P<0.05)while the colonic 5-HT4R mRNA expression elevated(P<0.05). Conclusion:Low-frequency auricular electro-acupuncture could ameliorate the visceral sensitivity in rats with visceral hypersensitivity,which might relate to the regulation of mRNA expression of CRF,nesfatin-1,CRF1R,CRF2R in hypothalamus and of 5-HT4R in colon.

Abstract:Objective:This study aims to observe the effects and mechanisms of transcription factor YY1 on the migration and invasion of pancreatic cancer cells. Methods:In this study,YY1 and SOX2OT expression were detected by using quantitative RT-PCR and Western blot. The role of YY1 in the migration and invasion of pancreatic cancer cells was studied by cell wound healing assays and transwell cell invasion assays. The functional recovery experiments were performed by overexpression or RNA interference of SOX2OT in YY1 overexpression or YY1 knockdown pancreatic cancer cells. Results:It was found that YY1 overexpression decreased,while YY1 knockdown increased SOX2OT expression in BXPC-3 and PANC-1 cells. Furthermore,YY1 overexpression suppressed,whereas YY1 knockdown enhanced,the migration and invasion properties of BXPC-3 cells in vitro. Overexpression of SOX2OT in YY1 overexpressed BXPC-3 cells or RNA interference of SOX2OT in YY1 knockdown cells can partly reverse the effect of YY on cell migration and invasion. Conclusion:The present study suggested that YY1 suppresses migration and invasion of pancreatic cancer cells in a SOX2OT-dependent mechanism.

Abstract:Objective:This study aims to investigate the value of detecting driver gene mutations in non-small cell lung cancer(NSCLC) patients using next-generation sequencing(NGS) technology. Methods:Somatic mutations in 150 NSCLC patients including EGFR,KRAS,BRAF,NRAS,Her-2 and PIK3CA were detected by Ion torrent personal genome machine(PGM). Sanger sequencing and ddPCR was used to test and verify the results of EGFR gene mutations. Results:According to NGS results,mutations were detected in EGFR(51.33%,77/150),KRAS(7.33%,11/150),BRAF(1.33%,2/150),NRAS(1.33%,2/150),Her-2(2%,3/150)and PIK3CA(4.67%,7/150). There were 57 samples without any somatic mutations in all genes,84 samples had one or more mutations in single gene,while 9 samples harboured mutations in two or more genes. The overall detection rate of EGFR mutation by NGS and Sanger sequencing was 51.33%(77/150)and 38.67%(58/150). The difference between the two methods was statistically significant(χ2=4.862,P=0.027). Compared to Sanger sequencing,the sensitivity and specificity of NGS assays was 98.28% and 78.26%,respectively. The overall detection rate of EGFR mutation by NGS and ddPCR was 51.33%(77/150)and 50%(75/150). There was no significant difference between the two methods(χ2=0.053,P=0.818). Compared to ddPCR,the positive concordance rate was 82.67%,the negative consistency rate was 80%,and the total concordance rate was 81.33%. Conclusion:Next-generation sequencing technology is more suitable for the diagnosis and treatment of NSCLC. Sanger sequencing and ddPCR can be useful supplements.

Abstract:Objective:This study aims to investigate the molecular mechanism of osteosarcoma chemotherapy sensitivity regulation of circ_0001246 via targeting miRNA30a. Methods:The MG63 and U2OS cells were randomly allocated into three groups in 6-well plates respectively,including non-transfection group(blank group), siRNA negative control transfection group(NC group),and siRNA transfection group(circ_0001246 siRNA). The transfection efficiencies were evaluated by fluorescence inverted microscope. Expression Level of circ_0001246 and miRNA30a were detected. Osteosarcoma chemotherapy sensitivity were detected by annexin V-FITC/7AAD apoptosis double staining. Function of circ_0001246 to adsorb miRNA30a were verified by luciferase reporter gene assay. Results: MiRNA30a expression in the circ_0001246 siRNA transfection group were significantly up-regulated compared to the NC group and blank group(P < 0.05). We found circ_0001246 and miRNA30a were inversely expressed. Osteosarcoma chemotherapy sensitivity in the circ_0001246 siRNA transfection group were significantly up-regulated compared to the NC group and blank group(P < 0.05). circ_0001246 could adsorb miRNA30a by direct base pairs. Conclusion: Circ_0001246 is an osteosarcoma related oncogene. Targeted inhibition of circ_0001246 expression in osteosarcoma can enhance the chemotherapy sensitivity of tumor cells to etoposide-VP16. One of the mechanisms is the negative regulation of the miRNA30a expression by the adsorption function of circ_0001246.

Abstract:Objective:The study compared the basic clinical information and hospitalization outcome of patients with eosinophil and non-eosinophilic inflammation type of acute exacerbation of chronic obstructive pulmonary disease(COPD)in order to guide early and accurate treatment. Methods:The basic clinical data of patients with severe acute exacerbation of COPD(AECOPD)admitted to the First Affiliated Hospital of Nanjing Medical University during 2017 were collected to analyze the differences between EOS-AECOPD and non-EOS-AECOPD groups as well as risk factors related to the condition. Results:There were 63 patients with EOS-AECOPD(25.6%)and 183 patients with non-EOS-AECOPD(74.4%). The proportion of critically ill patients was 17.5% and 35.5%,respectively. The mortality during hospitalization were 0 and 8.2%,respectively. Multi-regression analysis revealed that EOS ≥ 2% was a protective factor for the severity of the disease,while pulmonary heart disease,respiratory failure and neutrophil to lymphocyte ratio were independent risk factors. Conclusion:Patients with non-EOS AECOPD(PBE<2%)are more severe,and mortality of hospitalization is higher. The level of eosinophils may help to evaluate the severity of patients with AECOPD in clinical practice so as to guide early and active treatment for critically ill patients.

Abstract:Objective:To study the risk factors of pulmonary hypertension(PH)with supraventricular arrhythmias(SVA),and to improve the understanding of PH with SVA. Methods:The clinical data of 61 patients with PH admitted to the First Affiliated Hospital of NMU from January 2011 to December 2016 were retrospectively analyzed,and SVA was observed in 14 cases. Patients with SVA were included in the SVA group. Patients without SVA were included in the non-SVA group. The risk factors of SVA in PH were analyzed. Results:There were 14 patients in the SVA group,2 males and 12 females,with an average age of(49.21 ± 4.988)years. There were 47 patients in the non-SVA group,8 males and 39 females,with an average age of(38.02 ± 1.865)years. There were significant differences in the diameter of right atrium,the N-terminal pro B-type natriuretic peptide(NT-proBNP),ejection fraction of left ventricular,and age between two groups(P < 0.05). Multiple factor analysis suggested that the diameter of the right atrium was significantly related to PH with SVA. At the end of follow-up,the mortality rate in the SVA group was 28.57%,and the median survival time was 66 months;the mortality rate in the non-SVA group was 27.66%,and the median survival time was 84 months. Conclusion:The diameter of right atrium was significantly related to SVA in patients with PH,and was seemed to be an independent predictor of SVA in patients with PH.

Abstract:Objective:This study aims to explore the feasibility,safety,initial stability and early clinical efficacy of dual mobility cup total hip arthroplasty for the hip reconstruction. Methods:Total 25 patients with the fresh femoral neck fracture (9 male and 16 female),with age of (70.20±6.94) years old,were treated with dual mobility cup THA from January 2019 to June 2019. The operative duration,intraoperative blood loss,postoperative blood loss complications were recorded. All patients were followed up postoperatively. Visual analogue scule(VAS),Harris hip score(HHS) and Mayo hip score(MHS) were recorded to evaluate the improvement of hip local pain and recovery of hip function. Results:Patients were postoperative follow-up in time with an average of (13.55±0.99) months. The mean operative duration was (1.36±0.25) h;the mean blood loss was (275.00±77.17) mL and the mean postoperative blood loss was (236.11±76.32) mL;The X-ray of the postoperative hip joint showed that the joint prosthesis was in good position,the force line of the joint recovered satisfactorily,and the joint prosthesis was in close contact with the bone contact surface without any gaps. The mean VAS scores ranged from (5.61±0.97) points preoperatively to (0.11±0.12) points(t=26.92,P<0.001)at 12 months postoperatively. The average HHS ranged from (3.77±1.40) points preoperatively to (91.83±4.88) points(t=-86.73,P<0.001)at 12 months postoperatively. The average MHS was (89.10±5.22) points at 12 months postoperatively. None of the patients had any complications related to surgery,such as vascular nerve injury,infection,dislocation of prosthesis,vascular embolization. Conclusion:The preliminary clinical result of dual mobility cup total hip arthroplasty is satisfactory for the hip reconstruction,increasing the stability of the hip joint,and restoring the function of the postoperative hip joint.

Abstract:Objective:We aimed to identify the clinical effects of platelet-rich plasma(PRP)on knee osteoarthritis.Methods:We performed a matched case control research for patients with osteoarthritis in our hospital from October 2017 to June 2019,according to gender and age. Both PRP group and the control group enrolled 43 cases. All the patients were evaluated by the severity of osteoarthritis according to Keligren-Lawrence classification. The PRP group received an intra-articularly PRP injection intraoperatively,while the control group received arthroscopic treatment only. Both groups had the same rehabilitation plan after surgery. The pain and knee function were assessed by the VAS score and WOMAC score at baseline,1,3,6 and 12 months after the treatment. The PRP-related complications were recorded during the follow-up.Results:All enrolled patients were available for 12 months follow-up. No serious complication was found in both groups. The VAS and WOMAC scores of both PRP group and the control group after the treatment were significantly lower than those at the baseline. The VAS and WOMAC scores of PRP group were significantly lower than those of the control group at 1 and 3 months but not at postoperative 6 and 12 months after the treatment. For paitents with K-L grade 2,the VAS and WOMAC scores of PRP group were significantly lower than those of the control group at 1,3 and 6 months,but not at 12 months after the treatment. Meanwhile,for paitents with K-L grade 3,the VAS and WOMAC scores of PRP group were significantly lower than those of the control group only at 1 months,but not at 3,6 and 12 months after the treatment.Conclusion:For patients with osteoarthritis,PRP treatment could relieve pain symptoms and restore knee joint function with limited course. PRP has longer duration of efficacy in patients with lower K-L grade.

Abstract:Objective:This study aims to analyze the types of hyperthyroidism in pregnancy and to investigate its effects on pregnancy outcomes. Methods:Total 300 pregnant women were enrolled,who were diagnosed as hyperthyroidism in this study. To analyze the types of hyperthyroidism in pregnancy,according to the diagnostic criteria,there were 108 cases in Graves’ disease(GD)group,104 cases in gestationgal transient thyrotoxicosis(GTT)group,and 33 cases in hashitoxicosis group. Then 340 normal pregnant women were selected for control. The changes of thyroid function in pregnancy and its effects on the adverse outcomes of mothers and offspring in different groups were analyzed. Results:Of 300 pregnant women with hyperthyroidism,41.67% had hyperthyroidism before pregnancy and 58.33% were diagnosed with hyperthyroidism during pregnancy. Among 300 women,36.00% were GD,34.67% were GTT and 11.00% were hashitoxicosis. The proportion of pregnant women who have persistent hyperthyroidism was significantly higher in GD group than in GTT group. The incidences of pregnancy-induced hypertension,preeclampsia,preterm birth,and neonatal hypothyroidism were significantly higher in GD group than those in GTT group or in the control group. Conclusion:For women with active GD or previous history of hyperthyroidism,reasonable pre-pregnancy counseling and pregnancy planning are of great significance to reduce the occurrence of adverse outcomes of mothers and offspring.

Abstract:Objective:This study aims to explore the short-term effect of sublingual allergen-specific immunotherapy in children with allergic rhinitis. Methods:There were 71 subjects of 3-15 years old with allergic rhinitis who were sensitized to dermatophagoides farinae. They were treated with standardized sublingual dermatophagoides farinae immunotherapy drops. Then the short-term effects of the treatment were evaluated by comparing the outcome scores including the total rhinitis symptom scores,total medication scores and visual analogue scale(VAS)before and after the treatment for every three months until a year. Results:Total rhinitis symptom scores,total medication scores and VAS were all declined in 3 months,6 months,9 months and 12 months after the treatment compared with those before the treatment. Meanwhile,the three scores were all declined in 6 months and 9 months compared with those in 3 months and 6 months,respectively. Total rhinitis symptom scores and VAS were both declined while no difference was shown in total medication scores in 12 months compared with those in 9 months. Conclusion:In the short term,sublingual dermatophagoides farinae immunotherapy is an effective way to treat 3-15 years old children with allergic rhinitis who are sensitized to dermatophagoides farina,which can take effect in 3 months after the treatment,and becomes more and more effective in 6 month,9 months and 12 months.

Abstract:Objective:This study aims to investigate the effects of lithotomy on the hemodynamics of lower limbs in patients with gynecologic pelvic tumor whose lower limbs restored horizontally for 5 min after placing lithotomy position during the operation. Methods:From October 2018 to August 2019,101 patients with gynecological pelvic tumor surgery were enrolled in our study. They were randomly divided into two groups,51 patients in the experimental group and 50 patients in the control group. Experimental group received adjustable multi-function stirrup leg rack to obtain lithotomy position,and their legs were restored horizontally for 5 min every one hour. The control group received conventional leg rack until the end of surgery. The great saphenous vein pressure of the two groups patients were monitored before anesthesia(T0),at the beginning of lithotomy position(T1),1 h(T3),2 h after surgery(T3),and at the end of surgery(T4). The upper and lower limb of perfusion index(perfusion index,PI),pulse rate and blood oxygen saturation (SpO2) were recorded and compared. Results:Compared with T0,venous pressure of lower extremity in T1-T4 group increased significantly(P < 0.01). Compared with the control group,the fluctuation of venous pressure in the experimental group of T2~T4 was less,and the stability was significantly better than that in the control group(P < 0.05). However,there was no significant difference between two groups at T1. Compared with T0,the lower limb PI of T1-T4 in the two groups was significantly reduced(P < 0.05). Compared with the control group,the lower limb PI increased significantly(P < 0.05)at T2 and T3 but not T4 in the experimental group. There was no difference in upper limb PI,blood oxygen saturation and pulse rate between the two groups(P > 0.05). Conclusion:Lower limbs restoring horizontally for 5 min every hour can reduce lower limb hemodynamic changes for those lithotomy position surgery with more than two hours longer. It might be beneficial to prevent deep vein thrombosis.

Abstract:Objective:This stndy aims to evaluate the value of compressed sensing(CS) cine cardiac magnetic resonance(CMR) for left atrial(LA)functional assessment compared with traditional segmented cine imaging(regarded as gold standard). Methods:Forty healthy volunteers and thirty-one patients with reduced left ventricular diastolic function by echocardiography underwent segmented cine and CS cine imaging with steady-state free-precession sequence to cover LA during CMR. LA diameter,volume and ejection fraction(EF)were evaluated by CVI42(circle cardiovascular imaging,Canada). Two radiologists independently performed qualitative analysis of image quality and quantitative analysis of all above LA parameters. Results:The image quality was no significant difference between traditional and CS cine. There was an excellent correlation for LA diameter(ICC>0.90) and good correlation for LA volume,EF between both cines(ICC>0.80),except for active EF. There were no significant difference in LA 2- and 4-chamber transverse diameters derived from both cines. Compared to the gold standard,the CS cine overestimated LA minimal volume,but the difference was small[(4.45±11.18)mL,P=0.003]. The LA maximal volume,pre-active volume and passive EF obtained by the two methods had no statistical difference. CS cine more accurately distinguished volunteers from patients with abnormal LA function than segmented cine. Conclusion:CS cine can be used to accurately evaluate LA diameter and volume for LA functional assessment,especially passive EF of LA.

Abstract:Objective:This study aims to investigate the clinical value of shear wave elastography combined with contrast-enhanced ultrasound in the diagnosis of breast lesions. Methods:There were 159 patients,a total of 171 breast lesions enrolled by ultrasound in our hospital from January 2016 to September 2018. The conventional ultrasound,shear wave elastography and contrast-enhanced ultrasound were performed before operation. Taking the pathology as the golden standard,and the ROC curve was drawn. The diagnostic efficacy of conventional BI-RADS classification,shear wave elastography,contrast-enhanced ultrasound and the joint score in the diagnosis of benign and malignant breast lesions were compared,and the clinical value of shear wave elastography combined with contrast-enhanced ultrasound in the diagnosis of breast lesions was summarized. Results:According to the ROC curve,the diagnostic value of shear wave elastography,contrast-enhanced ultrasound and the joint method were obtained,respectively. The diagnostic sensitivity,specificity,accuracy and area under the curve of single method were 85.96%,75.44%,82.46%,0.807;94.74%,71.93%,87.13%,0.833 and 97.37%,75.44%,90.05%,0.864,respectively. The combination has the highest detection rate of malignancy and the highest diagnostic efficiency. Conclusion:Shear wave elastography combined with contrast-enhanced ultrasound improves the diagnostic efficacy of breast lesions and has great diagnostic value for benign and malignant breast tumors.

Abstract:Objective:This study aims to explore the application value of different combined schemes of S-Detect technique and conventional ultrasound in the evaluation of breast nodules. Methods:The routine ultrasound images of 175 breast nodules from May 2019 to April 2020 were collected. BI-RADS category was performed by low-,middle-and senior-aged sonographers before the surgery,and adjusted by four different schemes combined with S-Detect results. The pathological results were used as the gold standard after surgery to evaluate the S-Detect diagnostic value,and compare the diagnostic efficacy of sonographers with different seniority before and after combining different schemes. Results:The diagnostic specificity of S-Detect alone was higher than that of sonographers,and the difference was statistically significant(P < 0.01). AUC value of combined diagnosis of low- and middle-aged sonographers significantly increased,and it was statistically significant(P < 0.01),among which the combination of schemes Ⅲ and Ⅳ was significantly better than schemes Ⅰ and Ⅱ(P < 0.05);AUC value decreased after the combination of senior-aged physicians. Conclusion:S-Detect technique had high diagnostic specificity. Low- and middle-aged sonographers combined S-Detect technique with BI-RADS category could improve their diagnostic efficiency,among which the combination of scheme Ⅲ and Ⅳ performed better.

Abstract:Objective:To explore the feasibility of two time series models for predicting tuberculosis epidemic in Shanghai,so as to provide a scientific reference for the prevention and control of tuberculosis in Shanghai. Methods:The epidemiological data of tuberculosis from Jan.2007 to Jun.2018 in Shanghai was collected. The monthly cases of tuberculosis from Jan.2007 to Dec.2017 was fitted by multiple seasonal ARIMA model and the exponential smoothing model. We predicted the monthly number of tuberculosis cases from Jan. to Jun. 2018 using the established models and comparcd the results to the real values. Results:ARIMA(0,1,1)×(1,1,2)12 was the optimal ARIMA model whose RMSE was 76.27 and the sum of the relative error from Jan. to Jun.2018 was 0.402;The optimal model constructed by exponential smoothing was Holt-Winters additive exponential smoothing model with RMSE of 69.61 and a sum of relative error of 0.292 from Jan. to Jun.2018. Conclusion:The exponential smoothing model could be fitted more effectively and had higher predictive accuracy. In short,it has important guiding significance for the prevcntion and control of tuberculosis in Shanghai.

Abstract:Objective:This stndy aims to explore the death characteristics and trends caused by four main chronic noncommunicable diseases(NCDs)among residents between 30 and 69 years old in Wujin District of Changzhou from 2009 to 2019,to provide suggestions and basis for realizing the goal of decreasing the probability of premature mortality of“Health China 2030”major NCDs in this region. Methods:The mortality rate of four major NCDs were analyzed for residents between 30 and 69 years old,the average annual percent change(APC)was used to describe the change trend,and premature mortality probability of major NCDs was calculated on the basis of the life table,the effects of demographic factors and non-demographic factors were analyzed by differential decomposition analysis. Results:In 2009—2019,the crude mortality rate of major NCDs among residents between 30 and 69 years old decreased at the rate of APC=-2.01%(P<0.001). The standardized mortality of malignant tumor,cardio-cerebrovascular disease,chronic respiratory diseases going down with APC=-3.62%(P<0.001),APC=-3.80%(P<0.001),APC=-6.61%(P=0.004)respectively,while the standardized mortality of diabetes increased year by year(APC=4.19%,P=0.022). The premature mortality of major NCDs decreased from 12.32% in 2009 to 8.91% in 2019,while the premature mortality of diabetes showed a slightly upward trend. The decrease of crude mortality caused by non-demographic factors in the four major NCDs of residents aged 30 to 69 obviously outweighed the increase of crude mortality caused by demographic factors,but for the change of male diabetes mortality,both demographic factors and non- demographic factors contributed to the increase of mortality. Conclusion:The premature mortality of major NCDs residents between 30 and 69 years old is on the decline. However,in general,the prevention and control of major NCDs is still grim,and the prevention and control of diabetes is particularly important in terms of preventing premature death.

Abstract:Objective:This study aims to introduce the manufacture and use instruction for a simple and multifunctional device for mouse fixation. Methods:Discarded 50 mL centrifuge tubes were collected and processed into the fixation device according to the pre-designed program. A total of 60 mice were randomly and equally assigned into two groups and fixed with traditional methods(control group)and this fixation device(study group),respectively. Tail vein injections or tail-clamping blood sampling or intra-articular injections were performed. The procedure durations and success rate were recorded. Results:A single 50 ml centrifuge tube weighed about 12 g and mere 15 minutes were taken to manufact the fixation device. The procedure durations of tail vein injection,tail-clamping blood sampling and intra-articular injections in control and study groups were(32.10±1.97)s and(30.27±2.20)s,(30.10±1.73)s and(28.67±1.55)s,and(40.77±2.73)s and(38.73±2.16)s,respectively. The data between two groups were significantly different(P<0.01). The success rates of the three procedures in control group study group were 100% and 100 %,100% and 100%,and 83.33% and 95.00%,respectively. There were significant differences in procedure duration and success rate between two groups(P<0.05). Conclusion:The mouse fixation device possesses the advantages of easily available materials,simple manufacture,convenient transportation and multiple functions. It provides a new safe and reliable choice for mouse fixation in the experiment and is worthy to be promoted.

Abstract:Hypertension is closely related to the occurrence and development of cerebrovascular diseases,such as stroke and heart failure. Intensive blood pressure control can reduce the morbidity and mortality of related diseases. However,the awareness rate,diagnosis rate,treatment rate of hypertension and control rate of hypertension are still not satisfactory. The reasons of low control rate mainly include the unclear pathogenesis,the diversification of first-line drugs,and the varied response for different population. In recent years,metabolomics studies have provided new perspectives for the pathogenesis of hypertension,biomarker screening,pharmacological action of related drugs,efficacy and safety assessment of medication. In this paper,the research methods and the results of metabolomics research related to hypertension and antihypertensive drugs are reviewed,so as to provide reference for further research.

This paper reviews the animal models of special breast cancer at home and abroad,summarizes the background of the models,concludes the modeling methods,and discusses the characteristics of the models,so as to provide references for the research of breast cancer.

Abstract:Metabonomics is a popular research method in the area of tumor research. The theoretical basis is that tumor has metabolic characteristics different from normal tissues,and this metabolic changes are closely related to biological phenotype. Metabonomics uses high-throughput detection methods to identify the metabolic differences of biological samples,and combined with bioinformatics analysis,according to the variation in type or content of metabolites,to elucidate the remodeling mechanism of some key biochemical pathways in tumorgenesis and progression. It should be deeply analyzed or precisely intervened,so as to improve our understanding of cancer and the level of diagnosis and treatment. In this paper,we focus on the application of metabonomics in mechanism analysis of glioblastoma that is a subtype with the highest malignancy of glioma,from the perspective of metabonomics,to reveal the changes of some endogenous small molecule metabolites in the pathological process of glioblastoma and to provide important reference for its clinical diagnosis,treatment,prognosis and drug development.
