    • Establishment of ACE2⁃modified porcine fetal fibroblasts via CRISPR/Cas9 system

      Abstract:Objective:This study aims to establish angiotensin converting enzyme 2(ACE2)gene-modified porcine fetal fibroblast (PFF) cell lines via CRISPR/Cas9 system-mediated gene modification,which could serve as donor cells for the subsequent construction of a severe acute respiratory syndrome coronavirus type 2(SARS-CoV-2)insensitive pig model. Methods:①The ACE2 amino acid sequences of rats and chickens were compared by bioinformatic analysis. The ACE2 amino acid residues,which play a crucial role in the interaction between ACE2 and SARS-CoV-2 S protein,were replaced by the amino acid residues of chicken’s ACE2 at the corresponding sites. The chimeric ACE2 gene was synthesized and cloned into the knock-in vector. ②The sgRNA targeting the porcine ACE2 gene was designed,synthesized,and cloned into the pX330 vector to construct the ACE2 gene targeting vector. ③The ACE2 targeting vector,the ACE2 gene knock-in donor vector,and a neomycin-resistant plasmid were co-transfected into PFP. G418 was used to screen the resistant single-cell colonies,and sequencing was performed to identify the correct ones. Results:According to the amino acid sequence alignment,the rat ACE2 amino acid residues at positions 19 th,31st,34 th,35 th,42 nd,353 rd and 354 th were replaced by the amino acid residues of chicken ACE2. The chimeric ACE2 gene was synthesized,and the ACE2 knock-in vector was successfully constructed. Genotype analysis of G418 resistant colonies showed that single-cell colonies with expected modification were successfully obtained. Conclusion:The PFF cells with endogenous ACE2 knock-out and chimeric ACE2 knock-in are successfully generated using CRISPR/Cas9-mediated gene modification,which will lay a foundation for the construction of SARS-CoV-2 insusceptible pig models.

    • Di ⁃(2 ⁃ ethylhexyl)phthalate impairs the functions of human coronary artery endothelial cells by inhibiting PI3K/AKT signal pathway

      Abstract:Objective:This study aims to investigate the effects of di(2-ethylhexyl)phthalate(DEHP)exposure on PI3K/AKT signal pathway and human coronary artery endothelial cells(HCAECs),and to provide a theoretical basis for the treatment of HCAECs damaged by DEHP exposure. Methods:According to relevant literature research,HCAECs were treated with different concentrations of DEHP and divided into control group(DMSO)and experimental group(DEHP 128,256,384,512 μmol/L). The ability of cell proliferation was measured by CCK- 8 assay,the ability of cell tube formation was evaluated by in vitro tube formation assay,the level of apoptosis was evaluated by flow cytometry,and the levels of p-PI3K,PI3K,p-AKT,AKT and apoptosis-related proteins BAX,Bcl-2, cleaved Caspase-3 and Caspase-3 were detected by Western blot. Results:CCK-8 assay showed that DEHP inhibited the proliferation ability of HCAECs in a concentration- and time-dependent manner(P < 0.05). The results of in vitro tube formation assay showed that the morphology of cells in the experimental group was mostly isolated round and oval. The number of intersections formed was reduced compared with the control group(P < 0.05)when the DEHP concentration was ≥256 μmol/L. The results of flow cytometry showed that the proportion of apoptosis in the experimental group was higher than that in the control group(P < 0.05). Western blot results showed that the expression level of Bcl-2 in the experimental group was lower than that in the control group,while the levels of BAX and cleaved Caspase-3/Caspase-3 increased(P < 0.05). And the ratio of p-PI3K/PI3K and p-AKT/AKT were lower than those in the control group(P < 0.05)when DEHP≥256 μmol/L. Conclusion:DEHP can induce apoptosis and inhibit the proliferation and angiogenesis of HCAECs by inhibiting PI3K/AKT/ signal pathway.

      Abstract:Objective:This study aims to preliminary investigate the effect of plasma exosome and the content microRNA-25-3p (miR-25-3p)on myocardial fibrosis. Methods:The plasma exosomes of 10 healthy C57BL/6 mice(control group)and 10 mice underwent myocardial infarction surgery(cardiac fibrosis model group)were collected successively. The exosomes were identified by electron microscopy,nano tracking particle analysis and detection of marker protein;qPCR and Western blot were used to investigate effects of the two different exosomes on the fibrosis level of mouse cardiac fibroblasts stimulated by angiotensin Ⅱ(Ang Ⅱ); expression of miR -25-3p in exosomes from two groups were detected by qPCR;Western blot and immunofluorescence staining were used to present the effects of miR-25-3p mimic and inhibitor on differentiation and proliferation of cardiac fibroblasts. Results:The plasma exosomes of healthy mice attenuated the increased fibrosis level induced by Ang Ⅱ,while the plasma exosomes of myocardial fibrosis model mice lost this function and contained more miR-25-3p;miR-25-3p mimic promoted cardiac fibrosis,while miR-25-3p inhibitor showed the opposite effect. Conclusion:Plasma exosome of health mice can protect myocardial fibrosis;the up-regulated miR- 25-3p in plasma exosome promotes cardiac fibrosis.

      Abstract:Objective:This study aims to evaluate the effect of Parkin mediated mitophagy on globular adiponectin(gAPN) ameliorated H9C2 cardiomyocytes injury induced by intermittent hypoxia(IH)and the potential mechanisms. Methods:The cell IH model was established. H9C2 cell apoptosis was detected by flow cytometry. The mitophagy was detected by fluorescence colocalization of LC3 and mitochondrial red marker. The protein expression levels of Bax,Bcl-2 and Parkin were detected by Western blot. The mitochondrial membrane potential(MMP) was detected by JC-1 kit. Results:After IH exposure,the apoptosis of H9C2 cardiomyocytes,protein expression levels of Bax and Parkin,mitophagy were increased,MMP and Bcl-2 protein expression were decreased. After gAPN treatment,the apoptosis of H9C2 cardiomyocytes,Bax protein expression level were decreased,the MMP, protein expression levels of Bcl-2 and Parkin,and mitophagy were increased. Inhibiting Parkin gene expression decreased the mitophagy,and increased the mitochondrial injury and H9C2 cell apoptosis. Conclusion:gAPN ameliorated the IH induced H9C2 cardiomyocytes injury through upregulating Parkin mediated mitophagy.

    • The expression of TUBA1C and its biological functions in breast cancer

      Abstract:Objective:The current research aims to systematically analyze expression levels and prognostic values of tubulin gene superfamily members in breast cancer and identify novel functional microtubule genes. Methods:The Cancer Genome Atlas(TCGA) and the Kaplan -Meier plotter database were used to define the expression levels and prognostic values of tubulin gene superfamily members in breast cancer,respectively. Expression pattern and prognostic value of tubulin alpha 1c(TUBA1C)was subsequently confirmed on breast cancer tissue microarray(TMA)by immunochemistry(IHC)staining. Moreover,cell viability and apoptosis assays were conducted in MDA - MB - 231 and MCF - 7 breast cancer cells to investigate the functional role of TUBA1C in breast cancer. Results:Systematic analysis of tubulin gene superfamily revealed that TUBA1C was significantly overexpressed and had notable prognostic value in breast cancer. IHC analysis exhibited that the TUBA1C was overexpressed at the protein level in tumor tissues compared with para-tumor tissues(χ2 =6.929,P=0.008),and upregulated TUBA1C was associated with the advanced clinical stage(χ2 = 6.357,P=0.042)and worse overall survival(OS)(P=0.021). In addition,univariate(P=0.032)and multivariate Cox regression analyses (P=0.040)further demonstrated that TUBA1C was an independent prognostic factor in breast cancer. Besides,knockdown of TUBA1C notably suppressed cell proliferation and induced cell apoptosis. Conclusion:These results reveal that TUBA1C is a novel oncogene and a potential prognostic biomarker in breast cancer. Besides,silencing TUBA1C could inhibit breast cancer progression,which could be a potential target for therapy.

    • Establishment and validation of prognostic risk nomogram model among patients with premature ST⁃segment elevation myocardial infarction

      Abstract:Objective:This study aims to analyze the risk factors of patients with premature ST segment elevation myocardial infarction(STEMI)and to develop a nomogram model to predict the risk of postoperative major adverse cardiac events(MACE)in patients with premature STEMI. Methods:A total of 166 patients diagnosed with premature STEMI undergoing percutaneous coronary intervention(PCI)in the Affiliated Huaian No.1 People’s Hospital of Nanjing Medical University from January 2017 to December 2018 were selected as the research subjects,and they were followed up for 24 months. According to the occurrence of MACE,they were divided into MACE group(62 cases)and non-MACE group(104 cases). Risk factors were screened using LASSO regression with Cox regression analysis,and a nomogram prediction model was constructed. The efficacy of the model was evaluated by receiver operating characteristic curve(ROC),clinical decision curve(DCA),calibration curve,etc.,and the stability of the model was verified by Bootstrap self sampling. Results:The results of LASSO regression and Cox regression showed that neutrophil gelatinase associated lipocalin(NGAL),creatinine,time from AMI to opening of infarct related artery,left ventricular ejection fraction(LVEF),infarct related artery were significant risk factors for the occurrence of postoperative MACE in patients with premature STEMI(P < 0.05),and the nomogram prediction model was constructed using these 5 predictors. The areas under the ROC curves of the models were 0.95 (95% CI:0.89~1.00),0.94(95% CI:0.80~0.99),0.87(95% CI:0.82~0.93)at 6,12,24 months after PCI,respectively,which were larger than those of the individual risk factors. The calibration curve is close to the ideal curve. The DCA curves showed better performance of the nomogram prediction model in the 0.25~1 threshold probability range. Conclusion:In this study,a nomogram prediction model was constructed based on the significant risk factors for the occurrence of MACE after PCI in patients with premature STEMI,and after Bootstrap internal validation,the prediction model had better efficacy and stability,and could accurately predict the risk of MACE at 6,12,and 24 months after PCI in patients with premature STEMI. It is helpful to carry out individualized intervention treatment for high-risk patients with premature STEMI and improve their prognoses.

    • Application of three ⁃ dimensional speckle tracking imaging in the study of left ventricular diastolic function in coronary artery disease

      Abstract:Objective:This study aims to evaluate the left ventricular diastolic function in patients with coronary artery disease by three - dimensional speckle tracking imaging(3D - STI). Methods:Sixty - six patients with coronary heart disease were included and divided into two groups according to left ventricular end diastolic pressure(LVEDP):31 cases in the group with normal left ventricular diastolic function(LVEDP≤ 15 mmhg),and 35 cases in the group with left ventricular diastolic dysfunction. Thirty healthy volunteers were selected as the control group. The longitudinal strain(LS),circumferential strain(CS),area strain(AS)and radial strain(RS)of 17 segments of left ventricular were obtained by 4D auto LVQ software. Results:Four strains of diseased myocardium in left ventricular diastolic dysfunction group was significantly lower than those in normal left ventricular diastolic function group and control group(P < 0.05),LS and AS of diseased myocardium in normal left ventricular diastolic function group were lower than those in control group(P < 0.05). Pearson correlation analysis showed that the four strains had good correlation with LVEDP(P < 0.001). The analysis of receiver operating characteristic(ROC)curve showed that the area under the curve(AUC)of four strains(LS,CS,AS and RS)predicting left ventricular diastolic dysfunction were 0.76,0.76,0.86 and 0.72 respectively(P < 0.001). Logistic regression analysis revealed that the four strains could be used as risk factors for predicting left ventricular diastolic dysfunction,and their OR values were:LS 9.184(95% CI:1.394~30.496,P=0.021);CS 7.727(95%CI:1.175~20.841,P=0.033);AS 8.918(95%CI:1.294~19.484,P=0.022);RS 7.730 (95%CI:1.344~24.438,P=0.026). Conclusion:3D - STI can be used to evaluate left ventricular diastolic function in patients with coronary artery disease. LS and AS are more useful to find early stage of left ventricular diastolic dysfunction.

    • The correlation between the changes of psychophysiology and vagus nerve function and clinical characteristics in patients with Crohn’s disease

      Abstract:Objective:The aim of this study was to assess the disorder of psychopsychological and vagus nerve function in patients with Crohn’s disease(CD)and analyze the relationship with clinical characteristics of CD patients so as to provide evidence for the comprehensive treatment of CD. Methods:A total of 124 CD patients were enrolled in the First Affiliated Hospital of Nanjing Medical University between January 2014 and December 2018. A total of 104 healthy controls matched by age and sex were also included. Disease activities were evaluated by the Crohn’s disease activity index(CDAI). Anxiety and depression were assessed by Generalized Anxiety Disorder(GAD -7)and Patient Health Questionnaire(PHQ -9). Vagal nerve function was evaluated by heart rate variability (HRV)in 48 CD patients and 17 healthy controls. High frequency(HF),low frequency(LF),LF/HF and HF/(HF+LF)were analyzed. Pearson correlation analysis,Spearman correlation analysis and ordinal logistic regression analysis were used to analyze the possible link among the incidence of anxiety and depression,vagus nerve function and clinical features of CD patients. Results:Among the 124 CD patients,the incidence of anxiety and depression was 42.7%(53/124)and 27.4%(34/124),which was significantly higher than that in the control group[8.7%(9/104)and 16.3%(17/104)],P value were<0.001 and 0.046 respectively. Perianal surgery history(OR= 5.405,95%CI:1.213~24.085,P=0.027)and frequency of recurrence≥2(OR=3.887,95%CI:1.242~12.166,P=0.020)were positive factors for the escalation of anxiety grades. Enterectomy surgery history(OR=5.491,95%CI:1.356~22.243,P=0.017)and upper GI tract involvement(OR=4.774,95% CI:1.529~14.910,P=0.007)were positive factors for the escalation of depression grades. The average value of HF/(LF+HF)of CD patients was 0.490±0.151,which was significantly lower than that of control group(0.661±0.092, P<0.001). Vagal activity had negative correlation with symptom of anxiety(r=-0.377,P=0.008)and depression(r=-0.350,P=0.015). Conclusion:CD patients had increased incidence of anxiety and depression and showed vagus nerve dysfunction. The function of vagus nerve had negative correlation with severity of anxiety and depression in CD patients.

    • The prognostic value of esophageal dilatation in connective tissue disease with pulmonary arterial hypertension

      Abstract:Objective:This study aims to explore the prognostic value of esophageal dilatation in connective tissue disease with pulmonary arterial hypertension(CTD -PAH). Methods:A total of 170 patients with CTD -PAH diagnosed by echocardiography were collected from the Departments of Rheumatology,the First Affiliated Hospital of Nanjing Medical University from 2010 to 2022. Clinical characteristics,echocardiographic parameters,chest CT parameters and survival of patients with esophageal dilatation and without esophageal dilatation were compared. Kaplan-Meier and Cox methods were used for survival analysis. Esophageal dilatation is defined by coronal diameter of the esophagus lumen >9 mm at the infra - aortic portion of the esophagus. Results:The mortality of patients with esophageal dilatation was higher than that of patients without esophageal dilatation. The overall 1-,3- and 5-year survival rates of patients without esophageal dilatation were 100.0%,95.2%,84.8%,while the overall 1-,3- and 5-year survival rates of patients with esophageal dilatation were 93.6%,73.0%,62.3% . After adjustment for age,sex,PAH duration,and N - terminal pro - B type natriuretic peptide(NT-proBNP),esophageal dilatation remained an independent risk factor for prognosis of patients with CTD-PAH. Conclusion:Esophageal dilatation may be one of the indexes to predict the long-term poor prognosis of patients with CTD-PAH.

    • Analysis of coagulation function correlative indexes in asymptomatic hyperuricemia patients

      Abstract:Objective:This study aims to observe the changes of coagulation indexes in patients with asymptomatic hyperuricemia, and analyze its correlation with the increasing serum uric acid(SUA)level. Methods:Data were collected from 2 389 patients who have taken physical examinations in the Health Management Center of the First Affiliated Hospital of Nanjing Medical University from January 2018 to September 2021,including 1 497 males and 892 females. Due to the difference in SUA levels between males and females,they were discussed separately. The following coagulation indexes of patients with asymptomatic hyperuricemia were analyzed: prothrombin time(PT),activated partial thromboplastin time(APTT),fibrinogen(FIB),thrombin time(TT),D-dimer(DD)and platelet (PLT). The data of males and females were separately divided into four groups according to the SUA quartile level. With the increase of the SUA level,the changes of coagulation indexes and its correlation with SUA were analyzed. Results:The prevalence of asymptomatic hyperuricemia was significantly higher in males than that in females(P<0.001). Both in male and female asymptomatic hyperuricemia patients,data displayed shortened plasma PT(P<0.05),while the levels of FIB and PLT increased(P<0.05). With the increase of SUA level,there were statistically significant differences in PT,FIB and PLT among different groups in males(P<0.05), and there existed statistically significant differences in plasma PT among different female groups(P<0.05). The SUA level of male patients was negatively correlated with PT nor DD,and positively correlated with FIB and PLT(P<0.05). The SUA level of female patients was negatively correlated with PT,and positively correlated with FIB and TT(P<0.05). Conclusion:The high SUA levels can increase plasma viscosity,activate platelets and affect exogenous coagulation function,suggesting correct recognition and timely intervention of asymptomatic hyperuricemia in clinical diagnosis and treatment. Periodic physical examination is highly recommended to provide effective guarantee for early prevention of coagulation disorders.

    • Application of morning urine combined with pelvic ultrasound and serum gonadotropin levels in the primary diagnosis of girls with central precocious puberty

      Abstract:Objective:This study aims to investigate the diagnostic value of morning urine combined with pelvic ultrasound and serum basal urinary gonadotropin(Gn)levels of girls with central precocious puberty(CPP). Methods:Total 83 girls with precocious puberty attending our hospital were enrolled in this study. The age,bone age,the uterus ultrasonic,basal serum Gn levels and other related informations were collected. For girls with suspected CPP,gonadotropin - releasing hormone(GnRH)stimulation test was performed. Then 83 girls were divided into CPP group and non -CPP group according to the results of GnRH test. At the same time, morning urine from all patients were also collected. Then,the Gn levels were detected by immunochemiluminometric assays. SPSS 25.0 software was used for statistical analysis of the data,and receiver operating characteristic curve(ROC)was drawn. Results:①The levels of luteinzing hormone(LH)and follicle-stimulating hormone(FSH)of morning urine in CPP group were significantly higher than those in non-CPP group(P < 0.05). ②The LH levels in morning urinary were positively correlated with peak LH levels in the serum, uterine volume and ovarian volume(P < 0.001);urinary LH/FSH ratios were also positively correlated with peak LH/FSH ratios in the serum,uterine volume and ovarian volume(P < 0.001). ③The area under ROC curve(AUC),threshold,sensitivity and specificity of morning urine LH for diagnosis of CPP were 0.879,2.12 U/L,82.1% ,88.0% ,respectively;and AUC,threshold,sensitivity and specificity of uterine volume for diagnosis of CPP were 0.935,1.87 cm3 ,87.2%,88.0%,respectively;the sensitivity and specificity of serum basal LH(≥0.3 U/L)for CPP diagnosis were 89.7% and 80.0%,respectively. ④The AUC,sensitivity and specificity of the combination of morning urine LH,uterine volume and blood - based LH for diagnosis of CPP were 0.977,92.3% and 96.0% , respectively. Conclusion:Morning urine LH combined with uterine volume and blood-based LH can be used as preliminary screening indicators for CPP in girls,which is non - invasive and convenient. These indicators can further improve the preliminary diagnostic efficacy of CPP,and have certain clinical application value.

    • Epidemiological characteristics and temporal trends of pulmonary tuberculosis in Nanjingfrom 2005 to 2020

      Abstract:Objective:This study aims to analyze the epidemiological characteristics and temporal trends of pulmonary tuberculosis (PTB)in Nanjing from 2005 to 2020 and provide scientific evidence for tuberculosis prevention strategy. Methods:Data of registered PTB in Nanjing from 2005 to 2020 was collected through the National Tuberculosis Information Management System. The Joinpoint regression model was used to estimate the annual percent change(APC)as well as the average annual percent change(AAPC)and the 95% CI,respectively. Results:A total of 51 624 PTB were reported with the reported incidence dropped from 71.86/100 000 to 26.08/100 000 and presented overall declining trend,with an average annual decline of 6.47%(AAPC=-6.47,95%CI:-9.84~-2.97, P < 0.001). The reported incidence and the proportion of smear - positive PTB annually decreased by 8.72%(AAPC=- 8.72,95%CI: -14.71~-2.32,P= 0.008)and 3.37%(AAPC=-3.37,95%CI:-4.60~-2.12,P < 0.001),respectively. There was no significant change in reported incidence of bacteriological - positive PTB(AAPC=- 6.37,95% CI:- 15.21~3.41,P=0.194). However,the proportion of bacteriological-positive PTB decreased and then increased with 2016 as the turning point. The reported incidence of PTB in the males and females both presented significant declining trend(AAPC=- 6.80,95% CI:- 10.13~- 3.35,P < 0.001;AAPC=- 6.88,95% CI: -8.54~-5.18,P < 0.001),and there was no significant difference in the decline rate between the males and females(P=0.968). The proportion of the female cases increased(AAPC=0.99,95%CI:0.31~1.67,P=0.008),while the proportion of the male cases decreased (AAPC=-0.42,95%CI:-0.73~-0.10,P=0.013)annually. The reported incidence rate of all age groups of ≥ 30 years old showed a downward trend,while the reported incidence rate changes of groups of 0~< 15 and 15~< 30 years old had no statistical significance. In addition,the proportion of 15~<30 years old age group showed an increasing trend(AAPC=1.66,95%CI:0.28~3.07,P=0.019)while there was no significant change trend of all the other age groups. The PTB reported incidence in the main urban districts and the suburban districtsall showed decreasing trend(AAPC=-8.42,95%CI:-15.60~-0.62,P=0.035;AAPC=-5.37,95%CI:-9.13~-1.46, P < 0.008),and there was no significant difference with respect to the decline rate(P=0.478). However,the proportion of patients in the central urban area has shown a downward trend(P < 0.001)since 2009,while that in the non central urban area has shown an upward trend(P < 0.001). Conclusion:The overall reported incidence of tuberculosis in Nanjing showed a downward trend from 2005 to 2020 and the pathogen positive rate increased annually since 2016. More effort should be paid to the 15~<30 years old age group population,meanwhile the proportion of women and patients in non central urban areas is on the rise,which should be paid attention to.

      Abstract:Objective:This study aims to analyze the epidemiological characteristics of coronavirus disease 2019(COVID - 19) caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)Omicron variant and provide data support for prevention and control measures followed. Methods:The COVID-19 data during February 13,2022 to April 9,2022 were obtained from official website of Jiangsu Province Health Commission and municipal governments. Descriptive epidemiological methods were used to analyze the characteristics of cases. Results:As of April 9,2022,the spread of COVID - 19 caused by SARS - CoV - 2 Omicron variant in Jiangsu Province has been initially contained,with a cumulative incidence of 0.48/100 000 and a fatality rate of 0. It was characterized by a high proportion(76.3%)of asymptomatic patients,and most of the confirmed cases were mild. The median age was 42 years,and 16.9% infected individuals were under 20 years old. The median incubation period of Omicron BA.1 variant was 5.0(4.5,6.0)days,and that of BA.2 variant was 3.0(2.8,3.5)days,with significant difference(P<0.05). There was a high proportion of cluster cases(81.6%). Among them,the main type was family cluster. Conclusion:Omicron variant is highly infectious,with short incubation period and weakened virulence. At this stage,the fatality rate of infection caused by Omicron variant in Jiangsu Province was 0. The results of precise epidemic control measures implemented in Jiangsu Province were remarkable.

      Abstract:Objective:This study aims to establish and evaluate antibody quantitative detection methods of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),in order to provide serological data base for corona virus disease-2019(COVID-19)detection and prevention. Methods:The serum and throat swab samples of SARS-CoV-2 infected persons and their close contacts from January to February 2020 were collected,and the SARS-CoV-2 nucleic acid was detected by RT-qPCR,the SARS-CoV-2 specific antibody was detected by microfluidic method and colloidal gold method,respectively,and the consistency of the results was compared and analyzed. Results:The results of detection of SARS - CoV - 2 specific IgG antibody by microfluidic method and colloidal gold method were consistent with those by RT-qPCR,but there were significant differences in IgG positive rates between the early-middle and the late stages by the microfluidic method. Conclusion:As a novel quantitative way to detect SARS-CoV-2 antibodies,the microfluidic method is brief and accurate.

      Abstract:Objective:This study aims to establish percentile reference values and curves of body mass index(BMI)for children and adolescents aged 7 to 17 in Jiangsu Province,providing a scientific basis for evaluation of growth and development characteristics. Methods:Based on the“Surveillance for common disease and health risk factors among students project”,a stratified random cluster sampling method was used to collect 45 595 students aged 7 to 17 from 26 counties(cities or districts)of 13 cities in Jiangsu Province. BMI percentile reference values for children and adolescents in Jiangsu Province were obtained by using the skewness - median - coefficient of variation method(LMS method). The corresponding percentile curves were fitted and compared with the national level. Results:The BMI of boys and girls aged 7 to 17 in Jiangsu Province increased with age. Boys’BMI entered the growth peak at the age of 9,slowed down after the age of 12;girls’BMI entered the growth peak at the age of 10,and slowed down after the age of 14. The 5 th, 50 th,and 95 th BMI percentiles of children and adolescents in Jiangsu Province were higher than the national level throughout the age range. Conclusion:This study presents the distribution of BMI among children and adolescents in Jiangsu Province. BMI percentile reference values and growth curves of children and adolescents in Jiangsu Province were obtained based on the LMS method,and the fitting effect was good. The main percentiles of BMI for boys and girls in all age groups in Jiangsu Province were higher than the national level,and the median BMI of boys in each age group was higher than that of girls.

    • Research progress of gut microbiota and pregnancy⁃related diseases

      Abstract:The gut flora is an ecosystem which is composed of various microorganisms in the intestine and has a symbiotic relationship with the human body. These microorganisms participate in various functions such as metabolism,immunity and nutrition in the human intestine,and are closely related to the health of the human body. They are also inextricably linked to the occurrence and development of diseases. The composition of the gut flora of pregnant women has undergone significant changes,and the dysbiosis of the gut flora is also considered important in the occurrence and development of pregnancy-related diseases. Probiotics can improve the intestinal microenvironment and can be used to prevent or treat these pregnancy-related diseases. This article reviews the changes in the gut flora of women with pregnancy - related diseases such as gestational diabetes mellitus,preeclampsia,premature delivery, intrahepatic cholestasis of pregnancy and the effectiveness of probiotics for clinical prevention and treatment. This review is also expected to provide a basis for further research on the mechanism of gut flora affecting the health of pregnant women and higher-quality research on probiotics.

      Abstract:Endometriosis(EMS)has been recognized as a fibrotic disease in recent years,and the fibrosis process of EMS lesions is strongly associated with EMS-related pain,infertility and drug resistance. Myofibroblast(MFB)is the key cell of EMS fibrosis,MFB has a variety of cell sources in eutopic lesions and many activation pathways. Inhibiting MFB activation,promoting apoptosis or cell senescence can inhibit tissue fibrosis. In this paper,the research progress about the correlation between fibrosis and clinical manifestations of EMS,the sources and activation pathways of MFB in EMS,and targeting MFB to inhibit fibrosis are reviewed.
